Анотація:
Dalam Pembangunan system industry 4.0 salah satu yang menopang pembangunan adalah teknologi 3D Printing. Tujuan dari Penelitian ini adalah merancang mesin Printer 3D dengan Microkontroller Arduino MEGA 2560, RAMPS 1.4 Shield, Motor stepper NEMA 17, DVR8825 motor driver, Filament PLA, E3D v6 HotEnd, Memory Card, Smart LCD. Konstruksi rangka rancang bangun alat Printing 3D dengan melakukan perakitan rangka mesin printing 3D, Motor Stepper Mesin 3D, Limit Endstop XYZ, dudukan exstruder, dudukan bed, perakitan HotEnd untuk heater pemanas. Proses pencetakan dapat terhubung dengan PC atau menggunakan dukungan memory card, dan instruksi pengoperasian ditampilkan dalam monitor LCD. Penelitian ini menghasilkan printer 3D berbasis Arduino dengan teknologi FFF (Fused Filament Fabrication) yang akan memudahkan pengguna dalam mengoperasikan printer 3D dengan layar LCD dan Pencetakan tidak harus selalu terbuhung dengan PC karena menggunakan memory card yang dapat menyimpan file dan menghasilkan pencetakan yang lebih baik. Penelitian ini menggunakan metode penelitian model Linier Sequential Model (LSM). Model ini sering disebut dengan “Classic Life Cycle” atau model waterfall. Metode ini terdiri 5 tahapan yang berulang yaitu tahap analisis studi literatur, tahap desain/perancangan sistem, tahap perakitan hardware, tahap pengkodean, dan tahap pengujian (Pressman, R.S, 2012).

Анотація:
School failure is an object of debate in different educational and political spheres. Despite the discussions, it is a problem that is far from a resolution and deserves the attention of researchers. Research in the educational area has pointed out that the explanations for the phenomenon are centered on the student and/or his family. Thus, the aim of this theoretical essay is to problematize the conception that school failure is the sole and exclusive responsibility of the student, and the naturalizing and positivist views about the human being, in an attempt to overcome it. The theoretical and methodological framework to which our analysis sought support is the historical-cultural theory and, therefore, dialectical historical materialism. We specifically analyze the social and psychological dimensions of school failure beyond the accountability of the student and his family, considering the totality of social being and the objective conditions of concrete reality. Taking a critical look at the phenomenon of school failure implies revealing our difficulty as a society, embodied in our education systems, school institutions and education professionals in dealing with the challenges arising from the democratization of education, that is, with the access of the popular classes to the public school. Thus, school failure endsup being treated as an individual problem, exempting the others involved in the schooling process from responsibility.

AbstractAcetyl-CoA Carboxylase (ACCase) is considered to beone of the key enzymes in palm oil biosynthesis. Availabilityof genes encoding this enzyme would give some advantagesin the molecular breeding of oil palm. Over expression ofthe genes in the oil palm mesocarp might increase the oilproduction in this tissue. On the other hand, downregulating of ACCase could divert the central metaboliteAcetyl-CoA to other product such as PHB (Polyhydroxy-butyrate), one of the known biodegradable plastic. Thispaper reported the work of cloning of the full length codingsequence of biotin carboxylase (BC), one subunit of theACCase. Based on the DNA sequence of the BC conservedregion that had cloned previously, primers pairs weredesigned to amplify 5’- and 3’- cDNA ends of BC usingRACE-PCR. The RACE products of 5’- and 3’- cDNA endsof BC were cloned into E.coli, and the DNAs weresequenced and analysed. The full cDNA of BC was obtainedby reisolation of the cloned 5’- and 3’- cDNA ends followedby digestion using KpnI, ligation into pGEM-T vector andcloning into E.coli. Colony PCR was carried out to confirmthat the target gene has been cloned. The recombinantplasmid containing full cDNA of BC was then isolated forDNA sequencing.
