Повний текст джерелаThe prime number theorem, first proved in 1896 using complex analysis, gives the main term for the asymptotic distribution of prime numbers. It was not until 1949 that the first so-called "elementary" proof was published: it rests strictly on real analysis.In 1999, Wen Chao Lu obtained by an elementary method an error term in the prime number theorem very close to the one provided by the zero-free region of the Riemann zeta function given by La Vallée Poussin at the end of the 19th century. In this thesis, we make Lu's result explicit in order, firstly, to give the best error term obtained by elementary methods so far, and secondly, to explore the limits of his method

Plus précisément, on construit un test basé sur une quantité qui permet de canaliser toute l'information liant X à Y : la fonction de répartition conditionnelle de Y sachant (X = x) définie par F(y|x)=P(Y<=y|X=x). Notre test compare la valeur prise par l'estimateur polynômial local de F(y|x) à une estimation paramétrique du modèle supposé et rejette sa
validité si la distance entre ces deux quantités est trop grande. Dans un premier temps, on considère le cas où la fonction de répartition supposée est entièrement spécifiée et, dans
ce contexte, on établit le comportement asymptotique du test. Dans la deuxième partie du travail, on généralise ce résultat au cas plus courant en pratique où le modèle supposé contient un certain nombre de paramètres inconnus. On étudie ensuite la puissance locale du test en déterminant son comportement asymptotique local sous des suites d'hypothèses contigües. Enfin, on propose un critère de choix de la fenêtre d'ajustement qui intervient lors de l'étape d'estimation polynômiale locale de la fonction de répartition conditionnelle.

Повний текст джерелаThe main objective of this work was to study, using an in vivo approach, a number of proteins essential for haemostasis and coagulation, in pathophysiologicals conditions and by identifying potential determinants of their lifecycle. In the first part of the work, focused on the Von willebrand Factor (VWF)/ADAMTS13 couple, the critical role of VWF in the decrease of ADAMTS13 during sepsis has been highlighted. We also showed that VWF proteolysis by ADAMTS13 is not implicated in its clearance. In the second part of the thesis, the role of glycosylations on the function of both VWF and coagulation factor X (FX) has been studied in vivo. In FX, the major role of the two N-glycosylations present in the activation peptide of the molecule has been demonstrated. For VWF, we emphasized the fact that some O- glycosylations sites are involved in the maintenance of the protein plasma concentration while others are important for the functionality of the protein. This work, based on in vivo experimentations, underlines the importance of the relationship between VWF and ADAMTS13 during physiological processes like VWF clearance or pathophysological processes such as sepsis. This work also allowed us to understand the essential role of some glycosylations in the functionality of glycoproteins. Our approach, entirely using in vivo experiments, highlighted some unsuspected effects which pre-existing literature, based on in vitro experiments, did not indicate

Повний текст джерелаVon Willebrand factor (VWF) is an adhesive plasma protein which primary function is to promote platelet adhesion and aggregation at site of vascular injury. VWF deficient mice are a key element of this work. Indeed, study of VWF deficient mice revealed existence of new functions of this protein, in particular in leukocytes recruitment and in protein targeting to Weibel-Palade bodies. Thus, in a first part, we characterized leukocyte interaction with VWF under flow conditions, establishing implication of PSGL-1 and beta2 integrins on leukocyte surface. We next studied the fate of osteoprotegerin (OPG) and angiopoietin-2 (ang-2) in endothelial cells in absence of VWF, using an in vitro model of endothelial cells in which VWF synthesis is inhibited by small interfering RNA. Our results demonstrated that in absence of VWF, OPG and ang-2 are no longer detected in the cells. In a second part of our work, we also used VWF deficient mice as a tool, to study phenomenon that could only be investigated through in vivo methods. We therefore examined molecular basis of VWF clearance, and we identified VWF mutations associated with increased clearance responsible of von Willebrand disease. We finally tested the role of VWF in metastasis formation in mice, demonstrating its protective effect against metastasis development. In conclusion, this work has contributed to a better comprehension of these new aspects of thé biology and the function of VWF

Повний текст джерелаWe revisit the parametrix method in order to obtain a gaussian two-sided bound for the fundamental solution of a general parabolic operator which is not in a divergence form. Then we use the fact that the Neumann Green function of a general parabolic operator on a regular bounded domain can be constructed as a perturbation of the fundamental solution by a simple-layer potential in order to establish a Gaussian lower bound for this Neumann Green function. The key point of the proof lies in the time-regularising effect of the single-layer potential. We also prove that this method can be adapted to get a lower Gaussian bound for the Neumann heat kernel of the Laplace-Beltrami operator on an open subset of a compact Riemannian manifold. In a second part, we prove a new Gaussian upper bound for the Neumann heat kernel of the Laplace-Beltrami operator on a Lipschitz domain of a complete Riemannian manifold. The principal interest of this new upper bound is that we do not have the usual exponentiel terme in time in this upper bound. In a last part, as an application of the Gaussian estimates, we give a compactness result of isospectral potentials which is in relation to an asymptotic formule for the heat kernels
