Abstract A series of Cas9 variants have been developed to improve the editing fidelity or targeting range of CRISPR–Cas9. Here, we employ a high-throughput sequencing approach primer-extension-mediated sequencing to analyze the editing efficiency, specificity and protospacer adjacent motif (PAM) compatibility of a dozen of SpCas9 variants at multiple target sites in depth, and our findings validate the high fidelity or broad editing range of these SpCas9 variants. With regard to the PAM-flexible SpCas9 variants, we detect significantly increased levels of off-target activity and propose a trade-off between targeting range and editing specificity for them, especially for the near-PAM-less SpRY. Moreover, we use a deep learning model to verify the consistency and predictability of SpRY off-target sites. Furthermore, we combine high-fidelity SpCas9 variants with SpRY to generate three new SpCas9 variants with both high fidelity and broad editing range. Finally, we also find that the existing SpCas9 variants are not effective in suppressing genome instability elicited by CRISPR–Cas9 editing, raising an urgent issue to be addressed.

AbstractIn this paper, we propose to work in the 2.5D space of the scene to facilitate composition of new spherical panoramas. For adding depths to spherical panoramas, we extend an existing method that was designed to estimate relative depths from a single perspective image through user interaction. We analyze the difficulties to interactively provide such depth information for spherical panoramas, through three different types of presentation. Then, we propose a set of basic tools to interactively manage the relative depths of the panoramas in order to obtain a composition in a very simple way. We conclude that the relative depths obtained by the extended depth estimation method are enough for the purpose of compositing new photorealistic panoramas through a few elementary editing tools.

Beyond time frame editing in video data, object level video editing is a challenging task; such as object removal in a video or viewpoint changes. These tasks involve dynamic object segmentation, novel view video synthesis and background inpainting. Background inpainting is a task of the reconstruction of unseen regions presented by object removal or viewpoint change. In this paper, we propose a video editing method including foreground object removal background inpainting and novel view video synthesis under challenging conditions such as complex visual pattern, occlusion, overlaid clutter and variation of depth in a moving camera. Our proposed method calculates a weighted confidence score on the basis of normalized difference between observed depth and predicted distance in 3D space. A set of potential points from epipolar lines from neighbor frames are collected, refined, and weighted to select a few number of highly qualified observations to fill the desired region of interest area in the current frame from video. Based on the background inpainting method, novel view video synthesis is conducted with arbitrary viewpoint. Our method is evaluated with both a public dataset and our own video clips and compared with multiple state of the art methods showing a superior performance.

Purpose The purpose of this paper is to reveal the co-editing mechanism aiming at content creation, and an entry of online encyclopedia is taken as a case, for the purpose of promoting and enhancing the development of wiki-based digital humanities projects (WDHPs), specifically, the projects that focus on gathering contextual information in the culture heritage domain. Design/methodology/approach An exploratory study was conducted by three steps. A representative entry’s editorial records were reorganized to obtain a data set of discussion statements (n=608), based on which linked-structures were built, and PageRank algorithm was used to analyze the co-editing process. Skewness statistic was applied to measure the consensus of co-editing, and consensus evolution over time was explored. Linear or curve fitting was performed to analyze the correlation between consensus evolution and its influential factors. Findings In WDHPs, co-editing activity of content creation can be considered as a large-scale group discussion, consensus can evaluate the efficiency of co-editing, which evolves with time and is influenced by the number of statements, breadth and depth of argumentation structure. Taking “Mogao Grottoes” as an example, group discussions around 15 key issues dominate the content creating process, consensus is on a rise with time, finally reaches a relatively high level, and consensus evolution is more influenced by breadth than by depth of argumentation structure, which indicates that co-editing efficiency of “Mogao Grottoes” is fine and more argumentation in a depth manner should be guided. Practical implications For researchers of WDHPs, it is beneficial to apply online encyclopedia platform combining with consensus analysis to develop WDHPs. For designers of WDHPs, the elements related to argumentation structure can be absorbed into the design to promote co-editing in an effective manner. For DH researchers, the analytic procedure can be beneficial of revealing the interest of contributors in a specific DH field. Originality/value This research is novel in comprehensively understanding co-editing mechanism of content creation in WDHPs, resulting in a three-step analytic procedure of presenting co-editing process, evaluating and improving co-editing efficiency.

Adenine base editors (ABEs) enable precise and efficient conversion of target A•T base pairs to G•C base pairs in genomic DNA with a minimum of by-products. While ABEs have been reported to exhibit minimal off-target DNA editing, off-target editing of cellular RNA by ABEs has not been examined in depth. Here, we demonstrate that a current ABE generates low but detectable levels of widespread adenosine-to-inosine editing in cellular RNAs. Using structure-guided principles to design mutations in both deaminase domains, we developed new ABE variants that retain their ability to edit DNA efficiently but show greatly reduced RNA editing activity, as well as lower off-target DNA editing activity and reduced indel by-product formation, in three mammalian cell lines. By decoupling DNA and RNA editing activities, these ABE variants increase the precision of adenine base editing by minimizing both RNA and DNA off-target editing activity.

This article provides an in-depth examination of the medical rationale driving the acceptance of genome editing human embryos in two contexts: to cure disease and treat infertility. It describes complexities in genomics and outlines risks currently associated with genome editing, asserting the available evidence fails to demonstrate genome editing constitutes a curative therapy.

Background/Aims: Aberrant RNA editing, mediated by adenosine deaminases acting on RNA (ADAR), serves as a post-transcriptional event participating in tumorigenesis and prognosis. However, the RNA editing profiles during HCC progression and their clinical correlations remain unclear. Methods: Multiple tissue samples were collected from an advanced HCC patient. RNA-seq was performed to obtain the RNA editing profiles for each sample. Two RNA editing sites from CDK13 were further validated in 60 HCC patients; and their potential regulatory mechanisms were investigated. Results: In-depth analysis of the RNA-seq data revealed a significant number of editing sites (632-816) in coding regions for each tissue sample, showing branched evolution during tumorigenesis and metastasis. Two editing sites (Q103R and K96R) in CDK13 showed significant over-editing in tumor, and these phenomenon were validated in 60 HCC patients. Furthermore, the clinicopathological analysis revealed that these CDK13 over-editing sites were positively associated with TNM, PVTT and poor prognosis. In addition, the editing level of these sites were significantly correlated with the expression of ADAR1. Loss of function assays further proved that these CDK13 over-editing sites were mediated by ADAR1 in HCC cells. Conclusions: CDK13 RNA over-editing sites mediated by ADAR1 may serve as novel cancer driver events in HCC progression.

APOBEC3 enzymes contribute significantly to DNA mutagenesis in cancer. These enzymes are also capable of converting C bases at specific positions of RNAs to U. However, the prevalence and significance of this C-to-U RNA editing in any cancer is currently unknown. We developed a bioinformatics workflow to determine RNA editing levels at known APOBEC3-mediated RNA editing sites using exome and mRNA sequencing data of 1040 breast cancer tumors. Although reliable editing determinations were limited due to sequencing depth, editing was observed in both tumor and adjacent normal tissues. For 440 sites (411 genes), editing was determinable for ≥5 tumors, with editing occurring in 0.6%–100% of tumors (mean 20%, SD 14%) at an average level of 0.6%–20% (mean 7%, SD 4%). Compared to tumors with low RNA editing, editing-high tumors had enriched expression of immune-related gene sets, and higher T cell and M1 macrophage infiltration, B and T cell receptor diversity, and immune cytolytic activity. Concordant with this, patients with increased RNA editing in tumors had better disease- and progression-free survivals (hazard ratio = 1.67–1.75, p < 0.05). Our study identifies that APOBEC3-mediated RNA editing occurs in breast cancer tumors and is positively associated with elevated immune activity and improved survival.

AbstractAirborne expendable bathythermographs (AXBTs) are air-launched, single-use temperature–depth probes that telemeter temperature observations as VHF-modulated frequencies. This study describes the AXBT Real-Time Editing System (ARES), which is composed of two components: the ARES Data Acquisition System, which receives telemetered temperature–depth profiles with no external hardware other than a VHF radio receiver, and the ARES Profile Editing System, which quality controls AXBT temperature–depth profiles. The ARES Data Acquisition System performs fast Fourier transforms on windowed segments of the demodulated signal transmitted from the AXBT. For each segment, temperature is determined from peak frequency and depth from elapsed time since profile start. Valid signals are distinguished from noise by comparing peak signal levels and signal-to-noise ratios to predetermined thresholds. When evaluated using 387 profiles, the ARES Data Acquisition System produced temperature–depth profiles nearly identical to those generated using a Sippican MK-21 processor, while reducing the amount of noise from VHF interference included in those profiles. The ARES Profile Editor applies a series of automated checks to identify and correct common profile discrepancies before displaying the profile on an editing interface that provides simple user controls to make additional corrections. When evaluated against 1177 tropical Atlantic and Pacific AXBT profiles, the ARES automated quality control system successfully corrected 87% of the profiles without any required manual intervention. Necessary future work includes improvements to the automated quality control algorithm and algorithm evaluation against a broader dataset of temperature–depth profiles from around the world across all seasons.

With the continuous development and application of gene editing technologies, they have become valuable tools to study the mechanism of spermatogenesis. Gene editing technologies have provided us with a better understanding of the functions and potential mechanisms of action of factors that regulate spermatogenesis. This review summarizes the applications of gene editing technologies, especially CRISPR/Cas9, in deepening our understanding of the function of spermatogenesis-related genes and disease treatment. The problems of gene editing technologies in the field of spermatogenesis research are also discussed.

Human genome editing could be used to improve human intelligence, appearance, and physical strength. The ethical issues posed by the technology are causing public concern. Secularism views efforts to find moral truth do not depend on supernatural authority. The existence of a secular minority in Indonesia is considered an anomaly in public discourse and is left behind in the morally critical policy-making deliberations on genome editing regulations. The research explored Indonesian secular doctor’s view of human genome editing. This qualitative research used a non-ideal critical realism approach through in-depth interviews and literature review. Secular doctors say that human genomic editing has no different weight from other artificial selection. After conducting a risk-benefit analysis through moral calculus, secular doctors generally have positions and attitudes that support the research and implementation of human genomic editing technology.

Epi-illumination microscopy is a useful tool for studying morphology, but its use is limited by a small depth of field. Using digital editing, we have enhanced the depth of field for epi-illumination micrographs obtained for a number of shoot apices. For each specimen, an epi-illumination microscope fitted with Leitz Ultropak dipping cones was used to capture a series of digital images at successive levels of focus. Adobe® Photoshop® was used to produce one image by assembling only the focused areas of the image series. This stacking method offers a better way to present epi-illumination micrographs and increases the microscope’s usefulness as an alternative for conventional scanning electron microscopy. The method could be applied to images collected by other microscopical techniques.

Genome editing is an emerging, new breeding technology with numerous potential applications in plant breeding. In Europe, genome editing is regarded, in legal terms, as a genetic modification technique, hence plants obtained using these methods fall under the legislation for genetically modified organisms (GMOs). Despite the opportunities that genome editing brings to the plant sector, it also poses challenges to the regulatory system. For example, the enforcement of labelling and traceability requirements for GM foods and feeds may be impossible for small genome edits that are indistinguishable from natural mutations. In order to discuss potential adaptations of EU legislation with stakeholders from the Dutch plant breeding sector, five different scenarios of future regulation of plants obtained by means of genome editing were elaborated. These scenarios were discussed in depth, along with the potential applications of genome editing in plant breeding, as well as challenges and opportunities. Stakeholders particularly indicated their preference for new, future-proof legislation in the long term, which will also include products of novel technologies. Finally, we discuss potential short-term amendments to current legislation, including the exemption of certain small mutations, that would make it align with regulation of genome edited plants in non-EU countries.

Germline predisposition to bone marrow failure (BMF) and myelodysplastic syndromes (MDS) cause substantial economic and psychosocial burden and early mortality in affected individuals. Due to the limitations and risks of hematopoietic stem cell transplantation, there is an urgent need for new and innovative treatment approaches. Conventional lentiviral-based gene therapy is not feasible for most BMF/MDS predisposing genes because their uncontrolled overexpression is detrimental and lentiviral vectors might cause insertional mutagenesis. However, new technologies that precisely modify the endogenous allele can overcome these limitations. Base editing (BE) using cytosine base editors (CBE) and adenine base editors (ABE) as well as prime editing (PE) are advanced gene editing techniques with great therapeutic promise. The goal of this project is to develop a comprehensive map of gene-editable mutations in the GATA2, SAMD9, and SAMD9L genes as a preclinical effort to facilitate gene editing trials. Germline heterozygous mutations in these key predisposing genes lead to highly penetrant disorders characterized by progressive immunodeficiency and BMF/MDS risk, with particularly high prevalence in children, as recently reported by us (Sahoo, Nature Medicine 2021) and others. For in depth analysis, we focused on the 5 most recurrent mutations in respective genes and devise strategies to correct mutations in cellular models. We first created a comprehensive database encompassing reported and unpublished pathogenic mutations (single nucleotide variants [SNVs] and <20bp insertion/deletions[indels]). In total, we annotated 506 cases with GATA2, 195 cases with SAMD9, and 187 cases with SAMD9L mutations. SNVs were detected in 83.2% (421/506), 94.4% (184/195), and 93.1% (174/187) patients with GATA2, SAMD9, SAMD9L mutations, respectively, while remaining cases had indels. Among all GATA2 cases, 59.9% (303/506) mutations are editable by ABE, 4.5% (23/506) by CBE, while remaining 35.6% (180/506) can be “rescued” by PE ( Fig. 1A). For SAMD9, 34.4% (67/195) cases were found to be amenable to editing by ABE, 30.3% (59/195) by CBE, and 35.4% (69/195) using PE. For SAMD9L, 47.1% (88/187) are editable by ABE, 20.3% (38/187) by CBE, and 32.6% (61/187) using PE. Having a well-annotated mutation list, we proceeded to identify the top 5 most common mutations in each gene ( Fig. 1B). Remarkably, all 5 mutations in both GATA2 and SAMD9 and 4 out of 5 top mutations in SAMD9L and amenable to BE. Next, we explored in depth the target editing window, protospacer adjacent motif (PAM) sites, and bystander editing effect (utilizing BE that recognize the canonical [NGG], and non-canonical PAM sequences [NG and near PAMless SpRY-NRN/NYN]. This stepwise approach resulted in successful identification of at least one potential guide RNA for each mutation with predicted low bystander editing. Ongoing work includes guide RNA validation, off-target analysis, and assessment of gene-editing efficiency using model cell lines. In conclusion, we have undertaken a systematic analysis of the gene editing landscape for frequent germline driver mutations in BMF/MDS. Our findings reveal promising editing strategies for the majority of prevalent mutations in key predisposing genes GATA2, SAMD9 and SAMD9L using CBE, ABE, or PE editors, and lay a groundwork to enable future clinical translation.

Integrating technology into agricultural systems has gained considerable traction, particularly over the last half century. Agricultural systems that incorporate the public’s concerns regarding farm animal welfare are more likely to be socially accepted in the long term, a key but often forgotten component of sustainability. Gene editing is a tool that has received considerable attention in the last five years, given its potential capacity to improve farm animal health, welfare, and production efficiency. This study aimed to explore the attitudes of Brazilian citizens regarding the applications of gene editing in cattle that generate offspring without horns; are more resistant to heat; and have increased muscle tissue. Using a mixed-methods approach, we surveyed participants via face-to-face, using in-depth interviews (Study 1) and an online questionnaire containing closed-ended questions (Study 2). Overall, the acceptability of gene editing was low and in cases where support was given it was highly dependent on the type and purpose of the application proposed. Using gene editing to improve muscle tissue growth was viewed as less acceptable compared to using gene editing to reduce heat stress or to produce hornless cattle. Support declined when the application was perceived to harm animal welfare, to be profit motivated or to reinforce the status quo of intensive livestock systems. The acceptability of gene editing was reduced when perceptions of risks and benefits were viewed as unevenly or unfairly distributed among consumers, corporations, different types of farmers, and the animals. Interviewees did not consider gene editing a “natural” process, citing dissenting reasons such as the high degree of human interference and the acceleration of natural processes. Our findings raised several issues that may need to be addressed for gene editing to comply with the social pillar of sustainable agriculture.

The creation of genetically modified horses is prohibited in horse racing as it falls under the banner of gene doping. In this study, we developed a test to detect gene editing based on amplicon sequencing using next-generation sequencing (NGS). We designed 1012 amplicons to target 52 genes (481 exons) and 147 single-nucleotide variants (SNVs). NGS analyses showed that 97.7% of the targeted exons were sequenced to sufficient coverage (depth > 50) for calling variants. The targets of artificial editing were defined as homozygous alternative (HomoALT) and compound heterozygous alternative (ALT1/ALT2) insertion/deletion (INDEL) mutations in this study. Four models of gene editing (three homoALT with 1-bp insertions, one REF/ALT with 77-bp deletion) were constructed by editing the myostatin gene in horse fibroblasts using CRISPR/Cas9. The edited cells and 101 samples from thoroughbred horses were screened using the developed test, which was capable of identifying the three homoALT cells containing 1-bp insertions. Furthermore, 147 SNVs were investigated for their utility in confirming biological parentage. Of these, 120 SNVs were amenable to consistent and accurate genotyping. Surrogate (nonbiological) dams were excluded by 9.8 SNVs on average, indicating that the 120 SNV could be used to detect foals that have been produced by somatic cloning or embryo transfer, two practices that are prohibited in thoroughbred racing and breeding. These results indicate that gene-editing tests that include variant calling and SNV genotyping are useful to identify genetically modified racehorses.

Depth adjustment aims to enhance the visual experience of stereoscopic 3D (S3D) images, which accompanied with improving visual comfort and depth perception. For a human expert, the depth adjustment procedure is a sequence of iterative decision making. The human expert iteratively adjusted the depth until he is satisfied with the both levels of visual comfort and the perceived depth. In this work, we present a novel deep reinforcement learning (DRL)-based approach for depth adjustment named VCA-RL (Visual Comfort Aware Reinforcement Learning) to explicitly model human sequential decision making in depth editing operations. We formulate the depth adjustment process as a Markov decision process where actions are defined as camera movement operations to control the distance between the left and right cameras. Our agent is trained based on the guidance of an objective visual comfort assessment metric to learn the optimal sequence of camera movement actions in terms of perceptual aspects in stereoscopic viewing. With extensive experiments and user studies, we show the effectiveness of our VCA-RL model on three different S3D databases.

Lactococcus lactis is an important industrial microorganism and a widely used model object for research in the field of lactic acid bacteria (LAB) biology. The development of new L. lactis and related LAB strains with improved properties, including phage-resistant strains for dairy fermentation, LAB-based vaccines or strains with altered genotypes for research purposes, are hindered by the lack of genome-editing tools that allow for the easy and straightforward incorporation of a significant amount of the novel genetic material, such as large genes or operons, into the chromosomes of these bacteria. We recently employed a suggested system based on the CRISPR–Cas-associated transposon for the editing of the L. lactis genome. After the in-depth redesign of the system, we were able to achieve the stable incorporation of the fragments that were sized up to 10 kbp into the L. lactis beta-galactosidase gene. The efficiency of editing under the optimized conditions were 2 × 10−4 and 4 × 10−5 for 1 kbp and 10 kbp, respectively, which are sufficient for fast and easy modifications if a positive selection marker can be used.

A methodology of line-carved relief editing by mesh feature operations was proposed. Feature region of mesh model is firstly identified by curvature criterion. Secondly, profile lines are extracted by such feature operations as clustering, eliminating, retrieving and thinning of feature points. Thirdly, profile lines are optimized by trimming and smoothing operations. Finally, shape of carved curves could be edited by specifying the width and depth of profile lines, and a satisfactory line-carved relief is achieved by nonlinear compression.

To improve livestock and aquaculture-raised fish as food, targeted mutagenesis using genome editing technologies is becoming more realizable. Myostatin (Mstn), which functions as the negative regulator of skeletal muscle growth, is one of the major targets to improve the edible ratio of livestock and farmed fish. We previously reported that the deficiency of Pm-mstn, one of Myostatin paralogs, improves muscle growth and changes body shape in a finfish species, red seabream (Pagrus major), as a result of editing the gene by means of CRISPR/Cas9. In this study, we established Pm-mstnb-deficient red seabream, which is a null-allelic mutant of another paralogous gene of Myostatin in the species, and analyzed their phenotype in terms of growth traits and body shape. A comparison of all growth traits between Pm-mstnbwt/wt and Pm-mstnb-5/-5 revealed no significant differences. In addition, all metrics for body shape, defined as the ratios of body depth, body width, and depth of the caudal peduncle to body length, respectively, were also similar in Pm-mstnbwt/wt and Pm-mstnb-5/-5. Therefore, we concluded that Pm-mstnb does not function as a negative regulator of skeletal muscle growth in red seabream.

Some unusual experiences and situations involving twins that warrant media attention are also summarized. This research uses survey and in-depth interview for collecting the data. 87 students took a part in this study; then the result was explored by conducting in-depth interviews with unique samples of student. The result, students who have taken the mobile and social media journalism course already have the knowledge and ability to use mobile devices and social media platform. The challenge is when they uses a mobile device in the news production process and in the news editing process because of the characteristics of the device (small screen). Knowledge of the specifications of each editing application is very helpful as a mobile journalist. The class will be more ideal with learning how to reporting or hosting news which is very common on social media platform.

Background Major histocompatibility class I (MHC-I, human leukocyte antigen [HLA]-I in humans) molecules present small fragments of the proteome on the cell surface for immunosurveillance, which is pivotal to control infected and malignant cells. Immunogenic peptides are generated and selected in the MHC-I antigen processing and presentation pathway. In this pathway, two homologous molecules, tapasin and TAPBPR, optimise the MHC-I peptide repertoire that is ultimately presented at the plasma membrane. Peptide exchange on HLA-I by human TAPBPR involves the flexible loop region K22-D35, with the leucine at position 30 (L30) involved in mediating peptide dissociation. However, our understanding of the exact molecular mechanisms governing TAPBPR-mediated peptide exchange on HLA-I allotypes remains incomplete. Methods Here, in-depth re-analyses of published immunopeptidomics datasets was used to further examine TAPBPR peptide editing activity and mechanism of action on HLA-I. The role of the TAPBPR editing loop in opening the HLA-I peptide binding groove was assessed using a molecular dynamics simulation. Results We show that TAPBPR shapes the peptide repertoire on HLA-A, -B and -C allotypes. The TAPBPR editing loop was not essential to allow HLA-I to adopt an open state. L30 in the TAPBPR editing loop was typically sufficient to mediate peptide repertoire restriction on the three HLA-I allotypes expressed by HeLa cells. TAPBPR was also able to load peptides onto HLA-I in a loop-dependent manner. Conclusions These results unify the previously hypothesised scoop loop and peptide trap mechanisms of TAPBPR-mediated peptide exchange, with the former involved in peptide filtering and the latter in peptide loading.

Dynamic human movements such as dance are difficult to capture without using external markers due to the high complexity of a dancer’s body. This article introduces a marker-free motion capture and composition system for dance motion that uses multiple RGB and depth sensors. Our motion capture system utilizes a set of high-speed RGB and depth sensors to generate skeletal motion data from an expert dancer. During the motion acquisition process, a skeleton tracking method based on a particle filter is provided to estimate the motion parameters for each frame from a sequence of color images and depth features retrieved from the sensors. The expert motion data become archived in a database. The authoring methods in our composition system automate most of the motion editing processes for general users by providing an online motion search with an input posture and then performing motion synthesis on an arbitrary motion path. Using the proposed system, we demonstrate that various dance performances can be composed in an intuitive and efficient way on client devices such as tablets and kiosk PCs.

Sampling method used is purposive sampling. Data collection methods used in this study were in-depth interviews (In Depth Interview), documentation, participatory observation and FGD (focus group discussion). The data management methods in this study are editing, coding, and tabulating and the data analysis used is descriptive with a qualitative approach. The results of the study show that there are 3 prospects that can be developed for a Tourism Village in Burai Village, namely a floating hall, a flower garden and a lebak lebung. The contribution made by tourist villages to farmers' income is an average of 17.2%. Metode pengelolaan data dalam penelitian ini ialah editing, coding, dan tabulating dan analisis data yang digunakan ialah deskriptif dengan pendekatan kualitatif. Hasil dari penelitian menunjukkan terdapat 3 prospek yang dapat dikembangkan untuk Desa Wisata di Desa Burai yaitu balai apung, taman bunga dan lebak lebung. Kontribusi yang diberikan oleh desa wisata terhadap pendapatan petani, rata-rata sebesar 17,2%.

Inactivation of the CCR5 gene by CRISPR editing in human embryos, as recently attempted in China, was touted as a positive change for the babies involved since it was expected to impart resistance to HIV infection. However, it turns out that the absence of CCR5 is not neutral but actually decreases fitness, as shown by survival analysis of population data in the UK biobank. This underlines the pitfalls of genetic enhancement, and emphasizes that any germline modification must be preceded by in-depth studies to exclude unforeseen negative effects. ‡

A flat bed scanner can be used as a high-resolution digital camera with close-up capabitlities for photographing plant material such as leaves, flowers, and plant pests. The scanner is very useful as a diagnostic and instructional tool that is more portable and less expensive than a camera and dissecting microscope. The quality of the images can be very good and can be enhanced in post-production using image editing software. The main disadvantage is the shallow depth of field, which requires the object be on a single plane.

This study aims to find out and examine in depth the performance of civil servants in public services during the Covid-19 pandemic. The method used in this study is a qualitative research method. The steps used in data analysis techniques are editing, clarifying data, then drawing conclusions. Service during the pandemic is not the same as before Covid-19. Working from home during the Covid-19 pandemic did not go as expected, because service activities in the office are work that must be completed at the office, not at home.

AbstractRemarkable diversity in the domain of genome loci architecture, structure of effector complex, array of protein composition, mechanisms of adaptation along with difference in pre-crRNA processing and interference have led to a vast scope of detailed classification in bacterial and archaeal CRISPR/Cas systems, their intrinsic weapon of adaptive immunity. Two classes: Class 1 and Class 2, several types and subtypes have been identified so far. While the evolution of the effector complexes of Class 2 is assigned solely to mobile genetic elements, the origin of Class 1 effector molecules is still in a haze. Majority of the types target DNA except type VI, which have been found to target RNA exclusively. Cas9, the single effector protein, has been the primary focus of CRISPR-mediated genome editing revolution and is an integral part of Class 2 (type II) system. The present review focuses on the different CRISPR types in depth and the application of CRISPR/Cas9 for epigenome modification, targeted base editing and improving traits such as abiotic and biotic stress tolerance, yield and nutritional aspects of tomato breeding.

Wahana Honda is one of the businesses engaged in the distribution of Honda motorcycles and the service of Honda motorcycles is one of the services of Wahana Honda. This research uses descriptive research method with a qualitative approach. The method used is a case study by conducting in-depth interviews with informants. The results of this study indicate that the role of producers in the Wahana Honda Prime web series process consists of 3 stages, in pre-production which of course it is related in making themes, story ideas, cinematographic processes in the production and editing of images. In the production process the producer is fully responsible for controlling, supervising, and coordinating to all the crew involved for the smooth running of their production, carrying out production in accordance with the script and shooting script made by the producer. In post production there are 2 editing processes, namely offline and online. The producer ensures and supervises the production work in accordance with the client's direction and deliver the results in accordance with the agreed time.

Анотація:

The article is primarily aimed at presenting the tasks to be completed under the project The Sources for the History of the Czapskis in the 18th Century. Egodocuments by Family Members of the Pomeranian Voivode Piotr Jan (1685–1736) – a Textual and Historical Analysis and Editing financed by the National Programme for the Development of Humanities. The said project will culminate in three editions – of the diary of the travels of the Starost of Knyszyn Tomasz Czapski and of the letters of the Pomeranian Voivode’s wife Konstancja Czapska and her children – her sons: Tomasz, Paweł Tadeusz and her daughter Rozalia. The work will be conducted simultaneously by literary scholars, historians, linguists and an art historian, hence the sources prepared to appear in print will have been thoroughly analysed, characterised and published with due diligence. In the author’s opinion, implementation of such interdisciplinary scholarly enterprises is particularly important in the field of the humanities, as it breaks the hermetic seal of the conducted research, hence enabling an in-depth and multidimensional interpretation of old egodocuments.

Abstract Objective: To increase the extent of the depth of field and focal sharpness in highly magnified image series from cadaveric microneurosurgical anatomic study or live surgery photographs obtained with the operating microscope, providing a higher quality and more satisfactory photographic and imaging experience. Methods: We used a computer workstation, CombineZM software (Alan Hadley, United Kingdom, www.hadleyweb.pwp.blueyonder.co.uk/), an injected cadaver head specimen, and an operating microscope equipped with a digital single-lens reflex camera. Fifteen images were obtained of the dissection area through an anterior petrosal approach. The focus point was fixed to different points in each image. The images were loaded into CombineZM software for processing. Results: The stacking process of photographs with CombineZM freeware provides significant increase in extent of depth of field and wider area of image clarity, producing a sharp, high-quality image. Conclusion: An image processed from a stack of photographs from cadaveric microneurosurgical studies or from the operating microscope in live surgery can be rendered to show extended 3-dimensional depth of field and clarity. This method offers improvements for editing, displaying, and publishing neurosurgical anatomic images.

My research focuses on students who speak English as their first language and are currently enrolled in a technical writing class open to students from many majors. I will report in this post on the outcomes of teaching these students five distinct types of technical writing. In this case, the students in issue have English as their mother tongue. Previous experimental research has shown that it is useful to explicitly teach academic writing to adults who speak English as their first language. The findings of the research have proved this. In contrast, no research on technical writing has ever been conducted compared to this study's level of depth and breadth. In order to investigate these effects, I employed a strategy that included a few different research approaches. In order to present a more comprehensive, in-depth characterization of the 534 texts authored by 316 student authors, this approach consisted of a control-group quasi-experimental design with a qualitative analysis. These were the components that made up the strategy. According to the findings, the participants in the genre workshop created writings with a significantly higher sensitivity to audience, purpose, structure, design, style, and editing than those generated by participants who were taught using more traditional approaches. Participants displayed a superior awareness of audience, aim, and editing while working within the framework of technical genres in the job materials text type instead of the procedures text type. This was the case when comparing the two text types. When contrasting the two different forms of text, this was the result.

Capturing and editing full-head performances enables the creation of virtual characters with various applications such as extended reality and media production. The past few years witnessed a steep rise in the photorealism of human head avatars. Such avatars can be controlled through different input data modalities, including RGB, audio, depth, IMUs, and others. While these data modalities provide effective means of control, they mostly focus on editing the head movements such as the facial expressions, head pose, and/or camera viewpoint. In this paper, we propose AvatarStudio, a text-based method for editing the appearance of a dynamic full head avatar. Our approach builds on existing work to capture dynamic performances of human heads using Neural Radiance Field (NeRF) and edits this representation with a text-to-image diffusion model. Specifically, we introduce an optimization strategy for incorporating multiple keyframes representing different camera viewpoints and time stamps of a video performance into a single diffusion model. Using this personalized diffusion model, we edit the dynamic NeRF by introducing view-and-time-aware Score Distillation Sampling (VT-SDS) following a model-based guidance approach. Our method edits the full head in a canonical space and then propagates these edits to the remaining time steps via a pre-trained deformation network. We evaluate our method visually and numerically via a user study, and results show that our method outperforms existing approaches. Our experiments validate the design choices of our method and highlight that our edits are genuine, personalized, as well as 3D- and time-consistent.

Indoor mapping and modelling requires that acquired data be processed by editing, fusing, formatting the data, amongst other operations. Currently the manual interaction the user has with the point cloud (data) while processing it is visual. Visual interaction does have limitations, however. One way of dealing with these limitations is to augment audio in point cloud processing. Audio augmentation entails associating points of interest in the point cloud with audio objects. In coarse scan registration, reverberation, intensity and frequency audio cues were exploited to help the user estimate depth and occupancy of space of points of interest. Depth estimations were made reliably well when intensity and frequency were both used as depth cues. Coarse changes of depth could be estimated in this manner. The depth between surfaces can therefore be estimated with the aid of the audio objects. Sound reflections of an audio object provided reliable information of the object surroundings in some instances. For a point/area of interest in the point cloud, these reflections can be used to determine the unseen events around that point/area of interest. Other processing techniques could benefit from this while other information is estimated using other audio cues like binaural cues and Head Related Transfer Functions. These other cues could be used in position estimations of audio objects to aid in problems such as indoor navigation problems.
