The aim of this study is to find out what music students think about playing in music lesson and their reflections on the importance of genre in music education. The study covers music education in high school. I have used both of quanititative and qualititative methods in my study. The quantitative method vas in the form of queststionnaires and qualitative method in the form of my observations above all in the form of definitions, conceptions and theoretical constructions. The result shows that play the instuments is very important for students and rock and popmusic have a high status in music education. The majority of students prefer more to play the instruments than their have theory in the lesson. The most of music students in high school believ that rock and popmusic is very important things in music education. One importans aspect is that teacher’s commitment to the students musical development. Keywords: genre, ensemble, music education, play the instruments.

Повний текст джерелаIn recent years, public schools and higher education institutions in Texas have placed a growing emphasis on mariachi music programs. The purpose of this study was to gather data regarding the status of mariachi programs and the pedagogical practices and activities of selected mariachi instructors in kindergarten through 12th grade (K12) schools in Texas. The researcher examined the pedagogical practices, content knowledge bases, and specific skill sets that selected mariachi instructors considered to be important determinants of effectiveness in teaching mariachi music. Descriptive data about the selected mariachi instructors' class activities was also collected. The sample used for this study comprised 14 selected mariachi music instructors. The sampling process included compiling a list of mariachi programs throughout the state of Texas, organizing the list according to the seven All-State Area alignments recognized by the Texas Music Educators Association, and selecting two instructors-one from a junior high school and one from a high school-in each All-State Area. The method for gathering information on pedagogical practices and class activities included tape-recorded interviews and on-site observations ofthe selected mariachi instructors. Data from both the interviews and the observation sessions were aggregated for further analysis and for the development of categories and themes, as appropriate. The study and its reported outcomes constitute a status of mariachi programs across Texas. Data gathered from selected mariachi instructors provided information regarding program content and program scheduling in K12 schools and an overview of the instructors' backgrounds and bases of knowledge of the mariachi genre. Further, specific skills and skill sets regarding performance practices of the mariachi ensemble are discussed. It is anticipated that this accumulation of pedagogical information will be used by music educators currently involved in the mariachi genre and by other music educators in the development of their programs.

Resumo : Esta dissertação aborda o programa regional ¿Terra da Gente¿, da EPTV (Emissoras Pioneiras de Televisão), para uma reflexão sobre a construção da chamada pesca esportiva como prática ecologicamente correta e integrada ao desenvolvimento sustentável. Para tanto, foram analisados programas televisivos entre os anos de 2005 e 2006, além da versão na Internet e da Revista ¿Terra da Gente¿, bem como consultadas diversas fontes (livros, jornais, revistas e sites). O trabalho apresenta um breve histórico do surgimento das emissoras de TV no Brasil, especialmente da ¿Rede Globo¿ e a sua programação regional, tangenciando a discussão sobre o global e o local
Abstract : This thesis approaches the regional program ¿Terra da Gente¿ of the EPTV (Emitting Pioneers of Television) for reflection on the conception of recreational fishing as an environmentally safe practice integrating it to a sustainable development. Повний текст джерелаDuring the course of the work described in this thesis, ESTsequencing project was initiated in woody plant poplar forlarge scale gene discovery in a tree specie. Since most of thetree genomes are considerably large in size, EST sequencing wasconsidered to be a cost effective method for gene discovery.Initially, EST sequencing was performed on cDNA librariesprepared with mRNA isolated from woody tissues. This was firstsuch attempt to identify genes associated with the process ofwood formation on a large scale. At this stage, most of thesequence data was manually curated and a database wasestablished for storage of sequence information and making itavailable to biologist. One of the observations of this initialwork on sequencing of woody tissue cDNA libraries was the largenumber of ESTs displaying no similarity to any known gene inthe database prompting the suggestion regarding the existenceof tree specific genes. Subsequently, over 130,000 ESTs weresequenced from 19 different cDNA libraries and a set of perlscripts were written to facilitate efficient and automatedprocedure for data handling. In this automated pipeline, theoutput from sequencing machines as raw sequences would besubjected to quality control, blast and annotation processeseventually leading to storage of this data in an easilyaccessible, internet based database. The data from sequencingof cDNA libraries from leaves at two different stages ofdevelopment, namely, actively growing young poplar leaves andsenescing leaves was used to identify the changes in geneexpression occurring during the induction of senescence. Inorder to identify genes that may be involved in wood formationand to investigate whether there maybe tree specific genes, abioinformatics approach was taken by comparing EST compositionof cDNA libraries prepared from woody tissues from poplar,birch and pine and compared with the ESTs and genomic sequenceof Arabidopsis which under normal circumstances does not formwood. This comparison lead to the conclusion that there may befew if any tree specific genes finally using this data we couldidentify a set of genes whose expression was significantlyupregulated in woody tissues. Finally assembly of 130,000 ESTswas performed to obtain a glimpse into the genetic compositionof trees and this data was compared with that of Arabidopsisgenomic sequence to get a better understanding of thesimilarities and differences between trees and annuals at themolecular level.

The ability to synthesize custom de novo DNA constructs rapidly, accurately, and inexpensively is highly desired by researchers, as synthetic genes and longer DNA constructs are enabling to numerous powerful applications in both traditional molecular biology and the emerging field of synthetic biology, from the synthesis of large sets of novel proteins to the complete re-writing of bacterial genomes. However, the current cost of de novo synthesis--driven largely by reagent and handling costs-is a significant barrier to the widespread availability of such technology. The use of microfluidic technology greatly reduces reaction volumes and corresponding reagent and handling costs. Additionally, microfluidic technology enables large numbers of complex reactions to be performed in parallel, while facilitating the automation and integration of multiple processes in a single device. While microfluidic devices have been used to miniaturize a variety of chemical and biological processes, the benefits of such devices have yet to be realized in the area of de novo DNA synthesis. This thesis reports the first demonstration of gene synthesis in a microfluidic environment. A variety of DNA constructs with sizes as large as 1 kb were fabricated in parallel in a multi-chamber microfluidic device at volumes one to two orders of magnitude lower than those utilized in conventional bench top techniques. This thesis also reports on progress toward the direct synthesis of genes from hybrid microfluidic-DNA microarray devices, the integration of microfluidic gene synthesis with on-chip protein synthesis, and the microfluidic hierarchical synthesis of long DNA molecules.

The regulation of gene expression underlies the morphological, physiological, and functional differences between human cell types, developmental stages, and healthy and disease states. Gene regulation in eukaryotes is controlled by a complex milieu including transcription factors, microRNAs (miRNAs), cis-regulatory DNA and RNA. It is the quantitative and combinatorial interactions of these regulatory elements that defines gene expression, but these interactions are incompletely understood. In this thesis, I present two new methods for determining the quantitative specificity of gene regulatory factors. First, I present a comparative genomics approach that utilizes signatures of natural selection to detect the conserved biological relevance of miRNAs and their targets. Using this method, I quantify the abundance of different conserved miRNA target types, including different seed matches and 30-compensatory targets. I show that over 60% of mammalian mRNAs are conserved targets of miRNAs and that a surprising amount of conserved miRNA targeting is mediated by seed matches with relatively low efficacy. Extending this method from mammals to other organisms, I find that miRNA targeting rules are mostly conserved, although I show evidence for new types of miRNA targets in nematodes. Taking advantage of variations in 30 UTR lengths between species, I describe general properties of miRNA targeting that are affected by 30 UTR length. Finally, I introduce a new, high-throughput assay for the quantification of transcription factor in vitro binding affinity to millions of sequences. I apply this method to GCN4, a yeast transcription factor, and reconstruct all known properties of its binding preferences. Additionally, I discover some new subtleties in its specificity and estimate dissociation constants for hundreds of thousands of sequences. I verify the utility of the binding affinities by comparing to in vivo binding data and to the regulatory response following GCN4 induction.

Biological processes from differentiation to disease progression are governed by gene regulatory mechanisms. Currently large-scale omics and imaging data sets are being collected to characterize gene regulation at every level. Such data sets present new opportunities and challenges for extracting biological insights and elucidating the gene regulatory logic of cells. In this thesis, I present computational methods for the analysis and integration of various data types used for cell profiling. Specifically, I focus on analyzing and linking gene expression with the 3D organization of the genome. First, I describe methodologies for elucidating gene regulatory mechanisms by considering multiple data modalities. I design a computational framework for identifying colocalized and coregulated chromosome regions by integrating gene expression and epigenetic marks with 3D interactions using network analysis.
Then, I provide a general framework for data integration using autoencoders and apply it for the integration and translation between gene expression and chromatin images of naive T-cells. Second, I describe methods for analyzing single modalities such as contact frequency data, which measures the spatial organization of the genome, and gene expression data. Given the important role of the 3D genome organization in gene regulation, I present a methodology for reconstructing the 3D diploid conformation of the genome from contact frequency data. Given the ubiquity of gene expression data and the recent advances in single-cell RNA-sequencing technologies as well as the need for causal modeling of gene regulatory mechanisms, I then describe an algorithm as well as a software tool, difference causal inference (DCI), for learning causal gene regulatory networks from gene expression data.
DCI addresses the problem of directly learning differences between causal gene regulatory networks given gene expression data from two related conditions. Finally, I shift my focus from basic biology to drug discovery. Given the current COVID19 pandemic, I present a computational drug repurposing platform that enables the identification of FDA approved compounds for drug repurposing and investigation of potential causal drug mechanisms. This framework relies on identifying drugs that reverse the signature of the infection in the space learned by an autoencoder and then uses causal inference to identify putative drug mechanisms.

To better understand the tissue-specific regulation of chromatin state in cell-fate determination and development, we defined the tissue-specific expression of all 36 lysine methyltransferase (KMT) genes by endogenous mRNA detection in C. elegans. We found that most KMTs are expressed in only one or two tissues and that the germline is the tissue with the most general KMT expression. We discovered that the germline-expressed C. elegans ortholog of mammalian PRDM9, SET-1 7, promotes fertility through gene regulation in primary spermatocytes. SET-17 drives transcription of spermatocyte-specific genes from four genomic clusters to promote spermatid production. SET-1 7 is concentrated in stable, chromatin-associated nuclear foci at actively transcribed gene clusters, which we term spermatocyte transcription bodies. Our results identify the spatially restricted function of a PRDM9 ortholog in spermatocyte transcription and we propose that the spatial organization of chromatin factors might be a conserved mechanism in tissue-specific control of transcription.

Transmission of information from DNA to RNA to protein underlies the core of modem life forms. The advance in sequencing and genetic technologies has revolutionized the study of molecular biology, genetics and developmental biology enabling delineation of biological processes in unprecedented details. Through the study of epigenetics and posttranscriptional regulation of gene expression by high-throughput sequencing technologies in several biological processes, namely embryonic stem cells, somatic reprogramming, erythroid differentiation, epithelial-mesenchymal transition and cancer metastasis, this thesis work has identified novel players and regulatory mechanisms underlying these developmental processes and diseases. Furthermore, an attempt to engineer CRISPRzymes - protein fusions of RNA-guided DNA binding dCas9 - will enable experiments to directly test biological processes at defined genomic loci and expands the toolbox for synthetic biology and potentially opens up opportunities for novel therapeutics.

O presente trabalho tem por objetivo o estudo dos gêneros discursivos na alfabetização, por meio da análise de parte do material do programa “Ler e Escrever” da Secretaria Estadual de Educação do Estado de São Paulo. A preocupação central está relacionada à como o conceito de gênero é apresentado ao professor alfabetizador e de que forma articulam-se às práticas de alfabetização. Os conceitos de alfabetização e letramento vêm sendo amplamente discutidos e a utilização de textos em sala de aula é, atualmente, amplamente aceita. Entretanto, a questão dos gêneros aparece de forma pouco delineada nos materiais voltados para o ensino nas séries iniciais do Ensino Fundamental. Frente a isso, optou-se pela análise do material voltado ao professor do programa Ler e Escrever da Secretaria Estadual de Educação do estado de São Paulo, dada sua representatividade quantitativa, uma vez que é utilizada pela rede estadual, e por meio de convênio em diversas redes municipais do estado de São Paulo. Este programa tem como proposta a não dissociação entre alfabetização e letramento, por meio do uso de textos na alfabetização. Assim, buscamos verificar a concepção de gênero que subjaz a este material, o tratamento que os diferentes textos recebem no ensino, bem como sua articulação aos conteúdos específicos da alfabetização. A partir da análise realizada, pode-se perceber que a definição de gênero que orienta o trabalho do professor neste programa está centrada na forma que o mesmo assume, sendo desconsiderada a concepção de gênero proposta por Bakhtin, em que a situação de enunciação é fundamental para a compreensão do gênero, expresso como unidade entre forma de enunciação, forma de comunicação e o tema. Percebe-se ainda que, em virtude das bases teóricas que orientam o programa, há uma ausência de trabalho sistemático com os conteúdos específicos da alfabetização.
This study aims to investigate speech genres in literacy, through the analisys of parto f the “Ler e Escrever” program from State of São Paulo. The main concerns are related to how the concept of genre presents itself in literacy classes and also in which way it is related to literacy practices. Diferent concepts of literacy have been discussed and the use of texts in lietracy classroons has been fully accepted. However, the concept of genre appeas barely delimitated in materials focused on literacy. Due to these facts, it has been choosen to analize the teacher guide of the program named “Ler e Escrever” of the Educational State Secretary of the state of São Paulo, since it is widely used and very representative in numbers once it is adopted by state schools and also by some municipal ones. The purpose of this program is to teach language by focusing in texts and in the aspects of writen language without breaking them appart. Our goal is to investigate the genre concept beneath this material and also how it relates to literacy teaching. As a result of this study, one could note that the definition of genre that guides the material and the teacher practices is focused in the form assumed by each genre. In this way, it is a concepction that dismiss the unit proposed by Bakhtin, between form of comunication, form of enunciation and theme. It is also noted that, due to the theoretical bases of the purpose, there is na absence of sistematic teaching of the specific knowledge related to reading and writing skills.

This study dealt with the process of writing a personal letter by students of Youth and Adult’s Education program. Its main goal was to understand how the Portuguese Language teacher can contribute to the improvement of the writing process in the Youth and Adult’s Education, by writing a personal letter. These were the specific goals of this study: to identify and analyze the main difficulties presented by students of Youth and Adult’s Education program when writing a personal letter; to enable the student of the Youth and Adult’s Education program the understanding that the improvement of their writing will be relevant throughout their personal and professional lives; to evelop a writing strategy which can be applied in the classroom, aiming at a facilitator path between teacher and student in the process of writing a personal letter. The theoretical references were constituted by three axes: Youth and Adult’s Education program, Textual Genres and the personal letter based on authors such as Antunes (2009), Arroyo (2011), Cavalcante (2013), Creswell (2007), Demo (2008), Dolz, Noverraz and Schneuwly (2004), Ferraro (2009), Franco (2005), Freire (1993), Lüdke (1986), Marcuschi (2002, 2008, 2010), Oliveira (2014), Paiva (2015), Schneuwly (1994) and Vieira (2009), and also on papers such as the Portuguese National Teachers' Guide (1997/98/01) and the Law on Guidelines and Bases of National Education (9,394 / 96). As for its methodology, the research was structured in a qualitative approach, through a field study: the participant research, using instruments or collecting data, such as: the participant observation, the conversation wheel and the questionnaire. Data analysis took place through content analysis. Five students from the Youth and Adult’s education program took part in the study: 4 female and 1 male, all of them from the second segment, of a municipal school of Primary Education in Mamanguape-PB. After the theoretical constructions and the methodological path of the research, we presented the analysis of the productions of a personal letter, as well as the texture between the goals and the data collected in the research. The results of the analysis indicated that the written production of a personal letter, developed through a didactic sequence created by Dolz, Noverraz and Schneuwly (2004), enabled the participants of the research to acquire linguistic-discursive knowledge that is necessary for the construction of the genre personal letter, as well as helped them recognize and at the same time overcome the main difficulties they presented during the process of writing a personal letter. Finally, I present as a suggestion to teachers the systematization of a possibility of working with the personal letter in the Youth and Adult’s education program.
Este estudo tratou do processo de escrita da carta pessoal por estudantes da Educação de Jovens e Adultos. Apresentou como objetivo geral compreender como o/a professor/a de Língua Portuguesa poderá contribuir para melhoria do processo de escrita na Educação de Jovens e Adultos, fazendo uso da escrita da carta pessoal. Foram objetivos específicos desse estudo: identificar e analisar as principais dificuldades apresentadas por estudantes da Educação de Jovens e Adultos na produção da carta pessoal; possibilitar ao (à) estudante da Educação de Jovens e Adultos a compreensão de que a melhoria da sua escrita será relevante ao longo de sua vida pessoal e profissional; desenvolver uma estratégia de escrita que possa ser aplicada em sala de aula, visando a um caminho facilitador entre professor e estudante na produção escrita da carta pessoal. As referências teóricas se constituíram a partir de três eixos: a Educação de Jovens e Adultos, os Gêneros Textuais e a Carta pessoal fundamentado em autores e autoras como: Antunes (2009), Arroyo (2011), Cavalcante (2013), Creswell (2007), Demo (2008), Dolz, Noverraz e Schneuwly (2004), Ferraro (2009), Franco (2005), Freire (1993), Lüdke (1986), Marcuschi (2002; 2008; 2010), Oliveira (2014), Paiva (2015), Schneuwly (1994), Vieira (2009), e também em documentos como: nas orientações dos PCNs de Língua Portuguesa (1997/98/01) e a Lei de Diretrizes e Bases da Educação Nacional 9.394/96. No aspecto metodológico, a pesquisa foi estruturada na abordagem qualitativa, por meio de um trabalho de campo na pesquisa participante, fazendo uso de instrumentos ou procedimentos de coleta de dados como: a observação participante, a roda de conversa e o questionário. A análise de dados ocorreu por meio da análise de conteúdo. Foram participantes da pesquisa 05 estudantes da Educação de Jovens e Adultos: 04, do sexo feminino e 01 do sexo masculino, todos do segundo segmento, pertencentes a uma Escola Municipal de Ensino Fundamental do município de Mamanguape- PB. Após as construções teóricas e o caminhar metodológico da pesquisa, foram apresentadas às análises das produções da carta pessoal, bem como a tessitura entre os objetivos e os dados emanados da pesquisa. Os resultados das análises apontaram que, a produção escrita da carta pessoal, elaborada por meio da sequência didática desenvolvida por Dolz, Noverraz e Schneuwly (2004), possibilitou aos (às) participantes da pesquisa a aquisição de conhecimentos linguístico-discursivos necessários à construção do gênero carta pessoal, bem como ajudou-os(as) a reconhecer e ao mesmo tempo superar as principais dificuldades por eles apresentadas durante o processo de escrita da carta pessoal. Por fim, apresento como sugestão aos (às) professores (as) uma sistematização de uma possibilidade de proposta de trabalho com a carta pessoal na Educação de Jovens e Adultos.

ROS poden reaccionar amb diferent components cel•lulars, resultant en peroxidació d’àcids grassos, oxidació de proteïnes i danys al DNA. Els organismes unicel•lulars estan sotmesos a una àmplia varietat de canvis ambientals i a l’exposició de compostos tòxics. En resposta a l’estrés oxidatiu provocat pel peròxid d’hidrogen, S. pombe activa diferent vies de senyalització segons la severitat de l’estrés sofert, dut a terme per dos factors de transcripció, Pap1 i el bZIP Atf1 sota el control aquest últim de la MAPK. En el llevat S. pombe, Pap1 i Sty1 constitueixen un paper clau per lat resposta a l’estrés oxidatiu. La MAPK Sty1 s’activa sota l’efecte de diferent estressos, aixi com l’estrés oxidatiu o l’osmòtic. La modulació de la resposta gènica és un dels principals punts en aquesta resposta, hem centrat el treball d’aquesta tesis en la caracterització de la ruta de Sty1 com a sensor de l’estrés oxidatiu així com els diferents events moleculars que activen el programa transcripcional. També hem estudiat el paper de la traducció per l’Elongato en la inducció de la resposta a estrés.

Повний текст джерелаThis linguistic study aims to find, highlight and discuss how different genre features, discourses and identities are constructed in text based self-presentations of a customer club. The purpose is to see how these presentations are produced in the quest to recruit, retain and promote loyal customers. The study involves eight companies, whose customer clubs has been presented textually on each company's website. To find important features in the texts, the systemic-functional grammar is applied, and through interpersonal- and ideation text analyzes, the genre features and discourses can be identified. Parallels are also drawn to the genre features connection and creation in works of identity. A comparison to the advertising genre is also made. Based on this, a discussion of what linguistic methods customer clubs use to create a desirable identity both in themselves and their members, becomes a central part in the analysis. The result shows that there is a clear structure, in which customers are offered exclusive benefits and a coveted identity in exchange for enrolling in the club. In the presentations, members play a passive role that effortlessly gets or earns all the listed benefits while the customer clubs on the other hand, are portrayed as an active and resourceful distributor. A sales discourse also emerges here, which contrasts with a hidden consumption discourse. Negative aspects, such as costs and conditions, are made invisible and the presentation mainly focuses on describing the positive experience and all the benefits of being a member of the customer club.

Gene therapy has been heralded as a possible approach to a variety of diseases and conditions, ranging from cancer and heart disease to blindness and neurodegenerative diseases. However, progress in gene therapy requires a delivery system that can control when and where the therapeutic proteins will be generated. Our study was performed to determine the feasibility of attaching heat-inducible promoters to genes of interest in order to control activation of the gene in vivo via ultrasound-induced hyperthermia monitored by MRI thermometry. We first demonstrated that gene therapy-mediated gene expression could be spatially and temporally controlled with this method. Further studies were subsequently performed to determine if the activation of a particular heat-inducible gene, Hsp70b, could be quantified and predicted a priori during hyperthermia, thus allowing advance knowledge of the protein levels over time. Experiments indicated that as the temperature and duration of a hyperthermic shock increased, peak expression levels of Hsp70b mRNA also increased until a saturation level was reached. In addition, as the duration of a hyperthermic shock increased, the time during which Hsp70b mRNA remained elevated also increased. Most significantly, a correlation was found between total Hsp70b mRNA production generated by thermal shock and thermal dose, a predictor of dose often used in hyperthermia therapies. The relationship found between total Hsp70b mRNA production and thermal dose suggests that a real-time predictive model of therapeutic protein dose kinetics after ultrasound-induced hyperthermia for gene therapy is feasible. However, the creation of such a model would require further precision experimentation for which ultrasonically-induced hyperthermia is not suited. A final study was performed and found that Hsp70b was not activated by the mechanical stress caused by ultrasound. These results confirm that a predictive model applicable to ultrasonically-induced hyperthermia could be developed using waterbath techniques that will allow tighter control of temperature.

The program was transmitted live on the mornings by Radio Difusora, being presented by broadcaster Manuel Ferreira Canabarro, the Gauchinho, who was also mainly responsible for organizing the parties in association with the communities that were done in the afternoon, after the live broadcast by radio program Nossa Terra, Nossa Gente. The event lasted from 1982 to 2000 and occurred in the municipality of Marechal Cândido Rondon, where is the Radio Difusora, apart from Mercedes, Nova Santa Rosa, Quatro Pontes, Toledo, Ouro Verde do Oeste, São José das Palmeiras, Entre Rios do Oeste, Pato Bragado, Santa Helena, Terra Roxa, Guaíra, and the districts of these cities. Therefore, we discuss the various views of the subjects present in the radio program and at the parties, like the participants, organizers and musicians. Noteworthy is the role performed by the Radio Difusora, acting as an agent, very present in the daily region where the parties and the program were held. Therefore, discuss some issues on broadcasting in the town of Marechal Cândido Rondon, and especially, the objectives of the creation and operation of the program Nossa Terra, Nossa Gente. Also discussed will be the LP produced by Radio Difusora with songs of singers who presented the program. Among the sources used to research beyond the narrative and the LP, makes use of features of the Frente Ampla de Notícias newspaper and O Presente
O presente estudo aborda as narrativas das experiências vividas nos espaços do programa radiofônico itinerante Nossa Terra, Nossa Gente e das festas realizadas após a sua transmissão. O programa era transmitido aos domingos de manhã, ao vivo, pela Rádio Difusora, apresentado pelo radialista Manuel Ferreira Canabarro, o Gauchinho, que era, também, o principal responsável pela organização das festas realizadas, em associação com as comunidades, nas tardes de domingo, após a transmissão ao vivo pela rádio. O evento perdurou de 1982 a 2000 e acontecia no município de Marechal Cândido Rondon, onde se situa a Rádio Difusora, além dos municípios de Mercedes, Nova Santa Rosa, Quatro Pontes, Toledo, Ouro Verde do Oeste, São José das Palmeiras, Entre Rios do Oeste, Pato Bragado, Santa Helena, Terra Roxa, Guaíra e nos distritos desses municípios. Повний текст джерелаIn Bioinformatics, as in many fields of science, it is often necessary to analyze collections of data sets. These data sets are often composed of matched values for an independent and a dependent variable. For example, a chemist may examine the absorption spectra of a group of compounds, a sociologist might examine life expectancy versus income in different communities, or a bioinformatician might examine expression of individual nucleotides in a genome by an organism. In these examples, the independent variables are frequency, income, and position in the genome, respectively, and the dependent variables are absorption, life expectancy, and expression, respectively. Previously, such a group of data set may have been displayed visually by a collection of individual tiled line graphs, or by superimposing multiple line graphs on the same graph space. The Horizon Graph is a newly developed data visualization method for these sorts of data sets, and outcompetes its alternatives. A Horizon Graph is composed of a series of "bars", each of which displays a single data set. Bars code for the independent variable along the horizontal axis, and for the dependent variable using both color and the vertical axis. These bars are stacked atop one another vertically, resulting in a single plot that enhances a user's ability to recognize patterns within and between the data sets We wrote a program in Java that generates a Horizon Graph. The program accepts a collection of data sets describing mRNA expression of a common genomic region across multiple samples. It then generates a plot that represents the genetic splicing of the genomic region. The program may generate one of two types of Horizon Graph. The first plot type shows genetic splicing for the data sets. The second plot shows the splicing of every data set relative to the splicing of a "control" subset of the data sets. The program also offers a user several other options regarding the produced plot. These include the ability to mark contiguous portions of nucleotide positions. This can be used to mark regions of exogenous sequences. The individual tasks the program had to accomplish to generate a plot from data sets were separated into isolated Java "classes". These "classes" interacted through well-defined inputs and outputs, and were coordinated in another, separate "class". The resulting structure maximizes the ease for future users to use portions of the program to accomplish portions of its behavior in novel contexts. The program was demonstrated on a collection of data sets from the mef2d gene locus, each from a unique tissue type.

Gene therapy is a promising modality for the potential treatment of rare Mendelian diseases. To date a number of high profile proof-of-concept studies within the industry have demonstrated the significant disease-correcting promise of this therapeutic strategy. One of the major hurdles that remains for the commercialization of gene therapies is the lack of efficient manufacturing capabilities for the production of clinical-grade drug substance/drug product. The primary goals for this project were to decrease the biological contamination and cross-contamination risk associated with the biologic manufacturing process for viral gene therapy vectors and to adjust the process in order to optimize commercial profit. The project also included documenting the different existing processes for AAV production and developing a competitive analysis using information from ongoing clinical trials in the industry pipeline. The following process design steps were followed in order to fulfill the project objectives: (1) Define product specifications, analytical needs and market size, (2) Select production platform/process, (3) Collect data and create process flow diagram, (4) Perform material and energy balances, (5) Calculate costs: equipment and consumables, (6) Model the process in a spreadsheet, (7) Carry out sensitivity analyses, (8) Assess cost-effectiveness and risk, and (9) Develop recommendations. Five different AAV production platforms were identified and an AAV gene therapy landscape was generated. Also, the current process that Pfizer is planning to use was documented and an initial market sizing was performed. Finally, all the data necessary to model the process was collected and the cost-effectiveness and biological contamination and cross-contamination risk assessment were completed. This project confirmed that the use of a scalable line of single-use high cell density bioreactors for the production of AAV is cost-effective. This implies that sufficient AAV quantities can be manufactured for preclinical and clinical trials, using the process developed by Pfizer.

Повний текст джерелаThe study about television – heir of Medias like radio and cinema – doesn’t share the same scientific support with its predecessors. Specific TV work groups in Brazil, with Obitel as an example, became more frequent since 2000’s. This media style and aesthetic aimed studies are even rarer. This thesis suggests a starting point to the aesthetic discussion through metaTV, with the prime objective of stablishing an outline to the reflexivity aesthetic within Brazilian TV shows, verifying the possibility to set up a promise of a television genre that still needs to be defined. The reflexivity is, here, theorized from metaTV and conceptualized as an art field irradiating process. Its television form is verified upon a group of Brazilian shows, since 1980’s until 2000’s first decade. From a previous analysis, according to Metiérs and Techniques, Updates and Backstage and Program and Channel Promotion (SPIES, 2004), the corpus of this thesis is defined and constituted by Cena Aberta (REDE GLOBO, 2003), Profissão Repórter (REDE GLOBO, 2006) e No Estranho Planeta dos Seres Audiovisuais (TV FUTURA, 2008 – 2009). From this perspective, work with the possibility that the study will embody Natalie Burtin (1998) idea that metaTV would be the foundress of a television genre, as well as fulfill the aesthetic promise, argued by Virginie Spies (2004). In such verification, an analysis has been made, guided by style activation factors, like: Intermediality (CALDWELL, 1995), Plasticity, Morphology, Presentation Modes and Sounds, inspired by Elizabeth Bastos Duarte (2004). These analysis shows us that there are some arrangements among the possibilities offered by an aesthetic that inherits a big variety of audio-visual language characteristics shared with cinema that still maintain a great verbal language support boundage, wich points the persistent radio influence on television. This ensemble defines its audiovisuality (ROCHA et al, 2013) gives to television a peculiar way that reflexive promise can be accomplished.

The development of self-propagating gene drive containing organisms may have wide-reaching consequences, including for international trade. Given compelling potential applications in disease mitigation and pest management, countries may develop and release these self-propagating products of biotechnology in the near future. Such products will cross international borders through natural organism movement and through trade, potentially altering not only the populations and ecosystems of the country that developed the product, but also those of the countries to which they spread. Political and economic consequences will arise, some of which the World Trade Organization (WTO) will govern. The WTO's Agreement on Sanitary and Phytosanitary Measures (SPS Agreement) is an especially pertinent set of trade rules which permits trade restrictions aimed at protecting the health of a country's citizens or other species only when scientific evidence supports the perception of risk to the health of citizens or other species. In the early 2000s, the WTO evaluated the SPS Agreement for applicability to genetically modified crops, but no such analysis has yet been completed for gene drive containing organisms. In this thesis, I first summarize the status of gene drive development, highlighting divergent techniques and their status in laboratory experiments. I then explore potential scenarios for trade disputes involving gene drive containing organisms, with differing technological and ecological specifications. I examine the text of the SPS Agreement, as well as its past application to trade disputes, to evaluate relevance to gene drive containing organisms. Finally, I formulate suggestions to policy makers and technology developers for preventing international disputes on, and disruption of trade by, gene drive containing organisms.

Toxic algae are a growing concern in the marine environment. One unique marine diatom, Pseudo-nitzschia multiseries (Hasle) Hasle, produces the neurotoxin domoic acid, which is the cause of amnesic shellfish poisoning. The molecular characterization of this organism has been limited to date. Therefore, the focus of this thesis was to identify and initiate characterization of actively expressed genes that control cell growth and physiology in P. multiseries, with the specific goal of identifying genes that may play a significant role in toxin production. The first step in gene discovery was to establish a complementary DNA (cDNA) library and a database of expressed sequence tags (ESTs) for P. multiseries. 2552 cDNAs were sequenced, generating a set of 1955 unique contigs, of which 21% demonstrated significant similarity with known protein coding sequences. Among the genes identified by sequence similarity were several involved in photosynthetic pathways, including fucoxanthin-chlorophyll a/c light harvesting protein and a C4-specific pyruvate, orthophosphate dikinase. Several genes that may be involved in domoic acid synthesis were also revealed through sequence similarity, for example, glutamate dehydrogenase and 5-oxo-L-prolinase. In addition, the identification of sequences that appear novel to Pseudo-nitzschia may provide insight into unique aspects of Pseudo-nitzschia biology, such as toxin production. Genes whose expression patterns were correlated with toxin production were identified by hybridization to a microarray manufactured from 5376 cDNAs. 121 cDNAs, representing 12 unique cDNA contigs or non-redundant cDNAs, showed significantly increased expression levels in P. multiseries cell populations that were actively producing toxin.

The up-regulated transcripts included cDNAs with sequence similarity to 3-carboxymuconate cyclase, phosphoenolpyruvate carboxykinase, an amino acid transporter, a small heat shock protein, a long- chain fatty acid Co-A ligase, and an aldo/keto reductase. These results provide a framework for investigating the control of toxin production in P. multiseries. These transcripts may also be useful in ecological field studies in which they may serve as signatures of toxin production. Prospects for further application of molecular genetic technology to the understanding of the physiology and ecology of P. multiseries is discussed.

The promise of personalized medicine lies in its potential to fundamentally change healthcare. In the past, pharmaceuticals were prescribed on a "one size fits all" basis-patients with certain disease phenotypes were given what were thought to be appropriate drugs. There is growing evidence however that the effectiveness of these drugs may differ by individual and by sub-group; presumably due to fundamental genetic differences in disease and metabolic pathways. Drugs like Herceptin, Gleevec and Iressa are part of an emerging trend in the biopharmaceutical arena of drugs that are accompanied by genetic diagnostic tests and prescribed only for patients with genotypes in which the agents are most effective.

The aging process of the population is a world and Brazilian reality, and has a strong gender component, given the phenomenon of feminization of old age. Although the older population, especially women, are living longer, their growth is not accompanied by public policies that guarantee their rights. Among these, education emerges as a challenge regarding its targeting of women in the old age. Thus, the general and specific objectives of the study were: to analyze the meanings that the old women - participants of an EJA Program in Fortaleza - CE - attribute to education, to understand if this experience influences in their lives, from a gender perspective; Raise the motivations that drive the search for education during old age; To study the challenges that older women face in accessing formal education in relation to the gender issue; Unveiling the memories women have about their previous educational experiences; To see how women understand the EJA Program in which they participate. For this purpose, qualitative research was used, with the aid of bibliographic, documentary and field research, as well as the following techniques for obtaining information: direct observation and unstructured interview based on the Oral History Thematic method. As a field of research, an EJA Program was selected at a Municipal School in Fortaleza, with the participation of 5 (five) old female students from the school and the EJA, whose narratives were interpreted in the light of Hermeneutic-Dialectic. The perceptions, about the return or first access to the studies, were gathered in three (3) empirical categories: leisure / sociability, citizenship / belonging, freedom / autonomy. In this way, the meanings were as varied as possible, but all contributed to the need to expand educational spaces for women, where they can recognize their unique demands throughout all phases of their lives. From this change of thought and, consequently, of paradigm, it is that the school can embrace, democratically, men and women, of different ages, of class belongings the most diverse, of multiple beliefs and nationalities. This deconstruction requires the engagement of groups historically placed outside the right to education, which, recognizing themselves as protagonists, can contribute to the creation of a new societal order and a sociability based on gender equality without any kind of prejudice and / or discrimination.
O processo de envelhecimento da população é uma realidade mundial e brasileira, e que possui um contundente componente de gênero, haja vista o fenômeno de feminização da velhice. Embora a população velha, especialmente as mulheres, esteja vivendo mais, seu crescimento não é acompanhado de políticas públicas que garantam os seus direitos. Dentre esses, a educação emerge como um desafio no que tange ao seu direcionamento a mulheres na fase da velhice. Assim, os objetivos geral e específicos do estudo foram: analisar os significados que as mulheres velhas – participantes de um Programa de EJA em Fortaleza-CE – atribuem à educação, para compreender se essa experiência influencia em suas vidas, na perspectiva de gênero; levantar as motivações que impulsionam a busca pela educação durante a velhice; estudar os desafios que a mulher velha encontra ao ter acesso à educação formal, em relação à questão de gênero; desvelar as memórias que as mulheres possuem sobre suas experiências educacionais anteriores; verificar como as mulheres compreendem a Escola e o Programa de EJA do qual participam. Para tanto, lançou-se mão da pesquisa qualitativa, com auxílio das pesquisas de tipo bibliográfica, documental e de campo, além das seguintes técnicas para obtenção das informações: observação direta e entrevista não-estruturada baseada no método História Oral Temática. Como campo da pesquisa, selecionou-se um Programa de EJA em uma Escola Municipal de Fortaleza, com a participação de 5 (cinco) mulheres velhas alunas da EJA, cujas narrativas foram interpretadas à luz da Hermenêutica-Dialética. As percepções, sobre o retorno ou acesso pela primeira vez aos estudos, foram reunidas em 3 (três) categorias empíricas: lazer/ sociabilidade, cidadania/ pertença, liberdade/ autonomia. Dessa forma, os significados foram os mais variados possíveis, mas todos concorreram para a necessidade de ampliação de espaços educacionais para as mulheres, onde possam reconhecer suas demandas singulares no decorrer de todas as fases de suas vidas. A partir dessa mudança de pensamento e, consequentemente, de paradigma, é que a escola pode abraçar, democraticamente, homens e mulheres, de diferentes idades, de pertencimentos de classe os mais diversos, de múltiplas crenças e nacionalidades. Essa desconstrução requer o engajamento de grupos historicamente colocados à margem do direito à educação que, reconhecendo-se como protagonistas, podem contribuir com a criação de uma nova ordem societária e de uma sociabilidade pautada na igualdade de gênero sem nenhum tipo de preconceito e/ ou discriminação.

The materiality of the convergence of media due to digitalization processes of the production and content distribution means stands have configured new dynamics, both in the relationship between producers and receptors and in the texts which result from it. Our research study claims that these dynamics configure an ambience in which the logics of the open and closed systems operate interdependently and in which the space we define as „mediasphere‟ is constituted. This space is understood as a production, publication and consumer place in which the subjects of communication share texts and construct discourses, thus producing meanings which articulate symbolic goods. This study - entitled "Public Policies for Teacher Training and the Institutional Program of Scholarships for Teaching at the State University of Goiás in Iporá/GO: the perception of women and the impact on their lives" - was developed by research line State, Educational Policies and Institutions, of the Stricto Sensu Postgraduate Program in Education of the Pontifical Catholic University of Goiás (PUC/GO). The objective of this study is to study the Institutional Program of Initiatives for Teaching (PIBID) in execution (2014-2018) at the State University of Goiás (UEG), Campus Iporá, investigating what place it occupies in the reality of academic women, who seek there their professional qualification. Considering the importance of the PIBID in the transformation of the educational reality, by encouraging and enabling the initial training of teachers for regions of the interior of our country, and considering that women constitute the greatest demand in PIBID in Iporá, we sought to answer: Who are these women, and what understanding and evaluation do they make of the teaching profession and the training they receive and how does the Program intervene in their lives? The study adopted as a theoretical contribution the studies of Brzezinski (2008, 2011, 2014), Canan; (1995), Tardif and Raymond (2000), among others, who provided a foundation for Teacher Training and PIBID; (1998), and Cooper (1996), who have contributed to the discussion of the literature on the subject, and the authors, such as Louro (1997), Catani et al (2003), Costa and Bruschini (1992), Scott (1995), Saffioti (1979), Apple (1998) and Asuncion (1996), who subsidized the discussions on Gender, Woman and Profession Teacher. In order to begin to approach the theme, the following terms were used as descriptors that guided the bibliographic survey: Institutional Program of Initiation Scholarships; Teacher training; Profession Teacher; Genre; and Women. It was also decided to carry out an empirical research. The research at the State University of Goiás, Campus Iporá, counted on 21 participants, being: Five Area Coordinators (one from each course / subproject - Biological Sciences, History, Geography, Literature and Mathematics) and sixteen female PIBID scholarship holders. For data collection, the application of questionnaires was used. For the analysis of the data, the content analysis was adopted, within the Materialist Historical Dialectic perspective. Also included in the data collection procedures were the analysis of the PIBID projects and other legal documents that involve the Institution and the Program. The analyzes have a qualitative approach, since the main variants involved are the "voices" of the subjects and the projects and institutional documents about the implementation of PIBID. Thus, through the study, it was possible to identify that, although there are some misconceptions, the Program has contributed in a positive way in the life of the scholars participating in the research.
Este estudo – intitulado “Políticas Públicas para a Formação de Professores/as e o Programa Institucional de Bolsas de Iniciação à Docência na Universidade Estadual de Goiás em Iporá/GO: a percepção das mulheres e o impacto em suas vidas” – desenvolveu-se pela linha de pesquisa Estado, Políticas e Instituições Educacionais, do Programa de Pós-Graduação Stricto Sensu em Educação da Pontifícia Universidade Católica de Goiás (PUC/GO). Tem como objetivo estudar o Programa Institucional de Bolsas de Iniciação à Docência (PIBID) em execução (2014-2018) na Universidade Estadual de Goiás (UEG), Campus Iporá, investigando que lugar ele ocupa na realidade das mulheres acadêmicas, que buscam ali sua formação profissional. Na pesquisa, considerando a importância que o PIBID tem na transformação da realidade educacional, ao incentivar e viabilizar a formação inicial de professores para regiões do interior de nosso país, e considerando, ainda, que as mulheres constituem a demanda maior no PIBID em Iporá, buscou-se responder: Quem são essas mulheres, e que entendimento e avaliação fazem da profissão docente e da formação que recebem e como o Programa intervém em suas vidas? O estudo adotou como aporte teórico os estudos de Brzezinski (2008, 2011, 2014), Canan; Freitas (2007), Imbernon (2000), Moraes, Pacheco e Evangelista (2003), Nóvoa (1997, 2009), Tardif e Raymond (2000), dentre outros, que ofereceram fundamentação quanto à Formação de Professores e ao PIBID; e autores, dentre os quais Louro (1997), Catani et al (2003), Costa e Bruschini (1992), Scott (1995), Saffioti (1979), Apple (1998) e Assunção (1996), que subsidiaram as discussões sobre Gênero, Mulher e Profissão Professora. Para iniciar a abordagem do tema, aplicaram-se como descritores que orientaram o levantamento bibliográfico os termos: Programa Institucional de Bolsas de Iniciação à Docência; Formação de Professores; Profissão Docente; Gênero; e Mulheres. Optou-se, também, por realizar uma pesquisa empírica. A pesquisa na Universidade Estadual de Goiás, Campus Iporá, contou com 21 participantes, sendo elas: Cinco Coordenadoras de Área (uma de cada curso/subprojeto – Ciências Biológicas, História, Geografia, Letras e Matemática) e dezesseis mulheres bolsistas do PIBID. Para coleta de dados, empregou-se a aplicação de questionários. Para a análise dos dados, adotou-se a análise de conteúdo, dentro da perspectiva Materialista Histórica Dialética. Fez parte ainda dos procedimentos de coleta de dados a análise dos projetos do PIBID e demais documentos legais que envolvem a Instituição e o Programa. As análises têm abordagem qualitativa, uma vez que as principais variantes envolvidas são “as vozes” dos sujeitos e os projetos e documentos institucionais sobre a execução do PIBID. Assim, pelo estudo realizado, foi possível identificar que, embora apresente alguns desacertos, o Programa tem contribuído de forma positiva na vida das bolsistas participantes da pesquisa.

Повний текст джерелаA partir do problema da crise na sucessão geracional em unidades produtivas familiares e da reprodução social do modo de produção familiar, este estudo lança um olhar sobre os dilemas enfrentados pelas populações rurais e sobre as ações desenvolvidas pelo Estado para o atendimento de suas demandas, através das políticas públicas. As especificidades do processo de sucessão na agricultura familiar remetem a questões de gênero e geracionais, colocando os jovens e mulheres no centro da análise. Objetiva-se, num primeiro momento, traçar um detalhado diagnóstico da situação juvenil e feminina no meio rural, compreender suas demandas e o processo de formação da agenda de políticas para estes segmentos. Num segundo momento, busca-se caracterizar e analisar os diferentes programas e políticas desenvolvidas, seus impactos sobre a realidade rural, as avaliações que se têm sobre eles, elevando o conhecimento sobre os avanços e limites destas intervenções. Para o diagnóstico da realidade rural, realiza-se uma ampla revisão dos estudos recentes nesta temática, delimitando a análise à agricultura familiar do Rio Grande do Sul, o que inclui um trabalho empírico sobre os jovens agricultores familiares do município de Faxinal do Soturno/RS. Para a análise das políticas públicas, busca-se traçar o percurso histórico das ações desenvolvidas pelo Estado para o rural brasileiro, com especial atenção para a formação da agenda e a inclusão das pautas de jovens e mulheres da agricultura familiar, abordando as diferentes etapas que compõe o ciclo de políticas públicas. Ao final, as respostas institucionais são colocadas frente às demandas das populações rurais, permitindo um balanço sobre a efetividade das ações até então desenvolvidas. Particularidades da situação juvenil e feminina no meio rural ampliam os fatores explicativos da evasão do trabalho agrícola, apresentando novos elementos para o entendimento da reprodução social na agricultura familiar, o que aponta para a necessidade de adaptações no foco das políticas públicas para este público.
From the problem of the crisis in the generational succession in family production units and of the social reproduction of family farming, this study takes a look at the dilemmas faced by rural populations and on the actions taken by the state to meet their demands through public policy. The specifics of the process of succession in family farming refer to gender and generational issues, putting young people and women in the center of the analysis. It aims, at first, to draw a detailed diagnosis of juvenile and women's situation in rural areas, to understand their demands and the process of creation of the policy’s agenda for these segments. Secondly, it seeks to characterize and to analyze the different programs and policies that are developed, their impact on the rural reality, the evaluations that we have on them, raising awareness about the progress and limits of these interventions. For the diagnosis of the rural reality, we carried out a comprehensive review of recent studies on this topic, limiting the analysis to the family farm in Rio Grande do Sul, which includes an empirical work on the young farmers of Faxinal do Soturno/RS. For the analysis of public policies, we seek to trace the historical course of the actions taken by the State for the brazilian countryside, with special attention to the formation of the agenda and the inclusion of guidelines for youth and women in family agriculture, addressing the different steps that constitute the cycle of public policies. Finally, the institutional responses face the demands of rural populations, enabling an assessment of the effectiveness of actions hitherto developed. Special features of the youth and women's situation in rural areas expand the explanatory factors of evasion of agricultural work, introducing new elements to the understanding of social reproduction in family farming, which points out to the need for adjustments in the focus of public policies for this audience.

In order to fight against women's exclusion in peace processes and facilitate their involvement, the UN Security Council adopted on October 31st 2000 the 1325's Resolution entitled " Women, Peace and Security " which requires states and organizations involved in a conflict to involve women in all peace processes. Mammalian sexual reproduction requires sexual determination, sexual differentiation, and the production of haploid gametes. In this thesis, I examined the genomic evolution of the mouse Y chromosome, which instructs sexual determination, and genetic regulation of a program of gene expression for meiosis, a specialized cell cycle which gives rise to haploid gametes. Chapter 2 describes the study of the mouse Y chromosome. Contrary to popular theory that Y chromosomes should be degenerate and gene poor, we find that the mouse male-specific region of the Y chromosome (MSY) is almost entirely euchromatic and contains about 700 protein-coding genes. Almost all of these genes belong to three acquired, massively amplified gene families that have no homologs on primate MSYs but do have acquired, amplified homologs on the mouse X chromosome. We propose that lineage-specific convergent acquisition and amplification of X-Y gene families is a result of sex-linked meiotic drive. Chapter 3 describes the gene regulatory program of meiotic prophase. Meiotic prophase comprises a complex chromosomal program results in the production of haploid gametes. This must be supported by a program of gene expression via which the required genes are induced. We interrogated gene expression in fetal ovaries over time and space, and in mutants of Dazl and Stra8 - key genes required for meiotic initiation. We determined that genes are regulated in three classes. Class 1 genes are expressed independently of Stra8, class 2 genes are expressed partially independently of Stra8, and Class 3 genes are dependent on Stra8 to be expressed. All genes require Dazl to be expressed. We propose that the Stra8-independent genes may represent genes required to be expressed prior to or early during meiotic initiation. Following initiation of meiosis, we found that Stra8 is required to induce down-regulation of its own expression. We propose that induction of down-regulation of the initiating signal by itself serves to ensure timely cessation of and one-time activation of the chromosomal program of meiotic prophase.

Às vésperas de completar 100 anos em 2010, o pentecostalismo no Brasil se mostra como um fenômeno complexo, multifacetado, desafiando comunicadores, empresários da mídia, sociólogos, pastores, crentes, ateus e também pesquisadores da comunicação social, já que na contemporaneidade as experiências religiosas são impulsionadas pela adoção da cultura mediática. Essas experiências fermentam em programas como o Show da Fé comandado pelo líder da Igreja Internacional da Graça de Deus (IIGD), R. R. Soares, instituição religiosa objeto deste estudo. Visto acreditar que fenômenos sociais não são uniformes e tampouco que os produtos religiosos sejam livres de contradições, a pesquisa busca compreender a visibilidade mediática religiosa no tecido das relações sociais dos adeptos que assistem o Show da Fé. Ou seja, a pesquisa analisa a comunicação religiosa sob um outro ângulo, distinto da visão linear da transmisssão da mensagem, entendendo aquela comunicação como processo mediado pelas práticas sociais e culturais que se constroem no cotidiano, onde a constituição da subjetividade é resultado das maneiras pelas quais os sujeitos interagem entre si perante a mensagem. Assim, a pesquisa - um estudo de recepção na linha das mediações socioculturais - permitiu, entre outros aspectos, observar nos usos e hábitos de consumo televisivo a dialética denomiada por Martín-Barbero consumo objetivação de desejos/lugar de diferenciação social e processo ritual, evidenciando que o grupo de pertença exerce influência para a ressignificação do desejo do adepto. Foi possível identificar que a chave do gênero do programa Show da Fè é o poder de acionar ou despertar no imaginário dos adeptos um outro mundo, cujo senso de ordem interna, integração e direção faz com que o homem se sinta mais forte para suportar as dificuldades da existência.
On the verge of completing 100 years in 2010, Pentecostalism in Brazil turns out to be a complex, multifaceted, phenomenon, which challenges communication, media entrepreneurs, sociologists, preachers, believers, atheists and social communication reserachers, once nowadays new religious experiences are moved by the adoption of the mediatic culture. There are now widely shown experiences on broadcasts such as Show da Fé (The show of faith) hosted by R. R. Soares, the leader of Igreja Internacional da Graça de Deus (International Church of the Grace of God), which is the religious institution being this study's main topic. Having in mind the belief that social phenomenons are not uniform and also being aware that it is not possible to believe that the religious products are free from contradiction, the research opted to look for understanding the religious mediatic visibility through the thread of social relationships of those who adopt The show of faith. It means the research analyses the religious communication form a different angle, distinct from the linear view of message transmission, understanding that sort of communication as the process mediated by cultural and social practices which are built everyday, where the constitution of subjectivity is a way of analyzing the ways through which the individuals interact with the message. Thus, the research- a study on reception following the research line of the socio-cultural meditiation - allowed, among other aspects, to notice in the uses and habits of television consumerism the dialect named by Martin-Barbero as wish/place of social differentiation and ritual process objectification consumerism, evidencing that the group of belonging exerts influence for the re-seignification of the follower's wish. It was possible to identify that the key for the broadcast genre of Show da Fé (The show of faith) is the power... (Complete abstract click electronic access below)
