C-phycocyanin from the cyanobacterium, Mastigocladus laminosus, and its subunits have been treated with ρ-chloromercuribenzenesulfonate (PCMS). A single reactive site was found on the 13- subunit, and assigned to the single free cystein-β109. The concomitant spectral changes (absorp­tion, fluorescence, circular dichroism), together with the known close proximity of cys-β109 to chromophore β82, allowed an unambiguous assignment of the three spectrally, biochemically and functionally different chromophores to specific binding sites on the two peptide chains (α84: 616-618, β82: 622-624, β153: 598-600 nm).

AbstractRice straw was ensiled in laboratory containers of 11 capacity. Three concentrations of moisture (50, 60 or 70%) 3 concentrations of a commercial cellulase enzyme preparation (0, 5 or 10g/kg dry matter (DM)) and 3 types of straw processing (2 cm, 5 mm or 2 mm length) were used. The preparations were stored at room temperature (approximately 20°C) for 30 days. The straw treated with cellulase had a lower final pH (5.21, 4.87, 4.82; P<0.05), higher concentrations of lactic acid (198, 383, 367 mg/100g; P<0.05), a lower content of neutral detergent fibre (689, 630, 620 g/kg DM; P<0.05) and a higher solubility, measured as washing losses from nylon bags (152, 196, 212 g/kg DM; P<0.05) for the 0, 5 and 10 g/kg cellulase treatments respectively. The samples were subsequently incubated in nylon bags in the rumen of 3 sheep for 8, 16, 24, 48 and 72 h to estimate degradation rate and potential degradability using the expression p+a+b(1-e-ct) where p is degradability at time t and a, b and c are constants. While there was no effect of moisture content or physical form of the straw, the degradation rate constant (c) was significantly increased (P<0.05) by the addition of cellulase. The maximum potential (a+b) however was unchanged. The values for c were 0.0496, 0.0677 and 0.0847/h-1 and the values for (a+b) were 624, 621 and 628 g/kg for the 0, 5 and 10 g/kg cellulase enzyme additions respectively. It is concluded that the use of cellulase enzymes can assist in the preservation of wet straw and can result in improved degradation characteristics.

SummaryAntibodies directed against ADAMTS13 have been detected in the majority of patients with acquired thrombotic thrombocytopenic purpura (TTP). We have previously localized a major antigenic determinant within the spacer domain of ADAMTS13. To identify the amino acid residues of the spacer domain that are involved in binding of anti-ADAMTS13 antibodies, we constructed a series of fifteen hybrids (designated A-O) in which 5–10 amino acids of the spacer domain were exchanged for the corresponding region of ADAMTS1. Plasma from six patients with antibodies directed against the spacer domain was analyzed for reactivity with the ADAMTS13/ADAMTS1 chimeras. Exchange of amino acid residues 572–579 (hybrid C) and 657–666 (hybrid M) completely abolished the binding of antibodies from all six patients analyzed. Regions 580–587 (D), 602–620 (G, H), 629–638 (J), and 667–767 (N) contributed to binding of antibodies from patients 2, 4, and 5 (epitope present within regions CDGHJMN). Antibodies derived from patient 1 required region 602–620 (G, H) for binding (CGHM-epitope). For antibodies of patient 3, residues 564–571 (B), 580–587 (D), and 629–638 (J) were required (BCDJM-epitope), whereas replacement of residues 602–610 (G) and 629–638 (J) greatly diminished binding of antibodies from patient 6 (CGJM-epitope). Despite the presumably polyclonal origin of the antibodies present in plasma of patients, our results suggest that residues 572–579 (C) and 657–666 (M) comprise a common antigenic core region that is crucial for binding of anti-ADAMTS13 antibodies. Other regions that spatially surround this antigenic core further modulate binding of antibodies to the spacer domain.

Jalan Padang By Pass menghubungkan dua gerbang utama Provinsi Sumatra Barat, yaitu Bandar Udara Internasional Minangkabau dan Pelabuhan Teluk Bayur. Sejak tahun 2018, tipe jalan ini menjadi empat lajur dua arah terbagi. Fasilitas pelengkap pada jalan terbagi adalah median dan bukaan median. Penempatan dan jarak antar bukaan median pada jalan Padang ByPass terindikasi tidak memenuhi persyaratan teknik sehingga dapat membahayakan pengguna jalan. Untuk itu perlu dilakukan evaluasi geometrik dan kinerja. Pengumpulan data primer dilakukan melalui survei lapangan, yaitu pengukuran geometrik median dan bukaan median serta perekaman data volume dan waktu tunggu kendaraan melakukan putar balik di bukaan median. Perekaman data volume lalu lintas dan waktu putar balik dengan perekaman video dilakukan untuk jam sibuk pagi (06.00-08.00), jam sibuk siang (12.00-14.00) dan jam sibuk sore (16.00-18.00) WIB. Pembacaan data dari video menggunakan aplikasi FILMORA9. Hasil survei geometrik ditampilkan secara deskriptif dan dievaluasi dengan membandingkan kondisi geometrik standar yang ditetapkan dalam SNI 2444/2008. Evaluasi kinerja bukaan median didasarkan pada waktu tunggu mengikuti Peraturan Menteri Perhubungan Republik Indonesia nomor 96 tahun 2015. Hasil evaluasi geometrik median jalan By Pass disimpulkan sesuai standar SNI 2444/2018. Ukuran panjang bukaan median dalam wilayah studi tidak memenuhi SNI 2444/2018 yaitu 12 meter. Panjang bukaan median eksisting di lapangan adalah 5-11 m. Jarak antar median pada sta 7+682, 8+041, 8+251, 8+602 yaitu 255, m, 335,1 m, 199,1 m, 125,5 m. Kondisi ini tidak memenuhi standar SNI 2444/2018 karena kurang dari 500 m. Tiga bukaan median lainnya memenuhi syarat dengan jarak 565,4 m, 625 m, dan 841 m. Tingkat pelayanan C terjadi pada bukaan median sta 7+682 dan sta 8+602, sedangkan lima stationing lainnya dengan tingkat pelayanan B. Pada sta 7+682 tingginya volume kendaraan yang putar balik dengan panjang bukaan median 8,5 m terjadi waktu tunggu di antrian 16 detik/kendaraaan. Pada sta 8+602 panjang bukaan median hanya 5 m, cukup sulit bagi kendaraan ringan untuk melakukan manuver putar balik pada lokasi tersebut. Waktu tunggu di antrian 23 detik/kendraan dan panjang antrian 1081 m. Tingkat pelayanan C harus ditingkatkan menjadi B sesuai PerMenHub RI no.96/2015, yaitu dengan dengan menambah panjang bukaan median pada stasioning tersebut dan jika tidak memungkinkan bukaan median sebaiknya ditutup.

ABSTRACT Autonomously replicating sequence-binding factor 1 (ABF1) is a multifunctional, site-specific DNA binding protein that is essential for cell viability in Saccharomyces cerevisiae. ABF1 plays a direct role in transcriptional activation, stimulation of DNA replication, and gene silencing at the mating-type loci. Here we demonstrate that all three activities of ABF1 are conferred by the C terminus of the protein (amino acids [aa] 604 to 731). Furthermore, a detailed mutational analysis has revealed two important clusters of amino acid residues in the C terminus (C-terminal sequence 1 [CS1], aa 624 to 628; and CS2, aa 639 to 662). While both regions play a pivotal role in supporting cell viability, they make distinct contributions to ABF1 functions in various nuclear processes. CS1 specifically participates in transcriptional silencing and/or repression in a context-dependent manner, whereas CS2 is universally required for all three functions of ABF1. When tethered to specific regions of the genome, a 30-aa fragment that contains CS2 alone is sufficient for activation of transcription and chromosomal replication. In addition, CS2 is responsible for ABF1-mediated chromatin remodeling. Based on these results, we suggest that ABF1 may function as a chromatin-reorganizing factor to increase accessibility of the local chromatin structure, which in turn facilitates the action of additional factors to establish either an active or repressed chromatin state.

The effect of PEF on the whole liquid egg components, proteins and lipids, and the microstructure were studied and compared with pasteurisation. The effect of the refrigerated storage one week after the treatments was too studied. After all the treatments, only pasteurised samples showed water-soluble protein values significantly lower than the control and PEF treated samples, even after refrigeration. This could be related to the reinforcement of protein-protein interactions generated by the partial denaturalisation of proteins, observed by Cryo-SEM. Moreover, a water-soluble protein decrease was detected in the control and PEF treated samples after refrigeration, probably due to the aggregation of the egg lipoprotein during the storage. Furthermore, a slight lipolysis was observed in the control and PEF treated samples after refrigeration; but this effect was lower as higher was the PEF treatment. The study of the oxidation parameters showed an intermediate degradation of the lipids in treated samples, compared to the pasteurised eggs. These would reflect a higher microbiological stability of the PEF treated samples compared to the non-treated eggs.

The importance of regional adaptation of native plants is apparent in the selection of cultivars, especially in Cornus florida L., which grows in specific geographic areas of the United States where climate, soil and site characteristics are favorable. Selected from a seedling population in the western portion of the native range, `Ozark Spring' was compared with cultivars from other regions. Growth and flowering were determined under moderately stressful conditions in south central Kansas. The selection was one of the few which flowered following -30°C (-23°F) during the winter of 1982. Hardiness was confirmed at -29 to -31°C by University of Minnesota freezing tests. Performance of `Ozark Spring' has been superior to other cultivars in growth and bract development when planted in more exposed sites. The cultivar is expected to perform best in the Ozark region of Oklahoma, Arkansas, Missouri and eastern Kansas.

The mechanisms underlying increased angiotensinogen secretion in diabetic nephropathy are unknown. This study aimed to examine the mechanism of increased angiotensinogen secretion in mesangial cells. Additionally, we explored the effects of antioxidant compounds, such as isoflavones, on angiotensin secretion. Angiotensinogen expression and secretion were evaluated in mesangial cells treated with hydrogen peroxide. We investigated the effects of pretreatment with catalase, daidzein, and equol and inhibitors of mitogen-activated protein kinase, stress-stimulated kinase p38, or c-Jun NH2-terminal kinase. The 1,1-diphenyl-2-picrylhydrazyl radical scavenging assay revealed that daidzein and equol have antioxidant properties. Hydrogen peroxide stimulated angiotensinogen expression and secretion in mesangial cells in a concentrationdependent manner. Catalase, daidzein, and equol reduced the enhanced angiotensinogen expression and secretion induced by hydrogen peroxide. We examined the mitogen-activated protein kinase cascade to explore cell signaling mechanisms involved in angiotensinogen induction. We hypothesize that stress-stimulated kinase p38 and c-Jun NH2-terminal kinase were crucial in the mechanisms. We found that hydrogen peroxide enhanced angiotensinogen expression and secretion in mesangial cells. However, daidzein and equol decreased this enhancement. Increased angiotensinogen secretion will enhance stress-stimulated kinase p38 and c-Jun NH2-terminal kinase.

Based on the normal co-precipitation method to synthesize LiNi0.6Co0.2Mn0.2O2 cathode material, we propose a novel approach using a liquid nitrogen quenching method to synthesize Al2O3 coated LiNi0.6Co0.2Mn0.2O2 cathode material. In the whole process, liquid nitrogen was used four times to quench the materials from high temperatures (50 °C, 750 °C, 90 °C, 500 °C) to −196 °C rapidly in four stages. Various characterizations proved that this method could help to improve the electrochemical performance of lithium-ion batteries. Especially at 5 C rate current, after 100 cycles, the specific discharge capacities were 24.5 mAh/g (LNCM 622), 43.8 mAh/g (LNCM 622-LN), and 53.9 mAh/g (LNCM 622-LN@Al2O3). Liquid N2 quenching increased the charge/discharge capacities and the Al2O3 layer increased the cycle stability at high current, to finally obtain improved electrochemical properties.

We previously showed that allelic genes mo11 and mo12 used to protect lettuce crops against Lettuce mosaic virus (LMV) correspond to mutant alleles of the gene encoding the eukaryotic translation initiation factor 4E. LMV resistance-breaking determinants map not only to the main potyvirus virulence determinant, a genome-linked viral protein, but also to the C-terminal region of the cylindrical inclusion (CI), with a key role of amino acid at position 621. Here, we show that the propagation of several non-lettuce isolates of LMV in mo11 plants is accompanied by a gain of virulence correlated with the presence in the CI C terminus of a serine at position 617 and the accumulation of mutations at positions 602 or 627. Whole-genome sequencing of native and evolved isolates showed that no other mutation could be associated with adaptation to mo1 resistance. Site-directed mutagenesis pinpointed the key role in the virulence of the combination of mutations at positions 602 and 617, in addition to position 621. The impact of these mutations on the fitness of the virus was evaluated, suggesting that the durability of mo1 resistance in the field relies on the fitness cost associated with the resistance-breaking mutations, the nature of the mutations, and their potential antagonistic effects.

AbstractPostoperative infection rates were determined for gynecologic outpatient surgical procedures performed in a traditional operating room environment and a separate, recently opened, surgicenter within the same hospital. Infections were self-reported by attending surgeons responding to computer-generated line listings of their recent surgical procedures. Responses were obtained on 97.9% (612/625) of women having surgery in the operating room and 99.5% (629/632) of women with surgicenter procedures. The overall infection rate for reported women was 0.9% (11/1,241). The difference between operating room and surgicenter rates was not statistically significant. Postoperative infections occurred in 2.5% (3/118) of diagnostic laparoscopies with tubal lavage and 1.4% (3/214) of voluntary abortions by dilatation and evacuation and curettage (D&E&C). The five other infections were scattered among the remaining 25 procedure categories. Ten of the 11 infections were limited to the “clean-contaminated” wounds. No serious or life-threatening infections were encountered. The computer-assisted surveillance system worked well and was easily incorporated into the existing infection surveillance system. The degree of ascertainment of postoperative wound infections is unknown due to reliance on physician self-reporting. However, no patients requiring readmission for infection went unreported by the attending surgeons.

Transmission electron microscopy (TEM) was used in an investigation of the recrystallization behavior of amorphous surface layers on single crystal [0001] α-Al2O3. Conventional imaging and diffraction techniques were used to characterize the microstructures produced by annealing bulk specimens in the temperature range from 800°C to 1200°C. It was found that the amorphous Al2O3 first transforms to γ-Al2O3, a cubic transitional form, which in turn transforms to α-Al2O3. The γ to α transformation begins at the original crystalline-amorphous interface and proceeds toward the specimen surface. By measuring the position of the γ-α interface as a function of annealing temperature and time it was possible to determine the temperature-dependent velocity of the γ-α transformation front. Assuming a thermally activated process, the data are consistent with an activation energy of 3.6 eV for the transformation. However, no amorphous material remained, after even the shortest anneals (15 min) at the lowest temperature (800°C). These results indicate that the kinetics of the amorphous to γ transformation are faster than those of the γ to α transformation. Therefore a series of in-situ experiments was carried out to investigate the transformation of amorphous Al2O3 to γ-Al2O3. Amorphous Al2O3 surface layers, free of implanted impurities, were produced by implantation at −185°C with 2 × 1016 Al/cm2 at 90 keV and 3 × 1016 O/cm2 at 55 keV. Cross-sectioned specimens were prepared following established techniques.2 The specimens were annealed in-situ in a Philips CM12 operating at 120 kV with a Gatan Model 628 single tilt heating holder. Progress of the transformations was monitored with a Gatan model 622 video system.

Microspore embryogenesis (ME), a widely used haploid breeding method that can considerably shorten the breeding cycle, provides an efficient mean of cultivating many important Brassica crops, such as cabbage, Chinese cabbage, and oilseed rape. For cabbage, in many cases, short-term heat shock treatment can strongly increase the embryogenesis rate, however, the underlying mechanism of this effect has not been elucidated. In this study, we compared the proteomics of isolated microspores with samples pretreated at 32 °C for 24 h and 25 °C for 24 h using two cabbage accessions (Zhonggan 628 and 87–534) showing highly different embryogenic rates. The embryo yield was 19.7 embryos/bud in Zhonggan 628 after 32 °C treatment, while no embryoid was observed in Zhonggan 628 after 25 °C treatment as well as in 87–534 at both temperatures. We identified a total of 363 and 282 differentially expressed proteins (DEPs) for Zhonggan 628 and 87–534 via a label-free proteomics technology. There were 97 DEPs specifically identified only in Zhonggan 628 but not in 87–534 after 32 °C heat-shock treatment that may be related to heat shock-induced embryogenesis in vitro culture. These DEPs were primarily enriched in carbon metabolic process, protein synthesis and degradation process, and signal transduction. Based on protein-protein interaction and pathway enrichment analyses, we proposed that SGT1 homolog A and B(SGT1), heat shock 70 kDa protein 5 (HSP70), cell division control protein 48 homolog A (CDC48) and fatty acyl-CoA reductase (FAR) might play important roles in microspore embryogenesis. This proteomic study may contribute to our molecular understanding of cabbage microspore embryogenesis and help to build a high-efficiency haploid breeding system.

Abstract: This article addresses a prophecy found in vv. 2‒7 of the thirtieth Qurʾānic sūra, known as al-Rūm (“The Romans”). These verses report on the Romans’ (al-Rūm) involvement in a conflict against an unnamed enemy and predict its eventual outcome. The passage refers to the conflict between the Byzantines and Sasanians that lasted for about thirty years during the first three decades of the 7th c. (602‒628 CE). These verses are usually considered to be the only Qurʾānic allusion to a historical event that can be confirmed by sources external to the Islamic tradition. In this study I will argue that the prophecy on the Rūm has close parallels with other prophecies on the war that were circulating in the Middle East in the first half of the 7th c. The contextualization and comparison with other 7th c. prophecies will provide us with a better understanding of the Qurʾānic passage.

Abstract Inhibitory antibodies against ADAMTS13 have been detected in the majority of patients with acquired thrombotic thrombocytopenic purpura. Epitope-mapping studies revealed that antibodies binding to the cysteine-rich/spacer domains of ADAMTS13 are present in plasma of all patients analyzed so far. We have previously shown that a major antigenic determinant located within the spacer domain. To confirm these results we constructed a recombinant fragment consisting of the disintegrin/TSR1/cysteine-rich domains of ADAMTS13 and the spacer domain of ADAMTS1. The resulting ADAMTS13/ADAMTS1 chimera did not react with IgG present in plasma of a panel of patients with acquired TTP. To identify the amino acid residues that are involved in binding of anti-ADAMTS13 antibodies, we constructed a series of 15 hybrids (designated A–O) in which 5–10 amino acids of the ADAMTS13 spacer were exchanged for the homologous sequence of ADAMTS1. Plasma from 6 patients with antibodies directed against the spacer domain was analyzed for reactivity with the ADAMTS13/ADAMTS1 chimeras. Exchange of residues 572–579 (hybrid C) and 657–666 (hybrid M) of ADAMTS13 for the corresponding sequence of ADAMTS1 completely abolished the binding of antibodies from all 6 patients. Other regions of the ADAMTS13 spacer were also involved in binding of antibodies from patient plasma. Regions 580–587 (D), 602–620 (G,H), 629–638 (J), and 667–767 (N) of the ADAMTS13 spacer domain contributed to binding of antibodies from patients 2, 4, and 5 (epitope present within regions CDGHJMN). IgG derived from patient 1 required region 602–620 (G,H) for binding to the ADAMTS13 spacer (CGHM-epitope). For antibodies of patient 3, residues 564–571 (B), 580–587 (D), and 629–638 (J) were required (BCDJM-epitope), whereas replacement of residues 602–610 (G) and 629–638 (J) greatly diminished binding of antibodies derived from patient 6 (CGJM-epitope). Despite the presumably polyclonal origin of the antibodies present in plasma of the patients, our results suggest that residues 572–579 (C) and 657–666 (M) comprise a common antigenic core region within the spacer domain that is crucial for binding of anti-ADAMTS13 antibodies in all six patients. Amino acid residues derived from other regions that spatially surround this common antigenic core further modulate binding of antibodies to the spacer domain.

During mitosis, establishment of structurally and functionally sound bipolar spindles is necessary for maintaining the fidelity of chromosome segregation. Tumor-associated microtubule-associated protein (TMAP), also known as cytoskeleton-associated protein 2 (CKAP2), is a mitotic spindle-associated protein whose level is frequently up-regulated in various malignancies. Previous reports have suggested that TMAP is a potential regulator of mitotic spindle assembly and dynamics and that it is required for chromosome segregation to occur properly. So far, there have been no reports on how its mitosis-related functions are regulated. Here, we report that TMAP is hyper-phosphorylated at the C terminus specifically during mitosis. At least four different residues (Thr-578, Thr-596, Thr-622, and Ser-627) were responsible for the mitosis-specific phosphorylation of TMAP. Among these, Thr-622 was specifically phosphorylated by Cdk1-cyclin B1 both in vitro and in vivo. Interestingly, compared with the wild type, a phosphorylation-deficient mutant form of TMAP, in which Thr-622 had been replaced with an alanine (T622A), induced a significant increase in the frequency of metaphase cells with abnormal bipolar spindles, which often displayed disorganized, asymmetrical, or narrow and elongated morphologies. Formation of these abnormal bipolar spindles subsequently resulted in misalignment of metaphase chromosomes and ultimately caused a delay in the entry into anaphase. Moreover, such defects resulting from the T622A mutation were associated with a decrease in the rate of protein turnover at spindle microtubules. These findings suggest that Cdk1-cyclin B1-mediated phosphorylation of TMAP is important for and contributes to proper regulation of microtubule dynamics and establishment of functional bipolar spindles during mitosis.

Lithium-ion batteries have become the most promising energy storage devices in recent years. However, the simultaneous increase of energy density and power density is still a huge challenge. Ultrafast laser structuring of electrodes is feasible to increase power density of lithium-ion batteries by improving the lithium-ion diffusion kinetics. The influences of laser processing pattern and film thickness on the rate capability and energy density were investigated using Li(Ni0.6Mn0.2Co0.2)O2 (NMC 622) as cathode material. NMC 622 electrodes with thicknesses from 91 µm to 250 µm were prepared, while line patterns with pitch distances varying from 200 µm to 600 µm were applied. The NMC 622 cathodes were assembled opposing lithium using coin cell design. Cells with structured, 91 µm thick film cathodes showed lesser capacity losses with C-rates 3C compared to cells with unstructured cathode. Cells with 250 µm thick film cathode showed higher discharge capacity with low C-rates of up to C/5, and the structured cathodes showed higher discharge capacity, with C-rates of up to 1C. However, the discharge capacity deteriorated with higher C-rate. An appropriate choice of laser generated patterns and electrode thickness depends on the requested battery application scenario; i.e., charge/discharge rate and specific/volumetric energy density.

Strains 449T and 622 are both aerobic, Gram-stain-positive, short, rod-shaped bacilli that were recently isolated from the faeces of Tibetan antelopes on the Qinghai–Tibet Plateau in China. Their 16S rRNA gene sequences were most similar to those of Mycetocola zhadangensis ZD1-4T(97.9–98.0 %) and Mycetocola miduiensis CGMCC 1.11101T(97.3–97.4 %). Phylogenetic analysis of the 16S rRNA gene sequences further suggested that strains 449T and 622 represent a new lineage within the genus Mycetocola . The G+C content of strain 449T is 64.9 mol%. Optimal growth was achieved at pH 7.0 and 28 °C. Cells contained anteiso-C15 : 0 as the major cellular fatty acid, MK-10 and MK-11 as predominant menaquinones, diphosphatidylglycerol and phosphatidylglycerol as major polar lipids, and lysine as the diagnostic diamino acid. The DNA–DNA hybridization values of strains 449T and 622 were below the 70 % cut-off with respect to known strains of the genus Mycetocola . Based on these genotypic, phenotypic and biochemical characteristics, it seems rational to conclude that strains 449T and 622 belong to the genus Mycetocola and thus represent a novel species, for which the name Mycetocola zhujimingii sp. nov. is proposed. The type strain is 449T (=CGMCC 1.16372T=DSM 106173T).

El presente documento muestra a estudiantes y profesores la posibilidad de utilizar el lenguaje Java como herramienta para el aprendizaje de la programación en sustitución al tradicional lenguaje C. Se centra en el problema particular del uso de punteros en la implementación de algoritmos en C al intentar migrarlos a Java [1, 2, 3]. Muestra, mediante la utilización del patrón estructural Wrapper [4], que los algoritmos implementados en C son fácilmente traducibles a lenguaje Java [2] sin modificaciones considerables en la codificación. Finalmente sugiere implicaciones del uso de Java como lenguaje en el aprendizaje de la programación. Se pretende ir mucho más allá del acto investigativo y de la escritura del artículo, persiguiendo sembrar la importancia de arriesgarse a escribir, de proponer ideas, de crear y recrear la ciencia y la tecnología. En ocasiones, el estudiante y el profesor consideran como obvias [5] ciertas cosas dentro del acto educativo, el salón de clase, las cuales encierran, como en éste caso, situaciones ocultas que sólo con el tiempo se reflejan, situaciones que bien pueden ser tomadas por objeto de estudio de una investigación. The authors studied the physicochemical characteristics of new complex alloys containing, %: 11 – 30 Nb, 23 – 28 Si, 3 – 10 Al and 3 – 4 Ti. It was shown that complex alloys have the most favorable values of density and crystallization temperatures compared to standard ferroniobium (60 wt. % Nb). Complex alloys with a low concentration of niobium have acceptable crystallization temperatures and optimal density values (5740 – 6560 kg/m3). This allows the pieces of ferroalloy to be completely in the liquid steel when it is released into the ladle, and to be constantly in motion, which increases absorption of the leading components. When the niobium concentration increases to 30 %, phase composition of the alloy changes: a decrease in the proportion of the low-temperature FeSi phase with low density values and an increase in the proportion of the high-density ternary compound NbFeSi2 with a crystallization temperature of ~1713 °C. An increase in Nb concentration from 11 to 17 % leads to a decrease in the crystallization temperature, and a further increase to 30 % Nb, on the contrary, is accompanied by an increase in the liquidus and solidus temperatures to 1700 and 1610 °C, respectively, which is consistent with liquidus line in phase diagram of the Fe – Nb system with a minimum in Nb concentration range ~18 %. The best characteristics, both from the point of view of obtaining ferroalloys and use for alloying steel, belong to an alloy containing, wt. %: 17.1 Nb, 24.6 Si, 7.6 Al and 3 Ti. This alloy is characterized by the temperature of crystallization onset (1550 °C) below the liquid steel bath temperature and belongs to the category of low-melting alloys. It has optimal density values – 6390 kg/m3, which has a positive effect on the performance characteristics of niobium ferroalloys.

Abstract Lymphangiogenesis is induced in areas of chronic inflammation, including in both autoimmunity (associated with immunoreactivity) and in many solid tumors (associated with metastases and immunosuppression). We asked whether and how local VEGF-C-driven lymphangiogenesis alone could affect adaptive immunity to a model antigen. To this end, we designed a hydrogel system for sustained local delivery of the lymphangiogenic protein VEGF-C. These proteins were engineered to express a peptide tag which are covalently crosslinkable onto the hydrogel matrix, and are thus better retained by the hydrogels. Following implantation into healthy mice, local delivery of VEGF-C increased lymphangiogenesis at the implant site as well as at the draining lymph nodes compared to empty hydrogel controls. This promoted increased local expression of CCL21, similar to observations in lymphangiogenic tumors, and associated with increased recruitment of CCR7+ cells, most notably CD4 T cells expressing markers of naïve or memory phenotypes. To further examine the immunological relevance of these observations on antigen-specific T cell populations, hydrogels delivering both VEGF-C and the model antigen OVA were implanted into healthy mice which had previously received an adoptive transfer of OT-II T cells. These hydrogels also recruited CD4 T cells expressing markers of naïve and memory phenotypes. Our studies suggest a role for lymphangiogenesis in the recruitment of immune cells during inflammation.

ABSTRACT Aim This study investigated the effect of different fiber inserts (glass and polyethylene), bonding agents, and resin composites on the gingival margin microleakage of class V composite restorations. Materials and methods Sixty premolars were sterilized and mounted in acrylic resin bases. Class V cavities were prepared buccally and lingually, 1 mm below the cementoenamel junction, comprising 12 groups (n = 10). In the experimental groups fiber inserts were cut and placed at the gingival seat, while the control groups had no inserts. Combinations of two composite materials, Filtek-Z250 and Filtek-LS (3M-ESPE), and four bonding agents, Clearfil SE bond (Kuraray) (C), Scotch Bond Multipurpose (3M-ESPE) (SB), Prime and Bond NT (Dentsply) (PB), and Filtek-LS (3M-ESPE) (LS) were used. Restorations were incrementally inserted and polymerized for 40s. Specimens were then stored in distilled water for 7 days and thermocycled for 500 cycles. Teeth surfaces were sealed with nail polish except for 1 mm around restoration margins and immersed in 2% red procion dye. Teeth were then sectioned buccolingually and dye penetration was assessed with five-point scale. Data were statistically-analyzed by Kruskal-Wallis, ANOVA and Tukey's tests (α = 5%). Results Mean microleakage scores varied from 0.40 (Groups C, C with polyethylene, LS, LS with polyethylene) to 1.50 (SB). Conclusion Different bonding agents led to differences in microleakage scores where C and LS showed significantly lower microleakage than PB. SB had highest mean microleakage score, however, incorporation of fibers resulted in significant reduction in microleakage. Clinical significance Class V resin composite restorations bonded with a total-etch adhesive had a significant reduction in mean microleakage scores when glass or polyethylene fibers were placed at the gingival cavo-surface margin. Clustering algorithms has emerged as an important data mining technique for pattern recognition, data analysis and dimensionality reduction. Clustering is usually used in all fields to merge the same feature objects into a single group. The clustering method is incorporated with search algorithms to search the dataset from the databases. For large databases, there is a need of good clustering algorithm with high accuracy. Despite its high performance, the existing methods show some limitations. This paper focused on optimize the clustering method with a search structure for large multidimensional databases with dynamic indexes for the effective validation of construction projects. In the construction project validation to improve the clustering methods using Evolutionary Constrained Differential Optimization (ECDO) and Distributed Weighted Fuzzy C-Means algorithm to improve clustering performance than the centralized clustering approaches. The proposed methodology includes ECDO which presents new cluster tree indexing approach with cluster speed improvement in the validation of the construction projects. Distributed Fuzzy Possibilistic C-Means algorithm is proposed in improvement of cluster centre to overcome coincidence cluster problems. The comparative analysis stated that proposed ECDO model exhibits ~6% reduced memory utilization ~11% reduced scalability and ~8% minimized selectivity than the conventional WPCM model.

Abstract Alterations in the RAS-MAPK pathway are identified in nearly 30% of cancers and are associated with poor patient prognosis. Although available therapies target various nodes of the RAS-MAPK pathway, clinical anti-tumor activity is limited by paradoxical bypass mechanisms, feedback signaling and pathway reactivation, and genetic resistance mechanisms. NST-628 aims to overcome the limitations of traditional RAS-MAPK pathway inhibitors by stabilizing all RAF-MEK complexes in a conformation that blocks MEK phosphorylation, to sequester these nodes from dynamic signaling processes that promote RAF-mediated bypass signaling. In comparison with clinically approved RAS-MAPK pathway inhibitors, only NST-628 robustly and durably decreases pathway reactivation, as measured by MEK phosphorylation, in KRAS-G13D tumor models in vitro and in vivo. Immunoprecipitation of endogenous ternary complexes demonstrates that NST-628 promotes potent stabilization of CRAF-MEK, BRAF-MEK, and ARAF-MEK complexes with complete shutdown of MEK, ERK, and RSK phosphorylation. Moreover, NST-628 treatment completely prevents RAF paralog heterodimerization in RAS-driven cells. Dose-dependent panRAF-MEK complex stabilization was confirmed using recombinant pre-formed CRAF-MEK, BRAF-MEK, and ARAF-MEK complexes, and MEK and RAF paralog affinity increases in the presence of NST-628 were quantified via surface plasmon resonance (SPR). In this assay, NST-628 significantly increased the affinity of MEK with both inactive monomeric and active dimeric RAF complexes. In parallel, we structurally characterized NST-628 bound to CRAF-MEK, BRAF-MEK, ARAF-MEK complexes using x-ray crystallography and cryogenic electron microscopy (cryo-EM). NST-628 binds the interfacial allosteric pocket along the MEK activation loop, thereby preventing RAF-mediated activation of MEK. We also demonstrate RAF paralog-specific pocket features that NST-628 can engage, helping us rationalize the large increase in affinity observed for MEK and CRAF in the presence of NST-628. The increased stabilization of CRAF-MEK complex by NST-628 may contribute to the decreased pathway reactivation in specific biomarker-driven tumors. Through our robust biophysical and cellular characterization coupled with novel structural insights, we have defined the unique mechanism of action for NST-628 and provide insight into its best-in-class potential in RAS-MAPK activated cancers. Research the kinetic mechanism of grain growth of recrystallization of Cu-20Ni-5Sn alloy. The research result shows that the recrystallization temperature declines with the increase of cold-deformation. The recrystallization temperature is at about 500°C ~650°C respectively for 50% and 60% total cold deformation, and is about 470°C ~620°C respectively for 70% and 85% total deformation. The grains grow up with the increase of annealing temperature and holding time. The mathematic model of average grain size can be described as that Cu-20Ni-5Sn alloy annealed at 620°C ~680°C holding 2~10hours.

Abstract Alterations in the RAS-MAPK signaling cascade are common across multiple solid tumor types and are drivers for many cancers. Apart from approved mutation-selective inhibitors for BRAF Class I and KRAS G12C mutations, other mutations of RAS and RAF are not directly addressable by currently approved inhibitors and there is a need for therapeutic strategies having superior efficacy, durability and tolerability for the MAPK pathway. NST-628 is a potent pan-RAF-MEK molecular glue that prevents phosphorylation and activation of MEK by RAF. In this way, NST-628 overcomes the limitations of traditional RAS-MAPK inhibitors and drives deep, durable inhibition of the RAS-MAPK pathway. Cellular, biochemical, and structural analysis of RAF-MEK complexes show that NST-628 engages all isoforms of RAF and prevents the formation of BRAF-CRAF and BRAF-ARAF heterodimers, a differentiated mechanism from all current RAF-directed inhibitors. With its potent and durable inhibition of the RAF-MEK signaling complex, NST-628 demonstrates broad efficacy in cellular and patient-derived, in vivo tumor models harboring KRAS, NRAS, BRAF class II/III, and NF1 mutations. In addition, high intrinsic permeability across the blood-brain-barrier results in NST-628-mediated intracranial anti-tumor activity in orthotopic tumor models. Furthermore, NST-628 pharmacokinetic and metabolic profiles have been optimized for daily dosing in the clinic to maximize therapeutic index. Given its differentiated molecular mechanism, best-in-class drug-like properties, and superior safety profile in IND-enabling GLP studies, NST-628 is positioned to make an impact clinically to address unmet patient need. Abstract Neutrophils extracellular traps (NETs), which are DNA scaffolds in complex with granule proteins, could be detected in the pre-metastatic liver niches and were associated with a poor survival in breast cancer patients. However, the mechanisms of neutrophils infiltration and NETs formation in the pre-metastatic liver tissues remain poorly understood. Here, we dynamically characterized the liver micro-environment of normal and 4T1-bearing BALB/C mouse models with or without liver metastasis by the single cell RNA sequencing. Interestingly, we found that CXCL1 and CXCL2, derived from the liver macrophages, were dramatically elevated in pre-metastatic livers and induced the infiltration of peripheral neutrophils by CXCR2. Besides, liver macrophages were educated by cancer cells-derived cathepsin C to secrete the CXCL1 and CXCL2. Furthermore, we identified that the complement signals were over-activated in pre-metastatic livers and induced neutrophils to NETosis. Therapeutically, targeting CXCL1, CXCL2 and complement signals could inhibit the NET formation and effectively reduce breast cancer liver metastasis. Abstract The twelve title compounds have been prepared for the first time. Their CaCr2Al10-type structure (P4/nmm, Z = 4) was refined from single-crystal X-ray data for the three representatives TbMnAl10 (a = 1274.3(2) pm, c = 511.4(2) pm, R = 0.025 for 680 structure factors F and 43 variable parameters V), CeRe2Al10 (a = 1295.5(5) pm, c = 517.2(4) pm, R = 0.054 for 810 F and 46 V), and SmRe2Al10 (a = 1291.5(2) pm, c = 516.5(1) pm, R = 0.021 for 622 F and 46 V). The atomic positions of the lanthanoid and transition metal atoms are fully occupied. Significant deviations from the full occupancies were observed for two aluminum sites in TbMn2Al10 and for all five aluminum sites of the two rhenium-containing compounds, resulting in the compositions TbMn2Al9.63(2), CeRe2Al9.52(8), and SmRe2Al9.16(9). The cell volume of CeRe2Al10 and to a smaller extent also that of CeMn2Al10 indicate mixed or intermediate +III/+IV valencies of the cerium atoms in these compounds. The structural relationships between the three closely related body-centered tetragonal structures of ThMn12, CeMniAl8, DyFe6Al6 and the primitive tetragonal structure of CaCr2Al10 are briefly discussed.

622 Background: Expression of the proto-oncogene c-kit has been found in malignant tissue including a subset of breast cancers. c-Kit is also expressed in normal breast tissue and several authors found a loss of c-kit expression in breast carcinoma suggesting it might be involved in the growth control of mammary epithelium. Until now, only a few markers were described to be co-regulated with c-kit. To elucidate the possible role of c-kit in malignant transformation, we analyzed gene expression data of breast cancer patients. Methods: Tumor tissue of n=171 breast cancer patients were analyzed by gene expression profiling using Affymetrix Hg U133 Arrays (22,500 genes) and bioinformatic analyses. Tumor samples with high stromal and low epithelial cell content by gene expression profiling were excluded for further analysis. Validation was performed with n=100 independent samples. Results: A total of 10.5% of the tumors showed strong c-kit expression (2.5 fold above median). A careful dissection of global expression data revealed strong correlations of c-kit with the expression of a large cluster of genes containing several for whom c-kit coexpression was already described (HER1, CK-5/-17, PDGFR) as well as several members of the wnt signalling pathway, providing a possible novel link to mammary epithelial differentiation. Analysis of n=171 breast cancer samples according to this gene set allows the identification of putative “stem cell like” tumors (SCL) characterized by expression of several known stem cell markers. Surprisingly, a tight link of ER status and proliferation is restricted only to these SCL tumors but lost among non-SCL tumors. The clinical implications of our findings will be presented. Conclusions: For the first time these data bring together the description of two breast cancer subtypes identified by gene expression profiling with the actual stem cell model of the development of breast cancer. The solubility curves at 75 °C, represented in Jänecke's mode, of the quasi-ternary section K2SO4–KH2PO4–H2O and the limiting ternary system K3PO4–H3PO4–H2O of the reciprocal quaternary system H+, K+, PO43−, SO42−, H2O reveal an unusual increasing shape for the congruent salts KH2PO4 and K2HPO4. This shape is surprising when one considers the similarities that exist between the role of water content and that of temperature in polytherm systems. The effect of the representation mode on the shape of the solubility curves has been studied in order to explain this unusual behaviour. A thermodynamic treatment of this phenomenon is also proposed. Apart from approved mutation selective inhibitors for BRAF Class I and KRAS G12C mutations, other mutations of RAS and RAF are not directly addressable by currently approved inhibitors and there is a need for inhibitors with superior efficacy, durability and tolerability for the MAPK pathway in RAS- and RAF-driven cancers. NST-628 is a potent pan-RAF-MEK molecular glue that prevents phosphorylation and activation of MEK by RAF, leading to deep durable inhibition of MEK kinase activity and downstream ERK signaling. In a MEK1 immunoprecipitation assay, NST-628 treatment glues ARAF, BRAF, and CRAF with unphosphorylated MEK1, stabilizing an inactive RAF-MEK complex and deeply inhibiting phospho-ERK signaling. In an unbiased cell line panel screen (550 models), NST-628 demonstrates efficacy across multiple tumor types with RAS-MAPK alterations, including melanoma, lung, and pancreatic models. NST-628 is broadly efficacious in models with BRAF Class II/III mutations, KRAS-mutations (G12C, G12D, G12V, G12R, Q61H), and NRAS-mutations (Q61x, G12x) as well as showing anti-proliferative effects in NF1-mutant/deficient models. Cell lines with NRAS Q61 mutations were particularly sensitive to NST-628 inhibition (GI50 average=150 nM). In vivo, NST-628 has a balanced metabolic profile and predicted half-life and exposure in humans is compatible with low dose daily dosing. NST-628 (3-5 mg/kg QD treatment) led to tumor regressions in HCT116 (KRAS G13D colorectal) and IPC-298 (NRAS Q61L melanoma) xenograft models, 53% and 38% respectively, correlating to inhibition of both phospho-MEK and phospho-ERK in tumors. In comparison to the MEK inhibitor trametinib, NST-628 led to deeper tumor responses and on target pathway inhibition in both models as well as significantly greater tolerability as measured by body weight. In a mini-mouse trial utilizing patient derive xenograft (PDX) tumors harboring NF1, KRAS G12D/R, BRAF Class II/III, and NRAS Q61x mutations, NST-628 (3 mg/kg QD) dosing demonstrates broad anti-tumor responses across melanoma, lung, pancreatic, glioma, and ovarian models. These data collectively, together with the predicted effective human half-life of NST-628 being consistent with daily dosing, support best-in-class potential for NST-628 and initiation of clinical trials in patients with solid tumors harboring RAS- and RAF-mutations is planned.
