Анотація:

Between 1790 and 1794, Spanish and English expeditions to Nootka Bay reported two very different accounts of cannibalistic activities among the Nuu-cha-nulth. The British vividly described cannibalistic practices, whereas the Spanish observers claimed that the Nootka no longer practiced cannibalism. Many historians and anthropologists use these reports to debate the existence or absence of cannibalism among native peoples in the Pacific Northwest. This article argues that the differences reveal more about European concepts of race, culture, and society than whether cannibalism existed on the Northwest Coast. These differences capture the transition from past interpretations to Enlightenment ideals juxtaposed with national interests, highlighting how Spain and England saw their role in shaping the history of humanity.

Abstract Soil is a collection of minerals, organic matter and relatively loose sediments that are located above the bedrock. Based on the particle size, it is divided into four types, namely gravel, sand, silt and clay. The role of soil is very important in a construction because the soil functions as a support for the loads that are above it, so that the soil is expected to have a good bearing capacity. In this study, the soil taken is soft soil in Hambalang, Bogor where the soil conditions in the area are hard in the dry season and soft in the rainy season, so it is necessary to review the CBR (California Bearing Ratio) value. In this study, using waste from red brick as an additive to soft soil. CBR testing is appropriate (SNI 1744: 2012), with the CBR Laboratory test soaked and unsoaked. Along with the addition of% red brick powder, the CBR values of soaked and unsoaked (top and bottom) have increased, so that the addition of% content of red brick powder is 15% respectively got a value of 4.55% and 4.9% (soaked) and 17.94% and 19.70 (unsoaked). So, from this CBR test, it was found that the effect of adding red brick powder increases CBR tilapia soaked and unsoaked soft soil of Hambalang. Анотація:

Deux constats sont à l’origine de cet article : le sens attribué dans la vie courante en Haïti aux termes « nègre », « africain » et « citoyen », ensuite la distinction entre citoyens et Africains observée dans certains textes relatifs à l’abolition de l’esclavage en 1793 et 1794. L’auteur relève l’usage du vocable africain par les acteurs qui ont dominé la scène politique sous la révolution haïtienne. Il démontre que leur emploi de ce terme soulève un problème important, relatif au statut social, à l’identité et à la nationalité, pas seulement des anciens esclaves, mais de tous les indigènes. Les chefs mulâtres ne revendiquent aucune origine africaine et désignent les cultivateurs sous le vocable « africain », considéré comme infamant. Les leaders noirs se taisent sur leur origine africaine et sont aussi impitoyables que les chefs mulâtres envers les cultivateurs. Ces dirigeants perpétuent une image négative de l’Africain. Le courant indigéniste a essayé, au début du XXe siècle, de réhabiliter l’image de l’Africain aux yeux de l’Haïtien, mais sa reconstruction de l’histoire n’est pas scientifique, car il est impossible de faire des leaders noirs de la révolution des représentants des cultivateurs.

In 1822, Robert Wilmot, the undersecretary of state for the colonies, introduced a bill to unite Upper and Lower Canada in the British House of Commons. In doing so, he was proposing not just an intercolonial union but a new constitution. He believed that because the Canadas’ constitution was granted by an Act of Parliament in 1791 it could be changed by Parliament without colonial consultation or consent. Whig parliamentarians and colonists contested this interpretation and raised questions about consent and the status of colonial constitutions. These debates in the 1820s reveal just how muddied thinking about colonial constitutions and consultation had become. Lower Canadian opposition to the bill has received significant attention from historians, yet this attempt at constitutional change also forced Upper Canadians to take unexpected positions, including interpreting the 1791 act as a written charter. Through these transatlantic debates, they were continuing to fashion a settler interpretation of British constitutionalism. This article traces these ideas by examining the movement of news and rumours through emerging, intertwined colonial and imperial public spheres, which illustrate the permeability of the line between public and private information. Inserting this failed constitution-writing effort into the longer history of Canadian constitutions sheds light on the limits to imperial intervention and encourages a broader rethinking of Canadian constitutional histories and the role of empire in a long nineteenth century.

The article presents a brief overview of the main milestones in the biography of Fyodor Ivanovich Erdmann, also known as Friedrich Franz Ludwig Erdmann (1795-1862). The authors pay special attention to the main stages of his pedagogical and scholarly pursuits at Kazan University (1818-1845). After Chr.D. Frähn (1782-1851), Erdmann became the second visiting German professor of Oriental literature at the university and one of the last students of the famous German orientalist O.G. Tychsen (1734-1815). In 1819, at the age of 24, F.I. Erdmann moved to Kazan, Russia. During the first half of the XIX century, the invitation of representatives of European universities and various peoples of the Russian Empire to the city became an important academic and humanistic tradition that contributed to the development of Oriental studies at Kazan University. Following his dismissal from the university on May 11, 1845, Professor F.I. Erdmann was appointed Director of schools in Olonets region on February 7, 1847. Two months later, on April 8, 1847, Erdmann was transferred to the post of Director of schools in Novgorod region, a post which he held until his dismissal from service on August 20, 1855. Upon his death on November 14, 1862, the academic was buried in Kazan.

Fort peu y restèrent. En 1805, ou encore en 1820, la ville comptait environ 5 000 citadins étrangers. Quelques-uns devinrent citoyens —1806, dont trois femmes — entre 1792 et 1820 (tableau 1). Les problèmes posés à la ville étaient donc d'ordre institutionnel, financier et surtout politique, ce dernier point devenant bien plus difficile à résoudre que par le passé car, si le sujet britannique jurait allégeance à la personne du roi, le citoyen des États-Unis, lui, prêtait serment envers un corps politique corporatif et souverain dont il était membre. The trophic ecology (diet and head morphology) and migration patterns of two closely related salmonid fishes, Arctic char (Salvelinus alpinus (L., 1758)) and Dolly Varden (Salvelinus malma (Walbaum, 1792)), were examined in tributaries of Lake Aleknagik, southwestern Alaska, to test for differentiation between species. Schoener’s index of proportional overlap and multivariate analyses of diets suggested that these species had significantly different trophic niches. Arctic char and the largest individuals of both species had the most diverse diets, and sockeye salmon (Oncorhynchus nerka (Walbaum, 1792)) eggs dominated the diet of both species, especially Arctic char. Arctic char had larger jaws and wider heads than Dolly Varden of similar body length, which may contribute to interspecific diet difference. The species also differed in migration patterns; otolith microchemistry indicated that juvenile Arctic char were produced by nonanadromous mothers, whereas the mothers of the Dolly Varden had been to sea in the season prior to spawning. The species also segregate in spawning habitat (Arctic char in the lakes and Dolly Varden in streams), as well as in juvenile rearing habitat. Our study provides the first evidence of divergent feeding and migratory ecology between sympatric juvenile Arctic char and Dolly Varden, differences that may constrain hybridization and introgression between them.

The rainbow trout (Oncorhynchus mykiss (Walbaum, 1792)) is one of many salmonid species exhibiting a gradient of life histories including fluvial (stream-resident), anadromous (ocean-migrant), and adfluvial (lake-migrant) forms, the last of which is less extensively studied than the other two. Our goal was to determine the extent of diet segregation between fluvial and adfluvial rainbow trout. We collected stomach content and stable isotope data on rainbow trout sampled in stream and lake habitats in a southwestern Alaska watershed during summer and compared them with data on sympatric stream- and lake-specialist char species, Dolly Varden (Salvelinus malma (Walbaum in Artedi, 1792)) and Arctic char (Salvelinus alpinus (Linnaeus, 1758)), respectively. Rainbow trout in streams fed largely on aquatic insects, while those in the lake ate primarily benthic snails and amphipods. The trophic segregation of stream-resident and lake-migrant rainbow trout mirrored but was less extreme than the divergence of lotic Dolly Varden and lentic Arctic char in the same system. Spawning sockeye salmon (Oncorhynchus nerka (Walbaum in Artedi, 1792)) provided a nutrient subsidy in the form of eggs that supported rainbow trout in both stream and lake (littoral) habitats, causing their isotopic signatures to converge. This study augments knowledge of partial migration and trophic divergence within populations.

Il presente contributo si propone di indagare i modelli, le fonti e le forme di una parte della produzione poetica della livornese Fortunata Sulgher Fantastici, detta Temira Parraside in Arcadia. Dopo aver tracciato i contorni del contesto culturale nel quale si inserisce l’acclamata improvvisatrice, si propone una piccola rassegna dei testi di soggetto omerico e virgiliano nei suoi Componimenti poetici del 1785, poi ripubblicati in Poesie nel 1794. Si procederà dunque con l’analisi dei componimenti: attraverso i personaggi di Andromaca, Briseide, Achille, Penelope e Didone, si tesse una trama di ritorni al passato risultante da un uso sapiente e incrociato dei modelli, che rendono il discorso lirico di Sulgher perfettamente coerente con la cultura del suo tempo.

Abstract Die seelischen Folgen aus der unglücklichen Dreiecksgeschichte in Wetzlar umschreibt Goethe rückblickend als ,historia morbi‘ oder ,taedium vitae‘. Seine eigene Krankengeschichte verbindet er im Werther mit anderen Fällen. Nie zuvor hat ein Ich so rückhaltlos von sich selbst erzählt, ohne den Autor dabei völlig preiszugeben. Nicht der Held, sondern das Büchlein empfiehlt sich als ein Freund, der das Publikum über den Menschen aufklärt. Tragische Leidenschaft und Selbstmord wären bloß schrecklich und kein Fundament für Freundschaft, die daraus kausal entwickelte Geschichte hingegen ist fesselnd und erhellend. Die Innovation des Werther besteht in der Suggestion von Authentizität, erstens durch Versatzstücke aus der Wirklichkeit und zweitens durch Darstellung einer lebendig dramatisierten, ,inneren Geschichte‘ aus der Ich-Perspektive. Das empfiehlt den Roman für mehrere Bühnenadaptionen, von denen hier Willers ,,bürgerliches Trauerspiel“ von 1778 betrachtet wird. Goethes neuartige Darstellungsverfahren machen den Werther zum Kultbuch, das eine erste moderne ,,Ichzeit“ einleitet – noch vor M. Biller, R. Goetz, H. Hegemann, W. Herrndorf oder Chr. Kracht.In retrospect, Goethe describes the emotional consequences of the unfortunate love triangle in Wetzlar as ‘historia morbi’ or ‘taedium vitae’. In Werther, he connects his own medical history with other cases. Never before has an ego spoken so wholeheartedly about itself without revealing the author completely. Not the hero, but the little book is recommended as a “friend” who enlightens the public about the nature of man. Tragic passion and suicide would be merely horrific and not the foundation of friendship, but the reasonably arranged story is compelling and enlightening. Werther’s innovation consists in the suggestion of authenticity, firstly through set pieces from reality and secondly through the depiction of a vividly dramatized ‘inner story’ from the first-person perspective. This recommends the novel for several stage adaptations, of which Willer’s “bourgeois tragedy” from 1778 is considered here. Goethe’s innovative methods of presentation make Werther a cult book, introducing a first modern “ego time” before M. Biller, R. Goetz, W. Herrndorf or Chr. Kracht.

We analyzed the effect of total fertility rate (TFR) and crude birth rate (CBR) on the number of males per 100 females at birth, also called the secondary sex ratio (SR), and on the twinning rate (TWR). Earlier studies have noted regional variations in TWR and racial differences in the SR. Statistical analyses have shown that comparisons between SRs demand large data sets because random fluctuations in moderate data are marked. Consequently, reliable results presuppose national birth data. Here, we analyzed historical demographic data and their regional variations between counties in Sweden. We built spatial models for the TFR in 1860 and the CBR in 1751–1870, and as regressors we used geographical coordinates for the provincial capitals of the counties. For both variables, we obtained significant spatial variations, albeit of different patterns and power. The SR among the live-born in 1749–1869 and the TWR in 1751–1860 showed slight spatial variations. The influence of CBR and TFR on the SR and TWR was examined and statistical significant effects were found.

Abstract Although CD19-targeting CAR T cell therapy has shown remarkable success against B cell malignancies there is still room for improvement. To improve long-term responses amongst lymphoma patients, mechanisms underlying initial response and tumor relapses must be addressed. Recent studies have highlighted the importance of the quality of the starting cell material for CAR T cell production and the composition of the CAR T cell infusion product for clinical response. In our study, we have evaluated the clinical response to third generation anti-CD19 CAR T cell therapy in 23 patients with B cell lymphoma. Further, we evaluated the CAR T cell infusion products using targeted single-cell RNA sequencing and multi-color flow cytometry to find potential T cell subsets associating with clinical response. Patients were divided into responders (CR + PR) and non-responders (SD + PD) at the 1-month follow-up after treatment. The CAR T cell products of both responding and non-responding patients were dominated by CD8+ CAR T cells. When comparing cells from the two patient groups we found that the presence of effector-like CD8+ CAR T cells with a high cytotoxic and cytokine secretion and low dysfunction gene expression profile were associated with clinical response. Furthermore, CD8+ CAR T cells of responders displayed a higher polyfunctional gene expression profile, which has previously been shown to associate with clinical response. In contrast, CD8+ CAR T cells from the infusion products of non-responders displayed an elevated dysfunction gene expression profile. Our study reveals that single-cell RNA expression analysis of individual CAR T cell infusion product can identify subsets of effector cells correlating with clinical response. This can be used as a guidance in the CAR T cell production processes, to obtain CAR T cells with the desired characteristics. Tujuan dari penelitian ini adalah untuk mengetahui peningkatan nilai CBR tanah lempung dengan variasi pencampuran Serbuk Cangkang Kerang. Jenis tanah yang digunakan pada penelitian ini adalah tanah lempung yang diambil dari Jalan Gunung Sari, Kota Pekanbaru, Riau. Penelitian ini menggunakan serbuk cangkang kerang sebagai bahan stabilisasi tanah dengan persentase variasi campuran 0%, 25%, 30%, dan 35%. Metode yang digunakan dalam penelitian ini metode eksperimental yaitu menggunakan pengujian CBR laboratorium yang mengacu pada peraturan SNI-1744-2012. Dari hasil pengujian CBR laboratorium, variasi campuran serbuk cangkang kerang 0% diperoleh nilai CBR sebesar 1,40%, variasi serbuk cangkang kerang 25% diperoleh nilai CBR sebesar 4,20%, variasi serbuk cangkang kerang 30% diperoleh nilai CBR sebesar 7,10%, dan variasi serbuk cangkang kerang 35% diperoleh nilai CBR sebesar 6,80%. Nilai CBR maksimum terjadi pada variasi campuran serbuk cangkang kerang 30% sebesar 7,10%. Kesimpulan yang didapat adalah terjadinya peningkatan nilai CBR tanah yang distabilisasi dengan serbuk cangkang kerang sebesar 5,70% dengan nilai CBR 1,40% menjadi 7,10%.

O artigo tem como objetivo compreender os elementos que caracterizaram a postura do protestantismo norte-americano em relação aos divertimentos e à educação do corpo. Apesar de existirem muitas obras sobre Benjamin Franklin poucas exploraram a relação das suas contribuições com uma prescrição sobre os divertimentos e educação do corpo. As fontes utilizadas foram legislações da Pensilvânia e os escritos de Franklin. As análises evidenciam que, se por um lado as leis constituíam um quadro de condenação da ociosidade e dos divertimentos, por outro, os escritos trazem observações favoráveis a atividades como o xadrez e práticas físicas, justificando estes divertimentos a partir de uma moral utilitarista.

Abstract Chimeric antigen receptor (CAR)-T therapy has not shown significant therapeutic effects on brain tumors. As our data with a syngeneic mouse model of CAR therapy showed significant hypoxia in the brain tumor microenvironment, we sought to develop a novel strategy to improve the metabolic state of the CAR-T cells in the hypoxic tumor microenvironment. In mice bearing intracerebral SB28-EGFRvIII gliomas receiving an intravenous infusion of EGFR vIII-targeting CAR-T cells, we found mitochondrial ATP production in tumor-infiltrating CAR-T cells was significantly impaired compared to those migrating to the spleen. Therefore, we hypothesized that treating CAR-T cells with metabolic regulators that activate mitochondria before infusion would improve the therapeutic effect of CAR-T cells. Using a hypoxic incubator to mimic the tumor microenvironment of brain tumors, we screened drugs that would improve the metabolic state of CAR-T cells and maintain their anti-tumor effect. Although the mammalian target of rapamycin (mTOR) inhibitor rapamycin and AMP-activated protein kinase (AMPK) activator metformin are ineffective when treated as single drugs, a combination of these drugs (met+rap) prolonged the anti-glioma effect of CAR-T cells under hypoxic conditions. Furthermore, Seahorse assays revealed a higher mitochondrial spare respiratory capacity of met+rap-treated CAR-T cells compared to single drug-treated CAR-T cells or those without pre-treatment. A single intravenous infusion of CAR-T cells pre-treated with met+rap significantly prolonged the survival of mice bearing intracerebral SB28-EGRFvIII tumors compared with CAR-T cells without the pre-treatment. In mass cytometric analyses, a significantly higher number of glioma-infiltrating CAR-T cells were found in mice that received met+rap-treated CAR-T cells compared with mice that received control CAR-T cells. Interestingly, the glioma tissues infiltrated by met+rap pre-treated CAR-T cells were infiltrated by significantly fewer Ly6c+ CD11b+ monocytic myeloid-derived suppressor cells. As a major translational significance, because the CAR-T cells were pre-treated with the drugs during the in vitro expansion with no need for in vivo drug administrations, our strategy would circumvent any toxicity associated with in vivo treatments with the same medications. These data warrant a translational and clinical evaluation of met+rap pre-treated CAR-T cells in human settings and the development of a CAR-T cell trial using this approach. Abstract Background Chimeric antigen receptor T cells (CAR T) are potential therapies for rhabdosarcoma (RMS), the most common soft tissue sarcoma in children, where patients with relapsed or refractory disease have a dismal cure rate, and effective therapies are urgently needed. RMS tumors express high levels of the cell surface receptors FGFR4 and CD276, and both are direct targets of the PAX3-FOXO1 chimeric oncogene. However, CAR T have shown poor performance in solid tumors due to T cell exhaustion, limited persistence, and heterogeneous expression of target antigens. We hypothesize that engineering CAR constructs targeting FGFR4 and CD276 will enhance their activity and dual targeting will overcome the heterogenous target expression to effectively eliminate RMS tumors. Methods We modified and tested different hinge and transmembrane domains (HTM) or intracellular domain (ICD) of FGFR4 CAR constructs and developed multiple dual targeting Bicistronic CARs (BiCisCARs) against FGFR4 and CD276. We tested these CARs in aggressive mouse models and performed Cellular Indexing of Transcriptomes and Epitopes by Sequencing (CITE-seq) of tumor infiltrating CAR T cells to functionally characterize these constructs. Results FGFR4 targeting CAR construct with CD8HTM and 4-1BB ICD could eliminate low burden RMS but was less effective against orthotopic tumors. Replacing CD8HTM with CD28HTM improved the efficacy in a moderate burden RMS model but did not show any benefit against a more aggressive RMS559 orthotopic model. Further modification using the CD28 ICD significantly enhanced the activity in a large burden RMS model but was unable to eradicate all tumors. Similarly, CAR T cells targeting CD276 alone, showed significant anti-tumor activity in moderate burden RMS models but could not eliminate all tumors. The BiCisCAR with FGFR4.CD28HTM.CD28ICD and CD276.CD8HTM.4-1BBICD was the most potent construct, eradicating 100% tumors in all tested orthotopic models, and those with heterogenous expression of target antigens. CITE-seq and flow cytometry assays demonstrated that this BiCisCAR showed the most significant tumor infiltration, and persistence, with limited exhaustion. This BiCisCAR exhibited a synergistic effect on cytokine production and anti-tumor activity, compared to single targeting CARs. Moreover, biochemical characterization revealed that the combined use of both CD28 and 4-1BB ICDs in the BiCisCAR resulted in the activation of three TCR downstream signaling pathways including AKT, Erk1/2 and p65. Conclusions and Future DirectionsThus, we have developed a potent BiCisCAR with dual targeting of FGFR4 and CD276 that shows optimal biochemical activity, persistence, and limited exhaustion, and addresses heterogenous expression of target antigens. Air Conditioner is part in a car to achieve safety and security in driving. The purpose of this study was to determine the current of the air conditioner electrical system, identify the electrical circuit and analyze the electrical circuit of the air conditioner system. This research used experimental method. The air conditioner system simulator show current and voltage generated at blower speed 1: current 15.1 A, produces a temperature of 26 ℃, blower speed 2: current 15.2 A, produces a temperature of 25 ℃, blower speed 3: current 15.5 A, yields a temperature of 24 ℃. Whereas in the car the current and voltage generated by blower speed 1: current 17.4 A, produces a temperature of 21 ℃, blower speed 2: current 17.7 A, produces a temperature of 20 ℃, blower speed 3: current 17.9 A, produces a temperature of 19 ℃. So, the higher the current has an effect on the resulting performance of the air conditioner system. And the lower the current generated, the blower, magnetic clutch and extra fan performance is not optimal when the AC system is running. Sistem Air Conditioner merupakan bagian dari mobil untuk mencapai keyamanan dan keamanan dalam berkendara. Tujuan dari penelitian ini adalah untuk mengetahui arus sistem kelistrikan air conditioner, mengidentifikasi rangkaian kelistrikan dan menganalisis rangkaian kelistrikan sistem air conditioner. Metode penelitian yang digunakan dalam penelitian ini adalah metode eksperimental. Hasil pengujian arus dan tegangan yang dihasilkan pada simulator blower speed 1: arus 15,1 A, menghasilkan suhu 26℃, blower speed 2: arus 15,2 A, menghasilkan suhu 25℃, blower speed 3: arus 15,5 A, menghasilkan suhu 24℃. Sedangan pada mobil arus dan tegangan yang dihasilkan blower speed 1: arus 17,4 A, menghasilkan suhu 21℃, blower speed 2: arus 17,7 A, menghasilkan suhu 20℃, blower speed 3: arus 17,9 A, menghasilkan suhu 19℃. Jadi, semakin tinggi arus berpengaruh pada kinerja yang dihasilkan sistem air conditioner. Dan semakin rendah arus yang dihasilkan membuat kinerja blower, magnetic clutch dan extra fan menjadi tidak maksimal pada saat sistem AC bekerja.
