The persistence of HIV-1 viral reservoirs in the brain, despite treatment with combination antiretroviral therapy (cART), remains a critical roadblock for the development of a novel cure strategy for HIV-1. To enhance our understanding of viral reservoirs, two complementary studies were conducted to (1) evaluate the HIV-1 mRNA distribution pattern and major cell type expressing HIV-1 mRNA in the HIV-1 transgenic (Tg) rat, and (2) validate our findings by developing and critically testing a novel biological system to model active HIV-1 infection in the rat. First, a restricted, region-specific HIV-1 mRNA distribution pattern was observed in the HIV-1 Tg rat. Microglia were the predominant cell type expressing HIV-1 mRNA in the HIV-1 Tg rat. Second, we developed and critically tested a novel biological system to model key aspects of HIV-1 by infusing F344/N control rats with chimeric HIV (EcoHIV). In vitro, primary cultured microglia were treated with EcoHIV revealing prominent expression within 24 h of infection. In vivo, EcoHIV expression was observed seven days after stereotaxic injections. Following EcoHIV infection, microglia were the major cell type expressing HIV-1 mRNA, results that are consistent with observations in the HIV-1 Tg rat. Within eight weeks of infection, EcoHIV rats exhibited neurocognitive impairments and synaptic dysfunction, which may result from activation of the NogoA-NgR3/PirB-RhoA signaling pathway and/or neuroinflammation. Collectively, these studies enhance our understanding of HIV-1 viral reservoirs in the brain and offer a novel biological system to model HIV-associated neurocognitive disorders and associated comorbidities (i.e., drug abuse) in rats.

OBJECTIVES/GOALS: Ecotin is a potent periplasmic serin protease inhibitor that was first described in Escherichia coli (E. coli). Ecotin is known to inhibit a broad range of serine proteases including NE. Therefore, we hypothesized that ecotin can inhibit the NE activity present in CF clinical sputum samples. METHODS/STUDY POPULATION: To investigate the function of ecotin on CF sputum, orthologues from Campylobacter rectus and Campylobacter showae were recombinantly expressed and purified from E. coli to generate C. rectus ecotin, C. showae ecotin, and E. coli ecotin. Ecotin was added to CF sputum supernatant samples at various concentrations and NE enzymatic activity was measured by a fluorometric assay kit. In addition, CF sputum and healthy sputum was added to PMNs isolated from healthy donors for 3.5 hours and NE was measured with immunofluorescence staining. The sputum was washed two times prior to measuring NE released and on the PMNs, and PBMCs was used as a control. RESULTS/ANTICIPATED RESULTS: Our results indicate a clear inhibition of NE activity in CF sputum supernatant. C. showae ecotin showed the greatest inhibitory effect on NE activity in CF sputum supernatant. We also saw that CF sputum supernatant causing healthy PMNs to release more NE suggesting that NE in the CF airway may trigger more NE release from newly incoming PMNs to the lung. Our next steps will be to determine if ecotin can inhibit NE on the PMN surface and released from PMNs. DISCUSSION/SIGNIFICANCE: Neutrophil elastases are produced by PMNs to kill microbial pathogens in the lung, however in CF, the pathogens are unable to be killed by NE and instead causes severe lung damage. PMNs and NE has been shown to be elevated in the sputum of CF patients and there is currently no NE therapeutic inhibitor that has been effective in these patients.

Background We performed a systematic review of studies conducted in Latin America and the Caribbean (LAC) to assess the impact of oral diseases on oral health-related quality of life (OHRQoL). Materials and methods Searches were performed of the following PubMed, EMBASE, CINAHL, Scopus, and LILACS databases. Randomized clinical trials, quasi-experimental studies, cohort studies, case and control studies, and cross-sectional studies which included at least 100 participants evaluating the impact of oral diseases on OHRQoL were included. PROSPERO registry number: CRD42020156098. Results After exclusion of duplicates, 3310 articles were identified, 40 of which were included in this review. 90% of the studies were conducted in Brazil. The most commonly used OHRQoL measuring instruments were CPQ 11–14 (n = 9), ECOHIS (n-8) and B-ECOHIS (n = 8). The study designs included 32 cross-sectional, 2 cohort and 6 case and control studies. Most of the studies were conducted in children (n = 25) and adolescents (n = 9). Most studies identified an impact on OHRQoL in children, adolescents and adults with oral diseases. Moreover, greater oral disease severity had a greater impact on OHRQoL. Conclusions Most studies in LAC report a negative impact of diseases on OHRQoL. More longitudinal studies are required to confirm the results of these studies.

The paper provides data on the use of the Malus sieboldii Rehd. as a genetic donor of economically valuable traits in breeding low-vigorous clonal apple rootstocks. The method of embryo culture is proposed as a way to increase the efficiency of distant crosses. The studies were carried out using seeds obtained in three combinations of crossing (M. sieboldii x Malysh Budagovskogo, M. sieboldii x 60-160, M. sieboldii x 76-3-6). As sterilizing agents when introduced into culture in vitro, we used industrial disinfectant «Belizna (Whiteness)», the sanitizer «Ecocid C» + «Belizna» and the sanitizer «BioPag C» + «Belizna». It was found that additional pretreatment with «Ecocid C» (1.0%) and "BioPag C" (0.4%) for 50 minutes before using the disinfectant «Belizna» contributes to the production of sterile explants up to 61.5% compared to the control. The best results were obtained when combine using «Ecocid C» (1.0%, 50 min.) and «Belizna» (50%, 5 min.). After 28 days, due to the low number of developing embryos, the method of dissection of a part of the cotyledons of hybrid seeds was used. As a result, up to 65.3% of hybrid plants were obtained, compared to 4.2% of the control.

Abstract Ixodids suck blood from host animals and transmit pathogens that cause important medical and veterinary diseases. As synthetic conventional acaricide resistance becomes increasingly common, alternative tactics are likely to enhance control efforts. Laboratory bioassays assessed the lethality of CimeXa, a commercial silica gel desiccant dust, and EcoVia, a commercial product containing silica gel + thyme oil, against lone star tick, Amblyomma americanum (L.), larvae and nymphs. Both life stages were completely killed by CimeXa by 24 h, and EcoVia achieved total larval mortality within 1 h when they were initially immersed in the dusts and when they crawled across treated substrate. Larvae were killed faster than nymphs. Temporary immersion of A. americanum larvae and nymphs in aqueous suspensions of the dusts were less effective than exposure to the dusts applied dry. Larval and nymphal mortality associated with crawling on dried aqueous suspensions of the products was also not as strong as when the immature life stages crawled across substrate treated with dry dusts, and EcoVia was not more efficient than CimeXa. CimeXa and EcoVia can likely be used to protect cattle in a prophylactic context because silica gel does not degrade, and EcoVia might be effective at eliminating ticks that are feeding on cattle as well. We suggest that the two dust products be assessed for efficacy against other ixodid species, such as the southern cattle fever tick, Rhipicephalus microplus (Canestrini), which transmits the causal agents of babesiosis to cattle.

Genetic polymorphisms could explain the inter-individual differences in the oral health-related quality of life (OHRQoL) of children with anterior open bite (AOB). Objective: To assess the impact of AOB on OHRQoL in children and to evaluate whether MTR (rs1805087), MTRR (rs1801394), TGFβ1 (rs1800469) and TNF-α (rs1799964, rs1799724 and rs1800629) genes are potential biomarkers for OHRQoL in children with AOB. Study design: A cross-sectional study was performed with 173 children aged between 2–6 years. The Brazilian version of Early Childhood Oral Health Impact Scale (ECOHIS) was applied. Genetic polymorphisms were analyzed using real-time PCR. Mann-Whitney U-test and Chi-square were used. Results: The overall mean ECOHIS scores were 5.49 (SD= 5.72) and 3.45 (SD = 4.49) (p < 0.01) in the AOB and control groups, respectively. Children with the CC genotype of TNF-α (rs1799724) had a significantly higher psychological QoL level. The MTRR AA genotype group showed a lower QoL level in the child subscale (p = 0.006), function (p = 0.017), and psychological (p = 0.006) domains. There was no significant difference between OHRQoL and the genetic polymorphisms in MTR and TGFβ1. Conclusions: Genetic polymorphisms in TNF-α and MTRR are associated with the impact on the OHRQoL in children with AOB.

Objetivo: o diamino fluoreto de prata (DFP) é um produto seguro, eficaz e indolor para controlar a cárie dentária. Como requer consultas de curta duração, seu uso em pacientes epiléticos pode reduzir o risco de convulsões. Relato de caso: uma paciente de dois anos de idade com cárie severa na primeira infância envolvendo seis incisivos decíduos foi encaminhada à Clínica da Universidade. Seu histórico médico revelou a presença de paralisia cerebral e epilepsia com o uso diário de medicamentos para controlar a doença. Devido à menor idade da paciente, ao risco de convulsões durante um tratamento odontológico invasivo e demorado, e para evitar uma possível anestesia geral, o DFP 30% foi escolhido para paralisar as lesões de cárie. Um questionário de qualidade de vida (B-ECOHIS) foi aplicado antes e após o tratamento. Conclusão: o tratamento com SDF foi eficaz na paralisação da cárie dentária em uma criança epiléptica jovem, sem alteração em sua qualidade de vida.

The aims of the study were to characterize enamel, dentine and cementum surface aspect after using non-fluoride remineralization toothpastes containing calcium carbonate and calcium glycerophosphate by SEM evaluation, to determine chemical composition of enamel, dentin and cement after remineralization products application by EDX assessment, and to compare the remineralization capacity of the tested products. Ten extracted teeth having no dental caries, erosive or wear lesions were selected for the study. The teeth were split bucco-lingually and mesio-distally in four pieces. A free window of enamel, dentine, and cementum (4X4 mm) in the cervical area of the slices was preserved. In control group the slices were subjected to a artificial caries lesion formation. In study groups EcoBio� (President Company), Bio-natural (Dentissimo, Swiss Biodent), and Arthrodont Cassic (Pierre Fabre Oral Care) toothpastes were applied two times a day, for 3 minutes during caries lesion formation. The surface topography has been analyzed using a scanning electron microscope and the quantitative and qualitative chemical composition has been evaluated using an EDX detector. Interprismatic and intraprismatic mineral loss is clearly visible at higher magnification in enamel samples in control group. Some areas of demineralization are still present, and also small areas of extern deposits are visible on enamel, dentine and cementum samples in the groups were tested products were applied. All the tested toothpastes increased calcium and phosphorous ion concentrations in enamel, dentine and cementum. The highest remineralization effect was recorded in enamel by Dentissiomo, followed by Arthrodent and EcoBio toothpastes. In dentine and cementum the highest remineralization effect was registered by Arthrodent, followed by Dentissiomo and EcoBio toothpastes.

Introducción: La Calidad de Vida relacionada con Salud Oral (CVRSO) se refiere al impacto que las condiciones orales tienen en las actividades diarias de los individuos. Para recopilar esta información en niños, existen instrumentos específicos contestados por ellos mismos o por sus padres/apoderados, como ECOHIS. Dado que en Chile existe una alta prevalencia de caries en niños, es necesario evaluar cómo esta enfermedad afecta su calidad de vida, por lo tanto, el objetivo de la presente investigación es establecer la existencia de una asociación entre CVRSO y prevalencia y severidad de caries en niños de 5 años que asisten a colegios municipales del área norte de la Región Metropolitana (RM), aplicando ECOHIS. Materiales y Métodos: Este estudio observacional de corte transversal, evaluó 52 niños de 5 años, pertenecientes a 5 colegios del área norte de Santiago. El examen fue realizado por dos examinadores calibrados en criterios ICDASII y según recomendaciones de la OMS. Los apoderados firman un consentimiento informado y responden el cuestionario ECOHIS, tipo entrevista. Se calculó prevalencia e historia de caries, de acuerdo a criterios de detección OMS e ICDASII. La CVRSO fue determinada según puntaje ECOHIS. Se realizan asociaciones entre las variables y se establecen diferencias significativas entre los grupos. Resultados: La prevalencia e historia de caries fue de 55,8% y ceod=1,9 según criterio OMS y 86,5% y ceod=5,3 según ICDASII. Un 9,5% de apoderados reporta un impacto en la CVRSO, con una puntuación media de 4 puntos. Para ambos criterios de detección, la media de ECOHIS fue mayor para el grupo con caries, pero con diferencias significativas usando el criterio OMS. Asimismo, hubo una mayor puntuación ECOHIS en los grupos con mayor índice ceod, para ambos criterios de detección. Conclusiones: Existe una alta prevalencia de caries en niños de 5 años del área norte de la RM. La prevalencia y una mayor historia de caries impactan la calidad de vida de niños preescolares y sus familias, por lo que es necesario prevenir la enfermedad y tratarla en estadios tempranos.

Understanding the interaction of multiple drivers and their compounded effects on ecosystem dynamics is a key challenge for marine resource management. The Baltic Sea is one of the world’s seas most strongly impacted by effects from both human activities and climate. In the late 1980’s changes in climate in combination with intensive fishing initiated a reorganization of the Central Baltic Sea (CBS) food web resulting in the current sprat-dominated state. In the future, climate change is projected to cause drastic changes in hydrodynamic conditions of the world oceans in general, and the Baltic Sea in particular.   In this thesis, CBS food web responses to the combined effects of fishing, nutrient loads and climate were tested for the past (1974-2006) and projected into the future (2010-2098). A new food web model for the CBS (BaltProWeb) was developed using extensive monitoring data across trophic levels. This model described the past food web dynamics well, and was hence also used for future (2010-2098) projections. Different ensemble modeling approaches were employed when testing the food web response to future scenarios. The results show that regardless the climate change, the management of nutrient loads and cod fishing are likely to determine the food web dynamics and trophic control mechanisms in the future Baltic Sea. Consequently, the variation in the food web projections was large, ranging from a strongly eutrophied and sprat-dominated to a cod-dominated CBS with eutrophication levels close to today’s values. The results also suggest a potential risk of abrupt ecosystem changes in the future CBS, particularly if the nutrient loads are not reduced. Finally, the studies illustrate the usefulness of the ensemble modeling approach, both from the perspective of ecosystem-based management as well as for studying the importance of different mechanisms in the ecosystem response.

El presente trabajo de investigación se desarrollará en la empresa Ecovive SAC donde se plantea un sistema de gestión de inventario basada en la teoría de inventarios y control de la producción, utilizando tecnología QR para mejorar la gestión del inventario de productos o materiales, dado que en la actualidad presenta perdidas de materiales, no cuenta con la información necesaria para los reportes, no se registran en su totalidad las entradas y salidas de los materiales, demora en la generación de reportes para gerencia. Por ello ¿De qué manera se podrá mejorar la gestión de inventarios de productos de la empresa Ecovive SAC?, la presente investigación se justifica porque permitirá controlar mejor los materiales, y brindara información oportuna para la tomar de decisiones, disminución de la carga de trabajo, aumento de la productividad, disminución de los tiempos muertos, para lo cual se utilizara la metodología SCRUM porque es un marco de trabajo para la gestión y desarrollo de software basada en un proceso iterativo e incremental utilizado comúnmente en entornos basados en el desarrollo ágil de software. Para lograr el reconocimiento de los problemas de la empresa Ecovive SAC, se utilizó la técnica de la entrevista con el gerente, supervisor y personal operativo. También se utilizó fichas de observaciones.

Para recopilar esta información en niños, existen instrumentos específicos que pueden ser contestados por ellos mismos o por sus apoderados. ECOHIS y SOHO-5 son instrumentos para evaluar CVRSO. La diferencia que existe entre ambos es que SOHO-5 no solo aborda la visión de padres o tutores, sino que también el relato directo del niño. El objetivo de esta investigación es establecer si existe una correlación entre los resultados de SOHO-5 y ECOHIS en padres y sus niños de 5 años que asisten a colegios municipales del área norte de la región metropolitana. Materiales y Métodos: Este es un estudio observacional de corte transversal, que evaluó a 52 niños utilizando la encuesta ECOHIS y SOHO-5, todos pertenecientes a 5 colegios del área norte de Santiago. Las encuestas fueron realizadas por distintos grupos de alumnos de Odontología de la Universidad de Chile. Los apoderados firmaron un consentimiento informado y respondieron el cuestionario ECOHIS y SOHO-5, tipo entrevista. La CVRSO fue determinada según puntaje ECOHIS y SOHO-5. La correlación se cuantificó utilizando el coeficiente de Spearman. Resultados: De acuerdo a los resultados provistos por ECOHIS, el 90,5% de los padres no perciben un impacto en la CVRSO. El ítem de mayor prevalencia es “dolor” seguido de “la dificultad para comer”. En cuanto a los resultados de SOHO-5, un 93% de los encuestados muestra un bajo impacto de los problemas orales en la CVRSO. El ítem de mayor prevalencia reportado es “la dificultad para comer” seguido de “la dificultad para hablar”. Cuando se comparó ECOHIS con SOHO-5 total y SOHO-5 versión para padres se encontró una correlación moderada, directa y estadísticamente significativa. Mientras que, cuando se comparó SOHO-5 sección niños con ECOHIS y SOHO-5 versión para padres se observó una correlación débil, directa y no significativa para la primera. Conclusiones: SOHO-5 y ECOHIS se correlacionan directamente el uno con el otro. La distribución de respuestas de padres o tutores y el puntaje de calidad de vida obtenido comparativamente es similar para los dos instrumentos aplicados. Background Vaccination is the most cost effective and long-term solution to the global human immunodeficiency virus (HIV) pandemic. The HIV Gag and Tat proteins are attractive components of a HIV vaccine as immune responses targeting these proteins confer protective benefits against HIV infections in humans. This thesis has developed two innovative candidate HIV vaccines viz. a DNA vaccine encoding oligomerised and secreted Tat (pVAXsTat-IMX313), and a recombinant live human rhinovirus serotype A1 (HRV-A1)-based vaccine encoding Gag and Tat (rHRV-Gag/Tat). Methods To construct pVAX-sTat-IMX313, Tat was fused with the oligomerisation domain of IMX313 to form Tat heptamers and linked to the leader sequence of tissue plasminogen activator to ensure that the bulk of oligomerised protein is secreted. To develop the rHRVGag/Tat vaccine, initially, the full length tat gene and 5 discrete overlapping fragments corresponding to the full length gag gene were individually inserted into the junction between the HRV-A1 genes encoding structural and non-structural proteins (P1/P2 junction) to ensure that the exogenous HIV Gag or Tat proteins were separated from the recombinant polyprotein using the HRV encoded 2Aprotease enzyme. Thus, one recombinant HRV encoding Tat (rHRV-Tat) and 5 rHRVs each encoding a unique Gag fragment (rHRV-Gag1-5) were generated. The individual rHRVs were then mixed into a single cocktail vaccine (rHRVGag/Tat), purified and titrated for inoculation in mice. The immunogenicity of these vaccines was evaluated in female BALB/c mice that received up to five intradermal injections of pVAX-sTat-IMX313 (50 μg per dose) at 2 weekly intervals in one study. In another study, mice were vaccinated intranasally with 2 doses (5x106 TCID50/dose) of the rHRV-Gag/Tat followed by a single 50 μg booster dose of a cocktail DNA vaccine containing pVAX-sTat-IMX313 and pVAX-Gag-Perforin. Vaccine-induced immune responses were examined 2 weeks after the last dose by antibody ELISA, in-vitro Tat transactivation neutralization, IFN-γ ELISpot, KdGag197-205 tetramer staining and intracellular cytokine staining assays. Results Data showed that fusing Tat with IMX313 results in complete heptamerisation of Tat. Furthermore, the data suggested that pVAX-sTat-IMX313 vaccination elicited higher titers of serum neutralizing Tat-specific IgG, secretory IgA (sIgA) in the vagina and CMI responses, and showed superior control of ecotropic HIV (EcoHIV) infection, a surrogate murine HIV challenge model, compared with animals vaccinated with other DNA vaccines tested in this study. Human rhinovirus serotype A1 (HRV-A1) was successfully engineered into a replication-competent genetically stable recombinant vector to deliver a mucosally-targeted vaccine, rHRV-Gag/Tat, by inserting exogenous HIV gag and tat sequences into the HRV-A1 genome. Finally, intranasal administration of 2 doses of rHRV-Gag/Tat followed by a single DNA booster dose induced superior poly-functional Gag-specific CD8 T cell responses in the spleen (systemic) and mesenteric lymph nodes (mucosal), higher Tat-specific serum IgG and sIgA in the vagina, and effective control of EcoHIV infection compared to other vaccination regimens tested in this study. Conclusion First, the data support the inclusion of IMX313 as a molecular adjuvant for Tat-based HIV DNA vaccines. Second, the data demonstrated that intranasal vaccination with rHRV-Gag/Tat followed by a single DNA booster dose is effective in eliciting HIV-specific immunity panmucosally and systemically. Collectively, the data support further testing of the pVAX-sTat IMX313 and rHRV-Gag/Tat vaccines in macaques, preferably in a heterologous prime-boost vaccination strategy, and results from these studies might influence future HIV clinical trials.

<em>Abstract.</em>—A dynamic mass-balance ecosystem model (Ecopath with Ecosim) was used to investigate how relative changes in fishing mortality on sharks can affect the structure and function of Apalachicola Bay, Florida, a coastal marine ecosystem. Simulations were run for 25 years, wherein fishing mortality rates from recreational and trawl fisheries were doubled for 10 years and then decreased to initial levels. Effect of time/area closures on ecosystem components were also tested by eliminating recreational fishing mortality on juvenile blacktip sharks <em>Carcharhinus limbatus</em>. Simulations were run assuming mixed control and top-down control. In the mixed control, biomass of juvenile coastal sharks (finetooth shark <em>C. isodon</em>, spinner shark <em>C. brevipinna</em>, sandbar shark <em>C. plumbeus</em>), juvenile blacktip sharks, and bull sharks <em>C. leucas </em>declined up to 57% when recreational fishing mortality was doubled. Increases in biomass were also observed for the Atlantic sharpnose shark <em>Rhizoprionodon terraenovae </em>and, to a lesser extent, skates and rays. Increasing the fishing mortality imposed by trawl fisheries affected only a few elasmobranch groups, primarily skates and rays. Increases and decreases in biomass lasted only as long as fishing mortality was elevated, although a lag time was observed for some groups to recover to initial biomass. Simulating a time/ area closure for juvenile blacktip sharks caused increases in their biomass but decreases in juvenile coastal shark biomass, a competing multispecies assemblage that is the apparent competitor. Topdown control scenarios resulted in greater variation and magnitude of response than those elicited under mixed control, although the direction of the response was similar. In general, reduction of targeted sharks did not cause strong top-down cascades.
