This paper propose a new multiplexing technique which use the bipolar RZ signal to realize duty cycle division multiplexing (DCDM) ,we call it Absolute Polar Duty Cycle Division Multiplexing (APDCDM).Multiplexing and demultiplexing principle of APDCDM are analyzed. Here we have designed a optical fiber transmission system based on APDCDM of three users by Joint simulation with Optisystem and Matlab. Simulation result shows the feasibility of this system. By the analysis of signal energy, explain APDCDM can carry more users than DCDM.

Two kinds of graphene coatings are obtained by the graphene drop-coating drying method (DCDM) and the coating graphene conductive adhesive (CGCA). The effects of these two kinds of graphene coatings on the friction, wear, and voltage signals of the electrical contact interface are explored. The test results show that the presence of the graphene coating can effectively reduce the friction coefficient and friction force, and the graphene coating prepared by the DCDM possesses the best ability in reducing the friction coefficient. Although the presence of the graphene coating will lead to the increase in interface contact voltage at the initial stage, the voltage signal gradually becomes stable with the progress of friction and wear, suggesting that the graphene coating will not affect the stability of sliding electrical contact. Wear analysis results show that the graphene coating prepared by the DCDM has a good anti-wear effect, and the graphene particles in the abrasion area play the role of solid lubrication. Finite element analysis results show that the graphene coating will generate thermal expansion when electric current is applied, accordingly avoid the direct contact between the metal substrate, and, thus, reduce the interface friction and alleviate the wear degree of interface. However, the normal force fluctuation of the interface may increase.

Abstract The stability of particles in the cosmic soup is an important property that can affect the cosmic evolution. In this work, we update the constraints on the decaying cold dark matter scenario, when the decay products are effectively massless. We assume, as a base case, that all of dark matter is unstable and it can decay on cosmological time scales. We then extend the analysis to include the scenario where only a fraction of dark matter is unstable, while the remaining part is composed of the standard, stable, dark matter. We consider observations of cosmological probes at linear scales, i.e., Planck 2018 cosmic microwave background temperature, polarization, and lensing measurements, along with geometrical information from baryon acoustic oscillation (BAO) measurements from SDSS DR7, BOSS DR12, eBOSS DR16 and 6dFGS, to derive conservative constraints on the dark matter decay rate. We consider these dataset separately, to asses the relative constraining power of each dataset, as well as together to asses the joint constraints. We find the most stringent upper limit on the decay rate of decaying cold dark matter particles to be ΓDCDM < 0.129 × 10-18 s-1 (or, equivalently, the dark matter lifetime τ DCDM > 246 Gyr) at 95% C.L. for the combination of Planck primary anisotropies, lensing and BAO. We further explore one-parameter extensions of our baseline DCDM model. Namely, we vary the sum of neutrino masses, the curvature density parameter, and the tensor-to-scalar ratio along with the DCDM parameters. When varying the tensor-to-scalar ratio we also add data from the BICEP/Keck experiment.

The control of plant leaf diseases is crucial as it affects the quality and production of plant species with an effect on the economy of any country. Automated identification and classification of plant leaf diseases is, therefore, essential for the reduction of economic losses and the conservation of specific species. Various Machine Learning (ML) models have previously been proposed to detect and identify plant leaf disease; however, they lack usability due to hardware sophistication, limited scalability and realistic use inefficiency. By implementing automatic detection and classification of leaf diseases in fruit trees (apple, grape, peach and strawberry) and vegetable plants (potato and tomato) through scalable transfer learning on Amazon Web Services (AWS) SageMaker and importing it into AWS DeepLens for real-time functional usability, our proposed DeepLens Classification and Detection Model (DCDM) addresses such limitations. Scalability and ubiquitous access to our approach is provided by cloud integration. Our experiments on an extensive image data set of healthy and unhealthy fruit trees and vegetable plant leaves showed 98.78% accuracy with a real-time diagnosis of diseases of plant leaves. To train DCDM deep learning model, we used forty thousand images and then evaluated it on ten thousand images. It takes an average of 0.349s to test an image for disease diagnosis and classification using AWS DeepLens, providing the consumer with disease information in less than a second.

This research aimed to evaluate the interactions and effects of 2 and 4% addition levels of poultry slaughterhouse fat (chicken tallow) and soybean oil in diets for broiler chickens. Two experiments were carried out using one-day-old male Cobb chicks in an entirely random design with a 2x2 factorial scheme. In the first experiment, 560 chicks were used to evaluate performance and carcass characteristics. In the second experiment, 100 chicks were used to determine the nutrient digestibility, dietary energy utilization and the lipase and amylase pancreatic activity. There was no interaction between the fat sources and the addition levels for any of the analyzed variables, except for the digestibility coefficient of dry matter (DCDM), which was higher in diets added with 2% soybean oil when compared to chicken tallow. The addition of 4% fat in the diet, regardless of fat source, improved the digestibility coefficient of ethereal extract (DCEE) and increased weight gain and feed intake. Moreover, in the initial phase, the addition of 4% fat to the diet increased lipase activity when compared to diets with 2% addition, and a positive correlation between DCEE and pancreatic lipase activity was observed. In conclusion, there is no interaction between fat sources and addition levels, except for DCDM. Carcass characteristics are not influenced by any of the studied factors. The addition of 4% fat increases pancreatic lipase activity and improves DCEE, resulting in greater weight gain, regardless of the tested fat source, making chicken tallow a great alternative to soybean oil.

The role of octopamine (OA) in the feeding system of the pond snail, Lymnaea stagnalis , was studied by applying behavioural tests on intact animals, and a combination of electrophysiological analysis and morphological labelling in the isolated central nervous system. OA antagonists phentolamine, demethylchlordimeform (DCDM) and 2–chloro–4–methyl–2–(phenylimino)–imidazolidine (NC–7) were injected into intact snails and the sucrose–induced feeding response of animals was monitored. Snails that received 25–50 mg kg -1 phentolamine did not start feeding in sucrose, and the same dose of NC–7 reduced the number of feeding animals by 80–90% 1–3 hours after injection. DCDM treatment reduced feeding by 20–60%. In addition, both phentolamine and NC–7 significantly decreased the feeding rate of those animals that still accepted food after 1–6 hours of injection. In the central nervous system a pair of buccal neurons was identified by electrophysiological and morphological criteria. After double labelling (intracellular staining with Lucifer yellow followed by OA–immunocytochemistry) these neurons were shown to be OA immunoreactive, and electrophysiological experiments confirmed that they are members of the buccal feeding system. Therefore the newly identified buccal neurons were called OC neurons (putative OA containing neurons or OAergic cells). Synchronous intracellular recordings demonstrated that the OC neurons share a common rhythm with feeding neurons either appearing spontaneously or evoked by intracellularly stimulated feeding interneurons. OC neurons also have synaptic connections with identified members of the feeding network: electrical coupling was demonstrated between OC neurons and members of the B4 cluster motoneurons, furthermore, chemically transmitted synaptic responses were recorded both on feeding motoneurons (B1, B2 cells) and the SO modulatory interneuron after the stimulation of OC neurons. However, elementary synaptic potentials could not be recorded on the follower cells of OC neurons. Prolonged (20 to 30 s) intracellular stimulation of OC cells activated the buccal feeding neurons leading to rhythmic activity pattern (fictive feeding) in a way similar to OA applied by perfusion onto isolated central nervous system (CNS) preparations. Our results suggest that OA acts as a modulatory substance in the feeding system of Lymnaea stagnalis and the newly identified pair of OC neurons belongs to the buccal feeding network.

La música latinoamericana recoge un amplio espectro de tradiciones compartidas que desafían la noción convencional de fronteras nacionales y abren discusiones sobre factores de identidad personal, regional, nacional y continental. Este artículo toma como ejemplo un pasillo colombiano compuesto por una mujer de origen nariñense, el cual fue reinterpretado por un cantautor costarricense y territorializado como una pieza representativa del folclor de Guanacaste. La obra original se conoce en Colombia como el Cafetero, de la pianista pastusa Maruja Hinestrosa Eraso. La versión costarricense tiene el título de El hombre macho, y por muchos años ha sido atribuida al guanacasteco Adán Guevara Centeno. El objetivo del articulo apunta a profundizar y analizar los alcances de estas correspondencias, señalando ejes de relación que conecten las historias de la música en ambos países y cuestionando las posturas regionalistas en torno a estas composiciones. La metodología integra el análisis de fuentes escritas y sonoras, en articulación con un trabajo de campo que inició en Pasto (Colombia) y se complementó con una estancia de investigación en San José y Guanacaste (Costa Rica). Los resultados dan cuenta de los estrechos vínculos que se presentan en distintas músicas latinoamericanas, proponiendo una discusión fundamentada en dos conceptos relevantes para las obras musicales en cuestión: género y transnacionalidad.

As a new satellite sensor of the GaoFen (GF) series, GF-6 Wide Field of View (WFV) with the resolution of 16 m has the characteristics of wide coverage, high-frequency imaging and has four new bands of two red-edge, yellow, and purple compared with GF-1 WFV. In order to test the validity of the supplementary bands of GF-6WFV data for change detection of land use/land cover (LULC), this study applied the Double-constrained Change Detection Method (DCDM) that uses the double constraints (change vector intensity and correlation coefficient) for change detection on object-level. According to two GF-6WFV imageries acquired in the Xiong’an New Area in June of 2018 and 2019, feature analysis was performed to determine whether the new bands are helpful to detect the change of LULC first. Then, by coupling these selected features, the intensity of change vector and correlation coefficient were used as the double constraints to perform the change detection. The study demonstrates that the relevant features of the two red-edge bands can achieve the overall accuracy of 89% for change detection of LULC and improved by 2% comparing with using the corresponding temporal GF-1WFV data, while the purple and yellow bands cannot provide enough effective information for this detection. This study can provide theoretical support for the in-depth applications of GF-6 WFV data products in the change detection fields and has explored its applicability and potential in resource and environment monitoring, it is helpful to the further applications.

Abstract IL-21 promotes autoimmunity in the chronic graft versus host disease (cGVHD) model through both B and CD4 T cell intrinsic mechanisms. To determine the importance of IL-21R signaling in CD8 T cells in murine lupus, we assessed the effect of IL-21 administration on autoimmune parameters in the DBA-into-F1 model of cGVHD. In addition, the effect of IL-21R deficiency on CD8 T cells was assessed in the B6-into-F1 model of acute (a) GVHD using IL-21R sufficient and deficient donor (d) cells. cGVHD mice that received rmIL-21 exhibit attenuated autoimmune parameters with respect to host B cell expansion, anti-ssDNA autoAb production and renal disease along with enhanced dCD8 expansion and donor anti-host CTL activity. Phenotype of aGVHD induced with IL-21R-/- donor cells converted from an acute to a chronic pattern characterized by increased autoAb production, decreased host B cell elimination, impaired CTL activity and decreased dCD4 and dCD8 cell expansion. In both models, IL-21R signaling on dCD8 cells resulted in increased dCD8 expansion, granzyme B level and IFNg expression. These results suggest that lack of IL-21/IL-21R interaction on CD8 T cells converts acute to chronic GVHD, while in cGVHD, IL-21 administration counteracts the humoral component by enhancing CTL generation and host B cells elimination. Therefore, IL-21R signaling on CD8 T cells attenuates disease phenotype in murine lupus, thus opposing the enhancing effects of IL-21R signaling on CD4 T cells and B cells.

In plant cells, cytokinesis depends on a cytoskeletal structure called a phragmoplast, which directs the formation of a new cell wall between daughter nuclei after mitosis. The orientation of cell division depends on guidance of the phragmoplast during cytokinesis to a cortical site marked throughout prophase by another cytoskeletal structure called a preprophase band. Asymmetrically dividing cells become polarized and form asymmetric preprophase bands prior to mitosis; phragmoplasts are subsequently guided to these asymmetric cortical sites to form daughter cells of different shapes and/or sizes. Here we describe two new recessive mutations, discordia1 (dcd1) and discordia2 (dcd2), which disrupt the spatial regulation of cytokinesis during asymmetric cell divisions. Both mutations disrupt four classes of asymmetric cell divisions during the development of the maize leaf epidermis, without affecting the symmetric divisions through which most epidermal cells arise. The effects of dcd mutations on asymmetric cell division can be mimicked by cytochalasin D treatment, and divisions affected by dcd1 are hypersensitive to the effects of cytochalasin D. Analysis of actin and microtubule organization in these mutants showed no effect of either mutation on cell polarity, or on formation and localization of preprophase bands and spindles. In mutant cells, phragmoplasts in asymmetrically dividing cells are structurally normal and are initiated in the correct location, but often fail to move to the position formerly occupied by the preprophase band. We propose that dcd mutations disrupt an actin-dependent process necessary for the guidance of phragmoplasts during cytokinesis in asymmetrically dividing cells.

As a variant of minimum connected dominating set problem, two disjoint connected dominating sets (DCDS) problem is to ask whether there are two DCDS [Formula: see text] in a connected graph [Formula: see text] with [Formula: see text] and [Formula: see text], and if not, how to add an edge subset with minimum cardinality such that the new graph has a pair of DCDS. The two DCDS problem is so hard that it is NP-hard on trees. In this paper, if the vertex set [Formula: see text] of a connected graph [Formula: see text] can be partitioned into two DCDS of [Formula: see text], then it is called a DCDS graph. First, a necessary but not sufficient condition is proposed for cubic (3-regular) graph to be a DCDS graph. To be exact, if a cubic graph is a DCDS graph, there are at most four disjoint triangles in it. Next, if a connected graph [Formula: see text] is a DCDS graph, a simple but nontrivial upper bound [Formula: see text] of the girth [Formula: see text] is presented.

Background The digital Clock Drawing Test (dCDT) has been recently used as a more objective tool to assess cognition. However, the association between digitally obtained clock drawing features and structural neuroimaging measures has not been assessed in large population-based studies. Objective We aimed to investigate the association between dCDT features and brain volume. Methods This study included participants from the Framingham Heart Study who had both a dCDT and magnetic resonance imaging (MRI) scan, and were free of dementia or stroke. Linear regression models were used to assess the association between 18 dCDT composite scores (derived from 105 dCDT raw features) and brain MRI measures, including total cerebral brain volume (TCBV), cerebral white matter volume, cerebral gray matter volume, hippocampal volume, and white matter hyperintensity (WMH) volume. Classification models were also built from clinical risk factors, dCDT composite scores, and MRI measures to distinguish people with mild cognitive impairment (MCI) from those whose cognition was intact. Results A total of 1656 participants were included in this study (mean age 61 years, SD 13 years; 50.9% women), with 23 participants diagnosed with MCI. All dCDT composite scores were associated with TCBV after adjusting for multiple testing (P value <.05/18). Eleven dCDT composite scores were associated with cerebral white matter volume, but only 1 dCDT composite score was associated with cerebral gray matter volume. None of the dCDT composite scores was associated with hippocampal volume or WMH volume. The classification model for differentiating MCI and normal cognition participants, which incorporated age, sex, education, MRI measures, and dCDT composite scores, showed an area under the curve of 0.897. Conclusions dCDT composite scores were significantly associated with multiple brain MRI measures in a large community-based cohort. The dCDT has the potential to be used as a cognitive assessment tool in the clinical diagnosis of MCI.

Hospital readmission among people with diabetes is common and costly. A better understanding of the differences between people requiring hospitalization primarily for diabetes (primary discharge diagnosis, 1°DCDx) or another condition (secondary discharge diagnosis, 2°DCDx) may translate into more effective ways to prevent readmissions. This retrospective cohort study compared readmission risk and risk factors between 8054 hospitalized adults with a 1°DCDx or 2°DCDx. The primary outcome was all-cause hospital readmission within 30 days of discharge. The readmission rate was higher in patients with a 1°DCDx than in patients with a 2°DCDx (22.2% vs. 16.2%, p < 0.01). Several independent risk factors for readmission were common to both groups including outpatient follow up, length of stay, employment status, anemia, and lack of insurance. C-statistics for the multivariable models of readmission were not significantly different (0.837 vs. 0.822, p = 0.15). Readmission risk of people with a 1°DCDx was higher than that of people with a 2°DCDx of diabetes. Some risk factors were shared between the two groups, while others were unique. Inpatient diabetes consultation may be more effective at lowering readmission risk among people with a 1°DCDx. These models may perform well to predict readmission risk.

The parent-rated Developmental Coordination Disorder Questionnaire (DCDQ) has been revised to incorporate a wider age range, including adolescence. In this exploratory study, internal consistency and validity of the DCDQ-2007 was assessed using a community-based sample of 87 adolescents. Psychometric properties of the DCDQ-2007 were investigated and concurrent validity, sensitivity, and specificity were assessed with the MABC-2 as a criterion standard. The results demonstrated high internal consistency for the DCDQ-2007 and a relationship with the MABC-2 was found. The DCDQ-2007 met the recommended standard for sensitivity, although the confidence interval was large; however, it failed to meet the recommended standard for specificity. This has important implications concerning the suitability of the DCDQ-2007. Although promising psychometric properties were found within the current study, the applicability of the DCDQ-2007 as a screening measure for motor difficulties requires careful consideration.

This study investigated the relationship between the Developmental Coordination Disorder Questionnaire 2007 (DCDQ’07) and the Bruininks-Oseretsky Test of Motor Proficiency Second Edition (BOT-2) in Korea. This study also adjusted the cutoff score of the DCDQ’07 based on the BOT-2 for Korean children. A total of 256 children were recruited from communities in Korea. They were divided into two age groups: 8 to 9 years old and 10 to 12 years old. Children performed the BOT-2, and their parents completed the DCDQ’07. The correlation between the DCDQ’07 and the BOT-2 was analyzed. The adjusted DCDQ’07 cutoff score for Korean children was calculated using the BOT-2 as the criterion through a receiver operating characteristic curve. A significant correlation between the DCDQ’07 and the BOT-2 was found, indicating that Korean parents’ perception of children’s motor skills was related to their children’s actual motor proficiency. The adjusted cutoff score of the DCDQ’07 had a sensitivity of 72.7–85.7% and a specificity of 62.5–64.0%. This study demonstrated that children’s motor skills reported by Korean parents on the DCDQ’07 were valid based on a community sample. The adjusted cutoff score of the DCDQ’07 could be used to identify children suspected of having a developmental coordination disorder.

Background The Clock Drawing Test (CDT) has been widely used in clinic for cognitive assessment. Recently, a digital Clock Drawing Text (dCDT) that is able to capture the entire sequence of clock drawing behaviors was introduced. While a variety of domain-specific features can be derived from the dCDT, it has not yet been evaluated in a large community-based population whether the features derived from the dCDT correlate with cognitive function. Objective We aimed to investigate the association between dCDT features and cognitive performance across multiple domains. Methods Participants from the Framingham Heart Study, a large community-based cohort with longitudinal cognitive surveillance, who did not have dementia were included. Participants were administered both the dCDT and a standard protocol of neuropsychological tests that measured a wide range of cognitive functions. A total of 105 features were derived from the dCDT, and their associations with 18 neuropsychological tests were assessed with linear regression models adjusted for age and sex. Associations between a composite score from dCDT features were also assessed for associations with each neuropsychological test and cognitive status (clinically diagnosed mild cognitive impairment compared to normal cognition). Results The study included 2062 participants (age: mean 62, SD 13 years, 51.6% women), among whom 36 were diagnosed with mild cognitive impairment. Each neuropsychological test was associated with an average of 50 dCDT features. The composite scores derived from dCDT features were significantly associated with both neuropsychological tests and mild cognitive impairment. Conclusions The dCDT can potentially be used as a tool for cognitive assessment in large community-based populations.

Flying locusts perform a characteristic gliding dive in response to predator-sized stimuli looming from one side. These visual looming stimuli trigger trains of spikes in the descending contralateral movement detector (DCMD) neuron that increase in frequency as the stimulus gets nearer. Here we provide evidence that high-frequency (>150 Hz) DCMD spikes are involved in triggering the glide: the DCMD is the only excitatory input to a key gliding motor neuron during a loom; DCMD-mediated EPSPs only summate significantly in this motor neuron when they occur at >150 Hz; when a looming stimulus ceases approach prematurely, high-frequency DCMD spikes are removed from its response and the occurrence of gliding is reduced; and an axon important for glide triggering descends in the nerve cord contralateral to the eye detecting a looming stimulus, as the DCMD does. DCMD recordings from tethered flying locusts showed that glides follow high-frequency spikes in a DCMD, but analyses could not identify a feature of the DCMD response alone that was reliably associated with glides in all trials. This was because, for a glide to be triggered, the high-frequency spikes must be timed appropriately within the wingbeat cycle to coincide with wing elevation. We interpret this as flight-gating of the DCMD response resulting from rhythmic modulation of the flight motor neuron's membrane potential during flight. This means that the locust's escape behavior can vary in response to the same looming stimulus, meaning that a predator cannot exploit predictability in the locust's collision avoidance behavior.

Abstract Background DCDT2980S (DCDT) is an anti-CD22 monoclonal antibody (Ab) conjugated to MMAE, a potent anti-microtubule inhibitor. We have previously reported clinical activity and acceptable toxicity in pts with R/R B-cell NHL treated at DCDT doses of 1.8 and 2.4 mg/kg. The maximum tolerated dose (MTD) was defined as 2.4 mg/kg either as a single agent or in combination with RTX (375 mg/m2) every 21 days (q21d) (Advani et al. ASH 2012, abstract 59). In this report, we provide updated results from pts treated at 1.8 mg/kg as well as an expanded cohort treated at 2.4 mg/kg. Methods We evaluated safety, tolerability, pharmacokinetics (PK), and activity of DCDT q21d with or without RTX (375 mg/m2) in pts with R/R B-cell NHL. Expanded cohorts of pts with R/R diffuse large cell lymphoma (DLBCL) or indolent (i)NHL were evaluated at the single-agent MTD of 2.4 mg/kg q21d. Results Forty-six pts enrolled in DCDT ≥ 1.8 mg/kg (7 at 1.8 mg/kg, 39 at 2.4 mg/kg) and 16 in the DCDT+RTX cohort (5 at 1.8 mg/kg, 11 at 2.4 mg/kg). Median age was 66 yrs; 92% had an ECOG PS<2; median of 3 prior regimens (range 1-11); 100% prior RTX, 8% prior stem cell transplant. Patients received a median of 5 cycles (range 1, 24) of DCDT and 6 cycles (range 1, 15) of DCDT + RTX; 7 patients continue to receive study treatment. The DCDT+RTX safety profile did not differ from DCDT monotherapy and toxicities were mostly Grade 1-2. Grade ≥ 3 treatment-emergent adverse events (TEAEs) in ≥5% of pts included neutropenia (26%), hyperglycemia (10%), peripheral sensory neuropathy (10%), fatigue (5%), and diarrhea (5%). Febrile neutropenia and Grade 3-4 infection occurred in 1 pt and 7 pts respectively. The median time to first onset peripheral neuropathy (PN) was 50 days. Worsening PN (at least one grade increase) occurred in 18/33 pts (55%) with median time to worsening of 48 days. PN was managed with dose delays and dose reductions resulting in complete reversal in 5 (15%) pts. Serious AEs were seen in 37% of pts, the most common (≥ 3 pts) included pyrexia (3), pneumonia (3) and dyspnea (3). Treatment discontinuations for AEs were reported in 16 (26%) pts including 12 for peripheral neuropathy. Twenty-four pts (39%) had at least one dose delay including 10 for neutropenia and 5 for peripheral neuropathy. Twelve pts (19%) had at least one dose reduction including 8 for peripheral neuropathy and 7 for neutropenia. Three deaths occurring within 60 days of the last dose of DCDT judged unrelated to DCDT were reported. Exposure of Ab-conjugated (ac) MMAE, total Ab, and unconjugated MMAE increased with dose. Maximal concentrations of unconjugated MMAE were >100-fold lower than acMMAE with average Cycle 1 value of 5-7 ng/mL at the 2.4 mg/kg ADC dose level. Moderate accumulation of acMMAE and total Ab on the q21d dosing schedule was observed with no accumulation of free MMAE. Concurrent RTX administration did not impact DCDT PK. Overall objective responses were observed in 19/46 (41%) DCDT and 5/16 (31%) DCDT+RTX pts. Objective (OR) and complete responses (CR) by histology were as follows: The median PFS for DLBCL patients treated with DCDT or DCDT + RTX was 115 days. The median PFS for iNHL patients treated with DCDT or DCDT + RTX was 227 days. Conclusions DCDT alone or combined with RTX was generally well-tolerated. PN and neutropenia are the principal toxicities associated with DCDT. PN was reversible in some patients with dose delays and reductions. Encouraging antitumor activity was observed in heavily pre-treated pts with R/R NHL. Updated results from this Phase I study will be presented. The addition of rituximab does not appear to enhance the efficacy of DCDT based on the small number of patients treated with the combination and continues to be evaluated in an ongoing randomized Phase II study in patients with R/R DLBCL and FL of DCDT+RTX versus a CD79b-directed ADC (DCDS4501A) with the same linker-cytotoxic agent. Abstract Background DCDS4501A (DCDS), an anti-CD79b monoclonal antibody (Ab), is conjugated to the anti-mitotic agent MMAE. We previously determined a recommended Phase II dose (RP2D) of 2.4 mg/kg every 21 days (q21d), and clinical activity in R/R B-cell NHL at doses ≥ 1.8 mg/kg (Palanca-Wessels et al. ASH 2012). Here we update results from patients (pts) treated at 1.8 mg/kg and from the 2.4 mg/kg expansion cohort. Methods We evaluated ongoing safety, tolerability, pharmacokinetics (PK) and activity of DCDS with or without rituximab (RTX) at 375 mg/m2 q21d in pts with R/R DLBCL and indolent (i)NHL. Results Sixty pts were treated with DCDS (6 at 1.8 mg/kg, 45 at 2.4 mg/kg) and DCDS+RTX (9, DCDS at 2.4 mg/kg). Median age 68 yrs (range 20-86); 82% ECOG PS<2; median 4 prior regimens (range 1-14); 97% had prior RTX; 28% had prior autologous stem cell transplant. The DCDS+RTX safety profile did not differ from DCDS monotherapy. Patients received a median of 7 cycles (range 1-20) of DCDS and 10 cycles (range 1-17) of DCDS + RTX; 18 patients continue to receive study treatment. Treatment-emergent adverse events (TEAEs) included neutropenia (50%), diarrhea (45%), nausea (40%), pyrexia (38%), peripheral neuropathy (25%), peripheral sensory neuropathy (20%), and hypokalemia (20%). Grade ≥ 3 AEs in ≥5% of pts included neutropenia (43%), anemia (13%), thrombocytopenia (7%), hyperglycemia (7%), fatigue (5%) and diarrhea (5%). Grade ≥ 3 infection was reported in 8 (13%) pts. Twenty-two (37%) pts reported a serious AE. TEAEs related to peripheral neuropathy (PN) were reported in 32 (53%) pts with median time to first onset of 63 days. 22/32 pts (69%) had worsening PN with median time to worsening of 49 days. Grade ≥ 3 peripheral neuropathy/peripheral sensory neuropathy/peripheral motor neuropathy was reported in 5 (8%) patients. PN was managed with dose delays and dose reductions resulting in complete reversal in 7 (22%) pts. Treatment discontinuations for AEs were reported in 25 (42%) pts including 17 for PN. Seven pts (12%) had ≥ 1 dose reduction including 3 for PN and 2 for neutropenia. Twenty-six patients (43%) had ≥ 1 dose delay including 14 for neutropenia and 6 for PN. Six deaths were reported within 60 days of last study treatment assessed as unrelated to DCDS. Exposure of Ab-conjugated (ac) MMAE, total Ab, and unconjugated MMAE increased with dose. Maximal concentrations of unconjugated MMAE were >100-fold lower than acMMAE with an average Cycle 1 value of 5-9 ng/mL at the 2.4 mg/kg ADC dose level. PN was reversible in some patients with dose delays and reductions. Encouraging anti-tumor activity was observed in heavily pretreated pts with R/R NHL. Updated results from this Phase I study will be presented. An ongoing randomized Phase II study of DCDS+RTX versus a CD22-directed ADC (DCDT2980S) with the same linker-cytotoxic agent in patients with R/R DLBCL and follicular lymphoma will further assess the efficacy of DCDS in the treatment of NHL. An experiment was conducted to ascertain the nutrient composition of dried cashew apple meal (DCAM) and the effect of diets containing varying levels of the DCAM on the growth performance, economics of production and the internal organs of albino rats. Sixteen rats with a mean initial weight of 64.7 g were randomly allotted to four isoproteic and isocaloric diets in a Randomized Complete Block Design based on their weight and sex. The diets were: T0 (0% DCAM), T1 (5% DCAM), T2 (10% DCAM) and T3 (15% DCAM). Each treatment had four replicates. Feed and water were provided ad libitum. Data collected were analysed using ANOVA of GenStat Discovery Edition 4. No differences occurred between diets for daily feed intake, daily weight gain and FCR. It cost $0.009, $0.016 and $0.002 more for rats on T1, T2 and T3 respectively to gain 100 g of weight. No differences occurred between treatments for internal organs except for the relative weight of the empty GIT where rats on T3 had higher weight (P < 0.05). It was deduced that the DCAM could be useful in monogastric livestock feeding and could be added to a rate up to 15% of the diet.

AbstractDichorionic diamniotic (DCDA) twin pregnancies after single blastocyst embryo transfer have been reported recently, although a blastocyst ovum is generally believed to divide into monochorionic twin pregnancy. We investigated the incidence of DCDA twin pregnancy after single blastocyst embryo transfer and their zygosity. This prospective cohort study included 655 consecutive twin pregnancies that were managed from 2006 to 2014 at our institution. Chorionicity and amnionicity were determined using first-trimester ultrasonography and/or placental pathology. Zygosity was analyzed if the cases were DCDA twins after single blastocyst embryo transfer. Among 655 twin pregnancies, there were 348 DCDA cases, 295 monochorionic diamniotic (MCDA) cases and 12 monochorionic monoamniotic cases. Single blastocyst embryo transfer was performed in 43 cases. Six out of the 43 (14%) cases involved DCDA twin pregnancies and the other 37 cases involved MCDA twin pregnancies. Three DCDA twins born after single blastocyst embryo transfer, wherein frozen embryo transfer (FET) was performed in the natural cycle, were dizygotic, and the other three cases, wherein FET with hormone replacement therapy was performed, were monozygotic. DCDA twin pregnancy occurred in 14% (7% for monozygotic and 7% for dizygotic) of twin pregnancies after single blastocyst embryo transfer cases.

OBJECTIVE: The primary objective of this article is to provide readers with case reports of drug- or chemical-induced aseptic meningitis (DCAM) described in the medical literature. Background information regarding the classification of DCAM, incidence, proposed mechanisms, associated risk factors, clinical management, and sequelae is presented. DATA SOURCES: A MEDLINE search was used to identify pertinent background literature and case reports of DCAM. DATA EXTRACTION: All case reports of DCAM involve anecdotal information. A critical analysis of a causal relationship to the implicated drug or chemical and the appearance of meningeal involvement is presented. Animal data are included when pertinent. DATA SYNTHESIS: DCAM is a rare adverse event associated with numerous agents. Patients present with a variety of clinical signs and symptoms and laboratory findings of cerebral spinal fluid, when obtained, vary greatly. Most patients fully recover without sequelae. CONCLUSIONS: Numerous drugs and chemicals have been implicated in the medical literature as producing DCAM. Two proposed mechanisms of DCAM have been theorized: a delayed hypersensitivity-type reaction and direct meningeal irritation. The nonsteroidal antiinflammatory drugs, certain antibiotics, radiographic agents, and muromonab-CD3 have been most frequently implicated. There appears to be an association between the occurrence of the hypersensitivity-type reactions and underlying collagen vascular or rheumatologic disease.

To solve the problem of the low recovery rate of braking energy and the short driving range of electric vehicles, a novel mechanical-electric-hydraulic dynamic coupling drive system (MEH-DCDS) is proposed in this article. MEH-DCDS is a new power integration device that allows electric, mechanical, and hydraulic energy to be converted mutually. It comprises a swash plate plunger pump/motor and a permanent magnet synchronous motor. This article explains the structure and working principles of MEH-DCDS. We describe the dynamic characteristics of MEH-DCDS and analyze the pump and hydraulic motor in the MEH-DCDS hydraulic module. The simulation results show that the flow variation of the MEH-DCDS hydraulic module accords with the design concept of MEH-DCDS, and the pressure variation of high and low pressure accumulators also accords with the theoretical situation. The energy flow of Mechanical-Electric-Hydraulic Power Coupling Electric Vehicle (MEHPC-EV) under different working modes is expounded, and the mathematical model of its key components is established. Based on AMESim and Simulink, the article establishes a vehicle simulation dynamic model. The dynamic performance of MEHPC-EV in UDDS is analyzed by co-simulation. The simulation results show that the application of MEH-DCDS in electric vehicles is feasible. MEHPC-EV reduced battery energy consumption by 26.18% compared to EV. The research in this paper verifies the accuracy and superiority of the system, which has a significant reference value for the development and study of electric vehicles in the future.

1. Habituation is the declining responsiveness of a neural circuit (or behavior) to repetitive stimulation. Dishabituation (or arousal) can be brought about by the sudden presentation of an additional, novel stimulus. A clear example of arousal in the locust is provided by the visual system: the habituated response of the descending contralateral movement detector (DCMD) interneuron to repetitive visual stimuli can be dishabituated by a variety of other visual and tactile stimuli. 2. Application of octopamine to the locust brain and optic lobes dishabituates the DCMD in a manner similar to the effect of visual and tactile stimulation. 3. The locust CNS contains two pairs of octopamine-immunoreactive cells, the protocerebral medulla 4 (PM4) neurons, that could potentially mediate this dishabituation effect; PM4 neurons arborize in the optic lobe, they contain octopamine, and they respond to the same visual and tactile stimuli that dishabituate the DCMD. 4. To investigate whether PM4 activity dishabituates the DCMD, we recorded intracellularly from one of the PM4 neurons while recording extracellularly from the DCMD. When the PM4 neuron is injected with hyperpolarizing current to render it completely inactive, the DCMD exhibits its characteristic habituation to a moving visual stimulus. However, depolarizing the PM4 neuron, to produce action potentials at approximately 20 Hz, significantly increases the number of DCMD action potentials per stimulus. 5. The PM4 neurons may therefore play an important role in dishabituating the DCMD to novel stimuli. This effect is presumably mediated by PM4 neurons releasing endogenous octopamine within the optic lobe.

A double cleavage drilled compression (DCDC) specimen was analysed using the boundary element method. The DCDC specimen of FRP composites was proposed and used to evaluate the interfacial behaviour and fracture energy in model composites of carbon fibre reinforced polymer (CFRP). As a result, it was confirmed that the DCDC specimen had the mode I stress distribution for the hole offsetting displacement of b = 0, in which the specimen configuration was symmetrical, while it had the mixed-mode stress distribution for b = 0. The approach of calculating the interfacial fracture energy was established using both the analysis and experiments. Furthermore, the bridging fibre DCDC specimen was proposed and the effect of bridging fibres on the loading phase was made clear. It was shown that the DCDC test was a useful method to evaluate the interfacial behaviour of fibre/matrix in composites.

An integrated membrane distillation (MD) flowsheet, consisting of direct contact membrane distillation (DCMD) and vacuum membrane distillation (VMD) units, was proposed and analysed in terms of thermal performance and water recovery factor, for the first time. The same lab-scale membrane module (40 cm2) was used for carrying out experiments of DCMD and VMD at fixed feed operating conditions (deionised water at 230 L/h and ~40 °C) while working at the permeate side with deionised water at 18 °C and with a vacuum of 20 mbar for the DCMD and the VMD configuration, respectively. Based on experimental data obtained on the single modules, calculations of the permeate production, the specific thermal energy consumption (STEC) and the gained output ratio (GOR) were carried out for both single and integrated units. Moreover, the calculations were also made for a flow sheet consisting of two DCMD units in series, representing the “traditional” way in which more units of the same MD configuration are combined to enhance the water recovery factor. A significant improvement of the thermal performance (lower STEC and higher GOR) was obtained with the integrated DCMD–VMD flowsheet with respect to the DCMD units operating in series. The integration of DCMD with VMD also led to a higher permeate production and productivity/size (PS) ratio, a metric defined to compare plants in terms of the process intensification strategy.

The demand for organs for kidney transplantation (KTX) compels the use of high-risk donation after circulatory death donors (DCDs) and extended criteria donors (ECDs). Many deceased donors receive prehospital CPR, but the literature does not address CPR as a benefit to graft survival. We hypothesized that donor prehospital CPR correlates with improved graft survival with high-risk DCD/ECD kidneys. We retrospectively analyzed KTX recipients and their donor data from 2008 to 2013. A total of 646 cadaveric donors (498 SCDs, 55 DCDs, and 93 ECDs) facilitated 910 KTX. There were 223 KTX performed from 148 high-risk DCDs/ECDs (31 with CPR and 117 without CPR). The mean age of high-risk DCDs/ECDs with CPR was 44.94 versus 53.45 years without CPR (P = 0.005). The recipients of high-risk DCDs/ECDs revealed no significant difference in body mass index, length of stay, discharge Cr, CIT, or DGF with and without CPR. Graft survival at three years was significant with 0/50 failures from high-risk DCDs/ECDs with CPR versus 16/173 without CPR (P = 0.026). Our findings are limited as a single-center retrospective study; however, the result of significant three-year graft survival in high-risk DCDs/ECDs with CPR suggests that prehospital donor CPR should be further investigated for its contribution to the relative quality of the donor.

Numerous studies have revealed that the sparse spatiotemporal distributions of ground-level PM2.5 measurements affect the accuracy of PM2.5 simulation, especially in large geographical regions. However, the high precision and stability of ground-level PM2.5 measurements make their role irreplaceable in PM2.5 simulations. This article applies a dynamically constrained interpolation methodology (DCIM) to evaluate sparse PM2.5 measurements captured at scattered monitoring sites for national-scale PM2.5 simulations and spatial distributions. The DCIM takes a PM2.5 transport model as a dynamic constraint and provides the characteristics of the spatiotemporal variations of key model parameters using the adjoint method to improve the accuracy of PM2.5 simulations. From the perspective of interpolation accuracy and effect, kriging interpolation and orthogonal polynomial fitting using Chebyshev basis functions (COPF), which have been proved to have high PM2.5 simulation accuracy, were adopted to make a comparative assessment of DCIM performance and accuracy. Results of the cross validation confirm the feasibility of the DCIM. A comparison between the final interpolated values and observations show that the DCIM is better for national-scale simulations than kriging or COPF. Furthermore, the DCIM presents smoother spatially interpolated distributions of the PM2.5 simulations with smaller simulation errors than the other two methods. Admittedly, the sparse PM2.5 measurements in a highly polluted region have a certain degree of influence on the interpolated distribution accuracy and rationality. To some extent, adding the right amount of observations can improve the effectiveness of the DCIM around existing monitoring sites. Compared with the kriging interpolation and COPF, the results show that the DCIM used in this study would be more helpful for providing reasonable information for monitoring PM2.5 pollution in China.

H3K9 methylation directs heterochromatin formation by recruiting multiple heterochromatin protein 1 (HP1)-containing complexes that deacetylate histones and methylate cytosine bases in DNA. In Neurospora crassa, a single H3K9 methyltransferase complex, called the DIM-5,-7,-9, CUL4, DDB1 Complex (DCDC), is required for normal growth and development. DCDC-deficient mutants are hypersensitive to the genotoxic agent methyl methanesulfonate (MMS), but the molecular basis of genotoxic stress is unclear. We found that both the MMS sensitivity and growth phenotypes of DCDC-deficient strains are suppressed by mutation of embryonic ectoderm development or Su-(var)3-9; E(z); Trithorax (set)-7, encoding components of the H3K27 methyltransferase Polycomb repressive complex-2 (PRC2). Trimethylated histone H3K27 (H3K27me3) undergoes genome-wide redistribution to constitutive heterochromatin in DCDC- or HP1-deficient mutants, and introduction of an H3K27 missense mutation is sufficient to rescue phenotypes of DCDC-deficient strains. Accumulation of H3K27me3 in heterochromatin does not compensate for silencing; rather, strains deficient for both DCDC and PRC2 exhibit synthetic sensitivity to the topoisomerase I inhibitor Camptothecin and accumulate γH2A at heterochromatin. Together, these data suggest that PRC2 modulates the response to genotoxic stress.

OBJECTIVES The purposes of the current study were to examine the construct and predictive validity, and reliability of the developmental coordination disorder questionnaire’ 07 (DCDQ’ 07) for young children with developmental disabilities (DD).METHODS 135 parents of young children with DD completed an online questionnaire which consisted of demographic information, the DCDQ’ 07, and a physical activity behavior questionnaire. A one-way analysis of variance was conducted to evaluate motor difficulties across different types of young children with DD. Exploratory factor analysis and Cronbach’s alpha were used to examine construct validity and reliability of the DCDQ’ 07. Moreover, hierarchical multiple regression analysis was used to examine the association between the DCDQ’ 07 scores and physical activity behaviors in young children with DD.RESULTS The one-way analysis of variance indicated that the DCDQ’ 07 scores were significantly different across different types of young children with DD = 6.66, p <0.001). The exploratory factor analysis revealed a three-factor solution, accounting for 64.34% of the variance of the DCDQ’ 07. Internal consistencies of the three factors were acceptable. One factor, control during movement, was a significant predictor for physical activity behaviors in young children with DD (β = 0.37, p < 0.01).CONCLUSIONS The DCDQ’ 07 may be a reliable and valid measure in examining motor difficulties in young children with DD. To promote physical activity behaviors in young children with DD, teaching and reinforcing motor skills need to be encouraged.

In recent years, considerable attention has been focused on the development of new composite materials for application as drug delivery systems. In this field, calcium phosphate cements (CPCs) are often employed as support to delivery of drugs, but their behavior has some drawback related to the so-called burst effect. The aim of this work was to develop new CPCs formulations from synthesized tetracalcium phosphate (TTCP), dicalcium phosphate anhydrous TTCP and drug-containing hybrid microparticles (DCHM). The main function of these DCHM is providing nuclei of high concentration of drugs into the CPCs. The DCHM were synthesized via the sol-gel method from a bridged precursor of the type (H3CO)3 – Bridge – (OCH3)3 and aspirin (AS) as model drug. The inorganic polycondensation reached 89.5 % as calculated by 29Si NMR. The analysis by small angle X-ray scattering (SAXS) reveled a short range structural ordering in the DCHM at molecular level. Effective incorporation of AS inside the microspheres was detected by FTIR spectroscopy. In vitro tests of DCHM according to ISO 10993-5 revealed non-cytotoxic behavior. Four CPCs formulations containing 0, 1, 5 and 10 wt % of DCHM, were evaluated. The presence of DCHM did not modify neither the degree of conversion to low-crystallinity HA nor the measured setting times of the CPCs, however, the amount of incorporated microparticles considerably affected the degree of porosity (macropores of 200 µm) and interconnectivity of the cement matrix.

The Movement Assessment Battery for Children, 2nd Ed (MABC-2), comprised of motor tests and a questionnaire (Checklist), has been used by several nationalities to identify problems in children's motor behavior. However, the level of agreement between the motor tests and the checklist has been questioned. So, this study aimed to test the level of competition between the MABC-2 motor test and MABC-2 Checklist, controlled by the Developmental Coordination Disorder Questionnaire (DCDQ-BR) as the gold reference. Parents of 40 children and 16 teachers responded to the MABC-2 Checklist and the DCDQ-BR. Later 40 children were evaluated using the MABC-2 motor test. No significant congruence among a comparison of the MABC-2 motor tests, MABC-2 Checklist and DCDQ-BR was observed. The results indicated that there is no concurrent validity between the MABC-2 motor tests and the DCDQ-BR test, whereas the MABC-2 Checklist showed low levels of concurrent validity with the DCDQ-BR.

Background. Parent-report measures, such as the Developmental Coordination Disorder Questionnaire'07 (DCDQ'07), are used to identify developmental coordination disorder (DCD) in children. Early identification of this condition is important to mitigate its social-emotional and health consequences. Unfortunately, few French-language assessments are available to therapists working with francophone populations. Purpose. The aim of this study was to undertake a formal translation of the English DCDQ'07 and begin to examine its psychometric properties. Methods. The translation was done using Beaton, Bombardier, and Guillemin's (2000) guidelines for cross-cultural adaptation. Methodologies described by Haccoun (1987) and Vallerand (1989) were used to address the psychometric qualities of the translation. Findings. The DCDQ'07 and its French translation (DCDQ-FC) are equivalent, with excellent internal consistency and test-retest reliability. Concurrent and construct validity were adequate for a screening measure; however, low sensitivity was obtained with both measures. Implications. The DCDQ-FC is a valid translation for use with a French Canadian population.

Abstract Carmine is a kind of colorant which is widely used in food, beverage, medicine, cosmetics and tobacco industry. However, excessive use of carmine may lead to the risks of carcinogenic, teratogenic and mutagenic, which seriously threaten the health and safety of consumers. In this paper, DCDR technology is utilized to develop a quantitative method for the detection of carmine, which requires only a small volume deposition of analyte solution (several μL) on a suitable hydrophobic substrate. The conventional Raman spectrum of carmine aqueous solution and corresponding Raman spectrum using DCDR method were compared, illustrating a much higher sensitivity for DCDR method. The Raman spectra of carmine aqueous solution with different concentrations of 100, 50, 10, 8, 4 and 2μg/mL are acquired from the spots on the “coffee-ring” with DCDR method. Using DCDR method, a good linear relationship has been observed between the intensities of the two characteristic peaks, 1364cm−1 as well as 1572cm−1, and the concentrations of the solution, with the linear correlation coefficient of R2>0.99. The results illustrate that DCDR method has a good potential in the quantitative analysis of colorant like carmine, providing a promising technique for a rapid detection for food additives.

The Developmental Coordination Disorder Questionnaire (DCDQ) is a widely used and well-validated tool that contributes to the diagnosis of Developmental Coordination Disorder (DCD). The aim of this study was to further analyze the psychometric properties of the European Spanish cross-culturally adapted version of the Developmental Coordination Disorder Questionnaire (DCDQ-ES) in a sample of Spanish children aged 6–11 years and to establish reference norms with respect to age groups. Parents of 540 typically developing children completed the DCDQ-ES. A second sample of 30 children with probable DCD (pDCD) was used to test its discriminant validity. Confirmatory factor analysis supported the original three-factor structure and the internal consistency was excellent (Cronbach’s α = 0.907). Significant differences between age groups were found. The pDCD group scored significantly lower than the reference sample in the three subscales and DCDQ-ES total score (p < 0.001; AUC = 0.872). The DCDQ-ES is a reliable and valid tool for screening motor coordination difficulties in Spanish children and for identifying children with probable DCD. The findings of this research suggest that context-specific cut-off scores should be systematically utilized when using cross-cultural adaptations of the DCDQ. Age-specific cut-off scores for Spanish children are provided.

Motion dazzle describes high-contrast patterns (e.g. zigzags on snakes and dazzle paint on World War I ships) that do not conceal an object, but inhibit an observer's perception of its motion. However, there is limited evidence for this phenomenon. Locusts have a pair of descending contralateral movement detector (DCMD) neurons which respond to predator-like looming objects and trigger escape responses. Within the network providing input to a DCMD, separate channels are excited when moving edges cause areas of the visual field to brighten or darken, respectively, and these stimuli interact antagonistically. When a looming square has an upper half and lower half that are both darker than background, it elicits a stronger DCMD response than the upper half does alone. However, when a looming square has a darker-than-background upper half and a brighter-than-background lower half, it elicits a weaker DCMD response than its upper half does alone. This effect allows high-contrast patterns to weaken and delay DCMD response parameters implicated in escape decisions, and is analogous to motion dazzle. However, the motion dazzle effect does not provide the best means of motion camouflage, because uniform bright squares, or low-contrast squares, elicit weaker DCMD responses than high-contrast, half dark, half bright squares.

Background: Advantages of digital clock drawing metrics for dementia subtype classification needs examination. Objective: To assess how well kinematic, time-based, and visuospatial features extracted from the digital Clock Drawing Test (dCDT) can classify a combined group of Alzheimer’s disease/Vascular Dementia patients versus healthy controls (HC), and classify dementia patients with Alzheimer’s disease (AD) versus vascular dementia (VaD). Methods: Healthy, community-dwelling control participants (n = 175), patients diagnosed clinically with Alzheimer’s disease (n = 29), and vascular dementia (n = 27) completed the dCDT to command and copy clock drawing conditions. Thirty-seven dCDT command and 37 copy dCDT features were extracted and used with Random Forest classification models. Results: When HC participants were compared to participants with dementia, optimal area under the curve was achieved using models that combined both command and copy dCDT features (AUC = 91.52%). Similarly, when AD versus VaD participants were compared, optimal area under the curve was, achieved with models that combined both command and copy features (AUC = 76.94%). Subsequent follow-up analyses of a corpus of 10 variables of interest determined using a Gini Index found that groups could be dissociated based on kinematic, time-based, and visuospatial features. Conclusion: The dCDT is able to operationally define graphomotor output that cannot be measured using traditional paper and pencil test administration in older health controls and participants with dementia. These data suggest that kinematic, time-based, and visuospatial behavior obtained using the dCDT may provide additional neurocognitive biomarkers that may be able to identify and tract dementia syndromes.

Direct contact membrane distillation (DCMD) is a membrane distillation (MD) configuration where feed and distillate directly contact with a hydrophobic membrane. Depending on its operating conditions, the hydraulic pressures of the feed and distillate may be different, leading to adverse effects on the performance of the DCMD process. Nevertheless, little information is available on how hydraulic pressure affects the efficiency of DCMD. Accordingly, this paper investigates the effect of external hydraulic pressure on the process efficiency of DCMD. Gas permeabilities of MD membranes were measured to analyze the effect of membrane compaction by external pressure. Mass transfer coefficients were calculated using experimental data to quantitatively explain the pressure effect. Experiments were also carried out using a laboratory-scale DCMD set-up. After applying the pressure, the cross-sections and surfaces of the membranes were examined using a scanning electron microscope (SEM). Results showed that the membrane structural parameters such as porosity and thickness were changed under relatively high pressure conditions (>30 kPa), leading to reduction in flux. The mass transfer coefficients were also significantly influenced by the hydraulic pressure. Moreover, local wetting of the membranes were observed even below the liquid entry pressure (LEP), which decreased the rejection of salts. These results suggest that the control of hydraulic pressure is important for efficient operation of DCMD process.

Abstract AIM: RIP2 is a critical mediator of NLRs signaling for innate immune activation. GEF-H1 interacts with NLRs and is responsible for initiating tyrosine phosphorylation of RIP2 required for the activation of NF-κB. We identified DCAM-253, 2-dialkylamino-9-indazolyl-purine, as a potent small molecule tyrosine kinase inhibitor of Rip2. Results: Gene specific siRNA mediated knockdown and expression of signaling intermediates in macrophages, and HEK293 cells was used to determine the mechanisms by which DCAM-253 inhibits Rip2 mediated induction of cytokine expression. DCAM-253 prevented the recruitment of GEF-H1 to the Rip2/NLR signaling complexes and subsequent tyrosine phosphorylation of Rip2. DCAM-253 inhibited TNFα production induced by GEF-H1 dependent NLR activation. DCAM-253 was able to protect mice from intestinal inflammation induced by epithelial barrier disruption and microbial defense activation in the DSS-induced colitis model. Contol group Mice presented with transmural leukocyte infiltration and crypt damage with tissue destruction indicating severe persistent colitis. In contrast, low cellular infiltration and intact intestinal epithelium were observed in the colon sections of mice treated with 2 oral doses of DCAM-253. Conclusion: We have identified a new small molecule inhibitor, which protects mice from colitis and promotes mucosal recovery by preventing Rip2 tyrosine phosphorylation upon intestinal barrier disruption and subsequent innate immune activation.

Abstract Introduction: Desisting from disease directed treatment in the past weeks of life is a quality criterion in oncology service. Patients with advanced cancer have unrealistic expectations from chemotherapy and hold on to it as a great source of hope. Many oncologists continue futile and unnecessary treatments, instead of conveying to the patients the lack of benefit, resulting in delayed referral for palliative care (PC). Materials and Methods: This is a retrospective analysis of case records from June 2014 to December 2015. The primary objective was to study, how far back in time terminally ill cancer patients received definitive cancer directed therapy (DCDT). Apart from patient demographics, the diagnosis, stage, and details of DCDT, and death were captured. PC referral data were recorded. DCDT to death was taken as treatment-free interval (TFI). Analysis was performed using IBM SPSS Statistics for Windows, Version 20. Results: A total of 292 case records were evaluated. Seventy-three had inadequate treatment details. Hence, 219 records were analyzed. PC referral was done in 78.5% of patients. Only best supportive care (BSC) without any DCDT was given in 27 patients. The most common reason for BSC was a poor performance status in 92.5%. The median time from PC referral till death was 43.5 days (range: 1–518 days). Chemotherapy was the most common DCDT in 52.9% of patients. The median time from DCDT and death was 49 days (range: 0–359 days). Cervical and ovarian cancers patients had the longest TFI ; shortest in unknown primary. Most patients died at home (70.4%). Patients receiving PC preferred home or hospice as place of death. Of the 80 patients given hospice care, 39 (36.5%) died in the hospice. Conclusion: While DCDT needs to be started at the right time, it should also be discontinued when futile. Early involvement of the PC team, even while patients are on DCDT makes the transition smoother and more meaningful.
