Cell mechanics is involved in several biological processes such as adhesion, migration, or differentiation. Evaluating the mechanical properties of cellular elements is essential to understand their function in these processes and allows anticipating disruptions in case of pathological alterations. However, the techniques traditionally used to mechanically characterize cells do not allow eliminating interactions between different cellular elements during measurements. Picosecond acoustics offer a new opportunity thanks to the generation and detection of acoustic waves whose frequencies can reach the terahertz. We have developed an experimental protocol to perform picosecond acoustic measurements on living cells immersed in a culture medium. Using an imaging chamber, designed for direct and inverted microscopy, and a biocompatible opto-acoustic transducer, we can perform non-invasive measurements of mechanical properties with micrometric spatial resolution. We then studied the impact of fixation on the internal structure of the nucleus, using time-resolved Brillouin scattering. This study shows that the acoustic velocity in the nucleus increases with fixation. These changes are not homogeneous but mainly affect the elements of the nucleoplasm presenting the lowest Brillouin frequency. We also demonstrated the importance of maintaining hydration of fixed cells to avoid irreversible modifications caused by dehydration. Indeed, rehydrated cells do not recover the same mechanical properties as they had before dehydration. Finally, we studied, thanks to a statistical analysis on a large number of mesenchymal stem cells and osteoblasts, how the internal structure of the nucleus evolves with differentiation. By comparing cells incubated for 24 hours with those incubated for 14 days in an osteogenic medium, we observed that differentiated cells had a higher chromatin concentration than undifferentiated cells. This concentration results in an increase in Brillouin frequency, which is attributed to a change in nuclear rigidity

This manuscript presents the first steps of a new ultracold atoms experiment using strontium 84. The aim of this experiment is to study the relaxation dynamics of quantum gases initially prepared in an out-of-equilibrium state. This experiment will include a quantum gas microscope, allowing us to measure spatial correlation functions in two-dimensionnal systems. The current state of the construction allows us to generate both magneto-optical trap of strontium: along its wide transition at 461 nm and its narrow transition at 689 nm. Concurrently with the experimental setup, we carried out works on a reconstruction algorithm required for the future data processing of the microscope images. This manuscript details experimental aspects, justifying their choices, and presents the current state of work on the reconstruction algorithm. There are still steps to complete the experimental setup: add a chamber where we will make the measurements to the vaccuum system, set up the quantum gaz microscope and all the required optics to transport the atomic clouds between two vaccuum chambers, to reach Bose-Einstein condensation and to confine the atoms in two-dimensionnal optical traps

Este trabajo de tesis presenta un estudio experimental y teórico de la dinámica de fonones acústicos en multicapas a la escala nanométrica usando técnicas de acústica de picosegundos y dispersión Raman. Estudiamos las características de transmisión,generación y detección de fonones acústicos de ultra-alta frecuencia (en el rango GHz-THz) de superredes y nanocavidades de materiales semiconductores u óxidos dieléctricos. Diseñamos, optimizamos y caracterizamos dispositivos fonónicos aperiódicos y sistemas de nanocavidades acústicas acopladas. Finalmente utilizamos los efectos de confinamiento óptico para amplificar los procesos de generación y de detección de fonones acústicos coherentes y para modificar las reglas de selección. Por otra parte, desarrollamos modelos que nos permitieron calcular las respuestas ópticas y acústicas de estructuras excitadas por pulsos láser ultracortos, y evaluar la sección eficaz Raman en multicapas
This thesis presents an experimental and theoretical study of the acoustic phonon dynamics at the nanometric scale, by picosecond acoustic techniques and Raman scattering. We have studied the acoustic phonon (with frequencies in the GHz-THz range) transmission, generation and detection in superlattices and nanocavities made of semiconductor or dielectric oxyde materials. We have designed, optimized and characterized aperiodic phonon devices and coupled acoustic nanocavity systems. Finally, we have used the optical confinenment effects to enhance the coherent acoustic phonon generation and detection processes and to change the selection rules. In addition, we have developed models to computen the acoustic and optical responses of structures excited by ultrashort laser pulses, and to evaluate the Raman scattering cross section in nanometric multilayers

A l'intérieur du GaN, deux catégories de site d'incorporation d'ions de terre rare peuvent être distinguées, à savoir les ions de terre rare "isolés" (c'est-à-dire ne contenant aucun défaut dans leur voisinage) et les complexes associant un ion de terre rare avec un piège, celui-ci pouvant provenir du dopage lui-même ou d'un défaut cristallin. L'excitation non-résonante est réservée au second type de centres et a lieu par transfert d'énergie lors de la recombinaison non radiative d'un exciton lié sur un piège proche de la terre rare.
La comparaison d'échantillons de GaN dopés in situ et par implantation ionique par l'ion Eu3+ montre que ces films présentent chacun deux types de complexes "Eu3+-piège" dont l'un est commun à tous les échantillons. Les différences d'efficacité d'excitation respectives des deux complexes s'expliquent par la proximité du piège correspondant.
Grâce à des expériences pompe-sonde combinant des lasers impulsionnel et continu, le chemin d'excitation des ions de terre rare est confirmé expérimentalement et deux des mécanismes d'extinction de leur luminescence (la photo-ionisation de pièges et l'effet Auger avec des porteurs libres) sont étudiés.
L'excitation électrique du GaN:Er3+ est également présentée. Les études en fonction de la température, du courant parcourant l'échantillon ou de la tension de polarisation mettent en évidence l'excitation par impact des ions de terre rare.
