Molecular motors are essential agents of force production in the cells: they convert the chemical energy released by the hydrolysis of ATP into mechanical work. This thesis focuses on myosins, a family of molecular motors responsible for actin-based motility. Myosin VI is unique among all of the myosin superfamily members in that it moves in the opposite direction of all other known myosins. Previous work revealed myosin VI tail ability to fold back, constituting a potential auto-inhibited state that prevents motor activity. Several myosin VI partners, binding to the C-terminal tail of the myosin, have been identified and shown to recruit the motor for different functions. In the first chapter of this thesis, the mechanism allowing the regulation of myosin VI activity has been studied using biochemical and biophysical analysis (SAXS, light scattering, microscopy, binding assays and mutagenesis). GIPC1, the most studied myosin VI partners, promotes myosin dimerization and activation. During my PhD, I have been also involved in two other projects, in line with my thesis project, that have led to the publication of four articles. Two shorter chapters are therefore devoted to these projects. The second chapter of my thesis explores myosin VII activity regulation by its cellular partners. Finally, the third chapter is devoted to the allosteric regulation of myosins activity by small molecules

Le but de notre thèse est d'investiguer, dans les cellules thyroïdiennes de chien en culture primaire, l'implication des protéines de la famille Rho et de l'organisation du cytosquelette d'actine dans les actions diverses que la TSH exerce, via l'AMPc, sur la morphologie, la prolifération, la différenciation et la fonction des thyrocytes de chien en culture primaire.
Trois cascades conduisant à la mitogénèse coexistent dans la cellule thyroïdienne de chien: la voie de l'AMPc stimulée par la TSH ou la forskoline (activateur direct de l'adénylate cyclase), la voie des facteurs de croissance (tels que l'EGF, l'HGF) activant leur récepteur à activité tyrosine kinase et la cascade dépendante de la protéine kinase C activée par les esters de phorbol (TPA). Contrairement aux voies indépendantes de l'AMPc qui répriment l'expression des caractéristiques de différenciation, la cascade de l'AMPc stimule à la fois la prolifération, l'expression des gènes de l'état différencié et la fonction (iodation, formation d'H2O2, sécrétion hormonale).
Dans la cellule thyroïdienne de chien, les agents activant les cascades dépendantes et indépendantes de l'AMPc ont des effets différents sur l'organisation du cytosquelette d'actine. La TSH/AMPc et le TPA induisent une destruction des microfilaments d'actine et un "ruffling" membranaire, tandis que les autres agents (insuline, EGF, HGF, sérum) ne modifient pas le réseau de fibres d'actine (fibres de stress) présent dans les cellules quiescentes.
Parmi les protéines de la famille Rho, RhoA, Rac1 et Cdc42 sont les premières à avoir été identifiées et sont actuellement les mieux caractérisées. Nous montrons que la TSH, via l'AMPc, induit une diminution de la concentration de la forme active des protéines Rac1, Cdc42 et RhoA. En revanche, les autres agents mitogènes, tels que l'EGF et le TPA, qui activent des voies indépendantes de l'AMPc, n'affectent pas les taux de Rac1 et Cdc42 activés, mais augmentent le taux de RhoA-GTP. L'activation ou l'inactivation des protéines RhoA, Rac1 et Cdc42 est donc un nouvel élément distinguant les voies dépendantes et indépendantes de l'AMPc.
Grâce à deux toxines bactériennes, la toxine B qui inactive les protéines Rho et la toxine CNF1 qui au contraire les active, nous montrons que, dans les thyrocytes, celles-ci jouent un rôle critique dans l'organisation du cytosquelette, dans la transition G1-S, dans l'expression des gènes de différenciation Tg, ThOXs, NIS et TPO, mais pas dans la génération d'H2O2.
En effet, l'activité d'un ou plusieurs membres de cette famille est nécessaire à l'entrée des thyrocytes en phase S et à la phosphorylation de la protéine pRb, étape pré-requise à la transition G1-S. L'activation de ces protéines n'induit cependant pas, à elle seule, la prolifération. Nous mettons également en évidence l'existence d'un nouveau mécanisme par lequel ces protéines contrôleraient l'activité des complexes cycline D3-CDK4 indépendamment de leur assemblage. Par l'utilisation de la dihydrocytochalasine B, qui comme la toxine B via l'inactivation des Rhos, désorganise le cytosquelette, nous démontrons que l'intégrité de celui-ci n'est pas requise pour la progression des thyrocytes en phases G1 et S. L'inactivation des protéines Rho est par contre nécessaire à l'induction, par l'AMPc, de l'expression des gènes de différenciation incluant Tg, ThOXs, NIS et TPO, puisque ce processus est inhibé par la toxine CNF1. De plus, l'inactivation des Rhos par la toxine B, ainsi que le désassemblage des fibres de stress et du cytosquelette induit par la dihydrocytochalasine B, suffisent à imiter l'induction dépendante de l'AMPc de Tg et ThOXs, mais pas de NIS et TPO. La toxine B et la dihydrocytochalasine B imitent aussi l’effet de la voie TSH/AMPc sur l’accumulation de p27kip1. Enfin, nous montrons que l'augmentation de la production d'H2O2, nécessaire à la synthèse des hormones thyroïdiennes, ne requiert pas l'activité de la protéine Rac (ni des autres protéines de la famille Rho) alors que celle-ci joue un rôle déterminant dans la génération d'H2O2 dans le leucocyte.

Cell motility is involved in critical biological functions, and dysregulation of adhesion, migration and of the actin cytoskeletal can lead to severe disease like cancer. Therefore, it is essential to study the molecular mechanism driving the formation of sub-cellular structures involved in cell motility. The first step in mesenchymal cell migration is the forward protrusion of the lamellipodium which is a thin sheet of membrane-enclosed actin filaments (F-Actin) networks propelled by actin polymerization. The spatiotemporal coordination of F-actin regulators in the lamellipodium determines the polarity, architecture and movements of branched F-actin networks. This includes two interacting nanomachines, the WAVE regulatory complex (WRC) and the Arp2/3 complex. WRC activation is the central molecular event triggering Arp2/3 complex activation and thus, the initiation and formation of a branched F-actin network in the lamellipodium. WRC activation relies on the exposure of the cryptic Arp2/3-activating WCA domain located at the C-terminal extremity of the WAVE subunit tail. In vitro studies showed that two WCA domains are needed to efficiently activate the Arp2/3 complex.But how the local spatial organization of WRC at the molecular level translate into activation of the Arp2/3 complex triggering the morphogenesis of the lamellipodium is unknown. In other words, the stoichiometry and the spatial distribution required to translate WRC conformational activation to an efficient activation of Arp2/3 complex are essential missing information.The recent application of super-resolution microscopy (SRM) and single particle tracking (SRM) lead to a drastic rethinking of macromolecular assemblies, in particular structures involved in cell migration, including the lamellipodium. By tracking individual proteins and delivering images with spatial resolutions below the diffraction limit of light, these techniques give access to the nanoscale organization and dynamics of protein complexes in live cells.To reveal the molecular organization of WRC, we use DNA-PAINT, a SRM technique which allows spatial resolution below 10 nm. DNA-PAINT is based on hybridization of complementary DNA strands, one located on the target protein (docking strand) and the other on the dye (imager strand). DNA-PAINT enable absolute molecular counting in protein complexes (Quantitative-PAINT) and multi-color super-resolution imaging (Exchange-PAINT). This allowed us to assess stoichiometries, colocalizations and composition of nanomachines in the lamellipodium.Using Quantitative-PAINT, we showed that the stoichiometry of WRC at the lamellipodium tip of migrating mouse melanoma cell (B16) is one; while live super-resolution imaging based on RESOLFT nanoscopy revealed that these single WRC form discrete foci at the lamellipodium tip. Multicolor Exchange-PAINT super-resolution microscopy of the WRC core and its WCA domain, showed that its conformational activation induces the release of the WCA domain in a radius of 40 nm away from its core. Using stereotyped waveform protrusions, we correlated WRC molecular organization with the rate of membrane protrusions. We showed that the spatial distribution of individual WRC is below the radius of its conformational unfolding in regions of faster lamellipodial protrusion, increasing the possibility of WCA domain dimerization and thus activation of the Arp2/3 complex. This way, the WRC, functioning as an isolated complex, must be spaced at a distance less than its conformational unfolding to activate efficiently the Arp2/3 complex in the lamellipodium. Altogether, our results show that besides biochemical activation of signaling circuitry, the spatial organization of proteins is crucial for controlling their function in cells

The bacterium Myxococcus xanthus forms multicellular structures called fruiting bodies to resist to starvation conditions. Fruiting body formation implies a chemosensory-like system, the Frz system which regulates directional changes through the simultaneous pole-to-pole relocalization of two motility systems, (A) and (S). During my PhD, I have worked on the connection between the Frz chemosensory-like system and the downstream regulators MglA and MglB in the control of polarity inversion. The cell polarity axis is established by (i) a Ras-like small G protein, MglA, which constitutes a branch node in the regulation of A and S motility systems at the leading cell pole, and (ii) its cognate inhibitor MglB that localizes at the lagging cell pole. We showed that MglA interacts directly and specifically with the cytoskeleton to promote assembly and disassembly of the A-motility machinery. Using an evolutionary approach, we elucidated the modular architecture of the Frz system and the implication of four regulatory domains to (i) connect the Frz system to the MglAB proteins, (ii) filter and (iii) amplify the signal. We now propose a mechanism for polarity inversion in which the independent action of two response regulators at each cell pole perturbs the interactions between a small-G-protein and its cognate inhibitor to trigger the conversion of a stable polarity axis into a biochemical oscillator. Les ARNm codant les récepteurs GalR1 et GalR2 sont exprimés par les neurones à POMC (principalement par ceux situés dans la partie antérieure du noyau arqué) et cette expression est stimulée en présence de testostérone. Dans un second temps, nous avons étudié l'effet de la galanine sur (i) la libération de [bêta]-endorphine et (ii) les niveaux d'expression d'ARNm codant la POMC à partir de fragments d'hypothalamus médiobasaux maintenus en survie. La galanine diminue la libération spontanée de [bêta]-endorphine dès les 30-60 premières minutes d'incubation et cet effet implique l'activation de calcineurine. Une augmentation des niveaux d'ARNm codant la POMC et une accumulation de [bêta]-endorphine dans les tissus sont observées plus tardivement (60-120 minutes). Compte-tenu de cette observation, nous pouvons donc envisager que cet effet tardif de la galanine sur l'expression de l'ARNm codant la POMC puisse être consécutive à la diminution de [bêta]-endorphine dans le milieu de survie. En effet, la [bêta]-endorphine pourrait intervenir sur sa propre régulation comme le montrent nos travaux relatifs a l'expression de l'ARNm codant le récepteur [mu] par les neurones à POMC. Dans la deuxième partie de ce travail, nous avons étudié la régulation du système à POMC par un facteur astrocytaire, le TGF[bêta] 1, qui, pour agir sur sa cellule-cible, doit se fixer sur un complexe hétérodimérique formé d'un récepteur de type II et d'un récepteur de type I. Le but de notre travail a donc été de rechercher si les neurones à POMC avaient la potentialité d'exprimer ces récepteurs. Nos résultats montrent [. . . ]

Manganese superoxide dismutase (Mn SOD or SOD2) is an important enzyme in the antioxidizing defence, which seems to play an unclear role in the cancer development, according to the constitutive expression of its gene. However, the regulation of this constitutive expression is not totally known, particularly in the breast cancer cells. This work is based on a preliminary revealing that a protein, called Damaged DNA Binding 2 (DDB2), specifically binds the SOD2 gene promoter. The DDB2 is known for its involvement in the nucleotide excision repair. At first step, we characterized the specific DNA sequence recognized in the proximal area of the SOD2 gene promoter, on which a DDB2 monomer binds, in order to regulate negatively the Mn SOD transcription in the MCF-7 non metastatic breast cancer cells. Besides, DDB2 is not involved in the mechanism of SOD2 gene induction, when MCF-7 cells are exposed to induced substances. However, we showed that the lack of the DDB2 protein, associated with the lack of the AP-2a transcription factor, already known as a repressor of the SOD2 gene, lead to a high Mn SOD constitutive expression in the metastatic breast cancer cells. Furthermore, this high constitutive expression is mainly dependent of the Sp1 transcription factor. Secondly, we estimated the biological meaning of the regulation of the Mn SOD constitutive expression by the DDB2 in the breast cancer cells. Our results show that the DDB2 activates the positive ER breast cancer cell proliferation, by exercising its negative regulation on the Mn SOD expression. Thirdly, we tried to show the consequences on the negative ER breast cancer cell growth, which naturally and highly express the Mn SOD. Our results reveal that the antioxidizing enzyme plays an important role in the molecular mechanisms involved in the invasive capacities of the negative ER breast cancer cells. The high Mn SOD expression, associated in a decrease of the H2O2 detoxifying enzymes expression, enhance the negative ER breast cancer cell invasion and an increase of the matrix metallopeptidase-9 activity. The H2O2 elimination, with specific antioxidants, decreases both negative ER breast cancer cell growth and invasive capacities. This whole work contributes to better understand the Mn SOD importance and the mechanism of its gene regulation, in the tumoral growth and invasion. This work also reveals the Mn SOD and DDB2 as potential predictive factors of the breast cancer progress. Finally, the discovery of this new DDB2 biological activity opens a huge field of interesting perspectives in breast cancer research

Au cours de notre thèse, nous avons poursuivi la caractérisation de ces sites de liaison AP-1 et avons montré que les facteurs c Fos, JunB et JunD interagissent in vitro avec ces motifs. Pour chaque site, nous avons identifié des mutations qui abolissent la liaison des facteurs AP-1 sans altérer la séquence en acides aminés sous-jacente de la transcriptase inverse. Par des expériences de transfection transitoire, nous avons démontré que les sites AP 1 intragéniques sont entièrement responsables de l’activité enhancer PMA-dépendante du fragment 5103. De plus, l’activité PMA-inductible du fragment 5103 est inhibée par le mutant dominant négatif A-Fos à condition que les sites ne soient pas mutés. A l’inverse, l’expression ectopique de dimères forcés AP-1 affecte positivement l’activité enhancer du fragment 5103. Enfin, nous avons étudié le rôle biologique des sites AP-1 intragéniques dans la réplication virale et avons montré que ces sites contribuent positivement à l’infectivité du virus.
Durant la seconde partie de notre thèse, nous avons entamé la caractérisation physique et fonctionnelle du fragment 5105. Nos résultats de transfection transitoire montrent que l’activité PMA inductible du fragment 5105 est localisée dans le dernier tiers de ce dernier :le sous fragment 5105.3. L’analyse bioinformatique de cette région a permis de mettre en évidence un site de liaison pour les facteurs AP-1 in vitro. Des mutations ponctuelles permettent d’abolir la liaison des facteurs à leur site mais altèrent la séquence en acides aminés sous-jacente codant pour les protéines Tat et Rev. Nous avons montré que ce site est impliqué dans l’activité transcriptionnelle de ce fragment. L’expression ectopique du mutant dominant négatif A-Fos inhibe l’activité transcriptionnelle PMA-inductible du fragment 5105. Une analyse bioinformatique plus large nous a ensuite permis d’identifier in vitro, par retard de migration sur gel, 5 sites de liaison pour le facteur YY1 et 2 sites de liaison pour le facteur PU.1 dont les implications pour le virus restent encore à déterminer.

Apoptosis is an essential physiological process through which organisms are able to equilibrate their cell numbers and maintain tissues in healthy and functional conditions. Despite the recent advances in the field, little is known on the molecular mechanisms controlling individual cell decisions to either engage or avoid the activation of this pathway upon cancer treatment, which inevitably impacts on therapeutics development. To obtain a global view of the intervening proteins and their role on cell response dynamics to anticancer drugs, a new and detailed description of the apoptosis pathway at the receptor level was translated into a system of ordinary differential equations. The model was calibrated to single-cell data, from recent experiments on a population of HeLa cells exhibiting a highly heterogeneous response when exposed to the death-inducing ligand TRAIL. The sensitivities of the apoptotic reactions in our model were evaluated using the diversity of experimental behaviors observed in vitro. A series of computational tests and analyses were performed with our model to identify the origins of cell response heterogeneity. New features of the apoptotic pathway emerged from a comparison of different heterogeneity modeling approaches, detecting a set of key reactions to be further expanded. These analyses yield new biological insights and highlights the importance of refining regulation of death receptor complex activity, possibly through Caspase-10 as suggested from new experimental discoveries. This thesis offers a novel framework that can be used to uncover important biological insights using single-cell data of heterogeneous dynamical pathways
