Głównym celem powyższego artykułu było udowodnienie, że Bułhakow w swym Mistrzu i Małgorzacie – poprzez obrazy Jeszui Ha-Nocri, Piłata, Mateusza Lewiego i Kaify – bronił chrześcijaństwa personalistycznego i pełnego miłosierdzia, odrzucał zaś odżywające niestety i w naszych czasach, w latach 20. XXI wieku, „chrześcijaństwo średniowieczne”, posługujące się w swych argumentacjach i praktyce przemocą oraz mieczem. Taki był właśnie, zdaniem autora powyższego artykułu, ideowy cel Bułhakowa, a środki artystyczne mające temu służyć czerpał autor Mistrza i Małgorzaty z literatury religioznawczej i biblijnej. Wbrew szeroko rozpowszechnionym opiniom Bułhakow – inaczej aniżeli powieściowy Mistrz – korzystał podczas tworzenia wątku biblijnego głównie nie ze źródłowych prac w językach obcych, lecz z przetłumaczonych na rosyjski książek na ogół popularnonaukowych, często o zabarwieniu ideologicznym, autorstwa Ernesta Renana czy Frederica Farrara. Jednak to wcale nie dogłębna znajomość historii biblijnej, lecz przede wszystkim geniusz pisarski samego Bułhakowa – wzmocniony jeszcze jego lekturami w języku przede wszystkim ojczystym, ale także przecież tłumaczeń książek obcych – pozwoliły mu „odgadnąć”, tak jak to on sam dogłębnie pojmował, historię Jeszui Ha-Nocri, Piłata, Mateusza Lewiego czy arcykapłana Kaify. W ustaleniu albo przynajmniej zarysowaniu tego wszystkiego pomogła autorowi powyższego artykułu trzydziestopięcioletnia już praca badawcza nad Mistrzem i Małgorzatą, rozpoczęła jeszcze w połowie lat 80. we współpracy z Andrzejem Drawiczem i kontynuowana potem poszukiwaniami w archiwum Bułhakowa w Moskwie, a teraz zwieńczona – jak na razie – rodzinnym tłumaczeniem na polski Mistrza i Małgorzaty, które ukazało się wraz z nowym korpusem komentarzy w 2016 roku w krakowskim wydawnictwie Znak.

Artykuł porusza kwestię relacji pomiędzy filmem a literaturą na przykładach najsłynniejszej książki Michaiła Bułhakowa i inspirowanego nią, lecz będącego samodzielną wypowiedzią artystyczną obrazu Wojciecha Marczewskiego. Z jednej strony szkic stanowi zestawienie interpretacji i spostrzeżeń innych autorów, z drugiej – podejmuje próbę wskazania nowych odniesień komparatystyczno-intertekstualnych. Temu właśnie podporządkowano układ artykułu. W początkowych partiach omówiona została struktura postaci Cenzora, który skupia w sobie kilku powieściowych bohaterów: przede wszystkim Poncjusza Piłata, Berlioza, mistrza czy Judę z Kiriatu. Następnie ukazano sposób manifestowania się w filmie najważniejszych w Mistrzu i Małgorzacie, a niewystępujących na ekranie Jeszui oraz Wolanda, a także innych,pobocznych Bułhakowowskich charakterów. W kolejnej części tekstu analizie poddano motywy, cytaty, schematy stanowiące tworzywo dzieła Wojciecha Marczewskiego oraz reżyserską strategię prowadzącą do ich twórczego przekształcenia. Nadało to jego artystycznej wypowiedzi głębi, czyniąc ją wielopłaszczyznowym traktatem moralno-filozoficznym.

W artykule przedstawiono analizę perykopy zawierającej wołanie "Hosanna" w wersji Mt 21,9. Dla lepszego zrozumienia znaczenia tego słowa, które często wyjaśniane jest jako lityrgiczny okrzyk, omówiono szerszy kontekst niż ograniczający się zazwyczaj do Ps 118. W szczególności przeanalizowano kilka polskich wydań Biblii, a także starożytne tłumaczenia Septuaginta, Targum i Peszitta Starego i Nowego Testamentu. Przeanalizowano również liturgię żydowską nazywaną "Hoszanot" wykazując, że w modlitwach hebrajskie słowo "hosza na" jest używane w jego bezpośrednim znaczeniu. Ponadto, bazując na tym znaczeniu, odnotowano powiązanie wołania "Hosanna" z wersetami poprzedzającymi Ewangelii Mateusza, gdzie niewidomi wołali o zbawienie. Godnym odnotowania jest fakt, że niewidomi zostali uzdrowieni zarówno przed, jak i po analizowanym wydarzeniu. Ze względu na to, że kontekst wskazuje na misję prorocką Jezusa spełnioną w Świątyni, nieco uwagi poświęcono zagadnieniu proroctwa. Stwierdzono, że spełnienie wyroku Jesusa nad bezowocnym drzewem figowym zaznacza nieuniknione spełnienie proroctwa o zburzeniu Jerozolimy. Całościowy kontekst daje mocne podstawy do rozumienia tekstu w taki sposób, że Jezus podkreśla swoją tożsamość jako Boga zarówno słowami, jak i czynami, zaś spontaniczne wołanie tłumu "Hosanna" dostarcza kluczy do zrozumienia tego przesłania.

Albuca crispa is a new species of section Falconera series Trianthera with crispulate leaves from the Great Karoo, known at least since 1947 but overlooked until now. A second new species. A grandis. from the southwestern Cape was previously included in A. fragrans Jacq. (section Falconera series Falconera). It is a robust species that flowers in winter and early spring and the styles are rugulose with ± isodiametric epidermal cells, unlike typical A. fragrans which is a more slender species flowering in early summer and with derived, smooth styles with fusiform epidermal cells. The recent discovery of a flowering population matching the type of A. albucoides (Aiton) J.C.Manning Goldblatt (subgenus Osmyne) allows for a full description and illustration of this poorly know n and taxonomically neglected species that has often been included in A. suaveolens (Jacq.) J.C.Manning Goldblatt. Lastly, examination of the type of A. reflexa Krause Dinter from Namibia shows it to be conspecific w ith Drimia indica (Roxb.) Jessop.

The estimated population of congestive heart failure (CHF) patients in Pakistan is 28 millions.1 Besides epidemics of type 2 diabetes mellitus and coronary heart disease, South Asian countries are also be at an increased risk of heart failure at earlier ages than other racial/ethnic groups.2 Heart failure sub classified into three categories:
With preserved ejection fraction (LVEF>50%),
Mid-range ejection fraction (LVEF41-49%),
Reduced ejection fraction (LVEF<40%).3
According to studies in United States of American (USA) and United Kingdom (UK)4,5 heart failure with reduced ejection fraction (HFrEF) prevalence has increased due to ageing population, improved survival from myocardial infarction and high prevalence of co-morbid conditions like diabetes and obesity. With increasing number of young patients (<40 years) with acute myocardial infarction (AMI) 12%6 in our population, prevalence of heart failure with predisposing factors need to be explored.
Other than diagnosing and work up of these patients, the most challenging part is the pharmacological treatment by therapeutic agents proven to reduce morbidity and mortality in HRrEF. Registries have shown under-usage of angiotensin-converting enzyme (ACE) inhibitors, angiotensin II receptor blockers (ARBs), angiotensin receptor-neprilysin inhibitor (ARNI), Beta-blockers and mineralocorticoid receptor antagonists (MRA) in such patients.7 Reason of not acheving the outcomes were due to not attaining the target levels of drugs dosages.8
With recommendations from new guidelines new novel drug therapies i.e. sodium-glucose cotransporter-2 (SGLT2) inhibitors, most debatable questions from the physicians are keeping in view the hemodynamic status and kidney function. Questions coming to the Physicians minds include;9
Should all guideline directed medical therapies be started together or stage wise?
Which drugs should be titrated first?
How quick can one up-titrate B-blockers and ARNI?
At what level of kidney impairment should one stop ACE/ARB/ARNI/SGLT2 inhibitors?
When should one refer these patients for cardiac resynchronization therapy device (CRTD) or Heart Transplantation?
When should one repeat transesophageal echocardiography (TEE)?
Physicians need to have clear answers and stance on the above queries.
HFrEF is a major public health concern in our population especially with early onset of ischemic heart disease (IHD). Awareness, education and up to date knowledge regarding early diagnosis, work up and adjustments of drugs in such patients with proper follow up is important to reduce the ever rising morbidity and mortality in our population.

Prompted by continued imports and its impact on US troops in the Spanish–American War of 1898, US Surgeon-General George Sternberg sent a small Commission to investigate the problem in US-occupied Cuba. It comprised James Carrol, Jesse Lazear and Aristide Agramonte; its leader was Surgeon-Major Walter Reed. Whilst in Cuba they collaborated with Cuban Juan Guitéras of Havana's Sanitary Department and Surgeon-Major William Gorgas, American chief of the island's Sanitary Department.

American historiography puts forward a theory which looks upon the US history as consisting of a row of cycles. The pattern was detected by thinkers and historians like Ralph W. Emerson, Henry B. Adams, Arthur M. Schlesinger and others. A cycle includes two contradicting phases lasting approximately 15-20 years each. Their character and content are defined differently - by social interest/personal interest, liberalism/conservatism, democracy/capitalism. The common ground between all the oppositions is the vision of the cyclic regularity nature. During the social anxiety periods the energy breaks out, the nation stirs to action (Progressive Era (1890s-1910s), New Deal (30s), turbulent years (60s). When the social organism gets tired it demands a break to recover. The social rest time begins (Roaring Twenties, presidency of Dwight D. Eisenhower (1953-1961), Me Decade (80s)). The article takes as a premise that cinema reflects the rythmic variations - at the level of ideas, themes, types of characters, genres, plots, a visual style. The theory is tested by means of examination of the western - the oldest national American genre. The article analyses the western subgenre - films telling of legendary frontier outlaws, namely Jesse James regarded as American Robin Hood. The theory of cycles optics enables to track the transformation of James myth and his image. The main part of the article is devoted to the landmark film of the contemporary social anxiety phase The Assassination of Jesse James by the Coward Robert Ford (2007, dir. Andrew Dominik). A thorough review of the polyphonic text demostrates that whichever interpretation is prefered the intention of the authors to a radical reconsideration of the well known myth is obvious. Correlations and contrasts with the other Jesse James films reveal that the view on the criminal number one directly corresponds with the historical phase.

Le récit d’Is 6 est rédigé selon le schème des récits de la vocation. Il se compose de la vision (vv. 1-7) et de l’audience (8-13). La liturgie chrétienne propose aux auditeurs seulement la premières partie du chapitre en terminant par le v. 8: «Me voici, envoie-moi». Mais un vrai problème du chapitre se trouve dans sa deuxième partie où Dieu révèle au prophète une inefficacité de sa prediction. Les impératifs «appesantis le coeur de ce peuple» font l’impression que la prédication du messager divin causera l’endurcissement des Israelites, le manque de la conversion et du salut. Cependant ces impératifs sont à entendre comme futurs: ils dévoilent au prophète les difficultés au cours de sa mission, qui a pour but évidemment le salut du peuple. Et ce salut est bien visible dans le denier verset du récit qui parle de la survivance, après une punition terrible, d’un Reste, de la Semence Sainte. Ainsi la vocation finit non seulement par vision de la survivance d 'une petite partie du peuple, mais aussi de sa sanctification. Il est aussi très probable que dans l’appellatif zera’ qodes est compris le Messie personnel, du Rejeton de Jesse du chapitre 11, l et Ie Germe de Yahvé (semah Yahvé) du chapitre 4,2.Selon le récit de vocation d’Isaïe (Is 6, 1-13) l’aventure de la parole de Dieu se déploie dans le coeur humain. C’est pourquoi il vaut la peine de lire et de mediter toute la narration de la vocation d’Isaïe: la parole de Dieu entre dans le coeur de l’homme, le libère de l’endurcissement, et le conduit finalement à la conversion et la sainteté.

This essay examines the recurring preoccupation with geography in W. E. B. Du Bois’s and Jessie Redmon Fauset’s African American children’s magazine, The Brownies’ Book (1920–1921). Drawing in part on conventions established by early Black periodicals, including an emphasis on the rich global presence of non-Western peoples and places, many of the magazine’s features, from its stories and poems to its images and games, offered Black children a much wider view of their place in the world—both literally and imaginatively—than that provided by typical U.S. schoolroom atlases and geographies, which tended to have little to say (or show) about countries and continents outside North America and Europe. By aiming to develop in its readers alternative forms of geographic and political consciousness, The Brownies’ Book provocatively recast geography as a radical mode of knowledge available to Black children through cultural as well as cartographic forms, in the process remapping Black childhood itself.

The asymptotic behavior of the modulus of a characteristic function of a random variable, the distribution function of which is the classical singular Cantor function, is investigated. The emphasis is on calculating the upper bound of the modulus of the characteristic Cantor distribution function. The probabilistic measure corresponding to Cantor's distribution belongs to the class of Bernoulli's symmetric convolutions, the interest in which is considerable today. Bernoulli's symmetrical convolutions were actively studied by both domestic mathematicians: M. Pratsovyty, G. Turbin, G. Torbin, J. Honcharenko, O. Baranovsky and others, and foreign ones: Erdos P, Peres Y, Schlag W, Solomyak B, Albeverio, S and other. The value of the upper bound of the modulus of the characteristic function plays an important role in the problem of determining the Lebesgue structure of distributions of sums of probably convergent random series with independent discrete terms (random values of the Jessen-Winter type). The exact value of the upper bound of the module of the characteristic Cantor distribution function is found in the article.

Increasingly, research confirms that teacher and teaching quality are the most powerful predictors of student success. In short, principals ensure higher student achievement by assuring better teaching. To do this, effective administrators have a new teacher induction program available for all newly hired teachers, which then seamlessly becomes part of the lifelong, sustained professional development program for the district or school. What keeps a good teacher are structured, sustained, intensive professional development programs that allow new teachers to observe others, to be observed by others, and to be part of networks or study groups where all teachers share together, grow together, and learn to respect each other’s work.ResumoEste artigo apresenta escolas e distritos escolares com programas bem sucedidos de indução, todos facilmente replicáveis. Cada vez mais, a pesquisa confirma que o professor e a qualidade do ensino são os mais poderosos preditores do sucesso do aluno. Em suma, os diretores garantem maior desempenho dos alunos, garantindo melhor ensino. Para fazer isso, os administradores eficazes têm um novo programa de indução de professores disponível para todos os professores recém-contratados, que então se torna parte do programa de desenvolvimento profissional sustentado ao longo da vida para o distrito ou escola. The paper deals with the problem of deepening the Jessen-Wintner theorem for generalized Bernoulli convolutions of a special kind. The main attention is paid to the case when the terms of a random series acquire three values: 0, 1, 2. In the case when the probability that the term of a random series becomes 2 is 0, the corresponding generalized Bernoulli convolutions coincide with classic Bernoulli convolutions, which were actively studied domestic scientists (Pratsovyty M., Turbin G., Torbin G., Honcharenko Ya., Baranovsky O., Savchenko I. and others) as well as foreign researchers (Erdos P., Peres Y., Schlag W, Solomyak B., Albeverio S. and others). The problem of deepening the Jessen-Wintner theorem concerning the necessary and sufficient conditions for the distribution of a probably convergent random series with discrete additions to each of the three pure types, is extremely difficult to formulate and is not completely solved even for classical Bernoulli convolutions. The results of the study are a deepening in relation to the analysis of the Lebesgue structure of random series formed by s-expansions of real numbers. In the case when the corresponding Bernoulli convolution is generated by the sequence 3-n, we have a random variable with independent triple digits, which was studied by scientists in different directions: Lebesgue structure (Chaterji S., Marsaglia G.), topological-metric structure of the distribution spectrum (Pratsovityi M., Turbin G.), fractal analysis of the distribution carrier (Pratsovyty M., Torbin G.), asymptotic properties of the characteristic function at infinity (Honcharenko Ya., Pratsovyty M., Torbin G.). The paper presents certain sufficient conditions for the absolute continuity and singularity of the distribution, with certain restrictions on the stochastic distribution matrix and the asymptotics of the values of the random terms of the series. In the case when the Lebesgue measure of the set of realizations of the generalized Bernoulli convolution is different from zero, it is possible together with Levy's theorem to formulate criteria for belonging of the Bernoulli convolution distribution to each of the three pure Lebesgue types, namely: purely discrete, purely continuous or purely singular.

The Crisis was established in 1910 by the National Association for the Advancement of Colored People at the behest and under the editorship of W. E. B. DuBois. Its earliest years of publication are distinctive, as Anne E. Carroll has argued, for their dual focus on ‘protest[ing] against racial injustice and…affirm[ing] the achievement of African Americans’. Both building on and taking issue with recent research on the Crisis by Carroll and Russ Castronovo, this essay offers a materially based, object-oriented account of how the Crisis engaged an aspirational black middle class readership—and sought to make middlebrow literary texts matter—in its Easter 1912 issue. Instead of employing the methodologies of author-based literary study, I read Charles E. Chesnutt's ‘The Doll’ and Jesse Fauset's ‘Rondeau’ in tandem with and in relation to the magazine's non-literary and commercial content. Doing so not only brings into high relief the Crisis's outreach to an aspirational black middle class. It also helps us recognize the magazine's interest in having its readers find beauty in both a barbershop and in literary forms that cannot—or rather, should not—be subsumed within expansive new definitions of modernism.

Schwartz, “‘Dynamite Talk’: William Dean Howells, Racial Socialism, and a Legal Theory of Literary Complicity” (pp. 522–550) In the wake of the 1886 Haymarket bombing in Chicago, William Dean Howells famously stood alone among his peers and against nearly all public opinion by defending the accused, denouncing the court, and appealing for clemency. Scholars ever since have found it difficult to explain Howells’s transition from the often hidebound “Dean of American Letters” into an ardent—if provisional—activist. However, as Howells made clear from the start, his sympathies were not with the anarchists themselves but against the “civic murder” committed by the court. Taking the writer at his word, this essay returns to the transcript of the Haymarket trial in order to identify the catalyst behind Howells’s unlikely conversion. By linking the defendants’ tracts and speeches with subsequent political acts allegedly committed by their sympathizers, the prosecution had manufactured a juridical reconciliation that successfully collapsed all conceptual space between word and deed, thereby leaving every writer potentially liable for the social lives of their texts. In A Hazard of New Fortunes (1889), Howells’s literary response to both the bomb and the trial, the beleaguered author would attempt to unstitch this legal contiguity by appraising characters for their susceptibility to radical speech. Yet, by linking the degree of this complicity to ethnicity rather than politics, the novel ultimately upholds the legal logics that Howells had so vigorously opposed.

Background:Many systemic sclerosis (SSc) patients develop lung fibrosis, which contribute significantly to increased mortality1. Activated and proliferating fibroblasts are responsible for the excessive extracellular matrix (ECM) formation and stiffening of the connective tissue leading to skin and lung fibrosis. There is currently no effective treatment for the fibrosis in SSc and there is therefore a medical need for further understanding the pathogenesis of fibrosis. Fibrosis is associated with different growth factors, including tumor growth factor beta 1 (TGF-β1) and platelet derived growth factor-ab (PDGF-ab)2.Objectives:We investigated how stimulation with TGF-β1 and PDGF-ab affected the migration capacity and the ECM production using translational biomarkers of type I, III and VI collagens in healthy human dermal and lung fibroblasts.Methods:The fibroblasts were grown in DMEM media containing 0.4% fetal calf serum, ficoll (to produce a crowded environment) and ascorbic acid for up to 12 days. The cells were stimulated with TGF-β1 [0.04-1 nM] or PDGF-ab [3 nM] at treatment initiation and changed twice a week. Non-stimulated fibroblasts (w/o) were used as control. A wound was induced by scratching the cells at day 1 after treatment initiation and the migration was followed for 2 days. Type I, III and VI collagen formation (PRO-C1, PRO-C3 and PRO-C6, respectively) were evaluated by validated ELISAs (Nordic Bioscience) in supernatant from day 0, 4, 8 and 12. Statistical analysis included 2-way ANOVA and Dunnett’s test.Results:The PDGF-ab stimulated dermal fibroblasts migrated significantly more than the non-stimulated (p<0.0001) and TGF-β1 stimulated (p<0.001) dermal fibroblasts 48 hours after the scratch (migration app. 70%, 30% and 30% respectively). There was no difference between the migration of the non-stimulated, TGF-β1 and PDGF-ab stimulated lung fibroblasts after 48 hours, as all migrated to approximately 70%.TGF-β1 stimulation led to a significant increase in type I collagen formation (PRO-C1) in both dermal and lung fibroblasts from day 4 and onwards compared to w/o (p<0.0001). TGF-β1 also lead to a significant increase in type III collagen formation (PRO-C3) from day 8 in lung fibroblasts compared to w/o (p<0.0001). PDGF-ab stimulation led to a significant increase in type III collagen formation in dermal fibroblasts from day 8 compared to w/o (p<0.0001). PDGF-ab stimulation led to a significant increase in type VI collagen formation (PRO-C6) in both dermal and lung fibroblasts from day 4 and onwards compared to w/o (p<0.0001).Conclusion:PDGF-ab increased the migration activity of the dermal fibroblasts, where the lung fibroblasts had a general high migration activity. The dermal and lung fibroblasts showcase the same ECM production within both type I and type VI collagen formation. The two fibroblasts types did however react opposite each other regarding the type III collagen formation: the dermal fibroblasts responded to PDGF-ab stimulation, where the lung fibroblasts responded to the TGF-β1 stimulation. The clear differences in the ECM production between the dermal and lung fibroblasts can be important in the search for an effective treatment for fibrosis in SSc and related lung fibrosis.References:[1]McNearney, T. Abstract Background: Patients with relapsed small cell lung cancer (SCLC) have a poor prognosis and few therapeutic options, especially after standard treatment of platinum doublet chemotherapy +/- immunotherapy. SCLC is characterized with high replication stress. A previous single-arm phase 2 study showed anti-tumor activity with the addition of ataxia-telangiectasia–mutated and rad3-related (ATR) kinase inhibitor berzosertib to topotecan. This study aimed to investigate whether the addition of berzosertib to topotecan improves clinical outcomes in patients with relapsed SCLC. Patients and methods: The randomized, open-labeled phase 2 study recruited patients with SCLC and relapse after one or more prior therapies from 16 US cancer centers as members of the National Cancer Institute Cancer Therapy Evaluation Program. Patients previously treated with topotecan were not eligible. Eligible patients were randomly assigned to receive topotecan (arm 1: 1.25 mg/m2 intravenously on days 1 through 5) alone or with berzosertib (arm 2: 210 mg/m2 intravenously on days 2 and 5), in 21-day cycles. Randomization was stratified by sensitivity to first-line platinum-based chemotherapy. The primary endpoint was progression-free survival (PFS) in the intention-to-treat population. Secondary endpoints included overall survival (OS), objective response rate (ORR), and duration of response (DOR) in the overall population and in platinum-sensitive or -resistant patients. Results: Between December 2019 and December 2022, sixty patients were randomly assigned to receive topotecan alone (n=20) or in combination of with berzosertib (n=40). After a median potential follow-up of 21.3 months, there was no difference in PFS between the two arms [median: 3.0 months in arm 1 versus 3.9 months in arm 2, hazard ratio (HR) (95% CI): 0.80 (0.46 to 1.41), P = 0.44]. OS was significantly longer with the combination [5.4 months in arm 1 versus 8.9 months in arm 2, HR (95% CI): 0.53 (0.29 to 0.96), P = 0.033]. Of 57 evaluable patients, a numerically higher ORR was observed in patients receiving the combination: 5.6% (1/18, 95% CI: 0.1% to 27.3%) in arm 1 versus 25.6% (10/39, 13.0% to 42.1%) in arm 2. In an unplanned post hoc analysis, the depth of target lesion responses was significantly associated with better OS in patients on the combination arm (adjusted P = 0.022 by landmark analysis), suggesting that the deeper tumor regressions by the combination of berzosertib and topotecan may have eventually contributed to OS improvement. Adverse event profiles were similar between the two arms. Conclusions: In this randomized clinical trial, berzosertib plus topotecan did not improve PFS compared with topotecan alone in patients with relapsed SCLC. However, the combination significantly improved OS, suggesting potential efficacy of replication-stress targeted therapies in SCLC. Abstract Background: Cholangiocarcinoma (CCA) is a lethal and heterogenous malignancy of the biliary tree characterized by perineural invasion. About 85% of CCA show activation of YAP signaling, which promotes proliferation, anti-apoptosis, and therapeutic resistance. YAP signaling is mediated by Src family kinases (SFK) by phosphorylation of YAP (pYAPy357). NXP900 is a first-in-class, highly selective, SFK inhibitor with a novel mechanism of action which locks SFK in its closed and inhibited conformation, in contrast to other Src inhibitors such as dasatinib. A phase 1a study of NXP900 in patients with advanced solid tumors was recently initiated. Here, we examined treatment responses of CCA to NXP900 in vitro and in vivo. Methods: Cell viability was determined in seven CCA cell lines by CellTiter-Glo. Cell death and apoptosis were assayed by PI/Hoechst and Caspase-Glo 3/7 assay. CalcuSyn software was used to determine synergistic drug effects. Immunoblot analysis, RT-PCR, and IF staining of YAP localization were used to evaluate YAP knockdown. Clones of human CCA cell lines resistant to NXP900 were generated through escalating exposure to NXP900 over 6 months. Five patient-derived xenograft (PDX) tumors were expanded into flanks of NOD/SCID mice. Tumor bearing mice were randomized 1:1 with 5 mice per arm and treated with vehicle or NXP900 (40 mg/kg) once daily for up to 4 weeks. To determine drivers of sensitivity and resistance, predicted sensitivity scores based on tumor growth were linked to multi-omics (RNA seq and [phospho-]proteomics) of our PDX models. Results: All cell lines were sensitive to NXP900 with IC50 values between 7nM-15µM; IC50’s of resistant clones were 1000 times higher. NXP900 induced more cell death compared to vehicle, which appeared to be through apoptosis. NXP900 inhibited pSrc, decreased pYAPy357, and upregulated inactive pYAPs127. Correspondingly, decreased YAP target gene (Cyr61, NUAK, and CTGF) levels and a nuclear-to-cytoplasmic translocation were observed. NXP900/GemCis combination therapy increased cell death demonstrated a synergistic effect (Combination Indices <1) at all concentrations. In our PDX models, treatment was associated with a significant decrease in tumor growth in 3 models (mean fold change 3.1 vs. 14.2; 1.0 vs. 7.6; 1.4 vs. 2.6). The major resistant signature was the TRK signaling network, involved in nerve growth factor binding activity, while drivers of sensitivity included the SRC network. Conclusion: NXP900 demonstrated therapeutic activity in vitro and in human PDX models. We are currently performing multi-omic approaches in NXP900 sensitive and resistant cell lines to unravel determinants of activity and resistance. Fertilisation is less than expected with spermatozoa from bulls consuming toxic endophyte-infected (E+) tall fescue. The objective of this study was to evaluate motility characteristics of spermatozoa from bulls grazing tall fescue pastures using computer-assisted sperm analysis (CASA). Semen was collected from six Angus bulls (average age = 15.1 ± 0.04 months) during a three-month grazing study. Bulls grazed Kentucky 31 tall fescue (Festuca arundinacea Schreb.) infected with Neotyphodium coenophialum, an ergot alkaloid-producing endophyte (n = 3), or Jesup tall fescue with Max-Q™ (NTE), a non-ergot alkaloid producing endophyte (n = 3), and grouped by body weight and scrotal circumference to graze pastures from April 18 to June 26. Semen was collected once per week between 0600–0800 h beginning in mid-May and ending the last week of June. Gross motility and morphology was evaluated before extending with Bioxcell® animal protein-free formula (IMV, Aigle, France) and antibiotics (CSS 100, 2% of total volume). Extended semen was then evaluated using CASA to determine final dilution and packaged into straws (20 million sperm/straw), where equilibration occurred over 3 h in a cold room at 4°C. Straws were frozen for 7 min in static vapor of liquid nitrogen and plunged into goblets filled with liquid nitrogen. Semen was thawed and assessed using CASA at 0 and 3 h post-thaw. Data were analysed as a randomised block design with the fixed effects of treatment, blocking on semen collection date, utilising the mixed models procedure of SAS 9.2 (SAS Institute Inc., Cary, NC, USA). Data were tested for normality (Shapiro-Wilk W ≥ 0.90), and treatment differences were determined using F-protected least significant differences. Path velocity (P = 0.001) and progressive velocity (P = 0.003) were lower in spermatozoa from bulls grazing E+ during the last 2 weeks of collection in June independent of time of assessment post-thaw. Sperm head area decreased in size in spermatozoa from E+ grazing bulls at 3 h post-thaw (P = 0.04) compared with NTE grazing bulls. Percent of rapid (progressive % with path velocity >50 μm s–1) and medium (progressive % with path velocity <50 μm s–1 but > 30μm s–1) velocity spermatozoa was decreased for E+ grazing bulls compared to NTE grazing bulls (P < 0.0001 and P = 0.004, respectively) and was accompanied by an increase in static (immobile) spermatozoa from E+ bulls (P < 0.0001). These findings indicate that spermatozoa movement and velocity are impaired in bulls grazing E+ tall fescue pastures compared to bulls grazing NTE tall fescue pastures after the freeze and thaw process, which may explain decreased fertilisation and cleavage rates of oocytes co-incubated with these spermatozoa.
