Exogenous lactate administration has more recently been investigated for its various prophylactic effects. Lactate derived from potential functional foods, such as fermented oyster extract (FO), may emerge as a practical and effective method of consuming exogenous lactate. The current study endeavored to ascertain whether the lactate derived from FO may act on muscle cell biology, and to what extent this may translate into physical fitness improvements. We examined the effects of FO in vitro and in vivo, on mouse C2C12 cells and exercise performance indicators in mice, respectively. In vitro, biochemical analysis was carried out to determine the effects of FO on lactate content and muscle cell energy metabolism, including adenosine triphosphate (ATP) activity. Western blot analysis was also utilized to measure the protein expression of total adenosine monophosphate-activated protein kinase (AMPK), p-AMPK (Thr172), lactate dehydrogenase (LDH), succinate dehydrogenase (SDHA) and peroxisome proliferator-activated receptor gamma coactivator-1α (PGC-1α) in response to FO administration. Three experimental groups were formed: a positive control (PC) treated with 1% horse serum, FO10 treated with 10 μg/mL and FO50 treated with 50 μg/mL. In vivo, the effects of FO supplementation on exercise endurance were measured using the Rota-rod test, and Western blot analysis measured myosin heavy-chain 2 (MYH2) to assess skeletal muscle growth, alongside p-AMPK, total-AMPK, PGC-1α, cytochrome C and UCP3 protein expression. Biochemical analysis was also performed on muscle tissue to measure the changes in concentration of liver lactate, lactate dehydrogenase (LDH), glycogen and citrate. Five groups (n = 10/per group) consisted of a control group (CON), exercise group (Ex), positive control treated with Ex and 500 mg/kg Taurine (Ex-Tau), Ex and 100 mg/kg FO supplementation (Ex-FO100) and Ex and 200 mg/kg FO supplementation (Ex-FO200) orally administered over the 4-week experimental period.FO50 significantly increased PGC-1α expression (p < 0.001), whereas both FO10 and FO50 increased the expression of p-AMPK (p < 0.001), in C2C12 muscle cells, showing increased signaling important for mitochondrial metabolism and biogenesis. Muscle lactate levels were also significantly increased following FO10 (p < 0.05) and FO50 (p < 0.001). In vivo, muscle protein expression of p-AMPK (p < 0.05) and PGC-1α were increased, corroborating our in vitro results. Cytochrome C also significantly increased following FO200 intake. These results suggest that the effects of FO supplementation may manifest in a dose-response manner. FO administration, in vitro, and supplementation, in vivo, both demonstrate a potential for improvements in mitochondrial metabolism and biogenesis, and even for potentiating the adaptive effects of endurance exercise. Mechanistically, lactate may be an important molecule in explaining the aforementioned positive effects of FO.

Flavonoids are an abundant class of naturally occurring compounds with broad biological activities, but their limited abundance in nature restricts their use in medicines and food additives. Here we present the synthesis and determination of the antibacterial and antioxidant activities of twenty-two structurally related flavonoids (five of which are new) by scientifically validated methods. Flavanones (FV1–FV11) had low inhibitory activity against the bacterial growth of MRSA 97-7. However, FV2 (C5,7,3′,4′ = OH) and FV6 (C5,7 = OH; C4′ = SCH3) had excellent bacterial growth inhibitory activity against Gram-negative E. coli (MIC = 25 µg/mL for both), while Chloramphenicol (MIC = 25 µg/mL) and FV1 (C5,7,3′ = OCH3; 4′ = OH) showed inhibitory activity against Gram-positive L. monocytogenes (MIC = 25 µg/mL). From the flavone series (FO1–FO11), FO2 (C5,7,3′,4′ = OH), FO3 (C5,7,4′ = OH; 3′ = OCH3), and FO5 (C5,7,4′ = OH) showed good inhibitory activity against Gram-positive MRSA 97-7 (MIC = 50, 12, and 50 µg/mL, respectively), with FO3 being more active than the positive control Vancomycin (MIC = 25 µg/mL). FO10 (C5,7= OH; 4′ = OCH3) showed high inhibitory activity against E. coli and L. monocytogenes (MIC = 25 and 15 µg/mL, respectively). These data add significantly to our knowledge of the structural requirements to combat these human pathogens. The positions and number of hydroxyl groups were key to the antibacterial and antioxidant activities.

Neste trabalho, a atividade alelopática de Schinus terebinthifolius Raddi, a aroeira-vermelha, foi avaliada utilizando-se Lactuca sativa L., a alface, como planta alvo. Os bioensaios foram conduzidos em laboratório com extratos aquosos preparados a partir de folhas e frutos, resultando nos seguintes tratamentos: folha 5% (Fo5), folha 10% (Fo10), folha 20% (Fo20), fruto 5% (Fr5), fruto 10% (Fr10), fruto 20% (Fr20) e água destilada (Controle). A germinação foi acompanhada por seis dias e os resultados expressos em termos de Porcentagem de Germinação (PG), Índice de Velocidade de Germinação (IVG) e Biomassa das Plântulas (BP). Os dados foram submetidos à análise de variância (One-way ANOVA) seguida pelo teste de Tukey (p<0,05). Ápices radiculares foram processados em glicolmetacrilato para análise histológica qualitativa ao microscópio óptico. A germinação foi totalmente inibida pelos tratamentos de maior concentração (Fo20 e Fr20) e todas as variáveis analisadas apresentaram diferenças significativas com relação ao controle, exceto PG e BP no tratamento Fr5, indicando forte efeito alelopático da aroeira-vermelha. Necrose do ápice radicular e alterações celulares nas raízes submetidas aos extratos também foram evidenciadas. Os resultados sugerem que S. terebinthifolius tem potencial para ser utilizada como fonte de compostos bioativos no manejo de ervas daninhas em agroecossistemas.

Structural refinements from single-crystal X-ray diffraction data are reported for olivine with a composition of Fo100 (forsterite Mg2SiO4, synthetic), Fo80 and Fo62 (~Mg1.6Fe0.4SiO4 and ~Mg1.24Fe0.76SiO4, both natural) at room temperature and high pressure to ~8 GPa. The new results, along with data from the literature on Fo0 (fayalite Fe2SiO4), were used to investigate the previously reported structural mechanisms which caused small variations of olivine bulk modulus with increasing Fe content. For all the investigated compositions, the M2 crystallographic site, with its bonding configuration and its larger polyhedral volume, was observed to control the compression mechanisms in olivine. From Fo100 to Fo0, the compression rates for M2–O and M1–O bond lengths were observed to control the relative polyhedral volumes, resulting in a less-compressible M1O6 polyhedral volume, likely causing the slight increase in bulk modulus with increasing Fe content.

A total of 20 chickpea seed samples were collected from BARI, Gazipurand different locations of Savar, Dhaka district. Blotter method was used for detection of the associated fungi of chickpea seeds. Altogether 14 fungi comprising 12 genera namely Aspergillus flavus, Aspergillus niger, Aspergillus sp, Botryodiplodia theobromae, Botryti cinerea, Curvularia lunata, Chaetomium globosum, Cladosporium sp., Colletotrichum dematium, Fusarium oxysporum, Macrophomina phasaelina, Rhizoctonia solani, Rhizopus stolonifer and Stemphylium sarciniforme were isolated from chickpea seed samples. Among the fungi isolated fungi, F. oxysporumwas most prevalent with an average incidence of 18.95% and found in all the seed samples. The germination of seed samples varied from 55-90 % on blotter. The pathogenic fungi and other storage fungi like Aspergillus caused lower the germination of the seeds.A pathogenicity test was conducted with 20 isolates of F. oxysporum against their origin of chickpea seed samples in pot culture. The pathogenecity ranged from weak to highly pathogenic. The isolates FO 19, FO 17, FO 11 and FO 18 were highly virulent. The isolates FO 9, FO10 and FO 15 were virulent and rest of the isolates were moderately virulent. The isolates FO 2 and FO 3 were weak pathogen.J. Environ. AbstractThe Older Giant Dyke Complex is a differentiated alkaline intrusion of Proterozoic age (1154±16 Ma) and is the earliest of the late Gardar intrusions in the Tugtutôq-Ilímaussaq region. The dyke is approximately 20 km long by 0·5 km broad and comprises (i) marginal ‘border group’ rocks of alkali olivine gabbro, grading inwards to ferro-syenogabbro and (ii) an axial ‘central group’ of salic rocks ranging from augite syenite in the WSW to sodalite foyaite in the ENE.Chilled margins contain plagioclase (An53), olivine (Fo53), magnetite, ilmenite, and apatite as liquidus phases and later-crystallized augite (Di69Hd27Ac4) and biotite (Annite32). The coexisting Fe-Ti oxides indicate fO2 and T values just below the synthetic QFM buffer curve. In the border group, plagioclase cores zone into anorthoclase and soda-sanidine rims, olivines reach Fo16, pyroxenes Di32Hd59Ac9, and biotites Annite86. Interstitial pargasitic amphibole appears close to the innermost margins. In the central group, feldspars are all perthitic alkali feldspars and nepheline becomes a major, early crystallizing phase. Olivines range from Fo10-Fo4, in the augite-syenites where they coexist with ferro-salites Di50Hd47Ac3, but olivine is absent from foyaitic assemblages in which the pyroxenes range through aegirine-augite to pure aegirine. Interstitial amphiboles range from ferro-pargasite or hastingsite to katophorite and thence towards arfvedsonite, but are absent from the most differentiated rocks, whereas biotite occurs throughout the entire group in the range Annite71-Annite100.The parental magma, represented by the chilled margins, was a relatively anhydrous alkali olivine-basalt with an initial 87Sr/86Sr ratio of 0.70326. Its high Ti, P, Ba, and F contents are inferred to be features inherited from a primary magma, derived from the mantle as a small partial melt fraction which involved significant amounts of fluor-apatite and phlogopite. While all lithologies are considered as differentiates from this parental magma, there is both a well-defined field junction and a compositional hiatus between the border group and the central group rocks. Mineralogical considerations and REE patterns suggest that the later, more salic (?benmoreitic) magma from which the central group crystallized, related to the parental magma by ol-fsp-ap-FeTi oxide fractionation. Congelation in both border group and central group occurred by side-wall crystallization, but the salic magma became compositionally stratified, with upward concentration of alkalis and volatiles to produce a phonolitic upper facies which is preserved at the ENE end of the intrusion owing to subsequent axial tilting.

This study analyzes the soil water infiltration characteristics under muddy water irrigation and bio-organic fertilizer conditions in the current context of muddy water irrigation rarely being used in agricultural production and in combination with the problems of water resource shortages and low soil fertility in arid and semi-arid regions. An indoor one-dimensional soil column infiltration device was used for studying the effects of four muddy water sediment concentration levels (ρ0: 0; ρ1: 4%; ρ2: 8%; ρ3: 12%) and four bio-organic fertilizer levels (FO0: 0; FO1: 2250 kg·hm−2; FO2: 4500 kg·hm−2; sFO3: 6750 kg·hm−2) on soil water infiltration, evaporation characteristics, and leaching loss. The results demonstrated that a higher muddy water sediment concentration and fertilization level resulted in a smaller wetting front distance and cumulative infiltration amount within the same time, but the infiltration reduction rate (η) gradually increased. The three infiltration models (Kostiakov, Philip, and Horton) were fitted, and it was discovered that all three had good fitting results (R2 > 0.8), with the Kostiakov model displaying the best fit and the Horton model exhibiting the worst fit. The cumulative evaporation amount and evaporation time in muddy water irrigation and fertilization conditions was consistent with the Black and Rose evaporation models (R2 > 0.9), the Black model was proved to be higher than the Rose model. In comparison to ρ0, muddy water irrigation increased conductivity and total dissolved solids (TDS) in the leaching solution, but it reduced cumulative evaporation, soil moisture content, the uniformity coefficient of soil water distribution, and leaching solution volume. Compared with FO0, the application of bio-organic fertilizer increased soil water content and reduced soil water evaporation while also reducing the leaching solution volume, conductivity, and TDS in the leaching solution. The results of this research can provide scientific reference for the efficient utilization of muddy water irrigation and the rational application of bio-organic fertilizer.

Verticillium wilt of olive (VWO), caused by the widespread soil-borne fungus Verticillium dahliae Kleb., is currently the most serious disease affecting olive trees (Olea europaea L.) in all production areas. An integrated management strategy using eco-friendly approaches such as genetic resistance and biological control is considered the most advisable approach for controlling the disease in commercial olive orchards. This study evaluated a non-pathogenic strain of F. oxysporum (FO12) and the grape marc compost CGR03 for reducing inoculum density of V. dahliae and the disease progress in two olive cultivars with different VWO resistance levels. The experiment was conducted under semi-controlled conditions, using a naturally infested soil with two inoculum densities of V. dahliae. The biocontrol treatments (FO12 and CGR03) were previously selected out of 220 natural products as two of the most effective treatments against the pathogen. FO12 and CGR03 treatments significantly reduced pathogen inoculum density in comparison with that of the control (P = 0.05), with minimum microsclerotium amounts of 0.13 g-1 for FO12 and 0.53 g-1 for CGR03, during the experimental period. CGR03 reduced the progression of the disease compared with that in the control (P = 0.05), and FO12 achieved complete control of VWO, since no plants treated with this biological control strain developed VWO symptoms. This study highlights the effectiveness of these biocontrol treatments, and the potential use of eco-friendly approaches for control of VWO.

Rice (Oryza sativa L.) is a very important cereal worldwide, since it is the staple food for more than half of the world’s population. Iron (Fe) deficiency is among the most important agronomical concerns in calcareous soils where rice plants may suffer from this deficiency. Current production systems are based on the use of high-yielding varieties and the application of large quantities of agrochemicals, which can cause major environmental problems. The use of beneficial rhizosphere microorganisms is considered a relevant sustainable alternative to synthetic fertilizers. The main goal of this study was to determine the ability of the nonpathogenic strain Fusarium oxysporum FO12 to induce Fe-deficiency responses in rice plants and its effects on plant growth and Fe chlorosis. Experiments were carried out under hydroponic system conditions. Our results show that the root inoculation of rice plants with FO12 promotes the production of phytosiderophores and plant growth while reducing Fe chlorosis symptoms after several days of cultivation. Moreover, Fe-related genes are upregulated by FO12 at certain times in inoculated plants regardless of Fe conditions. This microorganism also colonizes root cortical tissues. In conclusion, FO12 enhances Fe-deficiency responses in rice plants, achieves growth promotion, and reduces Fe chlorosis symptoms.

Les cellules eucaryotes sont étroitement dépendantes des mitochondries pour leur production d’énergie. Par ailleurs, la chaîne respiratoire représente la principale source de radicaux libres, mutagènes pour l’ADN nucléaire ou mitochondrial. Ainsi, les fonctions mitochondriales sont au coeur du processus de vieillissement et de la physiopathologie de maladies à révélation relativement tardive, diabète de type 2 ou maladies neuro-dégénératives. Le rôle majeur de FoxO1 et de ses homologues dans la protection contre le stress oxydant et les dommages de l’ADN, et ainsi dans la détermination de la longévité est conservé phylogénétiquement. Nos travaux réalisés dans des cultures primaires d’hépatocytes de rat impliquent FoxO1 dans la régulation par rétrocontrôle des axes métaboliques et mitogéniques de la voie de l’insuline. Notamment, FoxO1 régule l’activité du suppresseur de tumeur p38, cette dernière augmentant en retour l’activité de la kinase de survie PKB. Le gène POLG1 code pour la polymérase de l’ADNmt, et des mutations de ce gène sont responsables de pathologies de maintenance de l’ADNmt, transmises selon un mode Mendélien. Le rôle potentiel de variants de ce gène dans l’accélération des processus morbides liés à l’âge justifient un dépistage rapide et large des mutations de ce gène. Nous avons mis en place ce criblage pour le gène POLG1 et deux autres gènes de maintenance de l’ADNmt, le gène Twinkle et le gène ANT1 en utilisant la technique de DHPLC. Le rôle crucial de FoxO1 dans la biogenèse mitochondriale illustre le trait d’union entre mes travaux, en souligne l’importance, et constitue une trame pour les études à venir
Eukaryotic cells heavily depend on mitochondria for their energy production. Oxidative metabolism is the main source of reactive oxygen species (ROS), which are mutagenic for mitochondrial and nuclear DNA. Therefore, mitochondrial functions lie at the heart of both the ageing process and the pathophysiology of late-onset diseases, including type 2 diabetes and neurodegeneration. Downstream of the insulin signaling pathway, the FoxO1 transcription factor and its homologs, encode for enzymes buffering ROS or repairing damaged DNA, and are involved in stalling the ageing process across evolution. Our experiments conducted in primary cultures of rat hepatocytes led us to conclude that FoxO1 controls and integrates key signaling pathways. Indeed, FoxO1 controls its own activity by sequentially activating the tumor suppressor p38, the mTORC2 complex and finally the survival kinase PKB. Mutations in the POLG1 gene, which encodes for the mitochondrial DNA polymerase, are found in Mendelian human diseases characterized by a defect in mtDNA stability and repair. Mutations or functional polymorphisms in such genes may promote the ageing process and a fast, reliable and large screening of these gene variants could be useful in humans. We have taken advantage of the DHPLC screening method to seek for mutations in POLG1, Twinkle and ANT1 genes, all of which are involved in mtDNA stability in humans. The recent involvement of FoxO factors in mitochondrial biogenesis provides a link between our interests, highlights their importance and marks the road for future studies
