W 2. połowie XVII w. do Warszawy przybyła grupa architektów pochodzących z położonej nad jeziorem Lugano Valsoldy, w północnej Lombardii. Sześć listów Carla Ceroniego do Samuela Pączkowskiego, administratora dóbr Krasińskich, odnalezionych w Archiwum Głównym Akt Dawnych w Warszawie przyniosło nowe informacje na temat prac prowadzonych w Węgrowie.

The chapel dedicated to Beata Lucia in the cathedral of Narni has been attributed to Nicola Michetti on the basis of both style and the fact that he had worked elsewhere for the patron, Cardinal Giuseppe Sacripante. The configuration of the altar surround, however, is not typical of Michetti's work. That structure, in fact, is by Carlo Rainaldi and originally stood in the chapel of S. Antonio di Padova in SS. Apostoli in Rome. Sacripante purchased the altar surround in October 1711 and had it set up in Narni sometime between 1712 and 1714. Michetti's contribution to the design of the chapel, therefore, is limited to the revetment of the walls, the windows, and the transverse oval dome. Rainaldi's altar surround is an important find. Conceived in 1652 and constructed between 1654 and c. 1656, it is the immediate forerunner of his urbanistic masterpieces of the early 1660s, S. Maria di Montesanto and S. Maria dei Miracoli, on piazza del Popolo.

The present text is the second instalment of the cycle of articles based on its author’s doctoral dissertation entitled The birth of cello as a solo instrument – instruments, prac-tice, and selected literature examples (Academy of Music in Łódź, 2017). It is an at-tempt to systematise the knowledge about the first cello literature creators and their compositions. The text includes descriptions (characterisations) of the first pieces writ-ten for the cello as early as in the 17th century along with short biographical sketches of their composers. The article characterises the main centres of development of cello mu-sic, such as Bologna and Modena, the places of work of Giovanni Battista Vitali, Do-menico Gabrielli, Petronio Franceschini, Giuseppe Jacchini, Bononcini brothers, Gio-vanni Battista Degli Antoni, Domenico Galii, Giuseppe Colombi, and others. These composers mainly focused around Bologna’s Accademia Filarmonica and Basilica of San Petronio, as well as the court of Francesco II d'Este in Modena. The article also pays attention to the circumstances in which the first attempts to write cello pieces were made. Moreover, it mentions singular historical items of early cello literature written in such centres as Rome, Venice or Naples, where Carlo Fedeli, Giovanni Lorenzo Lulier, Nicola Francesco Haym or other composers worked.

Gaetano Cattaneo (1771-1841), founder of the Numismatic Cabinet of Brera, inherited in 1815 from Giuseppe Bossi the historiographical materials that the secretary of the Brera Academy had collected with the aim to create a work dedicated to the history of Lombard art. Cattaneo also devoted himself to the project of a History of the arts and artists of the Lombard school, which never came to light, although its preparation is largely documented by his correspondence with Italian and foreign scholars, such as Cicognara, De Lazara and Passavant. A trace of Cattaneo’s work survives in an unpublished manuscript, kept in the Biblioteca Ambrosiana. In a schematic form, the document reconstructs the history of architecture in Lombardy from the Middle Ages to the sixteenth century, though it was supposed to be continued until the early nineteenth century. The information provided by Cattaneo about the origins of the different “epoche”, in which he divides the developments of Lombard architecture, as to the distinctive structural and stylistic characteristics or as to the masters and their main works offer a very interesting insight into the Milanese historiographical consciousness in the time between French domination and Restoration. The essay considers in particular the judgment on the different phases of the Middle Ages, with particular regard to the profile of Longobard architecture and the origins of the Milan Cathedral. In this case both local debates and the work of Seroux D’Agincourt are fundamental for Cattaneo. Another significant theme is the judgment on the architecture of the sixteenth century in Lombardy, which re-evaluates Leonardo, resizes Bramante and, above all, censors the protagonists of the Borromean age, from Galeazzo Alessi to Francesco Maria Richino. In this respect, the internal debate at the Brera school of architecture and the historiographical positions of Carlo Bianconi and Giuseppe Bossi played an important role, other significant latest sources such as Francesco Antonio Albuzzi and Venanzio De Pagave. The essay reports the complete edition of Cattaneo’s text according to the Ambrosian copy, which was probably made by his collaborator Carlo Zardetti.

Mozart’s Milan concert on 12 March 1770 secured for him his first commission to write an opera seria for Italy. The works usually thought to have been performed on this occasion are Fra cento affanni, k88, Misero tu, k73A, and Misero me . . . misero pargoletto, k77. This article offers new evidence for redating Fra cento affanni and argues that neither it nor Misero tu was performed at the event. Instead, the case is made for a programme consisting solely of concert arias with texts from the libretto of Metastasio’s Demofoönte, presented together as a kind of truncated ‘opera in aria’. In support of this hypothesis, the compositional circumstances surrounding Se tutti i mali miei, k83, and Ah più tremar, k71, are reassessed and possible performances by the singers Giuseppe Aprile, Antonia Bernasconi, Guglielmo d’Ettore, Lucrezia Agujari and Carlo Niccolini discussed.

In recent decades, architectural historians, preservationists, and the general public have shown a growing interest in Fascist-era buildings. Many of the most high-profile examples are those associated with the monumental excesses of the regime. However, new attention has also been focused on more modest buildings that are significant examples of interwar Italian modernism or Rationalism, including former party headquarters (case del fascio). Taking as primary examples works by Giuseppe Terragni, the architect most often associated with Rationalism, as well by Luigi Carlo Danieri and Luigi Vietti, whose interwar contributions to Italian modernism have been less often the focus of scholarly attention, this article traces the postwar histories of case del fascio with the aim of better understanding the ways in which architecture and politics intersect and some of the consequences of this for the contemporary Italian architectural landscape.

Reprinted in 1801, Vico's New Science (originally published 1744) had a profound impact on Bourbon Restoration culture, particularly in Italy and France, where it touched post-revolutionary readers profoundly. The reasons for Vico's revival in the early nineteenth century relate closely to the trauma of the political and social changes of that era. Vico's readership seems to have had significant peaks during periods of rapid social transformation: nineteenth-century readers reread the New Science in an attempt to find the reasons for revolutionary failure, and to relate the terror, the sense of displacement, failure and trauma to recognizable laws, promising that, after a crisis, a period of renaissance must necessarily follow. This article analyses the hermeneutic practices of some post-revolutionary readers of Vico (Carlo Cattaneo, Vincenzo Cuoco, Giuseppe Ferrari, Ugo Foscolo, Francesco Lomonaco) and suggests a comparison with the practices of readers during the Second World War (Eric Auerbach, Carlo Levi, Mario Fubini). By doing so, I propose an interpretation of Vico's New Science as a ‘posthumous’ book, acquiring special shades of significance when its readers experience the feeling that nothing will ever be like before, and meditate upon it in isolation, in fear, in exile, upon return from the front, and in prison.

Joseph (Giuseppe or Józef) Poniatowski (1816–1873), Polish prince, singer, opera composer and politician, spent all his life abroad: firstly in Italy, then in France and, finally, in England. His artistic output comprises twelve operas composed between 1839 and 1872; nine of them to Italian and three to French texts. Being an amateur composer, he notwithstanding succeeded in staging his operas in many operatic theatres of renown, including La Scala, Covent Garden, Teatro San Carlo, Teatro La Fenice and the Paris Opéra. The paper presents the composer’s output in the social and political contexts of his times. Prince Poniatowski started his international career as a plenipotentiary minister of Tuscany in Paris, London and Brussels; then he settled down in Paris and became a French citizen and even a French senator. He enjoyed the close friendship of Napoleon III with whom he went into exile to England after the Sedan defeat. In all of his three domiciles he presented his operas to the audiences. However, as shown by the press reviews, their reception changed from appreciation to indifference, which was caused by the different political and social backgrounds in the particular countries.

MEPO efficacy was evaluated in the first 12 months in terms of systemic disease and asthma control. The occurrence of any adverse event (AE) was recorded.Results:142 patients were included (38% males; median age 46.4 (IQR 36.7-54.4); 110 and 32 on MEPO 100 and 300 mg/month, respectively). General, ear-nose-throat, pulmonary, and neurological symptoms significantly decreased during treatment (table 1). MEPO accounted for a significant reduction in the BVAS (figure 1) and for a steroid sparing effect (figure 2). PROBLEMS WITH THE APPOINTMENT OF THE PAPAL NUNCIO TO POLAND, NOVEMBER 1935 – APRIL 1937SummaryThe article describes the appointment of Filippo Cortesi to the office of papal nuncio to the Republic of Poland, and reviews the role of the Polish diplomats accredited to the Holy See in this process. The appointment of a nuncio following the departure of Francesco Marmaggi was extremely complicated. The list of candidates was the resultant of a number of factors. The individuals whose names were on it had used influence with the pope, the Vatican’s secretary of state, or former nuncios to Warsaw. Both the Polish government and the bishops of Poland had a say in the final outcome. In addition there was also the volatile political situation in Europe at the time, especially in Spain and Yugoslavia, which exerted an effect on the process. In June 1936 Carlo Chiarlo received the nomination for the office. However, the unanimously negative position of the Conference of the Bishops of Poland, especially Cardinals Kakowski and Hlond, stopped the appointment at the last moment. The Polish Government did not want Ermenegildo Pellegrinetti to be appointed and tried to prevent it, considering Angelo Giuseppe Roncalli the ideal candidate. Eventually Pius XI appointed Filippo Cortesi, formerly nuncio to Argentina.

In the library of the Gioenian Academy are stored two copies of the book A Description of active and extinct Volcanoes, of Earthquakes, and of Thermal Springs (London 1848, II edition), bearing a dedication to the Academy by Dr Charles Daubeny. The author was a prominent chemist, geologist and botanist which made a particular endeavour to write A kind of Guide-book to the explorers of Districts in which Volcanic phaenomena occur abroad. Daubeny was elected to the prestigious Aldrichian Chair of Chemistry, University of Oxford (1834), was appointed to the Sherardian Chair of Botany (1834) and to the Sibthorpian Chair of Rural Economy (1840). He undertook interesting travels to Europe and North America (1819-1845), particularly three journeys to the south of Italy (1823-1845). He wrote Sketch of the Geology of Sicily (The Edinburgh Philosophical Journal, 1825), a booklet in two parts never translated into Italian. During the visit to the Etna region Daubeny had contact with local scholars such as Mario and Carlo Gemmellaro, Giuseppe Alessi and Carmelo Maravigna which supported his appointment as foreign associate of the Gioenian Society at Catania since 10 June 1824. He was the first of the squad of geologists as early foreign corresponding or honorary members. The relevance of his main book, Sicilian booklet and other works is discussed.

Naples in the last thirty years of the eighteenth-century was characterized by a fervent climate of theatrical experimentation. Although too often viewed as the last stronghold of Metastasian dramatic principles and traditions, the city was deeply influenced by the “reform culture” of Northern Europe. These exterior influences were bolstered by the contributions of local practitioners, whether composers, performers, and theorists. This essay is a brief consideration of how the ideas of “reform culture” affected contemporary Neapolitan theatrical practices and the emergence of new works in the city. A critical source for “reformed” theatrical philosophy was the work of Antonio Planelli (1747–1803), whose treatise Dell’opera in musica (1772) is a significant exploration and commentary on the dramatic stage of the Bourbon capital. Progressing from theatrical philosophy to existing practice, I will consider how the prevailing conditions animated the creation of the largely unknown cantata/pastorale/opera, La pietà d’amore (1782) by the singer, composer, and Calzabigi protégé Vito Giuseppe Millico (1737–1802), created expressly for Naples under the sway of reform principles and his direct collaborations with the poet of Orfeo, Alceste, and Paride ed Elena. My study concludes with an examination of the emergence of the so-called “Lenten tragedy” or azione sacra per musica, a theatrical form created in the exclusive environs of the Teatro di San Carlo, the royal theater of the Bourbon capital, yet imparting a new theatrical aesthetic and modes of representation for contemporary sacred genres consistent to select ideals of reform culture.

Jan Reisner (ca. 1655-1713) was a painter and architect. He was sent by King Jan III together with Jerzy Siemiginowski to study art at St. Luke Academy in Rome. He traveled to the Eternal City (where he arrived on February 24, 1678) with Prince Michał Radziwiłł’s retinue. Cardinal Carlo Barberini, who later became the protector of Regni Poloniae, was the guardian and protector of the artist during his studies in 1678-1682. In the architectural competition announced by the Academy in 1681 Reisner was awarded the fi prize in the fi class, and a little later he was accepted as a member of this prestigious university. He was awarded the Order of the Golden Spur (Aureatae Militiae Eques) and the title Aulae Lateranensis Comes, which was equivalent to becoming a nobleman. The architectural award was conferred by the jury of Concorso Academico, composed of the Academy’s principe painter Giuseppe Garzi, its secretary Giuseppe Gezzi, and the architects Gregorio Tommassini and Giovanni B. Menicucci. In the Archivio storico dell’Accademia di San Luca, preserved are three design drawings of a church made by Jan Reisner in pen and watercolor, showing the front elevation, longitudinal section, and a projection. Although they were made for the 1681 competition, they were labelled with the date 1682, when the prizes were already being awarded. Reisner’s design reflected the complicated trends in the architecture of the 1660s and 1670s, especially in the architectural education of St. Luke’s Academy. There, attempts were made to reconcile the classicistic tendencies promoted by the French court with the reference to the forms of mature Roman Baroque. As a result of this attempt to combine the features of the two traditions, an eclectic work was created, as well as other competition projects created by students of the St. Luke’s Academy. The architect designed the Barberini temple-mausoleum, on a circular plan with eight lower chapels opening inwards and a rectangular chancel. The inside of the rotund is divided into three parts: the main body with opening chapels, a tambour, and a dome with sketches of the Fall of Angels. Inside, there is an altar with a pillar-and-column canopy. The architectural origin of the building was determined by ancient buildings: the Pantheon (AD 125) and the Mausoleum of Constance (4th century AD). A modern school based of this model was opened by Andrea Palladio, who designed the Tempietto Barbaro in Maser from 1580. In the near future, the Santa Maria della Assunzione in Ariccia (1662-1664) by Bernini and Notre-Dame-de-l’Assomption (1670-1676) in Paris by Charles Errard could provide inspiration. In particular, the unrealized project of Carlo Fontana to adapt the Colosseum to the place of worship of the Holy Martyrs was undertaken by Clement X in connection with the celebration of the Holy Year in 1675. In the middle of the Flavius amphitheatre, he designed the elevation of a church in the form of an antique-styled rotunda, with a dome on a high tambour and a wreath of chapels encircling it. Equally important was the design of the fountain of the central church in Basque Loyola (Santuario di S. Ignazio a Loyola). In the Baroque realizations of the then Rome we find patterns for the architectural decoration of the Reisnerian church. In the layout and the artwork of the facades we notice the influence of the columnar Baroque facades, so common in different variants in the works of da Cortona, Borromini and Rainaldi. The monumental columnar facades built according to Carlo Rainaldi’s designs were newly completed: S. Andrea della Valle (1656 / 1662-1665 / 1666) and S. Maria in Campitelli (designed in 1658-1662 and executed in 1663-1667), and Borromini San Carlo alle Quatro Fontane (1667-1677). The angels supporting the garlands on the plinths of the tambour attic are modelled on the decoration of two churches of Bernini: S. Maria della Assunzione in Ariccia (1662-1664) and S. Andrea al Quirinale (1658-1670). The repertoire of mature Baroque also includes the window frames of the front facade of the floor in the form of interrupted beams and, with the header made in the form of sections capped with volutes. The design indicates that the chancel was to be laid out on a slightly elongated rectangle with rounded corners and covered with a ceiling with facets, with a cross-section similar to a heavily flattened dome. It is close to the solutions used by Borromini in the Collegio di Propaganda Fide and the Oratorio dei Filippini. The three oval windows decorated with C-shaped arches and with ribs coming out of the volute of the base of the dome, which were among the characteristic motifs of da Cortona, taken over from Michelangelo, are visible. The crowning lantern was given an original shape: a pear-shaped outline with three windows of the same shape, embraced by S-shaped elongated volutes, which belonged to the canonical motifs used behind da Cortona by the crowds of architects of late Baroque eclecticism. Along with learning architecture, which was typical at the Academy, Reisner learned painting and geodesy, thanks to which, after his return to Poland, he gained prestige and importance at the court of Jan III, then with the Płock Voivode Jan Krasiński. His promising architectural talent did gain prominence as an architect in Poland, although – like few students of St. Luke’s Academy – he received all the honors as a student and graduate.

This article investigates the growing proliferation of curtains and wall hangings as key elements in the design of art exhibitions in the years 1930–1955. To demonstrate how textiles were successfully employed as mediators on the threshold between architecture, design objects, and fine arts, I first examine the increasing use of curtains in the interwar period, Fascist Italy, and Nazi Germany to subsequently explore how the role of fabrics in both countries’ rationalist and neoclassicist architecture also played a significant part in exhibition design after the Second World War. I chart how the interest in textiles culminated in 1955, when glossy plastic curtains were integrated into the exhibition architecture at the first documenta in Kassel, Germany, one of the country’s most prestigious recurring art events to this day. During these politically turbulent decades, the exchange between exhibition designers in both countries was bound together by a profound reassessment of the relation between architecture, design, and art. The renewed consciousness of design as an integrated practice played a key role in 1930s architecture, also providing the foundation for the Bauhaus curriculum and the work of artists, designers, and architects (e.g., Wassily Kandinsky, Giuseppe Pagano, Le Corbusier, Carlo Scarpa, Willi Baumeister, Arnold Bode). I demonstrate that during this period textiles were essential for creating continuity between exhibitions and exhibits of vastly differing styles and contexts. The wall hangings, veils, and banners that were used as part of the monumental spaces created for the Fascist regimes in Italy and Germany were ultimately appropriated and turned into means to undermine the neoclassicist and rationalist style in a way that echoed, I argue, society’s neobaroque sensibility in the aftermath of World War II. Though the Federal Republic of Germany’s first two decades were characterized by the general will to educate its citizens in the aesthetics of internationalism, this effort and the concomitant return to the interwar period were accompanied by a strong resurgence in religiosity and desire for emotionally compelling experiences, which signify a partial disavowal of modernism’s most radical stipulations.

Abstract Background: Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal human malignancies: novel therapeutic approaches beyond conventional chemotherapy are still lacking and prognosis remains poor, even for resectable patients (pts). Furthermore, there is an almost complete absence of validated predictive factors. Consequently, robust biomarkers for the early diagnosis and the prognostic stratification are urgently needed in clinical practice, especially in the context of neoadjuvant and adjuvant settings. In the last years, evidence revealed the crucial role of miRNAs in cancer initiation and progression, as well as in the chemo-resistance mechanisms, suggesting their use as clinical biomarkers. Material and methods: In this pilot study, we performed a microarray analysis to characterize global miRNA expression profile from surgical tissue samples collected from 20 resected PDAC pts pooled into 4 groups according to different clinico-pathological features: nodal metastases (N+/N-) and tumor grading (G2/G3). Results: According to expression patterns, we identified, among 384 miRNAs, a significant different modulation for 11 miRNAs associated to G2 vs G3 and for 7 miRNAs in N+ vs N- disease, suggesting a possible specific signature reflecting histological grade and nodal metastasis occurrence, respectively. We focused on 2 up-regulated (miR-138-5p and miR-518-3p) and 3 down-regulated (miR-215-5p, miR-519a-3p and miR-576-5p) miRNAs in N+ pts, and on 3 up-regulated (miR-1-3p, miR-31-5p and miR-205-5p) in G3 pts, in order to verify their possible implication in the molecular changes behind tumor differentiation and spread, as well as their potential use for prognostic and therapeutic purpose. A bio-informatic analysis was also performed, using different in silico tools, to study both high affinity miRNA targets and cross-regulated pathways among the upand down-regulated miRNAs. The results identified several associated targets involved in multiple signaling pathways commonly dysregulated in cancer. Finally, BRCA1/2 and RB1 miRNAs-mediated-modulation is actually ongoing, considering the pivotal role of these genes in some PDAC pts. Conclusion: These preliminary data provide a strong rationale to further investigate miRNAs expression in larger cohorts of PDAC pts, possibly integrating validated tissue miRNAs data with circulating miRNAs, in order to identify strong (and easily accessible) potential biomarker(s) with prognostic and/or predictive significance. The building of the old College of Piarist in Chełm – located on Lubelska Street, near the late baroque Church of Holy Apostles the Messengers – is now the seat of the Wiktor Ambroziewicz Chełm Land Museum. Until now, it has not raised much interest among researchers and – appearing as a work of architecture devoid of expressive style features – has not been the subject of scientific reflection. This situation is changed by the disclosure of the preserved drawing from 1698, showing the building in a horizontal projection and axonometric view, stored in the Archivio Generale delle Scuole Pie in Rome. The information contained in written documents kept there allow to determine the time of construction of the building for the years 1698-1700. The project proves that the preserved edifice did not change substantially its one-story block, set on the plan of the letter H. The innovations concerned only the roof part over the main body, which was originally the Krakow roof, and the extension of one of the side wings in 1720-1724 (so that the college was connected to the church). Neither did the subsequent transformations significantly affect the internal divisions, be it in the two-and-a-half tract main corpus, with the cross-corridor communication system introduced therein, or in the single tract side wings. The shape of the building and the severity of the development of its facade, representing the baroque in its classicizing version, suggests the designer – Giuseppe Piola, an architect working in Warsaw at the turn of the 17th and 18th centuries, building, at the request of the Piarist order, also their church and monastery complex in Szczuczyn. However, the extension of the college wing made in the first half of the 18th century should probably be associated with the person of another capital architect – Carlo Antonio Bay, who at the same time, together with his son-in-law, Vincenzo Rachetti, also an architect, made calculations for the Piarist priests from Chełm for the profitability of their parcel located in the suburbs of the city of Lublin. The building in Chełm was a monastic college, and at certain times also a “profesorium”, in which Piarist clerics learned philosophy at a higher level of education. Contrary to some suggestions, there was never a public school run by the Piarists in this building. It was founded – as a Russian gymnasium – only after the January Uprising and the dissolution of the Scholarum Piarum community.

XVI i pocz. XVII w. Przykład pałacu Radziwiłłów-Koniecpolskich znakomicie ilustruje charakterystyczne w Warszawie zjawisko, gdy wczesnobarokowe wnętrza pałaców próbowano w XVIII w. unowocześnić by spełniały nowe wymogi reprezentacji.

Background:Few studies have examined the correlation between clinical demographic and laboratory parameters with peripherical radiological erosive disease in PsA pts. Objectives:To examine the association between clinical, demographical and laboratory data and the presence of radiographic erosions (RE) in the peripheral joints of psoriatic arthritis (PsA) pts. Methods:A cross-sectional study was conducted in consecutive patients with PsA afferring 7 rheumatological italian tertiary care centers. Demographical, clinical, laboratory and imaging data were collected according to a standardized protocol. A patient was considered as affected by erosive disease (ED) if at least one joint presented radiographic erosions at hand and/or feet rx examination. Patients with ED at early rx examination (before 5 y from disease diagnosis) were considered as early ED (EED) pts and pts without ED at 6 y or more rx examination from disease diagnosis were considered as not EED (NEED).The association between the presence of joint erosions and demographical, clinical and laboratory data was assessed using logistic regression analysis. The results were expressed in terms odds ratios (OR), and 95% confidence intervals (CI).Results:Rx hand and feet examination were available for analysis in 492/794 (39.9 % females, mean age 53.3 ± 13.2 y, mean PsA duration 16.9 ± 16.8 y, ED 171 pts). 48 pts had EED and 133 pts had NEED. At univariate analyses factors significantly associated with EED (p < 0.20) were PsA duration (OR=0.979,95%CI 0.953-1.006, p = 0.119), diagnostic delay (OR=1.077, 95%CI 1.018-1.138, p = 0.009), history of peripheral enthesitis (OR=2.308,95%CI 0.904-5.888, p= 0.080), hypertrigliceridemia (OR=2.756,95%CI 0.997-7.618, p = 0.0.051), hypercholesterolemia (OR=1.687, 95%CI 0.777-3.661, p = 0.186), hyperuricemia (OR=0.450, 95%CI 0.174-1.166, p = 0.10), use of biological agents (OR=1.712, 95%CI 0.873-3.355, p=0.118). Factors significantly associated with EED at multivariate regression analyses were diagnostic delay (OR = 1.11, 95% CI: 1.01, 1.22), history of enthesitis (OR = 3.15, 95% CI: 1.23, 8.22), use of therapy with biological agents (OR = 3.60, 95% CI: 1.31, 9.85) with protective effect of hyperuricemia (OR = 0.25, 95% CI: 0.07, 0.90).Conclusion:The presence of EED in a group of consecutive PsA patients is correlated to diagnostic delay and history of enthesitis. Abstract African Americans (AA) have higher incidence and mortality rates for several cancer types in comparison to their European American (EA) counterparts. Increasing participation in clinical research and patient registries, related to precision cancer medicine, will significantly improve cancer health equity. Many AA cancer patients are treated in community oncology clinics. Unfortunately, these health systems have limited access to Clinical Laboratory Improvement Amendments (CLIA) next generation sequence (NGS) germline and somatic DNA and RNA testing that are used to inform oncologists on the best treatment and/or clinical trial options for cancer patients. Indeed, AA CLIA NGS sample sets are poorly represented, which could presumably result in incomplete knowledge of genomic variants that could affect their treatment and overall outcomes. Hence, it is crucial to implement CLIA NGS efforts for all cancer patients. To address these disparities, Morehouse School of Medicine has formed the Comprehensive Approach to Reimagine health Equity Solutions (CARhES) consortium with Tuskegee University that has engaged community oncology practices in Alabama and Georgia - two of five Black Belt states. The CARhES consortium aims to implement precision cancer medicine to underserved and underrepresented communities that will improve the standard of cancer care by providing access to CLIA NGS testing, clinical trials, and personalized cancer care. Here we describe the first proof of concept of this approach with community oncology partners, i.e., Grady Health System, Wellstar Health System, Georgia Urology, Midtown Urology, and Maui Memorial Medical Center. At the time of consent, saliva, buccal, and tumor samples were collected from participants. Germline and somatic CLIA NGS was performed, and medical reports were returned to practitioners within 14 days. Prior to the COVID pandemic, the study enrolled over 880 patients with a 88% consent rate (n = 1000) in the first 11months of the program. At the start of the COVID pandemic, recruitment efforts were suspended for four months with a slow restart by June 2020. A decrease in the number of staff, office visits (67% reduction), and increase in COVID cases significantly limited recruitment efforts. During this slowdown, we established and improved eConsenting capabilities, which exist today. Community anxiety, due to the pandemic and SARS-CoV-19 vaccine efforts, resulted in a significant reduction in consent rates (88% to 60%). Nevertheless, this study began in April of 2019 and consented 1,750 participants in less than 2 years. Taken together, our study shows that a community-focused precision medicine approach requires meeting people where they are and providing them with access and understanding the benefit of clinical trial participation. The approximate 2,000 clinically annotated genomic AA datasets will greatly contribute to our understanding of cancer health disparities and among the first steps to democratize precision medicine. Abstract Introduction: Antibodies targeting PD-1 play a relevant role in the management of metastatic non-small cell lung cancer (NSCLC), although the majority of patients experience disease progression to treatment. We performed a pretreatment immune-related gene expression analysis to predict outcomes of patients with advanced stage NSCLC receiving pembrolizumab as first-line therapy. Methods: Pretreatment tissue samples from 48 metastatic NSCLC patients with PD-L1 high expression (> 50% 22C3 clone testing antibody), who undergone first line pembrolizumab therapy, were evaluated. Total RNA was isolated (RNeasy FFPE Kit Qiagen) and quantified (NanoDrop Fluorometer, Thermo Fisher Sci.). We performed a mRNA expression analysis of 770 immune-related genes (belonging to 21 pathways) by using the nCounter PanCancer Immune Profiling Panel (NanoString Tech.). Data normalization was performed in-house by using R functions. Patients were clustered by the K-means algorithm according to results of the gene expression analysis for each pathway. Advanced analyses were performed by nCounter Advanced Analysis plug in for nSolver Software (v. 2.0.134) and R package. Estimates of Overall Survival (OS) were done by Kaplan-Meier curves and tests of differences by log-rank test. Regression models were built including covariables. All tests were two-sided, with a statistical significance at p-value < 0.05. Results: The median age of patients was 72.5 years (IQR 64.3-77.0). Thirty-seven (77.1%) were male, 36 (75%) with non-squamous histology, 37 (77.1%) had an ECOG PS of 0 or 1, and 46 (95.8%) were former or current smokers. In 16 out of 48 cases (33.3%) the immune profile was performed on tumor-infiltrated lymph nodes. Disease Control Rate per RECIST 1.1 was 52.1% (16 partial and 2 complete responses, 7 disease stabilizations as best response). Median Progression-Free Survival was 2.96 months (95% CI 2.35-7.25), median OS was 13.43 months (95% CI 6.24-NR). The principal component analysis excluded 1 outlier subject by the gene expression analysis. Five gene expression signatures were significantly correlated with OS (Adhesion, Cytokines, Leukocyte Functions, Macrophage Functions, TNF Superfamily), all identifying 2 clusters of patients among each pathway. The 8-genes signature of the Leucocyte Functions pathway (CX3CL1, FUT7, HCK, IFNG, LCP1, SH2D1B, THBD, VEGFA) was the solely significantly correlated to OS regardless of age, sex, tumor histology, ECOG PS, smoke and site of diagnostic sampling (lymph node or non-nodal tissue), with a Hazard Ratio between the identified clusters of 0.29 (95% CI 0.24-0.60, p = 0.01). Conclusions: Our results support the hypothesis that pretreatment immune-related mRNA expression patterns, identified through the nCounter platform, may predict OS of patients with metastatic NSCLC treated with first line anti PD-1 therapy. Background:Radiofrequency Echographic Multi Spectrometry (REMS) is the first clinically available approach for direct non-ionizing measurement of bone mineral density (BMD) at lumbar spine (LS) and femoral neck (FN). Available scientific evidences describe BMD estimated by REMS as an accurate parameter for the diagnosis of osteoporosis [1].Objectives:To investigate the effectiveness of the T-score values provided by REMS scans at FN and LS in the identification of frail patients at risk for osteoporotic fractures and to compare the performance of REMS with the dual-energy X-ray absorptiometry (DXA) one. Methods:The patients underwent DXA and REMS scans at FN and at LS. Five clusters of fractures occurred during a median 3.5-year follow-up were identified whether involving the upper limb (forearm, elbow, humerus, wrist, hand), lower limb (tibia, ankle, metatarsus), thorax (shoulder blade, shoulder, rib), hip (femur or pelvis bones), or vertebrae. The ability of REMS and DXA T-score values to assess the incidence and site of fractures was evaluated through an analysis of covariance. Results:Seven hundred twenty-one Caucasian women were enrolled. Ninety-five fractures occurred, in particular 41 at upper limb, 16 at hip, 15 at thorax, 14 at lower limb, 9 at vertebrae. Patients characteristics are reported in table. Considering subcategories of fractured patients, there were not statistically significant differences for age, height, weight and BMI.In the analysis of covariance including age and BMI as covariates, the difference of T-score values between fractured and non-fractured patients is statistically significant for REMS and DXA at both sites.Lower FN T-score values were found for patients with fractures at hip or vertebra with respect to non-fractured patients both for REMS and DXA (p<0.001). Considering LS T-score, lower values were found for patients with fractures at hip, vertebra or upper limb with respect to non-fractured patients both for REMS and DXA (p<0.001, Figure). Conclusion:REMS T-score measured at axial sites is an effective parameter for identification of patients at the risk of incident fragility fractures, in particular occurring at hip, vertebra or upper limb in a population-based sample of female subjects.References:[1]Diez-Perez, Aging Clin Exp Res 2019;31(10):1375–89Table 1.Baseline patient characteristics, expressed as median (25th– 75thpercentiles).Patients with incident fragility fracturePatients without incident fragility fracturep-value*Age [years]70 (60-73)59 (54-64)<0.001Height [cm]159 (155-164)160 (156-165)0.08Weight [kg]63 (58-70)62 (57-69)0.42BMI [kg/m2]24.97 (23.13-26.86)24.24 (22.22-26.59)0.04FN REMS T-score-2.3 (-2.8 – -1.7)-1.8 (-2.3 – -1.1)<0.001FN DXA T-score-2.2 (-2.8 – -1.6)-1.7 (-2.3 – -1.1)<0.001LS REMS T-score-3.0 (-3.5 – -2.0)-2.0 (-2.8 – -1.0)<0.001LS DXA T-score-2.8 (-3.4 – -1.8)-1.9 (-2.7 – -1.0)<0.001* Wilcoxon ranksum testFigure.Boxplot of the distribution of T-score values estimated REMS and DXA at FN and LS among patients without incident fragility fracture and patients with incident fragility fractures at different sites.Note:G. Background:Schnitzler’s syndrome is an autoinflammatory disease characterized by monoclonal gammopathy and recurrent episodes of urticaria accompanied by clinical and laboratory signs of acute inflammation. Interleukin (IL)-1 inhibitors proved to be useful in the treatment, but data on long-term safety and efficacy of these agents are sparse. Objectives:To evaluate the retention rate of IL-1 inhibitors in patients with Schnitzler’s Syndrome. Methods:Retrospective analysis of an Italian multicenter cohort (9 Centers). All patients fulfilled Strasbourg diagnostic criteria. Data are expressed as median [IQR].Results:We identified 15 patients (8 females, 7 males) who received a total of 24 treatment courses with IL-1 inhibitor treatment (16 anakinra and 8 canakinumab) between January 2001 and December 2019, with a median treatment duration of 19 months [8.5-51.3]. Median age at diagnosis was 64.0 years [56.0-72.5] and median follow up was 5.0 years [2.0-8.0]. Before the biological treatment, all patients were treated with corticosteroids and 11 with at least one conventional synthetic disease-modifying antirheumatic drug (csDMARD): methotrexate (5), colchicine (5), cyclosporine (3), azathioprine (1), mycophenolate mofetil (1), cyclophosphamide (1).Fifteen patients received 16 courses of Anakinra, which was the 1st line biological treatment in 14 patients. Seven patients continued it with benefit, while 7 patients discontinued it: 3 for secondary inefficacy; 3 for adverse events (2 injection site reactions, 1 severe allergic reaction); 1 for secondary inefficacy and leukopenia. Anakinra was used as 2nd line treatment in 1 case (after tocilizumab failure); in 1 patient anakinra was resumed after temporary discontinuation and an attempt with infliximab. One patient died for multiple myeloma progression while on treatment with anakinra. The median duration of the courses with anakinra was 20.0 months [6.0-58.3].Seven patients received 8 courses of canakinumab (150 mg/8weeks in 5 cases and 150 mg/4weeks in 3). In 5 cases the drug was administered as 2nd line biological treatment (after anakinra failure) and in 2 cases as 3rd line treatment (1 after tocilizumab and anakinra failures and 1 after anakinra and adalimumab failure). In 1 patient, it was resumed after temporary discontinuation and an attempt with etanercept. One patient died while on treatment with canakinumab due to a presumably unrelated adverse event. The median duration of canakinumab treatment courses was 19.0 months [13.5-31.0].At last follow-up visit, all patients were on treatment with an IL-1 inhibitor: 8 with anakinra (7 at the dosage of 100 mg/day, 1 at the dosage of 200 mg/day) and 7 with canakinumab (2 at the dosage of 150 mg/8 weeks, 4 at the dosage of 150 mg/4 weeks and 1 at the dosage of 300 mg/4 weeks). Notably, in 3 patients the dosage of canakinumab was increased since the start of the treatment.Among 9 patients who were on treatment with prednisone at the start of the last IL-1 inhibitor, the prednisone median dose was 12.5 mg/day [10.0-18.8] while at the last follow-up visit it was 5.0 mg/day [0-7.5] (p= 0.02).The retention rate of IL-1 inhibitors was 73.4% [SE 9.4] at 1 year and 63.6% [SE 10.4] at 2 years (Figure 1a). There was no significant difference between the retention rate of anakinra (at 1 year: 67.0% [12.2]; at 2 years: 59.6% [12.9]) and canakinumab (at 1 year: 85.7% [13.2]; at 2 years 71.4% [17.1]) (log-rank test: p=0.41) (Figure 1b).Figure 1.a) Retention rate of IL-1 inhibitors (24 courses); b) Retention rate of canakinumab (8 courses) and anakinra (16 courses). Conclusion:In this multicentric cohort of patients affected by Schnitzler’s syndrome, the treatment with IL-1 inhibitors as 1st, 2nd or 3rd line biological treatment permitted a good disease control and corticosteroid reduction in patients who did not respond to csDMARDs and/or to prior other biological DMARDs. The optimal dosage of these drugs needs to be tailored for every patient.Acknowledgements:AIDA NetworkDisclosure of Interests:Francesca Crisafulli: None declared, Antonio Vitale: None declared, Carla Gaggiano: None declared, Lorenzo Dagna: None declared, Giulio Cavalli Speakers bureau: SOBI, Novartis, Paid instructor for: SOBI, Novartis, Consultant of: SOBI, Novartis, Rolando Cimaz: None declared, Ombretta Viapiana: None declared, Florenzo Iannone: None declared, Giuseppe Lopalco: None declared, Roberto Bortolotti: None declared, Masen Abdel Jaber: None declared, Carlomaurizio Montecucco: None declared, Sara Monti: None declared, Silvia Balduzzi: None declared, Giacomo Emmi: None declared, Paolo Airò: None declared, Franco Franceschini: None declared, Luca Cantarini Speakers bureau: SOBI, Novartis, Paid instructor for: SOBI, Grant/research support from: SOBI, Novartis, Micol Frassi: None declared

Abstract Introduction: Cyclin-dependent kinase 4/6 inhibitors (CDK4/6i) in combination with endocrine therapy (ET) are the standard first-line treatment for patients with hormone receptor-positive, human epidermal growth factor receptor 2 (HER2)-negative advanced breast cancer (HR+/HER2- aBC). HER2-low BC, which is defined by an IHC score for HER2 of 1+ or 2+ with negative ISH assay, accounts for more than half of all HR+/HER2- aBC cases, and it is associated with remarkable clinical benefit from the novel anti-HER2 antibody drug conjugate (ADC) trastuzumab-deruxtecan. Evidence on the prognostic impact of HER2-low status is controversial in both limited-stage and advanced BC. Here, we sought to investigate the possible prognostic relevance of HER2-low status in a population of aBC patients treated with CDK4/6i plus ET. Methods: We conducted a retrospective-prospective study in six Italian Cancer Centers to investigate the impact of HER2 status (low vs. 0) on the progression-free survival (PFS) and overall survival (OS) of consecutive HR+/HER2- aBC patients treated with CDK4/6i plus ET (aromatase inhibitors or fulvestrant) as a first-line therapy. In the main study analysis, we considered HER2 status in the last tumor assessment (i.e., primary tumor, or, when available, a metastatic lesion). We also performed a subgroup analysis including only patients with HER2 status evaluation in a metastatic lesion collected before CDK4/6i plus ET therapy initiation. The association between HER2 status (low vs. 0) and PFS or OS was evaluated using log-rank test and Cox regression modeling. Results: We evaluated 767 consecutive HR+/HER2- aBC patients treated with CDK4/6i plus ET between January 2017 and January 2022. Of these, 436 patients (56.8%) received CDK4/6i plus ET as a first-line therapy, and they were included in this analysis. Median age was 63 years (range 27-87), and 362 patients (83.0%) were postmenopausal. The majority of patients were treated with palbociclib (68.3%), while 91 (20.9%) and 47 (10.8%) patients received ribociclib and abemaciclib, respectively. Regarding HER2 status, 269 (62.9%) patients had HER2-low tumors, while 159 (37.1%) patients had HER2-0 neoplasms. HER2-low status was associated with significantly lower PFS when compared to HER2-0 status [median PFS (mPFS) 23.6 vs. 32.3 months, respectively; p=0.014]. HER2-low status was also associated with significantly worse OS (mOS 48.7 vs 58.3 months, respectively; p=0.025). These results were confirmed in multivariable models adjusting the impact of HER2 status for clinically-relevant covariates, namely estrogen receptor status, Ki-67, age, number of metastatic sites, presence of liver metastases, disease free interval, ECOG Performance Status. In this analysis, HER2-low status, compared with HER2-0 status, was independently associated with worse PFS [adjusted Hazard Ratio (aHR): 1.62; 95% confidence interval (CI): 1.17-2.24; p< 0.01] and OS (aHR: 1.74; 95% CI: 1.09-2.76; p=0.019). Subgroup analysis conducted in the subset of 256 patients with available metastatic tumor samples collected before CDK4/6i plus ET initiation confirmed that HER2-low status (n=157), when compared to HER2-0 status (n=99), was independently associated with worse PFS (mPFS 24.5 vs 35.2 months, p=0.01; aHR 2.07; 95% CI: 1.28-3.34, p< 0.01) and worse OS (mPFS 48.7 vs 72.3 months, p=0.027; aHR 3.12; 95% CI 1.44-6.77, p< 0.01). Conclusions: This multicenter Italian study revealed that HER2-low status has independent, negative prognostic value in patients with HR+/HER2- aBC treated with CDK4/6i plus ET in the first-line setting. Our results suggest that HER2-low status might be associated with different clinical benefit from standard anticancer therapies in specific clinical settings. The definition of treatment algorithms also taking into account HER2 status is a clinical priority in patients with HR+/HER2- aBC. e4519020The mark of violence is present in today's society. It is in this context that human rights education is fundamental. The research, of qualitative nature, aimed to apprehend and analyze, in compliance with human rights, the problematized aspects with regard to violence against women, supporting on the structured interview, projective modality, which used the device of photo reproduction so that the interviewees could talk, critically, about what they saw. For discussion, two textual productions related to the reading of two reproductions that “spoke” of violence against women, were considered and submitted to interpretative analysis. The texts expressed contradiction to what was put in the photo reproduction, problematizing the violations of rights and, at the same time, considering ethics, values, and human rights as fundamental components to the exercise of citizenship and, still, evoking the dignity of the human person for the constitution of scenarios sensitive to humanity. Among the least investigated personalities of the milanese erudition between the 18th and 19th century, we find Carlo Amoretti (1741-1816): polygraph, naturalist, Winckelmann’s first Italian translator and Leonardo scholar as prefect of the Ambrosiana Library. Starting from the manuscripts kept at the Istituto Lombardo – Accademia di Scienze e Lettere in Milan, this paper aims to shed a first light on the observations of Amoretti around 14th and 15th century artworks in the Lombard territory. In particular, his travel diaries provide unpublished insights into the debate on medieval and proto-Renaissance art that involved some of the leading antiquarians of the 19th century, with whom Amoretti himself was in contact: from Giuseppe Bossi to Seroux d’Agincourt, from Tommaso degli Obizzi to Stefano Borgia.
