AbstractThrough comparison of two families, Filipino and American, this essay finds that the axiomatic three-generational cycle of rise and decline, articulated famously by Andrew Carnegie, proved predictive for an American family firm but not for its Filipino counterpart. Over the span of a century, both families followed a surprisingly similar move from agriculture to food processing and then publishing. Thereafter, however, divergent state policies shaped different destinies for these two families. In the United States, impersonal enforcement of state security and economic regulation allowed the unchecked rise of finance capital that consolidated some 2000 US breweries, most of them family owned, into two transnational corporate conglomerates. In the Philippines, by contrast, persistent rent seeking by elite families, combined with personalised, partisan state economic enforcement, has allowed the continuing dominance of family-controlled corporations. Through comparison of two societies with close relations for over a century, we can see how state economic regulation can encourage the eclipse of major family firms in one society and the perpetuation of a political-economic oligarchy in another.

AbstractThe article explores the process behind the construction of the Chao Phraya Dam, the first World Bank-funded water infrastructure project in Thailand, developed during the 1950s. Employing Andrew Pickering's ‘dance of agency’ concept in examining the process of turning financial and technical assistance into a workable project, I argue that development infrastructure, like the Chao Phraya Dam, provides a space to explore the dialectic operations – accommodation and resistance – of agency and the unstable associations among diverse actors, expertise, institutions, and materials, as well as practices. Recounting the history of the dam in the making, I explore a series of entanglements through different dances of agency, namely initiation, assessment, mobilisation, negotiation, adjustment, confrontation, and settlement. Such a multiplicity of dancesinsideandin the makingof infrastructure reflects the techno-political entanglement encompassing the manifold negotiation and adjustment of conflicting goals, interests, recognition, and cooperation among different agencies. The dam, often portrayed as an engineering achievement of the state, is in fact the result of unanticipated relations and the responses to the temporal emerging forms of practices.

BackgroundTranscription factor A, mitochondrial (TFAM) is a transcription factor with essential function in the mitochondrial homeostasis, such as mitochondria biogenesis and mtDNA replication. Deregulation of TFAM expression has been linked to mitochondrial dysfunction. However, its role in the pathogenesis of rheumatic diseases has not been studied so far.ObjectivesWe aimed to study the role of TFAM in the pathological fibroblast activation in SSc.MethodsThe expression of TFAM in SSc skin fibroblast and skin biopsies was analyzed by immunofluorescence and Western blot. The role of TFAM in fibroblast activation was investigated by TFAM knockdown in cultured fibroblasts. The role of TFAM in skin and lung fibrosis was further studied in mice with fibroblast specific knockout of TFAM in three independent mouse models: Bleomycin-induced skin and lung fibrosis as well as TβRIact-induced skin fibrosis.ResultsDermal fibroblasts from SSc patients express lower level of TFAM in the skin and also after prolonged culture in vitro. The downregulation of TFAM impairs mitochondria homeostasis with decreased mitochondrial number, accumulation of damaged mitochondria with release of mtDNA, accumulation of deletions in mtDNA, metabolic reshaping with impaired OXPHOS and release of the mitokine GDF15. Long time, but not acute exposure of normal fibroblasts to TGFβ mimicked the finding in SSc fibroblasts with downregulation of TFAM and mitochondrial homeostasis disruption and damage. Knockdown TFAM of normal fibroblasts promotes fibroblast activation with increased myofibroblast differentiation and collagen release in a SMAD3 dependent manner. RNA sequencing demonstrated upregulation of pro-fibrotic genes and pathways. Mice with fibroblast-specific knockout of TFAM demonstrate are more sensitive to fibrotic stimuli such as bleomycin injection and TβRIact-overexpression and even demonstrate responses to NaCl instillation.ConclusionAlterations in the key mitochondrial transcription factor TFAM in response to prolonged activation of TGFβ and associated mitochondrial damage induce transcriptional programs that promote fibroblast activation and tissue fibrosis.Disclosure of InterestsXiang Zhou: None declared, Thuong Trinh-Minh: None declared, Cuong Tran Manh: None declared, Andreas Giessl: None declared, Christina Bergmann: None declared, Andrea-Hermina Györfi: None declared, Georg Schett: None declared, Jörg H.W. "The church has had to accept the national division of Europe since the Middle Ages and adapt to this situation. This issue is relatively unclear in the case of Tran-sylvania. N. Iorga stated about the Orthodox Christian consciousness that “it was so strong that it hindered the creation of a strong national consciousness”, and this would allow us to see in the ecclesiastical organization a form of expression of uni-tary organization of Romanian ethnicity in Transylvania. The time of Transylvani-an principalities and voivodeships shows us that most often the ecclesiastical leaders were also the political leaders (see the case of Prince Andrew Báthory who was Archbishop of Warmia – Poland); so, the two concepts of ethnicity and confession reflected the same historical reality during those times. The two concepts will be-come separated only later, after the emergence of confessions other than the Eastern rite. In support of our statement, we have the correspondence between the Hungar-ian kings and officials and the papacy. Before dealing with these perspectives, we shall pin down the terminology to grant the reader the possibility to understand the historical situations through a kind of thinking marked by the imprint of the Holy Scripture. Keywords: ethnicity, people, confession, dynamic status, national consciousness, Transyl-vania, the church. "

Traditional use of plants constitutes an important activity in Malagasy life, especially in the countryside. The Sihanaka group is found in the east of Lake Alaotra and is the dominant cultural group in the lake region, including in the Fokontany of Andreba Gara, near Park Bandro, in the east of Lake Alaotra, in Ambatondrazaka district where the study was conducted. The Sihanaka holds traditional knowledge on plant uses and makes use of this knowledge in their daily lives. The objective of this study was to describe the use of aquatic plants in the region to better understand the importance of the Alaotra wetlands in the lives of local people. Surveys conducted among the people of Andreba, including one traditional chief (the Tangalamena) and one healer, revealed 55 useful species of aquatic plants, grouped into 41 genera and 23 families. Most species were used for their medicinal and veterinary virtues (32 species), for animal food (17 species), as utensils and tools (16 species), for cultural uses (10 species), and for human food (9 species). Leaves (76%) are the most exploited plant parts. The species with the most uses were Cyperus papyrus subsp. madagascariensis (Willd.) Kük. with 26 types of use recorded, then Phragmites australis (Cav.) Trin. ex Steud. (19 types of use), Aeschynomene elaphroxylon (Guill. & Perr.) Taub. and Eichhornia crassipes (Mart.) Solms (12 types of use each). Nymphaea nouchali, Cyperus papyrus subsp. madagascariensis and Phragmites australis were the species most frequently cited by informants, indicating that they are important for the local population. The most frequently cited types of use included joro, namely ancestor’s invocation, food, materials for house walls construction, herbal teas to treat ailments, and fencing or demarcation of land boundaries. This study highlighted the importance of the Alaotra wetlands as a source of useful plants for the local population. Sustainable management of Lake Alaotra is therefore not just necessary for biodiversity conservation, but also for maintaining local livelihoods.

Abstract Homeostasis in the gut is achieved by impeccable functioning of tissue resident immune cells that reside and closely interact with the epithelium. The tissue resident immune cells of the intestinal epithelium are predominantly lymphocytes, known as intraepithelial lymphocytes (IELs) and mainly consist of TCRαβ T cells and TCRgd T cells. These IELs are well known to sense microbes, nutrients, and cellular stress but their role in intestinal cancer progression has been controversial. On one hand, gd Τ cells are the primary source of IL-17, an important proinflammatory cytokine that aid in colorectal cancer (CRC) progression. On the other hand, mice lacking gd Τ cells have been showed to have an increased risk of developing carcinogen-induced CRC. Here we used two different mouse model of colon cancer to investigate the role of gd T IELs in colon tumor growth. We showed that in both orthotopic implantation of cancer cells in the colon, and inflammation-driven colon cancer models, gd T cells are essentials to limit tumor growth. Indeed, mice lacking gd T cells had increased tumor incidence with high-grade adenomas compared to control mice. We also performed gene expression analysis of the adenomas in control mice and in mice lacking gd T cells. They revealed distinct transcription profiles with CRC-associated gene expression profile, including inflammatory genes and epithelial to mesenchymal transition genes, particularly enriched in adenomas from gd T cell deficient mice. Moreover, using human tissue microarrays, we investigated whether the presence of gd T cells in tumors of patients with advanced CRC could be used to predict disease outcomes. We revealed that patient disease-free survival was improved if gd T cells could be detected within tumors. Therefore, we showed that gd T cells are primordial in limiting colorectal cancer progression in both mice and humans. Abstract Introduction: Pediatric patients diagnosed with cancer report lower quality of life and increased psychological distress. Furthermore, African American pediatric oncology patients face unique and extensive obstacles upon diagnosis. The present study sought to identify patient-centered experiences following a cancer diagnosis in a sample of African American pediatric oncology patients. Methods: This study examined semi-structured interviews of pediatric African American oncology patients (N = 13; Mage = 13.54 years; 7-17 years). Interviews were audio recorded and transcribed. Response patterns examining patient perceptions of being diagnosed with cancer were derived through thematic analysis. Results: Four themes related to initial diagnosis emerged: negative emotions, quality of life, cognitions, and physical appearance. The majority of patients disclosed feeling fear, hurt, and sadness in relation to their cancer diagnosis. A myriad of cognitions surrounding initial diagnosis also emerged. Many patients reported being unaware of their diagnosis and reported a sense of confusion related to extended hospitalizations and taking medication. Patients disclosed worry and embarrassment regarding losing their hair (e.g., “my hair is my pride and joy,” and, “the first thing that did come to my mind was my hair. As a teenage girl, I was just like “my hair.”). Patients also expressed worries about quality of life and prognosis. Finally, a fear of the unknown, ability to attend school and activities, and possible death emerged as common themes. Conclusion: This study described perceptions of African American pediatric oncology patients during the initial diagnosis of cancer. Key themes included negative emotions, quality of life, cognitions, and physical appearance. A pattern between cognitions related to life changes due to diagnosis and resulting emotions such as fear emerged. This finding suggested a strong connection between the perceptual aspects of initial diagnosis and patient experiences through treatment, indicating that psychological interventions could serve as a tool to decrease negative emotions. The significant importance of patient-centered education delivered immediately at diagnosis emerged as a significant factor as many patients reported unawareness of their diagnosis (e.g., “I just didn’t know what was going to happen next, like all of the medicine, how they were going to treat it and all that”). Although level of patient education may differ based on family preferences, developmentally appropriate communication may enhance the treatment process for patients, and ultimately decrease psychological distress. These findings also indicate the cultural and historical importance of physical appearance with hair being an important piece of identity for African American patients. These findings highlight the relative importance of investigating patient-centered experiences and culturally appropriate interventions across the illness trajectory from new diagnosis to survivorship, especially among African American pediatric oncology patients. Abstract STRO-002 is an antibody-drug conjugate (ADC) composed of an anti-folate receptor alpha (FolRα) antibody conjugated to a tubulin-targeting hemiasterlin (SC209) warhead via a cleavable linker currently in clinical studies for ovarian and endometrial cancer. FolRα exhibits minimal expression in normal tissues but is overexpressed in several cancers such as ovarian, endometrial, and NSCLC. We previously showed that STRO-002 induces in vitro hallmarks of immunogenic cell death (ICD), such as increased calreticulin exposure and HMGB1 and ATP release in a FolRα dependent manner. Follow-up vaccination studies with murine MC38 tumor cells engineered to express human FolRα (MC38-hFolRa) were performed to further explore the significance of STRO-002-induced ICD and protective immunity in vivo. A single vaccination with STRO-002 or SC209 in vitro-treated MC38-hFolRa cells in immunocompetent mice resulted in 60% of animals rejecting a subsequent rechallenge with living MC38-hFolRa cells, while relapse was observed in most control mice. Including an additional booster vaccination further increased the proportion ( 90%) of animals remaining tumor free after rechallenge. These results demonstrate that tumor cells pre-treated with STRO-002 or SC209 undergo potent immunogenic cell damage which can, in turn, mount protective immunity in vivo. The ability of STRO-002 to induce ICD that elicits an effective and robust anti-tumor immune response may translate into several advantages in the clinic. Induction of ICD is likely to offer a complementary anti-tumor mechanism in combination therapy regimens such as with check point inhibitors or VEGF blockade. We have demonstrated that STRO-002 in combination with avelumab in MC38-hFolRα-bearing mice significantly enhanced efficacy and durable anti-tumor immunity. More recent rechallenge studies reveal that this combination treatment suppresses growth of not only MC38-hFolRα but also MC38 wild-type tumor cells, indicating formation of immunological memory with signs of epitope spreading. Additionally, co-administration of STRO-002 with anti-VEGF therapy demonstrates improved TGI compared to monotherapy (p < 0.0001) in human ovarian OV-90 tumors. The potent cytotoxic and immunostimulatory properties of STRO-002 may also contribute to enhancing efficacy in indications with low FolRα expression. Such tumor types include endometrial and NSCLC, in which STRO-002 has displayed potent monotherapy activity in preclinical studies. In a panel of endometrial PDX models, STRO-002 was significantly efficacious in 9/17 (53%) FolRα-positive tumors, including models with moderate and weak FolRα levels. In a NSCLC PDX model, a single dose of STRO-002 induced tumor regression and suppressed tumor growth for up to three months post-dose. In conclusion, the induction of ICD by STRO-002 has potential to increase anti-tumor activity in cancers with low FolRα expression. Citation Format: Cristina L. Background:Bone remodeling is a constant process maintained by the balance between osteoclast-triggered bone resorption and osteoblast-mediated bone formation. In inflammatory arthritis, such as rheumatoid arthritis (RA), the pro-inflammatory environment favors osteoclast differentiation and skews the balance towards resorption, leading to progressive bone erosion and bone loss. O-GlcNAcylation is a post-translational modification, which transfers a single N-acetylglucosamine molecule to the serine or threonine of the target protein. The modification is accomplished by a single pair of enzymes, O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA). Unlike other glycosylation, O-GlcNAcylation occurs in multiple cellular compartments, including the nucleus. Although O-GlcNAcylation is one of the most common modifications, its role in bone homeostasis is still poorly understood. Objectives:We aimed to investigate the role of O-GlcNAcylation in osteoclastogenesis under pro-inflammatory milieus. We also focused on dissecting the signaling pathways affected by O-GlcNAcylation during osteoclast differentiation. Methods:We examined the levels of O-GlcNAc during in vitro osteoclastogenesis by western blotting. The levels of O-GlcNAc in tissue from RA patients and experimental arthritis were detected by immunofluorescence. Pharmacological inhibition and genetic knockout were used to manipulate O-GlcNAcylaiton during osteoclastogenesis. RNA sequencing was performed to study O-GlcNAc-mediated pathways. Results:We demonstrate the dynamic changes in O-GlcNAcylation during osteoclastogenesis. The elevated O-GlcNAcylation was found in the early differentiation stages, whereas its downregulation was detected in the maturation process. TNFα elaborates the dynamic changes in O-GlcNAcylation, which further intensifies osteoclast differentiation.Targeting OGT by selective inhibitor and genetic knockout restrain O-GlcNAcylation and hinder the expression of the early differentiation marker Nfatc1. Inhibition of OGA, which forces high levels of O-GlcNAcylation throughout the differentiation, reduces the formation of multinucleated mature osteoclasts. Consistent with our in vitro data, suppressing OGT and OGA both ameliorate bone loss in experimental arthritis. We detected a reduced number of TRAP-expressing precursors and mature osteoclasts in the mice subjected to OGT inhibition. While inhibiting OGA only lowers the number of TRAP+F4/80– mature osteoclasts without affecting the number of TRAP+F4/80+ precursors.Transcriptome profiling reveals that O-GlcNAcylation regulates several biological processes. Increased O-GlcNAcylation promotes cytokine signaling and oxidative phosphorylation. The downregulation of O-GlcNAcylation is essential for cytoskeleton organization and cell fusion. Conclusion:We demonstrate that the dynamic changes of O-GlcNAcylation are essential for osteoclast differentiation. These findings reveal the therapeutic potential of targeting O-GlcNAcylation in pathologic bone resorption.Disclosure of Interests:Chih-Wei Chen: None declared, Yi-Nan Li: None declared, Thuong Trinh-Minh: None declared, ZHU Honglin: None declared, Alexandru-Emil Matei: None declared, Xiao Ding: None declared, Cuong Tran Manh: None declared, Xiaohan Xu: None declared, Christoph Liebel: None declared, Ruifang Liang: None declared, Min-Chuan Huang: None declared, Neng-Yu Lin: None declared, Andreas Ramming Speakers bureau: Boehringer Ingelheim, Roche, Janssen, Consultant of: Boehringer Ingelheim, Novartis, Gilead, Pfizer, Grant/research support from: Pfizer, Novartis, Georg Schett Speakers bureau: AbbVie, BMS, Celgene, Janssen, Eli Lilly, Novartis, Roche and UCB, Jörg H.W. Abstract Background: Peripheral neuropathy occurs in 20-25% of patients with an IgM paraprotein and 40-50% of patients with an IgM monoclonal gammopathy have a detectable anti-myelin associated glycoprotein (MAG) antibody. The presence of an anti-MAG antibody is often associated with a chronic, progressive symmetric demyelinating polyneuropathy that can lead to sensory deficits and gait dysfunction, significantly altering a patient’s quality of life. Prior clinical trials have failed to demonstrate an effective long-term therapy for anti-MAG neuropathy. Additionally, the best manner to assess the evolution of this type of neuropathy has not been elucidated. We designed this clinical trial to evaluate a potential therapy for anti-MAG related neuropathy based on previous reports of subjective improvement in patients with anti-MAG neuropathy treated with rituximab or Bruton’s tyrosine kinase inhibitors. We included multiple neurologic outcome measures so their responsiveness and performance could be assessed in a clinical trial setting. Methods: This is a single-arm open-label phase II trial investigating the use of acalabrutinib and an anti-CD20 monoclonal antibody for the treatment of an IgM-associated anti-MAG mediated neuropathy. Eligible patients must have positive anti-MAG antibody titers with an IgM monoclonal gammopathy of undetermined significance (MGUS) or Waldenström macroglobulinemia (WM). Patients must have a predominantly sensory neuropathy characterized by demyelinating features in nerve conduction studies with a modified Rankin score of ≥1 with progressive symptoms or a modified Rankin score of ≥2. The ECOG performance status must be ≤2. Acceptable organ and marrow function are required. Prior exposure to chemotherapy, Bruton Tyrosine Kinase inhibitors, or other therapies for WM are not permitted except for steroids, IVIg, or an anti-CD20 monoclonal antibody given at least 90 days before study drug initiation. Approximately 33 patients will be enrolled in this study. Patients will be treated with acalabrutinib 100 mg orally twice daily on days 1-28 of each cycle and treatment will be administered until disease progression or unacceptable toxicity. Rituximab (or a biosimilar) will be administered on days 1, 8, 15, and 22 of cycles 1 and 4. The primary endpoint of this trial is to evaluate the overall hematologic response rate (defined as ≥25% reduction in serum IgM) associated with the treatment. The key secondary endpoint is to evaluate the proportion of patients that achieve improvement or stability in neuropathic symptoms based on the Inflammatory Rasch-built Overall Disability Scale (I-RODS) patient-reported disability scale. Additional secondary neurologic endpoints include the INCAT disability score, exam-based outcomes (the MRC distal sum score and INCAT-ISS), measures of function (10-meter walk test, 9-hole peg test), a pain scale, a fatigue scale (FSS), and a neuropathy-specific quality of life scale (IN-QOL). Additional secondary endpoints include time to next treatment, overall survival, hematologic response based on MYD88 and CXCR4 mutational status, change in bone marrow disease burden, and proportion of adverse events associated with treatment. This study is currently open and accruing patients. I temanummerets siste bidrag anmelder Ragnhild Laird Iversen Innføring i KRLE-didaktikk. God lesing! Abstract Background Lyme borreliosis (LB) is an infectious, vector-borne disease caused by the spirochete Borrelia burgdorferi sensu lato and mainly transmitted by the Ixodes species ticks. Public surveillance of LB occurs in 9 of 16 federal states of Germany and remains a critical facet of disease epidemiology and trends. We describe the incidence, time trends, seasonality, and geographic distribution of LB in Germany using publicly reported surveillance data. Methods We obtained LB cases and incidence (2016-2020) from the online platform SurvStat@RKI 2.0, maintained by the Robert Koch Institute (RKI). Cases and incidence of LB were extracted by year, geographic area, week of notification, sex, age and by 2 case definition categories (clinically diagnosed LB, clinically diagnosed and laboratory-confirmed LB) reported by 9 states where LB notification is mandatory. The nine states include Bavaria, Berlin, Brandenburg, Mecklenburg-Vorpommern, Saxony, Saxony-Anhalt, Thuringia, Rhineland-Palatinate, and Saarland. Results During 2016-2020, the 9 federal states reported 63,940 LB cases, of which 60,570 (94.7%) were clinically diagnosed, and 3,370 (5.3%) also had laboratory confirmation, with an average of 12,789 cases annually. LB incidence rates were mostly stable over time. The average annual LB incidence was 37.2/100,000 person-years, ranging from 22.9 to 64.6/100,000 person-years among 9 states; from 16.8 to 85.6/100,000 person-years among 19 regions; and from 2.9 to 172.8/100,000 person-years among 158 counties. Incidence was lowest among persons 20‒24 years old (16.1/100,000 person-years) and highest among those aged 65‒69 years old (60.9/100,000 person-years). Most cases were reported between June and September, with a peak in July of every year. Annual incidence (per 100,000 person-years) of overall Lyme borreliosis notified in 158 counties (NUTS3), 2016-2020 Conclusion The risk of LB varied substantially at the smallest geographic unit and to a lesser degree by age group. Our results underscore the importance of presenting LB data at the most spatially granular unit and by age to allow implementation of efficient preventive interventions and reduction strategies. This study is aimed at evaluating the relationship among students’ skills and their employability based on surveying students using 5 point Likert scale. The survey includes 490 students whose major are foreign languages and the results show that there are differences in the level of awareness between first year students, third year students and last year students. Thanks to the data analysis, it is revealed that most students believe that the university reputation has an important role in their employability (Mean= 3.80), they can do any kinds of work provided that work matches their skills and experience (Mean=3.05); therefore, students need to have some other skills in order to become successful in finding their jobs. Among the skills mentioned in the survey, the skills for working in group, skills for adaptability, skills for listening to others, skills for risk acceptability, skills for public speaking and skills for critical thinking are more effectively contributing factors to employability in comparison with other skills.
