The Glyde–1 and Glyde Sidetrack–1 wells were drilled by Armour Energy in the Glyde Sub-basin of the McArthur Basin, NT, Australia in August 2012. This program was to evaluate the unconventional hydrocarbon potential of the Barney Creek Shale source rock and the conventional potential of the Coxco Dolomite of the McArthur Group. The Glyde wells discovered gas in both formations. Transtensional faults in this region allowed to form a series of fault-bounded depocentres. The target gas source of the Glyde discovery is located in 1640 Ma organic-rich black shales of the Barney Creek Formation. Weatherford was contracted to acquire both vertical and lateral advanced log suites and perform subsequent log interpretation to constrain the in situ minimum and maximum horizontal stress regimes to assist with maximising gas production from future lateral placement pilot programs in the Coxco Hydrothermal Dolomite (HTD) Play. Two stratigraphic and structural domains were defined by the observed features in the image log data; a dolostone dominated, fractured strata below an erosional surface. Above this stratigraphic timeline is a monotonous package of laminated, lower-energy Barney Creek Formation sediments. Observed changes in azimuths and dips of the measured beddings suggest a phase of compression after deposition of the Barney Creek Formation, resulting in gentle folding of the formations. The porous gas-charged HTD play is drilled in top of the anticline, which is further characterised by a significant number of conductive fractures, likely indicative of open fractures.

This study assesses the prospectivity of the world’s oldest known stacked petroleum systems from the Proterozoic greater McArthur Basin (Northern Territory, Australia), which has immense potential to host both conventional natural gas and oil, in addition to shale-gas accumulations. The Mesoproterozoic succession of the Beetaloo Sub-basin and surrounding region hosts the Territory’s premier shale-gas play and is at an advanced stage of exploration for shale hydrocarbon plays. However, there is also potential for natural gas in older sedimentary packages, with flows and shows reported in underlying Paleoproterozoic successions. At the continent-scale, four regional petroleum supersystems are identified and described in order to provide a platform for consistent nomenclature at the sedimentary package and group level; in ascending stratigraphic order; these are the Paleoproterozoic Redbank and McArthur supersystems, the Paleoproterozoic–Mesoproterozoic Lawn Supersystem, and the Mesoproterozoic Beetaloo Supersystem. The Redbank and Lawn supersystems are newly named and defined, and the Beetaloo Supersystem is renamed from the former Urapungan Supersystem. Eight possible conventional natural gas plays and six shale-gas plays are documented within the McArthur Supersystem, which incorporates Glyde Package successions of the McArthur Basin and the Birrindudu Basin. Petroleum play concepts are also described from this supersystem to assist with assessing the potential for gas resources. A better understanding of the petroleum systems of the greater McArthur Basin is critical to the targeting of areas for geoscience data acquisition in order to facilitate the reduction of exploration search space; and it enables a more rigorous assessment of the potential for conventional and unconventional hydrocarbon resources at local (play) and regional scales.

Identifying new targets and new drugs has always been a daunting task, especially in cancer research. This studyexamines the binding interaction and the drug-likeness properties of small molecules derived from marine sponge Dysideaherbacea to breast cancer receptors: epidermal growth factor receptor and estrogen receptor. The receptor’s interaction with the ligand was evaluated using the Schrodinger Glide package, and affinities were assessed based on the glide score. Ligand molecules that have higher binding affinity were evaluated further for their ADMET properties using Molinspiration.We found multiple ligands binding to these targets; however, Pyridine 3 carboxyamidewas found to have binding affinity to both the receptors. Compared to the other small molecules,further simulation studies could be taken up to ascertain its structural dynamics and ensuing in-vitro experiments that could prove growth inhibition of breast cancer cells.

Objective:Mitochondrial disorders linked to Thymidine Kinase 2 (TK2) deficiency is associated to long term treatment with antiviral nucleoside analogues such as AZT (AZidoThymidine). The accumulation of AZT-TP (AZidoThymidine-TriPhosphate) is suggested to affect DNA polymerase, resulting in mtDNA (mitochondrial DNA) depletion. Thus a potent and selective inhibitor for TK2 is essential to be predicted.Methods: The human mitochondrial TK2was modeled usingModeller and the structure was evaluated using Ramachandran plot. The ligand molecule of both the derivatives of deoxythymidine and thioureawere optimized usingChemsketch and were further docked using GLIDE Schrödinger package 2009.Results: The docking results of both Deoxythymidine and Thiourea derivatives against the target TK2 were analyzed to find the ligands with good dock score and Glide energy. The ligands were selected and induced fit docking of both the derivatives were carried out and the interactions were observed usingPyMol.Conclusion: The present study indicates that deoxythymidine derivatives interact significantly and hence serve as selective inhibitors against Human Mitochondrial TK2.Keywords: Thymidine kinase 2, Azidothymidine-triPhosphate, Docking, Grid-based ligand docking with energetics, Deoxythymidine, Thiourea

The current study investigated the antimicrobial and antitubercular activities of novel N-Pyrazolyl Benzamide derivatives. The study includes the synthesis, characterization, and ligand-based molecular docking of the designed molecules. Synthesis of N-Pyrazolyl Benzamide derivatives involves a two-step process in which 5-amino pyrazole (3) intermediate is produced by the condensation reaction of aryl hydrazine (1) and β-keto nitrile (2) in acidic conditions. The final N-Pyrazolyl Benzamide derivatives (5a-n) were synthesized by the reaction involving amide coupling between 5-amino pyrazole (3) and various substituted benzoyl halides (4a-n). The compounds (5a-n) were tested for antimicrobial activity against two gram-negative strains (E. coli and S. Typhi), two gram-positive strains (S. aureus and B. subtilis) and M. tuberculosis MTB H37Rv (ATCC 27294) strains were used for evaluating antitubercular activity. Molecular docking studies of designed derivatives were performed with the Schrodinger glide package against mycobacterial pantothenate synthetase (PDB ID: 4MQ6). All the tested compounds displayed moderate to good antimicrobial and antitubercular activity against the selected strains of bacteria. All the docked molecules were found occupying the active site and established a minimum of two hydrogen bond interactions in molecular docking studies

This paper presents a novel substrateless packaging solution for the D-band active e mixer MMIC module, using a waveguide line with a glide-symmetric periodic electromagnetic bandgap (EBG) hole configuration. The proposed packaging concept has the benefit of being able to control signal propagation behavior by using a cost-effective EBG hole configuration for millimeter-wave- and terahertz (THz)-frequency-band applications. Moreover, the mixer MMIC is connected to the proposed hollow rectangular waveguide line via a novel wire-bond wideband transition without using any intermediate substrate. A simple periodical nail structure is utilized to suppress the unwanted modes in the transition. Additionally, the presented solution does not impose any limitations on the chip’s dimensions or shape. The packaged mixer module shows a return loss lower than 10 dB for LO (70–85 GHz) and RF (150–170 GHz) ports, achieving a better performance than that of traditional waveguide transitions. The module could be used as a transmitter or receiver, and the conversion loss shows good agreement in multiple samples. The proposed packaging solution has the advantages of satisfactory frequency performance, broadband adaptability, low production costs, and excellent repeatability for millimeter-wave- and THz-band systems, which would facilitate the commercialization of millimeter-wave and THz products.

Background: Piperine or piperic acid was isolated from fruits of Piper nigrum and had been reported as pharmacological valuable bioactive constituents. Keeping in view, a series of piperic acid-based N heterocyclic’s derivatives were synthesized and evaluated for antibacterial activity. All these prepared ligands were docked to study the molecular interactions and binding affinities against the protein PDB ID: 5 CDP. Objective: To meet the real need of newer antibacterials, we designed and synthesized scaffolds with good antibacterial activity. The obtained antibacterials have been validated in terms of ligand-protein interaction and thus prove to build up as good drug candidates. Methods: Antibacterial activity of the compounds were carried out against bacterial strains; three Grampositive and three Gram-negative bacterial strains using agar well diffusion method. In silico molecular docking studies were carried out using Glide (grid-based ligand docking) program incorporated in the Schrödinger molecular modeling package by Maestro 11.0. Results: Compounds BC 28, BC 32, and BC 33 exhibits antibacterial activity along with Glide docking score of -8.580, -9.753 kcal/mol, and -8.813 kcal/mol, respectively. Docking studies explained hydrogen bonding, pi-pi, and hydrophobic interactions with amino acid residues which explain the binding affinity of the most docked ligand with protein. Conclusion: In the present study, substituted piperic acid was synthesized and evaluated as antibacterial compared with standard drug ciprofloxacin and results interpret that having nitrogen as heteroatom in the heterocyclic nucleus found to be more potent than the standard drug ciprofloxacin. On comparing, substitution with electron-donating groups generates excellent antibacterial potential against the bacterial strains. It was also proved that having substitution with electron-donating groups on meta and para position with triazoline ring system exhibits greater potential while compounds which have a meta- electron-donating substituent showed lesser activity with thiazole nucleus. In addition, structure-based activities of the prepared analogs were discussed under Structure-Activity Relationship (SAR) section.

In this article, I contend that philosophical reactions against ethno-philosophy, especially the arguments by professional African philosophers such as Paulin Hountondji and Kwame Anthony Appiah, cannot go unchallenged at a time when Africa is facing a myriad of problems such as disease, famine, ethnic conflicts, religious wars, and natural disasters which, in my view, stem from the continent’s failure to reflect on its past in the quest for lasting solutions. Having looked at the historical context of the emergence of ethno-philosophy or the project of cultural revivalism, and having closely examined the premises presented by Hountondji and Appiah against ethno-philosophy - which I consider to be unconvincing because of their tendency to glide into Western philosophical forms of thought - I argue that ethno-philosophy is just like Western philosophy, as it is based on a recognized form of reasoning, namely inductive reasoning, packaged in proverbs, riddles and other cultural resources. I also argue that religious beliefs are not an obstacle to the development of scientific thought in Africa; rather, they are an aid to it since both have complementary rather than opposing roles. Abstract Efficiently profiling the water column to achieve both high vertical and horizontal resolution from a moving vessel in deep water is difficult. Current solutions, such as CTD tow-yos, moving vessel profilers, and undulating tow bodies, are limited by ship speed or water depth. As a consequence, it is difficult to obtain oceanographic sections with sufficient resolution to identify many relevant scales over the deeper sections of the water column. This paper presents a new concept for a profiling vehicle that slides up and down a towed wire in a controlled manner using the lift created by wing foils. The wings provide a novel low-power method of propulsion along the cable by using the free stream velocity of the wire moving through the water in similar fashion to a sailboat sailing up wind. Scale model tests show a wide range of achievable profiling glide slopes for tow cable angles between vertical and 45°, and effective isolation of cable strum vibration from the towed vehicle body. The concept is not depth limited and will offer two-dimensional resolution that meets or exceeds current undulating tow bodies over the full water column. Additionally, this system could be used simultaneously with many other deep towed instrument packages to produce complementary datasets.

A graph-based genetic algorithm (GA) is used to identify molecules (ligands) with high absolute docking scores as estimated by the Glide software package, starting from randomly chosen molecules from the ZINC database, for four different targets: Bacillus subtilis chorismate mutase (CM), human β2-adrenergic G protein-coupled receptor (β2AR), the DDR1 kinase domain (DDR1), and β-cyclodextrin (BCD). By the combined use of functional group filters and a score modifier based on a heuristic synthetic accessibility (SA) score our approach identifies between ca 500 and 6,000 structurally diverse molecules with scores better than known binders by screening a total of 400,000 molecules starting from 8,000 randomly selected molecules from the ZINC database. Screening 250,000 molecules from the ZINC database identifies significantly more molecules with better docking scores than known binders, with the exception of CM, where the conventional screening approach only identifies 60 compounds compared to 511 with GA+Filter+SA. In the case of β2AR and DDR1, the GA+Filter+SA approach finds significantly more molecules with docking scores lower than −9.0 and −10.0. The GA+Filters+SA docking methodology is thus effective in generating a large and diverse set of synthetically accessible molecules with very good docking scores for a particular target. An early incarnation of the GA+Filter+SA approach was used to identify potential binders to the COVID-19 main protease and submitted to the early stages of the COVID Moonshot project, a crowd-sourced initiative to accelerate the development of a COVID antiviral.

Abstract The objective was to determine the magnitude of yearly differences in parasite load in growing ram lambs. Data were obtained from the Southwest Virginia Agriculture Research and Extension Center in Glade Spring, VA. The center conducts a forage-based ram growth test during the summer where rams also undergo a parasite challenge. Data consisted of 488 Katahdin rams tested from 2012 to 2018. Rams were dewormed at delivery, and at the start of the test, each ram received an oral dose of 5,000 H. contortus larvae adjusted for body weight. Fecal egg count was measured 70 d later when rams were on average (SD) 200 (18) d old. Fecal egg counts were not normally distributed. The Box-Cox procedure indicated that a log transformation was appropriate, but the residuals were not normally distributed for several linear models. A zero-inflated negative binomial generalized linear mixed model was used for data analysis with the glmmTMB package in R. The model included fixed effects of centered and scaled weight, centered and scaled age, year, birth type, and rearing type, and the random effect of consignor. Birth type, rear type, and age were not significant (P > 0.10). The least square mean (SE) fecal egg counts by year were 344 (118) for 2012, 623 (214) for 2013, 574 (195) for 2014, 1,125 (409) for 2015, 745 (253) for 2016, 408 (142) for 2017, and 239 (86) for 2018. Differences in summer precipitation could affect average parasite load. Despite similar total summer precipitation, 2012 had 1 extremely wet month whereas, 2015 had consistent precipitation throughout the summer. We conclude that producers should not compare fecal egg counts across years because the overall average may be multiple-fold different from year to year. Ranking rams within year will be more effective to select for improved parasite resistance.

Previous Sticky Wickets can be viewed at: www.biologists.com/JCS/caveman/index.html Back when Michael Jordan ruled the world of basketball, and everything else, a TV advertisement showed him effortlessly whirling around other players, gliding through the air and dunking the basketball. He then sat down and started guzzling an artificially colored saline solution. He was surrounded by young boys who looked up at him with rapture. Then, each of the boys promptly started to guzzle the same drink and cheerfully chanted in a mantra-style, ‘I wanna be like Mike! I wanna be like Mike!’ Society loves the cult hero, someone who is, or is packaged to appear, more intelligent, faster, taller, more beautiful and exciting, and funnier than you. Here is someone to look up to from your miserable, lowly rung on society's ladder - someone whose talents you can try to emulate, even if it is only in the soft drink that you buy. What better way to get through another grinding day at work than the knowledge that you are at least guzzling the same drink as Michael Jordan! And so it is in science. Young scientists today define science in terms of personalities, cult heroes and controversy between laboratories. In a discussion group for a class that I teach, students and faculty leaders critique papers that parallel the more didactic parts of the class. The discussion of each paper is started with a brief (historical) background and overview of the biological problem by one of the students. I noticed that the students presented the background by date and personality (as encouraged by one faculty leader!), and often highlighted personality conflicts: ‘In 1987, Phlemspengler showed that pigs could become airborne for short distances if they first leapt out of a first-floor window. Then, in 1988, Phlemspengler's rival, Snotely, reported that Phlemspengler's work was flawed because he had in fact tossed the pigs out of the window. Nevertheless, Snotely was intrigued with the possibility that pigs might fly. In 1989, she found that excess rolls of fat underneath the forelimbs of pigs could produce lift if splints were used to extend the forelimbs laterally from the body, and the pigs were then tossed out of a first-floor window. The big breakthrough came in 1992, when Phlemspengler's former graduate student Arsenmuth engineered a transgenic pig that had an enlarged sternum, a project that he was not allowed to work on as Phlemspengler's student. Arsenmuth showed that extensive exercise and pectoral muscle development, together with the forelimb fat rolls, provided sufficient strength and lift to enable the pigs to fly for short distances. At a recent meeting, Phlemspengler noted that Arsenmuth still had to toss the pigs from a first floor window in order to get them airborne and, therefore, he, Phlemspengler, was still the first discoverer of pig flight.’ It would be considerably more scholarly to summarize the evolution of ideas (one came from the other), to which each person contributed, and leave out names and anecdotal personality conflicts. ‘The evolution of pig flight came about through a series of body adaptations, starting with the use of splints to support the forearms and surrounding adipose tissue, and then the development of the sternum and pectoral muscles. However, it remains to be shown whether these pigs can sustain powered flight or simply glide after an assisted launch.’ Who said that the recitation of science should be exciting or presented in the format of an exclusive for the News of the World, National Enquirer or Das Bild? Unfortunately, the science cult figure looms large, and in some cases very large. Students want to grow up to be like. pick a star of genetics, cell or developmental biology - someone famous at a prize university, who gets the rock-star billing at meetings and has the greased pipeline into the top journals. And why not? Isn't this the way to get to the top (the top in terms of visibility, not necessarily scientific contribution), to emulate the success of someone else, to mould yourself to their pedigree and personality? Do not succumb to this form of societal inbreeding! It is not that Mike (or Gary, or Fiona) is not worth looking up to. However, it is better to be yourself and to develop your own set of principles, your own way of thinking and performing experiments, your own writing and presentation styles. Face it: in the end, you will never have the ability to perform the experimental equivalent of a gravity-defying, 360(o) spin tomahawk dunk, and you will definitely not look good in vest and shorts!

At the continent-scale, four regional petroleum supersystems are identified and described in order to provide a platform for consistent nomenclature at the sedimentary package and group level; in ascending stratigraphic order; these are the Paleoproterozoic Redbank and McArthur supersystems, the Paleoproterozoic–Mesoproterozoic Lawn Supersystem, and the Mesoproterozoic Beetaloo Supersystem. The Redbank and Lawn supersystems are newly named and defined, and the Beetaloo Supersystem is renamed from the former Urapungan Supersystem. Eight possible conventional natural gas plays and six shale-gas plays are documented within the McArthur Supersystem, which incorporates Glyde Package successions of the McArthur Basin and the Birrindudu Basin. Petroleum play concepts are also described from this supersystem to assist with assessing the potential for gas resources. A better understanding of the petroleum systems of the greater McArthur Basin is critical to the targeting of areas for geoscience data acquisition in order to facilitate the reduction of exploration search space; and it enables a more rigorous assessment of the potential for conventional and unconventional hydrocarbon resources at local (play) and regional scales. Shot put is a throwing exercise in track and field. Performance has a strong relationship with the thrower’s height, technique, and energy transferred. The purpose of this study was to compare basic kinematics, joint movement, shot mechanical energy, and joint energy of the male glide-style shot put between high-performance (HP) and low-performance (LP) athletes. Data for 12 right-handed male shot putters with glide-style was captured at the National Games held in Taipei city in 2013. The HP ( n = 6) and LP ( n = 6) groups were classified according to their best throwing distance in the competition. Additionally, the 2D kinematics of their joints and shots were obtained by Kwon 3D motion analysis. Finally, the kinematics and energy were calculated in Excel software package. Results showed significant differences in throwing distance, the relationship between throwing distance and body height index (RDH index), release velocity, and the shoulder-shot (BSX) angle at right toe-off ground instant (RTF) and the angle difference between the right toe-off instant and release instant (RI-RTF) between the HP and LP groups. However, the energy of the joints and shots were not significantly different between the two groups. The better throwers exhibited a faster release velocity, accompanied by a smaller change in the SSX angle, and the SSX angle can be used by the coach to assess the body’s forward or backward tilt during training.

Purpose According to the study of the space flight market, there is a demand for space suborbital flights including commercial tourist flights. However, one of the challenges is to design a mission and a vehicle that could offer flights with relatively low G-loads. The project of the rocket-plane in a strake-wing configuration was undertaken to check if such a design could meet the FAA recommendation for this kind of flight. The project concept assumes that the rocket plane is released from a slowly flying carrier plane, then climbs above 100 kilometers above sea level and returns in a glide flight using a vortex lift generated by the strake-wing configuration. Such a mission has to include a flight transition during the release and return phases which might not be comfortable for passengers. Verification if FAA recommendation is fulfilled during these transition maneuvers was the purpose of this study. Design/methodology/approach The project was focused on the numerical investigation of a possibility to perform transition maneuvers mentioned above in a passenger-friendly way. The numerical simulations of a full-scale rocket-plane were performed using the simulation and dynamic stability analyzer (SDSA) software package. The influence of an elevator deflection change on flight parameters was investigated in two cases: a transition from the steep descent at high angles of attack to the level glide just after rocket-plane release from the carrier and an analogous transition after re-entry to the atmosphere. In particular, G-loads and G-rates were analyzed. Findings As a result, it was found that the values of these parameters satisfied the specific requirements during the separation and transition from a steep descent to gliding. They would be acceptable for an average passenger. Research limitations/implications To verify the modeling approach, a flight test campaign was performed. During the experiment, a rocket-plane scaled model was released from the RC model helicopter. The rocket-plane model was geometrically similar only. Froude scales were not applied because they would cause excessive technical complications. Therefore, a separate simulation of the experiment with the application of the scaled model was performed in the SDSA software package. Results of this simulation appeared to be comparable to flight test results so it can be concluded that results for the full-scale rocket-plane simulation are also realistic. Practical implications It was proven that the rocket-plane in a strake-wing configuration could meet the FAA recommendation concerning G-loads and G rates during suborbital flight. Moreover, it was proven that the SDSA software package could be applied successfully to simulate flight characteristics of airplanes flying at angles of attack not only lower than stall angles but also greater than stall angles. Social implications The application of rocket-planes in a strake-wing configuration could make suborbital tourist flights more popular, thus facilitating the development of manned space flights and contributing to their cost reduction. That is why it was so important to prove that they could meet the FAA recommendation for this kind of service. Originality/value The original design of the rocket plane was analyzed. It is equipped with an optimized strake wing and is controlled with oblique, all moving, wingtip plates. Its post-stall flight characteristics were simulated with the application of the SDSA software package which was previously validated only for angles of attack smaller than stall angle. Therefore, experimental validation was necessary. However, because of excessive technical problems caused by the application of Froude scales it was not possible to perform a conventional test with a dynamically scaled model. Therefore, the geometrically scaled model was built and flight tested. Then a separate simulation of the experiment with the application of this model was performed. Results of this separate simulation were compared with the results of the flight test. This comparison allowed to draw the conclusion on the applicability of the SDSA software for post-stall analyzes and, indirectly, on the applicability of the proposed rocket-plane for tourist suborbital flights. This approach to the experimental verification of numerical simulations is quite unique. Finally, a quite original method of the model launching during flight test experiment was applied.
