The Dawesley Creek–Bremer River drainage system, in South Australia, is affected by the potentially costly problem of acid drainage from the Brukunga mine site. This paper contrasts geochemical model predictions of water quality with detailed measurement of water chemistry in the field. This information is also used in the ecological risk assessment code, AQUARISK, to predict the degree of ecological detriment. These predictions are then assessed against independent, field-based biomeasures of algal and bacterial communities to test the performance of the code. The study demonstrates that the assessment of ecological risk obtained using AQUARISK, when coupled with geochemical modelling, accords quite well with the independent data from the biomeasures. Results from the study also suggest that only a single water-quality guideline value is required for aluminium, the major toxicant in the Dawesley Creek–Bremer River system.

I review a variety of recent case histories illustrating the application of geophysics in mineral exploration in Australia. Geophysics is now an integral part of most programs. Examples are given of contributions by geophysics to all stages of mineral exploration, from regional area selection through to mine planning and development. Specific case histories summarized are as follows: (a) Olympic Dam copper‐uranium‐gold deposit, discovered using a conceptual genetic model and regional geophysical data; (b) Ellendale diamondiferous kimberlites, illustrating the use of low level, detailed airborne magnetics; (c) Ranger uranium orebodies, discovered by detailed airborne radiometric surveys; (d) geologic mapping near Mary Kathleen with color displays of airborne radiometric data; (e) mapping of lignite in basement depressions of the Bremer Basin, near Esperance, with INPUT; (f) White Leads, a lead‐zinc sulfide deposit discovered with induced polarization (IP) and TEM, near Broken Hill; (g) Hellyer, a lead‐zinc‐silver‐gold deposit discovered with UTEM; (h) application of geophysical logging near Kanmantoo; (i) Cowla Peak, a subbituminous steaming coal deposit mapped with ground TEM; and (j) Cook Colliery, where high‐resolution seismic reflection methods have successfully increased the workable reserves.

The Country Life Novel’s Gothic Machinery.National Identity and Fluctuating Exchange Value in Fredrika Bremer’s Life in Dalecarlia
When Fredrika Bremer in her novel I Dalarne (1845) (Life in Dalecarlia) represents Falu copper mine as the horrific ruin of classic gothic tales – a vast and life-threatening void, the home of dreadful crimes and secret passions – this is paradoxically part of a nationalistic literary strategy. Simultaneously, though, the novel writes the nation as a pastoral on country life and mores of the peasantry. The article investigates how the gothic and the pastoral idyllic respectively establish the nation and national identity on their separate terms and in relation to each other in accordance with their own particular inner logic.
The idyllic streaks both manifests and problematize local place as foundation for national identity, while the gothic exposes identity as arbitrary, and thereby subverts the notion of a citizen subject. This duality is linked to a market-based model of identity which the article argues is actualized by the novel itself. The condition of possibility is the emerging consumer society and the beginnings of industrialism – breakthrough changes which in the novel are thematized as tensions between tradition and modernity. At stake in this field of tension between the idyllic and the gothic stands the nation as a community of citizens: a collective subject.

This article examines the professional career of Governor Albert P. Brewer and the development of his vision for a public affairs research group, that exists to give objective facts about state and local governments. The research question of this article is if Albert Brewer’s goal was fulfilled. The careful planning and the quality reputation of the published research of Public Affairs Research Council of Alabama (PARCA) has fulfilled the goal that Brewer had in mind. A methodology in arriving at this conclusion can be found in investigating the published output and broad, favorable reception of that output. This article examines the major events in the career of Governor Albert Brewer. He was a state legislator, speaker of the house, lieutenant governor and governor of Alabama. Brewer had many accomplishments, including the creation of the Alabama Development Office (ADO), the Alabama Commission of Higher Education (ACHE), substantial increased funding for public schools, and an effort to produce a new state constitution. A goal of Brewer was to create the PARCA, as a source of objective data and other facts that all people could use. PARCA is a non-profit and non-partisan group. The goals of Brewer for PARCA have been met and the organization continues to flourish. Brewer is one of the most decent professionals to ever occupy the Alabama governor’s office. Brewer was governor and decided to pursue another term with the 1970 election. In his political campaigns he projected the goal of being inclusive as opposed to the strong desire of George Wallace to cause division between African American citizens and some other citizens. That is, Wallace pursued a campaign tactic of creating a racist environment, to get votes. That election is considered by historians to be one of the most racially charged elections. The result was that Brewer and Wallace were in a run-off, and Wallace won by less than 34,000 votes. Later, in 1972 Wallace survived an assassination attempt. Some people think that event changed him, as he did stop pursuing a racist ideology for the rest of his political career. In contrast, Brewer consistently pursued a more inclusive message in appealing to citizens to work together for progress. PARCA, a group championed by Brewer furthers that goal. To help policy makers, PARCA provides objective data, which people of all political views can refer to. Brewer’s goals of an organization that provides accurate information has been achieved.

Dendritic cells (DCs) are a type of antigen-presenting cells that play an important role in the immune response against Trypanosoma cruzi, the causative agent of Chagas disease. In vitro and in vivo studies have shown that the modulation of these cells by this parasite can directly affect the innate and acquired immune response of the host in order to facilitate its biological cycle and the spreading of the species. Many studies show the mechanisms by which T. cruzi modulates DCs, but the interaction of these cells with the Mexican strains of T. cruzi such as Ninoa and INC5 has not yet been properly investigated. Here, we evaluated whether Ninoa and INC5 strains evaded the immunity of their hosts by modulating the biology and function of murine DCs. The CL-Brener strain was used as the reference strain. Herein, it was demonstrated that Ninoa was more infective toward bone marrow-derived dendritic cells (BMDCs) than INC5 and CL-Brener strains in both BMDCs of BALB/c and C57BL/6 mice. Mexican strains of T. cruzi induced different cytokine patterns. In BMDCs obtained from BALB/c mice, Ninoa strain led to the reduction in IL-6 and increased IL-10 production, while in C57BL/6 mice Ninoa strain considerably increased the productions of TNF-α and IL-10. Also, Ninoa and INC5 differentially modulated BMDC expressions of MHC-II, TLR2, and TLR4 in both BALB/c and C57BL/6 mice compared to Brazilian strain CL-Brener. These results indicate that T. cruzi Mexican strains differentially infect and modulate MHC-II, toll-like receptors, and cytokine production in DCs obtained from C57BL/6 and BALB/c mice, suggesting that these strains have developed particular modulatory strategies to disrupt DCs and, consequently, the host immune responses.

The crude hydroalcoholic extract of Cassia sophera Linn. leaves were investigated for its possible analgesic and antipyretic activities in rodents. The peripheral analgesic activity of C. sophera leaves was studied using acetic acidinduced writhing in mice and central analgesic activity was evaluated by using heat-induced pain in mice. The antipyretic activity of HECS was studied in Brewers yeast-induced pyrexia in rats. All these experiments were done at the doses of 300 and 600 mg/kg body weight. Intraperitoneal administration of C. sophera hydroalcoholic extract (HECS) produced significant (P<0.01) reduction in number of writhing induced by acetic-acid. Moreover, in hotplate test, HECS significantly (P<0.01) raised the pain threshold at different time of observation (0-180 min) in comparison with control. The extract also showed significant as well as dose-dependent antipyretic activity in Brewers yeast-induced pyrexia in rats throughout the observation period of 4 h. Abstract CM is a globally renowned machine, designed to work as a mass production technology for underground coal. Different major coal producers across the globe are using this technology for decades to produce underground coal efficiently. India is also one of the major players globally in the arena of coal production and adopted this cutting edge technology since last decade by implementing at. few of the selective underground coal mining projects. Performance of CM technology is influenced by the geo-mining condition, fleets of other ancillary units and reliability of subsystems while implementation of this system depends largely on the extent of reserve. These aspects generate a scope of large scale research and development in this field. Overall Equipment Effectiveness (OEE) is the parameter to benchmark the equipment performance globally. OEE is the product of equipment availability, performance and product quality. This mining machine based paper focuses on the Overall Equipment Effectiveness (OEE) of the complete CM based operation to identify the vulnerable systems, which helps to design proper preventive maintenance programme. The CM based system is divided into few subsystems, such as; electrical, cutter, gathering arrangement, traction, hydraulic, chassis, feeder breaker, shuttle car, CM conveyor and out-bye conveyor. The downtime data used for this analysis is collected from an underground coal mine situated in the central part of India, belongs to one leading coal producing company of the country. From analysis it was found that, electrical systems and conveyors are among most vulnerable systems and deserves more care during maintenance. On the basis of these results recommendations are made to redesign the Preventive Maintenance Programme, in order to avoid the lower availability as well as lower OEE.

The project unites the artistic approach of creating metaphors and the scientific approach of theoretical inquiry. In this essay, artist and scientist show in a dialogical manner how art and science gain complementary insights by working with the same cognitive tools.

Bawang merah (Allium ascalonicum L.) merupakan salah satu komoditas penting dan cukup dominan di Kabupaten Brebes. Kebutuhan nasional terhadap bawang merah sejumlah 750.000 ton/tahun dan pada tahun 2019 kebutuhan tersebut diprediksi mencapai 1.060.400 ton. Kebutuhan ekspor bawang merah mencapai 2.500 ton/tahun dan baru terpenuhi 1.500 ton/tahun. Produktivitas rata-rata bawang merah nasional hanya sekitar 9,24 ton/ha, jauh dibawah potensi produksi yang berada diatas 20 ton/ha. Produktivitas bawang merah di Kabupaten Brebes saat ini hanya berkisar 80,00 - 137,72 kuintal/ha karena masih mengalami banyak kendala baik dari aspek penguasaan teknologi yang efektif namun ramah lingkungan, manajemen maupun modal. Kegiatan IbM ini dilaksanakan pada bulan April – Nopember 2017 dan bermitra kerja dengan Mitra IbM-1 dan Mitra IbM-2 di Desa Banjarsari, Kecamatan Brebes, Kabupaten Brebes. Tujuan dari kegiatan IbM adalah meningkatkan kemampuan mitra IbM dalam mengelola usaha budidaya mina-bawang baik dalam aspek teknologi budidaya maupun manajemen dalam rangka menguatkan aspek kewirausahaan dan kemandirian mitra untuk meningkatkan kesejahteraan. Target dari kegiatan IbM ini adalah: 1) Mitra mampu menerapkan teknologi biofilter-bioactive system di lahan yang memiliki keterbatasan ketersediaan air untuk menghasilkan produk bawang merah yang terbebas dari bahan kimia sehingga dapat meningkatkan kualitas produk sekaligus menekan biaya produksi; 2) Mitra mampu mengelola usahanya dengan lebih efisien. Teknologi biofilter, pupuk serta pembasmi hama dari bahan organik pada kegiatan IbM memungkinkan budidaya bawang dapat dilaksanakan 4 kali sedangkan sebelum kegiatan IbM hanya dapat dilakukan 3 kali karena kendala keterbatasan air. Selisih keuntungan Mitra IbM-1 dan IbM-2 setelah diadakannya IbM dengan sebelum ada kegiatan IbM masing-masing sebesar Rp. 122.974.928/hektar/tahun (meningkat 43,47%) dan Rp. 106.715.928/hektar/tahun (37,16%).

Brewer, Young, and Barnett reported a comprehensive psychometric evaluation of a new adult theory of mind measure (A-ToM) with a sample of high-functioning autism spectrum disorder (ASD) adults. Although correlations with existing theory of mind (ToM) instruments (i.e., the Strange Stories; the Frith- Happé animations) were reported, relationships with independent putative indicators of ToM development such as social–behavioral and interpersonal proficiencies were not examined. Here, we provide convergent validity data by examining the relations between A-ToM performance, and the social–behavioral skills and interpersonal relationships of ASD adults with IQs exceeding 85. ToM predicted interpersonal relationship quality via the mediating variable, social–behavioral skills, providing evidence of convergent validity for the A-ToM. Alternative models of the relationship between the three variables are described, as are the challenges associated with the interpretation of self-report social and interpersonal functioning measures.

Mouse model research is proliferating because of its readiness for genetic manipulation. Little is known about pulmonary vagal afferents in mice, however. The purpose of this study was to determine whether their pulmonary afferents are similar to those in large animals. Single-unit activity was recorded in the cervical vagus nerve of anesthetized, open-chest, and mechanically ventilated mice. We evaluated airway sensory activity in 153 single units; 141 were mechanosensitive, with 134 inflation receptors and 7 deflation receptors. The remaining 12 receptors were chemosensitive and mechanically insensitive, showing low basal firing frequency and behaving like C-fiber or high-threshold Aδ-receptors. In separate studies, phrenic activity was recorded as an index of respiratory drive to assess pulmonary reflexes. Lung inflation produced a typical Hering-Breuer reflex, and intravenous injection of phenylbiguanide produced the typical chemoreflex resulting in apnea, bradycardia, and hypotension. These reflexes were blocked by bilateral vagotomy. We conclude that mice possess a similar set of airway sensors and pulmonary reflexes as typically found in larger animals.

During oral arguments for the Patient Protection and Affordable Care Act, Justice Breyer on several occasions questioned whether the federal government could compel individuals to be vaccinated in the event of a national emergency where a highly contagious disease was sweeping through the country. This article does not seek to predict or analyze the legal implications of such an action; rather it argues that a national approach to such an emergency should be implemented. Recent concerns over the potential for H5N1, or “bird flu,” to become airborne illustrate the type of epidemic that Justice Breyer may have been envisioning. By broaching this subject now, instead of in the midst of an outbreak, adequate time is left to research appropriate solutions, allow for debate, and provide public education.While vaccination laws are typically promulgated on the state level under state police power, these compulsory laws are accompanied by exemptions that can undermine their effectiveness. For example, religious and philosophical exemptions have led to outbreaks of pertussis, or whooping cough, in multiple states. Considering the various state exemptions along with laws granting governors and health officials broad power to alter vaccination laws during emergencies, it is nearly impossible to predict how individual states will respond. Legally and ethically speaking, the rights of individuals are not absolute and cannot be utilized to subject others to harm.A federal compulsory vaccination law allows for balancing individual rights and public health, with the interests of the nation as a whole in mind.

The use of transgenic animals to manipulate milk composition has considerable potential, both for the production of biomedical proteins and for the direct manipulation of milk composition for the improvement of dairy animals and their products (for reviews, see Wall et al. 1992; Yom & Bremel, 1993). Promoters from a number of milk protein genes from a variety of species have been tested for their ability to direct the expression of foreign proteins to the mammary gland (for review, see Maga & Murray, 1995).β-Lactoglobulin (β-lg) is the major whey protein produced in ruminant milk and is part of the normal milk composition of most mammals except humans and rodents (Pervaiz & Brew, 1985). It is expressed at high levels in the mammary gland and is developmentally regulated. Transgenic mice have been produced using the complete ovine (Simons et al. 1987; Shani et al. 1992) and caprine (Ibañez et al. 1997) β-lg genes. In general, high levels of expression were obtained with the ovine β-lg gene, and expression was also seen in a position-independent manner (Whitelaw et al. 1992). Lower levels of expression were reported using the caprine β-lg gene. Here we report the production of transgenic mice using the bovine β-lg gene. We describe high expression, position-dependent, and copy number-related expression of bovine β-lg protein in the milk of six lines of transgenic mice.

Abstract In order to mine the state information of SF6 circuit breakers (CBs) interruption chamber and evaluate the contacts state, a method of measuring and calculating the dynamic capacitance between contacts in CBs opening process was proposed. The test CB is connected in series with a high frequency AC power supply, and the voltage and current of the circuit breaker are measured during the opening process. The fundamental frequency components of the voltage and current are extracted by the short-time Fourier transform, and the dynamic capacitance curve as a function of time is calculated. The results show that due to the variation of capacitance, there are a large number of harmonics with frequencies close to the fundamental frequency in terminal voltage and current. During the first two cycles after contacts separation, the capacitance drops sharply to 33.25 pF, then with the increase of the contact distance, the capacitance decreases much more slowly and tends to be constant, and the capacitance is 20.70pF after the completion of the opening.

Abstract Large pyrite-dominated gold deposits are hosted in hydrothermally altered metamorphic rocks in the Carolina slate belt of South Carolina and are partly covered by Coastal Plain sedimentary rocks. In this study, we investigate the utility of geophysical data including aeromagnetic, electromagnetic (EM), and land gravity surveys as exploration tools. Observed geophysical anomalies are correlated with rock properties such as resistivity, susceptibility, and density. Mineral concentrations were measured for 40 samples from 16 new drill holes, as well as densities and pyrite concentrations for 49,183 samples from 448 drill holes in the Haile ore zone. Regional positive gravity anomalies are observed over the Haile, Ridgeway, and Barite Hill mine areas, and residual high-pass filtered positive gravity anomalies are observed over all mine areas. New measurements for drill cores in the Haile mine area directly confirm high rock densities and high pyrite concentrations in the ore zone. In the Haile and Brewer mine areas, high-conductivity EM anomalies are observed over the ore zones as well as over nearby metasedimentary rocks. The high concentration of pyrite in the metasedimentary units and in the ore zones may explain the high conductivities observed. All gold deposits in the Carolina slate belt are hosted in similar geologic settings near the contact between the Persimmon Fork metamorphosed volcaniclastic rocks and Richtex metamorphosed sedimentary rocks. Density and conductivity contrasts between the Persimmon Fork and Richtex Formation rocks may permit mapping of the contact zone between the two units. The magnetic anomalies do not correlate with the mineralized zones but rather with granite and gabbro plutons and diabase dikes. A prominent east-northeast linear magnetic anomaly correlates with the Modoc shear zone that separates low-grade metamorphic rocks of the Carolina terrane from higher-grade metamorphic rocks of the Kiokee belt. We use the Modoc linear magnetic anomaly to predict the southeastern boundary of the Carolina slate belt where it is covered by Coastal Plain sedimentary rocks.

While Heidegger decried ethics as a distinct area of philosophical inquiry, a steady stream of secondary literature over the last three decades has mined his corpus for ethical insights. This literature tends to draw on his early or middle work and contrast his views with canonical normative theories. I bring Heidegger into conversation with philosophy of disability and feminist philosophy by focusing on the role of relationality and ability expectations. In section one, I provide a schematic of the dominant concept of ability in modernity: ability as personal power. Through the Bremen lectures, I then develop a Heideggerian concept of ability: ability as access. I conclude by discussing the stakes—ethical, philosophical, and political—of interpreting the question of the meaning of being as a question of ability as access to meaning.

Thirty years have passed since the mind-blowing publication in 1987 of the Vermont Longitudinal Study of Persons with Severe Mental Illness (Harding, Brooks, Ashikaga, Straus, & Breier), which led to Harding and Zahniser’s 1994 article, Empirical Correction of Seven Myths about Schizophrenia with Implications for Treatment. We need to systematically review what we know and what we do not know in the light of new evidence. We need to find ways to communicate the knowledge derived from academic research on schizophrenia and psychosis to professionals working with this population, and to people with schizophrenia and their families. Thus can we begin to break down the rock-solid prejudices that have been rooted in humanity for centuries.

Utilizing engineering principles, the style was completed considering cost, utility, simplicity of use, durability, and performance. The current study was planned to evaluate comparative anti-inflammatory, analgesic and anti-pyretic activities of two newly synthesized organo-antimony (v) ferrocenyl benzoate derivatives with piroxicam. Anti-microbial activity of these compounds was also screened against two microorganisms. Analgesic effect of test compounds was evaluated by formalin-induced paw licking test in mice. The test compounds at 50 and 100 mg/kg b.w. doses exhibited significant (p<0.001) reduction of paw licking in treated mice comparable with standard drug piroxicam. Anti-inflammatory activity was assessed against carrageenan-induced paw oedema. The compound A produced anti-inflammatory effects comparable with standard piroxicam in dose dependent manner whereas compound B showed better effects than piroxicam at dose of 100 mg/kg body weight. To investigate anti-pyretic activity, fever was induced by administration of Brewers yeast in mice. Compound A showed highly significant inhibition of pyrexia (p<0.001) comparable to piroxicam after 3 hours while compound B (50 and 100 mg/kg) produced relatively lower anti-pyretic effect than standard drug. Antibacterial activity determined by disc diffusion method showed that compound B was relatively more effective than compound A against Staphylococcus aureus and Klebsiella pneumoniae. It is conceivable that both the tested compounds possessed anti-inflammatory, analgesic, anti-pyretic and anti-microbial effects even after the structural modification of parent compound.Bangladesh Pharmaceutical Journal 19(1): 15-24, 2016

Abstract Surra's disease is caused by Trypanosoma evansi parasite has been established as one of the strategic infectious animal diseases. Drug resistance in this case is one of the major challenges in handle and control them. The aim of this study is to evaluate the provision drug resistance diminazene aceturate (Tryponil®) on Trypanosoma evansi isolate from Pemalang and Brebes Central Java province with in vivo test in mice. Total 50 mice, BALB / c strain, male, 2 months, body weight ± 30 gram are obtained from LPPT-UGM, adapted for one week. Mice were divided into 10 groups consist of 5 each. Each mouse was infected with Trypanosoma evansi by intraperitonial route. Treatment was given when mice had reached the level of parasitemia 108 – 109 trypanosoma / mL of blood this was predicted 24 hours post-infection (Eisler et al., 2001). The administration of the drug tryapanosidal was done intraperitonial with doses 1mg/kg, 3mg / kg, 5 mg / kg and 7mg / kg. Observation of parasitemia did every 2 times in one week till 60 days of observation. Parasitemia observation was performed using 3 techniques. The first method was native examination used a microscope, if the negative results would be followed by MHCT (Microhaematocrit centrifugation Technique) and BCT (Buffy Coat Technique) according to OIE (2012). Data obtained from the treatment group were the level of parasitemia, the number of deaths and the number of live mice from each test dose. The results are analysed by standard logit or probit. The results of this study showed the effects of the drug Dimianzene aceturate on both isolates varied. On Brebes Isolate was effective at doses of 7 mg / kg BW (100%) and 5mg / kg BW (80%), whereas in the effective dose Pemalang isolate at 3 mg dose / kg BW (80%), 5 and 7 mg / kg BW (100%). While at the lowest dose of 1 mg / kg obtained a level of effectiveness of 0% in both isolates. It could be concluded that both isolates have different pathogens and indicate resistance subpopulation to diminazene aceturate.Keywords : diminazene aceturate, in vivo, resistance, Trypanosoma evansi.

This study was undertaken to evaluate the prophylactic potential of proteoglycan (PG) administration in experimental arthritis. Female BALB/c retired breeder mice received two (2xPG50 and 2xPG100 groups) or three (3xPG50 group) intraperitoneal doses of bovine PG (50 μg or 100 μg) every three days. A week later the animals were submitted to arthritis induction by immunization with three i.p. doses of bovine PG associated with dimethyldioctadecylammonium bromide adjuvant at intervals of 21 days. Disease severity was daily assessed after the third dose by score evaluation. The 3xPG50 group showed significant reduction in prevalence and clinical scores. This protective effect was associated with lower production of IFN-γand IL-17 and increased production of IL-5 and IL-10 by spleen cells restimulatedin vitrowith PG. Even though previous PG administration restrained dendritic cells maturation this procedure did not alter the frequency of regulatory Foxp3+T cells. Lower TNF-αand IL-6 levels and higher expression of ROR-γand GATA-3 were detected in the paws of protected animals. A delayed-type hypersensitivity reaction confirmed specific tolerance induction. Taken together, these results indicate that previous PG inoculation determines a specific tolerogenic effect that is able to decrease severity of subsequently induced arthritis.

This article focuses on describing the implementation of mind mapping technique to increase students’ motivation in writing descriptive text. An English teacher and 36 students in a Junior high school in Brebes participated in this study. A qualitative design was used, and data were collected through classroom observation, questionnaire and interview. This study found that the teacher did three steps in implementing mind mapping: giving students image of mind mapping, designing the writing task, and asking students to finish the writing task. Meanwhile, interview data indicated that most students enjoyed learning descriptive text by means of mind mapping. Regarding students’ motivation, data from observation revealed that most students enjoyed the activities, as seen from their enthusiasm. Likewise, questionnaire data indicated that most students gave positive perceptions towards the use of mind mapping. Most of them strongly agree with the statement that this strategy can make the learning process become imaginative. In short, it can be concluded that the teacher has successfully applied mind mapping technique. It can make the students enjoy and more interested in learning descriptive text. Therefore, the writers suggest that teachers apply various strategies in teaching and learning process, especially mind mapping, to create an enjoyable classroom atmosphere. Meanwhile, other researchers are recommended to discover the use of mind mapping in teaching other text types

Abstract Abstract 3158 In addition to its well-known role in host defense, perforin-dependent cytotoxicity also has a critical immune regulatory function. Genetic defects of perforin underlie many cases of hemophagocytic lymphohistiocytosis (HLH), a fatal disorder of extreme immune activation. While the initial treatment of HLH is largely focused on suppressing dangerous inflammation, hematopoietic cell transplantation (HCT) is required for long-term correction of the underlying immune regulatory defect. Unfortunately, mortality after HCT in patients with HLH is unusually high for a non-malignant disorder, ranging from 20 to 50%. We developed a model of HLH in which perforin deficient (prf-/-) mice develop all of the features of HLH after challenge with lymphocytic choriomeningitis virus (LCMV). Recent studies from our group have demonstrated that transplantation of wild type (WT) bone marrow into prf-/- mice rescues them from the subsequent development of HLH after challenge with LCMV. In this model, perforin-dependent immune regulation appears to function in trans, with a critical threshold of 10% WT chimerism required to protect animals from HLH. This low threshold is consistent with clinical reports of very limited numbers of patients with long term mixed chimerism after HCT who appear to be protected from HLH recurrence. These data have also provided a clear target for gene therapy efforts. We have begun to test the feasibility of autologous gene correction using lentiviral vectors in this model system. Hematopoietic stem and progenitor cells (HSC) from prf-/- mice were transduced with lentivirus vectors expressing perforin under the control of ubiquitous or tissue-specific promoters. When transduced HSC were transplanted into lethally irradiated prf-/- mice, we have observed levels of in vivo gene correction ranging from 10–80%, depending on the promoter and transduction protocols utilized. We find that the transduced CD8 T cell and NK cell progeny display significant correction of their cytotoxic defects. Challenge of gene corrected prf-/- mice with LCMV has revealed significant correction of the underlying immune regulatory defect and protection from the development of HLH. However, reestablishing normal perforin-dependent immune regulation with gene corrected prf-/- HSC's appears to be somewhat less efficient than with mixed WT:prf-/- chimerism. Thus, further optimization of perforin gene control by lentiviral vectors appears to be warranted. Results of ongoing studies related to vector optimization and reestablishing physiologic perforin-mediated immune regulation will be presented. Because normal perforin expression in only a fraction of lymphocytes is sufficient to rescue individuals from HLH, this disorder is an attractive target for gene therapy approaches. Demonstration of long term gene correction and rescue from HLH in our animal model will pave a path for translational efforts testing gene therapy for HLH in clinical trials. A theory of traumatic memory was developed by Breuer and Freud in “Studies on Hysteria” based on Charcot’s theory on the traumatic origin of mental disorders. This was again developed in DSM-III in 1980 with the introduction of posttraumatic stress disorder (PTSD), where traumatic memory was conceptualized as a core of the syndrome and set in causal relationship with its manifestations. This implied that “trauma” tended to be seen as something static and reified, like a “thing” in the mind. It is shown that this conceptualisation diverts attention from the dynamic and reorganizing processes in the traumatized person’s mind, body and relations to others. The psychoanalytic conceptualization includes a wider spectre of the manifestations in posttraumatic conditions and it differs in the understanding of underlying processes. What is common for both childhood and adult trauma and their posttraumatic manifestations are deficiencies in symbolization processes related to the traumatic experiences. The signal anxiety function fails and the ego is overwhelmed by automatic or annihilation anxiety. The ability to distinguish between real danger and neutral stimuli which function as triggers fails. As anxiety related to trauma cannot be mentalized, fragments of the self are split off and evacuated. These experiences remain as fragmented bits and pieces that can express themselves in bodily pain, dissociated states of mind, nightmares and relational disturbances. Thus, the consequences of psychological trauma may be conceptualised as processes characterised by splitting off of not mentalized inner objects/introjects and parts of the mental apparatus. These processes are illustrated with a short clinical vignette.

After skin wounding, the repair process is initiated by the release of growth factors, cytokines, and bioactive lipids from injured vessels and coagulated platelets. These signal molecules induce synthesis and deposition of a provisional extracellular matrix, as well as fibroblast invasion into and contraction of the wounded area. We previously showed that sphingosine-1-phosphate (S1P) triggers a signal transduction cascade mediating nuclear translocation of the LIM-only protein Fhl2 in response to activation of the RhoA GTPase (Muller, J.M., U. Isele, E. Metzger, A. Rempel, M. Moser, A. Pscherer, T. Breyer, C. Holubarsch, R. Buettner, and R. Schule. 2000. EMBO J. 19:359–369; Muller, J.M., E. Metzger, H. Greschik, A.K. Bosserhoff, L. Mercep, R. Buettner, and R. Schule. 2002. EMBO J. 21:736–748.). We demonstrate impaired cutaneous wound healing in Fhl2-deficient mice rescued by transgenic expression of Fhl2. Furthermore, collagen contraction and cell migration are severely impaired in Fhl2-deficient cells. Consequently, we show that the expression of α-smooth muscle actin, which is regulated by Fhl2, is reduced and delayed in wounds of Fhl2-deficient mice and that the expression of p130Cas, which is essential for cell migration, is reduced in Fhl2-deficient cells. In summary, our data demonstrate a function of Fhl2 as a lipid-triggered signaling molecule in mesenchymal cells regulating their migration and contraction during cutaneous wound healing.

Hal ini diindikasikan dengan dirumuskannya sebuah konsep pengembangan blue economy untuk kelautan dan perikanan Indonesia. Pada tahun 2013 Kementerian Kelautan dan Perikanan melalui Balai Besar Penelitian Sosial Ekonomi Kelautan dan Perikanan (BBSEKP) melakukan identifikasi penerapan prinsip blue economy pada lokasi sasaran pengembangan Klinik Iptek Mina Bisnis (KIMBIS) di wilayah pesisir Kabupaten Brebes, Jawa Tengah. Penelitian dilakukan dengan menggunakan data primer dan sekunder yang dianalisis dengan pendekatan deskriptif-kualitatif. Hasil penelitian menunjukkan bahwa terdapat tiga usaha perikanan dan kelautan yang telah menerapkan prinsip-prinsip blue economy di lokasi penelitrian, yaitu: usaha longyam, polikultur, dan usaha pengolahan kulit ikan menjadi kerupuk. Untuk lebih meningkatkan tingkat penerapan prinsip blue economy pada ketiga usaha tersebut, perlu dukungan pemerintah baik berupa sarana maupun prasarana yang lebih baik dengan disertai upaya pendampingan yang lebih intensif.

In the context of hospital building, inpatient care has large portion in comparasion of the other facility areas within hospitals related to efficiency, such as outpatient services, emergency room, diagnostic and especially inpatient function group services. Even though inpatient care group do not have specific requirements for detailed design and building equipment, it requires efficiency considerations related to correlation with room layout. It is expected that by considering the level of efficiency of service to patients, design can fulfill technical requirements of health and medical aspects. Regarding designs for district-level private hospitals, demand optimization of placement and layout of inpatient care became main topic in this research. This is related to the value of investment in building area development and types of services provided according to inpatient services class. The method used is comparative study of two (2) private hospital design to find the mind factors that most influence of optimization of inpatient layout. The role of different isoforms of nitric oxide synthase (NOS) in the gastric mucosal hyperemia, induced by 155 mM luminal hydrochloric acid (pH ≈ 0.8) without a barrier breaker, was investigated. Rats were anesthetized with Inactin (120 mg/kg ip), and mice were anesthetized with Forene (2.2% in 40% oxygen gas at 150 ml/min); the gastric mucosa was exteriorized. Gastric mucosal blood flow was measured with laser-Doppler flowmetry (LDF) in rats treated with Nω-nitro-l-arginine (l-NNA; unspecific NOS inhibitor), l- N6-(1-iminoethyl)lysine [l-NIL; inducible (i) NOS inhibitor], or S-methyl-l-thiocitrulline [SMTC; neuronal (n) NOS inhibitor], 10 mg/kg, followed by 3 mg · kg–1 · h–1 iv, in iNOS-deficient (–/–) and nNOS(–/–) mice. mRNA was isolated from the gastric mucosa in iNOS(–/–) and wild-type (wt) mice, and real-time RT-PCR was performed. The effect of 155 mM acid on gastric mucosal permeability was determined by measuring the clearance of 51Cr-EDTA from blood to lumen. LDF increased by 48 ± 13% during 155 mM HCl luminally, an increase that was abolished by l-NNA, SMTC, or l-NIL. In iNOS wt mice, LDF increased by 33 ± 8% during luminal acid. The blood flow increase was attenuated substantially in iNOS(–/–) mice. RT-PCR revealed iNOS mRNA expression in the gastric mucosa in the iNOS wt groups. The blood flow increase in response to acid was not abolished in nNOS(–/–) mice (nNOS-sufficient mice, 39 ± 18%; heterozygous mice, 25 ± 19%; –/– mice, 19 ± 7%). Mucosal permeability was transiently increased during 155 mM HCl. The results suggest that iNOS is constitutively expressed in the gastric mucosa and is involved in acid-induced hyperemia, suggesting a novel role for iNOS in gastric mucosal protection.

Oxidative stress in diabetes mellitus triggers activation and increases in platelet counts that play a role in the formation of platelet plugs in the blood clotting process. If excessive blood clots occur, there is a risk of cardiovascular disease. Red seaweed extract contains carotenoids, phenols and their derivatives, fikobilin pigments, polysaccharide sulfates, and vitamins. The biggest phenol compound is flavonoids which are compounds that can produce platelet antia-gregation effects. The purpose of this study was to examine the effect of giving red seaweed extract to platelet counts in diabetic mice. The study group consisted of the treatment group of red seaweed extract, normal control, positive control and negative controls. The dosage of seaweed extract as much as 10 mg / 20 grBB is given in condition. The calculation of platelet count is carried out directly with the Brecher Cronkite method using a calculated room Improved Neubauer. The One Way ANOVA test results showed a significance value (p <0.05), that is, the group of diabetic mice given red seaweed extract had a lower platelet count of 280000 μl compared to diabetic mice given metformin as much as 379000 μl. The decrease is due to flavonoids, saponins, alkaloids, β carotene, vitamins and minerals able to stabilize the production of megakaryocytes and platelets, prevent the destruction of pancreatic β cells and prevent free radicals. It was concluded that red seaweed extract can reduce platelet counts of diabetic mice near normal values.

Aging is associated with stiffening of the large elastic arteries and consequent increases in systolic blood pressure (SBP), which together increase cardiovascular disease risk; however, the upstream mechanisms are incompletely understood. Using complementary translational approaches in mice and humans, we investigated the role of the gut microbiome-derived metabolite trimethylamine N-oxide (TMAO) in age-related aortic stiffening and increased SBP. Aortic stiffness was measured using carotid-femoral or aortic pulse wave velocity (PWV) in humans and mice, respectively. Study 1: Plasma TMAO concentrations were elevated ( P <0.001) in healthy middle-aged to older (6.3±5.8 µmol/L) versus young (1.8±1.4 µmol/L) humans and positively related to carotid-femoral PWV ( r 2 =0.15, P <0.0001) and SBP ( r 2 =0.09, P <0.001), independent of traditional cardiovascular risk factors. Study 2: Dietary supplementation with TMAO increased aPWV in young mice and exacerbated the already elevated aPWV of old mice, accompanied by increases in SBP of ≈10 mm Hg in both groups. TMAO-supplemented versus control-fed mice also had higher intrinsic mechanical stiffness of the aorta (stress-strain testing) associated with higher aortic abundance of advanced glycation end-products, which form crosslinks between structural proteins to promote aortic stiffening. Study 3: Ex vivo incubation of aortic rings with TMAO increased intrinsic stiffness, which was attenuated by the advanced glycation end-products crosslink breaker alagebrium and prevented by inhibition of superoxide signaling. TMAO induces aortic stiffening and increases SBP via formation of advanced glycation end-products and superoxide-stimulated oxidative stress, which together increase intrinsic wall stiffness. Increases in circulating TMAO with aging represent a novel therapeutic target for reducing risk of aortic stiffening-related clinical disorders.

Practical relevance: Many veterinarians – even those engaged in small animal reproduction – are uncomfortable with taking care of cat breeders or being in charge of catteries. Likely reasons for this are that feline reproduction is largely under-represented in undergraduate and graduate reproduction teaching, as well as in postgraduate education and science, whereas cat breeders themselves are often well informed and actively share knowledge with other cat breeders via the internet and social media. Clinical challenges: A variety of problems can exist within a cattery, and collaboration between veterinarian and breeder to solve these is ultimately beneficial for the breeder, for the veterinarian, and for the health and wellbeing of the individual cats and of the cattery as a whole. Aim: This review presents a comprehensive overview of aspects of cattery management that might negatively impact reproduction, including sanitation, hygiene and infectious disease control. It also discusses monitoring of reproductive performance, breeding recommendations, and the diagnostic and therapeutic approach to some common and specific problems. Equipment and technical skills: Reproductive management in the cattery requires no specialised equipment, as such. What it does need is an open mind, in terms of seeing how things are done, an open ear, for listening to the breeder, and the veterinarian’s clinical skills in palpation, auscultation and, where required, further examination of the animals. Keeping abreast of the latest information on infectious diseases, disinfection and genetics ensures proper advice is provided. Evidence base: Current knowledge of reproductive management in catteries is summarised in order to apply an evidence-based approach, whenever possible. Notwithstanding, much of the information remains empirical.

Serum amyloid A (SAA) is an acute phase protein whose expression is markedly up-regulated during inflammation and infection. The physiological function of SAA is unclear. In this study, we reported that SAA promotes cellular cholesterol efflux mediated by scavenger receptor B-I (SR-BI). In Chinese hamster ovary cells, SAA promoted cellular cholesterol efflux in an SR-BI-dependent manner, whereas apoA-I did not. Similarly, SAA, but not apoA-I, promoted cholesterol efflux from HepG2 cells in an SR-BI-dependent manner as shown by using the SR-BI inhibitor BLT-1. When SAA was overexpressed in HepG2 cells using adenovirus-mediated gene transfer, the endogenously expressed SAA promoted SR-BI-dependent efflux. To assess the effect of SAA on SR-BI-mediated efflux to high density lipoprotein (HDL), we compared normal HDL, acute phase HDL (AP-HDL, prepared from mice injected with lipopolysaccharide), and AdSAA-HDL (HDL prepared from mice overexpressing SAA). Both AP-HDL and AdSAA-HDL promoted 2-fold greater cholesterol efflux than normal HDL. Lipid-free SAA was shown to also stimulate ABCA1-dependent cholesterol efflux in fibroblasts, in line with an earlier report (Stonik, J. A., Remaley, A. T., Demosky, S. J., Neufeld, E. B., Bocharov, A., and Brewer, H. B. (2004) Biochem. Biophys. Res. Commun. 321, 936–941). When added to cells together, SAA and HDL exerted a synergistic effect in promoting ABCA1-dependent efflux, suggesting that SAA may remodel HDL in a manner that releases apoA-I or other efficient ABCA1 ligands from HDL. SAA also facilitated efflux by a process that was independent of SR-BI and ABCA1. We conclude that the acute phase protein SAA plays an important role in HDL cholesterol metabolism by promoting cellular cholesterol efflux through a number of different efflux pathways.

Indigo naturalis, which is extracted from the leaves and branches of Baphicacanthus cusia (Nees) Bremek, has traditionally been used to treat psoriasis. The current study aimed to examine a new mechanism of the components of indigo naturalis, including indirubin, indigo, and tryptanthrin. The anti-psoriatic effects were assessed by the proliferation biomarkers (Ki67, K16), cell cycle progression, ROS production, and interleukin profiling (ICAM-1, TNF-α, IL-6, and IL-8) in IL-22-treated HaCaT cells. Among the components, indirubin significantly decreased intracellular ROS production and lowered the production of ICAM-1, TNF-α, and IL-6 in IL-22-treated HaCaT cells. Indirubin, indigo, and tryptanthrin could decrease the proportion of Ki67-positive cells, but only indirubin decreased the proportion of cells entering the S phase and suppressed the expression of cyclin D1 and cyclin E1 in IL-22-treated HaCaT cells. Indirubin significantly suppressed the phosphorylation of STAT3 and ERK. In vivo, IL-22 was intradermally injected into mouse ears for six days and topically treated with 0.1% or 1% indirubin. In the IL-22-injected mice, treatment with indirubin inhibited epidermal hyperplasia. Immunohistochemistry and western blot analysis demonstrated the downregulation of K16 expression in psoriatic lesions. These results suggest that indirubin, which is a major component of indigo naturalis, may have therapeutic potential in an IL-22-induced psoriasis model.

A two-month-old, male intact, mixed-breed cat weighing 0.6 kg was presented with respiratory distress and anorexia. From the transthoracic echocardiographic, reduced fractional shortening (FS) and increased endocardial echogenicity were recognised with severe congestive heart failure (CHF). The kitten was administered an antibiotic and pimobendane under oxygen supplementation in an ICU cage. However, the respiratory condition worsened and the cat died the next day, and the subsequent necropsy and histopathology examinations confirmed endocardial fibroelastosis (EFE). There is a lack of information regarding the antemortem cardiac function evaluated by tissue Doppler imaging (TDI) in EFE cases. We report on the echocardiographic findings including the TDI in the EFE cat with a concomitant necropsy and histopathology confirmation in this paper. The echocardiographic findings showed presence of a ventricular false tendon within the left ventricle, a decrease in the left ventricular contractility (FS 11.1%, and a marked CHF). In this case, the echocardiographic findings were consistent with the human counterpart. However, these findings were like those of dilated cardiomyopathy and, hence, non-specific to EFE. As a result, veterinarians should keep in mind that endocardial fibroelastosis might be a possible reason for respiratory distress resulting from CHF with a low fractional shortening in young cats.
