Capacitor oil samples (PCBs > 90%wt) were treated in a bench scale experiment to investigate the destruction of PCBs during chemical destruction processes (a catalytic hydrodechlorination treatment with palladium carbon and additional treatment with potassium tert-butyloxide). Using those results, this study confirmed the decrease of PCBs and other undesirable dioxin-like compounds such as PCDD/Fs in treated samples during the treatment. Dioxin-responsive chemical-activated luciferase expression (DR CALUX®) AhR reporter gene bioassay was used to evaluate dioxin-like activity in the samples. During the treatment, the efficiency for PCB capacitor oil was around 99.99% or more in WHO-TEQ and CALUX-TEQ, whereas the sum of PCBs was reduced at a resulting efficiency of >99.9999%. In this study, a new cleanup procedure for separating PCBs from the mineral oil matrix was also developed for DR CALUX. The procedure consists of dimethylsulphoxide partitioning followed by silica gel-44% sulphuric acid reflux treatment and activated carbon chromatography. With the cleanup, CALUX-TEQ values were in good agreement with WHO-TEQ values and were as much as 3.3 times higher than WHO-TEQs for untreated/treated PCB-containing insulating oil samples. The DR CALUX results of mineral oil samples containing various PCB concentrations of 0.5–50 mg/kg (corresponding WHO-TEQs: 0.012–1.2 μg-TEQ/g) also correlated well with WHO-TEQs (CALUX-TEQ/WHO-TEQ ratio =1.0–3.0), which was consistent with the theoretical quantification limit of the CALUX. These results supported the validity of the proposed clean-up method.

ABSTRACT Fish consumption is the principal source of intake of organochlorinated compounds in humans. Compared with other types of foods of animal origin, fish contain the highest levels of polychlorinated biphenyls (PCBs), polychlorinated dibenzo-p-dioxins, and polychlorinated dibenzofurans, all of which are classified as highly toxic organochlorine compounds. Currently, lakes and fish farms in northern Italy are not regularly monitored for PCBs and dioxins in areas contaminated by industrial sources, partially because of the high costs of traditional analytical methods that limit the number of samples to be analyzed. The DR-CALUX cell bioassay is based on the uptake of the cellular aryl hydrocarbon receptor (AhR) for dioxins and dioxin-like compounds. The aim of this study was to assess the levels of dioxins and dioxin-like PCB contamination in Lake Maggiore and Lake Como, two lakes in northwestern Italy, and in nearby areas. The levels were quantified using the cell bioassay DR-CALUX and reference controls in two wild fish species, perch (Perca fluviatilis) and roach (Rutilus rutilus), and in a farmed species, rainbow trout (Oncorhynchus mykiss). Tissue samples collected from the farmed rainbow trout were also submitted to immunohistochemical analysis of CYP1A expression as a marker for environmental pollutant-induced liver damage. The levels of dioxins, furans, and dioxin-like PCBs were all below the maximum levels and action limits set by European Union Regulation, suggesting no risk for human health associated with the consumption of the fish species caught or farmed in these areas.

Biological-effects monitoring in estuarine environments is complex as a result of strong gradients and fluctuations in salinity and other environmental conditions, which may influence contaminant bioavailability and the physiology and metabolism of the organisms. To select the most robust and reliable biological-effect methods for monitoring and assessment programmes, a large-scale field study was conducted in two estuarine transects in the Netherlands. The locations ranged from heavily polluted harbour areas (the ports of Rotterdam and Amsterdam) to cleaner coastal and freshwater sites. Assessment methods used included a variety of biomarkers in flounder (Platichthys flesus) and a range of in vitro (sediment extracts) and in vivo bioassays. Multivariate statistical analysis was applied to investigate correlations and relationships between various biological effects and contaminant levels in flounder liver or sediments. Several biological methods seemed to be too much affected by salinity differences for routine use in estuaries. The most discriminative biomarkers in the study were hepatic metallothionein content and biliary 1-OH pyrene in fish. Mechanism-based in vitro assays DR-CALUX and ER-CALUX applied to sediment extracts for screening of potential toxicity were much more responsive than in vivo bioassays with macro-invertebrates using survival as an endpoint.

A topsoil sample obtained from a highly industrialized area (Taranto, Italy) was tested on the DR-CALUX® cell line and the exposed cells processed with proteomic and bioinformatics analyses. The presence of polyhalogenated compounds in the topsoil extracts was confirmed by GC-MS/MS analysis. Proteomic analysis of the cells exposed to the topsoil extracts identified 43 differential proteins. Enrichment analysis highlighted biological processes, such as the cellular response to a chemical stimulus, stress, and inorganic substances; regulation of translation; regulation of apoptotic process; and the response to organonitrogen compounds in light of particular drugs and compounds, extrapolated by bioinformatics all linked to the identified protein modifications. Our results confirm and reflect the complex epidemiological situation occurring among Taranto inhabitants and underline the need to further investigate the presence and sources of inferred chemicals in soils. The combination of bioassays and proteomics reveals a more complex scenario of chemicals able to affect cellular pathways and leading to toxicities rather than those identified by only bioassays and related chemical analysis. This combined approach turns out to be a promising tool for soil risk assessment and deserves further investigation and developments for soil monitoring and risk assessment.

To study somatostatin/dopamine (SS/D) synergy in a human cell system constitutively expressing SS and D receptors (SSR and DR, respectively), we characterized the expression of SSR and DR subtypes in the non-small-cell lung cancer line Calu-6, and then we evaluated the effect on cell proliferation of SS/D chimeric molecules (BIM-23A387 and BIM-23A370), which bind with high affinity both sst2 and D2R, and compared the results with those obtained by using SS-14 and subtype-selective SS analogs (SSA) and D agonists (DA). Because Calu-6 cells produce insulin-like growth factor (IGF) and IGF-binding protein (IGFBP) peptides, which play a role in the autocrine/paracrine control of cell growth, we also investigated the effects of chimeric compounds on secretion and expression of IGF system components. Relative high levels of sst2 and the long isoform of the D2R were detected by real-time RT-PCR and Western blot in Calu-6, together with sst5 and to a lesser extent sst3 and D4R. BIM-23A387 and BIM-23A370 significantly inhibited growth of Calu-6, whereas IGF-IGFBP secretion or expression was unaffected, suggesting a direct inhibitory effect. The inhibition of cell growth, measured by both [3H]thymidine incorporation and cell count, was significantly lower when individual SSA and DA control peptides or subtype-specific SSA and DA were tested. BIM-23A370 was more potent than BIM-23A387 ( P < 0.001). These findings show that SS/D chimeras can inhibit Calu-6 proliferation in an IGF-independent manner and suggest that this enhanced potency might be because of the induction of SSR/DR dimerization. The Calu-6 cell line, constitutively expressing SSR and DR, provides a suitable model to elucidate the mechanism of action of SSA and DA on regulation of cell growth and to characterize the interaction between SSR and DR.

Abstract Introduction Air pollution, water and soil pollution produce impaired quality of life and expose the population to contaminants that can cause cancer. One of the main pollutants are DLC, a group of highly toxic chemicals that enter the body in about 90% through our food, and are classified as type I carcinogens by the International Agency for Research on Cancer. In the U.S. and Europe during the twentieth century there was an increase in the incidence of NHL that has stabilized in recent years which is coincident with the implementation of strong dioxin emission regulations. In Chile there has been a higher incidence of NHL in recent decades so we decided to evaluate DLC levels in patients with NHL and compared them with a normal control population. Patients and Methods In order to achieve this goal a descriptive observational, case-control unpaired, epidemiological study was designed. Patients with NHL diagnosed in the Hospital of Valdivia in 2011 and 2012 were considered in the study. The control group was composed by individuals without cancer, from the same geographical area as the patients. All volunteers were asked if they wished to participate in the study using an informed consent. Blood sample without anticoagulant was obtained from each individual. Serum was aliquoted and stored at -20 ° C for later quantification of total lipids and DLC with sulfo-phospho-vanillin assay colorimetric method and DR-CALUX ® Bioassay, respectively (screening method approved by FDA and EEC). A nonparametric statistical hypothesis was constructed using Wilcoxon rank sum test to compare two independent samples considering a non-symmetrical distribution of the variable DLC (pg TEQ / total lipids) and a significance level of α= 5%. Results 51 patients with NHL and 100 controls entered the study. The mean ages of the LNH group and the control group were 59±14.5 and 39± 16.9 years, respectively. The male/female ratio was 0.52 and 0.82 in the LNH and control group, respectively. NHL cases were from southern regions of Chile matching that of controls in a 100%. The histopathology of NHL cases was in 90% of B lineage. The concentration of DLC was quantified in all cases and expressed in toxicity equivalents (TEQ) using as reference the 2,3,7,8, tetracloroparadibenzodioxine and corrected to the value of total serum lipids (pg TEQ / total lipids). In all samples analyzed DLC were detected. The DLC levels found for NHL cases and controls were 28.33 ± 28.57 and 15.02 ± 7.64 pg TEQ / total lipids, respectively. The median values for cases and controls were 19.02 and 12.67 pg TEQ / total lipids, respectively. The analysis with Wilcoxon rank sum test showed a statistically significant evidence (z = -4.44, p = 0.0000) (Figure 1). Conclusions The cases and controls analyzed came from a geographical area of 1200 km of extension without polluting industries having detectable levels of DLC in all them. DLC levels NHL cases were higher compared with controls. When comparing the results of the controls (15.02 pg TEQ / g lipid) with other studies in unexposed populations we find similar levels in Northern Australia and Taiwan, 10.9 and 15.2 pg TEQ / g lipid, respectively (Harden et al. 2007 and Chen et al., 2005). Levels are higher in areas with higher industrial activity as France, Slovakia and Belgium (Marchand et al. 2004 Chovancová et al 2012, Bilau et al., 2009), and lower for residents of Fukuoka and Athens (Masuda et al. , 2005 Costopoulou et al., 2006). The vast region from which the controls come from in our study, with an environment relatively free of pollution leads us to infer that DLC are basically food contaminants, since the majority of the population consume food products manufactured in areas of high industrial activity and distributed throughout the country. DLC studies in populations exposed to higher pollution (Viel et al., 2008) have shown increased incidence of NHL in those areas. This makes us pose the hypothesis of a potential pathogenic role of DLC in NHL that could be mediated by genetic mechanisms and / or deregulated B cell expansion. Анотація:
Objective: To present our ‘methylene blue-guided retroperitoneoscopy technique’ that we apply in cases where extravasation develops for various reasons and is complicated as a result in Percutaneous Nephrolithotomy surgery (PNL). Material and Methods: A total of 36 patients, who underwent PNL with the ‘methylene blue technique’ at Bakirkoy Dr. Sadi Konuk Training and Research Hospital between 2014 and 2020, were retrospectively evaluated. The study included only cases in which the ‘methylene blue-guided retroperitoneoscopy technique’ was used due to the inability to advance the Amplatz sheath to the targeted calyx due to renal scarring or a staghorn stone filling the targeted calyx, Amplatz sheath was withdrawn from the kidney with the guidewire due to the inattention of the assistant surgical team or contrast material extravasation. The patients’ demographic, preoperative, perioperative, and postoperative data were recorded, and stone-free and complication rates were noted. Results: The mean age of the patients was 45.1 (36-55) years, the mean body mass index was 27.8±4 kg/m2, the mean stone size was 3.4±0.7 cm, and the mean stone volume was 22.3±10.2 cm3. The mean operation time was calculated as 95.8±30.3 minutes. The stone-free rate was 68% on the postoperative first day and 75% on the third month. Eight patients (22.2%) underwent a secondary procedure (extracorporeal shock wave lithotripsy or flexible ureterorenoscopy). Conclusion: The ‘methylene blue-guided retroperitoneoscopy technique’ we apply under retroperitoneoscopic direct vision is a safe and practical option for re-accessing the kidney by locating the calyx, which is first partially or completely dilated. Diabetic retinopathy (DR) is a neurovascular disease characterized by the reduction of retina integrity and functionality, as a consequence of retinal pigment epithelial cell fibrosis. Although galectin-1 (a glycan-binding protein) has been associated with dysregulated retinal angiogenesis, no evidence has been reported about galectin-1 roles in DR-induced fibrosis. ARPE-19 cells were cultured in normal (5 mM) or high glucose (35 mM) for 3 days, then exposed to the selective galectin-1 inhibitor OTX008 (2.5–5–10 μM) for 6 days. The determination of cell viability and ROS content along with the analysis of specific proteins (by immunocytochemistry, Western blotting, and ELISA) or mRNAs (by real time-PCR) were performed. OTX008 5 μM and 10 μM improved cell viability and markedly reduced galectin-1 protein expression in cells exposed to high glucose. This was paralleled by a down-regulation of the TGF-β/, NF-kB p65 levels, and ROS content. Moreover, epithelial–mesenchymal transition markers were reduced by OTX008 5 μM and 10 μM. The inhibition of galectin-1 by OTX008 in DR may preserve retinal pigment epithelial cell integrity and functionality by reducing their pro-fibrotic phenotype and epithelial–mesenchymal transition phenomenon induced by diabetes.

All studies were conducted during the years 2020-2021 in the village of Kzo and its environs of Dohuk Governorate / northern of Iraq, the results of the survey led to the collection of 42 samples, Samples which all belongs to the genus Xylocopa , Six samples represent four species from all samples were sent to Dr Munazah Associate Professor, Department of Entomology, University of Agriculture and Science and Technology of Kashmir, India for classification , The result of classification showed that three of them were belong to the sub-genus Xylocopa represent two species,one male of species Xylocopa varentzowi The main character is that the front legs were contained a dense group of long white hairs on the outer part of the tibia, Male and female of species Xylocopa violacea the female were contained a density of black hairs on the front legs and more on the hind legs,The males of this species are distinguished by their antenna, in which the flagella consists of 11 segments and its segments is brown, but the ninth and tenth segments (9-10) which were characterized by their yellowish-orange color,and the other three specimens were belong to the sub-genus Proxylocopa represent two species, one female of species Xylocopa rufa were characterized by the calyx sita or hairs which covering edges of the abdominal region which were interrupted and unconnected in the middle, male and female of species Xylocopa olivieri were characterized by the hairs which were covered the edges of the abdominal tergit segments, pale yellow color, continuous and unbroken in the middle, The results of phenotypic studies of male and females samples of the four species Xylocopa varentzowi, Xylocopa violacea, Xylocopa rufa and Xylocopa olivieri is reported for the first time from the high altitudes of Duhok Governorate( more than 1,140 m above sea level), Iraq.

Abstract Excretion of toxicants accumulated from firefighter exposures through breastmilk represents a potential hazard. We investigated if firefighting exposures could increase the concentration of polybrominated diphenyl ethers (PBDEs) and aryl hydrocarbon receptor (AhR) activation in excreted breastmilk. Firefighters and non-firefighters collected breastmilk samples prior to any firefighting responses (baseline) and at 2, 8, 24, 48, and 72 hours after a structural fire (firefighters only). Five PBDE analytes (BDEs 15, 28, 47, 99, and 153) detected in at least 90% of samples were summed for analyses. The AhR in vitro DR CALUX® bioassay assessed the mixture of dioxin-like compounds and toxicity from breastmilk extracts. Baseline PBDEs and AhR response were compared between firefighters and non-firefighters. Separate linear mixed models assessed changes in sum of PBDEs and AhR response among firefighters over time and effect modification by interior or exterior response was assessed. Baseline PBDE concentrations and AhR responses did not differ between the 21 firefighters and 10 non-firefighters. There were no significant changes in sum of PBDEs or AhR response among firefighters over time post-fire, and no variation by interior or exterior response. Plots of sum of PBDEs and AhR response over time demonstrated individual variation but no consistent pattern. Currently, our novel study results do not support forgoing breastfeeding after a fire exposure. However, given study limitations and the potential hazard of accumulated toxicants from firefighter exposures excreted via breastfeeding, future studies should consider additional contaminants and measures of toxicity by which firefighting may impact maternal and child health.

Abstract: Nephrolithiasis is a disease which the symptom is indicated by the existence of a single or more solid mass of hard material like a stone which is found in the kidney tubule, calyx, infundibulum, kidney pelvis, and the whole of kidney calyx of the sufferer. Mostly, the doctors use imaging like ultrasonography to checkup the patients’ condition in order to ascertain the diagnosis of nephrolithiasis. Ultrasonography can give the spesific image if there is any stone located in the kidney. As a result, doctors will get some easiness in determining the patients diagnosis. The objective of this research is to figure out the kidney image resulted from ultrasonography of nephrolithiasis sufferers in Radiology Division, Medical Faculty, Samratulangi University/ Faculty Students Senate of Radiology, Public Service Corporation of Prof. Dr. R. D. Kandou Hospital, Manado, In the period of January 1st – June 30th 2014.The researcher used descriptive retrospective as the research method. By using medical notes found in Radiology Division, Public Service Corpration of Prof. Dr. R. D. Kandou Hospital, Manado, In the period of January 1st – June 30th 2014 as secondary data. Conclusion:The researcher then found that there were 105 cases of nephrolithiasis from totally result of kidney ultrasonography to the sufferers of nephrolithiasis. Many of the suferrers were men (62,9%) in average ages from 56 to 65 years old (36,2%). According to the location, kidney stone were found mostly in bilateral nepholithiasis (37,1%). The resercher also figured out that most of nephrolithiasis sufferers had a complication with chronic kidney disease (39,0%) and complication with hidronephrosis (19,0%). The patients who complain about pain on their weists should have kidney ultrasonography test to help the doctor to diagnose the causes, to avoid the possibity of another abnormal organ, and prevent the serious nephrolithiasis causes.Keywords: kidney ultrasonography, nephrolithiasisAbstrak: Nefrolitiasis merupakan suatu penyakit dengan gejala ditemukannya satu atau beberapa massa keras seperti batu yang terdapat di dalam tubuli ginjal, kaliks, infundibulum, pelvis ginjal, serta seluruh kaliks ginjal. Sehingga mempermudah dokter untuk menentukan diagnosis pasien. Kandou Manado Periode 1 Januari – 30 Juni 2014.Penelitian ini merupakan penelitian deskriptif retrospektif dengan memanfaatkan data sekunder berupa catatan medik yang terdapat di Bagian Radiologi BLU RSUP Prof. Dr. R. D. Kandou Manado Periode 1 Januari – 30 Juni 2014. Simpulan:Keseluruhan hasil Ultrasonografi ginjal pada penderita Nefrolitiasis ditemukan 105 kasus nefrolitiasis, dengan penderita nefrolitiasis lebih banyak terjadi padalaki-laki(62,9%).Penderita nefrolitiasis terbanyak pada kelompok umur 56 – 65 tahun (36,2%).Penderita nefrolitiasis berdasarkan letak batu yaitu nefrolitiasis bilateral (37,1%).Penderita nefrolitiasis dengan komplikasi CKD yaitus ebanyak (39,0%).Penderita nefrolitiasis dengan komplikasi Hidronefrosis yaitu sebanyak (19,0%). Penderita yang datang dengan keluhan rasa nyeri pada daerah pinggang sebaiknya dipastikan penyebabnya melalui pemeriksaan Ultrasonografi ginjal untuk membantu mendiagnosis, menyingkirkan kemungkinan kelainan pada daerah organ lainnya dan mencegah memberatnya penyebab nefrolitiasis.Kata kunci: ultrasonografi ginjal, nefrolitiasis

Abstract: Nephrolitiasis is a disease indicated by the existence of a single or more solid masses of hard material in the kidney tubule, calyx, infundibulum, kidney pelvis, and the whole parts of the kidney. Imaging such as non-contrast CT-Scan is usually used to ascertain the diagnosis of nephrolitiasis. This study was aimed to obtain the profile of non-contrast CT-Scan of the kidney performed on nephrolithiasis cases at Radiology Division of Medical Faculty University of Sam Ratulangi/Prof. Dr. R. D. Kandou Hospital Manado in the period of January 1 to Agust 30, 2016. This was a descriptive retrospective study using medical records at the Radiology Division. The results showed that there were 63 cases of nephrolitiasis in this study. The highest percentages were in males (71.43%) and age group of 56-65 tahun (25.39%). According to the location, most cases with bilateral nephrolitiasis (25.39%). Complication of hydronephrosis was found in 14.29% of cases. Conclusion: Based on the non-contrast CT-Scan of the kidney, most nephrolithiasis cases were found in males, age group of 46-55 years, bilateral nephrolithiasis, and without complication.Keywords: non-contrast CT-Scan of the kidney, nephrolitiasisAbstrak: Nefrolitiasis merupakan suatu penyakit dengan gejala ditemukannya satu atau beberapa massa keras yang terdapat di dalam tubuli ginjal, kaliks, infundibulum, pelvis ginjal, serta seluruh bagian ginjal. Kandou Manado periode 1 Januari-30 Agustus 2016. Jenis penelitian ialah deskriptif retrospektif dengan memanfaatkan data rekam medik di Bagian Radiologi RSUP Prof. Dr. R. D. Kandou Manado periode 1 Januari -30 Agustus 2016. Hasil penelitian mendapatkan 63 kasus nefrolitiasis dengan hasil CT-Scan ginjal non-kontras, lebih banyak terjadi pada laki-laki (71,43%) dan kelompok usia 46-55 tahun (33,33%). Berdasarkan letak batu, lebih sering ditemukan letak batu bilateral (36,68%). Komplikasi hidronefrosis ditemukan pada 14,29% kasus. Simpulan: Berdasarkan hasil CT-Scan non-kontras pada penderita nefrolitiasis didapatkan nefrolitiasis lebih sering ditemukan pada laki-laki, kelompok usia 46-55 tahun, dengan letak batu bilateral dan tanpa komplikasi.Kata kunci: CT-Scan tanpa kontras pada ginjal, nefrolitiasis

Abstract This study focusses on the adsorption efficiency of cotton pod (Gossypium arboreum) for biosorption of direct red (DR) 80 dye. The effect of dye concentration, pH, shaking time, adsorbent dose and temperature was considered to evaluate the adsorption efficiency of the cheapest raw material on the removal of dye. Various concentrations (10–50 ppm) were examined and maximum percentage removal of the adsorbent was obtained at pH 1 with adsorbent dose of 0.9 g and shaking time of 60 min. To enhance the adsorption efficiency of the adsorbent, the cotton calyx was treated with FeSO4.7H2O and investigated the removal of the raw adsorbent and the composite. It was examined that the removal efficiency of the composite is more than that of raw adsorbent. The composite was more efficient as dye removal reached up to 97%. FTIR of the composite shows an additional peak at 650 cm−1 for presence of Fe-O group and confirmation of composite formation. SEM micrograph showed that the particles in composite are more packed compared to the raw adsorbent. Mathematical models were applied and kinetic studies also have been done to provide better results regarding to the experimental data. It is concluded that the prepared adsorbent could be used as a tool for the removal of toxic pollutants from textile wastewater.

Abstract. Dechavez R, Calub ML, Genobata DR, Balacuit R, Jose R, Tabugo SR. 2022. Identification of culture-dependent microbes from mangroves reveals dominance of Bacillus including medically important species based on DNA signature. Biodiversitas 23: 5342-5350. Mangroves are among the most crucial plant hosts in the marine environment because of their notable role in the ecosystem and their benefits. Identification of plant-associated and culture-dependent microbes is key to having a rich reservoir of bioactive substances. DNA barcoding was done based on 16S rRNA gene and ITS region. In this study, seven (7) mangrove species were identified as hosts. Results show that most bacterial isolates from plant leaves were gram-positive bacteria belonging to phylum: Firmicutes and genus Bacillus. Phylogenetic inference reveals a diverse non-monophyletic group with medically and economically necessary species. Among the host plants, Sonneratia alba harbored the most species of bacteria and fungi. These include two medically important strains of B. cereus and B. thuringiensis and host-specific bacteria like B. altitudinis in S. alba, B. velezensis in Rhizophora apiculata, B. clausii in Avicennia marina and B. firmus in A. marina were revealed. Meanwhile, some bacteria like B. subtilis and B. megaterium were found in three host species and B. cereus was the most abundant being recorded in four host plants. Diverse strains of fungi, Aspergillus sp., A. nomiae, A. tubingensis and A. niger were also present. This study served as baseline data for future research on diversity and host ecology for possible drug discovery.
