Анотація:
Bacterial cryptic prophage (defective prophage) genes are known to drastically influence host physiology, such as causing cell growth arrest or lysis, upon expression. Many phages encode lytic proteins to destroy the cell envelope. As natural antibiotics, only a few lysis target proteins were identified. ydfD is a lytic gene from the Qin cryptic prophage that encodes a 63-amino-acid protein, the ectopic expression of which in Escherichia coli can cause nearly complete cell lysis rapidly. The bacterial 2-C-methyl-D-erythritol 4-phosphate (MEP) pathway is responsible for synthesizing the isoprenoids uniquely required for sustaining bacterial growth. In this study, we provide evidence that YdfD can interact with IspG, a key enzyme involved in the MEP pathway, both in vivo and in vitro. We show that intact YdfD is required for the interaction with IspG to perform its lysis function and that the mRNA levels of ydfD increase significantly under certain stress conditions. Crucially, the cell lysis induced by YdfD can be abolished by the overexpression of ispG or the complementation of the IspG enzyme catalysis product methylerythritol 2,4-cyclodiphosphate. We propose that YdfD from the Qin cryptic prophage inhibits IspG to block the MEP pathway, leading to a compromised cell membrane and cell wall biosynthesis and eventual cell lysis.

The article considers scenarios of the prospective interstate power grid (ISPG) expansion in Northeast Asia with possible economic recession in China because of COVID-19 and other factors. Features of optimization of new capacities addition of renewable energy sources in the ISPG are considered. A series of computational experiments with variations in the profiles of wind and solar power plants were carried out. The assessment of impact the intermittence of renewable energy power output on the structure of ISPG was evaluated, with taking into account of possible economic recession in China. A comparative analysis of solutions to the problem of optimization of intermittent generation capacity inputs for scenarios with the formation of an energy connection in northeast Asia and its absence is carried out.

It is well known that supplementation with high protein after exercise can effectively promote muscle synthesis and repair, while green tea is rich in catechins that have antioxidant effects. We aimed to explore the effects of green tea combined with isolated soy protein on increase muscle mass in resistance-trained mice. A total of 32 male ICR mice (8-weeks old) were divided into four groups (n = 8/group), sedentary control group (SC), isolated soy protein with green tea group (ISPG), resistance training group (RT), isolated soy protein and green tea combine with resistance training group (ISPG + RT). All mice received control or ISPG by oral gavage for four consecutive weeks. Forelimb grip and exhaustive swimming time were used for exercise performance evaluation. In biochemical profile, we analyzed lactate, ammonia, blood urea nitrogen (BUN), and glucose and muscle damage index creatine kinase (CK) after exercise as biochemical parameters of exercise fatigue. The grip strength, muscular endurance, and exhaustive swimming time of the ISPG + RT group were significantly increased than other groups (p < 0.05), and also significantly decreased in serum lactate and ammonia levels (p < 0.05, respectively). The ISP + RT group was not only increased in quadriceps weight, (p < 0.05) but also decreased EFP (p < 0.05). We recommend using a 4-week supplementation with ISPG, combined with RT, to increase muscle mass, exercise performance, glycogen storage, and reduce fatigue biochemical parameters after exercise. The benefits of long-term supplementation or application to human supplementation can be further explored in the future.

Isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP) serve as the universal precursors for the biosynthesis of terpenes. Besides the well-known mevalonate pathway, a second biosynthetic pathway conducive to IPP and DMAPP via 1-deoxy-d-xylulose-5-phosphate and 2C-methyl-d-erythritol-4-phosphate has been discovered recently in plants and certain eubacteria. 2C-Methyl-d-erythritol-4-phosphate, the first committed intermediate of the deoxyxylulose phosphate pathway, is converted into 2C-methyl-d-erythritol 2,4-cyclodiphosphate by the catalytic action of three enzymes specified by the ispDEF genes. The cyclic diphosphate is reductively opened by the IspG protein affording 1-hydroxy-2-methyl-2-(E)-butenyl-4-diphosphate. This compound can be converted into IPP as well as DMAPP by the catalytic action of IspH protein. The enzymes of this pathway are potential targets for novel antibacterial, antimalarial, and herbicide agents.

The role of the humoral immune response in protective immunity against listerial infection has been overlooked and is essentially unknown. This study aimed to discover the protein targets of Listeria monocytogenes that elicit an antibody response following infection in a rabbit model. A genomic expression library for L. monocytogenes was constructed and differentially screened to identify genes encoding proteins that reacted with antiserum from rabbits infected with live L. monocytogenes serotype 4b (RαL), but not with that from animals immunized with heat-killed bacteria (RαK). Thirty-one clones expressing proteins that reacted exclusively with RαL were identified and sequenced. Sequence analysis, together with Western blot analysis of the proteins expressed from positive clones, led to the identification of eight L. monocytogenes proteins as targets of humoral immune responses during listerial infection: three internalin members (InlA, InlD and InlC2) and five novel proteins of unknown function (designated IspA, IspB, IspC, IspD and IspE, respectively). Exhibition of humoral immune responses to these proteins in actively infected rabbits but not in animals receiving heat-killed L. monocytogenes suggested that they were induced or significantly upregulated in vivo during infection and thus are important in Listeria pathogenesis. With the exception of antibodies to InlA, this is the first demonstration of antibodies to the other seven proteins in infected hosts. These immunogenic proteins may be useful candidates for elucidation of the role of antibodies in protective immunity in the context of listerial infection, as well as potential targets for serodiagnostic reagents and vaccine and drug development.

AbstractDeletion analysis of several 17β-estradiol (E2)-responsive genes have identified GC-rich sites that are associated with hormone-induced transactivation in MCF-7 breast cancer cells. However, the role of individual specificity proteins (Sps) in mediating hormone-induced gene expression has not been unequivocally determined. In transient transfection studies using E2-responsive GC-rich promoters from the E2F1, carbamoylphosphate synthetase/aspartate transcarbamylase/dihydroorotase (CAD), and retinoic acid receptor α (RARα) genes, RNA interference using small inhibitory RNAs for Sp1 (iSp1), Sp3 (iSp3), and Sp4 (iSp4) decreased both basal and E2-induced transactivation. The contributions of individual Sp proteins to basal and E2-induced activity were promoter dependent. iSp1, iSp3, and iSp4 also significantly inhibited hormonal induction of E2F1, CAD, and RARα mRNA levels; however, the enhanced inhibitory effects of the latter two small inhibitory RNAs suggest that Sp3 and Sp4 play a major role in estrogen receptor α/Sp-mediated gene expression in MCF-7 cells.

Анотація:
Cao Phong orange (Citrus sinensis Lour) was grown in Cao Phong district, Hoa Binh province. Citrus sinensis is Vietnam’s most valued commercial fruit. In this study, 30 microorganim strains were isolated from the samples of root, stem, and leaf of Cao Phong orange. The ratio of endophytic actinomycetes was found in different parts of the plant: root, stem, and leaf was 37%, 40%, and 23%, respectively. The isolating HBC6-2 was selected because of its high output of α-glucosidase inhibitors from 6 different strains. Morphological, physiological, and biochemical characterization showed that HBC6-2 strain belongs to the Streptomyces sp. The 16S rRNA sequence of HBC6-2 indicated 99% identity to the corresponding sequence of Streptomyces costaricanus, and was registered on GenBank with the code MT 453944.1. Streptomyces costaricanus EBL.HB6 was able to produce melanin yelow pigment, and its aerial and substrate mycelia have brown and yellow-grey pigment on ISP2, ISP3, ISP4, and ISP5 cultivating medium, respectively. The optimal pH range was from 5-10 and temperature from 15-40°C and exhibited salt tolerance up to 3% and utilized the carbon sources such as fructose, xylose, arabinose, cellulose, and rhamnose. In the following investigation, Streptomyces costaricanus EBL.HB6 displayed the highest α-glucosidase inhibitory activity by 68.98% among Streptomyces sp. strains

Implantation serine proteinase 2 (ISP2), a S1 family serine proteinase, is known for its role in the critical processes of embryo hatching and implantation in the mouse uterus. Native implantation serine proteinases (ISPs) are co-expressed and co-exist as heterodimers in uterine and blastocyst tissues. The ISP1–ISP2 enzyme complex shows trypsin-like substrate specificity. In contrast, we found that ISP2, isolated as a 34 kDa monomer from a Pichia pastoris expression system, exhibited a mixed serine proteolytic substrate specificity, as determined by a phage display peptide cleavage approach and verified by the in vitro cleavage of synthetic peptides. Based upon the peptide sequence substrate selectivity, a database search identified many potential ISP2 targets of physiological relevance, including the proteinase activated receptor 2 (PAR2). The in vitro cleavage studies with PAR2-derived peptides confirmed the mixed substrate specificity of ISP2. Treatment of cell lines expressing proteinase-activated receptors (PARs) 1, 2, and 4 with ISP2 prevented receptor activation by either thrombin (PARs 1 and 4) or trypsin (PAR2). The disarming and silencing of PARs by ISP2 may play a role in successful embryo implantation.

Анотація:
The paper considers the problems of optimization of wind and solar power plants (WPP and SPP) expansion and it operating modes in the prospective interstate power grid (ISPG) in Northeast Asia, taking into account the intermittence of their power output. A series of experiments with variations in the profiles of WPP and SPP in the model nodes were carried out. Also authors carried out some additional experiments with the increase in fuel costs for fossil fuel power plants due to environmental restrictions. As a result, an assessment of the character of the changes in the solution of the model for new installed capacities and their costs, depending on the variations for the profiles of the contributions generation of WPP and SPP to cover daily electrical load graphs, is obtained.

Metode analisis gangguan kesehatan fisik menggunakan Korelasi Spearman’s rho. Hasil penelitian menunjukkan ISPU tertinggi yaitu 374 (sangat tidak sehat). Hasil penelitian menunjukkan kadar ISPU terhadap penyakit asma (p = 0,000), ISPA (p = 0,000), pneumonia (p = 0,005), iritasi mata (p = 0,000), iritasi kulit (p = 0,001) dan diare (p = 0,002), sehingga dapat disimpulkan bahwa kadar ISPU berhubungan signifikan dengan gangguan kesehatan fisik. Berdasarkan penelitian dapat disarankan kepada peneliti selanjutnya, perlu melakukan studi cohort lanjutan terkait dampak kebakaran hutan dan lahan terhadap gangguan kesehatan fisik.

Анотація:
This study aims to explore the anti-bacterial and toxicity activities from a rare actinobacterium isolated from mangrove, Mempawah District, West Kalimantan. The mangrove mud sample from Mempawah district was inoculated on ISP4 agar using a pour plate method. After 4 days of incubation, a colony of suspected actinobacterium was appeared, then isolated and coded as SM1P. SM1P was characterized based on morphological and biochemical traits and identified as a genus of Streptroporangium then called Streptroporangium sp. SM1P. Streptroporangium sp. SM1P was carried out anti-bacterial assay on both ISP1 agar and ISP4 agar media using the cross-streak method for the solid-state fermentation. The result showed that Streptroporangium sp. SM1P could inhibit Streptococcus sp. and Salmonella typhi on ISP1 agar and treptococcus sp., Escherichia coli, Vibrio cholerae, Staphylococcus aureus and Salmonella typhi on ISP4 agar. Streptroporangium sp. SM1P was cultivated on ISP1 broth and extracted using ethyl acetate, then evaporated to obtain crude extract. The crude extract was used for anti-bacterial assay (well-diffusion method for liquid-state fermentation) and toxicity assay (brine shrimp lethality test). The crude extract was active against 2 of the test bacteria (Streptococcus sp. and E. coli). The best medium and state fermentation for anti-bacterial assay were ISP4 agar with the condition of solid-state fermentation. The extract SM1P prepared on ISP1 broth showed toxic activity based on LC50 (106.094 µg/mL). Therefore, Streptroporangium sp. SM1P have a potential source to explore secondary metabolites having anti-bacterial and toxicity activities.

Hasil penelitian menunjukkan kadar ISPU terhadap penyakit asma (p = 0,000), ISPA (p = 0,000), pneumonia (p = 0,005), iritasi mata (p = 0,000), iritasi kulit (p = 0,001) dan diare (p = 0,002), sehingga dapat disimpulkan bahwa kadar ISPU berhubungan signifikan dengan gangguan kesehatan fisik. Dari hasil penelitian, tidak ditemukan peningkatan kasus stres akut, kecemasan dan PTSD pada periode bencana kabut asap. Unlike the pulmonary artery (PA), the pathophysiological changes of the pulmonary vein (PV) in the development of pulmonary hypertension (PH) remain largely unknown. In this study, we comprehensively investigated the structural and functional changes in the PV isolated from the chronic hypoxia (CH; 10% O2, 21 days)-induced PH rat model (CHPH). Results showed that CH caused an increase in right ventricular pressure but did not affect the mean pulmonary venous pressure and the left atrial pressure. Similar to the PA, vascular lumen stenosis and medial thickening were also observed in the intrapulmonary veins isolated from the CHPH rats. Notably, CH induced more severe loss in the endothelium of intrapulmonary veins than the arteries. Then, the contractile response to 5-HT and U46619 was significantly greater in the intrapulmonary small veins (ISPV) and arteries (ISPA) isolated from CHPH rats than those from normoxic rats but not in the extrapulmonary and intrapulmonary large veins. Treatment with nifedipine (Nif), SKF96365 (SKF), or ryanodine and caffeine either partially attenuated (Nif) or dramatically abolished (SKF or ryanodine and caffeine) 5-HT-induced maximal contraction in ISPV from both normoxic and CHPH rats. Because of the severe loss of endothelium in the PV of CHPH rats, the decrease in acetylcholine (ACh)-induced endothelium-dependent relaxation was significantly larger in ISPV than ISPA, whereas the sodium nitroprusside-induced endothelium-independent relaxation was not altered in both ISPA and ISPV. In conclusion, our results provide fundamental data to comprehensively define the PV system in CHPH rat model.

Анотація:
Xylocarpus granatum produces various bioactive compounds with diverse biological activities, one of them as an antioxidant. Endophytic actinobacteria can also produce bioactive compounds. This study aimed to explore the existence of the endophytic actinobacteria of X. granatum fruit and determine its antioxidant activity. The research method was carried out by isolating the endophytic actinobacteria from the fruit using selective media Humic Acid (HV). Furthermore, characterizing the obtained isolates using four different media, namely YSA, ISP2, ISP3, and ISP4, measured antioxidant activity for actinobacterial supernatants and actinobacterial ethyl acetate extracts using the DPPH method. The results showed that from X. granatum, 15 isolates of endophytic actinobacteria were obtained from fruit, seeds, and leaves. Endophytic actinobacteria isolates from X. granatum showed different colors of aerial mycelium, substrate mycelium, and spore chain type. XGF11, XGF12, and XGF4 isolates produced high antioxidant activity of the supernatant; 57.30%, 49.77%, and 41.90% inhibition, respectively. The antioxidant capacity of XGF12 ethyl acetate extract was 501.60 μg AEAC/mg extract and 465.47 μg AEAC/mg extract for XGF11 ethyl acetate extract. Based on 16S rRNA gene analysis, isolates XGF12 and XGF11 showed similarities with Streptomyces xylanilyticus, and isolated XGF4 showed similarities with Brevibacterium sediminis.

Background: To study and assess the potential laboratory biomarkers for foreseeing postoperative pancreatic fistula (POPF) after pancreatic resection medical procedures. Subjects and Methods: 64 patients were studied from Jan 2008 to Dec 2018. All patients undergoing Pancreatic resection surgeries like Pancreaticoduodenectomy (PD), distal Pancreatectomy (DP) for tumors of the periampullary region, Pancreatic body or chronic pancreatitis Clinical data of the patients, surgical approach, pancreatic parenchymal tissue consistency, histopathology of the specimen, white blood cell count and blood amylase levels tube Drain fluid amylase were recorded and analyzed. Correlations between these parameters and postoperative pancreatic fistula (POPF) were assessed. Results : 22 out of the 64 (34.37%) patients developed POPF. The fistula was graded according to the ISPG classification. It was Grade An of every 10 (45.45%) patients, grade B in 8 (36.36%), and grade C in 4 (18.18%),.On univariate and multivariate strategic relapse investigations, higher amylase levels in the stomach waste liquid on a postoperative day (POD)1 and higher serum amylase levels on POD4 indicated a critical relationship with fistula arrangement in the post-operation period (POPF) (P < 0.05). On ROC (recipient working trademark bend) examination, amylase cut-off degree of 2270.67 U/L in the stomach waste liquid was related with a 76.6% affectability and 82% particularity [area under the bend (AUC): 0.844; P = 0.009]. A cut-off serum amylase level of 53.2 U/L was related with a 74.6% affectability and 72.9% explicitness (AUC: 0.784; P = 0.05). Conclusion: Post-operative channel liquid amylase levels on day 1 and serum amylase level on day 4 speak to interesting biomarkers related to POPF improvement after the pancreatic medical procedure.
