ABSTRACT Mouse hepatoma × rat hepatocyte hybrids that segregate rat chromosomes were used to determine the chromosomal location of the rat genes encoding 11 β-hydroxylase and aldosterone synthase (Cyp11b1 and Cyp11b2 respectively). By means of species-specific restriction fragments and microsatellite markers both genes were mapped to rat chromosome 7. The Cyp11b1 microsatellite marker was subsequently found to vary in length between and within rat strains. Furthermore, we compared the sequences of Cyp11b1 markers in two genetically hypertensive strains of rat with their normotensive counterparts. Previous studies have indicated that 11β-hydroxylase activities in Milan and Lyon hypertensive strains are different from their respective genetic controls. The Cyp11b1 microsatellite regions from Lyon hypotensive and normotensive strains of rat were similar and were both shorter by 15 bases than that of the Lyon hypertensive strain. The Cyp11b1 marker in Milan hypertensive (MHS) and normotensive (MNS) strains differ from all the Lyon strains and from each other. The MHS marker is 12 bases shorter than that of MNS rats. These differences in microsatellite length may provide useful polymorphic markers in co-segregation studies of genetic hypertension in rats.

Purpose – Libraries face a “selling” job on the relationship between print and digital as the mode of delivery for content. Too often it is limply said that “everything is on the web” when we know that it is not or that licensed access to the content is not always available. The Lyon’s Declaration promotes freedom of access to information, yet libraries live and work with this paradox. How will the ambitions of the Lyon Declaration be met? So while it is ironic that everything is said to be on the web while it is not, we have powerful evidence of their dynamic purpose and value. This conference series has dealt with this confronting riddle, evolving and refining, but not quite finding the defining moment. The purpose of this paper is to explore the many interfaces here. By understanding this paradox of print and digital we should see the future of the repositories and libraries more clearly and position them more exactly. Design/methodology/approach – The paper explores the rich divergence of responses to the use and development of repositories and proposes future directions. It is an experiential paper as well as one guided by future planning perspectives. Findings – There is a need for a reconceptualisation of the role of print repositories blending in digital solutions together with the more traditional solutions. There is also a strong need for repositories to collaborate internationally in order to be able to render their own work and collections valid and effective in a much wider context. Originality/value – This paper is a series of observations and lessons. It is an extension of practical and managerial work in developing and managing repositories and their collections both in Australia and in Hong Kong. It is a collection views designed to stimulate and potentially guide library practitioners who are thinking and working in this area.

During a recent job as a field technician, I spent time in the burn area of the Beachie Creek Fire, which struck the Lyons-Detroit corridor in August 2020. The fire burned nearly 200,000 acres of old-growth forest, and countless people lost their homes. In the two years that have passed since the incident, however, the scarred hills have gradually grown brighter as foxgloves and other early-successional flora take hold. While wildfires can have devastating effects on human populations—effects that I do not at all intend to diminish—wildfires are a natural part of Oregon’s landscape, and they open up ecological niches for a plethora of wonderful meadow species that cannot tolerate the shady conditions created by old-growth canopies. This photo juxtaposes the human cost of wildfires—embodied by the abandoned, rusted trailer—with the ecological rejuvenation of open skies and ash-rich soil.

SUMMARYAim– To examine the associations of job acquisition and loss in a representative, prospective community sample of people with schizophrenia living in the UK, France and Germany.Method– A representative sample of twelve hundred and eight people with schizophrenia were recruited from selected secondary mental health services in the U.K, France and Germany and followed up for 2 years. Information on demographic details, psychotic symptoms and work status was collected.Results– The odds of getting jobs were increased by being resident in Marseille, Leipzig, Hemer and Heilbronn and by a higher regional general population employment rate. The odds were reduced by living in Lyon, a later illness onset, a longer length of illness, a continuous illness course and more severe negative psychotic symptoms. Previous vocational training reduced the odds of losing employment, whilst living in Lyon or Leipzig, harmful use of alcohol and more positive psychotic symptoms at baseline all increased the odds.Conclusions– In addition to illness related factors, area of residence and local labour market conditions appear to be important in explaining employment status change in people with schizophrenia.Declaration of Interest: All authors declare there are no conflicts of interest. The identification of mutations in the MEN1 gene causing MEN1 has represented a challenge since the cloning of the gene in 1997 because of the lack of mutation hot-spots in the gene and the lack of phenotype–genotype correlations. The use of denaturing high performance liquid chromatography (DHPLC), a high throughput, reliable and automated heteroduplex-based technique, is the ideal for mutation detection in MEN1. In this work, DHPLC was optimised for the screening of the nine coding exons and splice junctions of MEN1. Thanks to collaboration between two French laboratories recognised as reference centres for genotypic MEN1 diagnosis (Lyon and Lille), a blind retrospective study conducted in a cohort of 160 unrelated MEN1 probands with (or without) known germline mutations was undertaken to evaluate the sensitivity of DHPLC. We were able to detect 101 different sequence variations by DHPLC, distributed in the 10 analysed DNA fragments and corresponding to 100% of mutation detection compared with direct sequencing. 1·2% of samples were considered as false positive, exhibiting a heterogenous profile. DHPLC did not detect five cases of deletion or duplication of complete exons, neither did direct sequencing, showing the limits of the technique. Nevertheless, the method appeared to allow automated, rapid and low-cost mutation detection with high accuracy. Direct sequencing can be then applied to identify the sequence variations on the targeted DNA fragments showing heterozygous profile by DHPLC. In conclusion, genotypic diagnosis of MEN1 can benefit from DHPLC in terms of efficacy, rapidity and cost.

Changes in working patterns mean that the youngest generations in the work force, millennials and generation Z, are facing more organisational and job changes than previous generations. Specifically, millennials, defined as the generation born between 1982 and 1999 (Twenge, Campbell, Hoffman & Lance, 2010), experience between two and three times as many job and organisational moves as previous generations (Lyons, Schweitzer & Ng, 2015), with 60 per cent of millennials reporting that they are open to a different job opportunity (Gallup, 2016). Research suggests that key resources such as optimism and self-efficacy are important elements of career adaptability (Haenggli & Hirschi, 2020). The field of positive psychology, which was first introduced to the academic community in 2000 (Seligman & Csikszentmihalyi, 2000), aims to better understand and foster valued subjective experiences, such as wellbeing, resilience and optimism. More recently, positive psychology has integrated with coaching psychology to create a new approach to coaching, positive psychology coaching, which can be defined as ‘evidence-based coaching practice informed by the theories and research of positive psychology for the enhancement of resilience, achievement and wellbeing’ (Green & Palmer, 2019, p.10). With job and career changes becoming increasingly likely for the millennial generation, there is an opportunity to look at how positive psychology coaching can be used to support the experience of individuals within this generation, helping them to enhance outcomes. There has been some research conducted within this field, with one study suggesting that positive psychology coaching can be beneficial in boosting career confidence in potential career changers (Archer & Yates, 2017), however, the existing research is limited, and doesn’t specifically focus on the millennial generation. One positive psychology intervention that could be particularly relevant for this population is the Best Possible Self (BPS) intervention (King, 2001). This intervention encourages individuals to imagine a best possible future self, before then writing about what they imagined. Previous research has shown that the BPS intervention is linked to increased positive affect, increased optimism and increased levels of self-concordant motivation (Sheldon & Lyubomirsky, 2006), and there is also evidence to suggest that mental stimulation can support psychological adjustment to change (Rivkin & Taylor, 1999). This study endeavours to contribute to the research by understanding how a positive psychology intervention (specifically, the BPS intervention (King, 2001)) is experienced by millennials who are considering a career change, before considering the implications of this for career coaching.

In this reply to the commentaries of Lyon and Swenson, I clarify the arguments of my article, “On the nature and origins of cognition as a form of motivated activity.” In particular, I focus on the nature of self-preserving behavior, considered not only as a basic and universal form of cognitive behavior but also as a key example of cognition as form of motivated activity. I clarify that my approach affirms the importance of self-preserving behavior but questions the widespread assumption that this and other varieties of motivated behavior can be explained as the product of homeostatic mechanisms. I suggest that extremal properties can not only do a better job of explaining homeostasis-like behaviors, but they can also be extended to other kinds of motivated behavior that are not homeostasis-like. Finally, I emphasize the explanatory promise of extremal properties with respect to fundamental questions about the emergence of order in living and nonliving systems.

In the Federal Elections of August 1943 the United Australia Party(UAP), which had been in office for nearly ten consecutive years suffered a resounding defeat and the result was a landslide for Labor, giving it an absolute majority in both Houses. This thesis traces the course of the disintegration of the United Australia Party to try to fill a gap in the historiography by explaining how and why an apparently popular political party was so categorically rejected by the electorate in 1943 that it subsequently vanished from the political scene as if it had never existed.

Que signifie la lucidité dans la pensée de E. M. Cioran (1911-1995), sachant que son pessimisme radical laisse pourtant place à une jubilation non moins intense ? Cioran assigne une direction singulière à la pensée contemporaine, direction dont elle n'a pas fini de s'inquiéter : il est question de « recommencer la Connaissance » et de ménager néanmoins un « hédonisme à rebours ». A l'aide d'une double herméneutique (de l'infini et de la clairvoyance), cette thèse se propose d'enquêter sur la philosophie cioranienne du pire à l'aune de ses enjeux métaphysiques, et s'articule selon cinq moments distincts : en partant d'une détermination de ce que signifie précisément ce pessimisme, en s'interrogeant ensuite sur la distinction fondamentale et charnière de la chute dans le temps et du temps, puis en cernant les différents degrés de la lucidité elle-même, avant de retracer son ontologie négative, pour finalement aboutir à une étude des limites métaphysiques, heuristiques et affectives de cette lucidité. L'écartèlement de la conscience lucide est au cœur d'une refonte de la notion de sujet, scindé entre le soi et le moi, déchiré entre conscience et vie, partagé entre la tentation de mourir et la tentation d'exister, tiraillé entre la connaissance du mal et la jubilation. What is the meaning of “insight” in E. M. Cioran's philosophy (1911-1995), when one knows that his radical pessimism however leaves some space for jubilation which is all the more intense? Cioran ascribes a specific direction to contemporary philosophy, a direction which keeps worrying it: it is all about “starting Knowledge anew” and yet, to keep some “hedonism upside down”. Based on a double hermeneutics (of infinity and perceptiveness), this doctoral dissertation offers to investigate Cioran's philosophy of the worst by the yardstick of its metaphysical stakes; it is thus divided into five different parts: we will first begin by determining what Cioran's pessimism is really about, and we will question the fundamental and pivotal distinction between the fall in time and the fall from time; then, we will determine the various degrees of insight, before recounting its negative ontology, to finally offer a study of metaphysical, heuristic and affective limits to Cioran's insight. The quartering of consciousness is at the center of an overhaul of the notion of “subject”, split between the id and the ego, torn apart between consciousness and life, the temptation to die and the temptation to be, between the knowledge of evil and jubilation. The investigation followed in this dissertation seeks to fill in the space dug by the instable in-betweenness, endlessly swaying between an extreme and the other, and which scans the rhythm of an organic thought living at its own limits, a thought which is always on the existential go, and which is written between sense and silence

La période 1960 – 1990 a été agitée et féconde aux États-Unis malgré l’échec du projet de Grande Société et de lutte contre la pauvreté du président L.B. Johnson. L’Amérique a profité des retombées de sa domination économique et militaire pour financer l’éducation, le logement (G.I. Bill) ainsi que les arts (N.E.A.). De 1960 à 1990 la photographie est entrée massivement à l’université, dans les musées et dans le marché de l’art. Le paysage a toujours été un genre artistique privilégié aux États-Unis, de la peinture du dix-neuvième siècle à la photographie depuis son invention qui coïncide avec la découvertes de nouveaux territoires qui crée le pays. Le médium a documenté le développement territorial du pays puis s’est affirmé sur la scène de la photographie créative occidentale. Les années considérées vont voir la photographie créative américaine passer d’une période « romantique » tournée vers l’abstraction et le monde intérieur de l’artiste, conséquence des persécutions politiques de l’immédiat après-guerre, vers une réflexion ontologique et expérimentale, pour finalement traiter de problèmes de société tout en restant connectée aux réflexions esthétiques et philosophiques communes aux autres arts. Au sortir des années 1980, la photographie américaine de paysage, celle des grands espaces mais également celle des espaces humanisés, urbanisés, domine la scène internationale inspirant un renouveau du genre en Europe, au Canada, au Japon,…. Lancé par une exposition alors jugée mineure en 1975, le phénomène New Topographics est devenu planétaire et perdure. Ces quinze dernières années, de nombreuses expositions des paysagistes américains de cette période ont circulé à travers le monde, phénomène révélateur de leur rôle dans notre culture occidentalo – planétaire ainsi que pour l’histoire du médium
During the 1960 – 1990 period, in spite of the psychological and economical fall-outs of the various wars (Cold War, Korea and Vietnam ) undermining L.B. Johnson’s hopes and plans for a Great Society and his War on Poverty, the American government used its world supremacy and the derived wealth acquired in the wake of W.W. II (the USA was the only western country whose industrial production was impacted positively) to finance popular housing, adult education (G.I. Bill), and the arts (N.E.A.). During those years photography crashed the doors of academia, museum and art institutions, and entered the art market. Landscape has always been a major genre in the American visual arts, from the paintings of the nineteenth century (the Hudson River School, the Luminists) to photography. An interesting synchronicity can be observed between the birth, growth and coming of age of both the medium and the country. Landscape photography participated in the creation of an American identity. A century later, during what we can now call the Golden Age of American landscape photography from New Topographics in the 1970s to the advent of color photography in the 1980s, photographers turned their lenses back toward the east at the damage done and the state of the landscape left behind. The production of wall-size prints followed, competing for attention with paintings on the walls of museums and galleries that welcome them. Since the Culture Wars of the late 1980s and the 1990s, and the defunding of the arts that ensued, the rest of the world has caught up, influenced by the traveling exhibitions and publications of that generation of American photographers

Longtime Brubeck champion Jimmy Lyons, who had first brought Brubeck’s music to the public in 1949, saw the opportunity to premiere The Real Ambassadors at the 1962 Monterey Jazz Festival and set about booking all the artists featured on the soundtrack to perform their regular sets. He cleverly added a clause into each performer's contract stipulating an additional hour of performing on Sunday night with no other details. This would be to premiere the show. However, when Joe Glaser learned of the plan, he immediately contacted the Brubecks and instructed them to cancel the show. Armstrong’s wife, Lucille, who rarely meddled in her husband’s business affairs, joined the chorus demanding the show be cancelled, fearing her husband wasn't up to the demanding performance due to his health.

Phoebe Lyon illustrates the many different aspects of maternity and childcare leave as an educator in Japan. Her autoethnography acts as a vehicle for the author to reflect on her experience with the aim of helping others. The purpose of this chapter is to share questions that were important to the author in order to help others in a similar situation decide how they can best negotiate pre- and post-childbirth leave in Japan.

Les épreuves de chinois de traduction, de commentaire et de dissertation littéraires, dans les concours nationaux en France, comme le CAPES ou l’Agrégation, ou encore le concours d’entrée à l’ENS, sont aussi exigeantes en maîtrise de la langue chinoise qu’en ce qui concerne les connaissances littéraires et culturelles du monde chinois. Ces épreuves qui visent en premier à sélectionner les meilleurs éléments, peuvent aussi donner lieu à un questionnement : dans quelle mesure la compréhension d’un texte littéraire en langue étrangère est-elle possible, a fortiori par un candidat en situation de concours ? Comment celui-ci devrait-il mobiliser son savoir littéraire, historique et culturel, outre ses compétences linguistiques, pour fournir une interprétation plausible du texte ? Pour répondre à ces questions, nous nous basons sur des textes littéraires fournis comme sujets aux différents concours : J’aime Bill (1988) de Wang Anyi, à l’épreuve de l’ENS Lyon en 2018 ; Ailes de plomb (1980) de Zhang Jie à l’épreuve de l’ENS Lyon en 2019. En partant de ces sujets d’épreuve, nous procédons aux analyses des écarts entre l’attente du jury et les prestations fournies. Au-delà des conseils sur les écueils et les obstacles rencontrés par les candidats dans un tel exercice, nous proposons de mener une réflexion plus générale, sur les lacunes et les possibilités de l’enseignement de la littérature étrangère, en particulier pour une langue comme le chinois, réputée difficile pour les francophones.

Lead is useful, surprising, unpredictable, dangerous – and deadly. Lead is useful. Previous generations found it to be an essential part of civilized living: pipes, pewter, pottery, paints, and even potions were made with it. Toy soldiers were cast from it, port wine protected by it; grey hair was disguised with it, church roofs covered with it; cosmetics contained it, and cans of food were sealed with it. Lead is surprising. In 1859, Professor Lyon Playfair was taking the 18-year-old Prince of Wales, and future King Edward VII, round the chemistry laboratories of Edinburgh University when they came across a pot of molten lead. Playfair then carried out a remarkable demonstration: he poured the molten metal over the fingers of his assistant and to the Prince’s amazement the young man’s hand was unharmed. The Prince too wanted to try the same test, and after rinsing his hand in dilute ammonia solution, lead was poured over his fingers as well – again without scalding them. This demonstration was still being performed before stunned audiences in the 1950s, and the trick is to have the hand wet, so that as the metal hits the film of water on the skin it forms an instant layer of vapour that both protects the skin and causes the lead to bounce off in tiny droplets. Lead is unpredictable. In Shakespeare’s Merchant of Venice those who would seek to marry the fair Portia are offered three caskets from which to choose: gold, silver, or lead. The man who chooses the casket containing her portrait will win her hand in marriage. Of course the winning choice is lead, the casket with the curiously foreboding inscription: ‘Who chooseth me must give and hazard all he hath’ and her would-be suitor Bassanio reasons that this must be the right one saying: …‘. . . but thou, thou meagre lead, Which rather threat’nest than does promise aught, Thy plainness moves me more than eloquence, And here choose I; joy be the consequence!’ [The Merchant of Venice, III, ii]…

The design of modern aircraft engines increasingly involves highly sophisticated methodologies to match the current development pace. International company relations affect the collaboration between design offices all around the world. An important part of academic mission of modern engineering education is to produce graduates with skills compatible with industrial needs. Education may readjust accordingly to meet the higher requirements. However, a realistic scenario of the design process of an aircraft engine cannot possibly be transferred one-to-one into the student education process. A unique attempt to overcome this discrepancy was the International Gas Turbine Project. Within this project, undergraduate students have designed the cooling system of the HPT blades for a 30,000 lb thrust two-spool turbofan aeroengine. This project was collaboration between the Jet Propulsion Laboratory of TU Berlin, the Turbomachinery Group of EC Lyon and the Turbomachinery Laboratory of ETH Zurich. It also involved mentoring industry professionals from Rolls-Royce Deutschland, MTU, SNECMA and Alstom Power. Similar to modern aeroengine company structures, the design tasks included multi-component, multi-disciplinary and international interfaces of different educational systems. The student teams considered various aerothermodynamic and mechanical integrity aspects of the design. Particular attention was paid to design of the compressor, the secondary air system and the HP turbine including blade cooling. The three Universities integrated the project differently into their education curriculum and approached the tasks with different levels of software involvement. In this paper, the technical details of the design process, and the different approaches adopted are presented. Besides the application of turbomachinery-related knowledge, the impact of student interactions on the technical aspects of the project is discussed. The interfaces, including information management and the involvement of industrial partners are also addressed. Team spirit developed between the students from an initial competitive behavior to a final feeling of sitting in the same boat. It was observed that increased effort was required from academic staff in comparison to the conventional academic instruction. Nevertheless, students greatly benefited from the social interaction and an early training-on-the-job tuned to current industrial needs.
