In this thesis, I designed three open source Python libraries with the intention of creating a robust system that can accurately generate synthetic data. The goals of this thesis were to separate the different components in synthetic data generation into their own libraries. We identified these components as consisting of a way to transform the data, a way to model the data, and a way to recursively traverse the data set to model the relationships between the table as well as the data set itself. Once the libraries were implemented and functioning, we designed a program to run the synthetic data generation process in parallel on subsets of the original data. The goal of this program was to see if the overall modeling time could be reduced by modeling subsets in parallel and then averaging the parameters. In the end, we test how close these averaged parameters are to the original to see if this is a valid modeling technique.

Повний текст джерелаIn todayâ s ever-changing world of technology and information, a growing number of organizations and universities seek to store digital documents in an online, accessible manner. These digital library (DL) repositories are powerful systems that allow institutions to store their digital documents while permitting interaction and collaboration among users in their organizations. Despite the continual work on DL systems that can produce out-of-the-box online repositories, the installation, configuration, and customization processes of these systems are still far from straightforward.
Motivated by the arduous process of designing digital library instances; installing software packages like DSpace and Greenstone; and configuring, customizing, and populating such systems, we have developed an XML-based model for specifying the nature of DSpace digital libraries. The ability to map out a digital library to be created in a straightforward, XML-based way allows for the integration of such a specification with other DL tools. To make use of DL specifications for DSpace, we create a DL generator that uses these models of digital library systems to create, configure, customize, and populate DLs as specified. We draw heavily on previous work in understanding the nature of digital libraries from the 5S framework for digital libraries. This divides the concerns of digital libraries into a complex, formal representation of the elements that are basic to any minimal digital library system including Streams, Structures, Spaces, Scenarios, and Societies. We reflect on this previous work and provide a fresh application of the 5S framework to practical DL systems. Given our specification and generation process, we draw conclusions towards a more general model that would be suitable to characterize any DL platform with one specification.
We present this DSpace DL specification language and generator as an aid to DL designers and others interested in easing the specification of DSpace digital libraries. We believe that our method will not only enable users to create DLs more easily, but also gain a greater understanding about their desired DL structure, software, and digital libraries in general.

Повний текст джерелаThe project examines the information seeking behaviour of a small group of so-called Generation Y students at the Stellenbosch University as they undertake an academic 
assignment. There is consensus across the world that universities need to adapt to the needs of Generation Y students, brought up with high-level information technology, the internet and social networking. However, research shows that this does not mean that they are information literate. They have high-tech skills but often do not know how to analyse an information need or discriminate between information sources. Information scientist Carol Kuhlthau develop the ISP model upon which the study was based. Her model sees information seeking as a complex cognitive and affective process. Successful seekers have learned how to manage the process. University libraries need to adapt their information retrieval systems and services to meet the needs of their new kinds of students. The study, a small-scale intensive qualitative case study, hopes to provide insight into how they might do this. The researcher collected data while the participants were writing the assignment. Data gathering methods included interviews, journal writings and questionnaires.

Повний текст джерелаThe use and design of library spaces in higher education is a theme that has come to the forefront of user needs in the past few years. Academic libraries are redesigning their spaces to accommodate the 21st century user. The 21st century user is often labeled as Generation Y and requires a different set of knowledge and skills. In today’s knowledge economy, core skills include creativity, problem solving and collaboration. Libraries are creating space to accommodate the needs of these new users. They are developing group work areas, relaxation or pause areas, technology hubs and discussion rooms. This study has investigated how to redesign academic libraries, using Cape Peninsula University of Technology (CPUT) Cape Town branch as a pilot study. The researcher used the concept of library "as place" or "3ʳᵈ place" as a theory. The framework interrogated the ten qualities of a good library space. The researcher used a mixed method research design to conduct this study. A questionnaire of which multiple copies were made, interviews and internal statistics were used to gather data. The results of the study reflected that the library is functional, can be adapted, the space is accessible and varied, it is conducive to motivating and inspiring people, it does promote interactivity between users and library services, and is environmentally suitable, flexible and has a wow factor. All of these characteristics conform to McDonald’s ten qualities of a good library space.

An experimental study was performed to determine the effects on Corona Phase Molecular Recognition (CoMoRe) of bioconjugating a host of small molecules to DNA wrapped single-walled carbon nanotubes. In addition, the study observed the effects of the DNA sequence length on the subsequent effectiveness of the small molecules to alter the corona phase. The conjugation of small molecules was shown to alter both the intensity and the position of the fluorescence and absorbance profile. The length of the DNA sequence was found to change the small molecule's ability to alter the fluorecence spectra of the wrapped nanotubes. The EDC/sulfo-NHS reaction was done to conjugate the small molecules to two identical DNA sequences with varying lengthes. Through the methods of ultraviolet-visibile-near infrared absorption spectroscopy, near infrared fluorescence spectroscopy, and high-performance liquid chromatography characterization and structural analysis were performed. The results showed the successful conjugation of the small molecules to the amino-modified DNA and an alteration in the corona phase. The small molecules were found to bind to the DNA at multiple locations and the length of the sequence was found to have an effect on the corona phase.

Personalized on-demand robots have a vast potential to impact daily life and change how people interact with technology, but so far this potential has remained largely untapped. Building robots is typically restricted to experts due to the extensive knowledge, experience, and resources required. This thesis aims to remove these barriers with an end-to-end system for intuitively designing robots from high-level specifications. By describing an envisioned structure or behavior, casual users can immediately build and use a robot for their task. The presented work encourages users to treat robots for physical tasks as they would treat software for computational tasks. By simplifying the design process and fostering an iterative approach, it moves towards the proliferation of on-demand custom robots that can address applications including education, healthcare, disaster aid, and everyday life. Users can intuitively compose modular components from an integrated library into complex electromechanical devices. The system provides design feedback, performs verification, makes any required modifications, and then co-designs the underlying subsystems to generate wiring instructions, mechanical drawings, microcontroller code for autonomous behavior, and user interface software. The current work features printable origami-inspired foldable robots as well as general electromechanical devices, and is extensible to many fabrication techniques. Building upon this foundation, tools are provided that allow users to describe functionality rather than structure, simulate robot systems, and explore design spaces to achieve behavioral guarantees. The presented system allows non-engineering users to rapidly fabricate customized robots, facilitating the proliferation of robots in everyday life. It thereby marks an important step towards the realization of personal robots that have captured imaginations for decades.

Повний текст джерелаThis dissertation proposes, exemplifies, and validates the usage of course-subject co-occurrence (CSCO) data to generate topic maps of an academic discipline. CSCO is defined as course-subjects taught in the same academic year by the same teacher. This work is premised on the assumption that in the aggregate and for reasons of efficiency, faculty members teach course-subjects that are topically similar to one another. To exemplify and validate CSCO, more than 112,000 CSCO events were extracted from the annual directories of the American Association of Law Schools covering nearly eighty years of law school teaching in the United States. The CSCO events are used to extract and visualize the structure and evolution of law for the years 1931-32, 1972-73, and 2010-11—roughly, forty year intervals. Different normalization, ordination (layout), and clustering algorithms are compared and the best algorithm of each type is used to generate the final map. Validation studies demonstrate that CSCO produces topic maps that are consistent with expert opinion and four other indicators of the topical similarity of law school course-subjects. Resulting maps of the educational domain of law are useful as a reference system for additional thematic overlay of information about law school education in the United States. This research is the first to use CSCO to produce visualizations of a domain. It is the first to use an expanded, multi-part gold-standard to evaluate the validity of domain maps and the intermediate steps in their creation. Last but not least, this research contributes a metric analysis and visualizations of the evolution of law school course-subjects over nearly eighty years.

This report will try to solve this problem by creating own standard cell libraries, which in turn are more flexible since the user designs them. Having these libraries makes it possible to map VHDL or Verilog code to those libraries, using them instead of predefined cell libraries. The procedure of creating the libraries is quite time consuming, and thus the possibilities of making that procedure automatic, or as automatic as possible, has been examined. Unfortunately some manual labour has to be done, butthe process can be speeded up a lot by making parts of it automatic.

Повний текст джерелаDes avancées récentes dans le domaine du séquençage de prochaine génération ont ouvert une panoplie de façons de générer des données. Toutefois, chaque nouvelle méthode dévelopée est souvent appropriée à la caractérisation d’un seul type de phénomène ou de molécules. L’objectif de cette analyse est d’identifier la manière la plus appropriée de générer et traiter les données pour étudier les petits ARNs nucléolaires, snoRNAs. Récemment, ceux-ci ont été révélés comme des acteurs dans une variété de fonctions alternatives comme l’épissage alternatif, la résistance au choc oxidatif et l’état de la chromatine. Il est donc impératif de trouver une méthode qui puisse traiter une large quantité de données contenant les snoRNAs et leurs intéracteurs pour découvrir les rôles encore inexplorés des snoRNAs. Dans cette optique, un nouveau protocole a été élaboré. Cette nouvelle suite d’analyses s’appuie sur une reverse transcriptase isolée d’un intron de groupe II bactérien qui affiche une meilleure représentation des petits ARNs structurés comme les tRNAs et les snoRNAs. En effet, quand les données générées à travers la méthode de préparation des libraries pour petits ARNs standard est comparée à celle basée sur la reverse transcriptase bactérienne, cette dernière donne une meilleure représentation du compte des espèces. Ces avancées sont aussi présentes dans la méthode d’analyse informatique. La suite d’outils a été modifiée afin de permettre une meilleure détection des petits ARN non-codants. Ces modifications permettent de récupérer des millions de lectures par ensemble de données ce qui augmente le pouvoir prédictif de l’analyse.

Resumo: Devido à necessidade do desenvolvimento de fontes de energias renováveis é de grande interesse a descoberta de novas enzimas envolvidas na desconstrução da parede celular vegetal para a produção de bicombustíveis. A metagenômica é uma poderosa ferramenta para a descoberta de novos genes em comunidades microbianas que não são passíveis de cultivo pelas técnicas tradicionais. Neste contexto, o objetivo desta tese foi o desenvolvimento de estratégias metagenômicas para prospecção de novas enzimas atuantes na degradação da biomassa vegetal no metagenoma de solo de canavial bem como a caracterização funcional das mesmas. A biblioteca metagenômica construída com DNA extraído de um consórcio microbiano especializado na degradação de bagaço de cana-de-açúcar explodido a vapor e deslignificado foi empregada nos experimentos de triagem funcional de alto desempenho. Como resultado, foram identificados três clones positivos com atividade celulolítica e dois clones com atividade xilanolítica. A análise dos insertos de cada um dos clones resultou na localização de ORFs cujas sequências de aminoácidos apresentaram identidade com domínios conservados de glicosil hidrolases da família 5 (celulases E-1 e E-2), família 6 (celulase E-3), família 10 (xilanase X-1) e família 16 (glicosil hidrolase X-2). A celulase E-1 apresentou em sua estrutura além do domínio catalítico, E-1 Cat, um domínio de ligação a carboidratos, denominado E-1 CBM, que não apresentou identidade de sequência com domínios conservados conhecidos. A análise funcional do E-1 CBM revelou tratar-se de um CBM específico para cadeias de glucano com grau de polimerização mínimo de cinco unidades de glicose. Ensaios de atividade enzimática em diferentes substratos mostraram que E-1 Cat atuou especificamente na hidrólise das ligações glicosídicas do tipo ß(1,4) entre resíduos de glicose. Os maiores valores de atividade enzimática foram obtidos em pH 7,0 e temperatura de 50ºC. Os parâmetros cinéticos calculados em CMC foram Km igual a 6,05 ± 0,37 mg/mL, Vmax de 42,51 ± 1,2 ?mol/min/mg e eficiência catalítica kcat/Km de 4,06 mL/mg/s. A enzima apresentou termoestabilidade a 40ºC por cinco horas. A atividade enzimática de E-1 Cat em celulose cristalina e bagaço de cana-de-açúcar explodido a vapor resultou na liberação de açúcares solúveis, evidenciando sua potencial aplicação em processos de conversão da biomassa vegetal. Ensaios de atividade em diferentes substratos mostraram que X-1 apresentou maior atividade enzimática em xilana não ramificada, nas condições de pH e temperatura de 6,0 e 45ºC, respectivamente. Os parâmetros cinéticos calculados utilizando como substrato xilana de madeira de faia foram Km de 2,18 ± 0,13 mg/mL, Vmax de 1.435 ± 30,4 ?mol/min/mg e kcat/Km de 496,32 mL/mg/s. Em relação à termoestabilidade, a enzima se manteve estável a 40ºC e 50ºC por seis horas. A hidrólise de substratos complexos com X-1 resultou na liberação de xilooligossacarídeos, xilobiose e xilose, que são compostos que apresentam potencial aplicação nas indústrias alimentícias e de biocombustíveis. Os resultados obtidos neste estudo validaram a abordagem metagenômica desenvolvida para a descoberta de novos genes codificantes para glicosil hidrolases. Além disso, a estratégia descrita nesta tese pode ser estendida para a descoberta de uma miríade de bioprodutos de interesse biotecnológico
Abstract: Due to the necessity of development of renewable sources of energy, it is of great interest the discovery of novel enzymes involved in plant cell wall deconstruction for biofuels production. Metagenomics is a powerful tool for the discovery of novel genes in microbial communities that are not liable to cultivation by traditional techniques. In this context, the aim of this thesis was the development of metagenomic strategies for prospection of novel enzymes involved in plant biomass degradation in sugarcane field soil metagenome and functional characterization of the identified enzymes. The metagenomic library constructed with DNA extracted from a microbial consortium specialized in degradation of steam exploded delignified sugarcane bagasse was used in the experiments of high-performance functional screening. As a result, we identified three positive clones with cellulolytic activity and two clones with xylanolytic activity. The analysis of the inserts from each clone resulted in the location of ORFs whose amino acid sequences showed identity to conserved domains of glycoside hydrolase family 5 (cellulases E-1 and E-2), family 6 (cellulase E-3), family 10 (xylanase X-1) and family 16 (glycoside hydrolase X-2). Cellulase E-1 exhibited in addition to the catalytic domain, E-1 Cat, a carbohydrate binding module, called E-1 CBM, which showed no sequence identity with known conserved domains. Functional analysis of E-1 CBM showed that it is a CBM specific for glucan chains with a degree of polymerization of at least five units of glucose. Assays with a set of different substrates revealed that E-1 Cat hydrolyzed specifically ß(1,4) glycoside bonds between glucose residues. The highest value of enzymatic activity was obtained at pH 7.0 and temperature of 50°C. The kinetic parameters Km, Vmax and catalytic efficiency kcat/Km calculated using CMC were 6.05 ± 0.37 mg/mL, 42.51 ± 1.2 ?mol/min/mg and 4.06 mL/mg/s, respectively. The enzyme showed thermal stability at 40°C for five hours. The enzymatic activity of E-1 Cat in crystalline cellulose and steam exploded sugarcane bagasse resulted in the release of soluble sugars, demonstrating its potential application in processes of biomass conversion. The xylanase X-1 showed higher enzyme activity in debranched xylan, in reactions conducted in pH 6.0 and temperature of 45°C. The kinetic parameters Km, Vmax and catalytic efficiency kcat/Km calculated using beechwood xylan were 2.18 ± 0.13 mg/mL, 1,435 ± 30.4 ?mol/min/mg and 496.32 mL/mg/s, respectively. In relation to thermal stability, the enzyme was stable at 40°C and 50°C for six hours. The hydrolysis of complex substrates resulted in the release of xylo-oligosaccharides, xylobiose and xylose, which are compounds that have potential application in food and biofuels industries. The results of this study validated the metagenomic approach developed for the discovery of novel genes coding for glycoside hydrolases. Moreover, the strategy described in this work can be extended to the discovery of a myriad of byproducts of biotechnological interest

Повний текст джерелаEste trabajo de doctorado, desarrollado en la Universitat Politècnica de València (UPV), tiene como objetivo cubrir la primera fase del benchmark presentado por el grupo de expertos Uncertainty Analysis in Modeling (UAM-LWR). La principal contribución al benchmark, por parte del autor de esta tesis, es el desarrollo de un programa de MATLAB solicitado por los organizadores del benchmark, el cual se usa para generar librerías neutrónicas a distribuir entre los participantes del benchmark. El benchmark del UAM pretende determinar la incertidumbre introducida por los códigos multifísicos y multiescala acoplados de análisis de reactores de agua ligera. El citado benchmark se divide en tres fases: 1. Fase neutrónica: obtener los parámetros neutrónicos y secciones eficaces del problema específico colapsados y homogenizados, además del análisis de criticidad. 2. Fase de núcleo: análisis termo-hidráulico y neutrónico por separado. 3. Fase de sistema: análisis termo-hidráulico y neutrónico acoplados. En esta tesis se completan los principales objetivos de la primera fase. Concretamente, se desarrolla una metodología para propagar la incertidumbre de secciones eficaces y otros parámetros neutrónicos a través de un código lattice y un simulador de núcleo. Se lleva a cabo un análisis de incertidumbre y sensibilidad para las secciones eficaces contenidas en la librería neutrónica ENDF/B-VII. Su incertidumbre se propaga a través del código lattice SCALE6.2.1, incluyendo las fases de colapsación y homogenización, hasta llegar a la generación de una librería neutrónica específica del problema. Luego, la incertidumbre contenida en dicha librería puede continuar propagándose a través de un simulador de núcleo, para este estudio PARCSv3.2. Para el análisis de incertidumbre y sensibilidad se ha usado el módulo SAMPLER -disponible en la última versión de SCALE- y la herramienta estadística DAKOTA 6.3. Como parte de este proceso, también se ha desarrollado una metodología para obtener librerías neutrónicas en formato NEMTAB para ser usadas en simuladores de núcleo. Se ha realizado una comparación con el código CASMO-4 para obtener una verificación de la metodología completa. Esta se ha probado usando un reactor de agua en ebullición del tipo BWR. Sin embargo, no hay ninguna preocupación o limitación respecto a su uso con otro tipo de reactor nuclear. Para la cuantificación de la incertidumbre se usa la metodología estocástica Gesellschaft für Anlagen und Reaktorsicherheit (GRS). Esta metodología hace uso del modelo de alta fidelidad y un muestreo no paramétrico para propagar la incertidumbre. Como resultado, el número de muestras (determinado con la fórmula revisada de Wilks) no depende del número de parámetros de entrada, sólo depende del nivel de confianza e incertidumbre deseados de los parámetros de salida. Además, las funciones de distribución de probabilidad no están limitadas a normalidad. El principal inconveniente es que se ha de disponer de las distribuciones de probabilidad de cada parámetro de entrada. Si es posible, las distribuciones de probabilidad de entrada se definen usando información encontrada en la literatura relacionada. En caso contrario, la incertidumbre se define en base a la opinión de un experto. Se usa un segundo escenario para propagar la incertidumbre de diferentes parámetros termo-hidráulicos a través del código acoplado TRACE5.0p3/PARCSv3.0. En este caso, se utiliza un reactor tipo PWR para simular un transitorio de una caída de barra. Como nueva característica, el núcleo se modela elemento a elemento siguiendo una discretización totalmente en 3D. No se ha encontrado ningún otro estudio que use un núcleo tan detallado en 3D. También se usa la metodología GRS y el DAKOTA 6.3 para este análisis de incertidumbre y sensibilidad.
Aquest treball de doctorat, desenvolupat a la Universitat Politècnica de València (UPV), té com a objectiu cobrir la primera fase del benchmark presentat pel grup d'experts Uncertainty Analysis in Modeling (UAM-LWR). Es porta a terme una anàlisi d'incertesa i sensibilitat per a les seccions eficaces contingudes en la llibreria neutrònica ENDF/B-VII. La seua incertesa es propaga a través del codi lattice SCALE6.2.1, incloent les fases per col¿lapsar i homogeneïtzar, fins aplegar a la generació d'una llibreria neutrònica específica del problema. Després, la incertesa continguda en la esmentada llibreria pot continuar propagant-se a través d'un simulador de nucli, per a aquest estudi PARCSv3.2. Per a l'anàlisi d'incertesa i sensibilitat s'ha utilitzat el mòdul SAMPLER -disponible a l'última versió de SCALE- i la ferramenta estadística DAKOTA 6.3. Com a part d'aquest procés, també es desenvolupa una metodologia per a obtenir llibreries neutròniques en format NEMTAB per ser utilitzades en simuladors de nucli. Aquesta metodologia fa ús del model d'alta fidelitat i un mostreig no paramètric per propagar la incertesa. Com a resultat, el nombre de mostres (determinat amb la fórmula revisada de Wilks) no depèn del nombre de paràmetres d'entrada, sols depèn del nivell de confiança i incertesa desitjats dels paràmetres d'eixida. A més, las funcions de distribució de probabilitat no estan limitades a la normalitat. El principal inconvenient és que s'ha de disposar de les distribucions de probabilitat de cada paràmetre d'entrada. Si és possible, les distribucions de probabilitat d'entrada es defineixen utilitzant informació trobada a la literatura relacionada. En cas contrari, la incertesa es defineix en base a l'opinió d'un expert. S'utilitza un segon escenari per propagar la incertesa de diferents paràmetres termo-hidràulics a través del codi acoblat TRACE5.0p3/PARCSv3.0. En aquest cas, s'utilitza un reactor tipus PWR per simular un transitori d'una caiguda de barra. Com a nova característica, cal assenyalar que el nucli es modela element a element seguint una discretizació totalment 3D. No s'ha trobat cap altre estudi que utilitze un nucli tan detallat en 3D. També s'utilitza la metodologia GRS i el DAKOTA 6.3 per a aquesta anàlisi d'incertesa i sensibilitat.¿

Повний текст джерелаThe objective of this research is to evaluate the contribution of social networking sites of the Web in improving communication and the quality of services and products offered in academic libraries. This is an exploratory research with quantitative and qualitative approaches. We have submitted questionnaires to the libraries of UNESP (São Paulo State University), UNICAMP (University of Campinas) and USP (University of São Paulo) and conducted interviews with a sample of libraries of these institutions. The results show that the libraries are still experimenting with the use of social networks. We have observed that the tools provide new forms of communication and dissemination of information, services and products for libraries, as well as build capacity for the users. This results in innovations and adds value to the existing processes. At the same time, we have understood that the social networks are not characterized only as spaces for communication and the dissemination of information, but also of participation, collaboration and interaction, all of which are still little explored by these libraries. In spite of this, we believe that social networks can contribute to the improvement of communication and the quality of services and products offered by libraries
