Auxin plays a key role in different plant growth and development processes, including flower opening and development. The perception and signaling of auxin depend on the cooperative action of various components, among which auxin/indole-3-acetic acid (Aux/IAA) proteins play an imperative role. In a recent study, the entire Aux/IAA gene family was identified and comprehensively analyzed in Hedychium coronarium, a scented species used as an ornamental plant for cut flowers. Phylogenetic analysis showed that the Aux/IAA gene family in H. coronarium is slightly contracted compared to Arabidopsis, with low levels of non-canonical proteins. Sequence analysis of promoters showed numerous cis-regulatory elements related to various phytohormones. HcIAA genes showed distinct expression patterns in different tissues and flower developmental stages, and some HcIAA genes showed significant responses to auxin and ethylene, indicating that Aux/IAAs may play an important role in linking hormone signaling pathways. Based on the expression profiles, HcIAA2, HcIAA4, HcIAA6 and HcIAA12, were selected as candidate genes and HcIAA2 and HcIAA4 were screened for further characterization. Downregulation of HcIAA2 and HcIAA4 by virus-induced gene silencing in H. coronarium flowers modified the total volatile compound content, suggesting that HcIAA2 and HcIAA4 play important roles in H. coronarium floral scent formation. The results presented here will provide insights into the putative roles of HcIAA genes and will assist the elucidation of their precise roles during floral scent formation.

Coilin is a marker protein for the Cajal body, a subnuclear domain acting as a site for assembly and maturation of nuclear RNA-protein complexes. Using a yeast two-hybrid screen to identify coilin-interacting proteins, we have identified hCINAP (human coilin interacting nuclear ATPase protein), a nuclear factor of 172 amino acids with a P-loop nucleotide binding motif and ATPase activity. The hCINAP protein sequence is highly conserved across its full-length from human to plants and yeast and is ubiquitously expressed in all human tissues and cell lines tested. The yeast orthologue of CINAP is a single copy, essential gene. Tagged hCINAP is present in complexes containing coilin in mammalian cells and recombinant, Escherichia coli expressed hCINAP binds directly to coilin in vitro. The 214 carboxyl-terminal residues of coilin appear essential for the interaction with hCINAP. Both immunofluorescence and fluorescent protein tagging show that hCINAP is specifically nuclear and distributed in a widespread, diffuse nucleoplasmic pattern, excluding nucleoli, with some concentration also in Cajal bodies. Overexpression of hCINAP in HeLa cells results in a decrease in the average number of Cajal bodies per nucleus, consistent with it affecting either the stability of Cajal bodies and/or their rate of assembly. The hCINAP mRNA is an alternatively spliced transcript from the TAF9 locus, which encodes the basal transcription factor subunit TAFIID32. However, hCINAP and TAFIID32 mRNAs are translated from different ATG codons and use distinct reading frames, resulting in them having no identity in their respective protein sequences.

Infantile hypercalcemia type 1 (HCINF1), previously known as idiopathic infantile hypercalcemia, is caused by mutations in the 25-hydroxyvitamin D 24-hydroxylase gene, CYP24A1. The R396W loss-of-function mutation in CYP24A1 is the second most frequent mutated allele observed in affected HCINF1 patients. We have introduced the site-specific R396W mutation within the murine Cyp24a1 gene in knock-in mice to generate a humanized model of HCINF1. On the C57Bl6 inbred background, homozygous mutant mice exhibited high perinatal lethality with 17% survival past weaning. This was corrected by crossbreeding to the CD1 outbred background. Mutant animals had hypercalcemia in the first week of life, developed nephrolithiasis, and had a very high 25(OH)D3 to 24,25(OH)2D3 ratio which is a diagnostic hallmark of the HCINF1 condition. Expression of the mutant Cyp24a1 allele was highly elevated while Cyp27b1 expression was abrogated. Impaired bone fracture healing was detected in CD1-R396w/w mutant animals. The augmented lethality of the C57Bl6-R396W strain suggests an influence of distinct genetic backgrounds. Our data point to the utility of unique knock-in mice to probe the physiological ramifications of CYP24A1 variants in isolation from other biological and environmental factors.

AbstractIn our societies, the proportions of elderly people and of obese individuals are increasing. Both factors are associated with high health-related costs. During obesity, many authors suggest that it is a high chronic intake of added sugars (HCIAS) that triggers the shift towards pathology. However, the majority of studies were performed in young subjects and only a few were interested in the interaction with the ageing process. Our purpose was to discuss the metabolic effects of HCIAS, compare with the effects of ageing, and evaluate how deleterious the combined action of HCIAS and ageing could be. This effect of HCIAS seems mediated by fructose, targeting the liver first, which may lead to all subsequent metabolic alterations. The first basic alterations induced by fructose are increased oxidative stress, protein glycation, inflammation, dyslipidaemia and insulin resistance. These alterations are also present during the ageing process, and are closely related to each other, one leading to the other. These basic alterations are also involved in more complex syndromes, which are also favoured by HCIAS, and present during ageing. These include non-alcoholic fatty liver disease, hypertension, neurodegenerative diseases, sarcopenia and osteoporosis. Cumulative effects of ageing and HCIAS have been seldom tested and may not always be strictly additive. Data also suggest that some of the metabolic alterations that are more prevalent during ageing could be related more with nutritional habits than to intrinsic ageing. In conclusion, it is clear that HCIAS interacts with the ageing process, accelerates the accumulation of metabolic alterations, and that it should be avoided.

AbstractThe principles of the best interest of children and subsidiarity constitute the conceptual foundation of the Convention on Protection of Children and Co-Operation in Respect of Intercountry Adoption (HCIA). Subsidiarity in the HCIA dictates a priority for domestic adoption placements for children over intercountry adoption. This article argues against subsidiarity on two fronts. First, the analysis shows that the in-principle priority of domestic adoption cannot be justified on the basis of either heritage rights or state sovereignty. Second, the principle of subsidiarity in the HCIA is a procedural principle, one that stipulates the political/geographical location of the placement of children through a priority ordering. This does not comport with the principle of subsidiarity as it has been conceptualized in ethics and social philosophy, which gives normative structure to the process of decision-making by stipulating the proper level for decisional authority. Subsidiarity in this original sense holds that decisions regarding child welfare should be made at the lowest level possible, by those most affected by the decisions, unless doing so would not be the most suited to protecting and promoting the best interests of children. Appealing to subsidiarity in this theoretical version reveals at least two significant problems with HCIA placement policy and leads to the conclusion that subsidiarity in the HCIA must be formally revised as a structural principle of ethics that will not support the general priority of domestic adoption.

The reactions of piperonylic acid (HPip) and cinnamic acid (HCinn) with 4,4’-bipyridine (4,4’-bipy) have been assayed using the same synthetic methodology, yielding two binary cocrystals with different acid:4,4’-bipy molar ratios, (HPip)(4,4’-bipy) (1) and (HCinn)2(4,4’-bipy) (2). The melting point (m.p.) of these cocrystals have been measured and a remarkable difference (ΔT ≈ 78 °C) between them was observed. Moreover, the two cocrystals have been characterized by powder X-ray diffraction (PXRD), elemental analysis (EA), FTIR-ATR, 1H NMR spectroscopies, and single-crystal X-ray diffraction. The study of their structural packings via Hirshfeld surface analysis and energy frameworks revealed the important contribution of the π···π and C-H···π interactions to the formation of different structural packing motifs, this being the main reason for the difference of m.p. between them. Moreover, it has been observed that 1 and 2 presented the same packing motifs as the crystal structure of their corresponding carboxylic acids, but 1 and 2 showed lower m.p. than those of the carboxylic acids, which could be related to the lower strength of the acid-pyridine heterosynthons respect to the acid-acid homosynthons in the crystal structures.

1. Combined histochemical and biochemical single-fibre analyses [Staron & Pette (1987) Biochem. J. 243, 687-693], were used to investigate the rabbit tibialis-anterior fibre population. 2. This muscle is composed of four histochemically defined fibre types (I, IIC, IIA and IIB). 3. Type I fibres contain slow myosin light chains LC1s and LC2 and the slow myosin heavy chain HCI, and types IIA and IIB contain the fast myosin light chains LC1f, LC2f and LC3f and the fast heavy chains HCIIa and HCIIb respectively. 4. A small fraction of fibres (IIAB), histochemically intermediate between types IIA and IIB, contain the fast light myosin chains but display a coexistence of HCIIa and HCIIb. 5. Similarly to the soleus muscle, C fibres in the tibialis anterior muscle contain both fast and slow myosin light chains and heavy chains. The IIC fibres show a predominance of the fast forms and the IC fibres (histochemically intermediate between types I and IIC) a predominance of the slow forms. 6. A total of 60 theoretical isomyosins can be derived from these findings on the distribution of fast and slow myosin light and heavy chains in the fibres of rabbit tibialis anterior muscle.

Objective: The objective of the study was to compare pulmonary function tests results of hyperbaric chamber inside attendants (HCIAs) working in a hyperbaric chamber before and after sessions. Methods: A total of 68 health care personnel working as HCIAs in the hyperbaric oxygen therapy unit between June 2019 and September 2019 were included in the study. All participants experienced the pressure chamber for the first time. In spirometric evaluation, we measured forced vital capacity (FVC), forced expiratory volume at one second (FEV1), forced expiratory flow at 25%-75% of FVC (FEF25-75) and peak expiratory flow (PEF). In addition, FEV1/FVC ratio (FEV1%) was also calculated. Results: The mean FVC was found as 3.56 ± 0.66 (min-max: 2.17-5.63) before hyperbaric exposure and 3.44 ± 0.62 (min-max: 2.30-5.28) after the exposure (3.4%) (p<0.05). The mean FEV1 was found as 3.37 ± 0.63 (2.13-5.39) before the session and 3.24 ± 0.59 (min-max: 2.3-5.28) after the session (3.9%) (p<0.05). There was no statistically significant difference between the mean FEV1/FVC ratio, PEF and FEF25-75 measured before and after hyperbaric exposure. Conclusion: The results of this study indicated that among pulmonary function test parameters, decreases were found in FEV1, FVC, FEF25-75, PEF, but clinical significance has not been established.

1. Six adult rabbit soleus muscles were analysed by isolating histochemically identified fibre pieces from freeze-dried serial cross-sections. 2. By the use of this method, four fibre types (I, IC, IIC and IIA) were identified and analysed micro-electrophoretically. 3. Type I fibres contained the slow myosin heavy chain HCI and the slow myosin light chains LC1s and LC2s. 4. Type IIA fibres contained the fast myosin HCIIa with the fast light chains and, in addition, either LC1s or both LC1s and LC2s. 5. The C fibres (IC and IIC) represented intermediate populations between types I and IIC (IC) and between IC and IIA (IIC). They contained varied ratios of HCI/HCIIa with both sets of fast and slow light chains. With regard to myosin composition and isoforms of other myofibrillar proteins (M- and C-proteins, alpha-tropomyosin, troponin I), IC fibres resembled type I and IIC fibres resembled type IIA. 6. The presence of various myosin light and heavy chains within a specific fibre suggests a multiplicity of isomyosins. Without consideration of LC1sa and LC1sb differences, at least 54 possible isomyosins can be derived: type I fibres contain one isomyosin, types IC and IIC 54 possible isomyosins, and type IIA up to 18.

Myosin heavy chain isoforms and enzyme activities were compared between the costal and crural regions of the rat diaphragm. The percentage of heavy chain (HC) IIb in the crural region of the diaphragm was significantly (P less than 0.05) higher than that in the costal region (mean 7.3 vs. 3.0%), and the percentage of HCI was significantly lower in the crural than in the costal diaphragm (22.7 vs. 27.9%). The distributions of HCIIa and HCIId were relatively homogeneous in both regions. Succinate dehydrogenase activity in the costal diaphragm was 21% greater (P less than 0.01) than in the crural diaphragm. In contrast, there was no significant difference in the activity of phosphofructokinase in the crural and costal diaphragms. These results demonstrate that a difference in myosin heavy chain isoforms and oxidative capacity exists between the costal and crural regions of the rat diaphragm.

Background: Concomitant administration of herbal medicine and conventional may lead to severe metabolism-oriented herb-drug interactions. However, detecting herb-drug interaction is expensive and higher time-consuming. Several computer-aided techniques have been proposed in recent years to predict drug interactions. However, most of the methods cannot predict herb-drug interactions effectively. Methods: Canonical SMILES of bioactive compounds was gathered from the PubChem online database, and its inhibition details were gathered PKCSM from the webserver. Results: By searching the bioactive compound name in the search bar of “The Herb-CYP450 Enzyme Inhibition Predictor online database” (HCIP- http://hcip.in/), it will provide the liver enzyme inhibition profile of the selected bioactive compound. For example; Guggulsterone: CYP3A4 inhibitor. Conclusion: The Herb-CYP450 Enzyme Inhibition Predictor online database is very peculiar and easy to determine the inhibition profile of the targeted bioactive compound. This study examines and analyzes the effect of; (i) Human Capital in Islamic Perspective (HCIP) on Business Sustainability (BS); (ii) Strategic Management in Islamic Perspective (SMIP) on Business Sustainability, (iii) Mediation of Strategic Management in Islamic Perspective in relation to Human Capital in Islamic Perspective on Business Sustainability. This study uses Batik MSME respondents in Pekalongan City. Pekalongan is a Batik city that has the batik talents, qualified craftsmen, known worldwide and have been passed down from one generation to the next. The target sample is 150 MSMEs. It is conducted purposively with a return rate of 116 questionnaires (response rate 77%). The data is collected through questionnaires and interviews with several owners or managers of MSMEs. The statistical technique uses Partial Least Square (PLS 3.3). This study finds that; (i) Human Capital in Islamic Perspective has no significant effect on Business Sustainability; (ii) Strategic Management in Islamic Perspective has a significant effect on Business Sustainability, (iii) Strategic Management in Islamic Perspective mediates the effect of Human Capital in Islamic Perspective on Business Sustainability

Safety-net hospitals serving the poor and indigent in inner-cities have received inadequate research attention regarding the determinants of their financial performance in the changing health care environment. We analyze how the 1990–1992 financial performance of 275 such hospitals is related to exogenous and endogenous factors such as payer mix, service mix, staffing and ownership. Models of hospital financial performance are developed using operating margin, cost per discharge and revenue per discharge as measures of performance. Stepwise regression is used to test the model with data from the American Hospital Association (AHA) and Health Care Investment Analysts (HCIA). Results suggest that: 1) The profitability of inner-city hospitals appears positively related with technical complexity of care; 2) High interest and low operating surplus may constrain the addition of technically sophisticated services to enhance profitability; 3) There is some evidence that new governmental programs, e.g. Medicaid managed care and Medicaid Diagnosis Related Groups (DRGs), may not have improved operating margins, though Medicaid DRGs appear to have contained costs. Follow-up research is needed on this issue; 4) Given external fiscal realities, internal management strategies for inner-city hospitals require research, e.g. developing appropriate managed care systems and timely expansion of sub-acute services and; 5) Services such as AIDS treatment and community health education represent opportunities to respond to community needs, especially since unit cost of such services will decline with high volume.

Industrial Internet of Things (IIoT) is a new field of study that connects digital devices and services to physical systems. The IIoT has been utilized to create massive amounts of data from various sensors, and it has run into several problems. The IIoT has been subjected to a variety of hacks, putting its ability to provide enterprises with flawless operations in jeopardy. Businesses suffer financial and reputational losses as a result of such threats, as well as the theft of critical data. As a result, numerous Network Intrusion Detection Systems (NIDSs) have been created to combat and safeguard IIoT systems, but gathering data that can be utilized in the construction of an intelligent NIDS is a tough operation; consequently, identifying current and new assaults poses major issues. In this research work, a novel IIOT attack detection framework and mitigation model is designed by following four major phases “(a) pre-processing, (b) feature extraction, (c) feature selection and (d) attack detection”. Initially, the collected raw data (input) is subjected to pre-processing phase, wherein the data cleaning and data standardization operations take place. Subsequently, the features like “higher-order statistical features (Skewness, Kurtosis, Variance and Moments), technical indicator based features, mutual information, Improved Principal Component Analysis (IPCA)” based features are extracted from the pre-processed data. Further, from the extracted features, the most optimal features are selected using a new hybrid optimization model referred as Hunger Customized Individual Activity Model (HCIA) that hybrids the concepts of standard (Teamwork Optimization Algorithm (TOA) and Hunger Games Search (HGS)). The attack detection is carried out using the projected deep fusion model framework that encapsulates the Bi-GRU and Quantum Deep Neural Network (QDNN), respectively. The Bi-GRU and QDNN in the deep fusion model framework is trained with the optimal features selected using a new hybrid optimization model. The outcome acquired from Bi-GRU and QDNN is combined, and it will be the final detected outcome that portrays the presence/ absence of attacks in IIoT network. When an attack is being identified, the mitigation of such attack takes place via the Improved BIAT Framework. Further, the projected model is evaluated over the existing models to show its supremacy in the attack detection and mitigation process.

Гомогенные сплавы системы Ag-Pd являются эффективными катализаторами катодной реакции выделения водорода. Они обладают высокой механической прочностью и, в меньшей степени в сравнении с металлическим палладием, подвергается водородному охрупчиванию. Целью работы было установление кинетики выделения водорода на палладии и его гомогенных сплавах с серебром в щелочной водной среде, а также исследование их водородопроницаемости.Методами циклической вольтамперометрии и двухступенчатой катодно-анодной хроноамперометрии исследовано поведение Pd и сплавов системы Ag–Pd (XPd = 15–80 ат. %) в водном деаэрированном растворе 0.1M KOH. Циклические вольтамперограммы для Pd и Ag80Pd имеют схожий вид, однако при введении даже небольшого количества серебра (≤ 20 ат. %) в палладий скорость ионизации водорода снижается, а дальнейшее увеличение содержания серебра в сплаве приводит к его полному подавлению. Для Ag,Pd-сплавов с содержанием палладия менее 30 ат. % пикионизации водорода на вольтамперограммах не фиксируется. Зависимости пикового тока ионизации от скорости сканирования потенциала для всех изученных сплавов линейны и экстраполируются в начало координат, что говорит о наличии твердофазных диффузионных затруднений процесса. Наклон их прямых для сплавов Ag60Pd и Ag50Pd выше, чем для сплава Ag80Pd, что может свидетельствовать о проявлении на поверхности окисленных форм серебра. На всех изученных электродах зависимость потенциала пика тока от логарифма скорости сканированиялинейно возрастает, а это указывает на необратимый характер электрохимической стадии ионизации водорода, осложненной его твердофазной диффузией. Для расчета параметров водородопроницаемости сплавов потенциостатически получены катодные и анодные спады тока при различном времени наводороживания от 1 до 10 с. С увеличением продолжительности наводороживания наблюдается уменьшение амплитуд тока на катодныхи анодных ветвях хроноамперограмм. Используя результаты теоретического моделирования, описывающие процессы инжекции и экстракции водорода для электродов полубесконечной толщины, по линеаризованным в соответствующих критериальных координатах катодным и анодным спадам тока рассчитаны параметры водородопроницаемости. Константа фазограничного обмена и константа скорости инжекции атомарного водорода имеют максимум для сплава с содержанием палладия 80 ат. %. Константа скорости экстракции водорода изменяется линейно с уменьшением содержания палладия. Найдено, что значения параметра водородопроницаемости для Ag,Pd- сплавов в щелочной среде несколько ниже, чем в кислой. Контролирующей стадией реакции выделения водорода на Ag,Pd-сплавах (XPd ≤ 40 ат. %) в растворе 0.1М KOH является электрохимическая стадия ионизации атомарного водорода, осложненная его диффузией в твердой фазе. Параметры водородопроницаемости в сплавах системы Ag–Pd максимальны при содержании палладия ~80 ат. %,а потому такие сплавы могут быть использованы как эффективные материалы для очистки и хранения водорода. The early metastasis of cervical cancer is a multi-step process requiring the cancer cells to adapt to the signal input from different tissue environments, including hypoxia. Hypoxia-induced epithelial-to-mesenchymal transition (EMT) plays a critical role in the acquisition of the ability to invade surrounding tissue. However, the molecular mechanism underlying EMT in cervical cancer remains to be elucidated. Herein, we showed that HIF‑1α and ARNT are recruited to the hCINAP promoter and initiate hCINAP expression in hypoxia. Ablation of hCINAP decreased the migratory capacity and EMT of cervical cancer cells in hypoxia. Furthermore, hCINAP regulates EMT through Akt/mTOR signaling and inhibits hypoxia-induced p53-dependent apoptosis. Our data collectively showed that hCINAP may have essential roles in the metastasis of cervical cancer and could be a potential target for curing cervical cancer.

Abstract Targeting the specific metabolic phenotypes of colorectal cancer stem cells (CRCSCs) is an innovative therapeutic strategy for colorectal cancer (CRC) patients with poor prognosis and relapse. However, the context-dependent metabolic traits of CRCSCs remain poorly elucidated. Here we report that adenylate kinase hCINAP is overexpressed in CRC tissues. Depletion of hCINAP inhibits invasion, self-renewal, tumorigenesis and chemoresistance of CRCSCs with a loss of mesenchymal signature. Mechanistically, hCINAP binds to the C-terminal domain of LDHA, the key regulator of glycolysis, and depends on its adenylate kinase activity to promote LDHA phosphorylation at tyrosine 10, resulting in the hyperactive Warburg effect and the lower cellular ROS level and conferring metabolic advantage to CRCSC invasion. Moreover, hCINAP expression is positively correlated with the level of Y10-phosphorylated LDHA in CRC patients. This study identifies hCINAP as a potent modulator of metabolic reprogramming in CRCSCs and a promising drug target for CRC invasion and metastasis.

AbstractMutations in the chromodomain helicase DNA binding protein 2 (CHD2) gene are associated with neurodevelopmental disorders. However, mechanisms by which CHD2 regulates human brain development remain largely uncharacterized. Here, we used a human embryonic stem cell model of cortical interneuron (hcIN) development to elucidate its roles in this process. We identified genome-wide CHD2 binding profiles during hcIN differentiation, defining direct CHD2 targets related to neurogenesis in hcIN progenitors and to neuronal function in hcINs. CHD2 bound sites were frequently coenriched with histone H3 lysine 27 acetylation (H3K27ac) and associated with high gene expression, indicating roles for CHD2 in promoting gene expression during hcIN development. Binding sites for different classes of transcription factors were enriched at CHD2 bound regions during differentiation, suggesting transcription factors that may cooperatively regulate stage-specific gene expression with CHD2. We also demonstrated that CHD2 haploinsufficiency altered CHD2 and H3K27ac coenrichment on chromatin and expression of associated genes, decreasing acetylation and expression of cell cycle genes while increasing acetylation and expression of neuronal genes, to cause precocious differentiation. Together, these data describe CHD2 direct targets and mechanisms by which CHD2 prevents precocious hcIN differentiation, which are likely to be disrupted by pathogenic CHD2 mutation to cause neurodevelopmental disorders.

AbstractThe second order nonlinear optical (NLO) properties of two different ionic selfassembled multilayer (ISAM) films combined with Ag nanoparticles have been investigated. The plasmon resonances in the Ag particles concentrate the incident light, markedly increasing in the NLO efficiencies of the films. We find that the efficiency enhancement is significantly larger in conventional ISAM films compared to films made using a hybrid covalent ISAM technique (HCISAM), even though the intrinsic bulk second order non-linear susceptibility (χ(2)) is much larger for HCISAM films. We attribute this to the interfaces in HCISAM films being much easier to disrupt by external perturbations such as the metal deposition by which the nanoparticles are fabricated. We conclude that because the plasmon decay length is very short, the plasmonic enhancement of NLO effects primarily occurs at and near the film-particle interface. To discern the importance of the interfaces, we surrounded thin ISAM and HCISAM films with NLOinactive buffer layers, which confirmed this hypothesis, particularly in the case of HCISAM films.

Abstract Background Pharmacological therapy may be useful in the treatment of moderate to severe hypercalcemia in patients with infantile hypercalcemia-1 (HCINF1) due to pathogenic variants in the cytochrome P450 24 subfamily A member 1 (CYP24A1). Rifampin is an antituberculosis drug that is a potent inducer of cytochrome P450 3 subfamily A member 4 (CYP3A4), involved in an alternative catabolic pathway of vitamin D. The efficacy of rifampin in improving hypercalcemia was previously reported but many questions remain on the long-term efficacy and safety. Aim of the study is to test the long-term efficacy and safety of rifampin in a patient with HCINF1. Methods We report clinical, biochemical and imaging features of a 23-year-old man affected by HCINF1 with moderate hypercalcemia (12.9 mg/dL), symptomatic nephrolithiasis, nephrocalcinosis and impaired kidney function (eGFR 60 mL/min/1.73 m2) treated with rifampin for an overall period of 24 months. Kidney, liver and adrenal function were evaluated at every follow-up visit. Results In 2 months, rifampin induced a normalization of serum calcium (9.6 mg/dL) associated with an improvement of kidney function (eGFR 92 mL/min/1.73 m2) stable during the treatment. After 15 months, rifampin was temporally withdrawn because of asthenia, unrelated to impairment of adrenal function. After three months, the timing of drug administration was shifted from the morning to the evening, obtaining the remission of asthenia. At the end of follow-up, the nephrolithiasis disappeared and the nephrocalcinosis was stable. Conclusions Rifampin could represent an effective choice to induce a stable reduction of calcium levels in patients with HCINF1, with a good safety profile.

In modern applications, instruction cache misses have become a performance constraint, and numerous prefetchers have been developed to conceal memory latency. With today's client and server workloads, large instruction working sets require more. These working sets are typically large enough to fit in the Last Level Cache (LLC). However, the Level 1 Instruction (L1-I) cache has a high miss rate, which typically prevents the processor front-end from receiving instructions. Instruction prefetching is a latency hiding method that allows the LLC to send instructions to the L1-I cache. In order to design a high-performance cache architecture, prefetching instructions in the L1-I cache is a fundamental approach. When developing an efficient and effective prefetcher, accuracy and coverage are the most important parameters to be considered. This paper proposed a novel Hybrid Short Long History Table-based Cache Instruction Prefetcher (HCIP) for the L1-I cache. The HCIP makes use of a hybrid configuration of the two history-based prefetchers tables that are Long History Table (LST) and Short History Table (SHT). The transitive closure of the control flow graph is the PRE+PC table used in HCIP. In contrast to PIPS and NOPREF, HCIP indicates maximum coverage of 67% for the majority of the benchmarks given.

Auxin, an important plant hormone, induces the biosynthesis of various secondary metabolites by modulating the expression of auxin-responsive genes. In the ornamental plant Hedychium coronarium, linalool and methyl benzoate are biosynthesized by the terpene synthase (TPS) HcTPS5 and the benzoic/salicylic acid methyltransferase (BSMT) HcBSMT2, respectively. However, the transcriptional regulation of this process remains unclear. Here, we identified and functionally characterized the R2R3-MYB transcription factors HcMYB1 and HcMYB2 in regulating the biosynthesis of these floral aroma compounds. HcMYB1 and HcMYB2 are specifically expressed in flowers, their expression is correlated with the emission of volatile compounds in flowers, and is induced by auxin. Moreover, HcMYB1 and HcMYB2 interact with the HcBSMT2 promoter region. HcMYB2 activates the expression of the linalool synthase gene HcTPS5. In flowers with HcMYB1 or HcMYB2 silenced, the levels of floral scent compounds were significantly reduced, and HcBSMT2 and HcTPS5 were downregulated compared with the wild type. Moreover, HcMYB1 form protein-protein interaction with key scent-related HcIAA4 protein to regulate floral aroma production. Taken together, these results indicate that HcMYB1 and HcMYB2 play crucial roles in regulating the formation of scent compounds in Hedychium coronarium (H. coronarium) flowers in response to auxin signaling.

Background and Objectives: Medicinal plants have become popular in the search ofa cure for various hepatopathies with a considerable number of herbal products being used for the management of liver ailments. However, most, including Hura crepitans (HC), have not been toxicologically evaluated. The present study was aimed at determining the phytoconstituents of HC, their antioxidant potentials and safety in animals. Place and Duration: The study was performed at the Department of Biochemistry and Biotechnology, KNUST, Kumasi and subacute toxicity studies lasted for 14 days. Materials and Methods: Hydroethanolic extracts and fractions of the leaves and stem back of HC were assessed for their phytochemical constituent,DPPH and FRAP scavenging activities and total phenolic and total flavonoid content. The acute and sub acute oral toxicity of the hydroethanolic stem back of HCin animals was evaluated using standard methods to establish the LD50, effect on weight, haematological and biochemical parameters.Phytochemical characterization was achieved using standard methods. Results: The study showed that HC contained tannins, saponins, flavonoids, coumarins, glycosides, and triterpenoids. The hydroethanolic extract of the stem bark (EES) showed the greatest antioxidant potential. The LD50 estimate of the hydroethanolic extract of stem back was greater than 5000 mg/kg body weight/oral route, while the doses used in this study up to 1000 mg/kg body weight/oral route was safe. The extract generally did not cause any significant decrease in the weight of animals nor were there significant differences in relative organ weights in treated animals compared to the normal group. Haematology, liver function, kidney function, and serum electrolyte parameters were within the normal ranges in treated animals when compared to the control group. Conclusion: The results showed that oral administration of hydroethanolic extract of Hura crepitansin animals up to 1000 mg/kg body weight is considered safe as it causes no significant derangement in clinical and haemato-biochemical parameters measured.
