Повний текст джерелаA decade ago, my laboratory established the first role of type I IFNs receptor (IFNAR) endocytosis in the control of Jak/STAT signaling induced by type 1 IFNs (Marchetti et al., 2006). A salient question is now to elucidate why and how IFNAR endocytosis could control the Jak/STAT pathway. Two key players of endosomal sorting retained our interest: Hrs (Hepatocyte growth factor-Regulated tyrosine kinase Substrate) and STAM (Signal Transducing Adaptor Molecule). These two classical components of the ESCRT-0 (Endosomal Sorting Complexes Required for Transport-0) complex were ideally placed at the interface between signaling and membrane trafficking. By using a combination of molecular and cellular biology, biochemistry, and fluorescent microscopy, we could establish that STAM binds to the IFNAR complex at the plasma membrane to exert an inhibitory effect on Jak/STAT signaling. This inhibition is removed when IFNAR is delivered to the sorting endosome by interacting with Hrs upon IFN-α stimulation. Based on shRNA down-expression and pharmacological inhibition, we further involve the PTP1B (Protein Tyrosine Phosphatase 1B) as it activates Jak/STAT signaling upon IFN stimulation. We could also show that PTP1B activation is inhibited by STAM at the plasma membrane from experiments where IFNAR endocytosis was blocked by siRNA-mediated clathrin down-expression. This was further confirmed by protein-protein interaction experiments (Proximity Ligation Assay) showing that STAM was constitutively associated with IFNAR1, whereas the interaction between IFNAR1 and Hrs occured only at the sorting endosome. Our results therefore allow to draw a model where STAM is a constitutive handbrake on Jak/STAT signaling at the plasma membrane that is released after IFNAR endocytosis and delivery to the sorting endosome. We further show that Hrs/STAM interaction at the early endosome allows to selectively distinguish the activation of Jak/STAT signaling mediated by IFN-α or IFN-β

Повний текст джерелаThe modern rose variety ‘Jardin de Granville’, possesses proven activities against skin cell inflammatory and oxidant mechanisms and is devoted to cosmetic applications. The main goal of this study was to establish the molecular fingerprint of the different organs of ‘Jardin de Granville’. In this way, a standardized process for plant harvesting and sample extraction was developed giving access to the most exhaustive molecular fingerprint possible of the different organs. Several complementary analytical methods were implemented through HPTLC, HPLC-DAD-ELSD and UHPLC-HRMS, enabling to achieve the chromatographic fingerprint of the different organs and to identify the main constituents. These methods were selected to have increasing specificity and accuracy to bring progressive information on the molecule structure. Thus, more than 120 compounds were characterized in the different extracts. The second objective consisted in identifying specific phytochemical markers of the variety by comparing its fingerprint to those obtained from its two rose plant parents. In this way, two approaches were developed. The first one involves statistical analysis like PCA, HAC and ANOVA and allows comparing the whole sample chromatograms. The second approach performs extract chromatogram subtractions two by two and gives more detailed information. Both comparative methods led to the identification of the differential compounds existing between the different organ types which could be used to valuate some plant parts in particular. Some ‘Jardin de Granville’ specific markers were highlighted showing the method capacity to distinguish very close rose varieties, by comparing their molecular content

Повний текст джерелаBackground: Teleworking as a result of the ongoing Covid-19 pandemic has become a hot topic in recent years. HR, like all other professions, has been affected by this, HR is a complex profession that requires a lot of human interaction and therefore may be difficult to combine with a telework in all situations. Purpose: The aim is to interpret if there is a change in the HR role within organizations in the context of the increased teleworking as a result of Covid-19. Method: This study was conducted using a qualitative data collection in the form of semi-structured interviews and an interpretive approach. The data collection has since led to an empirical data analysis that identified four themes in the form of initial crisis work, the digital approach, communication in digital contexts and the future after covid-19. Conclusion: HR has had to work more operationally than before to be able to handle the problems that Covid-19 has caused. This has meant that the focus has shifted from the strategic roles that exist within HR. Therefore, the HR role has been affected but has not fundamentally changed.

One of the reasons has been that the HR function's strategic in HR-work can contribute to cost efficiency and creates value, well needed in a competitive environment. Knowledge-intensive organizations in growth are entirely dependent on its human capital, and should examine the need to maintain a studied approach to working with HR. Purpose: The purpose of this study is to understand the needs and expectations that are in place to implement a formalized HR function for knowledge-intensive companies in growth. We want to study the HR function's role and importance and nuanced the picture of the organization's incentives to implement a formalized HR function. Method: The paper consists of an inductive approach with a qualitative method. We conducted a case study of a medium sized company in growth in a knowledge-intensive industry. We have collected all the empirical data through qualitative interviews in the management-team and a questionnaire/survey to all employees in the company. Conclusion: The conclusion we reached is that the need to implement an HR function in knowledge- intensive companies in growth is context, expertise, size and leadership. Human capital is one of the major competitive advantages that tomorrow's organizations will face and the focus is on working systematically with HR and transform goals and strategies to become more efficient, value creation and strategic.

Повний текст джерелаThe Human Immunodeficiency Virus (HIV) is the etiologic agent of Acquired Immunodeficiency Syndrome (AIDS). AIDS is a disease which has a global distribution, and it is estimated that there are currently at least 36.9 million people infected with the virus. During the replication cycle, HIV promotes several changes in the physiology of the host cell to promote their survival and enhance replication. The fast progression of HIV-1 in humans and animal models is closely linked to the function of an accessory protein Nef. Among several actions of Nef, one is the most important is the down-regulation of proteins from the immune response, such as the CD4 receptor. It is known that this action causes CD4 degradation in lysosome, but the molecular mechanisms are still incompletely understood. Nef forms a tripartite complex with the cytosolic tail of the CD4 and adapter protein 2 (AP-2) in clathrin-coated vesicles, inducing CD4 internalization and lysosome degradation. Previous research has demonstrated that CD4 target to lysosomes by Nef involves targeting of this receptor to multivesicular bodies (MVBs) pathway by an atypical mechanism because, although not need charging ubiquitination, depends on the proteins from ESCRTs (Endosomal Sorting Complexes Required for Transport) machinery and the action of Alix, an accessory protein ESCRT machinery. It has been reported that Nef interacts with subunits of AP- 1, AP-2, AP-3 complexes and Nef does not appear to interact with AP-4 and AP-5 subunits. However, the role of Nef interaction with AP-1 or AP-3 in CD4 down-regulation is poorly understood. Furthermore, AP-1, AP-2 and AP-3 are potentially heterogeneous due to the existence of multiple subunits isoforms encoded by different genes. However, there are few studies to demonstrate if the different combinations of APs isoforms are form and if they have distinct functional properties. This study aim to identify and characterize cellular factors involved on CD4 down-modulation induced by Nef from HIV-1. More specifically, this study aimed to characterize the involvement of AP-1 complex in the down-regulation of CD4 by Nef HIV-1 through the functional study of the two isoforms of ?-adaptins, AP-1 subunits. By pull-down technique, we showed that Nef is able to interact with ?2. In addition, our data from immunoblots indicated that ?2- adaptin, not ?1-adaptin, is required in Nef-mediated targeting of CD4 to lysosomes and the ?2 participation in this process is conserved by Nef from different viral strains. Furthermore, by flow cytometry assay, ?2 depletion, but not ?1 depletion, compromises the reduction of surface CD4 levels induced by Nef. Immunofluorescence microscopy analysis also revealed that ?2 depletion impairs the redistribution of CD4 by Nef to juxtanuclear region, resulting in CD4 accumulation in primary endosomes. Knockdown of ?1A, another subunit of AP-1, resulted in decreased cellular levels of ?1 and ?2 and, compromising the efficient CD4 degradation by Nef. Moreover, upon artificially stabilizing ESCRT-I in early endosomes, via overexpression of HRS, internalized CD4 accumulates in enlarged HRS-GFP positive endosomes, where co-localize with ?2. Together, the results indicate that ?2-adaptin is a molecule that is essential for CD4 targeting by Nef to ESCRT/MVB pathway, being an important protein in the endo-lysosomal system. Furthermore, the results indicate that ?-adaptins isoforms not only have different functions, but also seem to compose AP-1 complex with distinct cell functions, and only the AP-1 variant comprising ?2, but not ?1, acts in the CD4 down-regulation induced by Nef. These studies contribute to a better understanding on the molecular mechanisms involved in Nef activities, which may also help to improve the understanding of the HIV pathogenesis and the related syndrome. In addition, this work contributes with the understanding of primordial process regulation on intracellular trafficking of transmembrane proteins.
