Повний текст джерелаAnti-cancer therapies, particularly immunotherapy, have made considerable progress in recent years; however, only a small number of patients derive significant and lasting clinical benefit. The incapacity of cytotoxic immune cells to infiltrate the tumor microenvironment explains in part this phenomenon. Immune infiltration depends in particular on the chemokine network, regulated in part by ACKRs. The aim of this thesis was to study the mechanisms involved in the regulation of ACKR2, which regulates the pro inflammatory chemokine network. In silico data showed that both murine and human ACKR2 promoters contain hypoxia response elements. In vitro, hypoxic colorectal, melanoma and breast cancer cells overexpressed ACKR2, which was no longer the case for cells with a deletion of HIF-1α. In vivo, ACKR2 expression was also decreased in HIF-1α-deleted tumors and associated with increased CCL5 and immune infiltration. Chromatin immunoprecipitation showed HIF-1α directly binds onto the HRE motifs of ACKR2 promoter in melanoma cells
