Abstract Abstract 1468 Background: Increased YB-1 expression correlates with poor prognosis, drug resistance and metastasis in several different cancers including B cell lymphoma. Phosphorylation and nuclear localization of YB-1 in response to growth factors leads to increased survival through expression of proteins such as survivin and multidrug resistance protein 1. Until now, its role in leukemia has not been investigated. We hypothesized that YB-1 expression is aberrantly regulated in pediatric pre-B acute lymphoblastic leukemia (pre-B ALL), and that YB-1 may be activated downstream of IL-7. This cytokine facilitates the differentiation and survival of pre-B cells and has been implicated in the drug resistance of pre-B ALL. Methods: YB-1 and IL-7Ra protein expression was investigated by flow cytometry in normal pre-B cells (CD19+CD10+CD20−), and mature B cells (CD19+CD10−CD20+) as well as diagnostic and relapsed pre-B ALL (CD19+CD10+/−). Cell surface and cytoplasmic expression was quantified by mean fluorescent intensity (MFI). Bone marrow from healthy donors was used as a source of normal pre-B cells, while mature B cells were derived from PBMCs; leukemic cells at presentation and relapse were obtained following local IRB approval and informed consent. Activation of YB-1 downstream of IL-7 stimulation (25 ng/ml) was examined in pre-B ALL cell lines or NSG (NOD scid gamma) mice-expanded pre-B ALL by Western blotting using anti-phosphoYB-1(S012). Pre-B ALL cell lines used in these experiments were 697, 380, RCH and RS-4;11. Signaling pathways were investigated by pre-treatment of cells with pharmacological inhibitors followed by Western analyses. For the transient overexpression of YB-1, pEGFP or a pEGFP/YB-1 fusion protein was electroporated into freshly isolated mature B cells (which have a low basal expression of YB-1) and YB-1 and IL-7Ra expression was assessed by flow cytometry after 24 h. Results: While intracellular YB-1 expression was significantly higher in leukemia samples at presentation compared to normal pre-B cells, the highest YB-1 levels were found in relapsed pre-B ALL (see figure, part A). All examined pre-B ALL cell lines had levels comparable to those of the relapse samples. Similarly, surface IL-7Ra (CD127) levels (MFI medians; upper-lower range) were increased in onset (221; 150–286), and relapsed (1840; 651–2030) ALL compared to normal pre-B cells (528; 333–2673). Overexpression of YB-1-GFP in normal mature B cells resulted in increased expression of IL-7Ra (see figure, part B), suggesting an link between the YB-1 and IL-7 signaling pathways. Activated YB-1 is phosphorylated on S102 and relocated to the nucleus. Addition of IL-7 to pre-B ALL cell lines led to phosphorylation of YB-1 within 30 min. Similar results were shown for patient-derived, NSG mice-expanded pre-B ALL samples. Intracellular immunostaining using Imagestream technology (Amnis) showed that IL-7 treatment of pre-B ALL cell lines increased nuclear YB-1 levels 4-fold. As PI3K and MEK1 are involved in signaling downstream of IL-7, we investigated their role in YB-1 signaling in both pre-B ALL cell lines and NSG-mouse expanded pre-B ALL using pharmacological inhibitors. Western analyses showed that inhibition of PI3K using LY294002 did not prevent IL-7-mediated phosphorylation of YB-1 but the MEK1 inhibitor U0126 did, indicating the involvement of MAPK (see figure, part C). Conclusion: We show that YB-1, which is highly expressed in pediatric pre-B ALL compared to normal pre-B cells, is expressed at even higher levels after relapse. We demonstrate a link between the YB-1 and IL-7 signaling pathways which could offer a novel target for the treatment of refractory leukemia. AbstractMelanoma is the most lethal dermal tumor, and a high recurrence rate and skin defects are two main serious problems. An antimelanoma material,which effectively inhibits tumor recurrence and possesses excellent biocompatibility, is urgently needed to treat melanoma. In this study, we developed a novel antitumor Yb3+ [Yb(NO3)3]containing chitosan hydrogel (Yb-CS hydrogel) by dissolving Yb(NO3)3 and chitosan in acetic acid solution and forming composite hydrogels by a freeze-drying process after adding NaOH to the mixed solution. In vitro studies demonstrated that the Yb3+ produces effect of inducing cell death in Yb-CS hydrogel. Moreover, we found that the Yb-CS hydrogel inhibited a focal adhesion kinase (FAK)-dependent signaling pathway and induced B-16 cell anoikis. However, the Yb-CS hydrogel was less effective on L929 normal mouse dermal cells. In vivo studies showed that the Yb-CS hydrogel inhibited the recurrence of melanoma in a mouse bare xenograft tumor model. We concluded that the Yb-CS hydrogel could potentially be used in the antimelanoma field, especially in the inhibition of melanoma recurrence.

Bulky monomeric lanthanoid(III) aryl oxides Ln(Odpp)3(Odpp = 2,6- diphenylphenoxide; Ln = Nd, Sm, Er, Yb or Lu) have been prepared by reaction between the lanthanoid metal, Hg(C6F5)2, and HOdpp in tetrahydrofuran (thf), and Yb(Odpp)3 has also been obtained from an analogous reaction of (PhCC)2Hg. Cleavage of Ln(η5-C5H5)3 (Ln = Nd or Yb) with HOdpp also yields Ln(Opdd)3. The aryl oxides have been isolated as [Ln(Odpp)3(thf)2] (Ln = Nd, Sm or Yb) or as the homoleptic complexes Ln(Odpp)3 (Ln = Nd, Er, Yb or Lu). Fusarium crown rot caused by Fusarium pseudograminearum is one of the most devastating diseases of wheat worldwide causing major yield and economic losses. In this study, strain YB-15 was isolated from soil of wheat rhizosphere and classified as Bacillus subtilis by average nucleotide identity analysis. It significantly reduced Fusarium crown rot with a control efficacy of 81.50% and significantly improved the growth of wheat seedlings by increasing root and shoot fresh weight by 11.4% and 4.2%, respectively. Reduced Fusarium crown rot may have been due to direct antagonism by the production of β-1, 3-glucanase, amylase, protease and cellulase, or by the ability of B. subtilis YB-15 to induce defense-related enzyme activities of wheat seedlings, both alone and in seedlings infected with F. pseudograminearum. Improved plant growth may be related to the ability of B. subtilis YB-15 to secrete indole acetic acid and siderophores, as well as to solubilize phosphorus. In addition, the genome of strain YB-15 was determined, resulting in a complete assembled circular genome of 4,233,040 bp with GC content of 43.52% consisting of 4207 protein-encoding genes. Sequencing the B. subtilis YB-15 genome further revealed genes for encoding carbohydrate-active enzymes, biosynthesis of various secondary metabolites, nutrient acquisition, phytohormone production, chemotaxis and motility, which could explain the potential of strain YB-15 to be plant growth-promoting bacteria and biological control agent. B. subtilis YB-15 appears to be a promising biocontrol agent against Fusarium crown rot as well as for wheat growth promotion.

The Y box is a conserved sequence in the promoter of major histocompatibility complex (MHC) class II genes, which contains a CCAAT sequence (CCAAT box). Previously, we partially purified the DNA-binding protein that recognizes the Y box of the I-Aβ gene and showed that it consisted of two components (factors A and B) both of which were necessary for optimal DNA binding. The genes for the heteromeric protein NF-Y (NF-YA and NF-YB), which binds to the I-Eα Y box have been cloned. We subsequently isolated the genes for NF-YA and NF-YB using oligonucleotides designed from the published sequences. NF-YA and NF-YB were tested for binding to the I-Aβ and I-Eα Y boxes. While neither NF-YA or NF-YB alone bound to the Y box, when the components were mixed the complex bound to the I-Aβ Y box with high affinity. Moreover, NF-YA and NF-YB could be complemented for binding to DNA by factor B or factor A, respectively. These results suggest that the active binding protein is NF-YA in factor A extracts and NF-YB in factor B extracts. Finally, antibodies against NF-YA and NF-YB were shown to induce a supershift when nuclear extracts were added to the double-stranded oligodeoxynucleotide covering the Y box of the I-Aβ gene. Antisense expression constructs of both NF-YA and NF-YB were made and their effect on expression from the I-Aβ promoter was tested. Either antisense construction, when transfected into cells, lowered the expression of a reporter gene linked to the I-Aβ promoter. This study provides direct evidence of the identification of NF-YA and NF-YB as the previously described factors A and B. Moreover, these results strongly implicate NF-Y in the expression of the MHC class II gene I-Aβ.

Herzinsuffizienz mit konsekutivem Herzversagen ist ein zentrales kardiovaskuläres Problem der heutigen Bevölkerung.Ursächlich ist insbesondere eine progrediente Herzhypertrophie. Die Calcium-Calmodulin abhängige Phosphatase Calcineurin (CnR) spielt hierbei in der Pathogenese eine entscheidende Rolle. CnR wird über seine Calciumbindungsstellen an der regulatorischen Untereinheit Calcineurin B (CnB) aktiviert.Um zu untersuchen, inwieweit CnB bei der Hypertrophie verschiedener Ätiologien reguliert wird, wurde in linksventrikulären Myokardbiopsien von Patienten mit Aortenstenose (AS= 14) bzw. aus explantierten Herzen mit Dilatativer Kardiomyopathie (DCM=27) und Koronarer Herzerkrankung (KHK=7) der mRNA-und Proteingehalt von CnB bestimmt und mit der Expression von ANP und BNP korreliert. Als Kontrollgruppe dienten 15 abgelehnte Spenderherzen mit normaler systolischer Funktion und gesunder Morphologie. In den Herzen der Kontroll-, DCM-, und KHK-Gruppen wurde der linksventrikuläre Fibrosegehalt bestimmt. Hierzu wurden eine extern standardisierte Real-Time-PCR-Technik und ein etabliertes Western Blot Verfahren angewandt. Die Ergebnisse werden im Median ± 25%/75%-Perzentile angegeben und mit dem Mann-Whitney-Test bzw. Korrelationsanalysen nach Spearman berechnet. Heart failure is a central cardiovascular problem for the current population. Cardiac hypertrophy is a central factor. The calcium-Calmodulin dependent phosphatase Calcineurin (CnR) plays a crucial role in the pathogenesis. CnR is activated via its calcium-binding site in the regulatory subunit Calcineurin B (CnB). In order to examine, to what extent CnB is regulated in different aetiologies of hypertrophy, we analysed CnB´s mRNA and protein in left ventricular samples from patients with aortic valve stenosis (AS = 14) and from explanted hearts with dilated (DCM=27) and ischemic (ICM=7) cardiomyopathy and correlated them with the expression of ANP and BNP. As a control, 15 rejected donor hearts with normal systolic function and non-pathologic changed morphology were used. Fibrosis of the left ventricle was determined in three groups: control , DCM and ICM. Therefore, we used an externally standardized real-time PCR and an established Western Blot. Die Therapie des häufigsten primären Hirntumors bei Erwachsenen, des Glioblastoms, gestaltet sich aufgrund der relativen Resistenz gegen Bestrahlung und Zytostatika schwierig und resultiert in einer äußerst schlechten Prognose. Ziel dieser Arbeit war die Identifikation von zellulären Faktoren, die bei einer Behandlung mit dem neuartigen onkolytischen Virus Ad5-Delo3RGD (ΔE1A-13S, ΔE19K, ΔE3, zusätzliches RGD-Motiv) eine Bedeutung hinsichtlich des Ansprechens auf die Therapie haben. Dazu erfolgte nach der Transduktion von Gliomzellen mit adenoviralen Vektoren, mit unterschiedlichem E1A-Status, eine Analyse der Modulation der globalen Genexpression. Die E1A-Deletante, Ad5-Ad312, war replikationsinkompetent in Gliomzellen und hatte infolgedessen nur einen marginalen Einfluss auf Genebene und keine biologischen Effekte auf Proteinebene. Das Wildtypvirus (Ad5-wt) mit intakter und das onkolytische Virus (Ad5-Delo3RGD) mit mutierter E1A-Region induzierten eine starke Modifikation der zellulären Genexpression in Gliomzellen. Die Transduktion der Gliomzellen mit Wildtypvirus bzw. onkolytischen Virus resultierte in einer bis zu 60 %-igen Hemmung der Sekretion des Angiogenesefaktors VEGF. Fernerhin konnte in Glioblastomzellen gezeigt werden, dass Ad5-wt und Ad5-Delo3RGD eine 50 bis 60 %-ige Inhibition der Sekretion von TGF-β2, sowie eine bis zu 65 %-ige Hemmung der Transkriptionsaktivität des TGF-β-Signalwegs induzierten. Damit reprimieren besagte Viren zelluläre Faktoren, deren Expression im Gliom mit einer schlechten Prognose korreliert ist. Aufgrund dieser Daten konnte für maligne Gliome ein Modell von Ad5-Delo3RGD-Therapie relevanten zellulären Faktoren entwickelt werden, welches den Zusammenhang zwischen TGF-β2, VEGF bzw. MIR-181 und der Virotherapie verdeutlicht. Mit Hilfe dieses Modells steigt das Verständnis um die antineoplastische Aktivität des onkolytischen Virus, und ermöglicht damit eine Verbesserung der Ad5-Delo3RGD-basierten Therapie.
Glioblastoma is the most common type of brain tumor among adults. The therapy is very difficult, due to its relative resistance to radiation and zytostatica, and often results in fatal prognosis. The main objective of this thesis was the identification of cellular factors associated with therapy response to adenoviral treatment using the newly developed oncolytic virus Ad5-Delo3RGD. Therefore, after viral transduction of glioma cells with adenoviral vectors, using E1A-wildtype and -mutant viruses, an analysis of the modulation of mRNA expression profiles was conducted. The E1A deletion mutant Ad5-Ad312 was replication-deficient in glioma cells and therefore had only marginal influence of host gene transcription level and no biological effect on protein level. Both, wildtype adenovirus type 5 (Ad5-wt), with intact E1A, and oncolytic virus (Ad5-Delo3RGD), with mutated E1A, induced strong modifications of gene expression profiles in glioma cells. The transduction of glioma cells with wildtype or oncolytic virus resulted in a 60 % reduced secretion of the angionesis factor Vascular Endothelial Growth Factor (VEGF). In addition it was shown that Ad5-wt and Ad5-Delo3RGD induced a reduced secretion of TGF-ß2 by 50 to 60 % and a repression of the SMAD2/SMAD3/SMAD4-specific transcription activity up to 65 %. Thus the viruses inhibit cellular factors, which expression is corelated to a weak prognosis. Based on this data a new model for malign glioma was developed, which illustrates the relationship between virotherapy and the cellular factors TGF-β2, VEGF and MIR-181, which are associated with treatment response to Ad5-Delo3RGD-based therapy. The model helps to understand the basic antineoplastic activity of the oncolytic virus and therefore should help to improve Ad5-Delo3RGD-based therapy.

Cette thèse porte sur l'étude par diffusion inélastique de neutrons des propriétés magnétiques de borures de terre rare : ceb#6, prb#6 et ybb#1#2. La difficulté de réalisation des expériences, notamment la faiblesse des signaux due en partie à la forte absorption des neutrons par le bore, nous a conduit à inclure, dans un programme d'analyse, la correction d'absorption. La partie principale de cette thèse a été l'étude des interactions magnétiques dans les phases ordonnées de ceb#6 et prb#6. Nous avons confirmé la présence d'interactions magnétiques quadrupolaires importantes, grâce à la mesure des excitations magnétiques dans la phase quadrupolaire de ceb#6 en appliquant un champ magnétique dont la valeur était comprise entre 0 et 6 t, et dans les phases double-k et simple-k basse température de prb#6. De plus nous avons montré qu'il semble exister une compétition entre les interactions magnétiques quadrupolaires et les interactions magnétiques dipolaires dans le compose ceb#6, alors que les interactions quadrupolaires dans prb#6 semblent au contraire renforcer la structure magnétique dipolaire. Les mesures effectuées avec le compose ybb#1#2 nous ont permis de donner une valeur de 60 k pour l'énergie du gap, cette valeur étant conforme à celles obtenues par des mesures de résistivité et d'optique. De plus nous proposons un schéma de niveau de champ cristallin, avec le niveau fondamental #8 et les niveaux excites #7 et #6 a respectivement 18 et 39 mev. En annexe, nous présentons également les résultats obtenus par diffusion inélastique de neutrons avec le compose bidimensionnel xy bani#2(po#4)#2. Un programme a été développé pour déconvoluer les spectres mesures de la fonction de résolution instrumentale à 4 dimensions. Les largeurs intrinsèques, ainsi déterminées, de la diffusion magnétique critique ne montrent aucune caractéristique des excitations non linéaires prévues par la théorie, mais peut être expliqué uniquement par une renormalisation des ondes de spin

In this research, homogeneous (R0.5A0.5)MnO3 (R = Gd, Yb; A = Ca, Sr) and (R0.5A0.5)NiO3 (A = Ce, Th) compounds with perovskite structure were synthesized by a polymeric citrate precursor method. (R0.5A0.5)TiO3 (A = Ca, Sr) was synthesized by a solid state method. (R0.5A0.5)MnO3 series samples have orthorhombic symmetry and (R0.5A0.5)NiO3 have monoclinic symmetry. The samples prepared by the polymeric citrate precursor method has a better homogeneity and the grain size is smaller than that prepared by the solid state method. Mn and Ti samples have M3+ and M4+ mixed valence state observed by XANES. The (R0.5A0.5)MnO3 samples are insulator, they has a spin-glass like property and the FC and ZFC susceptibility is irreversible. The (R0.5A0.5)NiO3 samples are paramagnetic metal. At 1 T magnetic field, both (R0.5A0.5)MnO3 and (R0.5A0.5)NiO3 have negative magnetoresistance (MR) ratio. The maximum MR ratio of (Gd0.5Ca0.5)MnO3 sample is found at 80-100 K,with a range of 60-65%. The maximum MR ratio of (R0.5A0.5)NiO3 sample is found at 180-200 K, with a range of 50- 60%. The (R0.5A0.5)TiO3 samples are paramagnetic Mott insulator.

In this thesis, series of (La1-xMx)2(Ce1-yNy)2O7 (M=Yb or N=V) were prepared by sol-gel method to study the influence of metal cation to their physical and electrochemical properties. For (La1-xYbx)2Ce2O7 series, single-phase were observed in XRD with a substitution range of 0 ≤ x ≤ 1 and their compositions were confirmed by ICP-AES/SEM-EDS. The refined cell constants decreased as the amount of substituted-Yb increased. According to the result from TPR, these materials react with hydrogen in the temperature of 400-750oC and their structures remain stable up to 900oC. Dense ceramic pellets for the materials were obtained through the optimal sintering condition, at 1600°C for 1 h under air, and the relative density of the pellets reached 95%. Total ionic conductivity increases significantly with x = 0.1 of substituted-Yb content. For (La0.9Yb0.1)2Ce2O7, the measurement of ionic conductivity via electrochemical impedance spectroscopy showed that the conductivities were in a range of 3.5×10-3 – 6.9×10-2 S/cm at 600-900oC. When x>0.1, the drop trend in total ionic conductivity with increasing Yb contents were attributed to the crystal phase transformation of pyrochlores, which could be speculated by XPS analysis. The total ionic conductivity rises as oxygen partial pressure increased, indicative of a p-type semiconductor. The electronic conductivity of (La1-xYbx)2Ce2O7 measured via four probe method were in a range of ~10-5 – 10-3 S/cm at 600-900oC, and exhibited no significant difference between the results measurement under O2 and H2. The Yb-substituted La2Ce2O7 exhibits high total conductivity and low electronic conductivity compare to that of electrolyte materials Y0.16Zr0.84O2 (YSZ), which is a potential electrolyte material for solid oxide fuel cell. For La2(Ce1-yVy)2O7 series, pure phase were observed in a range of 0 ≤ y ≤ 0.2 and their compositions were analyzed by ICP-AES/SEM-EDS. The refined cell constants decreased as V contents increased. From the TPR result, these materials started to react with hydrogen in the temperature range of 400-750oC and they exhibited structural stability up to 900oC. The dense ceramic pellets were sintered at 1500oC for 1h under air, and the corresponding relative density is over 95%. Total ionic conductivity increases with the substituted-V content. La2(Ce0.8V0.2)2O7 exhibited high total ionic conductivity of in the range of 3.7×10-3 - 10-1 S/cm at 600-900oC, which is higher than the total ionic conductivity of YSZ and similar to (La0.9Yb0.1)2Ce2O7.

Antes de retomar estos puntos conceptuales, comenzaré proponiendo una concepción inusualmente concreta de lo que podría significar «prioridad para los más desfavorecidos», basándome en el marco conceptual del socorro en casos de desastre. Luego, en la sección 2, después de discutir el significado de «exclusión social» (como relevante para la justicia), considero las similitudes y diferencias entre la fase de «recuperación» del socorro en casos de desastre y los esfuerzos (fuera del contexto del desastre) para reducir la exclusión social. Una diferencia clave es que la recuperación ante desastres se ocupa de regiones en las que el funcionamiento social se ha derrumbado por completo, mientras que la lucha contra la exclusión social se ocupa de restaurar la participación social de los individuos a niveles considerados normales o habituales para esa sociedad. En las secciones tercera y cuarta consideraré si la justicia social no requiere más que esta restauración relativa y si, en cambio, podría requerir la restauración de la participación social para producir los niveles más altos de capacidad sostenible para toda la población: a) dada su condición económica y social; recursos sociales en el momento y b) a medida que esos recursos se expanden en el tiempo (lo llamo un «óptimo igualitario»). As every school child knows, the difference between a solid and a liquid is that a liquid takes the shape of the container in which it is placed while the shape of a solid is independent of the shape of the container (providing the container is big enough). In other words, we must apply a force in order to change the shape of a solid. However, the thermodynamic functions described heretofore have no terms that depend on shape. In this chapter, we extend the thermodynamics discussed above to include such effects and therefore make it applicable to solids. However, since this is a thermodynamics, rather than a mechanics text, we focus more on the relationship between stress and thermodynamics rather than on a general description of the mechanical properties of solids. We start out discussion of mechanical deformation by describing the change of shape of a solid. We define the displacement vector at any point in the solid u(x, y, z) as the change in location of the material point (x, y, z) upon deformation: that is, ux(x, y, z) = x' - x, where the prime indicates the coordinates of the material that was at the unprimed position prior to the deformation. In linear elasticity, we explicitly assume that the displacement vector varies slowly from point to point within the solid where i and j denote the directions along the three axes, x, y, and z. Consider the small parallel-piped section of a solid with perpendicular edges shown in Fig. 7.1. We label the first corner as O, located at position (xO, yO, zO) and subsequent corners as A, B, . . . located at positions (xA, yA, zA), (xB, yB, zB), . . . The edge lengths are Δx, Δy, and Δz such that, for example, xA = xO + Δx. As a result of the deformation, the material originally at point O is displaced to point O' with coordinates (x'O, y'O, z'O).
