The purpose of this study was to examine the effect of Mindful Listening Instruction on Music Listening Sensitivity and Music Listening Enjoyment. The type of mindfulness investigated in this study was of the social-psychological type, which shares both commonalities with and distinctions from meditative mindfulness. Enhanced context awareness, openness to new information, situation in the present, awareness of novel distinctions, and awareness of multiple possible perspectives (cognitive flexibility) are components of social-psychological mindfulness. A pretest-posttest control group design was used for this study. Two different age groups of students were studied: fourth-grade students (N = 42) and undergraduate non-music major college students (N = 48). The fourth-grade participants in this study were selected from an elementary school in a large city in the Northeastern United States. The college students were selected from a large university in the Southeastern United States. Participants were randomized into either the experimental or control group. Gordon’s Intermediate Measures of Music Audiation and Advanced Measures of Music Audiation were used as a pretest for fourth-grade students and college students, respectively. The results showed no statistically significant differences between the experimental and control groups. Student demographical information was also collected and reported. The treatment consisted of 10 lessons for fourth-grade students. Five of the 10 lessons were used with the college students. For each age level, participants in both groups, Mindful Listening and Control, received instruction using listening-map-based and non-listening-map-based lessons from the Share the Music textbook series. Students in the Mindful Listening groups also received listening instructions designed to promote mindful listening. Music Listening Sensitivity was measured using the phrasing test from the Sensitivity portion of Gordon’s Music Aptitude Profile (MAP-P), as well as the researcher-created Anderson Test of Music Listening Sensitivity (ATMLS). Music Listening Enjoyment was measured using students’ ratings of their Listening Enjoyment after each lesson on a seven-point Likert-type scale. Results indicated that Mindful Listening Instruction yielded higher scores, which were statistically significant (at α = .05), for Music Listening Sensitivity (as measured by both the ATMLS and the MAP-P) and Music Listening Enjoyment for fourth-grade and college-student participants.

With modern culture moving towards convenience in terms of fresh produce, especially with meat products, science needs to constantly evolve to serve these customer needs. These needs however can sometimes be implemented too hastily without the proper assessment done for factors such as food safety. One such improvement is the use of modified atmosphere packaged minced meat. This form allows the minced meat to be kept for much longer than normal without freezing the product, providing the fast pasted consumer both the convenience of having fresh, unfrozen meat as well as an added shelf life. MAP works by disrupting the atmosphere within these packages, retarding the growth of the spoilage micro-organisms, thus causing them to require a longer time span to reach spoilage numbers. The problem however arises that although most of these techniques are tried and trusted on the products’ spoilage organisms, it does not take into consideration the effect MAP and the altered spoilage organism communities would have on a pathogen that might be present on the products. This study thus aimed to assess the effect of both factors on Escherichia coli O157:H7 (phase 1) and Staphylococcus aureus (phase 2) in minced meat kept at 5°C as well as trying to identify the major affecting factors. The two subject organisms were each inoculated into 2 different types of MAP packs and a non-atmosphere modified PVC overlaid minced meat sample at two differing concentrations of 105 and 102 to also assess the impact of high andlow initial pathogen presence. These packs were then analysed over a time period of 16 days to track the changing minced meat environment. APC, Pseudomonads, LAB and Enterobacteriaceae counts were all investigated along with the pathogenic counts. Apart from colony enumeration, the colour of the minced meat samples were also taken to determine the effect that these parameters have on the appearance of the product, as colour is often the first sensorial characteristic that determines the purchase of fresh meat products. pH was determined to ascertain the environmental changes occurring in the product and whether groups such as the LAB would change the environment to better suit their needs. Finally the atmospheric makeup was also measured to determine the effect of the MAP system and the change occurring in a closed system that could be attributed to the growth and respiration of the bacterial communities present. Apart from the main aim of the study, two additional studies were performed that arose during the planning and analyses of the two primary phases. Firstly the use of a quarter versus a full plate enumeration was studied to determine its accuracy as well as possibility of use in full studies to aid enumeration and decrease time and financial input. Here a direct comparison was done between the two techniques after which they were compared and assessed in their functionality for both homogenous and heterogeneous community enumeration on selective and non-selective media. The other secondary study focussed on the use of new technology for both the enumeration and tracking of genetically modified organisms in a variety of different environments. Here a bioluminescent imaging system was used on a genetically modified strain of E. coli to track its spread through minced meat, packaged either in a MAP or PVC overlaid pack, over 48 hours in an accelerated shelf life study. Enumeration of said organism was also undertaken whereby the intensity of emitted light would correspond to a defined count, enabling rapid enumeration of samples, whether overgrown or not. Copyright

This master's thesis focuses on the dynamic presentation of still photography using a depth map. This text presents an algorithm that describes the process of creating a spatial model which is used to render input photography so that the movement of virtual camera creates parallax effect due to depth in image. The thesis also presents an approach how to infill the missing data in the model. It is suggested that a guided texture synthesis is used for this problem by using rendering outputs of the model themselves as guides. Additional information in model allows the virtual camera to move more freely. The final result of the camera movement can be saved to simple video sequence which can be used for presenting the input photography.

iv In recent years, outbreaks of infections associated with raw and minimally processed fruits and vegetables have been reported. Possible sources for contamination are irrigation water, manure, wash water, handling by workers and contact with contaminated surfaces. Pathogens can occur on raw and minimally processed produce at populations ranging from 103 to 109 CFU/g and able to survive and sometimes grow under various storage conditions. The objective of this study was to analyse the growth/survival of Salmonella Enteritidis at spot-inoculated or stem-injected cherry tomatoes during passive modified atmosphere packaging (MAP), controlled atmosphere (CA) and air storage at 7 and 22°
C. Low density polyethylene (LDPE) with a package size of 10x10 cm2 for 25±
2 g tomatoes was used for MAP storage in which the gas composition equilibrated to 6% O2/ 4% CO2 and a carbon dioxide incubator was used for CA storage in which the CO2 level was monitored and maintained as 5% through the term of storage at 7 and 22°
C. During the research, the effect of ozone treatment (5-30 mg/L ozone gas for 0-20 min) was also considered for surface sanitation. The results demonstrate that S.Enteritidis can survive and/or grow during the storage of tomatoes depending on the location site of the pathogen on fruit, suspension cell density and storage temperature. During MAP, CA and air storage, S.Enteritidis with initial population of 7.0 log10 CFU/tomato survived on tomato surfaces with an approximate decrease of 4.0-5.0 log10 CFU/tomato in population within the storage period
however, in the case of initial population of 3.0 log10 CFU/tomato, cells died completely on day 4 during MAP storage and on day 6 during CA and air storage. The death rate of S.Enteritidis on the surfaces of tomatoes that were stored in MAP was faster than that of stored in air. Storage temperature was effective on the survival of S.Enteritidis for the samples stored at ambient atmosphere
cells died completely on day 6 at 7°
C and on day 8 at 22°
C. Stem scars provided protective environments for Salmonella
an approximate increase of 1.0 log10 CFU/tomato in stem-scar population was observed during MAP, CA and air storage at 22°
C within the period of 20 days. Cells survived with no significant change in number at 7°
C. The development of the microbial association in tomatoes was dominated by lactic acid bacteria (LAB). The pH values of the tomatoes changed approximately from 4.0 to 3.0 during the storage period. LAB grew well under all atmospheric conditions with or without the presence of S.Enteritidis. Gaseous ozone treatment has bactericidal effect on S.Enteritidis, inoculated on the surface of the tomatoes. 5 mg/L ozone gas treatment was not effective. 30 mg/L ozone gas treatment affected surface color.

When provided with spatial niches by growth in static nutrient medium, Pseudomonas fluorescens diversifies through adaptive radiation into several well-defined phenotype classes. One of these classes, named wrinkly spreader (WS) for its morphology on agar medium, forms a biofilm at the air-liquid interface through mutations in one of several loci including the genes wspF and awsX. These genes code for negative regulators of di-guanylate cyclases (DGCs). These DGCs catalyse synthesis of cyclic-di-GMP, a second messenger, overproduction of which effects physiological changes leading to overproduction of a cellulose polymer and the WS phenotype. Intriguingly, a diverse range of wspF mutations leads to diversity both in colony morphology and strain fitness.In this study, I investigate genetic and fitness diversity in wrinkly spreaders with the aim of identifying the causal factors that link genetic diversity and physiological factors with diversity in fitness. I approach the subject from several directions, examining the historical context of genetic diversity in wspF and awsX, distribution of control over output in the Wsp pathway and overall fitness effects of different causal factors. I investigate the genetic basis of wrinkly spreader evolution through generation of a large number of novel WS strains and exploration of the distribution of mutations in the wspF and awsX genes. In combination with this I calculate estimates of the past rates of mutation in these genes, derived from a phylogenetic investigation of a group of orthologues. I examine the response of the Wsp pathway to change in WspF function through a novel computational analysis that is capable of revealing valuable information on control in a biological system based purely on model structure. In addition I show how this analysis can be refined through specification of broad estimates of system parameters, thereby avoiding issues related to over-reliance on specific parameter values. Finally, I investigate the fitness implications of these factors, as well as a variety of others, through assays of fitness in a group of WS strains combined with machine learning analyses of predictive relationships between protein and mutation characteristics and experimentally measured strain fitness, and consider the implications of this analysis in the context of intermediate physiological effects.I find that mutations in the WspF protein that lead to the WS phenotype tend to be located in regions of historically strong conservation, the first time that any such pattern to WS mutations has been identified. Mutations in AwsX, on the other hand, do not fit such a pattern. Computational analysis of the Wsp pathway shows that, regardless of model parameters, pathway output is always more sensitive to changes in methylesterase activity by WspF than to changes in phosphorylation of WspF, which may explain the greater frequency of mutations fixed in vivo seen in the methylesterase domain. Despite these patterns, none of a wide range of mutation and sequence-based biochemical characteristics, including local rates of past evolution and size and position of mutations, exhibited any predictive power over WS fitness. Overall, the findings in this study point towards an essential role for complex pleiotropic effects in strongly modulating the fitness effect of different mutations in wspF.

High-precision optical systems are generally tested using interferometry, since it often is the only way to achieve the desired measurement precision and accuracy. Interferometers can generally measure a surface to an accuracy of one hundredth of a wave. In order to achieve an accuracy to the next order of magnitude, one thousandth of a wave, each error source in the measurement must be characterized and calibrated.Errors in interferometric measurements are classified into random errors and systematic errors. An approach to estimate random errors in the measurement is provided, based on the variation in the data. Systematic errors, such as retrace error, imaging distortion, and error due to diffraction effects, are also studied in this dissertation. Methods to estimate the first order geometric error and errors due to diffraction effects are presented.Interferometer phase modulation transfer function (MTF) is another intrinsic error. The phase MTF of an infrared interferometer is measured with a phase Siemens star, and a Wiener filter is designed to recover the middle spatial frequency information.Map registration is required when there are two maps tested in different systems and one of these two maps needs to be subtracted from the other. Incorrect mapping causes wavefront errors. A smoothing filter method is presented which can reduce the sensitivity to registration error and improve the overall measurement accuracy.Interferometric optical testing with computer-generated holograms (CGH) is widely used for measuring aspheric surfaces. The accuracy of the drawn pattern on a hologram decides the accuracy of the measurement. Uncertainties in the CGH manufacturing process introduce errors in holograms and then the generated wavefront. An optimal design of the CGH is provided which can reduce the sensitivity to fabrication errors and give good diffraction efficiency for both chrome-on-glass and phase etched CGHs.

Анотація:

Porphyrins consist of twenty-atom rings containing four nitrogen atoms and can be used as sensor to detect odours and gases.

This thesis investigates whether or not porphyrins can be used as functional materials on grid gate devices.  Drops of PVC embedded porphyrins were deposited on the surface of a grid gate which is a Metal Oxide Semiconductor (MOS) capacitor. In order to detect the gas sensing properties of the porphyrins a light addressable method called Scanning Light Pulse Technique (SLPT) has been used.

Drops of porphyrins were deposited with a stretched capillary tube (1 mm diameter).

The MOS capacitor has been exposed to nitrogen atmosphere as reference environment, while the target gases were carbon monoxide (100 ppm) and ammonia (500 ppm).

The result from the eight porphyrins is that one of them [Pt(II) TPP] has a response for both gases, ammonia induces a change in both the work function and surface resistance, while the carbon monoxide induces only a change in the surface resistance.

Salmonella enteritidis has recently emerged as a potential pathogen in poultry products. The growth of S. enteritidis in poultry is affected by several factors such as storage temperature, pH, water activity, modified atmosphere and the presence of preservatives. All of these factors may act alone or in combination with each other resulting in a synergistic, antimicrobial effect.
In this research, initial storage studies were done to determine the effect of various atmospheres (air, vacuum, oxygen absorbent and gas packaging) on the microbial changes of packaged chicken thighs followed by challenge studies with a strain of S. enteritidis$ sp{ rm{NAST}}$. Chicken thighs were packaged in Cryovac bags and stored at 4 and 12$ sp circ$C for up to 28d. Changes in headspace gas composition, pH, drip loss, color and odor were monitored at each sampling day.
The effect of various packaging treatments, dipping solutions (chitosan (0.2%w/v) and potassium sorbate (0.2%w/v)) and low dose irradiation (1.5 & 3.0 kGy) on the growth of S. enteritidis$ sp{ rm NAST}$ and on the shelf-life of chicken thighs stored at 4 and 12$ sp circ$C was also investigated. The study assessed the knowledge structure of Grade 11 physics students and their instructors using Pathfinder networks. Instructors’ structural knowledge was averaged to create a referent pathfinder network. Each student’s pathfinder network was compared with the referent pathfinder network in order to identify misconceptions. These misconceptions served as the basis for remedial instruction. The study was conducted in six sections of Grade 11. Three different types of remedial instruction based on three different chapters from the Grade 11 physics textbook were given to the students at three separate stages. In the first section, students were shown their own and referent pathfinder networks as an intervention during the first stage. The students were asked to reflect on the similarities and differences between them. The researcher gave written concept-oriented exercises based on the differences at the second stage, and multimedia concept-oriented instruction based on the differences was given to the students at the third stage. The order of instruction was counterbalanced in all the six sections. After each stage, students’ pathfinder networks were reassessed and the similarities between students’ and the referent pathfinder networks were calculated to measure the effect of a particular intervention. The study tried to determine which type of remedial instruction given to students best improved the knowledge structure of the students in the domain of physics. Results revealed that the similarity indices around the treatment concepts in the pathfinder networks of the students increased the most from pre- to post-intervention phase because of their reflections, followed by multimedia concept-oriented instruction and written concept-oriented exercises. Most likely, the major reason for this change was the interventions around the treatment concepts by the researcher at three different stages which stimulated and probably changed some of students’ misconceptions. To address the issue of validity, the similarity indices of control concepts in the students’ pathfinder networks were also checked for improvement. The result shows that there is no appreciable improvement in control concepts as there was no intervention around those concepts. Findings support the use of structural assessment of knowledge with pathfinder scaling technique to check the effectiveness of a classroom instruction.

Fucan is a term used to denominate L-fucose rich sulfated polysaccharides. The fucans have been studied due their pharmacological activities like antithrombotic, antiproliferative and antioxidant. We have extracted three fucan fractions from the brown seaweed Spatoglossum schr?ederi. These fucans were denominated Fuc B 1, Fuc B 1.5 and Fuc B 2. The chemical analyzes show that the fucans have very similar composition as demonstrated by agarose electrophoresis gel, sugar and sulfate content. The antiproliferative effect was determined by MTT and BrdU methodologies in CHO cells. The inhibition of proliferation effect of the three fractions was about 40%. Therefore this we proceed just with the Fuc B 2 due the higher yield. There is no apoptosis indication using the anexin V/propidium iodide test. We found a cell cycle phase G1 arrest. The western blotting show that the PKC; pFAK; pERK 1/2 are activated when the cells were treated with fucans. The treatement with inhibitor of MAPK PD98059 extinguished the fucan effect. These results indicates that fucan act by the ERK pathway inducing the cell death.

As fucanas t?m sido estudadas devido suas atividades farmacol?gicas: antitromb?tica, antiproliferativa e antioxidante. N?s extra?mos tr?s fra??es de fucanas da alga Spatoglossum schr?ederi. Essas fucanas, denominadas de Fuc B 1, Fuc B 1.5 e Fuc B 2, apresentam uma composi??o muito similar como demonstrado pela eletroforese em gel de agarose, e conte?do de a??car e sulfato. O efeito antiproliferativo foi determinado pelas metodologias de MTT e BrdU em c?lulas CHO. O efeito na inibi??o da prolifera??o das tr?s fra??es foi de cerca de 40%. Assim, procedemos somente com a Fuc B 2 devido seu maior rendimento. N?o houve indica??o de apoptose usando a marca??o com anexina V-FITC/ Iodeto de Prop?deo. Identificamos uma parada na fase G1 do ciclo celular. Os ensaios de western blotting mostraram que PKC, pFAK e pERK 1/2 s?o ativadas quando as c?lulas s?o tratadas com Fuc B. O tratamento com o inibidor de MAPK PD98059 aboliu o efeito da fucana. Esses efeitos indicam que a fucana age via ERK para inibir a prolifera??o das c?lulas.

Listeria monocytogenes is a pathogenic, psychrotrophic microorganism that is ubiquitous in nature. L. monocytogenes has been isolated from numerous meat products, both fresh and processed, the incidence of contamination varying greatly. The ability of Listeria to grow in meats depends on temperature, pH, water activity (a$ sb{ rm w}$), nutrients, species and numbers of competing microorganisms, gaseous conditions, and levels of additional barriers. Therefore, methods to control the growth of L.monocytogenes are of great importance to food processors since this organism can grow under a wide range of environmental and storage conditions. Two methods of control, in conjunction with temperature, were studied in this project: (i) modified atmosphere packaging (MAP) and (ii) chitosan, to determine the optimum levels of these "hurdles" needed to effectively control the outgrowth of L.monocytogenes in both model broth and agar systems and in fresh pork loin. On the basis of these preliminary studies, a combination of chitosan as a dipping solution and modified atmosphere packaging were investigated to control the growth of L.monocytogenes in fresh pork loin. Pork loin samples were dipped in a 0.2% chitosan solution for 60 seconds and packaged under various atmospheres in Cryovac bags and stored at 5, 10 and 15$ sp circ$C up to 28 days. Samples were monitored for physical, chemical and microbiological changes throughout the storage period. Optimum control over the growth of L. monocytogenes was achieved using a combination of 100%N$ sb2$ + an Ageless FX oxygen absorbent and dipping in a 0.2% chitosan solution. Based on these studies, a combination of 0.2% chitosan and MAP could be used to extend the shelf life of pork without adversely affecting color, odor and exudate loss while inhibiting the growth of the pathogenic microorganism, L.monocytogenes. The purpose of this work was both to explore the variety of methods to develop mobile applications and to create a requirement documentation and solution proposal to the case study Rundtursbuss. During the 10 weeks work-experience, I had the possibility to investigate a range of methods to develop mobile applications and apply it to a reference case. The questions regarding which method to choose when developing mobile application are not easy to answer, once there is not only one solution that can fit the needs of all the applications that will be developed at Scania. Each application needs to be evaluated not only from a technical perspective, but also from a user-centered way. One of the best ways to quickly evaluate a project is asking what that makes the project mobile The analysis of each criteria aided in the evaluation of the application requirements and in reaching the conclusion that the most promising methods to develop the application Rundtursbuss are either native or cross-compilation ones. In conclusion, it is crucial to emphasize that while Scania does not have a procedure on how to develop, maintain and coordinate the development of mobile applications, single initiatives will continue to emerge, which increase the cost and affect the quality of their IT products and services.

To identify a model legume experimental system for studying low temperature signaling and cold acclimation, cold-induced expression and regulation of homologues of alfalfa (Medicago sativa) cold acclimation-specific genes cas15 and cas30 were examined in M. arborea (relatively frost tolerant) and M. truncatula (relatively frost sensitive). Both cas15 and cas30 genes are present in the genomes of both species but whereas both genes are cold-induced in M. arborea, only cas15 is induced in M. truncatula. Cold-induced expression of these genes is inhibited by calcium chelators and channel blockers and by the membrane fluidizer benzyl alcohol. Treatment of leaves with dimethylsulfoxide, a membrane rigidifier, induced both genes at 25°C. A cold-activated MAP kinase activity was expressed in both species. These results suggest that M. truncatula, an annual, self-pollinated species may be successfully used as model experimental systems in studies of cold signaling and role of cas genes in cold acclimation in legumes.

Only a third out of all IT-projects are successful. This depends on the factor that the managers and the users which the IT-project is aimed towards aren’t properly involved in the design-process. Instead have the technical aspects of the project been in focus and not the users. Impact mappers solve these problems by collaborating with both the managers and the users. The method called Impact mapping has been in focus for this research project and how this method is used by impact mappers to help companies identify and achieve their goals. The impact mapper collaborates with the company to define the purpose of the IT-project. The collaboration between the impact mapper and the users enables the impact mapper to understand how the users interact with the IT-product. This process results in the creation of a so-called impact-map. The impact mapper uses the impact-map to create prototypes of the company’s new website. After that the prototypes will be sent to the web developers which will create the website. The managers are responsible for all decisions that are made during the project. The impact mapper recommends the users which should be prioritized based on how they can help the company achieve their goals. The impact mapper is therefore limited in his or her work. The impact mapper has a key role and needs to understand the users and how the business works. Through the usage of interviews we have investigated which opportunities and limitations that may arise when they collaborate with the managers and users. In this research project we have also identified which limitations and opportunities impact mapping has in the development of a new website.

En effektkartläggare rekommenderar enbart vilka målgrupper som kan tänkas prioriteras utifrån hurde kan bidra till att verksamhetens mål nås. Detta begränsar effektkartläggaren i sitt arbete. Sedan läggs även ett stort ansvar på effektkartläggaren gällande att förstå verksamheten och målgruppen. Genom användandet av intervjuer har det även undersökts vilka möjligheter och begränsningar effektkartläggare ser när de samarbetar med chefer och målgrupper. I detta forskningsprojekt har vi också tagit reda på vilka möjligheter och begränsningar effektkartläggning har rörande utvecklingen av en ny webbsida.

This thesis examines the utility of mathematical complexity measures for the analysis of stratospheric dynamics. Through theoretical considerations and tests with artificial data sets, e.g., the iteration of the logistic map, suitable parameters are determined for the application of the statistical entropy measures sample entropy (SE) and Rényi entropy (RE) to methane (a long-lived stratospheric tracer) data from simulations of the SOCOL chemistry-climate model. The SE is shown to be useful for quantifying the variability of recurring patterns in a time series and is able to identify tropical patterns similar to those reported by previous studies of the ``tropical pipe'' region. However, the SE is found to be unsuitable for use in polar regions, due to the non-stationarity of the methane data at extra-tropical latitudes. It is concluded that the SE cannot be used to analyse climate complexity on a global scale. The focus is turned to the RE, which is a complexity measure of probability distribution functions (PDFs). Using the second order RE and a normalisation factor, zonal PDFs of ten consecutive days of methane data are created with a Bayesian optimal binning technique. From these, the RE is calculated for every day (moving 10-day window). The results indicate that the RE is a promising tool for identifying stratospheric mixing barriers. In Southern Hemisphere winter and early spring, RE produces patterns similar to those found in other studies of stratospheric mixing. High values of RE are found to be indicative of the strong fluctuations in tracer distributions associated with relatively unmixed air in general, and with gradients in the vicinity of mixing barriers, in particular. Lower values suggest more thoroughly mixed air masses. The analysis is extended to eleven years of model data. Realistic inter-annual variability of some of the RE structures is observed, particularly in the Southern Hemisphere. By calculating a climatological mean of the RE for this period, additional mixing patterns are identified in the Northern Hemisphere. The validity of the RE analysis and its interpretation is underlined by showing that qualitatively similar patterns can be seen when using observational satellite data of a different tracer. Compared to previous techniques, the RE has the advantage that it requires significantly less computational effort, as it can be used to derive dynamical information from model or measurement tracer data without relying on any additional input such as wind fields. The results presented in this thesis strongly suggest that the RE is a useful new metric for analysing stratospheric mixing and its variability from climate model data. Furthermore, it is shown that the RE measure is very robust with respect to data gaps, which makes it ideal for application to observations. Hence, using the RE for comparing observations of tracer distributions with those from model simulations potentially presents a novel approach for analysing mixing in the stratosphere.

In questa tesi si utilizza il formalismo dei fibrati principali per descrivere le propriet`a topologiche globali di sistemi fisici classici e quantistici che presentano simmetrie legate all’azione del gruppo U(1). Nel primo capitolo `e contenuta una esposizione della teoria matematica dei fibrati, con un particolare riguardo ai fibrati principali ed alle strutture differenziali definibili su di essi (forme differenziali di connessione e curvatura). Nel secondo capitolo si impiega il formalismo precedentemente sviluppato per trattare le propriet`a del monopolo magnetico di Dirac e si ottiene una quantizzazione della carica magnetica sulla base di considerazioni di natura topologica. Inoltre, si mostra l’impiego dei fibrati principali U(1) nella costruzione di una descrizione Lagrangiana globale per sistemi quali una particella carica nel campo del monopolo e una particella classica con spin in campo magnetico. Nel terzo capitolo, si descrive teoricamente la comparsa di una fase geometrica (fase di Berry) in sistemi quantistici che evolvono adiabaticamente nel tempo, e si fornisce un’interpretazione geometrica di tale fase come olonomia in un fibrato principale U(1). Il moto di una particella quantistica con spin in campo magnetico quasi-statico e l’effetto Aharonov-Bohm vengono presentati come esempi tipici di manifestazione di una fase geometrica.

The regulation of the serum triglyceride responses to fat ingestion have been examined in normolipidemic men. To evaluate the existing methods for comparing chylomicron-triglyceride clearance, the oral and intravenous fat tolerance tests and a steady state duodenal perfusion method were compared. Good correlations (r > 0.8) were found between each of these methods. Since the intravenous fat tolerance test is independent of fat absorption, these data suggested that the serum triglyceride response to fat feeding was largely determined by the rate of chylomicron-triglyceride clearance. To determine the influence of the quantity and type of meal fat on postprandial serum triglyceride concentrations, the serum triglyceride responses to three different doses of dairy cream, and to standard doses of olive and sunflower oil were examined. For a given type of fat, the magnitude of postprandial lipemia (the integrated serum triglyceride excursion) varied directly with the quantity of fat in the meal. This finding suggested that the chylomicron- triglyceride clearance system(s) did not become saturated even after large fat meals. In addition, it appeared that the hormonal factors released in response to fat ingestion (some of which are known to increase lipoprotein lipase activity in vitro) did not increase the rate of chylomicron-triglyceride clearance. If the quantity of fat in a meal was fixed, then postprandial lipemia increased with increasing saturation of the triglyceride fatty acids. These differences did not appear to reflect differences in triglyceride absorption. Since acute fat feeding per se did not appear to stimulate chylomicron-triglyceride clearance, the effects of dietary proteins and carbohydrates were studied. The addition of up to 35g protein to a standard test meal did not affect postprandial lipemia. These results were supported by the observation that protein ingestion did not affect intravenous fat tolerance. Postprandial serum triglyceride concentrations were significantly influenced by carbohydrate ingestion. Fructose (50g) and sucrose (100g) markedly increased postprandial lipemia, although glucose ingestion did not. In agreement with earlier studies, glucose ingestion decreased serum triglyceride concentrations 2 hours after the meal. This effect was abolished by intraduodenal fat administration and by substituting starch for glucose in the test meal. The effects of glucose could be reproduced by iso-osmotic quantities of urea, however. These findings suggested that glucose ingestion did not increase chylomicron -triglyceride clearance. It is more likely that glucose delayed the absorption of triglycerides by slowing gastric emptying, and that this effect was partly related to the increased osmolarity of glucose- containing meals. The effects of chronic exercise on postprandial lipemia and chylomicron-triglyceride clearance were determined in endurance- adapted athletes. The serum triglyceride responses to large and small fat meals were lower in athletes than in sedentary men with comparable fasting triglyceride concentrations. These differences were not eliminated by a single bout of acute exercise in the sedentary men. The clearance of intravenously administered lntralipid, and chylomicron -triglyceride clearance assessed from steady state chylomicron-triglyceride concentrations during duodenal fat perfusion were faster in athletes than in the sedentary men. These data suggested that the low postprandial lipemia in athletes reflects increased chylomicron-triglyceride clearance caused by increased activity of the triglyceride clearing system(s). Given these considerations. it appears that the pathway(s) for chylomicron triglyceride clearance are extremely efficient in normal men and that these pathways are not subject to acute physiological regulation.

Adenosine is a nucleoside with varied pharmacological effects but its actions in man are sparsely documented. Cardiac and respiratory effects of adenosine in man were examined. Adenosine was found to increase coronary flow in patients without significant coronary atheroma. Adenosine restored sinus rhythm in some patients with supraventricular tachycardia. In those in whom sinus rhythm was not restored the underlying rhythm was atrial flutter. In patients with complete heart block adenosine decreased ventricular rate in a dose-related fashion. In subjects in sinus rhythm adenosine produced a transient dose-related bradycardia which was followed by a more sustained increase in sinus rate. These latter effects were compared but were found not to differ in young and elderly subjects. A dose-related respiratory stimulant effect of adenosine, which had not previously been widely appreciated, was observed. The possible relevance of this observation to the ventilatory response to hypoxia is discussed. Adenosine-induced respiratory stimulation was found not to differ in young and elderly subjects. Dipyridamole, an inhibitor of adenosine transport, potentiated adenosine-induced respiratory stimulation and bradycardia but not the subsequent tachycardia. Aminophylline, a competitive adenosine antagonist at cell-surface receptors, abolished adenosine-induced bradycardia but did not alter the tachycardia or respiratory stimulation. Adenosine-induced bradycardia in man may be explained by an action of adenosine on cell-surface receptors at one site, whereas such an explanation does not accord with the observations on tachycardia and respiratory stimulation. Adenosine administered proximal to the carotid circulation in man stimulated respiration, but infusion distal to those vessels had no such effect. These observations are consistent with the hypothesis that adenosine stimulates respiration in man by an action on the carotid body. Possible physiological, pathophysiological and therapeutic implications of these observations are discussed.

Geographic information systems (GIS) are tools used to facilitate locational decision making in interactive, graphic-based environments. GIS and interactive maps allow users to customize displays and manipulate data for accomplishing a variety of tasks, ranging from map interpretation to wayfinding and land use planning. Although originally adopted for professional use, GIS software is increasingly utilized by both expert and non-expert users. Despite the improved availability, training in cartographic design has not followed suit and studies claim that users often employ inefficient displays for task completion. Research on naïve realism indicates that people exhibit a bias for realistic depictions containing irrelevant, extraneous details, leading to increased cognitive load and decreased task performance. This dissertation explores how display realism affects decision making task performance when using a GIS. Prior studies examining naïve realism have primarily had users perform map reading and inference tasks with static displays. Natural resource management was selected as a test case because it often involves the use of geospatial tools and data and people with varying levels of GIS expertise. This research had expert and novice users utilize a GIS to perform site selection tasks for a natural resource management decision making scenario. The results indicate that increased display realism has a negative impact on task performance, especially with regards to task completion time. Individuals in both the expert and novice groups were influenced by naïve realism. It was observed that expert and novice users employed different strategies for task completion and the implications on task performance are discussed. Ultimately, the study results contribute to the theory of naïve realism and make recommendations that inform the use of task-appropriate graphic displays in an interactive mapping environment.

Connexins are transmembrane proteins that form hemichannels allowing the exchange of molecules between the extracellular space and the cell interior. Two hemichannels from adjacent cells dock and form a continuous gap junction pore, thereby permitting direct intercellular communication. Connexin 36 (Cx36), expressed primarily in neurons, is involved in the synchronous activity of neurons and may play a role in aberrant synchronous firing, as seen in seizures. To understand the reciprocal interactions between Cx36 and seizure-like neural activity, we examined three questions: a) does Cx36 deficiency affect seizure susceptibility, b) does seizure-like activity affect Cx36 expression patterns, and c) does acute blockade of Cx36 conductance increase seizure susceptibility. We utilize the zebrafish pentylenetetrazol (PTZ; a GABA(A) receptor antagonist) induced seizure model, taking advantage of the compact size and optical translucency of the larval zebrafish brain to assess how PTZ affects brain-wide neuronal activity and Cx36 protein expression. We exposed wild-type and genetic Cx36-deficient (cx35.5-/-) zebrafish larvae to PTZ and subsequently mapped neuronal activity across the whole brain, using phosphorylated extracellular-signal-regulated kinase (pERK) as a proxy for neuronal activity. We found that cx35.5-/- fish exhibited region-specific susceptibility and resistance to PTZ-induced hyperactivity compared to wild-type controls, suggesting that genetic Cx36 deficiency may affect seizure susceptibility in a region-specific manner. Regions that showed increased PTZ sensitivity include the dorsal telencephalon, which is implicated in human epilepsy, and the lateral hypothalamus, which has been underexplored. We also found that PTZ-induced neuronal hyperactivity resulted in a rapid reduction of Cx36 protein levels within. 30 minutes and one-hour exposure to 20 mM PTZ significantly reduced the expression of Cx36. This Cx36 reduction persists after one-hour of recovery but recovered after 3-6 hours. This acute downregulation of Cx36 by PTZ is likely maladaptive, as acute pharmacological blockade of Cx36 by mefloquine results in increased susceptibility to PTZ-induced neuronal hyperactivity. Together, these results demonstrate a reciprocal relationship between Cx36 and seizure-associated neuronal hyperactivity: Cx36 deficiency contributes region-specific susceptibility to neuronal hyperactivity, while neuronal hyperactivity-induced downregulation of Cx36 may increase the risk of future epileptic events.

Within the brain, cells (neurons) communicate with each other to pass along information. This communication is important for normal functions of the brain such as learning and memory, muscle movement, etc. Epilepsy is a disease of the brain that is caused by rapid over synchronized communication between cells. This leads to seizures which can include convulsions, loss of attention, and much more. Currently, 30% of patients suffering from epilepsy do not have a treatment option that works for them, it is, therefore, imperative to investigate new targets for treatment in this disease. Connexin36 is a protein in the brain that directly connects cells so they can pass information quickly between them. Connexin36, therefore, might make a good target for treatment. Previous work has aimed to understand this relationship but has been limited in their ability to look at the entire brain at any one time. The goal of this study was to understand the relationship between connexin 36 and brain hyperactivity across the whole brain simultaneously. To understand this relationship, we first determined what happened to brain activity if the protein was missing entirely after exposure to a seizure causing drug. We were asking: How does connexin 36 affect hyperactivity. We found that different regions of the brain responded differently without the connexin 36 protein. This suggests that one size does not fit all, and one must look at the whole brain to understand the effects of the connexin 36 protein. Next, we asked a similar question, but in the opposite direction, how does hyperactivity affect connexin 36? We found, in the short-term, hyperactivity reduced the amount of connexin 36 present in certain regions of the brain. This continued until 3 hours following exposure to the seizure causing drug Pentylenetetrazol (PTZ). Lastly, to determine if this short-term reduction in connexin 36 meant that an individual was more likely to experience hyperactivity. To do this, we used a connexin 36 blocking drug, then introduced the seizure causing drug at different concentrations. We found, at all concentrations, the connexin 36 blocking drug caused significant changes in neuronal activity, depending on the brain regions. Overall, our results showed that connexin 36 plays an important role in hyperactivity and that a short-term reduction in connexin 36 is detrimental, and may contribute to an increase in the possibility of subsequent hyperactivity.

Les matériaux composites à matrice céramique CMC, sont généralement formés d'au moins deux matériaux ayant une forte capacité d'adhésion. Ces matériaux sont principalement composés de renforts fibreux assurant la tenue mécanique de la structure et d'une matrice qui permet sa cohésion. Utilisées principalement dans le domaine de l'aéronautique, elles sont reconnues pour leur bonne tenue mécanique, leur réfractarité élevée tout en conservant une densité faible. Par contre, l'inconvénient majeur associé à ces matériaux est l'apparition de fissures qui sont dues soit au procédé de fabrication soit aux sollicitations mécaniques externes.Dans ce travail, une attention toute particulière est consacrée aux composites à matrice auto-cicatrisante dont la principale propriété est l'aptitude à "réparer" les effets de la fissure par formation d'un verre visqueux.Ces verres visqueux se forment au sein de la fissure grâce à l'oxydation des éléments qui constituent la matrice. Selon la température, différents verres peuvent être formés.Leur fonction est de reboucher les fissures de taille micrométrique de façon à ralentir la diffusion de l'oxygène en direction des fibres et éviter leur rupture par oxydation.Cependant, pour des systèmes en rotation rapide tels que les turbines basse pression des moteurs d'avion (pièce étudiée actuellement), on peut s'interroger quant à la mobilisation du verre visqueux cicatrisant dans un système complexe géométriquement et inhomogène du point de vue de la nature des surfaces. Pour approcher le comportement du verre cicatrisant dans un système modèle mais néanmoins réaliste, une approche de modélisation numérique a été entreprise. L'outil numérique utilisé pour cette étude est le code de calcul Thétis développé à l'I2M. Celui-ci est adapté à ce type de simulation puisqu'il permet la modélisation d'écoulements diphasiques incluant des phénomènes physiques complexes tels que le mouillage. Ainsi, l'objectif de ce travail est-il de déterminer les limites d'utilisation de ce type de matériau en fonction des conditions auxquels il est soumis en évaluant la mobilité du verre cicatrisant dans la fissure.

The purpose of this study was to examine two scaffolding methods on the performance of students in computer-based concept linking and retention of comprehension. After training and practice in concept mapping and CmapTools--a computer-based concept mapping program, 116 undergraduate students were randomly assigned to one of four treatment groups to work on a computer-based concept mapping task. Students in the no scaffolding (NS) group did not receive any scaffolding. Students in the linking phrase scaffolding (PS) group received linking words or phrases as scaffolding. Students in the articulation hint scaffolding (AS) group received a hint question as scaffolding, which asked them to elaborate on relationships between concepts in full sentences. Students in the linking phrase and articulation hint scaffolding (PAS) group received both scaffolding while working on the computer-based concept mapping task. One week after the treatment, students took a concept linking posttest, in which they constructed a concept map in CmapTools based on a web-based instruction on the human heart. After another week, they took another posttest on retention of comprehension about the heart. Two 2 X 2 factorial Analysis of Variance (ANOVA) were conducted to examine the main effects of linking phrase scaffolding and articulation hint scaffolding and any interaction effect between them on the performance of students in computer-based concept linking and retention of comprehension. The results showed no significant difference in the performance of students in both tests. However, the Pearson's correlation analysis showed that there was a positive correlation between students' performance in computer-based concept linking and retention of comprehension (γ = 0.447, p < 0.01).

The rationale for this study was to extend the knowledge about creative thinking among children with attention deficit hyperactivity disorder (ADHD) by investigating the effects of creativity training upon concept mapping complexity. To verify whether the CoRT Thinking Lessons can enhance creative ability and improve concept map complexity, a Pretest-Posttest Control Group Design was used in this study. Sixty four students who are fourth and fifth graders (age 9-10 years) and classified as having ADHD participated in the investigation. In order to ensure that concept mapping is a familiar technique to all participants, they all were given a training session in concept mapping. Next to that, they were asked to complete a concept map and Torrance tests of creative thinking (TTCT) as a pre-test measure. After that, they were assigned to either experimental or control group, each group consisted of thirty two students. The experimental group was given creativity training (20 hours of CoRT thinking lessons during ten weeks) and the control group received no creativity training. Finally, all sixty four participants completed a second concept map and the TTCT as a post-test measure. Data collected from participants were analyzed via the Pearson product-moment correlation coefficient, the t test for two independent samples, and the paired-sample t test. Results of the analysis indicated that participants who received creativity training developed significantly more complex concept maps than those participants who received no such creativity training. Thus, creativity training enhances both concept mapping complexity and creative ability of students with ADHD as measured by the TTCT. Implications for practice include recommendation for teachers to design and establish educational activities and environments in which creative abilities of children with ADHD can be develop.

Access to the to the Internet and to a variety of interactive mapping tools has increased interest among middle school teachers to use interactive maps in conjunction with learning activities. There is very little research in the area of interactive mapping in educational situations, specifically with regards to layer maps that combine multiple thematic layers on a single map. This study evaluates the relationship between the number of layers present on a web-based map and middle-school students' accuracy and timeliness using the map to answer geographic questions. Additionally, this study examines the specific effect of a hill shade on student response time and accuracy when answering questions do not require any terrain information. Tests were conducted in five Portland, Oregon area middle school classrooms using Blackboard CE8 to present maps and collect responses. The results of this research indicate no significant relationship between the number of layers present on a web map and middle school students' accuracy or response times while using the map to answer questions. The presence of a hillshade layers does not significantly impact the students' response times or accuracy while answering questions while using the map either.

This study investigated the use of varying formats for presenting complex visual information in the form of fictitious maps. There were three treatment formats, two which sequenced the map information by conceptual category (i.e., roads, geographic features, boundaries), and a third which used a normal intact map format. The two sequenced presentations differed in that the first was additive; that is, each visual in the sequence built upon the preceding one by adding a category of information until the complete map was displayed. The second simply displayed one conceptual category per visual. The cognitive style field dependence-independence was also examined as potentially related to map-learning ability based on the greater cognitive restructuring ability of the field independent orientation. lt was predicted that the achievement of field-dependent students would be higher with the sequenced map treatments, and the achievement of the field-independent students would be higher with the intact map treatment. The research questions were: 1. Does dividing map information into either an Additive sequence or a Part-by-Part sequence affect performance on a map-learning task? 2. Do students classified as held dependent, moderately field - independent, or held independent differ in their performance on a map-learning task? 3. Is there an interaction across visual design format with cognitive style? The sample for this study was composed of 92 four-year college students. Level of held dependence was measured by the GEFT. The analysis of the 3 X 3 research design was by a two-way analysis of variance with posttest scores (which measured immediate recall of map information) as the dependent variable. Map presentation format and cognitive style were independent variables. Results indicated there was no difference in achievement among the three map presentation formats, but that cognitive style was significantly related to posttest scores; that is, a higher level of held independence was associated with more effective map-learning performance.

Les plantes dans leur milieu naturel, sont constamment soumises à de multiples contraintes environnementales de nature biotique ou abiotique qui sont à l'origine de nombreuses pertes de rendement. Afin de répondre à ces stress, les plantes mettent en place des réponses adaptées qui reposent sur l'activation de voies de signalisations. L'une des voies importantes est celle impliquant la phosphorylation des protéines par les Mitogen Activated Protein Kinase (MAPKs). La régulation de l'activité des MAPKs est indispensable et dépend en partie de protéines phosphatases telles que les MAPK Phosphatases (MKPs). Ce travail a consisté à étudier les mécanismes de régulation de l'activité phosphatase de TMKP1, la seule MKP connue de blé dur, par les calmodulines et les protéines 14-3-3. Dans un premier temps, nous avons montré que l'activité phosphatase de TMKP1 est stimulée en présence des ions K+, Li+, Mg 2+ et surtout par le Mn2+. Des expériences de GST pull dawn ont permis de mettre en évidence une interaction, calcium dépendante, entre TMKP1 et la calmoduline (CaM) et nous avons démontré que l'activité de TMKP1 est inhibée par le complexe CaM/Ca2+. En revanche, ce même complexe stimule l'activité de TMKP1 en présence des ions Mn 2+. Ce mode de régulation d'une MKP par CaM/Ca2+ dépendant des ions Mo2+ est décrit ici pour la première fois. Dans un second temps, la présence au niveau de la séquence de TMKP1 d'un domaine de liaison aux protéines 14-3-3s, nous a incité à mener des essais d'immunoprécipitation à l'issue desquelles nous avons montré que TMKP1 pourrait interagir avec les 14-3-3s chez le Blé et que celles-ci stimulent l'activité phosphatase de TMKP1 in vitro. Enfin, l'implication de TMKP1 dans la réponse des plantes aux stress biotiques et abiotiques a été évaluée. Les tests de gennination ont révélé que la sur-expression de TMKP1 chez Arabidopsis permet une meilleure tolérance à la salinité. Ces mêmes plantes semblent également être plus résistantes à une infection bactérienne causée par Pseudomonas syringae que des plantes sauvages. Ces données suggèrent que TMKP1 agirait comme régulateur positif de la réponse d'Arabidopsis au stress salin et à l'infection P. syringae. L'ensemble des résultats obtenus ici a permis de dévoiler de nouvelles propriétés jamais décrites chez une MKP végétale et offre une nouvelle vision sur le rôle de ces phosphatases dans le contrôle des voies de signalisations MAPKs impliquées dans la réponse des plantes aux stress de l'environnement
Due to their sessile lifestyle, plants are constantly subjected to a variety of biotic and abiotic stresses causing tremendous yield losses. To survive under these conditions, plants have evolved different sigualing pathways allowing efficient stress responses. The Mitogen Activated Protein Kinase (MAPKs) are key sigual transduction molecules, which respond to varions external stimuli. However, the activity of MAPKs has to be strictly regulated by protein phosphatase such as MAPK Phosphatase (MKPs). ln the present study we describe the regulation ofTMKPI, a wheat MKP, by Calmodulins (CaM) and 14-3-3 proteins. We fust showed that phosphatase activity oTMKP1 is stimulated by K\ Li\ Mg2+ and especially by Mo2+. Using GST pull dawn assays, we demonstrated that TMKP1 binds to CaM in a Ca2+-dependent manner. Moreover, the CaM/Ca2+ complex inhibits the catalytic activity of TMKP1 in a CaM-dose dependent marmer. However, in the presence of Mo2+, this activity is enhanced by CaM/Ca2+ complex. Such effects were not reported so far, and raise a possible role for CaM and Mo 2+ in the regulation of plant MKPs during cellular response to extemal siguals. Moreover, a fine sequence analysis ofTMKPl revealed the presence of a conserved 14-3-3 mode 1 binding site. This finding incited us to perfonn co-immunoprecipitation assays on wheat protein extracts through which we provide proof-of-concept evidence for interaction of TMKP1 with 14-3-3 proteins. Interestingly, the phosphatase activity of TMKP1 was shawn to be enhanced by several plant and yeast 14-3-3 isoforms. Finally, the involvement of TMKP1 in plant stress responses was evaluated. Our data showed that the overexpression of TMKP1 in Arabidopsis resulted in a higher tolerance to salt stresses and to bacterial infection caused by P. syringae. Such findings suggest thal TMKP1 may act as a positive regulator of Arabidopsis responses to salt stresses and P. syringae infection. All together, our results provide novel functional properties for plant MKPs, and should add new knowledge to our understanding of the raie of these phosphatases in the control of MAPK signaling pathways controlling plant responses to various environmental stresses

In this work, melt pool size in process mapped in power-velocity space for multiple processes and alloys. In the electron beam wire feed and laser powder feed processes, melt pool dimensions are then related to microstructure in the Ti-6Al-4V alloy. In the electron beam wire feed process, work by previous authors that related prior beta grain size to melt pool area is extended and a control scheme is suggested. In the laser powder feed process, in situ thermal imaging is used to monitor melt pool length. Real time melt pool length measurements are used in feedback control to manipulate the resulting microstructure. In laser and electron beam direct metal additive manufacturing, characteristics of the individual melt pool and the resulting final parts are a product of a variety of process parameters. Laser or electron beam spot size is an important input parameter that can affect the size and shape of a melt pool, and has a direct influence on the formation of lack-of-fusion and keyholing porosity. In this work, models are developed to gain a better understanding of the effects of spot size across different alloys and processes. Models are validated through experiments that also span multiple processes and alloys. Methods to expand the usable processing space are demonstrated in the ProX 200 laser powder bed fusion process. In depth knowledge of process parameters can reduce the occurrence of porosity and flaws throughout processing space and allow for the increased use of non-standard parameter sets. Knowledge of the effects of spot size and other process parameters can enable an operator to expand the usable processing space while avoiding the formation of some types of flaws. Based on simulation and experimental results, regions where potential problems may occur are identified and process parameter based solutions are suggested. Methods to expand the usable processing space are demonstrated in the ProX 200 laser powder bed fusion process. In depth knowledge of process parameters can reduce the occurrence of porosity and flaws throughout processing space and allow for the increased use of non-standard parameter sets.

Hypsometric color is one of the most important cartographic design elements, and it affects regional height and terrains perception to map readers. Hypsometric color combined with hill shading was commonly used to present landform. There are different opinions in choosing hypsometric colors in geographic field and publishing field. What kind of hypsometric colors is more attractive to map readers? And more essentially, which one is more effective in transmitting landform information? Researchers used 15 maps (5 hypsometric colors, including 2 cartographic theory colors, 2 commonly used colors in publishing field, 1 commonly used color in atlas, and 3 different regions) to test readers with different backgrounds. By using visual experiment, this paper attempts to find out the favorite and acceptable color type. Moreover, using statistics analysis to find out the most effective color type for height information transmitting, and to analyze the behavior variation of map reader for different hypsometric color maps. The quantitative results suggested that the color used in common atlas was the favorable color to the subjects. Nevertheless, the color combined with hill shading which was originally assumed better feedback turned out to be in worse preference. In the height information transmitting parts, however, the colors concerned cartography exhibited the better performances, both in the accuracy and value of height. The research results showed the variation between acceptable degree of subjects and efficiency of height information transmitting in different hypsometric colors.

Test theory is an explanation of empirical relationships among examination data. The modern test theory is based on the Item Response Theory (IRT), which considers the parameters of test item and the response of test-receiver (including difficulty, discrimination, ability and so on),and the estimation of its test-receiver’s ability becomes more precise. Concept Effect Relation Map (CERM) constructed by data mining with naïve data preprocessing causes: (1) monotonous concept fuzzification result, (2) the association rules may not reflect the real concept relation and (3) the circulating association rules exit. In this thesis, we apply the IRT as the assessment of students’ concept learning response. With the consideration of the difficulty and the discrimination of test item, we propose an IRT-Based Data Preprocessing Concept Effect Relation Map Construction System. IRT-Based Data Preprocessing Concept Effect Relation Map Construction System includes two modules: the Data Preprocessing Module and the Data Mining Module. The former has four procedures: Test Item Analysis, Learning Response Index (LRI) Generator, Concept Decomposition/Aggregation and Fuzzy ACLR Generator, and the latter has two procedures: Association rule mining and concept map constructor. The experiment results of the proposed Approach show that the CERM construction can be improved and the number of circulated association rules generated can be reduced.

Sketch map is an important way to represent spatial information used in many geospatial reasoning tasks (Forbus, K., Usher, J., & Chapman, V. 2004). Compared with verbal or textual language, sketch map is a more interactive mode that more directly supports human spatial thinking and thus is a more natural way to reflect how people perceive properties of spatial objects and their spatial relations. One challenging application of sketch maps is called Spatial-Query-by-Sketch proposed by Egenhofer. Being a design of query language for geographic information systems (GISs), it allows a user to formulate a spatial query by drawing the desired spatial configuration with a pen on a touch-sensitive computer screen and get it translated into a symbolic representation to be processed against a geographic database (Egenhofer, M. 1997). During the period of sketch map drawing, errors due to human spatial cognition in mind may occur.
