Studies have shown that street network centrality measures are capable of explaining a significant proportion of pedestrian activity. These studies typically employ street centreline networks that differ significantly from the networks that pedestrians use to traverse the built environment. Presently, centrality approaches are rarely applied to dedicated pedestrian network (DPNs). This creates uncertainty regarding their ability to explain pedestrian activity when derived from DPNs. This study addresses that gap by investigating the extent to which centrality metrics derived from DPNs can explain observed pedestrian densities, both alone and when controlling for other built environment variables in metro station environments in Asia. In total, four DPNs were created centred on metro stations in Bangkok, Manila, Osaka, and Taipei chosen to represent different urban typologies. Multivariate results show that centrality metrics alone explain a mere 6–24% of observed pedestrian densities when calculated on DPNs. When all factors are considered, the contribution of centrality remained consistent in most study sites but is somewhat reduced with land-use variables and proximity to rail transit revealed as the strongest predictors of pedestrian density. Pedestrian design factors were also frequently associated with pedestrian density. Finally, stronger associations between centrality and pedestrian densities were observed in the denser, more complex pedestrian environments. These findings provide insight into the performance of centrality measures applied to DPNs expanding pedestrian network research in this area.

Diaphragmatic force, determined by stimulating the phrenic nerve while simultaneously measuring the pressures in a closed respiratory system, was assessed in five anesthetized dogs over a 5-h period to evaluate the inherent variability of this technique. Transdiaphragmatic pressure (Pdi) was measured at functional residual capacity during stimulation (120 Hz, 0.2-ms duration) of one phrenic nerve by either direct phrenic nerve stimulation (DPNS) or transvenous phrenic nerve stimulation (TPNS). An analysis of variance showed no significant (P greater than 0.50) change during the 5-h period. There was a significant correlation (r = 0.94, P less than 0.001) between Pdi obtained by TPNS and that obtained by DPNS. It is concluded that either DPNS or TPNS can be used to evaluate diaphragmatic strength over a 5-h period and that TPNS can be used in lieu of DPNS.

We developed dual pH-sensitive, fluorescent dye-loaded polypeptide nanoparticles (DPNs) for ratiometric sensing of pH changes. DPNs have good pH responses for solution and cells. Single acid activatable fluorescent dye doped nanoparticles can distinguish tumor tissue from normal tissue by monitoring the acidic extracellular environment.

Abstract The paper addresses the issue of meaning-making in a highly prolific and sprawling segment of English vocabulary – derivatives from precedent names (DPNs). The combination of cognitive linguistics methods applied to analyze the semantics of DPNs (Robin Hood cluster, Cinderella-based blends and Dorian Gray effect) permitted to account for their bias towards polysemy, which seems to be basically grounded in the process of metonymic zoom-in on the selected content in the event frame that describes the precedent name, oftentimes leading to domain extension and indeterminacy.

In a Drug Prescription Network (DPN), each drug is represented as a node and two drugs co-prescribed to the same patient are represented as an edge linking the nodes. The use of DPNs is a novel approach that has been proposed as a means to study the complexity of drug prescription. The aim of this study is to demonstrate the analytical power of the DPN-based approach when it is applied to the analysis of administrative data. Drug prescription data that were collected at a local health unit (ASL TO4, Regione Piemonte, Italy), over a 12-month period (July 2018–June 2019), were used to create several DPNs that correspond to the five levels of the Anatomical Therapeutic Chemical classification system. A total of 5,431,335 drugs prescribed to 361,574 patients (age 0–100 years; 54.7% females) were analysed. As indicated by our results, the DPNs were dense networks, with giant components that contain all nodes. The disassortative mixing of node degrees was observed, which implies that non-random connectivity exists in the networks. Network-based methods have proven to be a flexible and efficient approach to the analysis of administrative data on drug prescription.

Penelitian ini dilakukan atas dasar pasar saham pada tahun 2015 mengalami kondisi bearish, sehingga terjadi perubahan asumsi risiko dan return pada penelitian-penelitian portofolio saham optimal terdahulu. Tujuan dari penelitian ini adalah untuk mengetahui saham-saham yang masih berpotensi memberikan return positif pada kondisi pasar bearish dengan menggunakan Model Indeks Tunggal. Penelitian ini menggunakan data sekunder dari saham-saham yang yang terdaftar di BEI tahun 2015. Hasil penelitian menunjukkan bahwa dari populasi 528 emiten, terseleksi sampel sebanyak 38 emiten. Kemudian, terdapat 13 saham yang memenuhi kriteria, yaitu saham TOTO, EMTK, HMSP, AMAG, PLIN, BISI, INAI, MYOR, AKRA, AHAP, BNBA, DPNS, dan BFIN. Berdasarkan perhitungan, investor akan mendapatkan return portofolio sebesar 0,010047 dengan risiko portofolio sebesar 0,022006. Untuk berinvestasi, investor dapat membagi pada beberapa saham dengan proporsi pada saham sebesar 34,89% TOTO, 4,03% EMTK, 13,62% HMSP, 6,92% AMAG, 1,25% PLIN, 3,80% BISI, 5,33% INAI, 6,20% MYOR, 15,16% AKRA, 0,90% AHAP, 6,76% BNBA, 0,28% DPNS, dan 0,78% BFIN.

A common complaint with day-neutral strawberries is that they perform poorly in mid-summer heat. Since most modern day-neutral cultivars are derived from the same Fragaria virginiana ssp. glauca clone from Utah, we felt it prudent to search for alternate sources of day-neutrality that were more heat-tolerant. We compared the sexual and vegetative performance of nine F. virginiana clones from a wide range of environments including the Utah site, and four F. × ananassa day-neutral types (`Aromas', `Fort Laramie', `Ogallala', and `Tribute') under constant temperatures of 18, 22, 26, and 30 °C and 12-h days. `Aromas' and `Tribute' carry the Utah source of day-neutrality, while `Fort Laramie' and `Ogallala' are old cultivars that have a different, complex background. After a 4-week period of acclimation, we counted the number of crowns, inflorescences, flowers, stolons, and daughter plants that emerged over a 10-week period, and measured the dry weights of component parts. ANOVA tables revealed that temperature regime (T), genotypes (G), and T*G were significant for flower number (FLN) and total dry matter accumulation, while species and T*G were significant for daughter plant number (DPN). Mean FLNs across the four temperatures were 6.8, 3.7, 3.3, and 1.2, while mean DPNs were 0.7, 0.9, 0.7, and 1.8. F. virginiana clones averaged 3.8 FLNs and 1.8 DPNs, while the F. × ananassa clones averaged 4.1 FLNs and 0.2 DPNs. There was generally more variability among the F. virginiana clones than the F. × ananassa clones, but the F. × ananassa cultivars, `Fort Laramie' and `Ogallala', performed best at 30 °C. The Wasatch clone did not flower in any treatment, suggesting it is not day-neutral.

Deep penetrating nevi (DPNs) are rare melanocytic neoplasms consisting of pigmented spindled or epithelioid melanocytes with a distinctive wedge-shaped configuration showing activation of the WNT pathway, with unusual cyto-architectural features. It is unclear whether they show a distinct genomic profile associated with a diverse metastatic potential. We describe herein a cohort of 21 atypical DPNs analyzed by next-generation sequencing using the Ion AmpliSeq™ Comprehensive Cancer Panel. We found that β-catenin exon 3 was mutated in 95% and MAP kinase pathway genes in 71% of the cases. Less frequent mutations were observed in HRAS (19%) and MAP2K1 (24%). Isocitrate dehydrogenases 1 (IDH1) mutations, including R132C, V178I, and S278L, were identified in 38% of cases and co-existed with BRAF/HRAS mutations. The only case with progressive nodal disease carried alterations in the β-catenin pathway and mutations in IDH1 and NRAS (codon 61). By a comprehensive mutation analysis, we found low genetic heterogeneity and a lack of significant associations between specific gene mutations and histopathological features, despite atypical features. Whether the acquisition of an NRAS or IDH1 mutation in an atypical DPN may represent a molecular evolution implying a pathway to melanoma progression should be confirmed in a larger series.

AbstractThe operational backbone of modern organizations is the target of business process management, where business process models are produced to describe how the organization should react to events and coordinate the execution of activities so as to satisfy its business goals. At the same time, operational decisions are made by considering internal and external contextual factors, according to decision models that are typically based on declarative, rule-based specifications that describe how input configurations correspond to output results. The increasing importance and maturity of these two intertwined dimensions, those of processes and decisions, have led to a wide range of data-aware models and associated methodologies, such as BPMN for processes and DMN for operational decisions. While it is important to analyze these two aspects independently, it has been pointed out by several authors that it is also crucial to analyze them in combination. In this paper, we provide a native, formal definition of DBPMN models, namely data-aware and decision-aware processes that build on BPMN and DMN S-FEEL, illustrating their use and giving their formal execution semantics via an encoding into Data Petri nets (DPNs). By exploiting this encoding, we then build on previous work in which we lifted the classical notion of soundness of processes to this richer, data-aware setting, and show how the abstraction and verification techniques that were devised for DPNs can be directly used for DBPMN models. This paves the way towards even richer forms of analysis, beyond that of assessing soundness, that are based on the same technique.

Diabetic peripheral neuropathy (DPN) is a complication of diabetes mellitus that occurs in both type 1 and type 2 diabetics. This complication is estimated to be prevalent in approximately 50% of the diabetic population and is associated with significant morbidity and mortality. Painful DPNs affect a minority of patients with DPN but are often chronic, severe, and debilitating. Many pharmacologic agents—including but not limited to pregabalin, duloxetine, tricyclic antidepressants, abapentin, and carbamazepine—have been studied and proven effective for the treatment of pain associated with DPN. Data from these studies were extracted and evaluated to compile a list of agents that are effective in treating painful diabetic neuropathies.

This paper presents a new scheme of distributed pump-system for district heating network. As no circulating pumps for the heat sources and no throttle valves for the users, much energy is saved. And the control of hydraulic parameters can be adjusted without interfering with that of the thermodynamic parameters. The novel scheme is available for both looped pipeline networks (LPNs) with multiple heat sources and dendritic pipeline networks (DPNs) with single heat source. Moreover, for users located at the hydraulic intersection points of the looped pipe network with multi-heat sources, their water temperature can be less affected by their own water consumption. A case study was presented and about 22% power saved when the new scheme was employed.

It is therefore extremely important that the math teacher has a deep understanding of the functioning of numeral systems, in especial, the decimal positional numeral system (DPNS). In this article, we study the properties of numeral systems, in particular those related to the criteria of divisibility and the representation of real numbers in positional systems. We address the topic from three main aspects: its historical evolution; purely mathematical properties of positional numeral systems; and how the teacher can use the two previous aspects in their classes.

The purpose of this study was to determine the effect of the results of capital intensity and inventory intensity on tax avoidance in chemical sub-sector companies listed on the Indonesia Stock Exchange. The sample in this study used a purposive method with a total sample of 33 samples from 11 chemical sub-sector companies listed on the Indonesia Stock Exchange from 2017-2019 with company codes as follows AGII, BRPT, BUDI, DPNS, EKAD, INCI, MDKI, MOLI, SRSN, TPIA, UNIC. In this study, capital intensity and inventory intensity as independent variables, and tax avoidance as the dependent variable. And the results of this study indicate that capital intensity has a positive effect on tax avoidance, inventory intensity also has a positive effect on tax avoidance and simultaneously capital intensity and inventory intensity both have a significant effect on tax avoidance.

Traditionally Business Process Modeling has only focused on the control-flow perspective, thus allowing process designers to specify the constraints on the activities of the process: the order and potential concurrency of their execution, their mutual exclusivity, the possibility of being repeated, etc. However, activities are executed by different resources, manipulate data objects and are constrained by the state of such objects. This requires that the traditional notion of soundness, typically introduced for control-flow-only models, is extended so as to consider data. Intuitively, a (data-aware) process model is sound if (1) it does not contain deadlocks, (2) no more activities are enabled when the process instance is marked as completed and finally (3) there are no parts of the model that cannot be executed. Although several data-aware notations have been introduced in the literature, not all of these are given a formal semantics. In this paper, we propose a technique for checking the data-aware soundness for a specific class of such integrated models, with a simple syntax and semantics, building on Data Petri Nets (DPNs). These are Petri nets enriched with case variables, where transitions are guarded by formulas that inspect and update such variables, and are of the form variable-operator-variable or variable-operator-constant. Even though DPNs are less expressive than Petri nets where data are carried by tokens, they elegantly capture business processes operating over simple case data, allowing to model complex data-aware decisions. We show that, if a DPN is data-aware sound, the Constraint Graph is a finite-state automaton; however, a finite-state Constraint Graph does not guarantee data-aware soundness, but provides a finite structure through which this property can be checked. Finally, we investigate further properties beyond data-aware soundness, such as the problem of verifying that an actor participating in the business process can unilaterally enforce data-aware soundness by restricting the possible executions of a bounded DPN, assuming this actor to be able to control the firing of some transitions and decide the value of some of the case variables whenever these are updated.

Deep penetrating nevus (DPN) is an uncommon acquired melanocytic lesion with a distinct histopathological appearance that typically behaves in an indolent manner. The lesion is characterized by a symmetrical proliferation of epithelioid to spindled melanocytes associated with abundant melanophages and wedge-shaped extension to the deep reticular dermis and subcutis. Pronounced cytologic atypia and mitotic figures are usually absent, which helps distinguish DPN from melanoma with a deep penetrating growth pattern. Recently, the concept of atypical DPN has been proposed for lesions that demonstrate borderline histomorphologic features and may be associated with lymph node deposits but lack the copy number aberrations typical of melanoma by either fluorescence in situ hybridization or comparative genomic hybridization. While most of these lesions have a favorable clinical course, rare lesions may progress to melanoma. In this review, we summarize the current literature on atypical DPNs with uncertain behavior/metastatic potential and outline the characteristics that distinguish these lesions from conventional DPN and melanoma with DPN-like features.

In recent years, the study of social networks and the analysis of these networks in various fields have grown significantly. One of the most widely used fields in the study of social networks is the issue of link prediction, which has recently been very popular among researchers. A link in a social network means communication between members of the network, which can include friendships, cooperation, writing a joint article or even membership in a common place such as a company or club. The main purpose of link prediction is to investigate the possibility of creating or deleting links between members in the future state of the network using the analysis of its current state. In this paper, three new similarities, degree neighbor similarity (DNS), path neighbor similarity (PNS) and degree path neighbor Similarity (DPNS) criteria are introduced using neighbor-based and path-based similarity criteria, both of which use graph structures. The results have been tested based on area under curve (AUC) and precision criteria on datasets and it shows well the superiority of the work over the criteria that only use the neighbor or the path.

General ward nurses play a key role in discharge planning for end-stage cancer patients. It is necessary to assess the factors regarding their practice to promote discharge planning in accordance with end-stage cancer patients’ wishes. This study aimed to investigate the relationships between general ward nurses’ practice of discharge planning for end-stage cancer patients, self-efficacy, ethical behavior, attitude, knowledge and experience, perceived skills, and perceived barriers. A total of 288 general ward nurses from nine hospitals in a city in Japan completed the questionnaire. Path analysis was conducted to test the hypotheses. The results showed that nurses’ self-efficacy, ethical behavior (do-no-harm, do-good), knowledge (experience of attending home care seminars), and perceived skills (assertiveness) were positively and directly related to the practice of discharge planning. Nursing experience and perceived skills (assertiveness) were positively associated with discharge planning practice, while perceived barriers (death discussion) and attitude (degree of leaving it to discharge planning nurses (DPNs)) were negatively associated, with self-efficacy acting as a mediator. Thus, our findings show that it is important to enhance self-efficacy and nursing ethical behavior to improve the practice of discharge planning. Accordingly, education regarding home care, assertive communication skills, death discussion, and ethics is needed for general ward nurses.

Abstract Older adults are the fastest growing subset of complex patients with high medical, behavioral, and social needs. Understanding differences in disease progression patterns between complex and non-complex older adults is critical for understanding disease risk and tailoring patient-centered interventions. We identified complex patients as those having frequent medical encounters and multiple chronic conditions within the first year of the study period and non-complex patients as the converse. This study compares the disease progression patterns of (a) complex and (b) non-complex older adults by creating disease progression networks (DPN) from claims data of 762,362 patients (mean age = 73) from 2016 to 2020. We characterized the network size and density between the complex patient DPN (C-DPN) and non-complex patient DPN (NC-DPN), and compared disease progression incidence, time-to-progression, and age- and gender-related risk. Results show that the C-DPN was denser and had a wider range of values for risk of progression compared to the NC-DPN. This implies more varied disease progression patterns occurring in the complex adults. We were also able to compare (median) time-to-progressions of diseases relative to each subpopulation and found variation in disease progression time. Furthermore, k-means clustering on the network allowed us to identify highly connected diseases involved in many disease pathways that are prevalent among older adults. (e.g., lipoprotein disorders, hypertension, major depressive disorder). Our results suggest that DPNs can be used to identify important conditions and time-points for tailoring care to the complex and non-complex older adults.

This work aims to enhance the polarity of natural rubber by grafting copolymers onto deproteinized natural rubber (DPNR) to improve its compatibility with silica. Poly(acrylic acid-co-acrylamide)-grafted DPNR ((PAA-co-PAM)-DPNR) was successfully prepared by graft copolymerization with acrylic acid and acrylamide in the latex stage, as confirmed by FTIR. The optimum conditions to obtain the highest conversion, grafting efficiency, and grafting percentage were a reaction time of 360 min, a reaction temperature of 50 °C, and an initiator concentration of 1.0 phr. The monomer conversion, grafting efficiency, and grafting percentage were 91.9–94.1, 20.8–38.9, and 2.1–9.9%, respectively, depending on the monomer content. It was shown that the polarity of the natural rubber increased after grafting. The (PAA-co-PAM)-DPNR was then mixed with silica to prepare DPNR/silica composites. The presence of the (PAA-co-PAM)-DPNR and silica in the composites was found to improve the mechanical properties of the DPNR. The incorporation of 10 phr of silica into the (PAA-co-PAM)-DPNR with 10 phr monomer increased its tensile strength by 1.55 times when compared to 10 phr of silica loaded into the DPNR. The silica-filled (PAA-co-PAM)-DPNR provided s higher storage modulus, higher Tg, and a lower tan δ peak, indicating stronger modified DPNR/silica interactions and greater thermal stability when compared to silica-filled DPNR.

DNA–Protein cross-links (DPCs) are cytotoxic DNA lesions with a protein covalently bound to the DNA. Although much has been learned about the formation, repair, and biological consequences of DPCs in the nucleus, little is known regarding mitochondrial DPCs. This is due in part to the lack of robust and specific methods to measure mitochondrial DPCs. Herein, we reported an enzyme-linked immunosorbent assay (ELISA)-based method for detecting mitochondrial DPCs formed between DNA and mitochondrial transcription factor A (TFAM) in cultured human cells. To optimize the purification and detection workflow, we prepared model TFAM-DPCs via Schiff base chemistry using recombinant human TFAM and a DNA substrate containing an abasic (AP) lesion. We optimized the isolation of TFAM-DPCs using commercial silica gel-based columns to achieve a high recovery yield for DPCs. We evaluated the microplate, DNA-coating solution, and HRP substrate for specific and sensitive detection of TFAM-DPCs. Additionally, we optimized the mtDNA isolation procedure to eliminate almost all nuclear DNA contaminants. For proof of concept, we detected the different levels of TFAM-DPCs in mtDNA from HEK293 cells under different biological conditions. The method is based on commercially available materials and can be amended to detect other types of DPCs in mitochondria.

Soybean (SB) extracts markedly inhibit formation of diphenylnitrosamine (DPNA) from the precursors diphenylamine (DPA) and nitrite (NO2-). SB extracts interact with nitrite, as shown by the decrease in the nitrite level. The rate of interaction between SB extracts and nitrite was lower than the rate of inhibition of DPNA formation. Thus the reduction of DPNA yield by SB extracts is not caused mainly through their competition for the available nitrite, but it might be due to the adsorption of the DPNA formed to the SB proteins, internitrosation between SB constituents and DPNA formed, or through the breakdown of the DPNA.

Introduction. Xeroderma pigmentosum group C (XPC), essential component of multisubunit stem cell coactivator complex (SCC), functions as the critical factor modulating pluripotency and genome integrity through interaction with Oct-4/Sox2. However, its specific role in regulating pluripotency and multilineage differentiation of human dental pulp cells (DPCs) remains unknown. Methods. To elucidate the functional role XPC played in pluripotency and multilineage differentiation of DPCs, expressions of XPC in DPCs with long-term culture were examined by real-time PCR and western blot. DPCs were transfected with lentiviral-mediated human XPC gene; then transfection rate was investigated by real-time PCR and western blot. Cell cycle, apoptosis, proliferation, senescence, multilineage differentiation, and expression of Oct-4/Sox2/c-Myc in transfected DPCs were examined. Results. XPC, Oct-4, Sox2, and c-Myc were downregulated at P7 compared with P3 in DPCs with long-term culture. XPC genes were upregulated in DPCs at P2 after transfection and maintained high expression level at P3 and P7. Cell proliferation, PI value, and telomerase activity were enhanced, whereas apoptosis was suppressed in transfected DPCs. Oct-4/Sox2/c-Myc were significantly upregulated, and multilineage differentiation in DPCs with XPC overexpression was enhanced after transfection. Conclusions. XPC plays an essential role in the modulation of pluripotency and multilineage differentiation of DPCs through regulation of Oct-4/Sox2/c-Myc.

Oxidation of phenyl PtII complexes K[(dpms)PtIIPh2], 1, (dpms)PtIIPh(MeOH), 2, (dpms)PtIIPh(OH2), 3, and methyl PtII complex (dpms)PtIIMe(NH2Ph), 6, with O2 in aqueous or methanol solutions under ambient conditions leads to corresponding (dpms)PtIVR(X)OH complexes (R = X = Ph, 7; R = Ph, X = OH, 8; R = Ph, X = OMe, 9; R = Me, X = NHPh; 11; dpms = di(2-pyridyl)methanesulfonate). Complexes 7–9 could be isolated in high yield. Complex 11 as well as its phenyl analogue (dpms)PtIVPh(NHPh)OH, 10 can be prepared in high yield by oxidation of corresponding (dpms)PtIIR(NH2Ph) with H2O2 in methanol. Phenyl PtII complexes (dpms)PtIIPh(HX) derived from HX = aniline and DMSO, 4 and 5, respectively, are inert toward O2. The rate of oxidation of 1–5 with O2 decreases in the order 1 > 3 ~ 2 » 4, and 5 is unreactive. Methyl analogues are significantly more reactive compared with their phenyl counterparts. Proposed mechanism of oxidation with O2 includes formation of anionic species (dpms)PtIIR(X)– responsible for reaction with dioxygen. A hexanucleotide repeat expansion GGGGCC (G4C2) within chromosome 9 open reading frame 72 (C9orf72) is the most common genetic cause of amyotrophic lateral sclerosis and frontotemporal dementia (C9-ALS/FTD). This seminal realization has rapidly focused our attention to the non-canonical translation (RAN translation) of the repeat expansion, which yields dipeptide-repeat protein products (DPRs). The mechanisms by which DPRs might contribute to C9-ALS/FTD are widely studied. Arginine-rich DPRs (R-DPRs) are the most toxic of the five different DPRs produced in neurons, but how do R-DPRs promote C9-ALS/FTD pathogenesis? Proteomic analyses have uncovered potential pathways to explore. For example, the vast majority of the R-DPR interactome is comprised of disease-linked RNA-binding proteins (RBPs) with low-complexity domains (LCDs), strongly suggesting a link between R-DPRs and aberrations in liquid–liquid phase separation (LLPS). In this review, we showcase several potential mechanisms by which R-DPRs disrupt various phase-separated compartments to elicit deleterious neurodegeneration. We also discuss potential therapeutic strategies to counter R-DPR toxicity in C9-ALS/FTD.

Sox18 is a developmental gene that encodes transcription factors. It has been indicated as be a key gene affecting the growth and development of hair follicles, in which dermal papilla cells (DPCs) have been demonstrated to play an important role through their ability to induce the formation of hair follicles. Pre-laboratory studies have found that Sox18 is differentially expressed in the dermal papilla cells of different pattern types of Hu sheep. We speculated that Sox18 plays an important role in the dermal papilla cells of Hu sheep. In our study, we analyzed the effect of Sox18 on the induction ability of DPCs in order to elucidate the function and molecular mechanism of Sox18 in the DPCs of Hu sheep. We first identified the expression of Sox18 in the DPCs of Hu sheep by immunofluorescence staining. We then used alkaline phosphatase staining, cell morphology observations and RT-PCR to detect the effect of Sox18 on the induction of DPCs after overexpression of or interference with Sox18. We also used RT-PCR, WB and immunofluorescence staining to detect the effect of Sox18 on the Wnt/β-catenin signal pathway in DPCs. We found that Sox18 was specifically expressed in the DPCs of Hu sheep, and that Sox18 could enhance the alkaline phosphatase activity in the DPCs of Hu sheep and accelerate cell agglutination. The results of RT-PCR revealed that Sox18 promoted the mRNA expression of Versican, HHIP and FGFRI, and inhibited the mRNA expression of BMP4 and WIF1. Further studies showed that Sox18 promoted the expression of β-catenin and activated the Wnt/β-catenin signal pathway in DPCs. When the Wnt/β-catenin signal pathway of DPCs was activated, the induction ability of DPCs was enhanced. Overall, we believe that Sox18 could enhance the induction ability of DPCs in Hu sheep and regulate the induction ability of DPCs through the Wnt/β-catenin signal pathway.

Background: Skin is considered a reflection of a person's appearance, so it is normal if many people try to take care of their skin, especially facial skin, and retard the aging process. Dermoscopy is a quick, non-invasive technique that allows physicians to observe the skin. Different ethnic groups may show different signs of aging on the face. Medan is a multiethnic city and thus might show facial skin aging differently. This study aimed to evaluate facial skin aging through the Dermoscopic Photoaging Scale (DPAS) and assess the relationship between DPAS, Glogau and Fitzpatrick scales among various ethnic groups in Medan, Indonesia. Subjects and Methods: This cross-sectional study enrolled 155 subjects from 10 ethnicities in Medan. The examination of facial aging is based on DPAS using 11 criteria, the Glogau scale, and the Fitzpatrick skin type. Results: Malay and Karonese got the highest DPAS score. There was a significant difference between DPAS, Glogau, and Fitzpatrick in age (p = <0.001,<0.001 and 0.032), and Fitzpatrick scores in ethnicity (p <0.001). There was a strong and significant correlation between DPAS and Glogau scores (p < 0.001, r = 0.748), while DPAS and Fitzpatrick's scores showed a moderate and significant correlation (p < 0.001, r = 0.288). Conclusion: The highest DPAS scores were found in the Malay and Karonese ethnic groups. There were significant differences between DPAS, Glogau, and Fitzpatrick in age and Fitzpatrick scores in ethnicity. There was a strong and significant correlation between DPAS scores and the Glogau scale and a moderate and significant correlation between DPAS and Fitzpatrick scores in various ethnic groups.

A renewed interest has appeared in citizen co-production of public services due to financial pressure on governments. While social media are considered an important facilitator, many digital participatory platforms (DPPs) have been developed to facilitate co-production between citizens and governments in the context of urban development. Previous studies have delivered a fragmented overview of DPPs in a few socio-spatial contexts and failed to take stock of the rise of DPPs. This article aims to provide a more comprehensive picture of the availability and functionalities of DPPs. Through a systematic review, 113 active DPPs have been identified, analysed, and classified within a citizen-government relationship typology. Almost a quarter of these DPPs demonstrate a realistic potential for online and offline co-production between governments and citizens. The article critically analyses the characteristics of these DPPs and explores their real-world applications in urban development. The article concludes with directions for further research.

Dihydropyranocoumarins (DPCs) were isolated from Peucedanum japonicum Thunb as anti-obesity compounds in 3T3-L1 adipocytes assay; however, it is uncertain whether DPC exerts anti-obesity activity in vivo. Therefore, this study evaluated the oral intake of pure DPCs in mice fed a high-fat diet, and also attempted to enhance its activity by nanoparticulation. Increases in body weight gain and fat accumulation in white adipose tissues were significantly suppressed by the dietary intake of DPCs (1.943 mg/mouse/day). DPCs intake also significantly decreased the mean size of adipocytes and upregulated mRNA levels of thermogenesis-related genes. Nanoparticulation of DPCs with polylactic-co-glycolic acid (PLGA) dramatically increased its activity almost 100-fold over that of a non-nanoparticulated form. Thus, our findings clearly demonstrated the anti-obesity activity of DPCs in vivo and suggested that PLGA nanoparticle encapsulation was useful to enhance the anti-obesity activity of DPCs with the aim to develop natural and safe anti-obesity agents.

Considering the problem of health condition prognostics of complex equipment, a discrete input process neural networks (DPNN) model based on process neural networks (PNN) is proposed in this paper. DPNN utilizes vector inputs together with convolution operator to gain the capability of time and spatial aggregation operation, which is implemented with continuous function inputs and integral operator by PNN. Different from PNN, DPNN can use discrete samples as inputs directly, thus can avoid precision loss during procedures of data fitting and function expanding required by PNN. The application of DPNN to health condition prognostics of complex equipment is described through the prediction of the future health state of the civil aircraft engines, where the short-term and long-term predictions of the health condition represented by the exhausted gas temperature time series are conducted. Moreover, the performance of DPNN is compared with common artificial neural networks (NN) and PNN. The results show that DPNN has satisfied performance for health condition prognostics of civil aircraft engines, and DPNN performs better than both NN and PNN, which prove that DPNN is suitable for health condition prognostics of complex equipment.

Background: Arrival of 100s of new drugs into the market every year makes the rational use of drugs a challenge to the practitioners. The product promotion by pharmaceutical companies disseminates ambiguous drug information through medical representatives using drug promotion literatures (DPLs) which influence the physicians’ prescribing pattern. Objective of the study was to evaluate DPLs for accuracy, consistency, and validity using WHO criteria for ethical medicinal drug promotion.Methods: DPLs collected from different OPDs were analyzed and evaluated as per WHO criteria. References of DPLs in support of the claims were critically analyzed for their retrievability and validity.Results: 50 DPLs of 76 drugs were collected and analyzed. Of which 49 were FDCs and 27 single drug formulations. None of the DPLs fulfilled all the WHO criteria. Only 26% (13) fulfilled a maximum of 8 WHO criteria. Out of 88 references given in support of claims, 17% (15) of the references were irretrievable, 62 were from journals, 4 from textbooks and 7 from website. Almost all the DPLs had pictures of which only 50% (49) were relevant.Conclusions: Information provided is incomplete and biased. Hence, health care professionals must evaluate DPLs critically before considering the same for prescribing.

Curcumin is considered a potential anti-asthmatic agent owing to its anti-inflammatory properties. The objective of the present study was to prepare curcumin-containing poly(lactic-co-glycolic acid)-based microscale discoidal polymeric particles (Cur-PLGA-DPPs) and evaluate their anti-asthmatic properties using a murine asthma model. Cur-PLGA-DPPs were prepared using a top-down fabrication method. The prepared Cur-PLGA-DPPs had a mean particle size of 2.5 ± 0.4 μm and a zeta potential value of −34.6 ± 4.8 mV. Ex vivo biodistribution results showed that the Cur-PLGA-DPPs mainly accumulated in the lungs and liver after intravenous injection. Treatment with Cur-PLGA-DPPs effectively suppressed the infiltration of inflammatory cells in bronchoalveolar lavage fluid, and reduced bronchial wall thickening and goblet-cell hyperplasia compared to those in the phosphate-buffered-saline-treated control group. No significant changes in hematology and blood biochemistry parameters were observed after treatment with Cur-PLGA-DPPs. At equal curcumin concentrations, treatment with Cur-PLGA-DPPs exhibited better therapeutic efficacy than treatment with free curcumin. Our results suggest that the microscale Cur-PLGA-DPPs can be potentially used as a lung-targeted asthma therapy.

Hyperglycemia, oxidative stress, and inflammation play key roles in the onset and development of diabetic complications such as diabetic nephropathy (DN). Diphenyl diselenide (DPDS) is a stable and simple organic selenium compound with anti-hyperglycemic, anti-inflammatory, and anti-oxidative activities. Nevertheless, in vitro, the role and molecular mechanism of DPDS on DN remains unknown. Therefore, we investigated the effects of DPDS on tert-butyl hydrogen peroxide (t-BHP)-induced oxidative stress and lipopolysaccharide (LPS)-induced inflammation in rat glomerular mesangial (HBZY-1) cells and explored the underlying mechanisms. DPDS attenuated t-BHP-induced cytotoxicity, concurrent with decreased intracellular ROS and MDA contents and increased SOD activity and GSH content. Moreover, DPDS augmented the protein and mRNA expression of Nrf2, HO-1, NQO1, and GCLC in t-BHP-stimulated HBZY-1 cells. In addition, DPDS suppressed LPS-induced elevations of intracellular content and mRNA expression of interleukin (IL)-6, IL-1β and TNF-α. Furthermore, LPS-induced NFκB activation and high phosphorylation of JNK and ERK1/2 were markedly suppressed by DPDS in HBZY-1 cells. In summary, these data demonstrated that DPDS improves t-BHP-induced oxidative stress by activating the Nrf2/Keap1 pathway, and also improves LPS-induced inflammation via inhibition of the NFκB/MAPK pathways in HBZY-1 cells, suggesting that DPDS has the potential to be developed as a candidate for the prevention and treatment of DN.

Introduction: Several dry-powder inhalers (DPIs) contain lactose which may be contaminated with milk proteins. Confusion exists pertaining to DPI use in patients with cow’s milk protein allergy (CMPA). Methods: A computerized survey sent via e-mail to pediatric pulmonologists and allergologists. Results: A total of 77 out of 232 (33.2%) doctors replied, of whom 80.5% were pediatric pulmonologists. A total of 69 of 77 (89.6%) were specialists, 37.6% with more than 15 years of experience. The most commonly used DPIs were formoterol + budesonide and vilanterol + fluticasone. A total of 62 out of 77 (80.5%) responders knew these DPIs contained lactose. A total of 35 out of 77 (45.5%) doctors who replied did not know that DPI leaflets list CMPA as a contra-indication to DPI administration. Of these, 4 (11.4%) stated that they would instruct patients with CMPA to stop DPIs, and 7 (20%) would avoid recommending DPIs. A total of 42 out of 77 (54.5%) responders were aware of this warning, yet 13 of these 42 (30.9%) continued to recommend lactose-containing DPIs without hesitation and 18 of these 42 (42.8%) responders prescribed DPIs but considered allergy severity. Conclusions: Almost half of certified, experienced pediatric pulmonologists and allergologists were unaware of the warning to administer DPIs to patients with CMPA. Most doctors who do know of this warning still continue to prescribe these DPIs.

Deproteinization of natural rubber was achieved in the latex stage. The structure of deproteinized natural rubber (DPNR) was characterized by fourier transform infrared spectroscopy (FTIR). The thermo degradation of DPNR was studied by thermogravimetry analysis (TG) under air atmosphere and nitrogen atmosphere. The kinetic parameters apparent activation energies (Ea) of the thermal decomposition reaction been calculated from the TG curves using the method described by Broido. And the results were compared with the thermo degradation of natural rubber (NR) under the same conditions. The effect of proteins in natural rubber latex on thermal/ thermo-oxidative stability of NR was discussed. The results show that: the absorptions of the proteins in DPNR at 1546 ㎝-1, compared to NR, become significantly weaker, nearly disappear, which indicates most of proteins has been removed from NR. The thermo degradation of DPNR in nitrogen atmosphere is a one-step reaction. The initial degradation temperature (T0) 、the maximum degradation temperature(Tp) and the final degradation temperature(Tf)as well as the Ea of DPNR are higher than those of NR, which indicates that DPNR represents a better thermal stability than NR under nitrogen atmosphere. Thermo-oxidative degradation of DPNR and NR are two-step reaction. The characteristic temperatures (T0, Tp and Tf) of DPNR are lower than those of NR. The Ea during the First Step of Thermooxidative Degradation of DPNR are also lower than those of NR. These results prove that the thermo-oxidative stability of DPNR is worse than that of NR. Protein is the key role to the thermal stability of natural rubber.

ABSTRACT DNA-protein cross-links (DPCs) are unique among DNA lesions in their unusually bulky nature. We have recently shown that nucleotide excision repair (NER) and RecBCD-dependent homologous recombination (HR) collaboratively alleviate the lethal effect of DPCs in Escherichia coli. In this study, to gain further insight into the damage-processing mechanism for DPCs, we assessed the sensitivities of a panel of repair-deficient E. coli mutants to DPC-inducing agents, including formaldehyde (FA) and 5-azacytidine (azaC). We show here that the damage tolerance mechanism involving HR and subsequent replication restart (RR) provides the most effective means of cell survival against DPCs. Translesion synthesis does not serve as an alternative damage tolerance mechanism for DPCs in cell survival. Elimination of DPCs from the genome relies primarily on NER, which provides a second and moderately effective means of cell survival against DPCs. Interestingly, Cho rather than UvrC seems to be an effective nuclease for the NER of DPCs. Together with the genes responsible for HR, RR, and NER, the mutation of genes involved in several aspects of DNA repair and transactions, such as recQ, xth nfo, dksA, and topA, rendered cells slightly but significantly sensitive to FA but not azaC, possibly reflecting the complexity of DPCs or cryptic lesions induced by FA. UvrD may have an additional role outside NER, since the uvrD mutation conferred a slight azaC sensitivity on cells. Finally, DNA glycosylases mitigate azaC toxicity, independently of the repair of DPCs, presumably by removing 5-azacytosine or its degradation product from the chromosome.

ABSTRACT Fresh NR was adopted to prepare rubber with a nanomatrix structure. The nanomatrix structure was formed by graft-copolymerization of styrene onto deproteinized fresh NR (fresh DPNR) in the latex stage using tert-butyl hydroperoxide/tetraethylenepentamine as an initiator. Graft-copolymer consisting of fresh DPNR and polystyrene (fresh DPNR-graft-PS) was characterized by 1H-NMR spectroscopy and transmission electron microscopy. Under a suitable condition of the graft-copolymerization, conversion and grafting efficiency of styrene was 98 and 80 mol%, respectively. Morphology of fresh DPNR-graft-PS was observed by transmission electron microscopy. The rubber particles of about 1 μm in diameter were found to be dispersed in polystyrene matrix of 10–30 nm in thickness. Tensile properties of fresh DPNR were dramatically improved by forming the nanomatrix structure, compared with DPNR.

Introduction. Dental pulp cells (DPCs) are promising cell source for dental tissue regeneration. Recently, small molecules which optimize microenvironment or activate the reprogramming network provide a new way to enhance the pluripotency. Two promising bioflavonoids luteolin and apigenin were reported to enhance reprogramming efficiency in mouse embryonic fibroblast (MEF). However, their effect and underlying mechanism in cell fate determination of human DPCs remain unclear.Methods. To elucidate the effect of luteolin and apigenin on the cell fate determination of DPCs, we explored the cell proliferation, cell cycle, senescence, apoptosis, expression of pluripotency markers Oct-4, Sox2, and c-Myc, and multilineage differentiation capability of DPCs with luteolin or apigenin treatment.Results. We demonstrated that luteolin and apigenin inhibited cell proliferation, arrested DPCs in G2/M and S phase, and upregulated PI value and apoptosis. Moreover, luteolin and apigenin increased telomerase activity, maintained DPCs in a presenescent state, and activated the expression of Oct-4, Sox2, and c-Myc at a dose- and time-dependent pattern in DPCs even at late passages, albeit repressed lineage-specific differentiation.Conclusions. Addition of luteolin and apigenin in the culture medium might provide an effective way to maintain DPCs in an undifferentiated stage and inhibit lineage-specific differentiation.

General Data Protection Regulation (GDPR) focuses on important elements of data ethics, including protecting people’s privacy, accountability and transparency. According to the GDPR, certain public institutions are obliged to appoint a Data Protection Officer (DPO). However, there is little publicly available data from national EU surveys on DPOs. This study aimed to examine the scope of work, type of work, and education of DPOs in institutions in Croatia. During 2020-2021, this cross-sectional study surveyed DPOs appointed in Croatia. The survey had 35 items. The questions referred to their appointment, work methods, number and type of cases handled by DPOs, the sources of information they use, their experience and education, level of work independence, contacts with ethics committees, problems experienced, knowledge, suggestions for improvement of their work, changes caused by the GDPR, and sociodemographic information. Out of 5671 invited DPOs, 732 (13%) participated in the study. The majority (91%) indicated that they could perform their job independently; they did not have prior experience in data protection before being appointed as DPOs (54%) and that they need additional education in data protection (82%). Most DPOs indicated that they had none or minimal prior experience in data protection when they were appointed as DPO, that they would benefit from further education on data protection, and exhibited insufficient knowledge on basic concepts of personal data protection. Requirements for DPO appointments should be clarified; mandatory education and certification of DPOs could be introduced and DPOs encouraged to engage in continuous education.

Background: The study was aimed to critically analyse Drug Promotional Literatures (DPLs) using WHO guidelines. This would help to create awareness about DPLs amongst healthcare providers thus encouraging the improvement of healthcare system.Methods: This cross sectional observational study was carried out at Department of Pharmacology, Medical College Baroda. DPLs were collected & critically analysed for consistency, accuracy, validity of the provided information as per WHO guidelines.Results: Out of total 616 DPLs collected, 371 satisfied the inclusion criteria. None of the DPL was fulfilling all criteria according to WHO guidelines. Most of DPLs were having information regarding; generic name / INN (98.39%), brand name (100%), amount of active ingredient per dosage (94.07%), approved therapeutic uses (84.91%), dosage form (91.37%) and name & address of manufacturers (91.91%). Of all the DPLs, information provided for safety parameters like; name of active ingredient known to cause problem (11.59%), dosage regimen (32.88%), side effects & drug reaction (14.56%), major drug interactions (14.02%) and precautions, contraindications and warning (14.29%) seemed to be grossly neglected. Total of 431 claims were evaluated, of which the most common type of claim was efficacy (55.45%). Relevant references to claims were present in (48.74%) DPLs. Total 203 references were evaluated from 371 DPLs, of which maximum reference were from journal article (74.38%).Conclusions: From this study, it was concluded that pharmaceutical companies didn’t follow the WHO guidelines for ethical drug promotion, thus failing to fulfil the rational promotion of drugs. Given the present findings physicians should be cautious about drawing conclusions regarding medicine based on DPLs provided by pharmaceutical companies.

The interaction between the dermal papilla cells (DPCs) and epidermal hair matrix cells (HMCs) of hair follicles (HFs) is crucial for the growth and development of HFs, but the molecular mechanism is complex and remains unclear. MicroRNAs (miRNAs) are the key signaling molecules for cellular communication. In this study, the DPCs and HMCs of yak were isolated and cultured, and the differentially expressed mRNA and miRNA were characterized to analyze the molecular basis of the interaction between DPCs and HMCs during hair follicle (HF) development in yak. The mRNA differential expression and functional enrichment analysis revealed that there were significant differences between DPCs and HMCs, and they showed the molecular functional characteristics of dermal cells and epidermal cells, respectively. Multiple KEGG pathways related to HF development were enriched in the highly expressed genes in DPCs, while the pathways associated with microbiota and immunity were significantly enriched in the highly expressed genes in HMCs. By combining analysis with our previous 10× genomics single-cell transcriptome data, 39 marker genes of DPCs of yak were identified. A total of 123 relatively specifically expressed miRNAs were screened; among these, the miRNAs associated with HF development such as miR-143, miR-214, miR-125b, miR-31, and miR-200 were presented. In conclusion, the large changes in yak DPCs and HMCs for both mRNA and miRNA expression were revealed, and numerous specifically expressed mRNAs and miRNAs in DPCs or HMCs were identified, which may contribute to the interaction and cellular communication between DPCs and HMCs during HF development in yak.

Background: Drug promotional literatures (DPLs) are one of the well-known promotional activities of pharmaceutical industries which are sometimes inaccurate as well as of poor educational value. Angiotensin receptor blockers (ARBs) are one of the most commonly used antihypertensives. Therefore, this study was done to estimate the accuracy of DPLs on ARBs as per the World Health Organization (WHO) criteria. Aims and Objectives: The aims of this study were to estimate the accuracy of DPLs on ARBs as per the WHO and to estimate the DPLs for types of claims and appropriateness of claims. Materials and Methods: A cross-sectional observational study was carried out for 1 month. All the required information of selected DPLs on ARBs were recorded in a pro forma and were evaluated according to the WHO criteria. Results: In this study, a total of 20 (twenty) DPLs were evaluated only on ARBs. It was observed that none of the DPLs fulfilled all the WHO criteria. In this study, some DPLs made multiple claims, as much as five per DPL. Claims were, further, analyzed and divided into appropriate and inappropriate. We have observed that 65.96% claims were appropriate and 34.04% claims were inappropriate. Conclusion: This type of study can contribute to make prescribing practices rational as promotional activities influence the prescribing behavior of the health-care provider.

RESUMO Estudos indicam que fatores relacionados ao trabalho podem levar ao adoecimento de trabalhadores de unidades de terapia intensiva, mas estudos sobre distúrbios psíquicos menores (DPMs) em fisioterapeutas intensivistas ainda são escassos. Este estudo visa estimar a prevalência e os fatores associados a DPMs em fisioterapeutas intensivistas de uma grande cidade do estado da Bahia. Trata-se de um estudo epidemiológico de corte transversal que analisou uma população de 60 fisioterapeutas trabalhadores da terapia intensiva na cidade de Feira de Santana, Bahia. Um questionário autoaplicável avaliou dados sociodemográficos, características do trabalho, aspectos psicossociais do trabalho e presença de DPMs. A prevalência de DPMs encontrada foi de 41,7% e verificou-se associação entre as variáveis sociodemográficas, características do trabalho, hábitos de vida, aspectos psicossociais do trabalho e DPM. Observou-se elevada prevalência e uma diversidade de fatores associados aos DPMs entre os fisioterapeutas estudados. Os resultados apontam a necessidade de novos estudos epidemiológicos que possam identificar com mais precisão os fatores associados a DPMs nesses trabalhadores.

The main biologically active components of plants belonging to the genus Allium, responsible for their biological activities, including anti-inflammatory, antioxidant and immunomodulatory, are organosulfur compounds. The aim of this study was to synthetize the mixture of dipropyl polysulfides (DPPS) and to test their biological activity in acute hepatitis. C57BL/6 mice were administered orally with DPPS 6 h before intravenous injection of Concanavalin A (ConA). Liver inflammation, necrosis and hepatocytes apoptosis were determined by histological analyses. Cytokines in liver tissue were determined by ELISA, expression of adhesive molecules and enzymes by RT PCR, while liver mononuclear cells were analyzed by flow cytometry. DPPS pretreatment significantly attenuated liver inflammation and injury, as evidenced by biochemical and histopathological observations. In DPPS-pretreated mice, messenger RNA levels of adhesion molecules and NADPH oxidase complex were significantly reduced, while the expression of SOD enzymes was enhanced. DPPS pretreatment decreased protein level of inflammatory cytokines and increased percentage of T regulatory cells in the livers of ConA mice. DPPS showed hepatoprotective effects in ConA-induced hepatitis, characterized by attenuation of inflammation and affection of Th17/Treg balance in favor of T regulatory cells and implicating potential therapeutic usage of DPPS mixture in inflammatory liver diseases.

Maintenance of trichogenecity of dermal papilla cells (DPCs) have been a problem during cell therapy for androgenic alopecia, as they lose their regenerative potential in in vitro culture. Various spheroid culture techniques are used to increase and maintain trichogenecity of these cells. However, there are some critical drawbacks in these methods. Applying a hydrocell plate for sphere formation or hanging drop methods by hand would be difficult to control the size and cell density inside it. It would be difficult to commercialize or mass production for clinical therapy. In aim to address and overcome these drawbacks, we have introduced alginate sphere. The alginate sphere of DPCs were prepared by electrospinning at different voltages to control the size of sphere. Then the obtained alginate spheres were evaluated for cellular dynamics and density of DPCs under different conditions. In this study, we found that DPCs do not proliferate in alginate sphere. However, the number of DPCs were maintained and found to be in dormant state. Further, the dormant DPCs in the alginate sphere have upregulated DPC signature genes (SOX2, ALPL, WIF1, Noggin, BMP4 and VCAN) and proliferative capacity. Thus, we speculate that alginate sphere environment maintains the dormancy of DPCs with increased trichogenecity.
