Повний текст джерелаENGLISH ABSTRACT: Pseudococcus viburni (Signoret) (Hemiptera: Pseudococcidae) (obscure mealybug), is a common and serious pest of apples and pears in South Africa. Consumer and regulatory pressure to produce commodities under sustainable and ecologically compatible conditions has rendered chemical control options increasingly limited. Information on the seasonal occurrence of pests is but one of the vital components of an effective and sustainable integrated pest management system needed for planning the initiation of monitoring and determining when damage can be expected. It is also important to identify which orchards are at risk of developing mealybug infestations while development of effective and early monitoring tools for mealybug populations will help growers in making decisions with regards to pest management and crop suitability for various markets. It is also essential to determine the presence and efficacy of naturally occurring biological control agents in orchards so as to ascertain the potential of biological control as a viable alternative in orchards. However, under the current integrated pest management protocol, it has been difficult to determine this, due to the sporadic and relatively low incidence of mealybug infestations in some orchards, or by simply relying on naturally occurring field populations of biocontrol agents. Knowledge of the environmental conditions under which P. viburni population levels may become destructive is also essential for timing the release of insectary reared natural enemies as well as understanding the population ecology of this pest and its natural enemies. Information was gathered regarding the seasonal phenology of P. viburni and its natural enemies in pome fruit orchards in the Western Cape Province during the 2007/08 and 2008/09 growing seasons. Seasonal population studies showed that P. viburni has multiple overlapping generations with all life stages present throughout the year. The highest orchard infestations occurred during the summer period until early winter (January to early June). This was followed by a decrease in population from late June to November, before another increase in December. Presence-absence sampling of mealybugs on the host plant revealed that woody parts of the tree, such as the trunk and old stems were the most preferred sites for mealybug habitation, due to the availability of protected refuge sites. Migration of mealybug populations to newer growth and the upper sections of the tree crown, such as the new stems, leaves and eventually the fruit, was observed from December throughout the summer period until the early winter in June. Fruit colonization in both apples and pears commenced in January, when the fruit had developed a size sufficient for P. viburni to penetrate and occupy spaces such as the fruit core, calyx and stem end. There was no evidence of P. viburni occurring beneath the soil surface or on the roots of host trees. Two natural enemies of mealybugs, namely Pseudaphycus maculipennis (Mercet) and Coccidoxenoides perminutus (Girault), were found to be active in apple and pear orchards in the Western Cape. However, the status of C. perminutus as a parasite of P. viburni still needs to be verified despite evidence of emergence from P. viburni mummies, which was not sufficient enough to suggest that it is a useful biological control agent. Seasonal abundance trends of the two natural enemies revealed that their lifecycle is synchronized with that of the host. However, there was no evidence of P. maculipennis activity in Ceres. No predators were found during the course of this study. The rate of P. viburni parasitism at harvest was 46.52%, with P. maculipennis and C. perminutus constituting 98.966% and 1.034% of the parasitoids recovered from mealybug mummies, respectively. Studies on the use of pheromone traps as early monitoring tools for P. viburni showed that there was a positive and significant relationship between the fruit infestation and number of P. viburni adult males caught in pheromone-baited traps (r2 = 0.454). The action threshold level was estimated to be 2.5 male P. viburni caught per trap per fortnight at an economic threshold of 2% fruit infestation.
AFRIKAANSE OPSOMMING: Pseudococcus viburni (Signoret) (Hemiptera: Pseudococcidae) (ligrooswitluis), is ‘n algemene en ernstige plaag van appels en pere in Suid-Afrika. Druk deur verbruikers en regulasies om kommoditeite onder volhoubare en ekologies verenigbare toestande te produseer het chemiese beheeropsies toenemend beperk. Inligting oor die seisoenale voorkoms van plae is een van die essensiële komponente van ‘n effektiewe en volhoubare geïntegreerde plaagbestuurprogram. Dit is in die aanvanklike beplanning van monitering en om te bepaal wanneer skade verwag kan word. Dit is ook belangrik om boorde vroegtydig te identifiseer wat die risiko het om witluisbesmettings te ontwikkel. Die ontwikkeling van effektiewe en vroeë moniteringstegnieke vir witluisbevolkings sal produsente help met besluitneming rakende plaagbestuur en die geskiktheid van gewasse vir verskeie markte. Dit is ook noodsaaklik om die teenwoordigheid en effektiwiteit van biologiese beheer agente wat natuurlik in boorde voorkom te bepaal ten einde die potensiaal van biologiese beheer as ‘n lewensvatbare alternatief vas te stel. Seisoenale bevolkingstudies het getoon dat P. viburni verskeie oorvleuelende generasies het met alle stadia teenwoordig regdeur die jaar.

Megastigmus transvaalensis is an exotic wasp that attacks Brazilian pepper tree drupes Schinus terebinthifolius in native forests and areas of restoration and ecological restoration in Brazil. The parasitism begins with the oviposition M. transvaalensis in drupes S. terebinthifolius where the larvae hatch and remain internally feeding of nutrients and tissue, affecting germination. The aim of this work was to study the effect of temperature, rainfall and humidity in the population dynamics of M. transvaalensis and determine the parasitism rate and sex ratio of this wasp in drupes S. terebinthifolius. The study was performed with yellow sticky traps and collection of S. terebinthifolius drupes in seasonal semideciduous forest during August 2014 to September 2015, in the region of Sorocaba, São Paulo, Brazil. The capture of insects through the sticky traps proved satisfactory, and thus can be applied to the monitoring of small Hymenopteros. Populations of M. transvaalensis were negatively correlated with the maximum temperature and population peak in the fall of 2015, meaning that when the maximum temperature has reduced, there is a higher incidence of insect field. The parasitism rate of S. terebinthifolius drupes ranged from zero to 36.34% during this period, making this value an aggravating factor for the emergence of new individuals, since this phytophagous wasp has the potential to be spread throughout Brazil and It poses a threat to the natural regeneration of S. terebinthifolius. The sex ratio of M. transvaalensis was 0.42 and 0.08 in the laboratory field. The bio-ecology and the damage caused by M. transvaalensis in drupes S. terebinthifolius warrant further studies to integrated management
Megastigmus transvaalensis é uma vespa exótica que ataca drupas de aroeira-pimenteira Schinus terebinthifolius em florestas nativas e em áreas de recomposição e restauração ecológica no Brasil. O parasitismo inicia com a oviposição de M. transvaalensis nas drupas de S. terebinthifolius, onde as larvas internamente eclodem e permanecem alimentando-se dos nutrientes e tecidos, prejudicando a germinação. O objetivo desse trabalho foi estudar o efeito da temperatura, precipitação e umidade na flutuação populacional de M. transvaalensis e determinar o índice de parasitismo e razão sexual dessa vespa nas drupas de S. terebinthifolius. O estudo foi realizado com armadilhas adesivas amarelas e coleta de drupas de S. terebinthifolius em fragmento de floresta estacional semidecidual, durante agosto de 2014 a setembro de 2015, na região de Sorocaba, São Paulo, Brasil. A captura dos insetos através das armadilhas adesivas se mostrou satisfatória, podendo assim ser aplicada para o monitoramento de pequenos Hymenopteros. As populações de M. transvaalensis apresentaram correlação negativa com a temperatura máxima e pico populacional no outono de 2015, significando que, quando a temperatura máxima apresenta redução, há maior incidência do inseto em campo. O índice de parasitismo das drupas de S. terebinthifolius variou de zero a 36,34% no período avaliado, tornando este valor um agravante para o surgimento de novos indivíduos, uma vez que, essa vespa fitófaga tem potencial de ser disseminado por todo Brasil e representa ameaça para a regeneração natural de S. terebinthifolius. A razão sexual de M. transvaalensis foi de 0,42 no laboratório e 0,08 em campo. A bioecologia e os danos causados por M. transvaalensis em drupas de S. terebinthifolius justificam mais estudos visando o manejo integrado dessa vespa fitófaga.

Повний текст джерелаWith the rapid population growth and the reduction of areas for cultivation, the increased productivity of foods of all kinds is essential to meet the world's population demand. However post-harvest losses from the time of harvest until the arrival to the consumer, limits the supply of fruits to the consumer. The losses in the post-harvest of apples, strawberries and peaches, caused by the incidence of rot led by phytopathogenic fungi, is responsible for most of the losses. These rots are caused by various fungi, among these are the Colletotrichum sp., cause of bitter rot on apples, Botrytis cinerea causative agent of gray mold on strawberries and M. fructicola on peaches which causes brown rot. These phytopathogens are fungi with high ability to spread infection and therefore cause serious damage to fruits, generating losses in post-harvest. Usually the fungi control is conducted with the use of fungicides. However, the use of chemicals in the process concerns consumers, since there may be residues in fruits and the environment. Therefore, alternative methods like the resistance induction can be used to control the disease-causing microorganisms in the postharvest fruits. The induction consists on stimulating the plants defenses through (inducing) elicitor molecules, specifically the synthesis of compounds that act directly on the pathogen as phenols, protein-RPs, or producing structural reinforcement of tissues adjacent to the site of infection of the fungus. Currently, chitosan extracted from crustacean shells is an alternative elicitor molecule of low cost and no risk to the consumer that has been used in the induction of the resistance in postharvest fruits. This biopolymer has the ability to trigger the defensive responses of the fruits; apple, peach, and strawberry against fungi Colletotrichum sp., Botrytis cinerea and Monilinia fructicola, respectively. The inductor was applied at concentrations of 0.25, 0.5; 1.0 and 2.0% and on the control (distilled water). The treatments were arranged in a completely randomized design with four replications of 20 apple fruits, 20 strawberry fruits and 15 peach fruits. The fruits were selected and standardized, and subsequently the apples and peaches were subjected to treatment by immersion in solutions of chitosan, and strawberries were treated by spraying the chitosan-containing solution directly onto the fruits. After 24 hours, the fruits were inoculated with a solution containing conidia of the phytopathogen Colletotrichum sp. on apples, B. cinerea on strawberries and M. fructicola on peaches, in concentrations of [5.10]^(-3) conidia/ml directly on the fruit, with the help of a spray bottle or with a micropipette in the case of apples, directly inoculating the solution in an wound to the 2 mm bark. After completion of the treatments, the fruits were placed in BOD at 26 ± 1 ° C for apples and peaches, and 10 ± 1 ° C for strawberries, and evaluated after 24 hours to determine the following parameters; weight loss, physical and chemical analysis (solids and total soluble, titratable acidity, firmness, decay incidence) and biochemical (protein, reducing sugars, total sugars, anthocyanins, flavonoids, FAL, peroxidase, chitinase and β-glucanase). An initial sample of the fruits was taken to carry out the initial analyzes, using this data as comparative parameters. In a second experiment, the fungus (Colletotrichum sp, Botrytis cinerea and M. fructicola) were grown in the middle of culture containing the different concentrations of chitosan, to verify the existence of fungitoxic or fungistatic effect of the biomolecule in vitro. The fungi were previously cultivated in clean plates, and subsequently transferred to plates containing PDA medium with the chitosan concentrations (0, 0.25, 0.5, 1 and 2), and after 24 and 48 hours, were performed perpendicular measurements of the diameter of the colony to verify its mycelial growth. Data from experiments were submitted to analysis of normality and variance, and measures were compared by Tukey test and regression test (p = 0.05), with the assistance of Assistat software. The results demonstrated the interference of chitosan on the induction of resistance to control the incidence of bitter rot in postharvest Gala apple, activating the PRPs B-1-3 glucanase and control of Colletotrichum sp in vitro with fungistatic action. On strawberries, the inductor controlled the gray mold by activating the peroxidase, chitinase and β-1-3-glucanase, directly under the fungus B. cinerea in vitro. On peaches, the action was on the maintenance of fruit quality, on the induction, activating genes of chitinase, β 1-3 glucanase, and on the same way, on the in vitro of M. fructicola. Therefere, it has been concluded that chitosan has great potential in the induction of fruit resistance in post-harvest, activating the defense against pathogenic fungi, and directly over the latter with fungistatic activity and in vitro fungitoxic activity.

Повний текст джерелаENGLISH ABSTRACT: Fruit and Leaf Spot Disease (FLSD) of citrus, caused by Phaeoramularia angolensis, is found only in 18 countries in Africa, the Comores Islands in the Indian Ocean and Yemen in the Arabian peninsula. The major citrus export countries in Africa are Morocco, South Africa, Swaziland, and Zimbabwe. Zimbabwe is the only country affected by FLSD. FLSD is a disease of major phytosanitary and economic importance and its devastating effect on citrus is highlighted by the fact that the damage is cosmetic, which renders the fruit unmarketable. Total crop losses are not uncommon in Kenya. The aims of the present study, therefore, was was to determine the occurrence of P. angolensis in Zimbabwe and neighbouring Mozambique, to compare these isolates with the Cercospora Fresen. isolates from Swaziland and South Africa, to determine the epidemiology of the pathogen and to implement an effective control strategy to prevent the spread of FLSD. Leaf samples with citrus canker-like lesions collected in the early 1990’s in Zimbabwe were found to be infected by the fungus, Phaeoramularia angolensis. Surveys were undertaken to determine the spread and intensity of FLSD in Zimbabwe and Mozambique. In Zimbabwe, P. angolensis was limited to an area above the 19° south latitude, predominantly the moist areas and not the low-lying drier parts of the country. In Mozambique, no P. angolensis symptoms were found. Observations during the survey indicated that no proper management systems were implemented by Zimbabwean growers. A cercosporoid fungus causing a new Fruit and Leaf Spot Disease on Citrus in South Africa was identified. From morphological and rDNA sequence data (ITS 1, 5.8S and ITS 2), it was concluded that the new disease was caused by Cercospora penzigii, belonging to the Cercospora apii species complex. The genera Pseudophaeoramularia and Phaeoramularia are regarded as synonyms of Pseudocercospora, and subsequently a new combination was proposed in Pseudocercospora as P. angolensis. Cercospora gigantea was shown to not represent a species of Cercospora, while Mycosphaerella citri was found to be morphologically variable, suggesting that it could represent more than one taxon. A control strategy for the control of FLSD was evaluated in the study. The data showed that P. angolensis in Zimbabwe can be managed successfully by the removal of all old and neglected orchards, and on timely fungicide applications. Trifloxystrobin + mancozeb + mineral spray oil (20 g + 200 g + 500 ml/100 l water) applied in November, January and March was the most effective treatment. Three applications of benomyl + mancozeb + mineral spray oil (25 g + 200 g + 500 ml/100 l water) applied during the same period, was the second most effective treatment, and two applications (November and January) of trifloxystrobin + mineral spray oil (20g + 500 ml/100 l water) and difenoconazole (40 g) per 100 l/water applied twice in November and January, the third most effective treatment. The spore trap and weather data showed that P. angolensis needs high moisture and temperatures in excess of 25°C for disease development. It is concluded that P. angolensis in Zimbabwe can be managed successfully by implementing a holistic approach, which should be supported by the authorities, organised agriculture and all technical personnel involved in citrus production.
AFRIKAANSE OPSOMMING: Blaar- en vrugvleksiekte (BVVS) op sitrus, veroorsaak deur Phaeoramularia angolensis, kom in 18 lande in Afrika voor asook die Comores Eilande in die Indiese Oseaan en Yemen op die Arabiese skiereiland. Marokko, Suid Afrika, Swaziland en Zimbabwe is die belangrikste uitvoerders van sitrus in Afrika. Van dié lande het slegs Zimbabwe blaar en vrugvleksiekte op sitrus. Hierdie siekte is van fitosanitêre en ekonomiese waarde en die nadelige effek van die siekte, wat slegs kosmetiese van aard is, is venietigend aangesien vrugte onbemarkbaar is. Totale opbrengsverliese is nie ongewoon in lande soos Kenya nie. Trifloxystrobin (20 g) + minerale spuitolie (500 ml) per 100 l/water en difenoconazole (40 g) per 100 l/water, beide as twee bespuitings toegedien in November en Januarie, het die derde beste resultaat opgelewer. Die spoorlokval en klimatologiese data het getoon dat P. angolensis vogtige toestande en temperature hoër as 25°C benodig vir siekteontwikkeling. Die afleiding uit die studie is dat P. angolensis suksesvol beheer kan word indien ‘n holistiese benadering gevolg word en alle rolspelers naamlik die owerheid, georganiseerde landbou en tegniese personeel die proses ondersteun.

Повний текст джерелаENGLISH ABSTRACT: The export of South African fruit to some of its biggest international markets may be rejected if the phytosanitary pest, Phlyctinus callosus (Coleoptera: Curculionidae) is found in fruit consignments. An alternative to methyl bromide fumigation is needed and one of the most promising of the alternative treatments is phytosanitary irradiation as it is environmentally friendly, does not leave residues on food or in the environment and it is effective against a wide variety of insects. Field-collected weevils were treated with five doses of gamma irradiation (5, 10, 20, 40 and 80 Gy) and the fecundity and fertility of mating crosses of treated males and females with treated and untreated individuals of the opposite sex, were determined to evaluate the effect on P. callosus reproductive ability post-treatment. Results indicated that irradiation treatment did not affect fecundity, but fertility was significantly affected, decreasing as the irradiation dose increased. Females were more susceptible to the irradiation treatment than males, and after treatment with 80 Gy, eggs laid by females and mated with either treated or untreated males, did not hatch. A generic dose of 400 Gy for all insect pests except tephritid fruit flies and pupae and adult Lepidoptera is currently approved by USDA-APHIS (United States Department of Agriculture – Animal and Plant Health Inspection Services) for use on certain commodities. Results from the present study support the development of a species-specific dose for P. callosus, as well as the development of a group generic dose for the Curculionidae that is lower than 400 Gy. Effective phytosanitary irradiation treatments are only feasible if the treatment does not adversely affect fruit quality and the marketability of export fruit. Therefore, an investigation of the effects of irradiation disinfestations treatments on the quality of the new pluot cultivar, ‘Flavor Fall’ was made. Packed cartons were treated with three doses of gamma irradiation: 400 Gy, 900 Gy and 1400 Gy. After treatment fruit underwent a PD 7 dual temperature cold storage regime for 42 days and a shelf-life simulation for 7 days. The impact of insect-proof bags, sometimes required by importing countries to keep insects off packaged fruit, was also investigated. Respiration rate of the fruit was measured throughout and fruit quality evaluations were done after cold storage and after shelf-life. The results indicated that quality parameters measured at the end of cold storage, which would be after the fruit arrives at the export markets, were above the minimum standards for overseas markets. Gel breakdown was unacceptably high after the higher temperature exposure of shelf-life for fruit treated with the 900 and 1400 Gy doses. The insect-proof bags reduced shrivel, but resulted in higher incidence of gel breakdown. The use of irradiation, together with the use of the insect-proof bag, has potential as an alternative postharvest mitigation treatment for plums. Lastly, an investigation into potential rearing methods for P. callosus, including recommendation for the future, was made as the availability of a sustainable rearing method that ensures a consistent supply of high quality P. callosus adults would enable continuous research with greater numbers of this pest. The information generated in this study provides a greater understanding of the radiation biology of, not only this curculionid species, but the Curculionidae as a group, and is valuable in advancing the development of alternative postharvest control measures against this phytosanitary pest.
AFRIKAANSE OPSOMMING: Suid Afrikaanse vrugte uitvoere na van die grootste internasionale markte mag weg gewys word as die fitosanitêre pes, Phlyctinus callosus (Coleoptera: Curculionidae) in die versending gevind word. ‘n Alternatief vir metiel bromied beroking word benodig en een van die mees belowende alternatiewe behandelings is fitosanitêre bestraling aangesien dit omgewings vriendelik is, nie residue op kos of in die omgewing los nie, en effektief is teen ‘n wye verskeidenheid van insekte. Veldversamelde kalanders is behandel met vyf dosisse gamma bestraling (5, 10, 20, 40 en 80 Gy) waarna die vrugbaarheid van paringskruisings bepaal is deur kruisings tussen behandelde manlike en vroulike kalanders met behandelde en nie-behandelde individue van die teenoorgestelde geslag te maak, en so die na-behandelings effek op die voortplantings vermoeë van P. callosus te evalueer. Die resultate van die huidige studie ondersteun die ontwikkeling van ‘n spesie-spesifieke dosis vir P. callosus, so ook die ontwikkeling van ‘n generiese groep dosis vir Curculionidae wat laer as 400 Gy is. Effektiewe fitosanitêre bestralings behandeling is slegs moontlik indien die behandelings dosis nie nadelig vir vrugkwaliteit en die bemarking van uitvoer vrugte is nie. Dus is die effek wat bestralings bestryding behandeling op die kwaliteit van ‘n nuwe pluot kultivar, ‘Flavor Fall’ ondersoek. Laastens is ‘n ondersoek ingestel vir moontlike teeltegnieke vir P. callosus en aanbevelings gemaak vir toekomstike studies. Die beskikbaarheid van ‘n volhoubare teeltegniek wat konstante, hoë kwaliteit P. colossus individue verskaf sal aaneenlopende navorsing met groter getalle van die pes moontlik maak. Повний текст джерелаENGLISH ABSTRACT: A survey was conducted in the Western Cape Province during the 1999/2000 and 2000/2001 seasons on mealybugs occurring in vineyards. P/anococcus ficus (Signoret) was the dominant mealybug in vineyards during this time. During this study P. ficus was recorded for the first time on roots of grapevines, which has far reaching implications for the control of this important vine leafroll virus vector as control actions were focused on above ground control. Other mealybugs presently recorded in local vineyards included Pseudococcus /ongispinus (Targioni) and Ferrisia ma/vastra (McDaniel). Pseudococcus viburni (Maskell) and Ps. so/ani Ferris were found on weeds in vineyards. Natural enemies of P. ficus recorded most frequently were species of Nephus predatory beetles, and the parasitaids Coccidoxenoides peregrinus (Timberlake), Anagyrus sp. and Leptomastix dacty/opii (Howard). Developmental studies on P. ficus and C. peregrinus indicated that the intrinsic rate of increase (rm) was similar, peaking at 25°C (rm = 0.169 for P. ficus; rm = 0.149 for C. peregrinus). The net replacement rate (Ra) was higher for P. ficus than for C. peregrinus at all five temperatures tested. The Ra for P. ficus reached a maximum at 21°C (308.87) and C. peregrinus at 25°C for C. peregrinus (69.94). The lower and upper thresholds for development of P. ficus were estimated at 16.59 and 35.61°C respectively. The lower threshold for development of C. peregrinus was 8.85°C. These parameters indicated that both insects were well adapted to temperatures in the Western Cape Province. The lower minimum threshold temperature of C. peregrinus in relation to that of P. ficus suggests that C. peregrinus should be more active during winter and early spring than P. ficus. A central systematic presence-absence sampling system was developed for P. ficus. Monitoring three different plant parts on the vine indicated that new growth areas on vines adjacent to the main stem could serve as an early warning system for pending P. ficus bunch infestations. Intervention should be planned when 2 % of the stems are infested with P. ficus when using this system. Seasonal population studies of P. ficus and its natural enemies showed that stem infestation by P. ficus reached peak levels during January in Robertson and Stellenbosch and during February in the Hex River Valley. Vine mealybugs colonised new growth early in the season, followed by the leaves and eventually the bunches towards the end of the season. High stem infestations early in the season resulted in high bunch infestation levels at harvest. A density dependent relationship was evident between P. ficus populations and parasitoid populations, suggesting that the parasitoids played a mayor role in the biological control of P. ficus populations. Biological control was however only achieved towards the end of the season when damage to the crop had already occurred. Mass releases of C. peregrinus on P. ficus populations were done in order to augment biological control as an alternative to chemical control. Between five and six releases of 20 000 C. peregrinus per release were done at monthly intervals in three grapegrowing areas. Mass released C. peregrinus controlled P. ficus adequately in the Hex River Valley. Control of P. ficus using this approach was no worse than using chemical control in Robertson and Stellenbosch. C. peregrinus is commercially available and can therefore be used as an alternative to chemical control by producers. Degree day estimation was used to predict development of P. ficus populations. This information was used as an input in a P. ficus pest management model. Data acquired from P. ficus and ant monitoring were used as components to construct a decision chart. This chart can be used by producers to optimise the control of P. ficus populations using either chemical control or mass releases of C. peregrinus.

Planococcus ficus (Signoret) is tans die dominante witluisspesie in wingerde in die Wes Kaap Provinsie. P. ficus kolonies is op wingerdwortels gevind. Dié bevinding kan verreikende gevolge hê vir die beheer van dié plaag as "n belangrike rolbladvirus vektor aangesien beheer tot dusver gefokus het op bogrondse gedeeltes. Ander witluisspesies wat in wingerde gevind is, sluit in Pseudococcus /ongispinus (Targioni) en Ferrisia malvastra (McDaniel). Pseudococcus vibumi (Maskell) en Ps. so/ani Ferris is op onkruide in wingerde gevind. Dominante natuurlike vyande van P. ficus sluit predatoriese kewertjies van verskeie Nephus spp. en die parasitoïede Coccidoxenoides peregrinus (Timberlake), Anagyrus sp. en Leptomastix dacty/opii (Howard) in. Повний текст джерелаENGLISH ABSTRACT: The codling moth, Cydia pomonella (L.) has been a major pest of pome fruits since before the turn of the last century. However, despite its high economic profile little is known about the bionomics and . behaviour of this pest in apple orchards in South Africa, information required for the development of a sustainable integrated management programme. In field trials there was contingency between the time of year and the upper and lower half of the tree. First generation moths laid significantly more eggs in the bottom half of the tree while second and third generation moths laid significantly more eggs in the top half of the tree. The preferred oviposition sites on Granny Smith (GS) and Golden Delicious (GD) cultivars, in order of preference, were leaves, fruit and wood. More eggs were laid on the fruit ofGS spurs (35.6 %) than on those ofGD spurs (10.7 %). On fruit spurs there was a significant increase in the number of eggs on GD leaves and GS fruit over the season, whereas the number of eggs on GS leaves and GD fruit remained constant. On branches there was an increase in the number of eggs on GD and GS leaves, but not on the fruit or wood. The preferred oviposition site on the fruit was the fovea of the stalk insertion and the rounded cheek area surrounding the fovea. The distribution within different fruit bearing classes (1 - 4 fruit per spur) was random only for one fruit per spur, while on the other spur classes clustering occurred. In laboratory studies of the embryonic and immature stages there was a linear relationship between rate of development and constant temperatures of 15, 17,20,25 and 30·C ± l·C. The lower threshold temperatures for embryonic, larval and pupal development were 11.1, 7.9, 9.9°C respectively. The degree-days required to complete embryonic, larval and pupal development were 80.5, 345, and 279 respectively. The response of the different stages to constant temperatures was similar to that under fluctuating temperatures. At temperatures below 16°C or abouve 27°C moths did not mate and few eggs were laid. Moth longevity decreased with increasing temperature. There was seasonal variation in longevity and oviposition at constant and fluctuating temperatures. Summer adults produced significantly more eggs than spring adults at constant temperatures. At a constant temperature of2S·C and fluctuating temperatures there were five distinct larval instars. The similarity between the mean head capsule width and ranges for each instar reared on fruit of different stages of development at fluctuating temperatures indicates that fruit development and temperature have little influence on mean head capsule width. From sleeve-cage studies in the orchard there was no significant difference in the fecundity of spring and summer moths. In the beginning of October spring moths produced significantly fewer eggs than in November. Egg mortality increased from 8.2 %in spring to 21.2 %in summer. Failure of 1st instar larvae to penetrate the fruit ranged from 4.9 % to 19.5 %, while mortality oflarvae from egg hatch to emergence from the fruit ranged from 29.7 % to 42.9 %. Mortality of 5th instar larvae after emerging from the apples ranged from 0 % to 8.7 % and pupal mortality from 0 % to 3.5 %. On large 27-year old trees more overwintering larvaewere found on Golden Delicious (13.9) than on Granny Smith trees (5.7), with over 70 % oflarvae being found in pruning wounds on both cultivars. On small 7-year old Golden Delicious and Granny Smith trees the mean number oflarvae was 0.5 and 2.0 per tree. A combined mating disruption and insecticide control programme reduced codling moth resistant populations to levels requiring a minimum to no insecticide intervention for several seasons. The efficacy of a pheromone based strategy, number of pheromone treatments, number of dispenserslha and level of insecticide intervention required are strongly influenced by prevailing weather conditions. Fruit infestation in orchards under a mating disruption programme and under an insecticide programme were greater along the borders compared to the interior. The presence of horticultural mineral oil on the leaves and branches did not have a detrimental effect on oviposition nor was there any significant ovicidal effect. A significant ovicidal effect was obtained when applied after oviposition. In field trials, insecticides with lower levels of efficacy than the primary insecticide, azinphos-methyl, provided acceptable control when successfully incorporated into a spray programme which followed a policy of alternation of insecticides across generations. The least variation in the number of degree-days between biofix and first egg hatch of the spring flight was when the second trap catch (Biofix 2) was used as the biofix. A biofix based on the first evening when the temperature reached or exceeded 1TC at 18:00 after first trap catch also showed less variation than when the biofix was based on first trap catch. The mean number of degree-days accumulated between Biofix 2 and first egg hatch was found to be 139.1 ° D. The number of degreedays between the first and second flight biofixes varied between 531.2 and 488.87°D with a mean of 508.1°D.
AFRIKAANSE OPSOMMING: Kodlingmot (Cydia pomonella) is reeds sedert die vorige eeu 'n sleutelplaag van kemvrugte. Ten spyte van die hoë ekonomiese profiel, is daar min kennis betreffende die bionomie en gedrag van hierdie plaag in appelboorde in Suid-Afrika. Hierdie inligting is noodsaaklik vir die ontwikkeling van 'n volhoubare geïntegreerde bestuursprogram. Tydens veldproewe is 'n korrelasie tussen die tyd van die jaar en eierlegging in die boonste of onderste helfte van bome gevind. Eerste-generasie motte het betekenisvol meer eiers in die onderste helfte van die bome gelê, terwyl tweede- en derde-generasie motte meer eiers in die boonste helfte van die bome gelê het. In toenemende volgorde was die voorkeur eierleggingsposisies op Granny Smith (GS) en Golden Delicious (GD) appels die blare, vrugte en hout. Meer eiers is op vrugte van GS spore (35.6%) as op dié van GD spore (10.7%) gelê. Op vrugspore was daar 'n betekenisvolle toename in aantal eiers op GD blare en GS vrugte deur die seisoen, terwyl die getal eiers op GS blare en GD vrugte konstant gebly het. Op takke was daar 'n toename in aantal eiers op GD en GS blare, maar nie op vrugte of hout nie. Die voorkeur eierleggingsposisie op vrugte was die fovea van die steelaanhegting en die wang rondom die fovea. Die verspreiding tussen verskillende vrugdraende klasse (1 - 4 vrugte per spoor) was slegs in die een-Vrug-per-spoorklas ewekansig, terwyl daar in die ander spoorklasse groepering voorgekom het. In laboratoriumstudies van embrioniese en onvolwasse stadia is 'n lineêre verband tussen ontwikkelingskoers en konstante temperature van 15, 17,20,25 en 30°C±I°C gevind. Die onderste drempel-temperature vir embrioniese, larwale en papie-ontwikkeling was onderskeidelik II. 1°C, 7.f?C en 9.9°C. Graaddae benodig vir voltooiing van embrioniese, larwale en papie-ontwikkeling was onderskeidelik 80.5,345 en 279. Die respons van verskillende stadia by konstante temperature het ooreengestem met hul respons onder wisselende temperature. Die ooreenkoms tussen die gemiddelde kopkapsulewydte en wydte-reeks vir elke instar wat op vrugte van verskillende stadiums van ontwikkeling by wisselende temperature geteel is, dui daarop dat vrugontwikkeling en temperatuur weinig invloed op gemiddelde kopkapsule-wydte het. Tydens mou-hok studies in die boord is geen betekenisvolle verskil in die fekunditeit van lente- en somer-motte waargeneem nie. Vroeg in Oktober het lente-motte betekenisvol meer eiers as in November geproduseer. Eiermortaliteit het van 8.2% in die lente tot 21.2% in die somer toegeneem. Faling van 1ste instar larwes om vrugte te penetreer het van 4.9% tot 19.5% gewissel, terwyl mortaliteit van larwes vanaf uitbroei tot uitkoms uit die vrug van 29.7% tot 42.9% gewissel het. Mortaliteit van Sde instar larwes na uitkoms uit die vrug het van 0% tot 8.7% gewissel, en papie-mortaliteit van 0% tot 3.5%. Op groot, 27-jaar oue bome is meer oorwinterende larwes op Golden Delicious (13.9) as op Granny Smith (5.7) gevind, en meer as 70% van die larwes op beide kultivars is op snoeiwonde gevind. Op klein, 7-jaar oue bome was die gemiddelde aantallarwes op Golden Delicious en Granny Smithbome 0.5 en 2.0 onderskeidelik. Die teenwoordigheid van minerale olie op blare en takke het geen nadelige effek op eierlegging gehad nie en dit het geen betekenisvolle eierdodende effek gehad nie. Indien die olie ná eierlegging toegedien is, is daar wel 'n betekenisvolle eierdodende effek waargeneem. Tydens veldproewe het insekdoders met laer effektiwiteit as die primêre insekdoder, azinfos-metiel, aanvaarbare beheer verskaf indien dit suksesvol geïnkorporeer is in 'n spuitprogram deur 'n beleid van afwisseling van insekdoders oor generasies. Die kleinste variasie tussen die aantal graaddae tussen biofix en eerste uitbroei van eiers is gevind indien die tweede lokvalvangs as biofix gebruik is. 'n Biofix gebaseer op die eerste aand na die eerste lokval vangste wat die temperatuur 17°Cofhoër was teen 18:00, het ook 'n kleiner variasie getoon as die eerste lokvalvangs. Die aantal graaddae tussen die tweede en derde vlug biofix het tussen 531.2 en 488.87°D gewissel, met 'n gemiddelde van 508.1°D.

Повний текст джерелаENGLISH ABSTRACT: The banded fruit weevil, Phlyctinus callosus (Schönherr), is a key pest of apples, nectarines and grapevines in the southern areas of the Western Cape. The control of P. callosus is not satisfactory and the insecticides used to control this insect have not proved to be effective since the development of tolerance to pyrethroids and acephate. A control method that can be used, despite it being very labour-intensive, is that of tree trunk barriers. The use of such a method will prevent the weevils from reaching the fruit, as they are unable to fly. Alternative control options, such as the use of entomopathogenic nematodes, are urgently needed for the control of P. callosus. Entomopathogenic nematodes belonging to the Steinernematidae and Heterorhabditidae are ideal biocontrol agents for incorporation into an integrated pest management programme. In order to develop control strategies for P. callosus, large numbers and a predictable quantity of different weevil stages are needed. Especially large numbers of larvae are needed, as this is the stage that will be targeted with nematodes. One of the aims of the current study was to assess various artificial diets for rearing larvae of P. callosus. Though adult weevils were easily collected from orchards, it was very difficult to obtain large numbers of larvae. Modified versions of an agar diet, as well as different carrot based diets, were tested at 21°C. The highest percentage survival obtained for the agar diet was 50% and 60% for one type of carrot diet. A better rearing method proved to be that of planting full-grown carrots in pots, kept at 25°C, resulting in the attainment of the highest percentage survival rate of 90%. A study was undertaken to assess how long, and at what temperature, P. callosus eggs could be stored. A mean percentage hatch of 45.7% was obtained when eggs were stored at 4°C for 70 days. Eggs started hatching after 47 days and 10 days, when stored at temperatures of 11°C and 14°C, respectively. If the aim of the employment of such a method is only to delay egg hatching, the two temperatures (11°C and 14°C) will be suitable. For the following part of the study, several entomopathogenic nematode isolates were evaluated for their potential use as biological control agents against P. callosus. The susceptibility of P. callosus larvae and adults to nematode infection was assessed in the laboratory by screening for their mortality, using different nematode isolates. Larvae were found to be more susceptible to nematode infection than adults. Heterorhabditis isolates were found to cause higher levels of mortality than the Steinernema isolates during screening, when a concentration of 400 infective juveniles (IJ) per insect V was used. Biological characteristics, such as the effect of different temperatures on nematode activity and the minimum concentration of nematodes needed to obtain acceptable levels of control for P. callosus, were also investigated. The percentage mortality ranged from no infection to 75% after four days for the larvae, and the SF41 isolate of Heterohabditis zealandica was selected as the most promising isolate for further laboratory experiments. The vertical movement of nematodes in sand, compared with such movement in sandy loam soil, and the biology of H. zealandica in P. callosus larvae was also investigated in laboratory bioassays. After four days, the LD50 and LD90 values were 96 IJ/50 μl and 278 IJ/50 μl, respectively. Nematodes were found to be inactive at 11°C, with the highest mortality rate of P. callosus resulting from nematode infection being recorded at 25°C. A higher percentage mortality rate was obtained with the sandy loam soil (95.2%) than with the sand (77.5%). Heterorhabditis zealandica could successfully complete its life cycle in 6th instar P. callosus larvae. The study showed that P. callosus larvae are suitable hosts for H. zealandica, and that the control of P. callosus in the field by the selected isolate holds promise. The persistence of the SF41 isolate of H. zealandica at different concentrations was investigated in the last part of the study. The experiment took place in a blueberry orchard, subject to a high rate of infestation by P. callosus. Concentrations of 0, 20, 30 and 45 IJ/cm2 were topically applied, with persistence being evaluated for days 1, 35 and 84. Percentage persistence for 30 IJ/cm2 was calculated as 87.5% for days 35 and 84. The persistence of soil samples taken on day one, and kept in plastic containers at room temperature, was again evaluated on day 128, with the finding that both 30 IJ/cm2 and 45 IJ/cm2 caused 100% mortality of Tenebrio molitor (L.). Results indicated good persistence of H. zealandica after 84 days in field conditions, with a high maintenance of P. callosus populations. The study indicated the potential use of H. zealandica for the control of P. callosus, with the possibility of persistence for at least three months. Future research into the control of P. callosus with nematodes should aim to investigate the technical aspects of field application. The current study shows that entomopathogenic nematodes have potential for controlling the soil stages of P. callosus. The capacity to rear large numbers of P. callosus larvae in the laboratory, for later use in laboratory and field trials, is of key importance.
AFRIKAANSE OPSOMMING: Die gebande vrugtekalander, Phlyctinus callosus (Schönherr), is ʼn groot plaag in appel- en nektarienboorde sowel as wingerde in die suidelike gebiede van die Wes-Kaap. Phlyctinus callosus word nie voldoende beheer nie, en plaagdoders wat voorheen gebruik is om dié insek in toom te hou, het doeltreffendheid ingeboet weens weerstandontwikkeling teen piretroϊede en asefaat. ʼn Alternatiewe beheermetode is stamsperbande. Omdat die kalanders nie kan vlieg nie, moet hulle teen stamme uitklim om die vrugte te bereik. Stamsperbande versper dus die insekte se toegang tot die vrugte, maar is baie arbeidsintensief. Meer haalbare metodes vir die beheer van P. callosus is daarom dringend nodig, en die gebruik van entomopatogeniese nematodes blyk ʼn besliste moontlikheid te wees. Entomopatogeniese nematodes, wat tot die Steinernematidae en Heterorhabditidae behoort, is uitstekende biobeheermiddels vir insluiting by geϊntegreerde plaagbeheerprogramme. Eiers wat onderskeidelik by 11°C en 14°C geberg is, het ná 47 en 10 dae onderskeidelik begin uitbroei. Indien die doel is om die eiers slegs stadiger te laat uitbroei, sal hierdie twee temperature dus geskik wees. VII Hierna is verskeie entomopatogeniese nematode-isolate vir moontlike gebruik as biologiese beheermiddels vir P. callosus beoordeel. Phlyctinus callosus larwes en volwassenes se vatbaarheid vir nematode infeksie is in die laboratorium bepaal deur dit met behulp van verskillende nematodeisolate vir mortaliteit te toets. Dié toetse het getoon dat larwes meer vatbaar is vir nematode infeksie as volwassenes. In die proefnemings het die Heterorhabditis-isolate hoër mortaliteit as die Steinernema-isolate veroorsaak teen ʼn konsentrasie van 400 infektiewe larwes (IJ) per insek. Biologiese eienskappe, soos die uitwerking van verskillende temperature op nematode aktiwiteit, sowel as die minimum konsentrasie nematodes om aanvaarbare vlakke van beheer uit te oefen, is ondersoek. Konsentrasies van 0, 20, 30 en 45 IJ/cm2 is op die grond (uitwendig) toegedien, en oorlewing is op dag 1, 35 en 84 gemeet. Die persentasie oorlewing vir die 30 IJ/ cm2 konsentrasie was 87.5% op sowel dag 35 as 84. Oorlewing in grondmonsters wat op dag een ingesamel en by kamertemperatuur in plastiekhouers geberg is, is weer op dag 128 beoordeel. Daar is bevind dat sowel die 30 IJ/cm2 as die 45 IJ/cm2 konsentrasie 100% mortaliteit by T. molitor veroorsaak het. Heterorhabditis zealandica blyk ʼn goeie oorlewing te hê ná 84 dae in veld kondisies wat erg met P. callosus besmet is, en is dus ʼn moontlike beheermiddel vir P. callosus, met potensiële oorlewing vir minstens drie maande.

Brazil is the largest citrus producer, being one of its main exports of agro-industrial chain. One of the factors that affect the quality and production of the final product is the Brown Spot Alternaria, caused by the fungus Alternaria alternata. The disease causes leaf drop, dry branches and fruit drop in tangerines and their hybrids. To control this disease fungicides are used, however, the brown spot is a difficult to control disease, may be required from 7 to 10 applications of fungicide, appearance of resistance of fungi, intoxication in people and environment contamination owing the application of fungicides. Alternative methods such as biological control with applications antagonistic microorganisms and plant nutrition are options of smaller impact to the environment. Therefore, the objective of this work was to evaluate the potential of Trichoderma spp., Bacillus subtilis and fertilizer, in control of Alternaria Brown Spot in citrus plants Ponkan tangerine. The experiment was carried out in a randomized design, consisting of eight treatments with four replicates. The experiments were evaluated for the incidence of the disease. The data showed that the best control was the chemical treatment, but the treatments with Trichoderma spp., Trichoderma spp. + Fertilizer and Trichoderma spp. + Bacillus subtilis presented excellent control rates, and may even replace chemical treatment in the control of this disease.

Повний текст джерелаENGLISH ABSTRACT: The grain chinch bug, Macchiademus diplopterus, is an endemic pest of cultivated grain crops and wild grasses in the south-western Cape region of South Africa. In early summer when host plants dry out, adult grain chinch bugs aggregate in large numbers in shelter sites in surrounding areas and enter into aestivation. These shelter sites sometimes include the stalk or calyx ends of fruit, and shelter-seeking bugs can also contaminate export fruit cartons, consequently posing a phytosanitary/quarantine risk to importing countries. Presently, there are no feasible pre- or post-harvest control measures to manage this quarantine risk. The aggregating behaviour of grain chinch bugs suggests the involvement of pheromones. Therefore, investigating the chemical ecology of grain chinch bugs for potential use in control measures is the focus of the first research chapter of this study. Gas chromatography-mass spectrometry (GC-MS) was used to identify headspace volatiles collected from aggregating bugs. Olfactometer bioassays were conducted to assess the attractiveness of each gender to separate sexes, individual compounds and a mixture of the compounds as a formulated lure. The lure was tested in field trapping trials with delta and bucket traps. In the bioassays with the live insects the response of each gender to live females was greater than the responses of each gender to live males, suggesting that females may disseminate the pheromones more efficiently than males. The following eight volatile compounds were indentified from the GC-MS analysis: hexanal, (E)-2-hexenal, (E)-2-hexenol, (E)-2-hexenyl acetate, (E)-2-octenal, (E)-2-octenol, (E)-2-octenyl acetate and tridecane. In the bioassays with individual compounds, three of these eight compounds, hexanal, (E)-2-hexenal, and tridecane, elicited attraction of both females and males. The formulated lure was attractive to both males and females in the laboratory bioassay, but this attraction was not evident in the field. In the field, there was only one occasion when a significantly higher number of bugs were caught in baited traps compared to unbaited traps. Trap catches were very low compared to the actual level of infestation in the field which was evident from corrugated cardboard bands tied around tree trunks which contained many sheltering bugs. The low trap catches seen in the field were partly due to competition between the synthetic pheromone lure and the natural pheromones emitted by aggregating live insects. Also, the characteristic shelter-seeking behaviour of grain chinch bugs influenced trap catches, as more bugs were found in places that provide shelter, like cardboard bands and walls of the delta traps. This behavior of aestivating bugs could be used to the advantage of trapping bugs by integrating sheltering sites into traps in future trials. Also, the lure needs to be improved for optimum efficiency in the field. The second research chapter also addresses the quarantine risk posed by grain chinch bugs, by investigating the thermal biology of bugs to ultimately facilitate the development of effective post-harvest treatments. Critical thermal minimum and maximum temperatures (CTmin and CTmax) of both active and aestivating bugs were subjected to critical thermal limits analysis. The CTmin and CTmax of aestivating bugs were not affected by gender (p > 0.05). There was a decrease in CTmin from the active period into aestivation for both males (2.8°C to 1.0°C (± 0.1)) and females (2.1°C to 0.6°C (± 0.1)). Also, for CTmax there was an increase in tolerance from the active period into the aestivation period for both males (49.9°C to 51.0°C (± 0.1)) and females (49.9°C to 51.5°C (± 0.1)). To determine the plasticity of grain chinch bug thermal tolerance, aestivating bugs at 27 weeks into aestivation, were acclimated at different temperatures and photoperiods [18°C (10L:14D) and 26°C (16L:8D)] for a period of seven days. Both low (18°C) and high (26°C) acclimation temperatures and photoperiods increased CTmin of aestivating grain chinch bugs at 14 weeks from 0.8°C to -1.2°C and -0.1°C (± 0.1) respectively. However, CTmax was not altered by acclimation temperatures (p > 0.82). Field temperatures at collection sites were recorded to compare to grain chinch bugs thermal tolerance levels exhibited in the laboratory. These results, as well as the effects of acclimation treatments on the CTmin of bugs, have implications for post-harvest treatments, and understanding the quarantine risk posed to importing countries. The information generated from this study can be used to further advance the development of both effective pre-harvest and post-harvest control measures to reduce grain chinch bug quarantine risk.
AFRIKAANSE OPSOMMING: Die graanstinkluis, Macchiademus diplopterus, is 'n endemiese plaag van aangeplante graangewasse en wilde grasse in die Suidwes Kaap-provinsie van Suid-Afrika. In die vroeë somer wanneer gasheerplante uitdroog, soek groot getalle volwasse graanstinkluise skuiling in die omliggende gebiede en gaan in ʼn somerrusperiode. Hierdie skuilplekke sluit soms die stam of kelk eindes van vrugte in en graanstinkluise kan ook uitvoer-vrugte kartonne kontamineer. Gevolglik word lande wat vrugte uit Suid-Afrika invoer, aan die fitosanitêre kwarantynrisiko van stinkluisbesmetting blootgestel. Tans is daar nie haalbare voor- of na-oes beheermaatreëls om hierdie kwarantyn risiko te bestuur nie. Die aggregasiegedrag van graanstinkluise dui op die betrokkenheid van ʼn feromoon. ‘n Ondersoek van die chemiese ekologie van die graanstinkluis vir moontlike gebruik in beheermaatreëls is die fokus van die eerste gedeelte van hierdie studie. Gaschromatografie-massaspektrometrie (GC-MS) is gebruik om die vlugtige organiese verbindings in die bodamp van die saamgetrosde stinkluise te identifiseer. Olfaktometriese biotoetse is uitgevoer om die aantreklikheid van die insekte vir die teenoorgestelde geslag te bepaal, asook van die individuele verbindings en 'n mengsel van die verbindings as 'n geformuleerde lokmiddel in lokvalle. Die lokmiddel is getoets in veldproewe met deltatipe en emmertipe lokvalle. In die olfaktometriese biotoetse met die lewende insekte is die reaksie van beide geslagte teenoor lewende wyfies groter as die reaksie van die geslagte teenoor mannetjies, wat daarop dui dat wyfies die feromoon meer doeltreffend as mannetjies versprei. Die geformuleerde lokmiddel was aantreklik vir beide geslagte in laboratorium toetse, maar soortgelyke lok is nie in die veld gevind nie, waar daar net een keer 'n aansienlike groter getal graanstinkluise met lokmiddel gevang is in vergelyking met lokvalle sonder lokmiddel. Die getal graanstinkluise in lokvalle was baie laag in vergelyking met die werklike vlak van besmetting in die veld, wat duidelik geblyk het uit die getalle graanstinkluise wat skuiling gesoek het in die geriffelde karton bande wat om boomstamme vasgemaak was. Die lae lokvalvangste in die veld was deels te wyte aan die kompetisie tussen sintetiese feromoon en die natuurlike feromoon van saamgetrosde insekte. Die kenmerkende aggregasiegedrag van graanstinkluise het lokvalvangste beïnvloed, aangesien meer stinkluise gevind is in plekke wat skuiling bied, soos die kartonbande en die binnekant van die delta-lokvalle. Hierdie skuilings van graanstinkluise kan in toekomstige proewe uitgebuit word deur vir meer skuilplek in lokvalle voorsiening te maak. Die formulering en die aanbieding van die lokmiddle moet ook verbeter word vir 'n optimale doeltreffendheid in die veld. In die tweede hoofstuk word die kwarantynrisiko van die graanstinkluis aangespreek deur die ondersoek van die termiese biologie van stinkluise om uiteindelik die ontwikkeling van doeltreffende na-oes behandelings te fasiliteer. Kritiese termiese minimum en maksimum temperature (CTmin en CTmax) van beide aktiewe en rustende graanstinkluise is bepaal deur analise van die kritiese termiese beperkings van die insek. Die CTmin en CTmax van rustende graanstinkluise is nie geraak deur geslag nie (p > 0.05). Daar was 'n afname in CTmin van die aktiewe tydperk tot in rus, vir beide manlike (2.8°C tot 1.0°C (± 0.1)) en vroulike insekte (2.1°C tot 0.6°C (± 0.1)). Ook vir die CTmax was daar 'n verbetering in toleransie vanaf die aktiewe tydperk tot in die rusperiode vir beide manlike (49.9°C tot 51.0°C (± 0.1)) en vroulike insekte (49.9°C tot 51.5°C (± 0.1)). Om die aanpasbaarheid van die termiese toleransie van die graanstinkluis te bepaal, is graanstinkluise 27 weke na aanvang van die rusperiode geakklimatiseer by verskillende temperature en fotoperiodes [18°C (10L: 14D) en 26°C (16L: 8D)] vir 'n tydperk van sewe dae. Beide lae (18°C) en hoë (26°C) akklimatiseringstemperature en fotoperiodes het onderskeidelik die CTmin van rustende graanstinkluise op 14 weke verhoog van 0.8°C tot -1.2°C en -0.1°C (± 0.1). Daar is egter geen effek op CTmax deur akklimasie temperature nie (p > 0.82). Veldtemperature is ook bepaal om te vergelyk met graanstinkluis termiese toleransie vlakke wat in die laboratorium bepaal is. Hierdie resultate, sowel as die gevolge van die akklimasie behandelings op die CTmin van graanstinkluise, het implikasies vir na-oes behandelings, en begrip van die kwarantyngevaar wat dit inhou vir vrugte-invoerlande. Die inligting wat uit hierdie studie voortvloei, kan gebruik word om die ontwikkeling van beide effektiewe voor-oes en na-oes beheermaatreëls te bevorder en om die kwarantynrisiko wat graanstinkluise inhou, te verminder.

Sterile insect release is applied as a component of area-wide integrated pest management and includes the mass-rearing, sterilization and the release of the sterile insects. For sterile insect release, the improvements of rearing methods in terms of the quality of the diet ingredients and the economical aspect of the rearing method are examined. The effect of genetically modified maize meal, containing the Bacillus thuringiensis gene, in an artificial medium for codling moth rearing, is determined. The use of even a small amount of Bacillus thuringiensis resulted in larval mortality and prolonged development. These results are detrimental to a mass-rearing facility and must be considered by any rearing facility that uses genetically modified maize meal if the insect is sensitive to the gene. An alternative to maize meal in the artificial medium was tested and whole wheat flour was considered to be a suitable replacement. Agar agar is an expensive gelling agent used in the artificial medium. An alternative for agar agar (Kelcogel, Elastigel and carrageenen) is tested and the biological effect on codling moth is determined. Factors such as mortality, pupal and moth weight, longevity, fecundity and development time were used as quality parameters. Results showed that Elastigel was a suitable replacement for agar agar, with bigger pupae and moths, higher fecundity and increased longevity. The economical advantage of the replacement is a 40.91% reduction of the diet cost. The other gelling agents tested also gave acceptable results and can be considered if shortages of agar agar or Elastigel occur. A new method of mass-rearing codling moth larvae in a closed rearing system using large trays placed in a ventilated box is designed. This method is more cost and space effective as a smaller area is needed to rear a large number of moths. The risk of diet contamination is less because of the closed environment and more economical and effective air handling. This is the first report of its kind to describe the mass-rearing of codling moth in a closed environment and the risks involved in using genetically modified maize meal in an artificial diet for the codling moth. These results should be incorporated into existing mass-rearing facilities or taking into consideration when designing new mass-rearing facilities.
AFRIKAANSE OPSOMMING: Die kodlingmot, Cydia pomonella, is van ekonomiese belang vir die Suid-Afrikaanse kernvrugte bedryf. Die steriele insek tegniek word gebruik as ‘n komponent in area-wye geïntegreerde plaagbeheer en sluit in die massa-aanteel, sterilisering en vrylaat van steriele insekte. Vir die steriele insek tegniek is die verbetering van die massa-aanteel van die kodlingmot in terme van kwaliteit van die dieet en die ekonomiese aspek van die aanteel metode ondersoek. Die effek van genetiese gemanipuleerde mieliemeel wat die Bacillus thuringiensis geen bevat, in ‘n kunsmatige voedselmedium vir die aanteel van kodlingmot, is bepaal. Daar is gevind dat die gebruik van selfs ‘n klein persentasie Bacillus thuringiensis in die mieliemeel, mortaliteit en ‘n verlengde lewenssiklus in kodlingmot veroorsaak. Die gevolge is nadelig vir ‘n massa-aanteel fasiliteit en behoort in ag geneem te word vir enige insek wat op ‘n kunsmatige medium, wat mieliemeel bevat, geteel word, mits die insek sensitief is vir Bacillus thuringiensis. ‘n Alternatiewe bestanddeel vir mieliemeel, volkoringmeel, word aanbeveel. Agar agar is ‘n duur verdikkingsagent wat in kunsmatige mediums gebruik word. ‘n Alternatief vir agar agar (Kelcogel, Elastigel en carrageenen) is getoets en die biologiese effek op die kodlingmot is bepaal. Faktore soos mortaliteit, papie en mot gewig, langlewendheid, vrugbaarheid en lengte van lewenssiklus was gebruik as kwaliteit parameters. Resultate het getoon dat Elastigel ‘n geskikte plaasvevanger is van agar agar, met groter papies en motte, groter vrugbaarheid en langlewendheid. Die ekonomiese gevolg van die plaasvervanger, is ‘n vermindering van 40.91% van die dieetkoste. Die ander verdikkingagente wat is getoets is, het aanvaarbare resultate gelewer wat noodsaaklik is indien daar ‘n tekort van Elastigel of agar agar ontwikkel. ‘n Nuwe metode van massa-aanteel van kodlingmot larwes is bepaal. Die metode behels ‘n geslote sisteem, waar groter aanteel bakke in ‘n geslote, geventileerde boks geplaas word. Die metode is koste en spasie effektief en ‘n kleiner area word benodig om ‘n groter aantal motte te lewer. Die risiko van kontaminasie van die dieet word verminder as gevolg van die geslote sisteem wat gebruik word en meer ekonomiese en effektiewe lugversorging word gebruik. Hierdie is die eerste verslag van sy soort wat die massa-aanteel van kodlingmot in ‘n geslote sisteem beskryf en wat die risiko aandui van geneties gemanipuleerde mieliemeel in ‘n kunsmatige medium vir die kodlingmot. Hierdie resultate behoort in ag geneem te word vir reeds bestaande massa-aanteel fasiliteite of met die ontwerp van nuwe massa-aanteel fasiliteite.

Повний текст джерелаIn this scope, the development of an effective trapping method would be very useful to assess the bark-beetle presence at previously uninfested sites, or predator establishment after release or natural spread. We demonstrated the efficiency of oxygenated monoterpenes-baited kairomone traps to monitor R. grandis in various epidemiological conditions, including areas localized behind or at the limit of the pest’s distribution, or in areas where artificial releases were performed. Because the predator is strictly species-specific, another exciting possibility offered by the kairomone trapping is the indirect monitoring of the pest itself in areas of unknown status (e.g. areas under colonization, or considered as at risk at medium- term).
R. grandis is also considered as one of the most valuable natural enemies to strike aggressive North-American Dendroctonus species. In this respect, R. grandis has been recently applied in a neo-classical biological program against the red turpentine beetle D. valens, which invaded China from North America in the late 1990’s. In laboratory tests conducted on pine logs in the laboratory, or on living pine trees in the field, we demonstrated that R. grandis adults can successfully enter and reproduce into D. valens galleries.
In Europe, R. grandis is the only species regularly found in the brood systems of D. micans, where adults and larvae attack the gregarious larvae of their prey. In such enclosed systems, R. grandis’ functional response is therefore influenced by various interrelated components, such as the prey density, the predator density, or the prey distribution. Measuring the predator’s success in terms of larval survival and growth rates, we demonstrated the time spent by R. grandis larvae to wound and kill their prey to be the main factor limiting their development. This factor may be considerably influenced by the proportions of diseased, wounded or molting prey rise in the brood system, for instance as a result of an increase in prey density, or due to the presence of conspecific adults (which wound their prey but do not consume them entirely). Furthermore, our tests suggest that no cannibalism or noticeable intraspecific competition occurred between R. grandis larvae, whereas some lighter mode of competition probably took place.
R. grandis also exhibits a reproductive numerical response to prey density, which mainly relies on the perception of chemical stimuli and inhibitors released in the bark beetle brood system. In the current study, we developed a non-destructive approach to follow the dynamics of volatile compound production, using sequential sample collection on SPME fibers. Our tests demonstrated that the larval activity of D. micans or D. valens strongly influences the release of some oxygenated monoterpenes. However, our attempts to correlate the relative quantities of some identified chemicals to offspring production were less successful as it concerns the identification of potential oviposition stimuli and inhibitors.
The problematic rose by the progression of D. micans, as well as detailed results of each of the described above studies are discussed in the two published papers and the three manuscripts forming this thesis. Bringing all these studies together, several perspectives are also presented in the general discussion.

Ravageur des épicéas, Dendroctonus micans est toujours en voie d’extension en France, en Turquie, en Angleterre et au Pays de Galles. Dans la plupart de ces zones, le dendroctone est accompagné de manière naturelle par son prédateur monospécifique, Rhizophagus grandis. Depuis les années 1960, le prédateur a également fait l’objet d’une production de masse et de programmes de lâchers dans les zones d’arrivée récente du scolyte.
Dans le cadre de la lutte biologique contre D. micans, les gestionnaires forestiers doivent donc estimer au plus tôt la présence du ravageur dans des zones jusque là indemnes, mais également vérifier l’établissement du prédateur par progression naturelle ou résultant d’introductions délibérées. Dans la présente étude, nous avons démontré l’efficacité de pièges d’interception appatés à l’aide de monoterpènes oxygénés pour la capture de R. grandis. Celle-ci s’est faite dans différentes conditions épidémiologiques, incluant notamment des zones situées en arrière du front de progression du scolyte et des zones où des lâchers artificiels ont été réalisés. Comme R. grandis est strictement inféodé au dendroctone, un autre avantage de la technique est la possibilité de réaliser un dépistage indirect du ravageur dans les zones où son statut est incertain (zones en cours de colonisation, ou considérées comme à risque à moyen terme).
Par ailleurs, R. grandis est également considéré comme un des meilleurs ennemis naturels potentiels pour lutter contre d’autres espèces de Dendroctonus aggressifs. Dans cette optique, R. grandis a été récemment utilisé dans un programme de lute biologique contre D. valens, ravageur invasif arrivé en Chine dans la fin des années 1990 en provenance d’Amérique du Nord. Nous avons démontré la capacité de R. grandis à s’introduire et à se reproduire dans les galeries de D. valens lors de tests de laboratoire, mais aussi sur des arbres vivants en pinèdes.
En Europe, R. grandis est strictement inféodé aux galeries de D. micans, où larves et adultes du prédateur s’attaquent aux larves grégaires du scolyte. Dans ce système clos, la réponse fonctionelle de R. grandis est influencée par plusieurs facteurs étroitement corrélés, la densité de proies, la densité de prédateurs, et la distribution des proies. En mesurant l’efficacité de prédation de R. grandis en termes de survie des larves et de taux de croissance, nous avons démontré l’influence sur leur développement du temps passé par les larves à blesser et à tuer leurs proies. Ce facteur est par ailleurs fortement dépendant de la proportion de larves malades, blessées ou en cours de mue au sein du système ;une proportion qui peut augmenter en réponse à une augmentation de la densité de proies, ou lorsque des adultes sont présents (ceux-ci blessent les proies mais ne les consomment pas entièrement). Enfin, nos tests suggèrent qu’il n’existe que peu de cannibalisme ou de compétition intraspécifique entre larves de R. grandis, tandis que des modes de compétition moins importants prennent vraisemblablement place.
R. grandis présente également une réponse numérique reproductive à la densité de proies disponibles, principalement basée sur la perception de stimuli et d’inhibiteurs présents dans les galeries du scolyte. Par la collecte de composés volatils présents dans ces systèmes à l’aide de fibres SPME, nous avons développé une méthode non-destructive pour suivre la dynamique de production des médiateurs chimiques. Nos tests ont démontré que l’activité des larves de D. micans ou D. valens influence fortement la dynamique de production de certains monoterpènes oxygénés. En revanche, il n’a pas été été possible de corréler les différents composés identifiés au nombre de larves de R. grandis présentes dans le système.
La problématique soulevée par la progression de D. micans, de même que les résultats détaillés de chacune des études décrites ci-dessus sont discutés dans les deux papiers publiés et les trois manuscrits formant cette thèse. Les différentes perspectives apportées par ce travail sont également présentées dans la discussion générale.
