Experiments were performed at the Yuma Valley Agricultural Center to determine how timely harvest of cotton may affect aflatoxin contamination of cottonseed As the cotton was held in the field between the final irrigation and harvest, the quantity of aflataxin in the crop increased. Significant reductions in aflatoxin contents of seed were realized by harvesting in early September.

The overall goal of our study is to find effective and environmentally sound methods to reduce pre -harvest aflatoxin contamination of cottonseed in Arizona. The specific objectives are: 1) to screen a large number of bacteria for their ability to destroy the aflatoxin producing fungus, Aspeigillus flavus; 2) to test the efficacy and consistency of the recovered antagonistic bacterial isolates to reduce aflatoxin contamination of cottonseed in field trials; 3) to study the survival and competitiveness of the antagonists on cotton plants under prevailing field conditions; 4) to find innovative procedures to enhance survivability and competitiveness of the antagonists on cotton plants; and 5) to test the potential of the bacterial antagonists to reduce the population of A. flavus in field soils. Over 800 bacterial isolates, recovered from cotton field soils, cotton leaves, stems, and immature as well as opened bolls, were tested for ability to inhibit the growth of A. flavus on cottonseed. Six isolates partially or totally inhibited the fungus. All of these effective isolates prevented the fungus fioin infecting simulated pink bollworm exit holes in immature bolls in the field.

Bitter pit fruit in commercial consignments of apples still poses an economic threat to exporters from South Africa. Bitter pit develops pre-harvest, but gets progressively worse during storage and is only traceable once the lesions appear after storage. Accurate, early indications of bitter pit incidence will allow for remedial pre-harvest measures in the field, e.g. Ca foliar applications, to reduce the potential losses. Similarly, the automatic detection of a bitter pit fruit during packing will reduce financial losses by identifying unacceptable fruit before shipping. Fluorescence imaging is a fast, non-destructive technique, able to evaluate numerous fruits individually. Results of pre-harvest imaging on apples to identify fruit susceptible to bitter pit showed that pitted fruit were correctly classified, but misclassification of non-pitted fruit with fluorescence imaging was still too high. NIR-spectroscopy point meter readings could distinguish visible bitter pit lesions from healthy tissue. Important wavelengths associated with visible bitter pit were identified. This technique could also identify immature apples, more prone to bitter pit development. It could however not distinguish between bitter pit and non-pitted fruit when applied randomly on the calyx end of apples at harvest. Pre-harvest foliar applications to increase fruit Ca content and reduce bitter pit incidence, is a standard practice world wide. External Ca uptake by fruit was monitored to determine the efficacy of applications during different stages of fruit development. Two periods of efficient uptake of external Ca were identified, viz., cell division and the last few weeks before harvest. Foliar Ca applications from 40 days after full bloom were more effective in increasing fruit Ca content and reducing bitter pit incidence than at 80 days after full bloom, which was recommended previously. Mineral analysis of fruit has been used with variable success to predict bitter pit prior to harvest. The possibility of increasing the accuracy of existing predictive models by using analysis of individual fruit rather than pooled samples, was investigated. By improving the normality of different mineral distributions and decreasing the overlap between pitted and non-pitted fruit classes, it was attempted to improve the reliability of predictions based on variable threshold values. The Ca distribution showed a variation between pitted and nonpitted classes, but still a significant overlap between classes reduced the accuracy of the predictive capacity of this distribution. Even though our results produced a correct classification of 85% for non-pitted fruit, which can be useful, this was still below the required tolerance, of less that 2%, expected on the market. The effect of pruning and fruit bearing position on two-year-old wood on dry mass and Ca allocation of fruit was determined. ‘Golden Delicious’ fruit set was the lowest at the basal bearing position compared to the other positions evaluated and was contrary to expectations. Fruit in a terminal bearing position was superior to the basal position regarding total dry weight and fruit size. Distal wood possibly inhibited growth and set on the basal position via auxin distribution. Ca allocation differed between seasons and cultivars and could either be influenced by bearing position or presence or absence of re-growth.

ENGLISH ABSTRACT: As overpopulation threatens the world’s ability to feed itself, food has become an invaluable resource. Unfortunately, almost a third of the food produced for human consumption is lost annually. Pests including insects, phytopathogens and weeds are responsible for more than a third of the annual major crop losses suffered around the world. The majority of current post-harvest control strategies employ synthetic agents. These compounds, however, have been found to be detrimental to the environment as well as human health, which has led researchers to investigate alternative strategies. Biocontrol agents are environmentally compatible, have a lower toxicity and are biodegradable, making them an attractive alternative to the synthetic control agents. The lipopeptides produced by Bacillus spp. in particular, have shown great potential as biocontrol agents against various post-harvest phytopathogens. Most biocontrol strategies apply the biocontrol organism, for example Bacillus, directly, whereas this study focused on the use of the lipopeptide itself as an antifungal agent. This is advantageous as the lipopeptides are less sensitive to their surroundings, such as temperature and pH, compared to living organisms, allowing for the production of a standardized product. This study investigated the production of the Bacillus lipopeptides surfactin, fengycin and iturin under controlled batch conditions. Parameters increasing lipopeptide production were quantified, focussing on antifungal lipopeptides (iturin and fengycin), and lipopeptide production was optimized. Experiments were performed in a fully instrumented 1.3 L bench-top bioreactor and lipopeptide analyses were performed via high pressure liquid chromatography (HPLC) and liquid chromatography-mass spectroscopy (LC-MS). After screening four Bacillus spp., Bacillus amyloliquefaciens DSM 23117 was found to be the best antifungal candidate. This was based on it outperforming other candidates in terms of maximum antifungals produced, Yp/x,antifungals (yield per cells), and antifungal productivity. Nitrate, in the form of NH4NO3, was critical for lipopeptide production and an optimum concentration was observed above which the CDW (cell dry weight) no longer increased significantly and both μmax (maximum specific growth rate, h-1) and lipopeptide production decreased. For μmax, the optimum NH3NO4 concentration was 10 g/L and for lipopeptides it was 8 g/L. At these respective NH4NO3 concentrations μmax = 0.58 (h-1), the maximum antifungals (fengycin and iturin) were 285.7 mAU*min and the maximum surfactin concentration was 302 mg/L. The lipopeptides produced by B. amyloliquefaciens, the antifungals (fengycin and iturin) and surfactin, are secondary metabolites, regardless of the optimization treatment, i.e. increased NH4NO3 concentrations. Using 30% enriched air extended the nitrate utilization period, suggesting that when increasing supply concentration, more oxygen was available to act as electron acceptors, allowing nitrate to be used for lipopeptide production. The number of iturin and fengycin homologues generally increased with an increase in nitrate concentration. This suggested that process conditions, such as nitrate concentration, can be used to manipulate homologue ratios, allowing for the possibility to tailor-make biocontrol-agent upstream, during the production process, and possibly increase the efficacy of the biocontrol strategy. The lipopeptides produced by B. amyloliquefaciens showed complete inhibition against Botryotinia fuckeliana and diminished the growth capabilities of Botrytis cinerea. No inhibition was observed against Penicillium digitatum. These results indicate potential of the biocontrol strategy, although scale-up and fed-batch studies are recommended, especially when considering commercial implementation. Studies regarding the lipopeptide application method, i.e. a single application or multiple applications, should also be investigated as this will influence the efficacy of the lipopeptides against the target organisms.
AFRIKKANSE OPSOMMING: Met oorbevolking wat die wêreld se vermoë om die groeiende bevolking te onderhou belemmer, het dit noodsaaklik geword om huidige voedselbronne te beskerm. Daar word beraam dat een derde van die voedsel wat wêreldwyd geproduseer word vir menslike verbruik verlore gaan elke jaar. Verder is insekte, plantpatogene en onkruide verantwoordelik vir meer as ‘n derde van die verliese rakend jaarlikse oeste. Meeste bestaande na-oes beheermetodes maak gebruik van sintetiese stowwe. Ongelukkig kan hierdie verbindings nadelig wees vir die omgewing sowel as menlike gesondheid. Navorsers het hulsef dus toespits daarop om alternatiewe beheermetodes te ondersoek. Bio-beheermetodes is omgewingsvriendelik sowel as bio-afbreekbaar, wat hulle ideale alternatiewe maak vir die sintetiese stowwe. Die lipopeptide, die antifungiese middels (fengycin en iturin) en surfactin, geproduseer deur B. amyloliquefaciens is sekondêre metaboliete, ongeag van die optimerings-behandelinge wat toegepas word, soos ‘n verhoging in NH4NO3 konsentrasie. “Summer slump” is a decline in growth of alfalfa usually beginning in July in areas where maximum daily temperature exceeds 100 °F, such as the low elevation deserts of Southwestern U.S (Fig. 1). In more temperate regions, there is a gradual decrease in alfalfa yield in successive harvests throughout the year, but the yield decline in the summer is not as sharp as in hot summer regions. The term summer slump has also been applied to reduction in growth of perennial cool season grasses such as tall fescue during the summer.

Resumo: Botrytis cinerea Pers, causador da doença conhecida como mofo cinzento, é o principal problema para a conservação pós-colheita de uvas de mesa. A utilização do dióxido de enxofre (SO2) é a prática pós-colheita mais comum para o controle desta doença. Pesquisas buscam alternativas a este produto devido às reações que causa em pessoas alérgicas, danos que pode causar nos frutos e às restrições ao seu uso em sistemas de produção orgânico. Foram avaliados os efeitos da aplicação de uma atmosfera de 40% de CO2 por 24 ou 48 horas (pré-armazenagem) combinado com armazenagem em atmosfera controlada (AC) (12% O2 + 12% CO2) no controle de B. cinerea, e nos atributos de qualidade de uvas 'Flame Seedless' e 'Crimson Seedless'. Também foram avaliados, em uvas 'Crimson Seedless', e os efeitos da associação deste tratamentos com aplicações pré-colheita de cloreto de cálcio (CaCl2) ou dióxido de cloro (ClO2). Os tratamentos foram aplicados em uvas orgânicas infectadas de três formas: infectadas naturalmente, superficialmente inoculadas com conídios e inoculadas com uma baga coberta de micélio. Uvas 'Flame Seedless', naturalmente infectadas, tratadas com 40% de CO2 por 48 horas + AC apresentaram redução da podridão pós-colheita, de 22% para 0,6%, após 4 semanas, e de 100% para 7,4%, após 7 semanas. O pré-armazenamento em 40% de CO2 sozinho também limitou a incidência de mofo cinzento em frutos infectados naturalmente e em uvas inoculadas artificialmente, porém foi menos eficaz do que quando seguido pelo armazenamento em AC. A aplicação de CaCl2 ou ClO2 em pré-colheita reduziu a incidência do mofo cinzento em uvas 'Crimson Seedless' inoculadas com uma solução de conídios, porém não houve controle quando os cachos de uva foram inoculados com micélio. A aplicação de CaCl2 e ClO2 reduziram o mofo cinzento de 45% para 23,2% e 15.6%, respectivamente, em cachos inoculados com conídios e avaliados após 6 semanas armazenadas 0oC. O pré-tratamento com 40% CO2 + CA não afetou as características físico-químicas e sensoriais de uvas 'Crimson Seedless' ou 'Flame Seedless'. Em experimentos in vitro os tratamentos com 40% CO2 por 24 ou 48 h limitaram o crescimento micelial até 72 horas após o tratamento. A germinação dos conídios de B. cinerea foi retardada por 12h. Os resultados mostram que o pré-tratamento com 40% CO2 + CA possui grande potencial para ser adotado como prática comercial para conservação de uvas orgânicas
Abstract: Gray mold, caused by Botrytis cinerea Pers, is the main postharvest decay of table grapes. The use of sulfur dioxide (SO2) is the common post-harvest practice for its control. Several researchers are looking for alternative methods of control, because SO2 can cause allergic reactions, damage fruits and also it cannot be applied in organic production system. In this thesis, it was evaluated the effects of applying an atmosphere of 40% CO2 for 24 or 48 hours (pre-storage) combined with controlled atmosphere storage (CA = 12% O2 + 12% CO2) in the control of B. cinerea, and the effects in the quality and sensory attributes of 'Flame Seedless' and 'Crimson Seedless' table grapes. In addition, it was evaluated the efficacy of CaCl2 or ClO2 application in pre-harvest alone or in combination with pre-storage of 40% CO2 for 24 h + CA, to control gray mold on 'Crimson Seedless' table grapes, and the determination of the impact of these treatments on fruit quality. The treatments were applied in certified organic table grapes naturally infected, surface inoculated and nesting inoculated (inoculated with an infected berry). After 4 weeks of storage, the pre-storage in 40% CO2 for 48 hours + CA reduced postharvest rot from 22% to 0.6%, and after 7 weeks, the decay was reduced from 100% to 7.4% in 'Flame Seedless' naturally infected. The pre-storage in 40% CO2 alone also reduced the incidence of gray mold in fruits naturally infected and in artificially inoculated, but it was less effective than combined treatment. The application of CaCl2 or ClO2 pre-harvest reduced the incidence of gray mold on grapes 'Crimson Seedless' inoculated with a spore solution, but there was no control when fruits were nesting inoculated. After 6 weeks at 0oC, the application of CaCl2, and the ClO2 in fruits surface inoculated, reduced the gray mold from 45% to 23.2% and 15.6%, respectively. The pretreatment with 40% CO2 + CA did not affect quality and nor sensory attributes for both varieties tested. In vitro experiments, the treatment with 40% CO2 for 24 or 48 h limited mycelial growth for at least 72 hours after treatment. Conidial germination of B. cinerea was delayed for 12 hours. Our results showed the potential that pre-treatment with 40% CO2 associated with CA has to be adopted in commercial practice for preservation of organic grapes

A queixada (Tayassu pecari) é um ungulado de grande importância tanto no sentido ecológico pelo seu rol vital na estruturação e composição das comunidades vegetais e para a conservação de ecossistemas bem como no sentido sócio-econômico, por ser fonte de alimento e renda para os caçadores de subsistência na Amazônia Peruana. Não obstante varias pesquisas relataram seu declínio em decorrência de sobre extração e citando a presença de epidemias como um problema potencial. Com o objetivo de avaliar a presença de doenças em populações de queixadas de vida livre e caracterizar os padrões de caça nesta espécie foi desenvolvido o presente estudo durante os anos 2008 e 2009 na Região de Madre de Dios. Foram coletados 103 soros provenientes de animais abatidos para caça de subsistência de duas comunidades indígenas e de animais capturados no interior de três áreas naturais protegidas. Os resultados evidenciaram a presença de anticorpos às doenças de brucelose, leptospirose, toxoplasmose, existindo diferencia na prevalência entre os locais de coleta. Esta corresponde à primeira avaliação sanitária de queixadas na Amazônia Peruana. Como parte complementar foram monitoradas as atividades de caça dos caçadores de subsistência das duas comunidades para caracterizar a área de caça utilizada, a produtividade, a pressão de caça e avaliar se a extração era sustentável. A caça das queixadas aparenta ser sustentável e a população caçada esta sendo adequadamente extraída em termos de sustentabilidade em longo prazo.
The peccary (Tayassu pecari) holds an important place in the Amazonian ecosystem. As seed dispersers they have measurable effects on their habitat, partly guiding structure and composition of the vegetation community. Additionally, their socio-economic role is unsurpassed as one of the most important sources of bushmeat for subsistence hunters in the Peruvian Amazon. Various studies have alarmingly reported the decline of several populations, possibly a result of overextraction due to hunting. However, passing reference was also made to disease and its possible role in population fluctuations. The following study, taking place in Madre de Dios during 2008 and 2009, evaluates the presence of disease in three wild peccary populations. It is the first of its kind in the Peruvian Amazon. A complimentary study characterizing human hunting patterns of peccaries in two indigenous communities is also reported. Exactly 103 serum samples from hunted or live-captured animals were obtained for the study. The results indicate the presence of antibodies to brucelose, leptospirose, and toxoplasmose, in wild populations, the distribution of which is not equal among the three sites. The complimentary study characterizes human hunting patterns of peccaries, focusing on the delimitation of hunting grounds and the quantification of hunting pressure, annual animal productivity, and sustainability of hunting activities. Hunting pressure was found to be well within the limits of sustainability.

INTRODUÇÃO: O Estado de São Paulo é o maior produtor de cana-de-açúcar dentre os demais estados da federação, alcançando uma área de 5.768.184 hectares (ha) cultivada no ano de 2013. Diversos estudos desenvolvidos recentemente no Brasil apontam para os impactos negativos à saúde humana no processo de colheita por meio da queima da cana-de-açúcar. Em 2007, foi assinado o Protocolo de Cooperação (Protocolo do Etanol Verde) entre o setor sucroalcooleiro e o governo estadual visando à antecipação das metas de redução de queima de cana-de-açúcar por meio do processo de mecanização da colheita. Diante deste cenário, faz-se necessária uma análise sobre os impactos à saúde humana neste período de implantação de práticas sustentáveis. OBJETIVO: Este trabalho tem como objetivo avaliar o impacto da produção canavieira, após a assinatura do Protocolo Etanol Verde (2007), em alguns municípios do noroeste paulista por meio da análise das evoluções dos métodos de colheita da cana-de-açúcar possíveis associações com as doenças do aparelho respiratório. METODOLOGIA: . A área de estudo foi definida por meio de técnicas de análise espacial utilizando o software ARCGIS 10.1 com criação de shapefile de incidência de internações por doenças do aparelho respiratório por município e calculado o Índice Local de Moran, possibilitando a identificação dos municípios com maiores incidências e que formam aglomerados espaciais (clusters). Foram consolidados dados secundários de internações por doenças respiratórias em todos os municípios do Estado de São Paulo entre os anos de 2008 e 2012, levando em consideração o mês mais seco do ano (julho) para composição da incidência. Concomitantemente, os aspectos socioeconômicos e físicos relevantes foram caracterizados na área de estudo. Em seguida, foi realizada a análise espacial da distribuição dos métodos de colheita de cana-de-açúcar nos 25 municípios selecionados e foram definidas variáveis de risco associadas à distribuição espacial da colheita e que potencializam o impacto na saúde respiratória da população, levando-se em consideração o decreto 47.700, SMA - SAA 004/08 e outras referências. The study area was defined by spatial analysis using ArcGIS 10.1 software to create shapefile incidence of hospitalizations for respiratory diseases by county and calculated the Local Moran Index, allowing the identification of municipalities with higher incidences and forming spatial clusters (clusters). Secondary data on admissions for respiratory diseases were consolidated in all counties of the State of São Paulo between 2008 and 2012, taking into account the driest month of the year (July) for composition of incidence. Concomitantly, the relevant socioeconomic and physical aspects were characterized in the study area. Resumo: O manejo inadequado é um fatores que afetam a qualidade de produtos agrícolas, conduzindo a perdas devido aos danos mecânicos e o desenvolvimento microbiano, agravado pelo armazenamento inadequado. No caso do figo, devido à limitada proteção oferecida pela epiderme e presença do ostíolo, o beneficiamento utilizando água não é recomendado, trazendo em pauta ações capazes de promover a desinfecção superficial sem afetar a qualidade do produto. Dentre os métodos físicos mais promissores para conservação de frutas estão a refrigeração, radiação ultravioleta (UV-C) e atmosfera modificada. Por outro lado, a utilização de métodos combinados apresenta vantagens devido sua ação sinérgica e complementar. Neste sentido, acredita-se que o uso combinado de métodos físicos pode trazer benefícios para a conservação de figos, principalmente voltada para o mercado externo, auxiliando na manutenção da qualidade do produto e redução da carga microbiana. Este trabalho teve como objetivo avaliar o desempenho da combinação dos tratamentos de atmosfera modificada passiva, atmosfera enriquecida com 100% de CO2 e radiação UV-C na conservação do figo 'Roxo de Valinhos'. As principais etapas foram as seguintes: (1) isolamento dos fungos causadores de podridao em figos 'Roxo de Valinhos' e teste de patogenicidade, utilizando métodos de inoculação de deposição de suspensão de esporos na epiderme do figo, com e sem perfuração, e com deposição sobre papel filtro, avaliando-se a incidência e a severidade dos patógenos com cálculo do Índice de Doença (ID); (2) identificação do nível de tolerância do figo à exposição da radiação UV-C de 0,00 kJ.m-2; 0,70 kJ.m-2; 1,32 kJ.m-2; 2,64 kJ.m-2 e 4,00 kJ.m-2, armazenados à 10°C e 20°C, utilizando como parâmetro de qualidade a aparência visual; (3) avaliação do efeito da radiação UV-C no crescimento in vitro e in vivo dos fungos Rhizopus stolonifer, Lasiodiplodia theobromae e Alternaria alternata, utilizando-se valores de energia de radiação iguais a 0,00 kJ.m-2; 2,64 kJ.m-2; 4,00 kJ.m-2; 8,00 kJ.m-2 e 16,00 kJ.m-2 para os ensaios in vitro e iguais a 0,00 kJ.m-2; 0,70 kJ.m-2; 1,32 kJ.m-2 e 2,64 kJ.m-2 para os ensaios in vivo, a 10ºC e 20ºC; (4) avaliação de desempenho do filme de polipropileno (PP) com espessuras de 25'mi'm, 30'mi'm e 50'mi'm na manutenção da qualidade de figos, avaliada pelos parâmetros perda de massa (PM), sólidos solúveis (SS), acidez titulável (AT), aparência visual e redução da incidência de podridão; (5) avaliação do efeito de atmosfera com 100% de CO2 por 4h, 8h e 12h, à 10ºC e 20ºC, na manutenção da qualidade e diminuição da carga microbiana no figo, avaliada pelos índices PM, SS, AT e aparência visual e (6) combinação do filme de PP de 30'mi'm com radiação UV-C de 1,32 kJ.m-2 e com a exposição à atmosfera com 100% de CO2 durante 4h. Os resultados evidenciaram que a utilização isolada do filme de PP de 30'mi'm, radiação UV-C e exposição à 100% de CO2 mostrou-se eficiente na manutenção da qualidade das frutas, sobretudo quando se utiliza o armazenamento refrigerado. O uso combinado do filme de PP de 30'mi'm com atmosfera modificada com 100% de CO2, representa uma alternativa para o armazenamento do figo, mesmo quando não se utiliza refrigeração, em função da redução da carga microbiana e manutenção da qualidade
Abstract: The main factor affecting quality of agricultural products is the inadequate management, leading to considerable losses due to mechanical damage and fungal contamination, aggravated by improper storage. The fig vulnerability is amplified due to the presence of the ostiole, a peculiar fig orifice, acting as an open door to contamination. Since aqueous sanitization is not recomended, other methods should be searched to promote surface sanitization without affecting the quality of the product. Some promising methods are cooling, ultraviolet radiation (UV-C) and modified atmosphere. A combined method also has advantages because of their synergistic effect on inhibiting microbial growth, and thus maintaining the nutritional and visual qualities of the product. Moreover, the combined use of physical methods, to improve the fig shelf life, potentially can increase business opportunities, specially in the international market. The main objective of this work was to evaluate the performance of the combination of modified atmosphere and UV-C radiation on 'Roxo de Valinhos' fig conservation. To attain this general goal several intermediate steps were performed: (1) fungi rot identification and associated pathogenicity using inoculation by deposition on filter paper to evaluate disease incidence and contamination severity; (2) UV-C radiation fig tolerance identification using energy levels of 0,00 kJ.m-2; 0,70 kJ.m-2; 1,32 kJ.m-2; 2,64 kJ.m-2 and 4,00 kJ.m-2, followed by storage at 10ºC and 20ºC; (3) evaluation of the effect of UV-C radiation on Rhizopus stolonifer, Lasiodiplodia theobromae and Alternaria alternata fungi growth, for in vitro and in vivo tests, using energy irradiation of 0,00 kJ.m-2; 2,64 kJ.m-2; 4,00 kJ.m-2; 8,00 kJ.m-2 and 16,00 kJ.m-2 for the in vitro test, and 0,00 kJ.m-2; 0,70 kJ.m-2; 1,32 kJ.m-2 e 2,64 kJ.m-2 for the in vivo test, at 10ºC and 20ºC; (4) evaluation of the polypropylene (PP) in maintaining the quality, using film thicknesses of 25'mi'm, 30'mi'm and 50'mi'm; (5) evaluation of the fig exposition to 100% CO2 modified atmosphere for 4h, 8h and 12h, followed by storage at 10ºC and 20ºC, in maintaining quality and keeping the microbiological load low and (6) evaluation of the combined use of 30'mi'm PP film with UV-C irradiation of 1,32 kJ.m-2 and 100% CO2 modified atmosphere during 4h. Results have shown that the use of the 30'mi'm PP film, radiation UV-C of 1,32 kJ.m-2 and exposition to 100% of CO2 for 4h was effective in maintaining the quality of the fruits, specially when refrigerated storage is employed. It was concluded that combined use of the 30'mi'm PP film with high CO2 concentration represents an alternative to non refrigerated fig storage, due to the reduction of the microbial load

O objetivo do trabalho foi avaliar produtos alternativos, considerados naturais, como o extrato de própolis verde e óleos essenciais, no controle pós-colheita da antracnose em mangas ‘Palmer’. Foram extraídos dez óleos essenciais de plantas e avaliados, inicialmente, quanto ao potencial antifúngico contra o Colletotrichum gloeosporioides in vitro. Dentre os óleos testados, os de Thymus vulgaris, Cymbopogon citratus e Eucalyptus globulus foram selecionados pelos seus destacados efeitos inibitórios. Na etapa in vitro foram realizados quatro experimentos, em que foram testadas concentrações de extrato de própolis verde e dos óleos, utilizando seis repetições por tratamento. Na etapa in vivo, foram realizados quatro experimentos em que mangas ‘Palmer, foram imersas em cera associada a diferentes concentrações de extrato de própolis verde e dos três óleos essenciais selecionados na etapa anterior. Após a secagem e formação dos revestimentos, os frutos tratados com própolis foram armazenados 12 ºC por 12 dias e mais 3 dias em temperatura de ambiente (22± 2 ºC e 65± 5% UR), e os tratados com óleos essenciais por 14 dias sob refrigeração, e por mais 3 dias a temperatura de ambiente. Os resultados foram submetidos à análise de variância e o efeito dos tratamentos, quando significativo, por meio do Teste F. As médias foram comparadas pelo teste de Tukey (P≤0,05). O própolis verde a partir de 2%, o óleo de capim-limão a partir de 250 ppm e o de tomilho a partir de 10 ppm, se mostraram eficientes em controlar o crescimento do C. gloeosporioides in vitro. O óleo de eucalipto comum retardou, mas não inibiu o crescimento do C. gloeosporioides in vitro. Todos os óleos se mostraram eficientes em inibir a germinação de esporos de C. gloeosporioides. Os tratamentos com cera e cera associada aos óleos ou extrato de própolis reduziram o crescimento da lesão in vivo, sendo que a maior inibição (70%) do patógeno foi conseguida com o extrato de própolis verde na concentração de 2,5%. Todos os produtos testados se mostraram eficientes na manutenção da qualidade de frutos de mangas ‘Palmer’ inoculadas com C. gloeosporioides.
The objective was to evaluate alternative products, considered natural such as green propolis extract and essential oils, in the post-harvest control of anthracnose in 'Palmer' mangos. Ten essential oils from plants were extracted and evaluated, initially, for their potential as antifungal agents against Colletotrichum gloeosporioides in vitro. Among the oils tested, the Thymus vulgaris, Cymbopogon citratus and Eucalyptus globulus were selected for their outstanding inhibitory effects. In the in vitro stage four experiments were conducted, in which green propolis extract and oil concentrations were tested using six replicates per treatment. In the in vivo phase, four experiments were performed in which ‘Palmer’ mangos were dipped in wax associated with different concentrations of green propolis extract and the three essential oils selected in the previous step. After the drying process and formation of coatings, the fruits treated with propolis were stored at 12°C for 12 days and then another 3 days at room temperature (22 ± 2°C and 65 ± 5% RH), and those treated with essential oils were stored for 14 days under refrigeration and for 3 days at room temperature. The results were subjected to variance analysis and a review of the effect of treatments, when significant, by test F. Means were compared by Tukey test (p ≤ 0.05). The green propolis from 2%, lemongrass oil from 250 ppm and thyme from 10 ppm were effective in controlling the growth of C. gloeosporioides in vitro. Common eucalyptus oil was inhibited, but it did not inhibit the growth in vitro of C. gloeosporioides. The procedures with wax and associated wax with oil or propolis extract reduced the growth in vivo of the injury, and the greater inhibition (70%) of the pathogen was achieved with green propolis extract at 2,5% concentration. All products tested have proven effective in maintaining fruit quality of 'Palmer' mangos inoculated with C. gloeosporioides.

Magistrantūros studijų baigiamajame darbe pateikiami ekologiškai auginamų juodojo serbento veislių fenologinių fazių eigos, krūmo augumo, kekių ir derliaus kokybinių rodiklių bei pažeidimo grybinėmis ligomis ir serbentine erkute tyrimo duomenys. Darbo objektas – 10 ekologiškai auginamų juodojo serbento veislių: ‘Titania’, ‘Geo’, ‘Smaliai’, ‘Minaj Šmyriov’, ‘Blizgiai’, ‘Vakariai’, ‘Katiuša’, ‘Gagatai’, ‘Almiai’ ir perspektyvus selekcinis numeris ‘Aldoniai’ (Vakariai x 70-88-9-1). Darbo metodai: Grybinės ligos ir serbentinė erkutė įvertinti maksimalaus pasireiškimo metu. Atskirai apžiūrėjus kiekvieną krūmą, grybinių ligų ir serbentinės erkutės pažeidimai vertinti 6 balų skale (0 – ūgliai, lapai ir pumpurai sveiki; 5 – pažeista virš 75 proc. lapų, ūglių ir pumpurų). Krūmo augumas vertintas prasidedant ir baigiantis vegetacijai. Juodojo serbento kekės ir derliaus kokybė bei produktyvumas nustatytas įvertinus kiekvieno krūmo derlių, uogų nokimo darnumą ir kekių rodiklius. Darbo rezultatai. Veislės ‘Smaliai’ vaiskrūmiai ekologinei sodininkystei tinka dėl vėlyvo žydėjimo, pražysta tik gegužės 10 dieną, tai vertinga veislės savybė, nes vėlai žydinčių serbentų žiedai apsisaugo nuo pavasarinių šalnų pavojaus ir tuo pačiu, išsiskiria stabiliu kasmetiniu derliumi. In the thesis of Master degree studies is presented the study data about the process of phenological phases of the species of ecologically cultivated blackcurrant, bush height, qualitative indicators of bunches and harvest, also the data about affection by diseases and blackcurrant gall mite. Object of the work – 10 species of ecologically cultivated blackcurrant: ‘Titania’, ‘Geo’, ‘Smaliai’, ‘Minaj Smyriov’, ‘Blizgiai’, ‘Vakariai’, ‘Katiusa’, ‘Gagatai’, ‘Almiai’ and perspective selection number ‘Aldoniai’ (Vakariai x 70-88-9-1). Methods of the work: Fungal diseases and blackcurrant gall mite were assessed during maximum manifestation. Affections by fungal diseases and blackcurrant gall mite after inspection of each bush separately were assessed in accordance with the scale of 6 points (0 - shoots, leaves and buds are healthy; 5 – over 75 percent of leaves, shoots and buds are affected). The bush height was assessed at the beginning and the end of vegetation. Assessment of the quality of blackcurrant bunch and harvest was performed through application of ordinary methods. Introdução: A queima da cana de açúcar para a colheita ocorre para facilitar o trabalho de corte, reduzir o volume de resíduos, controlar pragas, evaporar e concentrar o açúcar na haste e eliminar animais perigosos do canavial. A queima da cana de açúcar e seus efeitos à saúde humana são o objeto de diversas pesquisas na área da saúde pública brasileira e mundial. Objetivos: Estudar, por diferentes métodos, a relação entre as queimadas de cana de açúcar e a incidência de doenças respiratórias na mesorregião de Presidente Prudente. Verificar a evolução das queimadas e da produção de cana de açúcar na região, no período de 2008 a 2011. Metodologia: Revisão bibliográfica. Composição de banco de dados geográficos com as variáveis em estudo. Análise dos dados. Elaboração de mapas temáticos e gráficos das variáveis. Cálculo do índice de Moran local das variáveis de estudo. Análise de estatística espacial com os dados de focos de queimadas, hectares de cana de açúcar colhidos com queima prévia e internações. Elaboração de mapas temáticos para comparação e análise. Descrição dos resultados. Resultados: A revisão bibliográfica, entre outras coisas, mostrou diferentes impactos à saúde devido a queima de cana de açúcar. A produção de cana de açúcar aumentou na região de estudo nos últimos anos, consequentemente aumentando a quantidade de queimadas em oposição ao previsto na legislação. Os aglomerados espaciais onde há a colheita com queima prévia coincidiram com os de maior incidência para doenças respiratórias. Identificou se uma área, na porção norte do território estudado de maior vulnerabilidade à poluição atmosférica e, consequentemente, com maior incidência de internações. Conclusões: No período de 2008 a 2011 ocorreu aumento no número de hectares colhidos com queima prévia, associado ao aumento da produção de cana de açúcar. Este efeito não se repetiu nas internações por doenças respiratórias para as faixas etárias selecionadas. Novas pesquisas são necessárias para uma melhor correlação entre o aumento da utilização da queima como método de colheita e as internações respiratórias na região.
Introduction: The burning of sugarcane for harvest occurs to facilitate the cutting work, reduce the volume of waste, control pests, evaporate and concentrate the sugar in the stem and remove dangerous animals from a sugarcane field. The burning of sugar cane and its effects on human health are the subject of several studies in the area of the brazilian and global public health. Objectives: To study, by different methods, the relationship between the burning of sugarcane and the incidence of respiratory illnesses in the middle region of Presidente Prudente. Checking the progress of fires and sugar cane production in the region, from 2008 to 2011. Methods: Literature review. Geographic database composition with the study variables. Data analysis. Preparation of thematic maps and charts of the variables. Calculation of the local Moran index of the study variables. Spatial statistics analysis with data from outbreaks of fires, hectares of sugar cane harvested with previous burning and hospitalizations. Preparation of thematic maps for comparison and analysis. Description of the results. Results: The literature review, among other things, showed different health impacts due to burning of sugar cane. The production of sugar cane increased in the study region in 9 recent years, thus increasing the amount of fires in opposition to the law. The clusters where there is the harvest with previous burning coincided with the highest incidence of respiratory diseases. Identified if an area in the northern portion of the territory studied most vulnerable to air pollution and consequently a higher incidence of hospitalizations. Conclusions: In the period 2008-2011 there was an increase in the number of acres harvested with previous burning, associated with increased production of sugar cane. This effect was not repeated in admissions for respiratory diseases for selected age groups. Further research is needed to better correlation between the increased use of burning as harvesting method and respiratory hospitalizations in the region.

A banana é um fruto consumido no mundo todo, sendo que o Brasil é o quarto maior produtor. A degradação do amido e a síntese de sacarose são responsáveis pelo adoçamento do fruto e, portanto, pela qualidade do mesmo. Os produtores de banana do Vale do Ribeira enfrentam dificuldade de produzir frutos de qualidade, principalmente devido a Sigatoka Negra e a Sigatoka Amarela que são as duas principais doenças da cultura bananeira. Estas duas doenças causam necroses nas folhas diminuindo a área fotossinteticamente ativa da planta podendo diminuir o peso dos cachos, das pencas e até dos frutos. Além disso, as Sigatokas também causam alterações no metabolismo pós-colheita do fruto acelerando seu amadurecimento. No intuito de aprofundar os conhecimentos sobre as alterações causadas pelas Sigatokas nos frutos, estudos foram realizados visando o metabolismo amido-açúcares por se tratar de um processo metabólico importante para a qualidade dos frutos. Para isso, foi conduzido, em 2008, um experimento no Vale do Ribeira aonde três áreas foram marcadas: uma com a presença exclusiva da S. Amarela, outra com predominância da S. Negra e outra área com controle eficiente das doenças. Nestas áreas foi acompanhado o nível de infestação das plantas através do índice de severidade que é uma estimativa visual da doença. Também foram marcadas plantas no momento da antese para que sua idade fisiológica pudesse ser estimada. Pencas foram colhidas em duas idades fisiológicas diferentes: 700 Graus Dia (GD) e 960 GD. Os frutos dessas pencas tiveram suas respiração e produção de etileno medidas diariamente até a senescência. Também foram congeladas amostras ao longo do amadurecimento que, posteriormente foram analisadas quanto aos teores de amido, açúcares solúveis, oligossacarídeos e atividade das enzimas α-amilase e β-amilase utilizando os reagentes Ceralpha e Betamyl (Megazyme) e amido-fosforilases visualizadas em P.A.G.E-nativo. As Sigatokas causaram desordens fisiológicas diferentes na pós-colheita da banana, mas ambas as doenças causaram mudanças nos padrões de respiração e produção de etileno, vida-verde, e no acúmulo e degradação do amido. Exceto pela vida-verde, que foi igualmente afetada por ambas as doenças, os outros fatores foram mais severamente afetados pela Sigatoka Negra, enquanto os frutos oriundos de plantas infectadas pela Sigatoka Amarela foram muito semelhantes aos frutos-controle. Tanto o acúmulo quanto a degradação do amido e enzimas, principalmente a β-amilase e a isoforma plastidial das amido-fosforilases, envolvidas no processo de degradação foram afetados pela Sigatoka Negra.
Banana is one of the most consumed fruits in the world and Brazil is the fourth largest producer of bananas in the world. The degradation of starch and sucrose synthesis are responsible for the sweetening and the quality of fruits. Banana producers in the Vale do Ribeira, southeastern state of Sao Paulo, have difficulty to achieve fruits with high quality, mainly due to Black Leaf Streak Disease and Sigatoka Disease, which are the two main disease problems of banana production. These two diseases cause necrosis in the leaves, reducing the photosynthetic active area of the plant and can also decrease the weight of bunches and fruits. Moreover, both diseases also cause alterations in post-harvest ripening of fruits. In order to better understand the changes caused by these diseases in the fruit ripening, studies were conducted on the starch-to-sugar metabolism, because of its importance for fruit quality. The experiment was conducted in 2008, in the Vale do Ribeira, where we selected three plots: one with the exclusive presence of S. Disease, another one with predominance of Black Leaf Streak disease and another area with an effective disease control. Disease infestation rate were determined by monitoring Severity Index, which is a visual estimate of the disease. To determine the physiological age of fruit, banana plants were marked at the flowering time and the mean temperature were measured daily. The fruits were harvested in two different physiological ages: 700 degree-days (GD) and 960 GD. The samples were also frozen during ripening. In these samples were analyzed the contents of starch, soluble sugars and oligosaccharides; and measured the activities of α-amylase, β-amylase and starch-phosphorylases. Leaf spot diseases caused different physiological disorders in post-harvest banana, but both diseases caused changes in patterns of respiration, ethylene production, green-life, accumulation and degradation of starch. Except for the green-life, which was similarly affected by both diseases, other factors were more severely affected by the Black Leaf Streak disease while the fruits of plants infected by the Sigatoka disease were very similar to control fruit. Both the accumulation and degradation of starch and enzymes, particularly of isoform β-amylase and the plastidial starch phosphorylase, involved in the degradation process were affected by Black Leaf Streak disease.

Some species of sundew (Drosera spp.), harvested from the wild for use in homeopathic cold remedies, are now endangered. This research consisted of two experiments in hydroponic production of sundew. The first investigated the effects of four nitrogen (N) fertilization levels (0.0, 5.1, 25.5 or 51.0 mg/L added N) and three harvest methods (after two months, four months, and sequential harvest at two and four months) on the growth of Drosera adelae (F. Muell.). In addition, the effect of N on the production of a medicinal active ingredient in D. adelae was examined. The second experiment investigated effects of the same nitrogen treatments in combination with three substrates (long fiber sphagnum, peat or rockwool) on growth. The growth rate of D. adelae was greatest at low levels of nitrogen fertilization, and slowed after two months of growth. Plants that were sequentially harvested had the lowest growth rate among all the harvest method treatments. Young plants and plants that received little or no N fertilizer had a greater water concentration than the other plants. Substrate and N level interacted, but generally peat and rockwool produced greater yields than did sphagnum. The medicinal compound plumbagin and a similar but unidentified compound were detected in the leaf extracts of D. adelae. However, whether N fertilization influenced the concentration of these compounds remains inconclusive.

O presente estudo buscou identificar as diversas características da goiaba de polpa branca -\'Kumagai\' e polpa vermelha -\'Pedro Sato\' ensacadas no campo, em relação à qualidade físico-química, à incidência de doenças pós-colheita e ao custo de produção. O experimento foi realizado no município de Valinhos, interior do estado de São Paulo, e as análises laboratoriais foram feitas na Escola Superior de Agricultura \"Luiz de Queiroz\". Com o objetivo avaliar a qualidade físico-química, as goiabas foram ensacadas em dois estádios de desenvolvimento, com 1,5 e 3,0 cm de diâmetro. O ensacamento foi feito com saco de papel branco simples, saco de papel-branco duplo, tecido-não-tecido (TNT), e frutos sem ensacamento, marcados como controle. A metodologia utilizada avaliou o peso do fruto, o diâmetro do fruto, a coloração da casca, a coloração da polpa, a firmeza da polpa, o teor de sólidos solúveis, a acidez titulável e o teor de ácido ascórbico. Após a colheita, os frutos foram mantidos por uma semana em temperatura ambiente controlada (25º C) e avaliados quanto à incidência doenças pós-colheita, de pinta-preta (Guignardia psidii) e antracnose (Colletotrichum spp.); presença ou ausência de larvas de moscas-das-frutas (Anastrepha spp. e Ceratitis capitata) e acúmulo de resíduos químicos aplicados no controle de insetos e de doenças. Por fim, avaliou-se a sustentabilidade econômica da produção de goiabas, pela comparação do custo de produção entre os sistemas de cultivo com e sem ensacamento dos frutos. Para a cultivar Kumagai, o ensacamento com papel duplo proporcionou precocidade na maturação dos frutos e para a cultivar Pedro Sato os ensacamentos com papel simples e com TNT foram os que proporcionam essa precocidade. O ensacamento atuou de forma a evitar a incidência de antracnose para as duas qualidades de goiaba, entretanto, propiciou o aumento na incidência de pinta-preta na pós-colheita de frutos da cultivar Kumagai. O custo total para a produção de goiabas ensacadas foi 8,6% mais elevado do que para a goiaba não ensacada, no entanto, ambas as formas de produção apresentaram-se viáveis economicamente.
This research sought to identify several characteristics of white pulp gravas - \'Kumagai\' and red pulp guavas - \'Pedro Sato\', in relation to physic-chemical quality, incidence of post-harvest diseases and production costs. In order to fulfill the objectives, the experiment was conducted in Valinhos - SP, and the laboratorial analysis was performed at Escola Superior de Agricultura \"Luiz de Queiroz\". To evaluate physic-chemical qualities of two varieties of guava the fruits was bagged at two different development stages, 1,5 and 3,0 cm of fruit diameter. The bagging was realized using a simple white paper bags, double white paper bags and non-textured fabric (TNT). The non-bagged fruits were noted as control. The used methodology evaluated the weight of single fruit, fruit diameter, skin and pulp color, pulp firmness, sugar concentration, acidity, and ascorbic acid levels. In post-harvest guavas, the fruits were kept for one week at controlled temperature (25 ºC) and evaluated for the occurrence of the black spot disease (Guignardia psidii) and anthracnose (Colletotrichum spp.); for the presence or absence of larvae of fruit flies (Anastrepha spp., and Ceratitis capitata); and the abundance of chemical residues, applied to control insects and diseases in guavas. Finally, the economic sustainability of guava production was evaluated, through the comparison of the production costs of both bagging and non-bagging systems. The main results suggests that bagging with double white paper bags induced a shortening in the ripening process of \'Kumagai\' guavas; while in the \'Pedro Sato\' guavas, the shortening ripening process was induced by a simple white paper and TNT bags induced. The bagging process avoided anthracnose incidence in both varieties, although the bagging has resulted in the occurrence of black spot in post-harvest of Kumagai cultivar fruits. The production cost of bagged fruits was 8.6% higher than non-bagged fruit, however, both production systems have shown to be economically sustainable.

O Brasil é considerado o maior produtor de citros e o maior exportador de suco de laranja. Doenças de pós-colheita representam uma grande perda na citricultura, sendo que para muitos frutos a serem exportados, existe uma exigência para que os mesmos estejam isentos de resíduos químicos. Em relação a alguns patógenos de importância em pós-colheita podemos destacar, Guignardia citricarpa (Mancha-pretados- citros), Penicillium digitatum (Bolor-verde) e Colletotrichum gloeosporioides (Antracnose). Dada a importância econômica que representa esse complexo de doenças dos frutos cítricos, tanto em termos de comprometimento da qualidade dos frutos, limitações às exportações e dificuldade de controle, a busca de alternativas adicionais que possam viabilizar a capacidade produtiva dos produtores e garantir a obtenção de frutos com excelentes padrões de qualidade torna-se imprescindível. Nesse contexto, pode-se inserir o emprego de medidas de controle alternativas que não englobam o controle químico clássico. Sob esse ponto de vista inclui-se o controle através do uso de agentes bióticos e abióticos e a indução de resistência em plantas. Portanto neste trabalho, foi estudada a viabilidade do controle de doenças pós-colheita em citros, envolvendo a ação direta sobre os patógenos através do uso dos extratos etanólicos de albedo (mesocarpo) de laranja doce (Citrus sinensis var. Valência) e flavedo (exocarpo ou epicarpo) de limão-Tahiti (Citrus aurantifolia Swing var. Tahiti). O extrato do albedo apresentou efeito antifúngico sobre G. citricarpa e o flavedo do limão “Tahiti” sobre C. gloeosporioides, além de se demonstrar a existência de compostos voláteis com efeito tóxico. A segunda parte envolveu o controle e a indução de resistência em frutos, através do uso dos agentes bióticos Lentinula edodes e Agaricus blazei e do agente abiótico ácido jasmônico. Foi possível observar que o extrato aquoso do albedo (C. sinensis), flavedo (C. aurantifolia), L. edodes e A. blazei diminuíram o aparecimento de novas lesões causadas por G. citricarpa, porém não apresentaram efeitos sobre P. digitatum e C. gloeosporioides em frutos de C. sinensis var. Valência quando tratados em pós-colheita. Dessa maneira, no presente trabalho demonstrou-se a viabilidade de um possível controle alternativo de doenças pós-colheita em citros, buscando-se novos agentes que atuem como indutores de resistência ou de controle direto sobre os fitopatógenos.
Brazil is considered the biggest citrus producer and the biggest orange juice exporter. Post-harvest diseases represent a great loss in the citriculture, and for many fruits to be exported they should be free of chemical residues. In relation to some pathogens of importance in post-harvest it can be mentioned Guignardia citricarpa (black-spot-ofcitrus), Penicillium digitatum (green-mold) and Colletotrichum gloeosporioides (anthracnose). Because of the economical importance that represents this disease complex in citric fruits, in terms of compromising fruit quality, limitations to the exports and control difficulties, the search for alternative control measures that can make possible improve the producing capacity of the producers and the obtaining of fruits with excellent quality are indispensable. Thus, in this context it can be included measures of alternative control that do not include the chemical control. Under this point of view, control include the use of biotic and abiotic agents and the resistance induction in plants. Therefore, it was studied the viability of the control of post-harvest diseases in citrus, involving the direct action on the patogens by using ethanolic extracts of albedo (mesocarp) of sweet orange (Citrus sinensis var. Valência) and flavedo (exocarp or epicarp) of lemon-Tahiti (Citrus aurantifolia Swing var. Tahiti). The results showed that the extract of the albedo exhibited antifungal activity on G. citricarpa and the flavedo of the "Tahiti" lemon on C. gloeosporioides, and it was also demonstrated the existence of volatile compounds toxic to the fungus. The second part involved the control and resistance induction in the fruits, by using the biotic agents Lentinula edodes and Agaricus blazei and the abiotic agent jasmonic acid. It was possible to observe that the aqueous extracts from the albedo (C. sinensis), flavedo (C. aurantifolia), L. edodes and A. blazei reduced the formation of new lesions caused by G. citricarpa, however they did not exhibit effects on P. digitatum and C. gloeosporioides in fruits of C. sinensis var. Valência when treated in post-harvest. Thus, in the present work it was demonstrated the viability of possible alternative control measures of diseases in post-harvest of citrus, indicating the need of looking for new agents to act as resistance inducers or agents to directly control on the phytopathogens.

Pure essential oils have been purified and chemically characterized from cinnamon, clove and black pepper. All these oils have antifungal properties and when applied to apple fruits in post harvest can efficiently control Botrytis cinerea, Alternaria alternata or Penicillium expansum. In addition, application of these compounds or Trichoderma culture filtrate can induce systemic resistance mechanisms in apple fruit. This has been confirmed through phytoalexins and proteomic analysis. The accumulation of the phytoalexins scopoletin, umbelliferone, and scoparone was induced by the three essential oils or Trichoderma CF. Proteomic analysis indicated many changes in protein production caused by the treatment with the oils, Trichoderma CF and the pathogens. The differential spots (TOTD) obtained were up to 166 in clove plus P. expansum treatment. Twenty nine of the most interesting differentially expressed proteins were further analysed by MALDI-TOF MS. The protein groups identified included: pathogenesis-related proteins belonging to the PR-10 sub-family, some antimicrobial proteins, enzymes involved in the biosynthesis of antimicrobial compounds pathway and ribosomal proteins. The most interesting essential oil was the one extracted from black pepper because it was not phytotoxic on tomato seedlings and apple fruits at all the tested concentrations and showed a good disease control activity. A new bio-formulate for post-harvest application, based on a combination of three different active ingredients (black pepper oil, Trichoderma CF and a natural ISR-inducing agent also used as emulsifier) was designed and successfully tested as a synergistic mixture.

The distribution of total phenolics, antioxidant capacity, monomeric anthocyanin, and ascorbic acid contents in the peel and flesh of Red Delicious, Granny Smith, and Fuji apples during a six-month storage period were determined. In addition, the effect of 1-MCP (1-Methyl cyclopropene) on these parameters of the edible portion of apples during storage was also investigated. Two different assays were employed to determine antioxidant activities: the spectrofluorometric-based Oxygen Radical absorbing Capacity (ORAC) and the spectrophotometric-based Ferric Reducing Antioxidant Power (FRAP). Total phenolics were measured using the Folin-Ciocalteau method. A determination of monomeric anthocyanin content was also performed using the pH-differential method for Red Delicious and Fuji apples. Antioxidant activity was predominantly observed in the aqueous fraction and was attributed to polyphenolics. Antioxidant activities were highest in the peel, with Red Delicious peel having the highest values, presumably due to anthocyanin pigments. At zero time month storage, the ORAC, FRAP, total phenolics, and total monomeric anthocyanin contents of Red Delicious peel were: 37.7 μmol T.E./g, 62.7 μmol T.E./g, 6.63 mg/g GAE, and 26.4 mg/100g, respectively. The three tested cultivars were significantly different with respect to ORAC, FRAP, total phenolics, and monomeric anthocyanin contents. 1-MCP treatment did not have a significant influence on antioxidant activities and monomeric anthocyanin contents of the three cultivars during storage. Storage did not have significant influence on ORAC values of the edible portion of apple, however it had significant influence on the FRAP values. The contribution of ascorbic acid to the total antioxidant capacity was small.

Pitaya (Hylocereus spp.) is one of the important emerging fruit crops in Taiwan. In recent years, major post-harvest losses encountered throughout the storage time are mostly due to post-harvest diseases caused by many species of fungal pathogens. Therefore, this study investigated the potential fungal pathogen species on pitaya young fruits in the field which might cause post-harvest disease. The result showed that the number of total potential fungal pathogens isolated from fruit surface increased with time. Colletotrichum, Fusarium, Alternaria, Phomopsis and Botryosphaeriaceae were frequently observed on pitaya young fruits. For disease incidence during storage, anthranose (Colletotrichum spp.) and Fusarium fruit rot (Fusarium spp.) had the highest incidence, followed by black spot (Alternaria spp.) and Phomopsis fruit rot (Phomopsis spp.). However, disease incidences varies between different orchards. Early bagging in the field could reduce the disease severity after harvesting but the result was not significantly different. Accroding to the result, fungal pathogens, Colletotrichum gloeosporioides TC1, Gilbertella persicaria GP1, Alternaria sp. PL3, Fusarium oxysporum F213124 and Phomopsis helianthi F213077, were selected for futher testing. Antifungal effects of three coating materials and twenty one plant essential oils were tested against the five fungal pathogens by the checkboard method in vitro. M-Pede® (potassium salt of fatty acid product) and potassium salts of palm oil had the same antifungal effect, but chitosan was the most effective. At a concentration of 10000 mg·L-1, chitosan inhibitory rates of PL3, F213124 and F213077 mycelial growth were 100%. The results of essential oils antifungal activity showed that 500 μL·L-1 clove leaf oil, cinnamon leaf oil and thyme oil had 100% inhibition rate on spore germination and mycelial growth of all tested isolates. The use of clove leaf oil as a plant protection preparation is most cost-effective. The chemical composition of clove leaf oil analyzed by GC-MS and GC-FID showed that the major component was eugenol (84.65%), and inhibition experiments identified it as the main antifungal ingredient. Moreover, effect of chitosan combined with clove leaf oil on mycelial growth were also determined and their combined effects on all tested isolates were additional. The harvested pitaya fruits were soaked in chitosan or clove leaf oil alone or in combination to control artificially inoculated TC1 or GP1. 1% (10000 mg·L-1) Chitosan combined with 0.05% (500 μL·L-1) clove leaf showed markedly decreased disease incidence and severity, and were similar in effect to the fungicide cyprodinil+fludioxonil (fludioxonil 125 a.i. mg·L-1+ cyprodinil 187.5 a.i. mg·L-1). Furthermore, non-inoculated pitaya fruits soaked in 1% chitosan combined with 0.05% clove leaf were more effective in delaying the occurrence of disease compared to cyprodinil+fludioxonil, and did not affect the weight loss, firmness and total soluble solids, which effectively maintained fruit quality. These results could provide valuable information in formulating fruits protectants for post-harvest pitaya fruit in the future.

Maize is a major calorie source for people living in Sub-Sahara Africa. In this region, Aspergillus flavus causes ear rot diseases in maize, contributing to food insecurity due to aflatoxin contamination. The biological control principle of competitive exclusion has been applied in both the United States and Africa to effectively reduce aflatoxin levels in maize at harvest by introducing atoxigenic strains that out-compete toxigenic strains. The goal of this study was to determine if the efficacy of preharvest biocontrol treatments carry over into the drying period, which is often delayed in Sub-Sahara Africa by the complexities of postharvest drying practices and lack of modern drying machinery. Maize was collected from fields in Texas and North Carolina that were treated with commercial biocontrol, and control fields that were untreated. To simulate moisture conditions similar to those experienced by farmers during drying in Sub-Sahara Africa, we adjusted the grain to 20% moisture content and incubated it at 28 ℃ for 6 days. Although the initial number of infected kernels in most samples were high, less than 24% of kernels were infected with Aspergillus flavus and aflatoxin levels were low (<4ppb). Both toxigenic and atoxigenic strains increased and spread through the grain over the incubation period, and aflatoxin levels increased, even in samples from biocontrol-treated fields. Our molecular analysis suggests that applied biocontrol strains from treated fields migrate to untreated fields. The results also indicate that the population of toxigenic A. flavus in the harvested grain will grow and produce aflatoxin during the drying period when moisture is high. Therefore, any potential postharvest reduction in aflatoxin accumulation will depend on how effective the biocontrol strain was at displacing the toxigenic populations prior to harvest.

The South African Citrus Industry is the second largest exporter of citrus, after Spain. The industry is under pressure to supply high quality fruit as well as to expand into new, high paying markets. However, high paying markets such as Japan and the USA require cold sterilised fruit as obligatory quarantine treatments against Mediterranean fruit fly (Ceratitis capitata) in order to reduce any possible spread of the pest. Citrus fruit originated from tropical climates and hence are chilling susceptible. Chilling injury symptoms appear as dark brown spots, pitting and/or decay when fruit are transferred to shelf temperatures; thus reducing the marketability of citrus fruit. Therefore, there is need for methods to mitigate chilling injury. Previous studies have shown silicon to mitigate many forms of stress without any hazardous effect on human health. Thus, the aim of the study was to investigate the potential of post-harvest silicon dips in mitigating chilling symptoms in citrus fruit. Briefly, fruit from two sources (Ukulinga Research Farm and Ithala Farm) were dipped in different silicon concentrations (0, 50, 150, and 250 mg ℓ-1) for 30 minutes and thereafter stored at -0.5 or 2⁰C for up to 28 days with weekly evaluation for chilling injury symptoms. Total antioxidants were determined using FRAP, ABTS, and DPPH assays under spectrophotometer. In addition, sugars, ascorbic acid, phenolics and flavonoids were analysed using High Performance Liquid Chromatography (HPLC). Fruit from Ukulinga Research Farm showed significantly higher total antioxidants (ascorbic acid total phenolics and specific flavonoids hesperidin and naringin) and sugars relative to fruit from Ithala Farm. Low concentrations of silicon dips significantly reduced the appearance of chilling injury symptoms by inducing an enzymatic conversion of glucose to ascorbic acid, thereby increasing the antioxidant capacity of chilling susceptible fruit. Moreover, silicon increased the concentration of total antioxidants, total phenolics and total flavonoids. High silicon concentrations had a negative effect on post-harvest quality of lemons by increasing fruit weight loss and electrolyte leakage, resulting in appearance of chilling symptoms. In conclusion, the study showed that silicon had potential to reduce chilling injury. However, high silicon concentrations raised concern, in particularly, on fruit appearance.

New emerging markets such as Japan and the United States require cold sterilisation of South African citrus fruit as a phytosanitary standard against fruit fly. However, citrus fruit are chilling susceptible, with lemons being the second-most chilling susceptible after grapefruit. Chilling injury is a physiological rind disorder; the occurrence of which is despite its prevalence in horticultural commodities, not well understood. Therefore, the aim of this study was to investigate physiological compounds regulating chilling susceptibility or resistance in citrus fruit, with special emphasis on lemons. Furthermore, the potential of hot water dips or “molybdenum soaks” to maintain a certain level of physiological compounds which determine manifestation of chilling injury symptoms in citrus fruit was investigated. Moreover, it was attempted to create an understanding of the order in which physiological compounds mitigate chilling injury. Lemon fruit from different farms known to be chilling susceptible or resistant were obtained during the 2007 and 2008 harvest season. Thereafter, fruit were treated by soaking for 30 min in 1μM NaMo04.2H20 solution followed by a 2 min HWD 47 or 53°C. Treated fruit were waxed, weighed and stored at -0.5°C for up to 28 days and sampled for chilling injury evaluation 7, 14, 21, or 28 days into cold storage. A second evaluation was carried out five days after withdrawal from cold storage to allow development of chilling injury symptoms as a shelf-life simulation. After the second evaluation fruit were peeled, peel freeze-dried, milled using mortar and pestle and stored at -21°C for further physiological analysis. Freeze-dried peel was analysed for soluble sugars (glucose, fructose, sucrose), vitamin C (ascorbic acid), vitamin E (α-tocopherol), β-carotene, polyamines (putrescine, spermine, spermidine), specific flavanones (naringin and hesperidin) using HPLC-UV-Vis detector and proline, total antioxidant assays (FRAP, ABTS, DPPH), total phenolics, total flavonoids, lipid peroxidation using spectrophotometry, as well as for the heat shock protein (HSP70) using electrophoresis and silver-staining. Chilling susceptibility of lemon fruit varied with fruit source; those sourced from Ukulinga and Eston Estates were chilling resistant, while fruit from Sun Valley Estates showed chilling injury symptoms after 28 days of cold storage plus five days shelf-life. Furthermore, hot water dips (HW) 53°C, 1 μM Molybdenum (Mo) and 10 μM Mo plus HW 53°C significantly reduced chilling injury symptoms compared with the control and HW 47°C. In addition, Sun Valley Estates fruit also showed higher fruit weight loss compared with non-chilling resistant lemons. The alignment of higher fruit weight loss during storage with chilling susceptibility ascertains the use of weight loss as a non-destructive parameter for chilling susceptibility. With respect to flavedo sugars, glucose was found to be the dominant soluble sugar with multi-functional roles during cold storage. This plays a significant role in mitigating cellular stress. Chilling susceptible lemons from Sun Valley Estates had low flavedo glucose concentrations and, therefore, little conversion of glucose to ascorbic acid was possible resulting in a low antioxidant capacity. However, treatments with HW 53°C and Mo soaks seemed to enhance the enzymatic conversion of glucose to ascorbic acid leading to a higher antioxidant capacity in the flavedo of such treated fruit. Furthermore, glucose also feeds into the pentose phosphate pathway which is coupled with the shikimate pathway synthesizing secondary metabolites, especially of the phenolics group. The decrease in glucose was aligned to the levels of total phenolics, but not to that of β-carotene, naringin and hesperidin through 28 days into cold storage period. Moreover, as glucose also feeds into shikimate pathway, simultaneously an increase in proline flavedo concentration was observed. Proline is an antioxidant synthesized from glutamate; as cellular glucose decreases so does the total antioxidant capacity during cold storage. Ascorbic acid is a dominant and potent antioxidant in lemon flavedo as proven with the FRAP, ABTS and DPPH assays. Chilling resistant fruit have significantly higher ascorbic acid conversion. Furthermore, ascorbic acid also acts to generate the α-tocopheroxy radical to further important membrane-bound antioxidant, vitamin E (α-tocopherol equivalent). Furthermore, the DPPH assay was found to be effective in quantifying total antioxidants in lemon flavedo since it detects both lipophilic and hydrophilic antioxidants compared with the ABTS and FRAP assays which are bias to the estimation of liphophilic or hydrophilic antioxidants, respectively. The hot water and molybdenum treatments increased total antioxidants (DPPH assay) with reduced lipid peroxidation 7 days into cold storage and therefore, reduced chilling symptoms in fruit from Sun Valley Estates. The capacity of antioxidant to scavenge reactive oxygen species (ROS) was increased during cold storage and membrane stability significantly improved. Furthermore, putrescine as low valency polyamine was reduced as such compound acted as precursor to the synthesis of the high valency polyamines, spermine and spermidine. Chilling susceptible lemons from Sun Valley Estates showed increased soluble-conjugated polyamines as a response to stress. Furthermore, HW 53°C, 1 μM Mo and 10 μM Mo plus HW 53°C significantly increased the protein concentration and, therefore, likely also the occurrence of proteins with 70kDa (as estimator of HSP70). Additionally, the concentration of conjugated high valency polyamines was also increased, resulting in reduced chilling injury symptoms. The effect of ROS has only been viewed as damaging, while recently their role has also been viewed as stress acclamatory signalling compounds when produced concentrations below critical damaging threshold. Therefore, hot water dips seems to signals synthesis of total protein which include HSPs which then act throughout cold stress to protect other protein and channel other damaged proteins towards proteolysis. While molybdenum increased ROS production below damaging critical threshold, with ROS signalling stress acclimation by further signalling production of bioactive compound with antioxidant properties.

Phytophthora lateralis is the causal agent of cedar root rot, a fatal forest pathogen whose principal host is Chamaecyparis lawsoniana (Port-Orford-cedar), a predominantly riparian-restricted endemic tree species of ecological, economical, and cultural importance to coastal Oregon and California. Local scale distribution of P. lateralis is thought to be associated with timber harvest and road-building disturbances. However, knowledge of the landscape-scale factors that contribute to successful invasions of P. lateralis is also important for effective land management of Port-Orford-cedar. P. lateralis is able to infest in wet conditions via stream networks (zoospore) and dry conditions via road networks (resting spore). This study tested the hypothesis that vehicles spread P. lateralis by relating its distribution to traffic intensive, anthropogenic disturbances (i.e. a road network, timber harvest) over a 31-yr period in a 3,910-km² portion of the Rogue River-Siskiyou National Forest in the Siskiyou Mountains of Oregon. Indices of road disturbance (presence/absence, configuration, length, density, road-stream network connectivity) and timber harvest (presence/absence, area, density, frequency) were related to locations of infested cedar populations from a USFS survey dataset using a geographic information system (GIS). About 40% of 934 7th-field catchments were infested with the pathogen. Total road length of the study site was 5,070 km; maximum road density was 8.2 km/km2 and averaged 1.6 km/km² in roaded catchments (n = 766). Timber activities extracted 17,370 ha (2,338 cutting units) of forest across 509 catchments; 345 catchments were cut ≥ twice. Maximum harvest density was 0.92 km²/km² ([mean] = 0.04). Both road networks and timber harvest patchworks were significantly related to cedar root rot heterogeneity. Chi-squared contingency tables showed that infestation rates were 2.2 times higher in catchments with roads compared to roadless catchments and 1.4 times higher in catchments with road-stream intersections compared to those that were unconnected. Infestation was twice as likely in catchments with both harvest and road presence than road presence alone. Single-variable logistic regression showed that a one percent increase in harvest density increased infestation odds 25% and a one-unit (km/km²) increase in road density increased infestation odds 80%. Road and stream network configuration was also important to pathogen distribution: 1) uninfested catchments are most likely to be spatially removed from infested, roaded catchments, 2) only 11% of 287 roaded catchments downstream of infested, roaded catchments were uninfested, and 3) only 12% of 319 catchments downstream of infested catchments were uninfested. Road networks and timber harvest patchworks appear to reduce landscape heterogeneity by providing up-catchment and down-catchment access to host populations by linking pathogenic materials to the stream network. Timber harvest data suggest that while infestation risk to Port-Orford-cedar populations remains high, management policies may have curbed infestation risk in timber-harvested catchments; if this is a result of specific P. lateralis mitigation policies adopted in the late 1980's or broader, region-wide conservation policies (i.e. the Northwest Forest Plan) is yet unclear.
