A presente pesquisa detém-se sobre a experiência desenvolvida pela Associação de Corais Infantis “Um Canto em Cada Canto”/ACIC a partir de histórias de vida e formação de artistas-educadores/as, também chamados regentes populares, que fizeram parte do trabalho de arte-educação realizado junto a 27 corais infantis na periferia de Fortaleza e cerca de 300 corais infantis no interior do Ceará entre 1988 e 2008. Cotejando a história de vida desses artistas-educadores/as da ACIC com a metodologia de trabalho da entidade que inclui três momentos: 1.amorização do grupo 2.trabalho com a música 3.o brincar/o brinquedo, a pesquisa qualitativa de cunho colaborativo realizada a partir de ciclos reflexivos e de entrevistas semiabertas identificará tanto os processos de auto, hetero e ecoformação na práxis educativa desses sujeitos, que têm na gênese do trabalho da ACIC um processo de formação que passa pela experiência do Polo de Música de Messejana, quanto os aprendizados para um possível trabalho com arte na escola. Breast cancer is the most frequently diagnosed cancer among women around the world. The hormone 17-estradiol (E2) is strongly implicated as a causative agent in this cancer. Since estrogen acts as a complete carcinogen, agents that interfere with the carcinogenic actions of E2 are required. Most agents effective against experimental mammary carcinogenesis have been employed as pure compounds disregarding the synergy that exists between several phytonutrients in a whole food. In these studies we have taken a unified approach, by employing a pure phytonutrient ellagic acid and whole foods that contain the phytonutrient at various levels berries, in the prevention of E2- induced mammary cancer in ACI rats. We have also used a tiered approach by screening several phytochemicals in vitro and implementing these results in both short- and long-term studies. Initially, several phytochemicals were tested as pure compounds against oxidative DNA damage induced by 4-hydroxy estradiol and CuCl2. Ellagic acid, was the most effective agent (andgt;98% reduction). In a short-term in vivo study, both dietary blueberry and strawberry (5% w/w), were ineffective in reducing the baseline oxidative DNA damage in the livers of CD-1 mice. However, red raspberry (5% w/w) was highly effective (50% reduction) and ellagic acid (400 ppm) was moderately effective (25% reduction). Further both diets up-regulated hepatic DNA repair genes in a similar fashion. In a long-term estradiol-induced mammary carcinogenicity study in ACI rats, dietary berries (2.5% w/w) and ellagic acid (400 ppm) reduced both tumor volume and tumors per animal to different extents (50-75%). One mechanism by which these dietary interventions inhibit mammary tumorigenesis may be via modulation of E2 metabolism, especially at the early stages of carcinogenesis. At 6 weeks after E2 treatment both berries and ellagic acid or berries alone significantly offset E2- induced changes in CYP1B1 and CYP1A1 expressions respectively. In addition, no toxicity or adverse effects are observed when rodents were fed either berries (1 - 5%) or ellagic acid (400 ppm). These data taken collectively support the possibility of using natural foods such as berries as an adjuvant to current pharmacological therapies in the prevention and treatment of breast cancer.

Microfibrillated cellulose (MFC) is an alternative material that has been widely studied to enhance the mechanical properties of a polymer matrix due to a number of perceived advantages over traditional plant fibre forms. Mechanical properties of MFC networks were found to depend on parameters such as the modulus of fibrils, bonding strength, porosity, degree of crystallinity, contact area of fibrils and possibly the modulus of the cellulose crystals of the raw materials (cellulose I or II). Even though the longer processing time used to produce MFC was found to yield networks with fewer fibre aggregates, finer fibrils and higher density, some properties, for instance thermal stability and degree of crystallinity, decreased due to the degradation of fibrils caused by the harsh treatment. The aims of this thesis were to assess the mechanical properties and interfaces of composites produced using of a range of MFC materials, prepared using different treatments and from different sources. Raman spectroscopy has been used to detect the molecular orientation of cellulose chains within an MFC network, and to monitor the deformation micromechanics of MFC networks. The Raman band initially located at ~1095 cm-1 obtained from MFC networks was observed to shift towards a lower wavenumber position upon the application of tensile deformation. The intensity of this band as a function of rotation angle of MFC networks was similar, indicating randomly oriented networks of fibrils. From the Raman band shift rate data, the effective moduli of MFC single fibrils produced from pulp were estimated to be in the range of 29-41 GPa. Poly(lactic acid) (PLA) composites reinforced with MFC networks were prepared using compression moulding. Enhanced mechanical properties of MFC reinforced composites were reported, compared to neat PLA films. The mechanical properties of these composites were found to mainly depend on the interaction of the PLA matrix and the reinforcement phase. The mechanical properties of the composites reinforced with dense networks were shown to be dominated by the network properties (fibril-fibril interactions), while matrix-fibril interactions played a major role where more opened networks were used to reinforce a polymer matrix. The penetration of the matrix into the network was found to depend on the pore sizes, fibre width and porosity within the network. It was found that the matrix easily penetrates into the network with a range of mean fibril dimensions, rather than for networks with only fine fibrils present. The stress-transfer process in MFC reinforced PLA composites was monitored using Raman spectroscopy. Greater Raman band shift rates with respect to tensile deformation for the composites were observed compared to pure MFC networks. This indicates that stress is transferred from the PLA matrix to MFC fibrils, supporting the enhancement of the mechanical properties of the composites.

This thesis presents evidence for a novel mechanism of regulation of pyruvate dehydrogenase (PDH) kinase by fatty acids and also results of a study of muscle triacylglycerol concentration. In animals regulation of PDH complex activity is central to the selection of respiratory fuels and to the conservation of glucose during carbohydrate deprivation. The principal means of regulation of PDH complex is interconversion of phosphorylated (inactive) and dephosphorylated (active) forms effected by PDH kinase and PDH phosphatase. Earlier in vitro studies by others had identified both shorter term (min) and longer term (hours) mechanisms of activation of PDH kinase by fatty acid. In the present study PDH kinase activity (as measured by rates of ATP-dependent inactivation of PDH complex in extracts) was shown to be increased when rat heart mitochondria were incubated with palmitoyl-L-carnitine [PC] (and other CoA utilising respiratory substrates). The activation of PDH kinase persisted through removal of respiratory substrate following incubation with CCCP. A comparable effect of PC was also demonstrable in heart mitochondria from 48h-starved rats (i.e. the mechanism may be distinct from that which increases PDH kinase activity in starvation). Rates of ATP-dependent inactivation of PDH complex were also increased when extracts of rat heart mitochondria were incubated with palmitoyl-CoA (PCoA); the increase was comparable with that seen on incubation of intact mitochondria with PC. The PC effect in intact mitochondria and the PCoA effect in mitochondrial extracts may not be identical as PCoA further increased PDH kinase activity in extracts from mitochondria incubated with PC. The Protestant Ethic is an ideology which espouses a belief that success is a reward for hard work and determination, whilst a lack of success results from moral failings of self indulgence and a lack of self discipline (Quinn & Crocker, 1999). Thus, positive outcomes will be received by those who deserve them, have worked hard and are morally superior, but those who demonstrate a lack of self discipline, self indulgence and are morally flawed will experience negative outcomes. It has been suggested that Protestant Ethic values provide the foundations for the formation of anti-fat attitudes (Crandall, 1994; Hoverd & Sibley, 2007). Drawing primarily on this framework, this thesis examines the social perceptions of obesity employs a number of theories relevant to the aim of each study in the programme of research. Consisting of seven chapters, this thesis had four main aims. First, to examine UK adults’ perceptions of obesity using implicit and explicit measures, employing Attribution Theory (Weiner, 1995) and System Justification Theory (Jost & Banaji, 1994) to explain beliefs about the controllability of obesity and anti-fat attitudes reported by individuals across a range of weight categories. Second, to examine the portrayal of obesity in UK newspapers, employing the Attribution theory (Weiner, 1995), System Justification Theory (1994) and Cultivation Theory (Gebner & Gross, 1976) to explain newspaper portrayal of the causes of obesity, self stigmatisation by the overweight and obese, and why individuals may perceive media portrayals as a reflection of reality. Third, to examine the effects of media portrayals of obesity, drawing on the Protestant Ethic values and Cultivation Theory (Gebner & Gross, 1976) to explain changes self and other perception that occur when exposed to media portrayals of obesity. Fourth, to examine the effectiveness of counter-conditioning as an attitude change intervention, using the Elaboration Likelihood Model (Petty & Cacioppo, 1986) to explain the anti-fat attitude formation and maintenance. Chapter 3 introduces the first study which examines implicit and explicit perceptions of obesity in UK adults aged 18-65 years. A cross-sectional sample of 2380 participants was recruited from across the UK. Participants completed implicit and explicit attitude measures of perceptions of obesity, and responses were analysed to examine how attitudes and beliefs compare across a range of demographic variables, such as age, sex and exercise frequency. This research highlighted that certain adult populations are more likely to report anti-fat attitudes about obesity, associate more negative characteristics with being fat and perceive obesity to be more controllable (e.g., high food consumption). The findings demonstrate evidence of beliefs that obesity is controllable explained using Attribution Theory (Weiner, 1995) and that the overweight and obese report anti-fat attitudes supporting the proposals of System Justification Theory (Jost & Banaji, 1994). This research also demonstrated that anti-fat attitudes are more negative than in previous reports and that differences were evident between implicit and explicit attitudes, highlighting the importance of measuring both forms of attitude. Alongside other research in this area, the study highlights attitude change as a potential aspect of obesity treatment and management. This consequently warrants attention in future research. Chapter 4 presents the second study which examines the portrayal of obesity in six UK newspapers over the period of a year, comparing broadsheet and tabloid depictions. This research demonstrated the difference in portrayal of the condition between broadsheet and tabloid newspapers, for example there were more articles about obesity and interventions to treat excessive fat in tabloid newspapers. Evidence also highlights the potential for media portrayals to contribute to negative perceptions of obesity with sparse commentary on genetic influences which may lead to misconceptions concerning the causes of obesity and the extent of media focus on obesity. Findings also demonstrate that Protestant Ethic values are evident within newspaper portrayals of obesity, promoting moral judgement of the overweight and obese. Portrayals of overweight and obesity are discussed using the Attribution Theory (Weiner, 1995) and the System Justification Theory (Jost & Banaji, 1994). The majority of appearance related research has examined television and magazine portrayals of obesity. Therefore by examining newspaper portrayal of obesity this adds to and extends current knowledge, for instance presenting evidence of the portrayal of obesity as a moral transgression. Chapter 5 presents study three which examines the effects of media portrayal on self and other perception, using three different sources to increase interpersonal awareness through exposure to others’ physical appearance and behaviour. Participants (n = 29) were exposed to three stimuli (UK tabloid newspaper article, UK broadsheet newspaper article and an obesity related UK television programme). Results indicated that the broadsheet and tabloid newspaper articles had deleterious effects on other perception, which are explained using the Protestant Ethic as these portrayals include information indicative of the ‘sins’ of gluttony and slothfulness. This research provides much needed novel information as the majority of appearance related research has examined the effects of general television programmes and fashion magazines on body related perceptions and on thinness and self perception. Chapter 6 presents study four which examines the effects of exposure to images of overweight and obese individuals on perceptions of obesity. A sample of 28 participants attended the laboratory on three separate occasions to complete implicit and explicit measures of attitudes towards obesity. In visit 1, baseline measures were obtained, followed, in visits 2 and 3 by two experimental conditions where participants were exposed to images of overweight and obese celebrities or members of the general public in counterbalanced order. It was initially expected that the images of overweight and obese celebrities would reduce anti-fat attitudes through their positive affiliation; however, this was not observed, but interestingly anti-fat attitudes worsened. On the basis of these results, it was suggested that formation of anti-fat attitudes occurs through the central route of processing of the Elaboration Likelihood Model (Petty & Cacioppo, 1986). This implies that anti-fat attitudes may be robust and resistant to change. Results highlight a number of potential avenues for future research as well as providing important information on the effects that exposure to images of overweight and obese individuals may have on perceptions of obesity. Not only did this research suggest at the complex nature of perceptions of obesity, but also demonstrates the need for long term intervention to change anti-fat attitudes. Chapter 7 provides a discussion of the overall research programme, including applied implications that derive from the findings, strengths and limitations, and future research suggestions. Applied recommendations include intervening with media portrayal of obesity given the deleterious portrayal observed in study 2, the development of effective interventions to modify anti-fat attitudes, and increasing understanding of the condition to ensure that psychological and social aspects of the condition are appreciated alongside biological knowledge. As described above, this thesis also offers a number of potential research avenues for future research that appear warranted on the basis of the findings of the four studies. This thesis has made a number of useful contributions to research examining psychosocial aspects of obesity, demonstrating the pervasive nature of anti-fat attitudes, the link between Protestant Ethic values and negative perceptions of obesity, and the fruitfulness drawing on a number of theories to explain anti-fat attitude formation and maintenance.

Age-related hearing loss (ARHL), or Presbycusis, is the most prevalent sensory impairment observed in the elderly. It is a progressive, symmetrical, age-related sensorineural hearing loss, most pronounced at higher frequencies. ARHL is a multifactorial disease, with contribution from both environmental and genetic factors. To date, little progress has been made in determining the genetic loci involved. The aim of my doctorate studies is to elaborate upon the genetics underlying ARHL through the identification and characterization of ENU-induced mouse models of ARHL. This approach has identified trombone, a recessive model of ARHL arising from the Harwell Ageing Screen. Recurrent auditory phenotyping at 2, 6, 9 and 12 months of age shows that affected animals display elevated ABR thresholds from 9 months of age, when compared to littermates, and these are further increased at 12 months of age. Genome mapping studies identified a 12.5Mb critical region on chromosome 2 and next generation sequencing identified a T>C mutation in the novel deafness gene Slc4a10, causing a leucine to proline substitution in the encoded protein. Immunohistochemical staining of cochlear sections demonstrates that Slc4a10 is expressed in the type II and V fibrocytes of the spiral ligament of wildtype mice, whereas no labelling is observed in Slc4a10trmb/trmb mice. In addition, ultrastructural studies show progressive sensory hair cell loss (inner and outer) in the Slc4a10trmb/trmb mice from >6 months of age. Furthermore, histological assessment of the lateral wall identified strial thinning in the Slc4a10trmb/trmb mice. Given the expression pattern and morphological changes observed, endocochlear potentials were measured in these mice. This identified that Slc4a10trmb/trmb mice have a chronically low endocochlear potential compared to their wildtype and heterozygous littermates. My findings establish the presence of Slc4a10 in the inner ear and suggest an important role for this sodium-coupled bicarbonate transporter in normal auditory function. I hypothesize that trombone is a novel model of strial presbycusis and further functional characterization of this model promises to increase our understanding of the pathobiology associated with age-related hearing loss.

The causes of childhood cancers are not well understood. That infections are believed to play an important role in childhood cancer development is of particular interest in sub-Saharan Africa, where infectious diseases are common. The objectives of this thesis were to identify childhood cancers associated with HIV, malaria, EBV and HHV-8, and to investigate child and maternal factors associated with Burkitt lymphoma and Kaposi sarcoma. In Blantyre, Malawi, 305 children diagnosed with cancer and 212 of their mothers, were recruited. Risk factor data were collected using a brief questionnaire and blood samples tested for infections. Case-control analyses were conducted to compare 148 Burkitt lymphoma cases and 22 Kaposi sarcoma case with a control group comprising 104 children with cancers other than those known to be associated with HIV. The prevalence of HIV was 6% among children with Burkitt lymphoma and 2% in controls (OR=12.4, 95% CI 1.3 to 116.2). Tumour risk increased with increasing litres of antibodies against EBV and malaria. In comparison with those who had low titres against both EBV and malaria, the highest risk of Burkitt lymphoma was among those with high titres against both infections (OR=13.2, 95% CI 3.8 to 46.6). Reported use of mosquito nets was protective against Burkitt lymphoma. The prevalence of HIV was 81% among children with Kaposi sarcoma (OR=762.7, 95% CI 44 to 13376), and risk increased with increasing HHV-8 antibodies. Prevalence of infections was also examined among children with other cancer types and no associations were identified, although the number of cases was small. Few maternal factors were found to be associated with cancer in children. This research demonstrates that infections play a particularly important role in increasing the risk of Burkitt lymphoma and Kaposi sarcoma in children in sub- Saharan Africa. Prevention or early treatment of these infections may be vital in the control of childhood cancer.

A novel hydrogen atom source, involving the 193.3 nm photolysis of nitrous oxide in the presence of hydrogen, is described. The system is shown to provide a very clean source of hydrogen atoms, particularly at temperatures above 400K. The H + C2H4 addition reaction is studied as a function of pressure (50-750 Torr He) and temperature (285-604K). Resonance fluorescence was used to monitor hydrogen atom concentrations in real time, following excimer (ArF) photolysis of the N20/H2 precursor. The data are analysed using the Troe factorisation method. A model is developed, which allows the accurate calculation of the rate constant for the H + C2H4 reaction over a wide range of temperature and pressure. The direct observation of equilibration in the H + C2H4 = C2H5 system is reported for the first time, at temperatures between 775 and 850K. Hydrogen atom concentrations were monitored by resonance fluorescence, following 193.3 nm photolysis of the ethylene substrate. The equilibrium constant for the system Kc is determined much more precisely than has previously been possible. The standard heat of formation of the ethyl radical, ΔHof, 298(C2H5), is evaluated from the measured values of Kc(T), using Third Law methods and the known molecular and atomic parameters of H, C2H4 and C2H5 giving ΔHof 298(C2H5) = 28.36 ± 0.40 kcalmol . The quoted errors refer to 95.45% confidence limits and include contributions from the scatter of the data and from estimates of possible systematic errors. An excimer laser flash photolysis/ gas chromatographic end product analysis apparatus is described. The design of the photolysis cell allows its entire contents to be transferred to the analysis column, and enables total C3 hydrocarbon yields as low as 2.6x1012 molecules to be detected. A study of the photolysis of acetone at 193.3 nm is presented. The dependence of product yields on pressure (10-500 Torr He), temperature (300,600K), laser pulse energy and the presence of radical scavengers (NO,C2H4) is described. Under all experimental conditions, methyl radicals account for >95% of the radical products of the photolysis. The ArF photolysis of acetone is demonstrated to provide a very clean source of methyl radicals, suitable for use in conventional laser flash photolysis /kinetic spectroscopy experiments, at temperatures of up to 825K. Experiments are described, in which ethane and ethane/propylene mixtures, diluted to ∿5% in nitrogen, were pyrolysed in a quartz jetstirred reactor. Temperatures and conversions comparable to those achieved in industrial pyrolysis were employed. A kinetic model of ethane pyrolysis is developed, which qualitatively reproduces the experimental results. It is shown that the model predictions are extremely sensitive to a number of poorly defined rate constants.

In this thesis, the smartphone is installed on vehicle as a tool to detect potholes on the road. The data collected and pre-processed by smartphone is then analysed to obtain the result. The history and status of pothole detection study is firstly discussed. The traditional pothole detection method has two disadvantages: 1, low efficiency; 2, limited detection area. To work out a new method, the suspension models and tyre models are studied. Also the axis-correction and de-noise in signal process discussed for data analysis. Based on these, the requirement of detection system is analysed, and the features of embedded systems are discussed, which leads to the conclusion that smartphone is the best hardware for pothole detection. Then the smartphone (3 mobiles are chosen: Samsung Note 1, Nexus 7 and iPhone 5) is fixed on vehicle (Nissan Micra K11 and Saab 93) as experiment platform. The Tay Road Bridge is chosen as experiment road. The software Sensor Insider Pro and Sensor Data are chosen to collect data. Simulations about vehicle and tyre are done by MATLAB Simulink. From the simulation the relationship between the bouncing height and the speed of vehicle is obtained, and is used as threshold in pothole detection. The experimental data from acceleration sensor and gyroscope is processed by MATLAB using axis-correction and wavelet transform. Together with velocity data gathered by GPS and threshold (calculated from speed), the position of pothole is obtained. Comparing with road, the detection has good accuracy, which proves the feasibility of this pothole detection method. At the end of thesis, other potential field of application of smartphone is discussed.

This thesis is concerned with the debate between absolutists and relativists about generality. Absolutists about quantification contend that we can quantify over absolutely everything; relativists deny this. The introduction motivates and elucidates the dispute. More familiar, restrictionist versions of relativism, according to which the range of quantifiers is always subject to restriction, are distinguished from the view defended in this thesis, an expansionist version of relativism, according to which the range of quantifiers is always open to expansion. The remainder of the thesis is split into three parts. Part I focuses on generality. Chapter 2 is concerned with the semantics of quantifiers. Unlike the restrictionist, the expansionist need not disagree with the absolutist about the semantics of quantifier domain restriction. It is argued that the threat of a certain form of semantic pessimism, used as an objection against restrictionism, also arises, in some cases, for absolutism, but is avoided by expansionism. Chapter 3 is primarily engaged in a defensive project, responding to a number of objections in the literature: the objection that the relativist is unable to coherently state her view, the objection that absolute generality is needed in logic and philosophy, and the objection that relativism is unable to accommodate ‘kind generalisations’. To meet these objections, suitable schematic and modal resources are introduced and relativism is given a precise formulation. Part II concerns issues in the philosophy of mathematics pertinent to the absolutism/relativism debate. Chapter 4 draws on the modal and schematic resources introduced in the previous chapter to regiment and generalise the key argument for relativism based on the set-theoretic paradoxes. Chapter 5 argues that relativism permits a natural motivation for Zermelo-Fraenkel set theory. A new, bi-modal axiomatisation of the iterative conception of set is presented. It is argued that such a theory improves on both its non-modal and modal rivals. Part III aims to meet a thus far unfulfilled explanatory burden facing expansionist relativism. The final chapter draws on principles from metasemantics to offer a positive account of how universes of discourse may be expanded, and assesses the prospects for a novel argument for relativism on this basis.

Human cancers exhibit the common phenotype of elevated glycolytic metabolism, which causes acidification of the tissue microenvironment and may facilitate tumour invasion. In this thesis, we use mathematical models to address a series of open problems underlying the glycolytic tumour phenotype and its attendant acidity. We first explore tissue-scale consequences of metabolically-derived acid. Incorporating more biological detail into a canonical model of acidity at the tumour-host interface, we extend the range of tumour behaviours captured by the modelling framework. We then carry out an asymptotic travelling wave analysis to express invasive tumour properties in terms of fundamental parameters, and find that interstitial gaps between an advancing tumour and retreating healthy tissue, characteristic of aggressive invasion and comprising a controversial feature of the original model, are less significant under our generalised formulation. Subsequently, we evaluate a potential role of lactate---historically assumed to be a passive byproduct of glycolytic metabolism---in a perfusion-dependent metabolic symbiosis that was recently proposed as a beneficial tumour behaviour. Upon developing a minimal model of dual glucose-lactate consumption in vivo and employing a multidimensional sensitivity analysis, we find that symbiosis may not be straightforwardly beneficial for our model tumour. Moreover, new in vitro experiments, carried out by an experimental collaborator, place U87 glioblastoma tumours in a weakly symbiotic parameter regime despite their clinical malignancy. These results suggest that intratumoural metabolic cooperation is unlikely to be an important role for lactate. Finally, we examine the complex pH regulation system that governs expulsion of metabolically derived acid loads across tumour cell membranes. This system differs from the healthy system by expression of only a few key proteins, yet its dynamics are non-intuitive in the crowded and poorly perfused in vivo environment. We systematically develop a model of tumour pH regulation, beginning with a single-cell scenario and progressing to a spheroid, within a Bayesian framework that incorporates information from in vitro data contributed by a second experimental collaborator. We predict that a net effect of pH regulation is a straightforward transmembrane pH gradient, but also that existing treatments are unable to disrupt the system strongly enough to cause tumour cell death. Taken together, our models help to elucidate previously unresolved features of glycolytic tumour metabolism, and illustrate the utility of a combined mathematical, statistical, and experimental approach for testing biological hypotheses. Opportunities for further investigation are discussed.

This thesis reports the study of single photon sources that emit one infrared wavelength photon at a time, creating cavity quantum electrodynamical effects for applications such as quantum information processing. This work considers two major single photon sources: a) InAs single quantum dots and b) single carbon nanotubes, which both emit in the infrared range. Photonic crystal slabs and photonic crystal waveguides are served as distinctive passive devices with manipulated photonic band-gaps to control the propagating light. A simulation of leaky modes of two-dimensional photonic crystal slabs is introduced to constrain model parameters in the device design. Fullerenes are used as fluorescent material to achieve resonance of a leaky mode with excitation 1492 nm and emission at 1519 nm and to see enhancement of the PL. We include novel characterization techniques and PL measurements to show sharp emission peaks from single quantum dots and successfully couple them to micro-cavities. The strong coupling effect is observed and is amongst the best examples of cavity-dot structures achieved to date. Single-walled carbon nanotubes have shown anti-bunched light emission, thus we systematically study them as another possible candidate of single photon sources. PLE spectra show clear evidence of the existence of excited states, and time evolution measurements reveal the disorder induced diffusion, which separate the tubes into a series of quantum dots. These strongly confined states are concluded as the origin of the possibility that single-walled carbon nanotubes are single photon sources.

Hepatitis C virus (HCV) causes severe illness in millions of people worldwide, but the epidemic strains responsible for most infections arose within the past hundred years and represent only a small part of total HCV diversity. In this thesis I combine laboratory and computational methods to study HCV in Africa. I aim to characterize its current genetic diversity and its historical transmission prior to the global HCV epidemic. In Chapter 2 I begin by screening samples from the Democratic Republic of the Congo (DRC) for HCV and the related human pegivirus. I find high HCV sequence diversity, including a putative new subtype, and find significantly higher HCV prevalence in those born before 1950. Chapter 3 continues this screening, and combines the sequences obtained with those from online databases. Using molcular clock methods I estimate that genotype 4 originated in central Africa around 1733, and that multiple lineages, including subtype 4a which dominates the HCV epidemic in Egypt, have moved to north Africa since ~1850. In Chapter 4 I analyse sequences sampled from an elderly population in Kinshasa to estimate HCV’s transmission history there during the 20th century. The results indicate a rapid increase in HCV transmission between 1950 and 1970 in multiple independent lineages. Possible causes of this increase are discussed. This study population also exhibits high HCV genetic diversity, including the second genotype 7 sample discovered to date. Finally, Chapter 5 uses a range of sequencing techniques, including RNAseq, to characterise two putative HCV recombinants from Cameroon. I confirm that both sequences are recombinants, and generate a full genome sequence for one. I also develop new tools to distinguish between dual infection and recombination in next-generation sequencing data, and discuss how recombination might affect HCV diversity and treatment.

This thesis consists of three chapters that analyze theoretically the role of income inequality in consumer markets. Each chapter introduces distributional considerations into an economic model where previously inequality did not play a major role. Chapter one uses a consumer search model to show under what conditions the distribution of income within a community is related to the type of firms that exist within that community, impacting the level of prices. We show that if time and money costs of search are high enough, only the middle class have incentive to search and therefore are the most aggressive shoppers. Using a supply side model, we argue that firms located in more informed communities are more likely to enter the market as large low-priced retailers. Connecting these two results, the model shows under what conditions the size of the middle class can have a negative relationship with the level of prices. Chapter two demonstrates how firm pricing strategy and determinants of household location can interact to determine city structure. In this city, consumers and firms live on a continuous line interval. The model consists of two types of firms; many high-cost perfectly competitive firms located in the Central Business District, and one large low-cost "Superstore", choosing its price strategically. We show how the shopping habits of the consumer population, as determined by the relative price of the Superstore and the Corner Stores, can contribute to the various income segregation outcomes described in previous literature. In addition we consider the impact of city population structure on the pricing decision of a monopolist facing a competitive fringe. Chapter three uses a simple model of banking services to consider how deposit-taking banks price for their services and choose the type of deposit customers that they target. This chapter goes beyond previous theoretical work on consumer banking, identifying the role of household income in the access to deposit services. We show that a higher rate of return on investments available to banks lowers financial exclusion, increasing the profitability of low-income consumers for deposit-taking institutions. This suggests that the possibility of financial exclusion increases in periods of recession. The chapter demonstrates how an increase in income dispersion can lead to a greater proportion of consumers excluded from mainstream banking.

This project problematizes the general understanding of China's birth planning regime. By providing an analytical history of a two-child policy experiment, it moves beyond previous work to generate granular accounts of key policymaking decisions. A 'critical junctures' approach centred around what has been called the 'Experiment of Eight Million People' was leveraged to unlock this alternative chronology within China's national fertility policy. The Experiment was a two-child pilot that governed select rural counties for more than thirty years, but it has not yet been systematically examined in the English-language literature. Tools from critical junctures theory, or the idea that explicating possible inflection points builds understanding of policy mechanisms, provide a framework to identify the times when the two-child design appeared most likely to be on the cusp of national expansion. Data from Chinese media, declassified internal government documents, secondary literature, and interviews with elite Chinese academics and policymakers are then leveraged to provide detailed descriptions of the events that predicated the policy outcomes at these points. This process yielded the first English account of the mechanisms that triggered China's change to a national two-child policy in 2015. Finally, results from this analysis were used to hypothesize about which political factors were most central in facilitating or obstructing changes in a Chinese social policy. Beyond Chinese birth planning specifically, the project has implications for the study of political factors enabling or constraining social policy change, welfare policy in East Asian and authoritarian states, and the degree to which population policies serve a social function.
