A growing tendency to study Australian popular musics as an aspect of Australian cultural studies has begun to bear fruit in the form of long overdue appraisal of the socio-cultural and musical significance of these musics. Yet in our justified enthusiasm for the exciting horizons made visible by this development, it is possible to forget that music research can serve more traditional but nevertheless important functions. In the study of popular musics, in particular, knowing how music was played can be more important than knowing what piece of repertoire was performed. For example, discovering what was ‘done’ to music in performance can tell us about the sound of unrecorded genres and can facilitate (for whatever reason) the reconstructed performance of these genres. Knowing that specific performance gestures (such as ‘flattened’ or ‘blues’ notes) were applied can of course increase our understanding of the social, cultural, and even the political nature of specific musics. In this article I am mostly concerned with applying this notion of ‘doing things’ to music to the retrieval of musical information about a specific ensemble. This more or less indigenous ensemble, which was promoted in 1918 as ‘Australia's First Jazz Band’, has captured the imagination of Australian jazz writers and is widely considered to represent the beginning of Australian jazz (Bissett 1979, p. 9).

In this study, the author investigated factors underlying vocal improvisation achievement and relationships with the singers' musical background. Participants were 102 college students in Australia and the United States who performed 3 jazz improvisations and 1 free improvisation. Jazz improvisations were rated on rhythmic, tonal, and creative thinking criteria; free improvisations were rated only on creativity criteria. The results are as follows: (a) A significant difference was found between jazz and free improvisation achievement; (b) extensive jazz experience, especially study and listening, was found to be significantly correlated with vocal improvisation achievement; (c) 3 factors were found to underlie jazz improvisation: jazz syntax, vocal creativity, and tonal musicianship; and (d) 3 factors were found to underlie free improvisation: musical syntax, vocal creativity, and scat syllable creativity.

For young Australians in the early 1960s America was the icon of pop music and fashion. This was the result of the projection of America through the mass media and the numerous American rock'n'roll acts that were brought to Australia by Lee Gordon, an American entrepreneur who lived in Sydney (Zion 1984). This overall tendency led the American, A. L. McLeod, to observe when writing about Australian culture in 1963 thatin general, Australian popular music is slavishly imitative of United States models; it follows jazz, swing, calypso or whatever the current fashion is in New York or San Francisco at a few months distance. (McLeod 1963, p. 410)Yet by late 1963 the potency of America was in decline. For while the Californian surf music craze made a somewhat delayed impact, especially in Sydney, the popularity of the Beatles was gathering momentum. This can be traced crudely through the Top Forty lists of the day: in Sydney the song ‘From Me To You’ entered the charts on 12 July 1963 and eventually reached number six (Barnes et al. 1979, p. 50).

Recent research on practice-based doctorates in Australia has revealed an institutional preference for ‘theorised’ research approaches aimed at situating studies of practice within established academic paradigms. In this article we examine how the aim of communicating with artistic peers steers the research design and the production of text-based artefacts for a group of practice-based doctoral students working on jazz topics (n = 11) away from theorised approaches and towards what is commonly referred to as a ‘commentary’ approach. This finding reveals the extent to which the values of an artistic community can influence the scope of what is discussed within practice-based doctorates and highlights the need for ongoing discussion related to how such values might best interface with what institutions view as best-practice research frameworks.

The live music pub and club scene has historically been regarded as the source of a distinctively Australian rock/jazz culture, and the basis for global recording success. This paper examines the history of live venue practices as a case study of a local cultural industry that often existed outside of traditional policy structures and meanings of the arts industries. Confronted with a loss of performance opportunities for local musicians, it is argued that traditional cultural policy mechanisms and platforms used for cultural nationalist outcomes are no longer relevant. Rather, policy intervention must engage with administrative obstacles to live creativity, specifically the series of local regulations that have diminished the viability of live venues. The decline of the rock/jazz pub continues in the face of current federal government support for touring musicians. A closer inspection of the local administration of cultural practice remains the best means of understanding the devaluation of the social and industrial value of live performance.

Over the past two decades the topic of graduate outcomes has increasingly informed the discourse on the changing nature of universities. For conservatoires and university music departments the global shift in audience demand away from western classical music and jazz styles (traditionally the cornerstone tertiary music programs) to contemporary commercial music (CCM) has added an extra dimension to the graduate outcomes discussion with respect to vocation preparation and musicians’ portfolio careers. Few studies have tracked the career paths of music graduates across time with none focused on jazz/contemporary singers. This report discusses the findings from a snapshot study of Jazz/Contemporary Voice graduates (2001 to 2012) of one Australian conservatoire. The purpose of this research was to better describe this population of graduates in terms of employment outcomes, the dynamics of their employment activities, employment-seeking strategies and the relevance of university coursework to their employability. Their responses have implications for tertiary music training programs.

The one-to-one teacher–student relationship is a common learning configuration within jazz education. However, opportunities to learn through engagement in ensemble performances and industry-level recording opportunities with esteemed jazz performers are rare classroom environments the tertiary jazz music institutions offer. This qualitative study examines ‘real-world’ jazz performance contexts within an Australian tertiary music course, exploring students’ learning experience spanning three diverse collaborative projects. Bandura’s Social Cognition Theory is utilized to elucidate an ecological system of musical development, where learning occurs in a social context within dynamic, reciprocal interactions between learners, environment and students’ adaptive behaviours that are bounded by context, culture and learner history. Findings from pre- and post-participation interviews reveal student and educator perspectives of engaging in authentic experiential learning situations. A stratum of positive influences impacting students included metacognitive, behavioural, emotional affordances, as well as the cultivation of a wider social, environmental and cultural/creative confidence and an expanding collaborative community influencing individuals’ learning decisions. Students and educator participants expressed professional-level expectations, real-world outcomes, and a deeper musical connection and understanding by students of the guest artist/composers’ intention, musical aesthetic and expert band direction. The authors maintain that inclusion of experience-based education and embedding of authentic professional industry experience and creative music-making contexts within educational settings enhance the learning of students and potentially enculturate richer musicianship in students and their developing creative communities.

This study explores performance-related medical disorders (PRMD) among a sample of tertiary-trained jazz pianists. Participants included both Australian and US pianists (n=214), including current and former tertiary students, professional pianists, and teachers. This mixed methods (quantitative and qualitative) exploratory and descriptive study used survey and case studies to provide baseline data for further research. Students reported a past and present period prevalence of 63% for pain and 41% for PRMD (injury attributed to practice or performance) with the forearm being the body part most affected, usually by fatigue. Diagnosis and treatment were reported as often unsatisfactory mainly due to lack of knowledge of PRMD by teachers and by professional health providers, and also to limited access to specialist PRMD services where these exist. Although teacher knowledge of PRMD is quite low, students still seek advice primarily from their teachers. The current study highlights a need to address the issue of PRMD among jazz pianists and their teachers more strategically, both in its prevention and in diagnosis and treatment.

AbstractThis paper argues that management education needs to consider a trend in learning design which advances creative learning through an alliance with art-based pedagogical processes. A shift is required from skills training to facilitating transformational learning through experiences that expand human potential, facilitated by artistic processes.This creative learning focus stems from a qualitative and quantitative analysis of an arts-based intervention for management development, called Management Jazz, conducted over three years at a large Australian University.The paper reviews some of the salient literature in the field, including an ‘Artful Learning Wave Trajectory’ Model. The Model considers four stages of the learning process: capacity, artful event, increased capability, and application/action to produce product. Methodology for the field-based research analysis of the intervention outcomes is presented. Three illustrative examples of arts-based learning are provided from the Management Jazz program. Finally, research findings indicate that artful learning opportunities enhance capacity for awareness of creativity in one's self and in others, leading, through a transformative process, to enhanced leaders and managers. The authors conclude that arts-based management education can enhance creative capacity and develop managers and leaders for the 21st century business environment.

The study and practice of improvisation in music departments of Australian colleges and universities tends to be dominated by jazz and other African-American styles. However, the School of Music of the Victorian College of the Arts in Melbourne has developed a course of study with a different focus. While rooted in the fundamentals of jazz performance, the philosophy of the course is that students at the end of the twentieth century should endeavour to develop their own musical ‘voice’. An important means of assisting this development is the encouragement for students to compose their own music as the basis for improvisation. In many cases personal concerns and events form the basis for these original pieces and allow performers to develop their own compositional concepts. This is also a significant means of allowing the music to reflect the era and culture of the performers. This article outlines an investigation of ten students conducted over the three years of their degree studies. The investigation aimed to ascertain the conditions under which a personal voice might be acquired and the extent to which composition was employed in the participants' major performances.

Music institutions predominantly utilize the one-to-one lesson in developing and supporting music students’ learning of skill and knowledge. This article explores the effect that interpersonal interaction plays in shaping pedagogical applications between teacher and student. Observing the learning of improvisation within this individualized social context, dynamic systems theory (DST) is used to explore how learning and development of musical improvisation skills are shaped by interpersonal behaviors and learning relationships. Through the dimensions of teacher “action” and “affiliation”, this phenomenological study investigates the interactive behaviors of five expert Australian educator/improvisers and their students. Interpretative phenomenological analysis is utilized to identify modes of behavioral interactions that evolve through dynamic and fluid interplay. Interactive behavior positions and facilitates the delivery of skill, knowledge, teacher attitudes and understandings of the improvisation process. An interpersonal model of behaviors within the instrumental music lesson is posited, and implications for ongoing critical reflection of interpersonal and pedagogical approaches in the one-to-one context are suggested.

In recent decades, the heritage sector has become increasingly precarious amid the rise of austerity neoliberalism, impacting both the efficacy of heritage institutions and the labour experiences of those who run them. While scholarly literature has regularly examined these impacts for mainstream heritage institutions, little work considers volunteer-run, do-it-yourself (DIY) community heritage organisations. This article takes a serious leisure perspective to explore what constitutes ‘good work’ for volunteers in a DIY heritage institution, the Australian Jazz Museum (AJM). Drawing on interviews with 26 AJM volunteers, we discuss some of the ‘rewards’ and ‘costs’ of career volunteering in this institution. Our research suggests that the conditions for good work are contingent on the efforts of volunteers in management roles, while the conditions for bad work are heightened by austerity policies affecting funding opportunities. The case study also highlights the need to consider the value of work beyond remuneration.

We investigate the roles of the acoustic parameters intensity and spectral flatness in the modeling of continuously measured perceptions of affect in nine diverse musical extracts. The extract sources range from Australian Aboriginal and Balinese music, to classical music from Mozart to minimalism and Xenakis; and include jazz, ambient, drum n' bass and performance text. We particularly assess whether modeling perceptions of the valence expressed by the music, generally modeled less well than the affective dimension of arousal, can be enhanced by inclusion of perceptions of change in the sound, human agency, musical segmentation, and random effects across participants, as model components. We confirm each of these expectations, and provide indications that perceived change in the music may eventually be subsumed adequately under its components such as acoustic features and agency. We find that participants vary substantially in the predictors useful for modeling their responses (judged by the random effects components of mixed effects cross-sectional time series analyses). But we also find that pieces do too, while yet sharing sufficient features that a single common model of the responses to all nine pieces has competitive precision.

The 1920s fascination with jazz music and dance provoked deep concern and anxiety about the future direction of society in different corners of the world. Jazz was readily associated with various social evils that were considered a threat to the cultural and moral wellbeing of nations as well as the mental and physical health of both individuals and society in general. Negative reception of jazz was not limited to the United States but seems to have expanded around the globe just as rapidly as the appeal of the music and movements loosely identified at the time as jazz. The article identifies defining elements of this negative discourse to explore their mechanism and bearings in particular localized settings, namely, in Australia and Iceland. The focus is on how the perceptions of jazz as a social evil contributed to conceptualizing ideas of social reform and how the demonization of jazz played a role in defining national character and culture. Australia and Iceland serve as examples for understanding the way in which the negative discourse gained a foothold in society by relating to pre-existing social anxieties, political objectives and historical context.

This paper presents new data extracted from the National Survey of Jazz Instrumentalists and Vocalists. The survey was administered to 209 professional jazz musicians who resided and performed in Australia during 2009–2010. Presented here are five statistically significant characteristics which differentiate vocalists’ experiences from other jazz musicians. These are: the singers’ preference for learning by imitation, their use of chords to find starting notes, their reliance on aural feedback, their greater sense of personal risk in improvisation, and their desire to be comfortable when performing lyrics. The results are accompanied by suggestions as to how jazz educators may respond to the findings.

The root-lesion nematode Pratylenchus thornei causes substantial loss to bread wheat production in the northern grain region of Australia and other parts of the world. West Asia and North Africa (WANA) wheat accessions with partial resistance to P. thornei were analysed for mode of inheritance in a half-diallel crossing design of F1 hybrids (10 parents) and F2 populations (7 parents). General combining ability was more important than specific combining ability as indicated by components of variance ratios of 0.93 and 0.95 in diallel ANOVA of the F1 and F2 generations, respectively. General combining ability values of the ‘resistant’ parents were predictive of the mean nematode numbers of their progeny in crosses with the susceptible Australian cv. Janz at the F1 (R2 = 0.86, P < 0.001, 8 crosses), F2 (R2 = 0.83, P < 0.001, 9 populations) and F∞ (R2 = 0.71, P < 0.05, 5 doubled-haploid populations). The F2 and F∞ populations showed relatively continuous distributions. Heritability was 0.68 for F2 populations in the half-diallel of resistant parents and 0.82–0.92 for 5 ‘resistant’ parent/Janz doubled-haploid populations (narrow-sense heritability on a line mean basis). The results indicate polygenic inheritance of P. thornei resistance with a minimum of from 2 to 6 genes involved in individual F∞ populations of 5 resistant parents crossed with Janz. Morocco 426 and Iraq 43 appear to be the best of the parents tested for breeding for resistance to P. thornei. None of the P. thornei-resistant WANA accessions was resistant to Pratylenchus neglectus.

Plant parasitic nematodes are a major biotic cause of wheat yield loss in temperate wheat-growing regions. Previous studies using Australian germplasm and/or synthetic hexaploid lines have identified quantitative trait loci (QTLs) for root lesion nematode resistance on chromosomes 2B, 6D, and 7A. This study examines Pratylenchus thornei resistance in 2 Middle-Eastern landraces (AUS13124 and AUS4926), using doubled haploid populations generated by crossing with the susceptible Australian cultivar Janz. Single marker regression and QTL analysis identified resistance loci on chromosomes 2B, 3B, 6D, and 7A, and a susceptibility locus on chromosome 1B. The 2B and 6D loci, which have been reported to explain up to 19% and 24% of variation, respectively, in previous studies, made smaller contributions in the Middle-Eastern varieties, explaining 2–13% (2B) and 1–6% (6D) of phenotypic variation in these populations. The previously reported 7A locus (P. neglectus resistance) was detected through single marker regression only (AUS13124 × Janz – LRS = 4.1, P = 0.04292; AUS4926 × Janz – LRS = 9.6, P = 0.00195), with genotype at the microsatellite marker Xgwm350.3 accounting for 3–23% of phenotypic variation. The previously unreported resistance QTL, located on chromosome 3B, explained up to 24% of phenotypic variation, and the susceptibility locus on chromosome 1B explained up to 21%. The 3B locus was detected in both the AUS13124 × Janz (max. LRS = 20.13) and AUS4926 × Janz (max. LRS = 11.19) populations, and the 1B locus was detected in the AUS4926 × Janz population (max. LRS = 18.82) only.

Background: Prognosis of Acute Myeloid Leukaemia (AML) is guided by chromosomal and molecular profiling. Standard regression approaches may not be optimal for identifying complex gene-gene interactions. Aims: We investigated use of machine-learning (ML) by recursive partitioning (RP) and random forest (RF) analysis on two large AML datasets to develop a hierarchical prognostic risk model. Methods: This study included 2074 non-APL AML cases aged ≤70 from National Taiwan University Hospital (NTUH; n=759) and the German-Austrian AML Study Group (AMLSG; n=1315)(Table 1). Statistical analysis performed in R (version 3.5.1) with RP performed using the rpart (version 4.1-13); RF analysis used randomForestSRC (version 2.7.0) with complexity parameter 0.0007 and minimum subgroup 20. Missing data imputed, branch-points without prognostic significance trimmed. Kaplan-Meier analysis utilized for survival with patients with incomplete data excluded from survival analysis. Overall survival (OS) outcomes were classified into Good (5-year OS &gt;55%), Intermediate (5-year OS &gt;40 - 55%), Poor (5-year OS &gt;20 - 40%) and Very Poor (5-year OS ≤20%). Results: Median age 48 (range 15 - 70) years. Cohorts were evenly matched except higher rate of allograft in complete remission (CR) or CR with incomplete blood count recovery (CRi) for AMLSG (31% vs 18%)(Table 1). CR or CRi achieved in 70%. Median OS for the entire cohort 2.3 years. Machine-learning to develop a prognostic decision-tree for OS based on random 2:1 selection of cases (training-set) and validated on remaining cases (validation-set). Each cohort produced similar RP-trees (Figure 1). Through ML, a decision tree was developed that hierarchically categorized cytogenetic and molecular factors into groupings that most accurately predicted survival. AML features used in the tree included complex karyotype (CK), inv(16), CEBPAdmut, inv(3)/t(3;3), FLT3-ITD, spliceosome mutations (U2AF1, SRSF2 or SF3B1), NPM1mut (in the absence of FLT3-ITD), t(8;21), and other AML identified by single genetic lesions including MLL translocations, NRASmut, isolated TP53mut or isolated ASXL1mut (Table 2). 261 patients (12.5%) were not classified by the model due to incomplete cytogenetic or mutational data and were excluded from survival analysis. In the CK group, a negative effect on prognosis (CR and OS) was conferred by inclusion of high-risk monosomies or structural chromosomal alterations (-7, del(7q), chromosome 17 abnormalities, -5 or del(5q)) or TP53mut, whereas CK cases lacking these lesions had a better prognosis (median OS 5.5 vs 25.1 months, p&lt;0.001 and CR 36 vs 70%, p&lt;0.001). In CEBPAdmut AML, ML highlighted the favorable impact of NRASmut (present in 27%) on survival. In patients with FLT3-ITD, very poor prognosis seen with co-occurrence of spliceosome mutations. High FLT3-ITD allelic ratio (AR &gt;0.5) also associated with very poor prognosis in the absence of both spliceosome lesions and NPM1mut (median OS 11.6 months). A poor prognostic category comprising triple mutant FLT3-ITD, NPM1mut and DNMT3Amut was identified (median OS 13.2 vs 41.7 months, p=0.002). Cases with spliceosome mutation (SRSF2, U2AF1, SF3B1) had a very poor prognosis if accompanied by either ASXL1mut or potential heterozygous loss of ASXL1 on 20q (either by del(20q) or -20)(median OS 11.8 vs 21.6 months, p=0.003). In NPM1mut disease, ML identified good prognosis for patients who with NRASmut, whereas co-mutation with IDH1mut conferred an intermediate prognosis. In patients with t(8;21) AML, KIT mutations identified patients with intermediate prognosis (median OS 36.6 months vs NR, p=0.02). There were 49 patients (2.4%) with RUNX1mut not categorized to other groups and RUNX1mutper se did not emerge as a prognostic discriminator in this analysis. In comparison to ELN2017 classification, ML reclassified 45% of cases - 30% good, 26% intermediate, 26% poor and 18% very poor (compared to 39% good, 31% intermediate and 30% poor). In establishing the accuracy of predicting death at any time for individual cases the error rate was 35% for our ML model compared to 45% for the ELN2017 classification and 51% for ELN based cytogenetic analysis alone. Conclusion: By combining large datasets from two AML groups using machine-learning, an AML decision-tree was able to refine the accuracy of assessing survival prognosis compared to ELN 2017 classification by providing further prognostic granularity. The effect of frost damage on wheat grain quality was examined in samples of an Australian Prime Hard wheat cultivar Janz. Wheat grain samples that were lightly frosted, severely frosted and unfrosted were obtained from the Riverina district of New South Wales. Each frosted sample was separated by sieving into 2 fractions by size using a commercial grader that was equipped with an aspirator. As the degree of frost damage in the samples increased, grain size decreased, flour extraction decreased, flour ash increased, flour colour deteriorated, dough strength decreased, baking quality decreased, α-amylase activity increased and falling number decreased. Quality assessment of the separated grain fractions showed that the large grains (&gt;2 mm) in both lightly frosted and severely frosted crops were equal or better than the unfrosted sample in all tested quality parameters. The large grain fraction fully met the Australian Prime Hard receival standards and was of quality commensurate with the grade. Separation of grain by size was calculated to be commercially viable for up to 50% frost damage. A commercial flourmill purchased frosted feed grain, followed the sieving procedure, and produced large grain consistent with unfrosted wheat that was subsequently used for normal processing.

At the present stage of development of world music, the accordion and button accordion occupy one of the leading positions. Formation of instruments on the professional concert stage is an integral part of both world and Ukrainian musical culture, as evidenced by the composition, a large number of performing competitions and festivals that take place not only in Ukraine but also in Europe, Asia and America, and Australia. A significant phenomenon of the latest wave of development of accordion and button accordion art is the crystallization of pop and jazz in the works of composers and performers. From the second half of the twentieth century, the popularity of pop and jazz music among performers and listeners (due to its brightness and accessibility) opened new horizons, genre and stylistic searches which have largely affected the trends in the development of world accordion and button accordion art, including pop and jazz. The composer’s activity of leading performers is becoming a widespread phenomenon of modern pop-jazz accordion movement. Among them we note V. Podgorny, V. Zubytsky, V. Vlasov, A. Haidenko, O. Nazarenko, B. Myronchuk, A. Stashevsky and others. Well aware of the specifics of technical-expressive, acoustic and textural capabilities of modern accordion and button accordion, domestic composers-accordionists have created a large number of bright works of pop and jazz direction. Thus, the intensification of composer’s work and the emergence of new works of pop and jazz in the accordion and button accordion art of modern times have determined the relevance of the topic of this article. The purpose of the article is to identify the main stylistic and genre features of the French musette in the works of A. Haidenko. One of the most significant examples of this genre is the series «Paris Secrets» of five waltzes in the style of French musettes for accordion by A. Haidenko. Such a work as a «musette» has not yet been mastered by any of the domestic composers, especially in the form of a cycle. The composer rethought the basics of the French folk instrumental genre in terms of professional accordion performance. This synthesis transforms the genre of the musette, embodies the pop genre in terms of academic art. The use of professional performance capabilities distinguishes the artist’s works from other compositions of this style, with the general availability of musical material to a wide range of listeners. Typical melismatics of French musettes is organically and professionally implemented by A. Haidenko in the whole cycle. The melismatics is based on beamed ascending and descending grace notes on chord tones, “singing” grace notes on separate notes, chromatic grace notes (imitations of “transitions” to the sound), mordents. Sudden dynamic contrasts, shift of strong bars, chord introduction, virtuosity, brightness of phrases and sentences are typical for A. Haidenko’s musettes. The series of five waltzes for accordion “Paris Secrets” in the style of French musettes by A. Haidenko is a unique heritage not only of the domestic, but also of the world original repertoire.

Introduction Molecular MRD assays targeting NPM1 mutant (mut) transcripts have an established role for monitoring treatment efficacy in patients with NPM1mut AML. Approximately 25% of NPM1mut patients show persistent MRD level in the bone marrow (BM) at the end of treatment (EOT), which is associated with a higher risk of relapse (Ivey, NEJM 2016; Kronke, JCO 2011). Molecular persistence at low copy number (MP-LCN) is defined by the European LeukemiaNet (ELN) as MRD positivity in patients in morphological complete remission (CR) with &lt;1000-2000 transcripts per 105ABL and a relative increase of &lt;1 log between any two positive samples collected after EOT (Schuurhuis, Blood 2018). The UK NCRI working group recently reported the impact of NPM1mut MRD burden and FLT3-ITD status on risk of relapse after allogeneic stem cell transplantation (Dillon, Blood 2020), however the clinical relevance of NPM1mut MP-LCN in patients who are not transplanted is unknown. We aimed to characterize the natural history of persistent NPM1mut MRD after chemotherapy and to identify factors associated with subsequent disease progression. Methods Consecutive patients with newly diagnosed NPM1mut AML receiving at least 2 cycles of intensive chemotherapy were included if MRD remained detectable at EOT. UK patients were all enrolled in the NCRI AML17 and AML19 studies. Australian patients were retrospectively identified. BM samples were analyzed by either RT-qPCR (n=75) (Ivey, NEJM 2016) or ultra-deep next-generation sequencing (UD-NGS, n=5, excluded from Cox model). We applied the ELN definitions for molecular progression or relapse (considered together as molecular failure [MF]), MP-LCN and complete molecular remission (CRMRD-). Kaplan Meier survival estimates were calculated from EOT to death (OS), and/or morphologic relapse (RFS), and/or MF (EFS). The optimal NPM1mut expression threshold was pre-determined by recursive partitioning. Variables with p&lt;0.1 from univariate analysis were included in the final multivariate Cox model and confirmed by backward selection method. Copy numbers are expressed per 105ABL copies (for RT-qPCR) or as percentage of total reads (for UD-NGS). Results A total of 80 patients had NPM1mut detectable at a median of 37 days (range 19-90; 12 [15%] were &gt;60 days) from last course of chemotherapy; characteristics summarized in Table 1. The majority of patients (89%) had received 3-4 cycles of chemotherapy. At EOT, the median NPM1mut transcript level by RT-qPCR was 11 copies (range 0.3-8699; 3 were &gt;2000) and median reduction from baseline was 4.7 log (range 2.1-6.0). A total of 34 patients (45%; 4 not monitored) fulfilled the criteria for MP-LCN. With a median follow-up time of 24 mo (range 8-77), 27 (34%) spontaneously achieved a sustained CRMRD- for &gt;6 months, 31 (39%) had MF and 13 (16%) had morphologic relapse without preceding MF (due to rapid relapse [n=8], lack of monitoring [n=3], extramedullary or NPM1 wild-type relapse [n=1 each]) (Figure 1). Nine patients (11%) had persisting MP-LCN (included transient MRD negativity): median interval between EOT to the last known positive sample was 12 mo (range 3-32), and all patients currently remain alive and without morphologic relapse by the data cut-off date July 13, 2020. Next, we examined the variables associated with MF and/or morphologic relapse. In univariate analysis, factors associated (p&lt;0.1) with disease progression were white cell count, FLT3-ITD, &lt;4 cycles of chemotherapy received, EOT NPM1mut ≥15 copies, and &lt;4 log reduction from baseline (Table 2). In multivariate analysis (and confirmed by backward selection), only baseline FLT3-ITD (HR 2.5, 95% CI 1.3-4.6) and EOT NPM1mut MRD &lt;4 log reduction from baseline (HR 3.2, 95% CI 1.7-6.2) retained significance. Patients with none, either, or both of these features had a median EFS of not reached, 5.8 and 1.4 mo, respectively (Figure 2). The corresponding 2-year RFS was 73%, 54% and 0% (median 3.5 mo). Conclusions We describe the natural history of patients with NPM1 mutated AML who remain MRD positive after completing intensive chemotherapy. Almost half either spontaneously achieve MRD-negativity or have stable low-level expression without relapse with minimum follow up of 8 mo. Patients with both FLT3-ITD and &lt;4 log reduction in transcript levels at the end of therapy are at very high risk of disease progression and warrant consideration of pre-emptive treatment. Introduction The goal of betibeglogene autotemcel (beti-cel; LentiGlobin for β-thalassemia) gene therapy in patients with transfusion-dependent β-thalassemia (TDT) is lifelong, stable production of functional adult hemoglobin (Hb) sufficient for transfusion independence (TI) and reduction in ineffective erythropoiesis. 60 patients with TDT have been treated with beti-cel across 2 completed phase 1/2 studies (HGB-204, HGB-205) and in 2 ongoing phase 3 studies (HGB-207, HGB-212). After 2-yrs of follow-up in these 4 parent studies, patients were invited to enroll in a 13-yr long-term follow-up study, LTF-303 (NCT02633943). Interim results of patients enrolled in LTF-303 with follow-up as long as 6 years are reported. Methods Autologous CD34+ cells were transduced with BB305 lentiviral vector and infused into patients following single-agent, pharmacokinetic-adjusted busulfan myeloablation. Transduction in the phase 3 studies used a refined manufacturing process compared to the phase 1/2 studies. LTF-303 assessments include Hb, peripheral blood vector copy number (PB VCN), assessment of erythropoiesis and iron overload, quality of life, adverse events (AEs), replication-competent lentivirus (RCL), and insertion site analysis. Data are analyzed as median (min - max). Results As of 3 March 2020, all 32 patients who completed the parent studies (age at enrollment in parent study: 20 [12 - 35] yrs) enrolled in LTF-303 (22 treated in phase 1/2 studies, 10 treated in phase 3 studies). Follow-up post-infusion was 49.1 (23.3 - 71.8) months. PB VCN was detected in all patients at last follow-up (Phase 1/2: 0.4 [0.07 - 4.0] c/dg; Phase 3: 2.0 [0.13 - 3.0] c/dg). Gene therapy-derived Hb, HbAT87Q,in patients treated in the phase 1/2 studies was stable over time: 6.4 (0.5 - 10.1), 6.7 (0.4 - 10.1), 6.6 (0.5 - 10.7), and 7.1 (2.8 - 11.2) g/dL at months 24 (n=22), 36 (n=22), 48 (n=22), and 60 (n=10). Median HbAT87Q at month 24 in patients treated in the phase 3 studies was 9.5 (0.9 - 12.4) g/dL (n=10). Of the 32 patients enrolled in LTF-303, TI (defined as a weighted average Hb ≥9 g/dL without packed red blood cell transfusions for ≥12 months) was achieved in 14/22 (64%) patients treated in phase 1/2 (12 achieved TI during parent study, 2 during LTF-303) and in 9/10 (90%) patients treated in phase 3 (all achieved TI in parent study). All patients remain TI at last follow-up for 39.4 (19.4 - 69.4) months. Weighted average Hb during TI was 10.4 (9.4 - 13.3) and 12.5 (11.9 - 13.5) g/dL in patients treated in the phase 1/2 and phase 3 studies, respectively. In patients who achieved TI in the phase 3 studies, soluble transferrin receptor decreased from 144.1 (65.9 - 235.3) nmol/L at baseline to 54.1 (24.7 - 67.1) nmol/L at Month 24. Patients who achieved TI in HGB-207 had an improved health state today score from 65 - 96 at baseline to 90 - 100 at month 24 (n=8) on the EQ-5D-3L or EQ-5D-Y instrument. All patients were on iron chelation before beti-cel infusion, but post-infusion, only 26/32 (81%) patients restarted iron chelation; of these, 11 have since discontinued chelation. Phlebotomy was used for iron removal in 7/32 patients (22%; 3 patients treated in phase 1/2, 4 patients treated in phase 3) including 3 patients who also used iron chelation. Following an initial increase in liver iron concentration (LIC) after infusion, LIC in patients who achieved TI decreased, particularly in patients with an elevated baseline LIC (Figure). The median decrease in LIC from baseline to month 48 in patients who achieved TI was a 38% reduction (85% reduction to 269% increase; n=13). No drug-product-related AEs were reported &gt;2 years post-infusion. Serious AEs during LTF-303 included gonadotropic insufficiency, ectopic pregnancy, gall bladder wall thickening/polyp, bacteremia with neutropenia. and major depression (all n=1). No deaths, RCL, or insertional oncogenesis were reported. Insertion site analysis as assessed every 6 months until month 60 showed unique insertions accounted for &lt;30% of all insertions indicating polyclonal hematopoiesis. Summary These results demonstrate the durability and stability of response after beti-cel gene therapy in patients with TDT. Sustained levels of HbAT87Q and effective iron reduction with phlebotomy and/or iron chelation have resulted in improved hematologic parameters and iron burden. The paucity of gene therapy-related AEs observed beyond 2 years post-infusion study suggests a favorable long-term safety profile. Background Patients with transfusion-dependent β-thalassemia (TDT) may experience transfusional iron overload and end-organ damage. While potentially curative, allogeneic hematopoietic stem cell (HSC) transplantation is limited by transplant-related risks and donor availability. Transplantation of autologous CD34+ cells encoding a βA-T87Q-globin gene (LentiGlobin gene therapy for β-thalassemia) may overcome some of these limitations. βA-T87Q-globin is incorporated into adult hemoglobin (Hb), forming gene therapy-derived HbAT87Q, which can be distinguished from other Hb species. The phase 1/2 Northstar study (HGB-204; NCT01745120) using the original manufacturing process evaluated the safety and efficacy of LentiGlobin in adolescents and adults with TDT (≥100 mL/kg/yr of red blood cells [RBCs] or ≥8 RBC transfusions/yr) and non-β0/β0 or β0/β0 genotypes. Methods HSCs were mobilized with G-CSF and plerixafor and collected via apheresis. CD34+ cells were transduced with BB305 lentiviral vector. After busulfan myeloablation, patients were infused with transduced cells. Primary efficacy endpoints were sustained production of ≥2 g/dL HbAT87Q between months 18 and 24 and transfusion independence (TI; weighted average Hb ≥9 g/dL without RBC transfusions for ≥12 months). Patients were monitored for 2 years and subsequently enrolled in the 13-year long-term follow-up study, LTF-303 (NCT02633943). Results are shown as median (min ‒ max) unless otherwise indicated. Results Eighteen patients were treated (age: 20 [12 - 35] yrs) and followed for 40.7 (29.3 - 53.8) months as of 13 December 2018. In the 2 years prior to enrollment, patients had an annualized transfusion volume of 169.0 (124.0 - 273.0) mL/kg/yr and pre-transfusion weighted mean nadir Hb of 9.3 (7.0 - 10.1) g/dL. Neutrophil and platelet engraftment occurred at 18.5 (14 - 30) and 39.5 (19 - 191) days, respectively. No patient had graft failure. Grade ≥3 non-hematologic adverse events (AEs) reported in ≥25% of patients after infusion were stomatitis, febrile neutropenia, and pharyngeal inflammation. No replication-competent lentivirus or death has been reported. The vector integration site profile in all 18 patients has remained polyclonal. The number of unique integration sites (UIS) identified was 1646 (190 - 2888), 1677 (151 - 6935), 2484 (984 - 5511), 1773 (1260 - 2693) at Months 12 (n=18), 24 (n=18), 36 (n=11), 48 (n=4), respectively. The highest mean (SD) frequency of any UIS in patients across all visits was 11.5% (5.8%). No oncogenesis has been reported. In Northstar, 16/18 (89%) patients achieved the primary endpoint of ≥2 g/dL HbAT87Q between months 18 and 24. Eight of 10 (80%) patients with non-β0/β0 genotypes achieved and maintained TI; current duration of TI was 38 (21.2 - 45.3) months (Figure 1). The weighted average total Hb during TI was 10.3 (9.1 - 13.2) g/dL. Total Hb and HbAT87Q remained stable over time. Total Hb in patients with non-β0/β0 genotypes who achieved TI was 10.3, 10.4, 10.6, and 11.1 g/dL at Months 12 (n=8), 24 (n=8), 36 (n=7), 48 (n=3), respectively. Transfusion volumes were reduced by 73% and 43% in the 2 patients still receiving transfusions. Three of 8 (38%) patients with β0/β0 genotypes achieved TI with a current duration of 16.4 (16.1 - 20.8) months. Weighted average total Hb during TI was 9.9 (9.5 - 10.1) g/dL and HbAT87Q was 8.0 - 8.9 g/dL at last visit. One additional patient was transfusion-free for 13.7 months; however, total Hb was <9 g/dL. The 4 other patients had a transfusion volume reduction of 53% (10% - 72%). Patients who achieved TI resumed iron chelation 13 (2 - 15) months after infusion and all remain on iron chelation as of last follow-up. Serum ferritin and liver iron content (LIC) (Figure 2A, 2B) were reduced in patients who achieved TI by 55% (16 - 78%) and 56% (38 - 83%) from screening to Month 48 (n=4), respectively. Of these 4 patients who had a Month 48 visit, LIC values were 0.8 - 7.1 mg/g at Month 48 compared to 4.8 - 11.5 mg/g at screening. In patients who achieved TI, cardiac T2* ranged from 27.0 - 39.0 msec at screening and 31.4 - 57.6 msec at last visit. Summary With up to 4.5 years of follow-up after LentiGlobin gene therapy, generally stable HbAT87Q levels and durable TI were observed in 8/10 and 3/8 patients with TDT and non-β0/β0 and β0/β0 genotypes, respectively. Iron burden has improved over time in patients who achieved TI. The safety profile of LentiGlobin remains consistent with myeloablative conditioning. Root-lesion nematodes (Pratylenchus thornei Sher and Allen and P. neglectus (Rensch) Filipijev and Schuurmans Stekhoven) cause substantial yield loss to wheat crops in the northern grain region of Australia. Resistance to P. thornei for use in wheat breeding programs was sought among synthetic hexaploid wheats (2n = 6x = 42, AABBDD) produced through hybridisations of Triticum turgidum L. subsp. durum (Desf.) Husn (2n = 4x = 28, AABB) with Aegilops tauschii Coss. (2n = 2x = 14, DD). Resistance was determined for the synthetic hexaploid wheats and their durum and Ae. tauschii parents from the numbers of nematodes in the roots of plants grown for 16 weeks in pots of pasteurised soil inoculated with P. thornei. Fifty-nine (32%) of 186 accessions of synthetic hexaploid wheats had lower numbers of nematodes than Gatcher Selection 50a (GS50a), a partially resistant bread wheat. Greater frequencies of partial resistance were present in the durum parents (72% of 39 lines having lower nematode numbers than GS50a) and in the Ae. tauschii parents (55% of 53 lines). The 59 synthetic hexaploids were re-tested in a second experiment along with their parents. In a third experiment, 11 resistant synthetic hexaploid wheats and their F1 hybrids with Janz, a susceptible bread wheat, were tested and the F1s were found to give nematode counts intermediate between the respective two parents. Synthetic hexaploid wheats with higher levels of resistance resulted from hybridisations where both the durum and Ae. tauschii parents were partially resistant, rather than where only one parent was partially resistant. These results suggest that resistance to P. thornei in synthetic hexaploid wheats is polygenic, with resistances located both in the D genome from Ae. tauschii and in the A and/or B genomes from durum. Five synthetic hexaploid wheats were selected for further study on the basis of (1) a high level of resistance to P. thornei of the synthetic hexaploid wheats and of both their durum and Ae. tauschii parents, (2) being representative of both Australian and CIMMYT (International Maize and Wheat Improvement Centre) durums, and (3) being representative of the morphological subspecies and varieties of Ae. tauschii. These 5 synthetic hexaploid wheats were also shown to be resistant to P. neglectus, whereas GS50a and 2 P. thornei-resistant derivatives were quite susceptible. Results of P. thornei resistance of F1s and F2s from a half diallel of these 5 synthetic hexaploid wheats, GS50a, and Janz from another study indicate polygenic additive resistance and better general combining ability for the synthetic hexaploid wheats than for GS50a. Published molecular marker studies on a doubled haploid population between the synthetic hexaploid wheat with best general combining ability (CPI133872) and Janz have shown quantitative trait loci for resistance located in all 3 genomes. Synthetic hexaploid wheats offer a convenient way of introgressing new resistances to P. thornei and P. neglectus from both durum and Ae. tauschii into commercial bread wheats.
