Abstract. The Gravity Recovery and Climate Experiment (GRACE) mission has yielded data on the Earth's gravity field to monitor temporal changes for more than 15 years. The GRACE twin satellites use microwave ranging with micrometre precision to measure the distance variations between two satellites caused by the Earth's global gravitational field. GRACE Follow-on (GRACE-FO) will be the first satellite mission to use inter-satellite laser interferometry in space. The laser ranging instrument (LRI) will provide two additional measurements compared to the GRACE mission: interferometric inter-satellite ranging with nanometre precision and inter-satellite pointing information. We have designed a set of simulated GRACE-FO data, which include LRI measurements, apart from all other GRACE instrument data needed for the Earth's gravity field recovery. The simulated data files are publicly available via https://doi.org/10.22027/AMDC2 and can be used to derive gravity field solutions like from GRACE data. This paper describes the scientific basis and technical approaches used to simulate the GRACE-FO instrument data.

For 15 years, the Gravity Recovery and Climate Experiment (GRACE) mission have monitored total water storage (TWS) changes. The GRACE mission ended in October 2017, and 11 months later, the GRACE Follow-On (GRACE-FO) mission was launched in May 2018. Bridging the gap between both missions is essential to obtain continuous mass changes. To fill the gap, we propose a new approach based on a remove–restore technique combined with an autoregressive (AR) prediction. We first make use of the Global Land Data Assimilation System (GLDAS) hydrological model to remove climatology from GRACE/GRACE-FO data. Since the GLDAS mis-models real TWS changes for many regions around the world, we further use least-squares estimation (LSE) to remove remaining residual trends and annual and semi-annual oscillations. The missing 11 months of TWS values are then predicted forward and backward with an AR model. For the forward approach, we use the GRACE TWS values before the gap; for the backward approach, we use the GRACE-FO TWS values after the gap. The efficiency of forward–backward AR prediction is examined for the artificial gap of 11 months that we create in the GRACE TWS changes for the July 2008 to May 2009 period. We obtain average differences between predicted and observed GRACE values of at maximum 5 cm for 80% of areas, with the extreme values observed for the Amazon, Alaska, and South and Northern Asia. We demonstrate that forward–backward AR prediction is better than the standalone GLDAS hydrological model for more than 75% of continental areas. For the natural gap (July 2017–May 2018), the misclosures in backward–forward prediction estimated between forward- and backward-predicted values are equal to 10 cm. This represents an amount of 10–20% of the total TWS signal for 60% of areas. The regional analysis shows that the presented method is able to capture the occurrence of droughts or floods, but does not reflect their magnitudes. Results indicate that the presented remove–restore technique combined with AR prediction can be utilized to reliably predict TWS changes for regional analysis, but the removed climatology must be properly matched to the selected region.

As a major contributor to global mean sea-level rise, the Greenland ice sheet (GrIS) and the patterns of its mass change have attracted wide attention. Based on Gravity Recovery and Climate Experiment (GRACE)/GRACE Follow-On (GRACE-FO) gravimetry data, we computed monthly non-cumulative mass change time series of the GrIS, which agree with those from the mass budget method confirming the reliability of GRACE-FO-derived mass change. Over the GrIS, mass was mainly gained in winter, followed by spring. It primarily lost mass in summer, with the percentage of summer mass loss versus the corresponding annual mass loss ranging from 61% to 96%. We report that spring mass loss has become more frequent since 2015, and autumn mass gain occurred more frequently after 2014. By separating mass gain from mass loss at the annual timescale, we find that both the mass gain and mass loss showed a slightly increasing trend during 2003–2020, which might be a response to the ongoing Arctic warming. Summer mass variations highly correlated with the summer North Atlantic Oscillation index are dominated by temperature-associated precipitation and meltwater runoff. This study suggests that long-term observations would be necessary to better understand patterns of the GrIS mass variations in future.

Subtle gravitational e ects can be measured by precisely monitoring the position of a test mass. Often this is done by measuring the displacement between two or more test objects. The Gravity Recovery and Climate Experiment (GRACE) satellites do just this, by continuously tracking changes in their separation with micron-level sensitivity. These displacement measurements are used to infer the gravitational potential of the Earth, which has enabled scientists to monitor key aspects of our climate since their launch in 2002. It is planned that the GRACE Follow-On satellites will include a laser ranging instrument as a technology demonstrator to improve the displacement measurement. Before science operation commences and measurements can begin, the laser on each satellite needs to be precisely pointed towards the opposite satellite, and thus the satellites must undergo an initial acquisition scan after launch to establish the laser link. This thesis is concerned with developing technology for the GRACE Follow-On laser ranging instrument and exploring interferometric techniques for future satellite missions. In the following chapters, we experimentally demonstrate an acquisition system with GRACE Follow-On-like parameters, requiring no additional hardware but relying on the photodetectors and signal processing equipment already required for science operation. This strategy was developed with multiple collaborators over several years led by C. Mahrdt at the Albert Einstein Institute. To establish the laser link, ve degrees of freedom must be optimized (pitch and yaw for each beam, and the frequency di erence between the two lasers). Laser steering and frequency scanning patterns are combined with a fast Fourier transform-based peak detection algorithm run on each satellite to nd the signal. We successfully demonstrate both stages (commissioning and reacquisition) of the proposed acquisition strategy. One of the core components needed for the GRACE Follow-On laser ranging instrument is the triple mirror assembly (TMA), a modi ed corner cube that symmetrically routes the laser beam around existing hardware about the satellite's center of mass. A prototype triple mirror assembly was designed and constructed by local and international collaborators, and we present optical tests demonstrating three of the performance requirements of the prototype. The path length stability of a beam traveling through the TMA was measured in a test bed resembling the measurement con guration of the GRACE Follow-On interferometer. The parallelism between the incoming and outgoing beams to/from the TMA is measured to the arc second level. Additional measurements quantify changes in the parallelism as the TMA prototype is heated and cooled. Finally, we give a brief overview of digitally-enhanced interferometry, a developing technique for optical metrology which has signi cant advantages over a conventional heterodyne system and could be employed for future space missions. We present an experimental demonstration of the multiplexing capability of the technique, showing an improved displacement sensitivity between measurement points when information from several sensors is combined to suppress errors due to laser frequency noise. We discuss an option for the technique to be applied to future inter-satellite measurement architectures and examine possible simpli cations to the optical bench layout.

From the angle of emergent literacy perspective, the researcher conducted a case study to investigate lower-grade pupils’ development of Mandarin reading and writing. The two subjects, with abundant experience of reading, drawing, writing, and speaking, graduated from kindergarten which adopted learning center approach and had not received formal instruction not only of reading and writing but also of Mandarin Phonetic Symbols until they entered elementary school. The purpose of the research was to understand the subjects’ characteristics and progress in reading and writing in elementary school and to explore the transitional process between preschool literacy experiences and literacy learning in primary education. The methods included participant observation methods, field notes, video- and audio-recordings, interviews, collections of the pupils’ works and relevant documents as well as accomplishments in Mandarin courses. Fields of research contained the situations wherein cases participated in Mandarin courses and after-class reading and writing activities. Research findings revealed the following information: 1)The progress of learning reading Although those who had learned Mandarin Phonetic Symbols were less frustrated after enrolment, they still made commonly seen mistakes. Some of the rules could be absorbed through direct teaching and drills. While the two subjects met with setbacks at the beginning of learning Mandarin Phonetic Symbols, they ultimately can comprehend and employ the rules of pin-yin system. One of the subjects, Duo-Duo, learned Mandarin Phonetic Symbols with a certain amount of lexical acquisition which helped her reduce anxiety about learning Mandarin Phonetic Symbols and reading; meanwhile, it also accelerated reading performance. In the aspect of reading strategies, the two subjects advanced from reading aloud to silent reading, and they could interchangeably apply the two strategies by utilizing fingers to assist in pointing, read word by word as well as correlate symbols with sounds. Reading comprehension was concerned with the subjects’ decoding speed, the amount of lexical acquisition, and real-life experiences. 2)The progress of learning writing In the initial stage, the case who wrote Mandarin Phonetic Symbols would verbally write out based on the property of symbols. From a viewpoint of emergent literacy perspective, the phenomenon would not influence language competency and meaning conveyed. After being provided with formal instructions of characters, the subjects gradually replaced Mandarin Phonetic Symbols with characters. As for written language, the patterns that two subjects mostly relied on were sounds and shapes in the task of word recognition whereas least were semantic meanings. Oral expression served as their important language experience related to background knowledge of organizing words and sentences. However, higher-level organizational abilities needed to be acquired by participating in more literacy activities. Furthermore, writing and reading were reciprocally associated with each other, and plentiful literacy experience would reflect in oral and writing competency. 3)Contextual factors that influenced learning of reading and writing Literacy development would vary with individual differences. Attitudes and styles were cultivated through interactions with surroundings which further affected the subjects’ literacy learning. Moreover, family played a role in supporting their initial and life-long literacy experiences; preschool education provided learning and support suitable for their differences and age; primary education leaded the subjects to learn formal language forms. The connection of family and school education composed supportive system of literacy development. Lastly, enactment of language education policies and emphasis on literacy achievement in our cultural context also worked upon language experience of the subjects and all pupils. These above-mentioned systems interrelated to each other and influenced the subjects’ literacy development to different extents. The findings showed that language education of preschool should supply the children with rich literacy context which included drawing, listening, storytelling, oral expression, and so on. In this way, even if traditional literacy skills were not specifically taught, these children still could have concept and abilities of reading and writing, by which would benefit their literacy development. The more important thing was that under the cultural context in Taiwan emphasizing formal language instructions, the two subjects developed interest in daily literacy practice during preschool stage and extended such an interest, which become parts of their life, to primary education. These cases learned reading and writing on the basis of their own specific strengths, and their disinterest of learning Mandarin Phonetic Symbols, recognizing words and writing did not affect language development. In their sophomore, one of the subjects, Jiang-Jiang, gradually revealed higher-level language ability mastering tips of reading and writing whose performance was related to personal attributes and experience at home and school.

This study aims to explore the design strategies, development processes, possible obstacles and solutions to a story follow-up writing. By understanding the changes of students before and after a story follow-up writing, this study would provide a learning process to researcher. The action research approach was adopted and 28 eight-grade students were participated for this study. The subject was follow-up writing with four units of worksheet. The quantitative data were collected and analyzed through students’ writing attitude scale. Meanwhile, the qualitative data were collected and analyzed through interviews with the participants, interviews with the collaborative researchers, teacher reflection notes and students’ feedbacks. The analyses of these data revealed the results as below: 1. By using kinds of stories would correspondent with students who have each reading hobbies and would improve students’ writing motivation. 2. “Telling stories, follow-up writing and comparing” is a very helpful course that could improve students’ writing attitude. 3. The researcher gained the teaching growth and enhanced the professional capacity from the action research. The researcher makes the following recommendations for teachers based on the findings: 1. Choose a story, which is closed to life. 2. Teacher could tell the story orally, which would make students interested in writing. 3. Increase the class numbers of follow-up writing course. 4. Focus on the skill shift after course. 5. The positive feedback from teacher help students improve the writing attitude 6. Provide students an opportunity to assess their writing to each other. 7. Seek research partners and organize a teaching team to design more perfect course. The suggestions for future researchers are listed as follows: 1. Increase the difficulty level of follow-up writing. 2. Explore the changes of writing skill of students on follow-up writing.

The study was to learn how an After-school Caring Service Teacher program for grade 2 students in an elementary school through the design and implementation of a multiplied teaching program, to enrich the service contents of the after-school caring service. The research subjects of the study were the grade 2 students of an elementary school in Taipei City, who were joining an after-school caring service program of that school. The research method adopted in the study was action research. The researcher with her partners and the supervising administrator, from the three pre-set reference courses of“Picture Books of Character”,“the Joyful Physical Fitness”and“the Creative Aesthetic Labor”, formed a series of actions including the design, implementation, assessment, review and recommendations, with the hopes that the students, in addition to the making of the homework and subject guidance, would explore their extra-curriculum activities to promote the growth of character and to raise the interest of learning and the enlightenment of creativeness. We also hope that with this study, we can further understand the problems the teachers facing and their solutions, and we might gradually constitute a usable and effective after-school caring service program through reflection and correction. We concluded our data-analysis as follows: 1.The diversified after-school caring service program would help to raise the percentage of homework-completion. 2.A proper time-frame for the after-school caring service would take both the homework-making and diverse learning into consideration. 3.The growth of the character education could be seen through the teaching with picture books presenting the promotions and growths of seven item about characters. 4.The character education with the adoption of joyful physical fitness teaching is effective in cultivating the respect, fairness and teamwork spirit of the students. 5.The teaching of creative aesthetic labor had greatly helped in raising the students’ interest toward learning and enlightening the students’ creativity. 6.Due to the heavy tasks for teachers in after-school caring service, teachers would only be able to realize targets of their program-designing in the form of group. Based on the above-mentioned conclusion, the researcher would like to propose concrete suggestions here as the reference for teachers, administrators and future researchers.

Like other church fathers, Hilary sometimes articulates the will as entirely competent; sin is entirely our fault because our will is strong enough to prevent it. However, the ‘anthropological household’ demonstrates another strain in his thought, where the will is affected by the Fall and is now subject to disobedience (infidelitas). Similarly, he sees that humans find it difficult to will to follow God, but suggests that God gives grace for this as long as the human shows the initiative (initium fidei). This is then compared to other early Christian writers. Although they do admit other constraints on human action, Hilary appears to be the first Latin author expressly using the fallen will. He appears to follow Origen, who does articulate constraint in terms of the will. Hilary’s expressions may thus arise under the influence of Stoic thought, which reached him through Origen’s material.

Although seldom as overt as male–male competition, female–female competition is equally serious in terms of lifetime impacts and may occasionally become violent. Here I follow a female lifetime, exploring the kinds, intensity, and impact of female–female competition.

A year later, the band's self-titled debut album burst onto the Billboard charts, capturing the imagination of fans and music critics worldwide. The fact that the group had formed in the sleepy southern college town of Athens, Georgia, only increased the fascination. Soon, more Athens bands followed the B-52's into the vanguard of the new American music that would come to be known as "alternative," including R.E.M., who catapulted over the course of the 1980s to the top of the musical mainstream. As acts like the B-52's, R.E.M., and Pylon drew the eyes of New York tastemakers southward, they discovered in Athens an unexpected mecca of music, experimental art, DIY spirit, and progressive politics--a creative underground as vibrant as any to be found in the country's major cities. In Athens in the eighties, if you were young and willing to live without much money, anything seemed possible. Cool Town reveals the passion, vitality, and enduring significance of a bohemian scene that became a model for others to follow. Grace Elizabeth Hale experienced the Athens scene as a student, small-business owner, and band member. Blending personal recollection with a historian's eye, she reconstructs the networks of bands, artists, and friends that drew on the things at hand to make a new art of the possible, transforming American culture along the way. Management of prostate cancer remains controversial, in large part because of its wide heterogeneity in terms of aggressiveness and prognosis. Early detection efforts based on prostate specific antigen (PSA) and aggressive treatment of high-risk cancers have yielded major improvements in mortality rates, but overtreatment of low-risk cancers—those unlikely to cause symptoms or threaten life if they were never detected—is associated with high rates of avoidable toxicity and cost. Prostate cancer can be effectively risk-stratified based on tools (e.g. nomograms, CAPRA score) integrating the PSA level, Gleason grade, clinical stage, and extent of biopsy tissue involvement. Most men with low-risk tumours are eligible for active surveillance, a programme of careful monitoring based on PSA and follow-up biopsies. Men with higher-risk cancers are best served with radical prostatectomy or radiation therapy.

Many studies have supported the idea that human interaction relies on cooperation and joint action, implying that everyday communication has primarily a social motivation. According to Grice, interlocutors are expected to meet the informational needs of their interactional partner(s) in both accuracy and informativeness. However, conversional principles incompatible with Grice’s maxims have been found to be implicitly applied in traditional societies from Madagascar, the Pacific Islands, and Mesoamerica. This chapter considers the management of epistemicity among the Yucatec Mayas of Mexico, focusing on the function and use of evidential particles and the broader cultural context into which they fit. Another is to present the implicit rules that adults and children follow to evaluate and endorse claims of knowledge. Such rules might explain why Yucatec Mayas tend to be linguistically accurate in stating and evaluating knowledge sources. Because they expect others to lie or withhold information, they constantly monitor how their and others’ assertions and information are shared.

This chapter gives a constructive account of saving encounter with divine grace, through the Spirit, in the context of embodied experience. It draws on the insights of affect theory and the “material turn” in religious studies to argue for the ongoing experiential plausibility of the doctrine of sin in the contemporary world. The chapter demonstrates that theologians in recent decades have tended to follow Lindbeck and Stendahl, amongst others, in making assumptions about the plasticity of human experience through the instruments of language and discursive practice, and that these assumptions require substantial qualification. The second half of the chapter builds on these insights, together with the theology of Martin Luther, to describe Christian experience of grace in terms of an affective pedagogy effected by the Spirit through the instruments of the law and the gospel. It concludes by showing how this account of grace can make religious sense of a wide variety of experiences of affective plight in the contemporary world.

AbstractThe recovery of Earth’s time variable gravity field from satellite data relied heavily on Satellite Laser Ranging (SLR) before the recent GRACE and GRACE Follow-On satellite gravity missions. Currently, the monthly gravity solutions from GRACE/GRACE-FO provide important global information about the temporal variations of gravity field. However, there are a few low-degree coefficients derived from GRACE/GRACE-FO that are not well determined, because of the satellite gravity mission configuration and issues with the accelerometer data. These low-degree coefficients can be determined reasonably well using SLR data from the dedicated SLR satellite configuration and can be used to replace the less well-determined values from GRACE/GRACE-FO. A more rigorous and consistent approach is to directly combine SLR and GRACE/GRACE-FO gravity solutions in a simultaneous solution. This paper presents a combination strategy for gravity field recovery from combined SLR and GRACE/GRACE-FO mission data. To correctly account for all correlations, the combination is performed at the information (normal) equation level. The coefficients C20 and C30 are determined mainly from SLR by renaming C20 and C30 parameters in GRACE/GRACE-FO information equations. The results show that the combined products are improved in comparison with the nominal GRACE/GRACE-FO gravity solutions. The gravity field products are evaluated by comparing different gravity solutions through coefficient-wise comparison, equivalent water height variations and mass changes over selected areas.

AbstractSatellite gravity missions, like GRACE and GRACE Follow-On, successfully map the Earth’s gravity field and its change over time. With the addition of the laser ranging interferometer (LRI) to GRACE-FO, a significant improvement over GRACE for inter-satellite ranging was achieved. One of the limiting factors is the accelerometer for measuring the non-gravitational forces acting on the satellite. The classical electrostatic accelerometers are affected by a drift at low frequencies. This drawback can be counterbalanced by adding an accelerometer based on cold atom interferometry (CAI) due to its high long-term stability. The CAI concept has already been successfully demonstrated in ground experiments and is expected to show an even higher sensitivity in space.In order to investigate the potential of the CAI concept for future satellite gravity missions, a closed-loop simulation is performed in the context of GRACE-FO like missions. The sensitivity of the CAI accelerometer is estimated based on state-of-the-art ground sensors and predictions for space applications. The sensor performance is tested for different scenarios and the benefits to the gravity field solutions are quantitatively evaluated. It is shown that a classical accelerometer aided by CAI technology improves the results of the gravity field recovery especially in reducing the striping effects. The non-gravitational accelerations are modelled using a detailed surface model of a GRACE-like satellite body. This is required for a realistic determination of the variations of the non-gravitational accelerations during one interferometer cycle. It is demonstrated that the estimated error due to this variation is significant. We consider different orbit altitudes and also analyze the effect of drag compensation.

Why does God’s Spirit grant the gift of faith to some but not to others? Erasmus solved the dilemma by admitting minimal freedom to the human side, a wrong use of free choice is to be blamed. This is the way of maintaining credibility in the justice of God. Luther’s solution to the dilemma was a distinction between the concepts of “the revealed God,” Deus revelatus, and of “the hidden God,” Deus absconditus. On the notitia level, i.e. in regard to knowing who is elected, we are in total darkness; it is a secret of the hidden majesty. We are restricted to the usus level of election, i.e. to revelation which says that God wills everyone to be saved. God’s will does not follow any human logic of justice, God’s will itself is the norm for itself and cannot be subjected to any rule outside itself. Asking why God does what he does is a concern arising from religious pride; the sovereignty of the divine will utterly annihilates speculation about any grounds for bargaining with God. Luther follows the paradigm of “the theology of the cross”: Anyone who has become “desperate about him/herself” is, paradoxically, already in the state of grace. This paradox brings about certainty of salvation: God has taken the question of salvation completely “outside ourselves” into his hands, this results in peace in the scruples of salvation, the believer is liberated from “the pestilence of uncertainty.” Luther prefers the Biblical term “election” to the philosophical concept “predestination.”

This chapter summarizes the themes of the book as a whole by returning to a primary issue raised at the start of the volume, namely, how to understand the roots of human evil. It follows a broadly Thomistic understanding of evil as lacking in agency, mirrored in Sergii Bulgakov’s suggestive terminology of shadow sophia. The risks apparent in Neoplatonism within Bulgakov are countered through a focus on practical wisdom and its counterfeit in distorted forms, alongside the biblical imagery of folly. The different vices discussed throughout the work are revisited again in their relationship to each other in order to develop a broader sense of the subtlety and energy of evil as it arises in individuals and communities. Some tentative conclusions are reached about what Ricoeur terms ‘the fault’, which cannot be reduced to purely scientific explanations, as well as his provocation to lament. At the same time, scientific engagement enlarges the understanding of virtues and the accompanying vices. The afterword opens up the discussion to the volume which will follow on the work of grace in the moral life.

Chapter four, “Unwilling Feeling,” reads John D. Caputo’s material theology and his conception of the insistence of God alongside Sara Ahmed’s work on what she names “affect aliens” and willfulness to offer biblical scenes of affect alien prophets. Jonah and Martha embody such moody prophecy in this scene. The chapter constructs and applies an affect hermeneutic to and with biblical texts in order to read for what might happen when we follow moodiness to unexpected theological conclusions. Jonah’s and Martha’s moodiness in their biblical tales reveal not what’s wrong with them, but rather serve as a lament against problematic theological interpretations and conscriptions of each character. These affect alien prophets exist as blockage; their existence stops up or slows down the normative flow. These biblical characters prophetically persist by remaining moody impediments to the story. To gravely attend to such prophets is to embrace alternate flows or undercurrents within the biblical story. Such an embrace invites us to look for alternate flows within our contemporary stories.

Introduction: Uterine sarcoma accounts for nearly 3% of all uterine malignancies. They have 4 major pathology includes endometrial stromal sarcoma high grade, ESS low grade, uterine leiomyosarcoma (uLMS) and undifferentiated uterine sarcoma (UUS). Recent WHO classification 2014, recognizes low grade ESS and high grade ESS as distinct entity. They differ from endometrial carcinoma in their aggressive nature and poor prognosis. We review our database and found total 44 eligible patient treated at our institute. Materials and Methods: Its retrospective analysis of computer based database of our institute from January 2009 to December 2015. We analyzed demographic, pathological, treatment and survival data. Results: Total 44 patient treated for uterine sarcoma at our institute. Among these 16 were operated at our institute during study period. Here we reporting results of operated patients at our institute. The histological diagnosis LMS (5/16), ESS-L (4/16), MMMT (3/16), UUS (3/16) and ESS-H (1/16). Stage distribution was stage I, (6/16) stage II, (5/16) stage III, (3/16) stage IV, (0/16) and unknown stage (2/16). Two patients underwent completion surgery for outside myomectomy. The adjuvant treatment was CT in 3/16, CT with RT in 7/16, HT in 4/16 and one lost to follow up with one was put on observation. Median follow up is 30 month with 14 patients alive and one lost to follow up. At last follow up 4 patients alive with metastatic disease and 10 patients alive with no evidence of disease. Seedless cultivars dominate the table grape industry. In these cultivars it is mandatory to apply gibberellin (GA) to stimulate berry development to a commercially acceptable size. These cultivars differ in their sensitivity to GA application, and it frequently results in adverse effects such as decreased bud fertility and increased fruit drop. Our long term goals are to (1) understand the molecular basis for the differential sensitivity and identify markers for selection of sensitive cultivars (2) to develop new strategies for targeted manipulation of the grape berry response to GA that will eliminate the need in GA application and the undesirable effects of GA on the vine, while maintaining its desirable effects on the berry. Both strategies are expected to reduce production cost and meet growing consumer demand for reduced use of chemicals. This approach relies on a comprehensive characterization of the central components in the GA signaling cascade in the berry. Several key components in the GA signaling pathway were identified in Arabidopsis and rice, including the GA receptors, GID1s, and a family of DELLA proteins that are the major negative regulators of the GA response. GA activates its response pathway by binding to GID1s, which then target DELLAs for degradation via interaction with SLY, a DELLA specific F-box protein. In grape, only one DELLA gene was characterized prior to this study, which plays a major role in inhibiting GA-promoted stem growth and GA-repressed floral induction but it does not regulate fruit growth. Therefore, we speculated that other DELLA family member(s) may control GA responses in berry, and their identification and manipulation may result in GA-independent berry growth. In the current study we isolated two additional VvDELLA family members, two VvGID1 genes and two VvSLY genes. Arabidopsis anti-AtRGA polyclonal antibodies recognized all three purified VvDELLA proteins, but its interaction with VvDELLA3 was weaker. Overexpression of the VvDELLAs, the VvGID1s, and the VvSLYs in the Arabidopsis mutants ga1-3/rga-24, gid1a-2/1c-2 and sly1-10, respectively, rescued the various mutant phenotypes. In vitro GAdependent physical interaction was shown between the VvDELLAs and the VvGID1s, and GAindependent interaction was shown between the VvDELLAs and VvSLYs. Interestingly, VvDELLA3 did not interact with VvGID1b. Together, the results indicate that the identified grape homologs serve as functional DELLA repressors, receptors and DELLA-interacting F-box proteins. Expression analyses revealed that (1) VvDELLA2 was expressed in all the analyzed tissues and was the most abundant (2) VvDELLA1 was low expressed in berries, confirming former study (3) Except in carpels and very young berries, VvDELLA3 levels were the lowest in most tissues. (4) Expression of both VvGID1s was detected in all the grape tissues, but VvGID1b transcript levels were significantly higher than VvGID1a. (5) In general, both VvDELLAs and VvGID1s transcripts levels increased as tissues aged. Unfertilized and recently fertilized carpels did not follow this trend, suggesting different regulatory mechanism of GA signaling in these stages. Characterization of the response to GA of various organs in three seedless cultivars revealed differential response of the berries and rachis. Interestingly, VvDELLA3 transcript levels in the GA-unresponsive berries of cv. Spring blush were significantly higher compared to their levels in the highly responsive berries of cv. Black finger. Assuming that VvDELLA2 and VvDELLA3 are regulating berry size, constructs carrying potential dominant mutations in each gene were created. Furthermore, constitutive silencing of these genes by mIR is underway, to reveal the effect of each gene on the berry phenotype.

The objectives of this project were to develop nondestructive methods for detection of internal properties and firmness of fruits and vegetables. One method was based on a soft piezoelectric film transducer developed in the Technion, for analysis of fruit response to low-energy excitation. The second method was a dot-matrix piezoelectric transducer of North Carolina State University, developed for contact-pressure analysis of fruit during impact. Two research teams, one in Israel and the other in North Carolina, coordinated their research effort according to the specific objectives of the project, to develop and apply the two complementary methods for quality control of agricultural commodities. In Israel: An improved firmness testing system was developed and tested with tropical fruits. The new system included an instrumented fruit-bed of three flexible piezoelectric sensors and miniature electromagnetic hammers, which served as fruit support and low-energy excitation device, respectively. Resonant frequencies were detected for determination of firmness index. Two new acoustic parameters were developed for evaluation of fruit firmness and maturity: a dumping-ratio and a centeroid of the frequency response. Experiments were performed with avocado and mango fruits. The internal damping ratio, which may indicate fruit ripeness, increased monotonically with time, while resonant frequencies and firmness indices decreased with time. Fruit samples were tested daily by destructive penetration test. A fairy high correlation was found in tropical fruits between the penetration force and the new acoustic parameters; a lower correlation was found between this parameter and the conventional firmness index. Improved table-top firmness testing units, Firmalon, with data-logging system and on-line data analysis capacity have been built. The new device was used for the full-scale experiments in the next two years, ahead of the original program and BARD timetable. Close cooperation was initiated with local industry for development of both off-line and on-line sorting and quality control of more agricultural commodities. Firmalon units were produced and operated in major packaging houses in Israel, Belgium and Washington State, on mango and avocado, apples, pears, tomatoes, melons and some other fruits, to gain field experience with the new method. The accumulated experimental data from all these activities is still analyzed, to improve firmness sorting criteria and shelf-life predicting curves for the different fruits. The test program in commercial CA storage facilities in Washington State included seven apple varieties: Fuji, Braeburn, Gala, Granny Smith, Jonagold, Red Delicious, Golden Delicious, and D'Anjou pear variety. FI master-curves could be developed for the Braeburn, Gala, Granny Smith and Jonagold apples. These fruits showed a steady ripening process during the test period. Yet, more work should be conducted to reduce scattering of the data and to determine the confidence limits of the method. Nearly constant FI in Red Delicious and the fluctuations of FI in the Fuji apples should be re-examined. Three sets of experiment were performed with Flandria tomatoes. Despite the complex structure of the tomatoes, the acoustic method could be used for firmness evaluation and to follow the ripening evolution with time. Close agreement was achieved between the auction expert evaluation and that of the nondestructive acoustic test, where firmness index of 4.0 and more indicated grade-A tomatoes. More work is performed to refine the sorting algorithm and to develop a general ripening scale for automatic grading of tomatoes for the fresh fruit market. Galia melons were tested in Israel, in simulated export conditions. It was concluded that the Firmalon is capable of detecting the ripening of melons nondestructively, and sorted out the defective fruits from the export shipment. The cooperation with local industry resulted in development of automatic on-line prototype of the acoustic sensor, that may be incorporated with the export quality control system for melons. More interesting is the development of the remote firmness sensing method for sealed CA cool-rooms, where most of the full-year fruit yield in stored for off-season consumption. Hundreds of ripening monitor systems have been installed in major fruit storage facilities, and being evaluated now by the consumers. If successful, the new method may cause a major change in long-term fruit storage technology. More uses of the acoustic test method have been considered, for monitoring fruit maturity and harvest time, testing fruit samples or each individual fruit when entering the storage facilities, packaging house and auction, and in the supermarket. This approach may result in a full line of equipment for nondestructive quality control of fruits and vegetables, from the orchard or the greenhouse, through the entire sorting, grading and storage process, up to the consumer table. The developed technology offers a tool to determine the maturity of the fruits nondestructively by monitoring their acoustic response to mechanical impulse on the tree. A special device was built and preliminary tested in mango fruit. More development is needed to develop a portable, hand operated sensing method for this purpose. In North Carolina: Analysis method based on an Auto-Regressive (AR) model was developed for detecting the first resonance of fruit from their response to mechanical impulse. The algorithm included a routine that detects the first resonant frequency from as many sensors as possible. Experiments on Red Delicious apples were performed and their firmness was determined. The AR method allowed the detection of the first resonance. The method could be fast enough to be utilized in a real time sorting machine. Yet, further study is needed to look for improvement of the search algorithm of the methods. An impact contact-pressure measurement system and Neural Network (NN) identification method were developed to investigate the relationships between surface pressure distributions on selected fruits and their respective internal textural qualities. A piezoelectric dot-matrix pressure transducer was developed for the purpose of acquiring time-sampled pressure profiles during impact. The acquired data was transferred into a personal computer and accurate visualization of animated data were presented. Preliminary test with 10 apples has been performed. Measurement were made by the contact-pressure transducer in two different positions. Complementary measurements were made on the same apples by using the Firmalon and Magness Taylor (MT) testers. Three-layer neural network was designed. 2/3 of the contact-pressure data were used as training input data and corresponding MT data as training target data. The remaining data were used as NN checking data. Six samples randomly chosen from the ten measured samples and their corresponding Firmalon values were used as the NN training and target data, respectively. The remaining four samples' data were input to the NN. The NN results consistent with the Firmness Tester values. So, if more training data would be obtained, the output should be more accurate. In addition, the Firmness Tester values do not consistent with MT firmness tester values. The NN method developed in this study appears to be a useful tool to emulate the MT Firmness test results without destroying the apple samples. To get more accurate estimation of MT firmness a much larger training data set is required. When the larger sensitive area of the pressure sensor being developed in this project becomes available, the entire contact 'shape' will provide additional information and the neural network results would be more accurate.
