A prototype system for the cleaning and decontamination of poultry transport containers was previously developed and evaluated as a means of eliminating foodborne pathogens entering poultry processing plants. While decontamination of the containers once with the use of either hot water (up to 70°C) or sodium hypochlorite (up to 1,000 ppm) resulted in significant reductions in the numbers of coliforms and the elimination of small numbers of Salmonella, complete removal of pathogens was not attained. Therefore, the present study was conducted to determine whether repeated decontamination of the same containers could eliminate coliforms and Salmonella consistently. Individual five-tier containers consisting of galvanized steel frames and fiberglass floors were identified (n = 6) and decontaminated once per day for five consecutive days after being used to haul broilers from farms to the processing plant. Two types of containers were tested in this study: one had previously been used for broiler transportation, and the other had new floors. After each transport, the containers were first precleaned with a cleaning agent using a high-pressure jet (6,094 kPa) to remove debris and to loosen biofilms from surfaces. The containers were then immersed in an aqueous solution of 1,000 ppm of sodium hypochlorite at 70°C for 2 min. Samples obtained from the container surfaces before and after each cleaning and decontamination were analyzed to obtain coliform and Salmonella counts. Coliforms were completely eliminated from both types of containers following one decontamination treatment. Because no Salmonella were detected on the containers, the effect of decontamination in the elimination of Salmonella was not determined. Similar treatments on five successive days also resulted in poultry transport containers that were essentially free of Salmonella and coliforms. This decontamination system involving a combination of heat and sodium hypochlorite can be used as a standard method for cleaning poultry transport containers in the poultry industry. It is recommended that such containers be cleaned after each use to avoid the potential risk of a buildup of significantly higher loads of pathogenic microorganisms and their biofilms.

Abstract. The Agulhas Current Time-series Experiment mooring array (ACT) measured transport of the Agulhas Current at 34∘ S for a period of 3 years. Using along-track satellite altimetry data directly above the array, a proxy of Agulhas Current transport was developed based on the relationship between cross-current sea surface height (SSH) gradients and the measured transports. In this study, the robustness of the proxy is tested within a numerical modelling framework using a 34-year-long regional hindcast simulation from the Hybrid Coordinate Ocean Model (HYCOM). The model specifically tested the sensitivity of the transport proxy to (1) changes in the vertical structure of the current and to (2) different sampling periods used to calculate the proxy. Two reference proxies were created using HYCOM data from 2010 to 2013 by extracting model data at the mooring positions and along the satellite altimeter track for the box (net) transport and the jet (southwestward) transport. Sensitivity tests were performed where the proxy was recalculated from HYCOM for (1) a period where the modelled vertical stratification was different compared to the reference proxy and (2) different lengths of time periods: 1, 3, 6, 12, 18, and 34 years. Compared to the simulated (native) transports, it was found that the HYCOM proxy was more capable of estimating the box transport of the Agulhas Current compared to the jet transport. This was because the model is unable to resolve the dynamics associated with meander events, for which the jet transport algorithm was developed. The HYCOM configuration in this study contained exaggerated levels of offshore variability in the form of frequently impinging baroclinic anticyclonic eddies. These eddies consequently broke down the linear relationship between SSH slope and vertically integrated transport. Lastly, results showed that calculating the proxy over shorter or longer time periods in the model did not significantly impact the skill of the Agulhas transport proxy. Modelling studies of this kind provide useful information towards advancing our understanding of the sensitivities and limitations of transport proxies that are needed to improve long-term ocean monitoring approaches.

Most of the hydrogen storage vessels meant for vehicles to run the electric motor via a fuel cell during transport are designed for drives of only a few tenths of kilometers per day. The present report, however, describes a vessel model that is conceived to hold the hydrogen energy only for short periods during transport, such as a few hours. This would include transport via a light-duty vehicle, a taxi, or a bus, which would load liquid hydrogen at a station every morning for the day. This is a simple model based on the novel concept of Double Open Vessel (DOV), in which the liquid H2 is loaded inside an open container inserted inside another open container. The walls of this DOV are constituted of simplified linings that allow the entry of thermal heat nearly a hundred times greater than that allowed by the cryo-compressed vessels with higher insulation. After loading, the liquid evaporates, while the gas flows around its initial container into which it was loaded, passes through a few porous plugs, and is gradually released towards the Fuel Cell (or toward an ignition motor). Such a counter-flow of the gas creates a retroaction effect that insulates the inner container, thereby delaying the increases in temperature and pressure. The successive porous plugs installed in the space between the two containers form a system of barrages that regulate the gas flow through successive expansions of decreasing pressures. In addition, these catalytic plugs convert a portion of the loaded hydrogen into its ortho variety, acting as a heat pump, while temporarily storing the other portion. Collectively, these effects maintain the internal pressures below 150 bar. The proposed design for the DOV models is convenient to manufacture and has a lighter weight, and consequently, a low cost.

To store CO2 in geological reservoirs, expansion valves have been used to intentionally release supercritical CO2 from high-pressure containers at a source point to lower-pressure pipelines and transport to a selected injection site. Using expansion valves, however, has some shortcomings: (i) the fluid potential, in the form of kinetic energy and pressure which can produce mechanical work or electricity, is wasted, and (ii) due to the Joule-Thomson cooling effect, the reduction in the temperature of the released CO2 stream might be so dramatic that it can induce thermal contraction of the injection well causing fracture instability in the storage formation. To avoid these problems, it has been suggested that before injection, CO2, should be heated to a temperature slightly higher than that of the reservoir. However, heating could increase the cost of CO2 injection. This work explores the use of a Tesla Turbine, instead of an expansion valve, to harvest the potential of CO2, in the form of its pressure and kinetics, to generate mechanical work when it is released from a high-pressure container to a lower-pressure transport pipeline. The goal is to avoid throttling losses and to produce useful power because of the expansion process. In addition, due to the friction between the gas and the turbine disks, the expanded gas temperature reduction is not as dramatic as in the case when an expansion valve is used. Thus, as far as CO2 injection is concerned, the need for preheating can be minimized.

To elucidate the mechanisms involved in Mg2+ transport at the apical and basolateral poles of the renal tubular epithelium, apical and basolateral plasma membrane vesicle preparations were derived from kidney tissue of freshwater- and seawater-adapted Mozambique tilapia Oreochromis mossambicus. Brush-border preparations were enriched 15.8-fold in alkaline phosphatase activity and consisted almost exclusively of right-side-out membrane vesicles. Basolateral membrane preparations were enriched 7.5-fold in Na+/K+-ATPase activity and contained resealed vesicles and leaky membrane fragments. Mg2+ association with brush-border and basolateral plasma membranes, traced using radioactive 27Mg, occurred in an osmotically active space. In all instances, Mg2+ binding to the vesicular membrane was low compared with the vesicular uptake. Mg2+ equilibration across the vesicular membrane of brush-border preparations was rapid and sensitive to the presence of extravesicular Ca2+, suggesting that the apical membrane of the renal epithelium contains a transport pathway for divalent cations. Application of various ionic gradients did not affect vesicular Mg2+ transport in apical and basolateral membrane preparations, suggesting the presence of an ion-coupled transport mechanism. ATP or ATP--S did not stimulate Mg2+ fluxes, indicating that Mg2+ transport does not proceed via an ATP-driven or activated transporter. In these aspects, vesicular Mg2+ transport was similar in seawater and freshwater preparations. These results suggest that the apical membrane of renal epithelial cells lacks an active secretory Mg2+ transport mechanism. We propose that the Mg2+ conductivity of the apical membrane reflects a route for downhill Mg2+ entry and is involved in renal Mg2+ reabsorption.

1. The transport of K+, Na+ and Cl− across the three morphologically distinct regions of the tobacco hornworm midgut was measured under open-circuit and short-circuit conditions. Using a saline which contained physiological levels of haemolymph ions, amino acids and sugars, it was shown that all three sections actively secrete K+ and Cl− and absorb Na+. 2. The anterior section maintained the highest short-circuit current (Isc), transepithelial potential difference (PD) and net K+ secretion. The middle section had the lowest Isc, PD and K+ secretion, but absorbed Na+ at the greatest rate. The posterior section had the greatest rate of Cl− secretion. 3. Omission of K+ depressed the Isc. Subsequent addition of K+ stimulated the Isc to control levels in the middle and posterior sections, but not in the anterior section. Omission of Cl− or Na+ also inhibited the Isc. Reintroduction of Cl− had no stimulatory effect and, although reintroduction of Na+ stimulated the Isc, control levels were not attained. 4. Unlike the results reported in previous studies, the net K+ transport exceeded the Isc in all three midgut sections. The deficit in Isc was not made up by the transport of Na+ and Cl−. The results are discussed with respect to proposed models of ion transport across this epithelium.

We have selected Ia variants from the Ia+ (H-2d) M12.4.1 B cell lymphoma that are negative on the cell surface for one or both Ia isotypes. The molecular analysis of two such independently selected cell lines, M12.A2 and M12.C3, is reported here. This analysis revealed that the genes encoding Ad beta (M12.A2) and Ed beta (M12.C3) contained identical single-nucleotide transitions that resulted in the substitution of Ser (mutant) for Asn (wild-type) at residue 82/83 of the extracellular NH2-terminal (membrane distal) beta 1 domain. This conservative substitution caused a cytoplasmic accumulation of I-A or I-E molecules in the respective cell line although predicted secondary-structure analysis suggests a minimal effect on protein conformation. Thus, the mutation appears to have either created a negative signal that stops transport or eliminated a positive signal that is required for transport and targeting to the cell surface.

The paper covers the mechanism of lubrication layer formation by fuels containing synthetic hydrocarbons and alcohols. Development of alternative fuels containing FAME, alcohols, and synthetic hydrocarbons has increased the interest in the mechanism of lubrication of fuelling systems parts. Fuel lubricity tests have been conducted using the HFRR and BOCLE testing rigs. Fuels under testing, both for CI engines and for aviation turbine ones, contained synthetic components: saturated hydrocarbons both of even and odd number of carbon atoms, and butanol, isomers. These components have been added to conventional fuels, such as diesel fuel and Jet A-1 fuel at the concentration of 0–20% (V/V). All fuels under testing contained commercially available lubricity improvers (carboxylic acid). Test results were analysed using model αi described in [L. 6, 7]. As a result of the analysis, it has been found that the liquid phase, which is a lubricating film, should contain agglomerates or molecular clusters responsible for the transport of energy introduced into lubricating film by electrons emitted from metal surface. The mechanism enabling a description of the effect of base fuel without lubricity improvers on efficiency of such additives has been suggested.

Abstract Low-level winds along the Californian coast during spring and early summer are typically strong and contained within the cool, well-mixed marine boundary layer (MBL). A temperature inversion separates the MBL from the warmer free troposphere. This setup is often represented by a two-layer shallow-water system with a lateral boundary. Near a prominent point such as Point Conception, California, the fast-moving MBL flow is supercritical and can exhibit distinct features including a compression bulge and an expansion fan. Measurements from the University of Wyoming King Air research aircraft on 19 May 2012 during the Precision Atmospheric MBL Experiment (PreAMBLE) captured wind in excess of 14 m s−1 off of Point Conception under clear skies and wind ~2 m s−1 east of San Miguel in the California Bight. A compression bulge was identified upwind of Point Conception. When the flow rounds the point, the MBL undergoes a near collapse and there is a spike in MBL height embedded in the general decrease of MBL height with greater turbulence just downwind that is associated with greater mixing through the inversion layer. Lidar and in situ measurements reveal that transport of continental aerosol is present near the pronounced MBL height change and that there is a complex vertical structure within the Santa Barbara Channel. Horizontal pressure gradients are obtained by measuring the slope of an isobaric surface. Observations of wind and pressure perturbations are able to be linked through a simple Bernoulli relationship. Variation of MBL depth explains most, but not all of the variation of the isobaric surface.

The uptake and metabolism of glucose, alanine and lactate were assessed in red blood cells (RBCs) of the American eel Anguilla rostrata. l-Lactate was metabolized at the highest rates as assessed by O2 consumption and CO2 production, followed by glucose and alanine (rates were approximately half of those observed for lactate). A saturable (Km 10.36±0.60 mmol l-1, Jmax 27.42±2.16 µmol 3-OMG l-1 cell water min-1), sodium-independent but cytochalasin-B-sensitive carrier for d-glucose was observed, which was stereospecific and inhibited by other hexoses. These characteristics are in agreement with those reported for the GLUT-1 glucose carrier of human and Japanese eel erythrocytes. These cells also contained a saturable carrier for l-lactate in the concentration range 0­10 mmol l-1 (Km 6.74±0.36 mmol l-1, Jmax 2.29±0.09 mmol lactate l-1 cell water min-1) whereas, at higher concentrations (10­40 mmol l-1), transport occurred by simple diffusion. The carrier was stereospecific, sodium-independent, fully inhibited by alpha-cyano-4-hydroxycinnamate, DIDS and pyruvate, but less sensitive to SITS, IBCLA and pCMBS. We suggest that this carrier is similar to the H+/monocarboxylate carrier found in mammalian RBCs. Despite the fact that l-alanine transport did not saturate, transport was stereospecific because it was inhibited by d-alanine. These experiments do not, therefore, exclude the existence of an alanine carrier in the eel RBC. The rates of substrate uptake exceeded the ability of the RBC to metabolize the substrate (using 1 mmol l-1 extracellular concentration), with uptake rate/metabolic rate ratios being 2 for alanine, 5 for glucose and 151 for lactate. These experiments indicate that uptake does not limit the ability of the American eel RBC to utilize glucose, alanine or lactate, but that the mechanism(s) of substrate uptake is species-specific.

By the end of oocyte development, the ovaries of Penaeus semisulcatus have accumulated almost equal amounts (approximately 16 mg lipid g-1 protein) of phospholipids and triacylglycerols. The phospholipids consist mainly of phosphatidylcholine (75–80 %) and phosphatidylethanolamine (20–25 %). Approximately 30 % of the total fatty acid content of both phospholipids and triacylglycerols is made up of polyunsaturated fatty acids. In fractions obtained by centrifugation of ovarian homogenates, most of the increase in levels of ovarian lipids during ovarian maturation was associated with an increase in triacylglycerol levels in the floating fat fraction and of phospholipids in the infranatant fraction. The presence of polyunsaturated fatty acids in the ovaries indicates the occurrence of lipid transport to the ovary during oocyte maturation. The gradual decrease in the relative abundance of polyunsaturated fatty acids as the ovaries matured supports previously published results suggesting intra-ovarian synthesis of saturated and mono-unsaturated fatty acids. Most of the lipids found in the female haemolymph (64.8 %) were recovered in the high-density lipoprotein fraction after density ultracentrifugation. The haemocyanin fraction recovered from this stage of fractionation contained substantial amounts of lipid (16.8 %) that could be removed by further sequential centrifugation at a higher NaBr density, leaving less than 0.9 % of the total haemolymph lipids associated with this fraction. While 16.2 % of the lipids were recovered from the very high-density lipoprotein fractions, these lipoproteins carried only 64–89 microg lipid mg-1 protein compared with 538.9 microg lipid mg-1 protein in the high-density lipoprotein fraction, indicating that the high-density lipoproteins are more likely to be the main transporters of lipids to the ovary. However, the contribution of very high-density lipoproteins to lipid transport cannot be ruled out at this stage. In this study, we present two models for lipid transport to the ovary based on the abundance of phospholipids and triacylglycerols in the haemolymph and on the amounts of polyunsaturated fatty acids accumulated within the ovary during vitellogenesis.

SUMMARYThis study examines calcification in planulae and polyps of the hydroid Hydractinia symbiolongicarpus. We observed that established colonies produce a crystalline mat on their substratum and that crystals visible by polarized light microscopy occur in the vacuoles of the gastrodermal cells of both polyps and planulae. The crystalline mat was found by infrared spectroscopy to contain calcium carbonate in the form of aragonite. The composition of the vacuolar crystals and the cellular mechanisms for manufacturing them were explored by alteration of calcium levels in the environment and by the use of pharmacological agents (acetazolamide, caffeine, DIDS, diltiazem, nifedipine, procaine, Ruthenium Red, ryanodine and verapamil) that affect cellular uptake and transport of calcium and bicarbonate. The results indicated that the crystals in the vacuoles contained calcium carbonate. The gastrodermal cells are hypothesized to serve as a physiological sink for excess calcium that enters the organism during motility, secretion and metamorphosis of the planula, and to create a crystalline substratum for the colony of polyps.

Crude homogenates and plasma-membrane-enriched fractions were prepared from the epithelium of the gills, epipodites and branchiostegites of intermoult European lobsters Homarus gammarus, and Na(+)/K(+)-ATPase, Ca(2+)-ATPase and Na(+)/Ca(2+) exchange activities were quantified in these tissues. Lobsters were kept in sea water (salinity 35) or were adapted to dilute sea water (22.1). The lobster hyperregulates haemolymph osmolarity and Ca(2+) levels in both media. Homogenates of the podobranchs, arthrobranchs and pleurobranchs had comparable Na(+)/K(+)-ATPase specific activities, and mean activities increased significantly for all three types of gills when the animals were kept in dilute sea water. In the epipodites and branchiostegites, Na(+)/K(+)-ATPase specific activities exceeded those in the gills, and exposure to dilute sea water greatly enhanced these activities. In sea water, 80 % of the total Na(+)/K(+)-ATPase activity is associated with the gills and epipodites (each tissue containing 40 %) and 20 % with the branchiostegites; in dilute sea water, the gills contained approximately 25 %, the epipodites 40 % and the branchiostegites approximately 35 % of the total activity, indicating the relative importance of the epipodites and branchiostegites for ionic hyperregulation in dilute media. In plasma membrane vesicles isolated from the gills, epipodites and branchiostegites, Ca(2+) transport driven by ATP and by a Na(+)gradient was demonstrated. Exposure to dilute sea water enhanced Na(+)/Ca(2+)exchange and Ca(2+)-ATPase activities in the epipodites and branchiostegites; in the gills, however, Ca(2+) transport activities decreased. The role of these tissues and enzymes in Na(+) and Ca(2+) handling by the lobster is discussed.

SUMMARY Female German cockroaches usually carry their oothecae until they hatch. The success of embryogenesis may be dependent on the water-balance relationship between females and their developing oothecae. Oothecae detached from females early in embryogenesis often fail to develop, especially in low-humidity environments. Experiments reported here using tritiated water have confirmed the transport of a significant amount of water to the ootheca from the female during embryogenesis; 18% of the tritiated water injected into gravid females was recovered in their oothecae after 24 h. We describe a structural basis of water absorption by the oothecae. An area located on the proximal end of the ootheca (adjacent to the `escutcheon-shaped vaginal imprint') contains small pores that penetrate the oothecal covering to access regions of the chorion lying beneath these pores. Experiments using microparabiotic chambers to examine transport of tritiated water and water-soluble materials across the escutcheon region of the oothecal covering, along with dark-field birefringent microscopy of the chorion, support the hypothesis that this chorionic network is capable of conducting water throughout the interior of the ootheca. Furthermore, the structural arrangement and intimate association of the female vestibulum with the oothecal pore field contained in the region of the escutcheon-shaped vaginal imprint appear to provide an efficient conduit to the chorion. The overall structural relationship might be a means for maintaining water balance between females and their oothecae during embryonic development. Evidence presented here supports observations that this species represents an important link in the transition from oviparity to ovoviviparity by internalization of cockroach oothecae.

Potassium transport characteristics were investigated in three species of freshwater bivalves: a corbiculid, Corbiculafluminea, and two unionids, Carunculina texasensis and Ligumia subrostrata. Using 42K, all three were found to take up potassium from dilute artificial pondwater ([K+] about 0.05 mmoll+1). The influx (Ji) was 0.72μequiv g−1dry tissue h−1 in the corbiculid, significantly higher than the value of about 0.40μequiv g−1dry tissuer g−1 in the unionids. The K+ uptake displayed saturation kinetics in the range 0.05-0.36 mmoll−1: in Co. fluminea, there was a Jmax of 3.56μequiv g−1 dry tissue h−1 and the affinity coefficient (Km) was 0.27mmoll−1; in Ca. texasensis, Jmax had a value of 1.8 μequiv g−1dry tissue h−1 and Km was 0.16mmoll−1. Using K+-free artificial pondwater containing 0.03-0.04 mmoll−1 Rb+, the Rb+ influx was 0.41μequiv g−1 dry tissue h−1 in the corbiculid and 0.28 μequiv g−1 dry tissue h−1 in Ca. texasensis. All animals lost K+ during the rubidium flux studies, and since they contained no Rb+, the Rb+ efflux was zero and the net flux was equal to the influx. The Jmax values for Rb+ were lower than the corresponding values for potassium: in Co. fluminea, Jmax was 1.4μequiv g−1 dry tissueh+1, significantly higher than in Ca. texasensis, which had Jmax of 0.84μequiv g−1 dry tissue h−1. The rubidium Km (approx. 0.05 mmoll−1) values were significantly lower than corresponding values for potassium. Salt depletion increased the rubidium transport rate fourfold for both Co. fluminea and Ca. texasensis. High rates of net K+ uptake may account for the bivalves' inability to tolerate elevated environmental potassium.

A new double-seeded insert (DSI) technique is described for culture of branchial epithelial preparations from freshwater rainbow trout on filter supports. DSI epithelia contain both pavement cells and mitochondria-rich (MR) cells (15.7+/−2.5 % of total cell numbers). MR cells occur singly or in clusters, are voluminous, open apically to the ‘external environment’ and exhibit ultrastructural characteristics similar to those found in the ‘chloride cells’ of freshwater fish gills. After 6–9 days in culture with Leibovitz's L-15 medium on both surfaces (symmetrical conditions), transepithelial resistance (TER) stabilized at values as high as 34 k capomega cm(2), indicative of electrically ‘tight’ epithelia. The density of MR cells, the surface area of their clusters and transepithelial potential (TEP; up to +8 mV basolateral positive, mean +1.9+/−0.2 mV) were all positively correlated with TER. In contrast, preparations cultured using an earlier single-seeded insert (SSI) technique contained only pavement cells and exhibited a negligible TEP under symmetrical conditions. Na(+)/K(+)-ATPase activities of DSI preparations were comparable with those in gill filaments, but did not differ from those of SSI epithelia. Replacement of the apical medium with fresh water to mimic the in vivo situation (asymmetrical conditions) induced a negative TEP (−6 to −15 mV) and increased permeability to the paracellular marker PEG-4000. Under symmetrical conditions, unidirectional Na(+) and Cl(−) fluxes were in balance, and there was no active transport by the Ussing flux ratio criterion. Under asymmetrical conditions, there were large effluxes, small influxes and evidence for active Cl(−) uptake and Na(+) extrusion. Unidirectional Ca(2+) fluxes were only 0.5-1.0 % of Na(+) and Cl(−) fluxes; active net Ca(2+) uptake occurred under symmetrical conditions and active net extrusion under asymmetrical conditions. Thus, DSI epithelia exhibit some of the features of the intact gill, but improvements in culture conditions are needed before the MR cells will function as true freshwater ‘chloride cells’.

Recognition of self–peptide-MHC (pMHC) complexes by CD4 T cells plays an important role in the pathogenesis of many autoimmune diseases. We analyzed formation of immunological synapses (IS) in self-reactive T cell clones from patients with multiple sclerosis and type 1 diabetes. All self-reactive T cells contained a large number of phosphorylated T cell receptor (TCR) microclusters, indicative of active TCR signaling. However, they showed little or no visible pMHC accumulation or transport of TCR–pMHC complexes into a central supramolecular activation cluster (cSMAC). In contrast, influenza-specific T cells accumulated large quantities of pMHC complexes in microclusters and a cSMAC, even when presented with 100-fold lower pMHC densities. The self-reactive T cells also maintained a high degree of motility, again in sharp contrast to virus-specific T cells. 2D affinity measurements of three of these self-reactive T cell clones demonstrated a normal off-rate but a slow on-rate of TCR binding to pMHC. These unusual IS features may facilitate escape from negative selection by self-reactive T cells encountering very small amounts of self-antigen in the thymus. However, these same features may enable acquisition of effector functions by self-reactive T cells encountering large amounts of self-antigen in the target organ of the autoimmune disease.

The contribution of the secondary circulatory system to acid-base regulation and epithelial ion transport was evaluated qualitatively in freshwater-acclimated rainbow trout. The dorsal aorta (DA) and the lateral cutaneous vessel (LCV) (which is considered to be the venous drainage of the secondary system) were chronically cannulated and the fish were exposed to environmental hypercapnia (2 % CO2) after establishment of normocapnic control values. Fluid sampled from the LCV contained much less haemoglobin (0.14 g 100 ml−1) and fewer blood cells (packed cell volume, PCV, 1.2-1.7%) than DA blood ([Hb] 8.2-8.9 g 100 ml−1, PCV 27.2-32.5%) regardless of ambient CO2 levels, indicating highly limited access of red blood cells to the secondary circulatory system through anastomoses connecting it to the primary system. There was no significant difference between the two sampling sites for any of the acid-base variables (pH, PCO2 [HCO3−]) and most plasma ion concentrations ([Na+], [Ca2+], [Mg2+] and [Cl−]) during normocapnia, although plasma [K+] and osmolarity were significantly lower in LCV fluid. Upon exposure to hypercapnia, the pH of LCV fluid became significantly higher than that of DA plasma because of the higher [HCO3−] in the LCV plasma than in the DA plasma. The LCV plasma concentration of Cl− was significantly lower than that of DA plasma during hypercapnia. These data suggest that the secondary circulatory system draining the body surface is involved in ionic acid-base regulation in fish, probably via Cl−/HCO3− exchange processes across the extrabranchial epithelium of the body surface.

External application of the 3',5'-cyclic monophosphates of inosine,cytidine, uridine and thymidine stimulated the fluid secretion rate (FSR)of Malpighian tubules isolated from Drosophila melanogaster. The evidence suggested that the cyclic nucleotides acted intracellularly in some capacity. Receptors of the 'purinergic' type appeared not to be major contributors to fluid secretion; of three purinergic agonists tried,adenosine, adenosine 5'-monophosphate (AMP) and adenosine 5'-triphosphate(ATP), only adenosine had an effect, but this was not observed consistently. None of the purinergic agonists interfered with the stimulation of the FSR by adenosine 3',5'-cyclic monophosphate (cAMP). The maximum stimulation of the fluid-secretion rate by any cyclic nucleotide was approximately double the unstimulated (control) rate. Tubules stimulated to less than maximal FSR by one cyclic nucleotide could be stimulated maximally by an appropriate concentration of another cyclic nucleotide. Malpighian tubules bathed in solutions that contained either[3H]cAMP or [3H]cGMP accumulated radioactivity to a level many times that in the medium. Accumulation of radioactivity by tubules bathed in 430 nmol l-1 [3H]cAMP was suppressed by 1 mmol l-1 non-radioactive cyclic nucleotides in the order cAMP>>cGMP>cIMP>cCMP; neither cTMP nor cUMP suppressed the accumulation of [3H]cAMP. Approximately 35 % of the[3H]cAMP and 80 % of the [3H]cGMP that entered the Malpighian tubule cells was metabolised to compounds that were not identified. It was concluded that cyclic nucleotides enter the Malpighian tubule cells by at least one transport mechanism which is particularly sensitive to purine-based nucleotides.

J774 macrophage-like cells express organic anion transporters that promote the efflux of fluoroquinolone antibiotics such as norfloxacin (NFX) from these cells. Gemfibrozil (GFZ) blocks organic anion transport in J774 cells, thereby facilitating the intracellular accumulation of NFX (Cao, C., H.C. Neu, and S.C. Silverstein. 1991. J. Cell Biol. 115:467a [Abstr.]). To determine whether GFZ enhances the efficacy of fluoroquinolone antibiotics against intracellular bacterial pathogens, J774 cells were infected with Listeria monocytogenes and incubated in medium containing a fluoroquinolone antibiotic in the presence or absence of GFZ. Intracellular growth of L. monocytogenes was evaluated by lysing J774 cells and assaying for colony-forming units of Listeria. GFZ intensified the bacteriostatic effect of 4 micrograms/ml NFX and rendered 8 micrograms/ml bactericidal for L. monocytogenes. GFZ had a similar potentiating effect when used in combination with 2 micrograms/ml ciprofloxacin (CFX). CFX plus GFZ was bactericidal for intracellular L. monocytogenes. Treatment of J774 cells with NFX plus GFZ markedly reduced the cytotoxic effect of the bacteria on these cells. Over 55% of cells treated with 8 micrograms/ml NFX alone were dead 16 h after infection, whereas only 5% of cells treated with 8 micrograms/ml NFX plus GFZ were dead at 16 h. Similarly, GFZ potentiated the ability of 2 micrograms/ml to protect J774 cells against the cytocidal effect of Listeria. NFX in combination with GFZ limited cell-to-cell spread of L. monocytogenes. In antibiotic-free medium, > 99% of J774 cells contained intracellular L. monocytogenes at 14 h after infection. NFX alone in the medium did not change this outcome. However, 4 micrograms/ml NFX plus GFZ decreased bacterial spread by approximately 40% at 24 h postinfection, and 8 micrograms/ml NFX plus GFZ prevented all spread beyond the initially infected cell population. These results suggest that GFZ could be used clinically to enhance the efficacy of fluoroquinolone and of other anionic antibiotics against bacteria that grow and/or reside within macrophages and/or other cells.

Batch process control is typically used for repeated chemical reaction tasks. It starts with a measured liquid material filling operations followed by a controlled reaction leading to the discharge or transport of processed quantities of material. The input materials is contained in vessel reactor and subjected to a sequence of processing activities over a recipe predefined duration of time. Batch systems are designed to measure, process, and discharge a varying volume of liquid from drums, tanks, reactors, or other large storage vessel using a programmable logic controller (PLC). These systems are common in pharmaceutical, chemical packaging, Beverage processing, personal care product, biotech manufacturing, dairy processing, soap manufacturing, and food processing industries. This paper briefly discusses the fundamental techniques used in specifying, designing, and implementing a PLC batch process control [1, 2]. A simplified batch process is used to illustrate key issues in designing and implementing such systems. In addition to the structured PLC ladder design; more focus is given to safety requirements, redundancy, interlocking, input data validation, and safe operation. The Allen Bradley (AB) SLC 500 PLC along with the LogixPro simulator are used to illustrate the concepts discussed in this paper. Two pumps are used to bring in material during the tank filling and a third pump is used to drain processed product. The three pumps are equipped with flow meters providing pulses proportional to the actual flow rate through the individual pipes. The tank material is heated to a predefined temperature duration followed by mixing for a set time before discharge. Batch control systems provides automated process controls, typically and universally using PLC’s networked to HMI’s and other data storage, analysis, and assessment computers. The overall system perform several tasks including recipe development and download, production scheduling, batch management and execution, equipment performance monitoring, inventory, production history and tracking functionalities. Flexible batch control systems are designed to accommodate smaller batches of products with greater requirements / recipes variation, efficiently and quickly. In addition to providing process consistency, continuous batch process control quality improvements are attained through the automatic collection and analysis of real-time reliable and accurate event performance data [3, 4].

Purpose In many coastal areas where there are problematic soils, pavement construction on the soil is difficult because of the low shear strength and high consolidated. Also, given that the container terminals constitute more than 70% of the port area and as pavement in these areas is subject to heavy loads due to the long-term container storage, wheels of transport and movement equipment, the pavement must tolerate a distributed loading of at least 4 ton/m2 in accordance with the type and weight of the containers imposed on the pavement. This study aims to investigate a variety of common pavement designs in coastal areas of southern Iran. The pavement type and characteristics of the subgrade layers are the same for each port; the thickness of different pavement layers is designed. Design/methodology/approach Due to problematic soil in the pavement subgrade, heavy and long-term container loading and the associated equipment, port pavement enjoys great importance. Findings The designed pavements are modeled by ABAQUS finite element software. The pavements are subject to a static load imposed by the corner casting container and resulted a distributed load 4 tons/m2. The results from data analysis show that the concrete block pavements influenced by the containers static loads of 3%–20% have less vertical displacement on the subgrade than other pavements (rigid and flexible). Originality/value This paper is modeling 3 port pavement in Iran. Based on field evaluation and simulation actual loading on pavement.

Impinging jets have become an indispensable measure for cooling applications in gas turbine technology. The present study seeks to explore the flow field dynamics inside an enigine-relevant cooling passage of trapezoidal cross-section. The investigated geometry produces a highly complex flow field which was investigated employing particle image velocimetry (PIV). The experiments were accompanied by numerical simulations solving the Reynolds-averaged Navier–Stokes (RANS) equations with FLUENT using the low-Re k-ω-SST (shear stress transport) turbulence model. Additionally, time-resolved pressure measurements were performed utilizing Kulite pressure transducers. The spectral analysis of the transient pressure signal in conjunction with a proper orthogonal decomposition (POD) analysis of the PIV data allows for a detailed insight into the effects of geometric constraints on the fluid dynamic processes inside the geometry. The results are presented for a jet Reynolds number of 45,000 and display a qualitatively fair agreement between the experiments and numerical simulations. Nevertheless, the simulations predict flow features in particular regions of the geometry that are absent in the experiments. Despite the lack of conspicuous high energy modes, the flow was well suited for a POD analysis. Depending on the considered PIV plane, it could be shown that up to 25% of the flow field's total turbulent energy is contained in the first ten POD modes. Additionally, using the first 20 to 60 POD modes sufficed to reconstruct the flow fields with its governing features.

IntroductionIf we follow the line of much literature surrounding airports and urban mobility, the emphasis often falls on the fact that these spaces are designed to handle the mega-scale and super-human pace of mass transit. Airports have rightly been associated with velocity, as zones of rapid movement managed by enormous processing systems that guide bodies and things in transit (Pascoe; Pearman; Koolhaas; Gordon; Fuller & Harley). Yet this emphasis tends to ignore the spectrum of tempos and flows that are at play in airport terminals — from stillness to the much exalted hyper-rapidity of mobilized publics in the go-go world of commercial aviation.In this photo essay I'd like to pull a different thread and ask whether it's possible to think of aeromobility in terms of “uneven, differential mobility” (Bissell 280). What would it mean to consider waiting and stillness as forms of bodily engagement operating over a number of different scales and temporalities of movement and anticipation, without privileging speed over stillness? Instead of thinking mobility and stillness as diametrically opposed, can we instead conceive of them as occupying a number of different spatio-temporal registers in a dynamic range of mobility? The following is a provisional "visual ethnography" constructed from photographs of air terminal light boxes I have taken over the last five years (in Amsterdam, London, Chicago, Frankfurt, and Miami). Arranged into a "taxonomy of differentiality", each of these images comes from a slightly different angle, mode or directionality. Each view of these still images displayed in billboard-scale light-emitting devices suggests that there are multiple dimensions of visuality and bodily experience at play in these image-objects. The airport is characterized by an abundance of what appears to be empty space. This may be due to the sheer scale of mass transport, but it also arises from a system of active and non-active zones located throughout contemporary terminals. This photo series emphasises the "emptiness" of these overlooked left-over spaces that result from demands of circulation and construction.1. We Move the WorldTo many travellers, airport gate lounges and their surrounding facilities are loaded with a variety of contradictory associations and affects. Their open warehouse banality and hard industrial sterility tune our bodies to the vast technical and commercial systems that are imbricated through almost every aspect of contemporary everyday life.Here at the departure gate the traveller's body comes to a moment's rest. They are granted a short respite from the anxious routines of check in, body scans, security, information processing, passport scanning, itineraries, boarding procedures and wayfaring the terminal. The landside processing system deposits them at this penultimate point before final propulsion into the invisible airways that pipe them into their destination. We hear the broadcasting of boarding times, check-in times, name's of people that break them away from stillness, forcing people to move, to re-arrange themselves, or to hurry up. Along the way the passenger encounters a variety of techno-spatial experiences that sit at odds with the overriding discourse of velocity, speed and efficiency that lie at the centre of our social understanding of air travel. The airline's phantasmagorical projections of itself as guarantor and enabler of mass mobilities coincides uncomfortably with the passenger's own wish-fulfilment of escape and freedom.In this we can agree with the designer Bruce Mau when he suggests that these projection systems, comprised of "openings of every sort — in schedules, in urban space, on clothes, in events, on objects, in sightlines — are all inscribed with the logic of the market” (Mau 7). The advertising slogans and images everywhere communicate the dual concept that the aviation industry can deliver the world to us on time while simultaneously porting us to any part of the world still willing to accept Diners, VISA or American Express. At each point along the way these openings exhort us to stop, to wait in line, to sit still or to be patient. The weird geographies depicted by the light boxes appear like interpenetrating holes in space and time. These travel portals are strangely still, and only activated by the impending promise of movement.Be still and relax. Your destination is on its way. 2. Attentive AttentionAlongside the panoramic widescreen windows that frame the choreography of the tarmac and flight paths outside, appear luminous advertising light boxes. Snapped tightly to grid and locked into strategic sightlines and thoroughfares, these wall pieces are filled with a rotating menu of contemporary airport haiku and ersatz Swiss graphic design.Mechanically conditioned air pumped out of massive tubes creates the atmosphere for a very particular amalgam of daylight, tungsten, and fluorescent light waves. Low-oxygen-emitting indoor plants are no match for the diesel-powered plant rooms that maintain the constant flow of air to every nook and cranny of this massive processing machine. As Rem Koolhaas puts it, "air conditioning has launched the endless building. If architecture separates buildings, air conditioning unites them" (Koolhaas). In Koolhaas's lingo, these are complex "junkspaces" unifying, colliding and coalescing a number of different circulatory systems, temporalities and mobilities.Gillian Fuller reminds us there is a lot of stopping and going and stopping in the global circulatory system typified by air-terminal-space.From the packing of clothes in fixed containers to strapping your belt – tight and low – stillness and all its requisite activities, technologies and behaviours are fundamental to the ‘flow’ architectures that organize the motion of the globalizing multitudes of today (Fuller, "Store" 63). It is precisely this functional stillness organised around the protocols of store and forward that typifies digital systems, the packet switching of network cultures and the junkspace of airports alike.In these zones of transparency where everything is on view, the illuminated windows so proudly brought to us by J C Decaux flash forward to some idealized moment in the future. In this anticipatory moment, the passenger's every fantasy of in-flight service is attended to. The ultimate in attentiveness (think dimmed lights, soft pillows and comfy blankets), this still image is captured from an improbable future suspended behind the plywood and steel seating available in the moment —more reminiscent of park benches in public parks than the silver-service imagined for the discerning traveller.3. We Know ChicagoSelf-motion is itself a demonstration against the earth-binding weight of gravity. If we climb or fly, our defiance is greater (Appleyard 180).The commercial universe of phones, cameras, computer network software, financial instruments, and an array of fancy new gadgets floating in the middle of semi-forgotten transit spaces constitutes a singular interconnected commercial organism. The immense singularity of these claims to knowledge and power loom solemnly before us asserting their rights in the Esperanto of "exclusive rollover minutes", "nationwide long distance", "no roaming charges" and insider local knowledge. The connective tissue that joins one part of the terminal to a commercial centre in downtown Chicago is peeled away, revealing techno-veins and tendrils reaching to the sky. It's a graphic view that offers none of the spectacular openness and flights of fancy associated with the transit lounges located on the departure piers and satellites. Along these circulatory ribbons we experience the still photography and the designer's arrangement of type to attract the eye and lure the body. The blobby diagonals of the telco's logo blend seamlessly with the skyscraper's ribbons of steel, structural exoskeleton and wireless telecommunication cloud.In this plastinated anatomy, the various layers of commercially available techno-space stretch out before the traveller. Here we have no access to the two-way vistas made possible by the gigantic transparent tube structures of the contemporary air terminal. Waiting within the less travelled zones of the circulatory system we find ourselves suspended within the animating system itself. In these arteries and capillaries the flow is spread out and comes close to a halt in the figure of the graphic logo. We know Chicago is connected to us.In the digital logic of packet switching and network effects, there is no reason to privilege the go over the stop, the moving over the waiting. These light box portals do not mirror our bodies, almost at a complete standstill now. Instead they echo the commercial product world that they seek to transfuse us into. What emerges is a new kind of relational aesthetics that speaks to the complex corporeal, temporal, and architectural dimensions of stillness and movement in transit zones: like "a game, whose forms, patterns and functions develop and evolve according to periods and social contexts” (Bourriaud 11). 4. Machine in the CaféIs there a possible line of investigation suggested by the fact that sound waves become visible on the fuselage of jet planes just before they break the sound barrier? Does this suggest that the various human senses are translatable one into the other at various intensities (McLuhan 180)?Here, the technological imaginary contrasts itself with the techno alfresco dining area enclosed safely behind plate glass. Inside the cafes and bars, the best businesses in the world roll out their biggest guns to demonstrate the power, speed and scale of their network coverage (Remmele). The glass windows and light boxes "have the power to arrest a crowd around a commodity, corralling them in chic bars overlooking the runway as they wait for their call, but also guiding them where to go next" (Fuller, "Welcome" 164). The big bulbous plane sits plump in its hangar — no sound barriers broken here. It reassures us that our vehicle is somewhere there in the network, resting at its STOP before its GO. Peeking through the glass wall and sharing a meal with us, this interpenetrative transparency simultaneously joins and separates two planar dimensions — machinic perfection on one hand, organic growth and death on the other (Rowe and Slutsky; Fuller, "Welcome").Bruce Mau is typical in suggesting that the commanding problem of the twentieth century was speed, represented by the infamous image of a US Navy Hornet fighter breaking the sound barrier in a puff of smoke and cloud. It has worked its way into every aspect of the design experience, manufacturing, computation and transport.But speed masks more than it reveals. The most pressing problem facing designers and citizens alike is growth — from the unsustainable logic of infinite growth in GDP to the relentless application of Moore's Law to the digital networks and devices that define contemporary society in the first world. The shift of emphasis from speed to growth as a time-based event with breaking points and moments of rupture has generated new possibilities. "Growth is nonlinear and unpredictable ... Few of us are ready to admit that growth is constantly shadowed by its constitutive opposite, that is equal partners with death” (Mau 497).If speed in part represents a flight from death (Virilio), growth invokes its biological necessity. In his classic study of the persistence of the pastoral imagination in technological America, The Machine in the Garden, Leo Marx charted the urge to idealize rural environments at the advent of an urban industrialised America. The very idea of "the flight from the city" can be understood as a response to the onslaught of technological society and it's deathly shadow. Against the murderous capacity of technological society stood the pastoral ideal, "incorporated in a powerful metaphor of contradiction — a way of ordering meaning and value that clarifies our situation today" (Marx 4). 5. Windows at 35,000 FeetIf waiting and stillness are active forms of bodily engagement, we need to consider the different layers of motion and anticipation embedded in the apprehension of these luminous black-box windows. In The Virtual Window, Anne Friedberg notes that the Old Norse derivation of the word window “emphasizes the etymological root of the eye, open to the wind. The window aperture provides ventilation for the eye” (103).The virtual windows we are considering here evoke notions of view and shelter, open air and sealed protection, both separation from and connection to the outside. These windows to nowhere allow two distinct visual/spatial dimensions to interface, immediately making the visual field more complex and fragmented. Always simultaneously operating on at least two distinct fields, windows-within-windows provide a specialized mode of spatial and temporal navigation. As Gyorgy Kepes suggested in the 1940s, the transparency of windows "implies more than an optical characteristic; it implies a broader spatial order. Transparency means a simultaneous perception of different spatial locations" (Kepes 77).The first windows in the world were openings in walls, without glass and designed to allow air and light to fill the architectural structure. Shutters were fitted to control air flow, moderate light and to enclose the space completely. It was not until the emergence of glass technologies (especially in Holland, home of plate glass for the display of commercial products) that shielding and protection also allowed for unhindered views (by way of transparent glass). This gives rise to the thesis that windows are part of a longstanding architectural/technological system that moderates the dual functions of transparency and separation. With windows, multi-dimensional planes and temporalities can exist in the same time and space — hence a singular point of experience is layered with many other dimensions. Transparency and luminosity "ceases to be that which is perfectly clear and becomes instead that which is clearly ambiguous" (Rowe and Slutsky 45). The light box air-portals necessitate a constant fluctuation and remediation that is at once multi-planar, transparent and "hard to read". They are informatic.From holes in the wall to power lunch at 35,000 feet, windows shape the manner in which light, information, sights, smells, temperature and so on are modulated in society. "By allowing the outside in and the inside out, [they] enable cosmos and construction to innocently, transparently, converge" (Fuller, "Welcome" 163). Laptop, phone, PDA and light box point to the differential mobilities within a matrix that traverses multiple modes of transparency and separation, rest and flight, stillness and speed.6. Can You Feel It?Increasingly the whole world has come to smell alike: gasoline, detergents, plumbing, and junk foods coalesce into the catholic smog of our age (Illich 47).In these forlorn corners of mobile consumption, the dynamic of circulation simultaneously slows and opens out. The surfaces of inscription implore us to see them at precisely the moment we feel unseen, unguided and off-camera. Can you see it, can you feel it, can you imagine the unimaginable, all available to us on demand? Expectation and anticipation give us something to look forward to, but we're not sure we want what's on offer.Air travel radicalizes the separation of the air traveller from ground at one instance and from the atmosphere at another. Air, light, temperature and smell are all screened out or technologically created by the terminal plant and infrastructure. The closer the traveller moves towards stillness, the greater the engagement with senses that may have been ignored by the primacy of the visual in so much of this circulatory space. Smell, hunger, tiredness, cold and hardness cannot be screened out.In this sense, the airplanes we board are terminal extensions, flying air-conditioned towers or groundscrapers jet-propelled into highways of the air. Floating above the horizon, immersed in a set of logistically ordained trajectories and pressurized bubbles, we look out the window and don't see much at all. Whatever we do see, it's probably on the screen in front of us which disconnects us from one space-time-velocity at the same time that it plugs us into another set of relations. As Koolhaas says, junkspace is "held together not by structure, but by skin, like a bubble" (Koolhaas). In these distended bubbles, the traveler momentarily occupies an uncommon transit space where stillness is privileged and velocity is minimized. The traveler's body itself is "engaged in and enacting a whole kaleidoscope of different everyday practices and forms" during the course of this less-harried navigation (Bissell 282).7. Elevator MusicsThe imaginary wheel of the kaleidoscope spins to reveal a waiting body-double occupying the projected territory of what appears to be a fashionable Miami. She's just beyond our reach, but beside her lies a portal to another dimension of the terminal's vascular system.Elevators and the networks of shafts and vents that house them, are to our buildings like veins and arteries to the body — conduits that permeate and structure the spaces of our lives while still remaining separate from the fixity of the happenings around them (Garfinkel 175). The terminal space contains a number of apparent cul-de-sacs and escape routes. Though there's no background music piped in here, another soundtrack can be heard. The Muzak corporation may douse the interior of the elevator with its own proprietary aural cologne, but at this juncture the soundscape is more "open". This functional shifting of sound from figure to ground encourages peripheral hearing, providing "an illusion of distended time", sonically separated from the continuous hum of "generators, ventilation systems and low-frequency electrical lighting" (Lanza 43).There is another dimension to this acoustic realm: “The mobile ecouteur contracts the flows of information that are supposed to keep bodies usefully and efficiently moving around ... and that turn them into functions of information flows — the speedy courier, the networking executive on a mobile phone, the scanning eyes of the consumer” (Munster 18).An elevator is a grave says an old inspector's maxim, and according to others, a mechanism to cross from one world to another. Even the quintessential near death experience with its movement down a long illuminated tunnel, Garfinkel reminds us, “is not unlike the sensation of movement we experience, or imagine, in a long swift elevator ride” (Garfinkel 191).8. States of SuspensionThe suspended figure on the screen occupies an impossible pose in an impossible space: half falling, half resting, an anti-angel for today's weary air traveller. But it's the same impossible space revealed by the airport and bundled up in the experience of flight. After all, the dimension this figures exists in — witness the amount of activity in his suspension — is almost like a black hole with the surrounding universe collapsing into it. The figure is crammed into the light box uncomfortably like passengers in the plane, and yet occupies a position that does not exist in the Cartesian universe.We return to the glossy language of advertising, its promise of the external world of places and products delivered to us by the image and the network of travel. (Remmele) Here we can go beyond Virilio's vanishing point, that radical reversibility where inside and outside coincide. Since everybody has already reached their destination, for Virilio it has become completely pointless to leave: "the inertia that undermines your corporeity also undermines the GLOBAL and the LOCAL; but also, just as much, the MOBILE and the IMMOBILE” (Virilio 123; emphasis in original).In this clinical corner of stainless steel, glass bricks and exit signs hangs an animated suspension that articulates the convergence of a multitude of differentials in one image. Fallen into the weirdest geometry in the world, it's as if the passenger exists in a non-place free of all traces. Flows and conglomerates follow one another, accumulating in the edges, awaiting their moment to be sent off on another trajectory, occupying so many spatio-temporal registers in a dynamic range of mobility.ReferencesAppleyard, Donald.
