Hyposidra talaca (Walker) is an important pest of tea plant. H. talaca can cause losses of between 40–100% in the dry season if proper control is not carried out. H. talaca has natural enemies such as predators, parasitoids, and pathogens. One of the entomopathogens is NPV. This study aims to obtain molecular characteristics through DNA polymerase sequences and determine the virulence level of NPV isolates from H. talaca. The same species from different locations can have genetic variability. Therefore, molecular characterization by polymerase chain reaction (PCR) on DNA polymerase sequences is one way to study the genetics of HytaNPV. NPV was isolated from infected H. talaca larvae collected from the field. The DNA isolates were used as templates for PCR for DNA polymerase gene amplification with an amplicon target of ±1,000 bp. A sequencing process followed the PCR provides nucleotide sequence. HytaNPV DNA polymerase sequencing results were aligned with GenBank’s BLAST data to provide information on the relationship of HytaNPV to NPVs isolated from other regions. Based on molecular character analysis using DNA polymerase gene sequence, HytaNPV Bogor has a homology level of 93.9% with HytaNPV isolated from India. HytaNPV Bogor has a genetic relationship with the NPV that infects Buzura suppressaria from China and Australia. HytaNPV Bogor is similar to the NPV that infects H. talaca from India. The bioassay of HytaNPV isolate against H. talaca showed the highest LT50 value of 1.92 days was found in concentration of 1.58 x 107 POBs/ml in second instar larvae.

Surveys conducted during the epidemic defoliation in Sal forests of Kanha Tiger Reserve, Mandla Forest Division, Madhya Pradesh, revealed the occurrence of four insect defoliators belonging to the order Lepidoptera, viz. Caviria sericea Moore (Lymantriidae), Hyposidra talaca Walker (Geometridae) Trabala vishnou Lefroy (Lasiocampidae) and Trypanophora semihyalina Kollar (Zygaenidae). The defoliation period was observed to be rainy season of the year (July-October). Defoliation intensity was noticed to be varied greatly. Complete defoliation of Sal trees was recorded and some ranges were observed to be severely affected. Among the insect species recorded, C. sericea is the first record of occurrence of Sal defoliator.

The present paper reports the occurrence of insects damaging Eucalyptus in nursery stage. Out of nine insects recorded, seven species observed as occasional defoliating species, viz. Achaea janata (Linnaeus) (Lepidoptera : Noctuidae), Catapsilia crocale Cramer (Lepidoptera : Pieridae), Dasychira grotei Moore (Lepidoptera : Lymantriidae), Eurema hecabe Linnaeus (Lepidoptera : Pieridae), Hyposidra talaca (Walker) (Lepidoptera : Geometridae), Lymantria sobrina Moore (Lepidoptera : Lymantriidae) and Spodoptera litura (Fabricius) (Lepidoptera : Noctuidae), one rare sap-sucking species, Chrysocoris purpureus Westwood (Heteroptera : Pentatomodae) and one gall making species, Leptocybe invasa Fisher & LaSalle (Hymenoptera : Eulophidae). Among the nine species collected, six defoliators and one sap sucker are new addition to the Eucalyptus entomology. Further, H. talaca is the first record of occurrence on Eucalyptus in India. On the basis of percentage infestation/damage caused by above insects, L. invasa, commonly known as blue gum chalcid wasp, was recorded only as a key insect pest in nursery stage of Eucalyptus. The results have been discussed in the light of the present observations.

Abstract Background The black inch worm (BIW), Hyposidra talaca Walker (Lepidoptera: Geometridae), is a pest that defoliates tea leaves in India, posing a significant threat to the tea industry. Nucleopolyhedrovirus (NPV) is capable of infecting larvae of this species, which has raised the possibility of its use as a biocontrol agent. Results Rearing larvae in a semi-synthetic artificial diet produced healthy adults, which is sufficient for mass culture of H. talaca to support one of the IPM components using baculovirus. In artificial diets, the NPV was evaluated for its insecticidal activity against H. talaca. The bioassay findings of inoculated H. talaca nucleopolyhedrovirus virus (HytaNPV) at various concentrations showed that it was effective in killing the BIW. Purified polyhedral inclusion bodies (PIBs) were estimated to a concentration of 1 × 1010 PIBs per ml by mixing with water, and various concentrations of 0.25, 0.5, 1, 2, 5, 7.5, 10 ml/l were evaluated against BIW. Both laboratory and field studies revealed that HytaNPV is an eco-friendly and ecologically safe agent for controlling BIW. Besides no residue was estimated in made tea after the seventh day of exposure, and it is nontoxic to non-target species. Conclusion It was found that NPV is environmentally beneficial for the control of pests on tea plants and in production of pesticide-free tea. Tea ecosystems can reduce their reliance on conventional insecticides by using HytaNPV as an alternative bio-insecticide.
