ABSTRACT The d105 dominant-negative mutant form of the herpes simplex virus 1 (HSV-1) single-stranded DNA-binding protein, ICP8 (d105 ICP8), inhibits wild-type viral replication, and it blocks both viral DNA replication and late gene transcription, although to different degrees (M. Gao and D. M. Knipe, J. Virol. 65:2666–2675, 1991; Y. M. Chen and D. M. Knipe, Virology 221:281–290, 1996). We demonstrate here that this protein is also capable of preventing the formation of intranuclear prereplicative sites and replication compartments during HSV infection. We defined three patterns of ICP8 localization using indirect immunofluorescence staining of HSV-1-infected cells: large replication compartments, small compartments, and no specific intranuclear localization of ICP8. Cells that form large replication compartments replicate viral DNA and express late genes. Cells that form small replication compartments replicate viral DNA but do not express late genes, while cells without viral replication compartments are incapable of both DNA replication and late gene expression. The d105 ICP8 protein blocks formation of prereplicative sites and large replication compartments in 80% of infected cells and formation of large replication compartments in the remaining 20% of infected cells. The phenotype ofd105 suggests a correlation between formation of large replication compartments and late gene expression and a role for intranuclear rearrangement of viral DNA and bound proteins in activation of late gene transcription. Thus, these results provide evidence for specialized machinery for late gene expression within replication compartments.

ABSTRACT Members of the Arenaviridae family have been isolated from mammalian hosts in disparate geographic locations, leading to their grouping as Old World types (i.e., lymphocytic choriomeningitis virus [LCMV], Lassa fever virus [LFV], Mopeia virus, and Mobala virus) and New World types (i.e., Junin, Machupo, Tacaribe, and Sabia viruses) (C. J. Peters, M. J. Buchmeier, P. E. Rollin, and T. G. Ksiazek, p. 1521–1551, in B. N. Fields, D. M. Knipe, and P. M. Howley [ed.], Fields virology, 3rd ed., 1996; P. J. Southern, p. 1505–1519, in B. N. Fields, D. M. Knipe, and P. M. Howley [ed.], Fields virology, 3rd ed., 1996). Several types in both groups—LFV, Junin, Machupo, and Sabia viruses—cause severe and often lethal human diseases. By sequence comparison, we noted that eight Old World and New World arenaviruses share several amino acids with the nucleoprotein (NP) that consists of amino acids (aa) 118 to 126 (NP 118–126) (RPQASGVYM) of LCMV that comprise the immunodominant cytotoxic T-lymphocyte (CTL) epitope forH-2 d mice (32). This Ld-restricted epitope constituted >97% of the total bulk CTLs produced in the specific antiviral or clonal responses ofH-2 d BALB mice. NP 118–126 of the Old World arenaviruses LFV, Mopeia virus, and LCMV and the New World arenavirus Sabia virus bound at high affinity to Ld. The primary H-2 d CTL anti-LCMV response as well as that of a CTL clone responsive to LCMV NP 118–126 recognized target cells coated with NP 118–126 peptides derived from LCMV, LFV, and Mopeia virus but not Sabia virus, indicating that a common functional NP epitope exists among Old World arenaviruses. Use of site-specific amino acid exchanges in the NP CTL epitope among these arenaviruses identified amino acids involved in major histocompatibility complex binding and CTL recognition.

Following the attacks on the New York World Trade Center on September 11, 2001, the EMDR Humanitarian Assistance Program initiated a response establishing the New York City Disaster Mental Health Recovery Network. The network provided coordination and assistance to local psychotherapists who volunteered to provide treatment to individuals directly affected by the tragedy. The psychotherapists utilized both the EMDR standard protocol and the EMDR Recent Events protocol during the initial aftermath and ongoing recovery at the World Trade Center site. The development of the network is reviewed, and detailed descriptions are provided regarding three cases to illustrate the use of the EMDR Recent Events protocol. The research findings reported by Silver, Rogers, Knipe, & Colelli (2005) that demonstrated support for EMDR as a postdisaster treatment are summarized. Further research is recommended.

Abstract. Macrofaunal and foraminiferal analyses have shown the Woodbine Shale in South Cumbria to be of latest early Asbian age. Ostracods and solid foraminifera from the Woodbine Shale are described and illustrated for the first time. Several ostracods including Rectobairdia cf. dorssenata, Healdia ‘penchfordensis’, Bairdia cf. leguminoides, Pustulobairdia cf. confragosa and Microcheilinella subcorbulinoides, previously only known from late Asbian or younger strata, have now been shown to range into the early Asbian. The numerous bairdiids, heavily ornamented ostracods, trilobites, brachiopods and abundant foraminifera suggest a deep, subtidal, marine environment of deposition for the Woodbine Shale. Foraminifera from the underlying Lower Urswick Limestone suggest an early Asbian age for this formation whilst the overlying Upper Urswick Limestone is shown to be of late Asbian age. The foraminiferal assemblages from these beds can be correlated directly with those in the early Asbian Upper Potts Beck Limestone and the late Asbian Knipe Scar Limestone in the Asbian stratotype section at Little Asby Scar, Cumbria.

Purpose The purpose of this paper is to describe the Eye movement desensitisation and reprocessing (EMDR) treatment of an adult female patient detained within a high secure hospital with complex mental health difficulties, including complex trauma, factitious disorder, self-injury and a history of offending. The EMDR treatment addressed the patient’s urges to engage in severe and sometimes life-threatening self-injury, a primary motive of which was to access physical healthcare interventions within a general hospital. The paper describes the wide-ranging benefits of the treatment and incorporates feedback from the patient and clinicians within her multi-disciplinary team (MDT). Design/methodology/approach Four triggers for self-injury were processed during the therapy using the DeTUR Protocol (Popky, 2005, 2009) and the Constant Installation of Present Orientation and Safety (CIPOS, Knipe, 2009a) method. In total, 18 one hour therapy sessions were delivered plus three follow-up sessions to continue to offer support and complete the post-treatment evaluation. Findings The level of urge for each trigger was reduced to 0 which the patient defined as “no urge to self-injure”. Benefits went well beyond self-injury with reported positive impacts on mood, thinking, sleep, concentration, memory and experience of flashbacks. Practical implications This case report demonstrates that the EMDR DeTUR Protocol together with the CIPOS method can be extremely valuable in the treatment of patients who self-injure. Originality/value The case report offers an important contribution to an area that requires much further research.

ABSTRACT The herpes simplex virus genome can enter a repressed transcriptional state (latency) in sensory neurons of the host nervous system. Although reduced permissiveness of the neuronal environment is widely accepted as a causal factor, the molecular pathway(s) directing and maintaining the viral genome in the latent state remains undefined. Over the past decade, the field has been strongly influenced by the observations of Kosz-Vnenchak et al., which have been interpreted to indicate that, in sensory neurons in vivo, a critical level of viral DNA synthesis within the neuron is required for sufficient viral immediate-early (IE) and early (E) gene expression (M. Kosz-Vnenchak, J. Jacobson, D. M. Coen, and D. M. Knipe, J. Virol. 67:5383-5393, 1993). The levels of IE and E genes are, in turn, thought to regulate the decision to enter the lytic cycle or latency. We have reexamined this issue using new strategies for in situ detection and quantification of viral gene expression in whole tissues. Our results using thymidine kinase-null and rescued mutants as well as wild-type strains in conjunction with viral DNA synthesis blockers demonstrate that (i) despite inhibition of viral DNA replication, many neurons express lytic viral proteins, including IE proteins, during acute infection in the ganglion; (ii) at early times postinoculation, the number of neurons expressing viral proteins in the ganglion is not reduced by inhibition of viral DNA replication; and (iii) following a reactivation stimulus, the numbers of neurons and apparent levels of lytic viral proteins, including IE proteins, are not reduced by inhibition of viral DNA replication. We conclude that viral DNA replication in the neuron per se does not regulate IE gene expression or entry into the lytic cycle.

The process of cutting is widely used during the stockpiling and preparation of fodder - the processing of stems by the direct application of the force that arises at the cutting edge of the knife. Almost 85% of stem fodder are ground fodder for the stockpiling of which the forage harvesters are used. Grinding of leaf mass is one of the most energy-consuming operations in the general process of stockpiling and the final quality of the fodder depends on it. The energy intensity of the grinding process is influenced by both the constructive and operating parameters of the machine, the cutting pair, and the physical and mechanical properties of the leaf-stem mass. Therefore, the development of grinding machines, the biter-knife in particular, should be carried out on the basis of detailed studies of the process of cutting grass mass, taking into account the current power and energy parameters. The object of the study in this article was the process of cutting a herbal mass with a biter-knife grinding machine containing an active disk knives. The purpose of the work was to establish the analytical dependence of the destructive cutting force in the interaction of the blade of the disk knive of biter-knife grinding machine with herbal mass. The task of the work was: to analyze the researches and publications on establishing the effort of cutting a layer of leaf stem and influence of various factors on its magnitude; to establish a mathematical model of calculation the critical effort of cutting a layer of grass mass with a cutting pair: double finger of the feed rotor - a disk knife during the cutting with sliding. The research methodology was based on the analysis of the interaction of the working parts of the grinding machine with herbal mass using the methods of theoretical mechanics. In the scientific work the interaction of single-blade blade of a disk knife of a biter-knife grinding machine with a leaf-stem mass is considered under the condition of double-backed cutting with sliding. The force interaction of the knife`s blade with the grass mass was analyzed and the dependence (3) of the calculation of the critical destructive cutting force Rkr was determined. From the obtained expression it is established that its magnitude is influenced by both the constructive parameters of the cutting steam and the technological parameters of the grinding machine. Also the magnitude of the destructive force is affected by the transformed angle of sharpening of the blade of the disk knife and the sliding angle.

The focus of this study was on how different knife characteristics affect the consumer’s ability to slice vegetables. The aim of this study was to investigate if there is a difference in cutting between a chef knife and a santoku knife, a ceramic knife and a stainless-steel knife, and a sharp and dull knife in terms of muscle activation, body part discomfort, time, and slice performance. The results show for the consumer that knife characteristics do not affect the user’s performance.

The sharpness of the knife used for slaughter is of the utmost importance from an animal welfare perspective. The quantification of knife sharpness is almost impossible in abattoirs. The sharpness of the knife blade used to slaughter an animal, as well as its effects on animals’ pain and stress levels, is an important area of investigation that needs to be addressed. The objective of this study was to evaluate the effects of knife sharpness on blood biochemical parameters, plasma catecholamines, and electroencephalographic (EEG) responses. Twenty Brahman crossbred steers were either subjected to slaughter with a sharp knife (n = 10) or a commercial sharp knife (n = 10); knife sharpness was measured with the ANAGO® sharpness tester. There was significant increase in adrenaline (p < 0.0001), glucose (p = 0.0167), creatinine kinase (p = 0.0123) and lactate dehydrogenase (p = 0.0151) at post-slaughter compared to pre-slaughter in commercial sharp knife group than in thesharp knife group. A significant increase was observed in the median frequency (p < 0.0001) and total power (p < 0.0001) of the EEG, the parameters for pain and stress, in the animals slaughtered with the commercial sharp knife than those slaughtered with the sharp knife. Thus, EEG results also supported the hormonal and biochemical results. From the results, it is concluded that animals slaughtered with a sharp knife experienced the least amount of pain and stress compared to those slaughtered with a commercial sharp knife.

Backgrounds. With respect to the knife’s design in colorectal endoscopic submucosal dissection (ESD), diameter, water jet function, and electric power are important because these relate to efficient dissection. In this study, we analyzed a novel, narrow ball tip-typed ESD knife with water jet function (Flush knife BT-S, diameter: 2.2 mm, length: 2000 mm, Fujifilm Co., Tokyo, Japan) compared to a regular diameter knife (Flush knife BT, diameter: 2.6 mm, length: 1800 mm).Methods. In laboratory and clinical research, electric power, knife insertion time, vacuum/suction amount with knife in the endoscopic channel, and water jet function were analyzed. We used a knife 2.0 mm long for BT-S and BT knives.Results. The BT-S showed faster mean knife insertion time (sec) and better vacuum amount (ml/min) compared to the BT (insertion time: 16.7 versus 21.6,p<0.001, vacuum amount: 38.0 versus 14.0,p<0.01). Additionally, the water jet function of the BT-S was not inferior. In 39 colorectal ESD cases in two institutions, there were mean 4.7 times (range: 1–28) of knife insertion. Suction under knife happened 59% (23/39) and suction of fluid could be done in 100%.Conclusions. Our study showed that the narrow knife allows significantly faster knife insertion, better vacuum function, and effective clinical results.
