The relationship between pre-flowering climatic conditions and sugary disease incidence was quantified in grain and forage sorghum genotypes at two sowing periods (mid November and mid December). The trials were carried out over the 2001/02 and 2002/03 growing seasons, in Lages, Santa Catarina State. Four commercial male-fertile sorghum hybrids (BR 600-forrage, BR 700-grain, BR 701-forage/silage and BR 800-forage) and one male-sterile inbred line (BR 001-A) were evaluated. When each genotype reached the flowering stage, 50 panicles were marked and sprayed with a suspension of Claviceps africana (1,000 spores mL-1). Air temperature, humidity and rainfall were recorded through out the growing cycle. Low temperatures three to four weeks prior to flowering, increased susceptibility. At both sowing periods, average night temperatures lower than 15°C during the critical period of pre-flowering turned the fertile hybrids as susceptible as the male-sterile inbred to ergot infection. The tested hybrids differed in their ability to tolerate pre-flowering cold stress. Seed set in uninoculated heads under pollination bags was also reduced, suggesting that increased susceptibility to sugary disease was the result of low temperature induced sterility.

Impaired endothelial vasodilatation may contribute to the exaggerated blood pressure (BP) responses to exercise in individuals who are overweight/obese. The present study investigated whether consumption of cocoa flavanols, which improve endothelium-dependent flow-mediated dilatation (FMD), can modify BP responsiveness to exercise. Twenty-one volunteers (eight females and thirteen males, 54·9 (se2·2) years, BMI 31·6 (se0·8) kg/m2, systolic BP 134 (se2) mmHg, diastolic BP (DBP) 87 (se2) mmHg) were randomised to consume single servings of either a high-flavanol (HF, 701 mg) or a low-flavanol (LF, 22 mg) cocoa beverage in a double-blind, cross-over design with 3–7-d washout between treatments. Two hours after cocoa consumption, FMD was measured, followed by continuous beat-to-beat assessment (Finapres™) of BP before and during 10 min of cycling at 75 % of age-predicted maximum heart rate. Averaged data from two assessments on each type of beverage were compared by analysis of covariance using pre-exercise BP as the covariate. Pre-exercise BP was similar after taking LF and HF (153 (se3)/88 (se3)v. 153 (se4)/87 (se2) mmHg, respectively,P>0·05). However, the BP response to exercise (area under BP curve) was attenuated by HF compared with LF. BP increases were 68 % lower for DBP (P = 0·03) and 14 % lower for mean BP (P = 0·05). FMD measurements were higher after taking HF than after taking LF (6·1 (se0·6) %v. 3·4 (se0·5) %,P < 0·001). By facilitating vasodilation and attenuating exercise-induced increases in BP, cocoa flavanols may decrease cardiovascular risk and enhance the cardiovascular benefits of moderate intensity exercise in at-risk individuals.

ImportanceBRAF variants are associated with tumor progression; however, the prevalence of BRAF variant subtypes and their association with disease characteristics, prognosis, and targeted therapy response in patients with intrahepatic cholangiocarcinoma (ICC) are largely unknown.ObjectiveTo explore the association of BRAF variant subtypes with disease characteristics, prognosis, and targeted therapy response in patients with ICC.Design, Setting, and ParticipantsIn this cohort study, 1175 patients who underwent curative resection for ICC from January 1, 2009, through December 31, 2017, were evaluated at a single hospital in China. Whole-exome sequencing, targeted sequencing, and Sanger sequencing were performed to identify BRAF variants. The Kaplan-Meier method and log-rank test were used to compare overall survival (OS) and disease-free survival (DFS). Univariate and multivariate analyses were performed using Cox proportional hazards regression. Associations between BRAF variants and targeted therapy response were tested in 6 BRAF-variant, patient-derived organoid lines and in 3 of the patient donors of those lines. Data were analyzed from June 1, 2021, to March 15, 2022.InterventionsHepatectomy in patients with ICC.Main Outcomes and MeasuresThe association of BRAF variant subtypes with OS and DFS.ResultsOf 1175 patients with ICC, the mean (SD) age was 59.4 (10.4) years and 701 (59.7%) were men. A total of 20 different subtypes of BRAF somatic variance affecting 49 patients (4.2%) were identified; V600E was the most frequent allele in this cohort, accounting for 27% of the identified BRAF variants, followed by K601E (14%), D594G (12%), and N581S (6%). Compared with patients with non-V600E BRAF variants, patients with BRAF V600E variants were more likely to have large tumor size (10 of 13 [77%] vs 12 of 36 [33%]; P = .007), multiple tumors (7 of 13 [54%] vs 8 of 36 [22%]; P = .04), and more vascular/bile duct invasion (7 of 13 [54%] vs 8 of 36 [22%]; P = .04). Multivariate analysis revealed that BRAF V600E variants, but not overall BRAF variants or non-V600E BRAF variants, were associated with poor OS (hazard ratio [HR], 1.87; 95% CI, 1.05-3.33; P = .03) and DFS (HR, 1.66; 95% CI, 1.03-2.97; P = .04). There were also broad differences among organoids with different BRAF variant subtypes in sensitivity to BRAF or MEK inhibitors. Conclusions and RelevanceThe findings of this cohort study suggest that there are broad differences among organoids with different BRAF variant subtypes in sensitivity to BRAF or MEK inhibitors. Identifying and classifying BRAF variants may be able to help guide precise treatment for patients with ICC.

Purpose This study aims to examine the setting of International Standards on Auditing (ISA) 701 on disclosing key audit matters (KAMs) to explore the role of standard setting in maintaining or reconstituting the relationship of the auditing profession with preparers and users of financial reports. Design/methodology/approach This study draws on concepts from the sociology of the professions literature and the regulatory space metaphor. Data comprises comment letters and other documents pertaining to the setting of ISA 701. Findings The study shows that the KAM reporting requirement is part of the ongoing re-calibration of the regulatory arrangements governing auditing, which started in the early 2000s. This study interprets standard setting as a site for negotiating the relationships between linked ecologies in the audit regulatory space, namely, the auditing profession, preparers of financial statements and users of audited reports. This study identifies three processes involved in setting ISA 701, namely, reconstitution of the rules governing auditors’ reports as a link between the three ecologies, preserving boundaries between the auditing profession and preparers and negotiation aimed at balancing competing interests of the interrelated ecologies. Originality/value The study offers insights into the role of regulatory rule setting as a central medium through which the adaptive relationship of the profession with its environment is negotiated.

Internal tandem duplication (ITD) mutations of the juxtamembrane domain–coding sequence of the FLT3 gene are found in up to 34% of patients with acute myeloid leukemia (AML) and are associated with a poor prognosis. FLT3/ITDs result in constitutive activation of the tyrosine kinase domain and transform growth factor–dependent cell lines. FLT3 activation leads to antiapoptotic and proliferative signals, but little is known about the impact of FLT3/ITDs on differentiation. This study was designed to investigate the effect of FLT3/ITD expression on the differentiation of the 32Dcl3 (32D) myeloblastic cell line to neutrophils in response to granulocyte colony-stimulating factor (G-CSF). Expression of FLT3/ITD completely blocked morphologic differentiation and induction of myeloperoxidase (MPO), lysozyme, and CCAAT/enhancer-binding protein ε (C/EBPε) in response to G-CSF. Wild-type FLT3 and vector-transfected 32D cells were able to differentiate, although the maturation of FLT3-transfected cells was delayed by FLT3 ligand (FL) stimulation. CEP-701, a potent FLT3 tyrosine kinase inhibitor, overcame the morphologic block in differentiation caused by FLT3/ITD expression and allowed G-CSF induction of myeloid maturation markers. These findings suggest that blocking differentiation may be one of the mechanisms by which FLT3/ITDs contribute to leukemogenesis. CEP-701 and other FLT3 inhibitors may be useful for overcoming the block to differentiation (as well as the block to apoptosis) in the leukemic cells of patients with AML.

Background:For treating Rheumatoid Arthritis (RA) it’s clear that Treat to Target has presented the best strategy. The best outcome for patients has been attained by accepting clinical remission as the final objective in treatment. The development of new drugs for the treatment of RA has dramatically modified the approach clinicians have on the disease, however, the treatment of inflammatory conditions represents a challenge due to the multiple factors can affect its response. It is known that non-adherence represents an increase in mortality, morbidity and healthcare costs.1,2.Emphasis should be made on one of the most simple and effective variables that impact the effectiveness of treatment: adherence to treatment itself. Objectives:This work aimed to evaluate the impact of methotrexate adherence on treatment outcome in patients with RA.Methods:An observational, cross sectional study was performed using medical records from the Rheumatology Clinic at University Hospital from UANL Jose Eleuterio Gonzalez, Monterrey, México; the data was collected from 03/16/2018 to 01/29/2020. RA patients visits included fulfilled the 2010 ACR/EULAR classification, had their disease activity measured by Disease Activity Score 28 ESR (DAS28 ESR) and were treated with methotrexate as their primary disease modifying antirheumatic-drug (DMRAD). Patient disease activity was classified in 4 groups according to the DAS28 ESR score as <2.6 remission, 2.6-3.2 low activity, >3.2-5.1 moderate activity, >5.1 high activity.Adherence to medication was evaluated utilizing a self-reported assessment that was applied per patient’s visit. Patients were classified into 4 groups according to the percentage of the prescribed doses of Methotrexate that they abided to. Statistical analysis was performed using IBM SPSS 21 statistical package. Descriptive analyses were performed with frequencies (%) and the correlation between the calculated disease activity versus patient adherence to treatment was calculated using Pearson correlation coefficient.P-values <0.05 were considered statistically significant. Results:A total of 795 patients visits were included. They were 92.33% female, mean (SD) age was 52.44 (±12.99) (Table 1). The most frequent routes for methotrexate were Oral 603 (75.84%), Subcutaneous 113 (14.21%) and Intramuscular 79 (9.93%). A significant correlationr(795) =0.-183, (p<0.001) was found between Disease Activity according to DAS28 ESR and Adherence classification according to prescribed doses takenTable 1.Baseline CharacteristicsN= 795Age, mean SD52.44 (±12.99)Sex, n (%) Female734 Male61Methotrexate (mg/week)Average Dose19.94Median Dose20Intramuscular79 (9.93%)Subcutaneous113 (14.21%)Oral603 (75.84%)Disease Activity accordingto DAS28 ESR, n (%)Remission343 (42.3)Low activity101 (12.5)Moderate activity292 (36.0)High activity59 (7.3)Adherence classification according to prescribed doses taken, n (%)Absent (<25%)20 (2.5%)Poor (25-49%)19 (2.3%)Regular (50-74%)55 (6.8%)Adequate (>75%)701 (86.5%)Conclusion:Adherence to treatment with methotrexate therapy has a direct impact on RA treatment outcome. The evaluation of adherence to the prescribed treatment should be addressed before modifications in DMRAD therapy in patients with Rheumatoid Arthritis. Before considering treatment failure in a patient treated with methotrexate, adherence to treatment must be evaluated. According to our results, self-reported adherence appears to be a cost and time effective method to care for patients.References:[1]Monchablon C, et. al. Uncontrolled kochia plants that regrow after small-grain harvest can produce substantial numbers of seeds. An average of 4,100 seeds per plant were produced between harvest (late July to mid August) and the first killing frost (late September) at three locations in Montana. Field experiments were conducted to determine the optimal timing of postharvest herbicide applications to prevent kochia from producing viable seeds. Herbicide treatments were applied at three timings from late August to mid September. The most effective treatments were glyphosate (631 g/ha) and paraquat (701 g/ha) applied at the second application timing (late August to early September). These treatments reduced kochia seed production by 92% or greater at each site. Kochia regrowth by this time had sufficient leaf area for herbicide absorption, but few viable seed had been produced. Herbicide treatments at the first and third application timings were generally less effective and more variable in reducing kochia seed production. Sulfentrazone (157 g/ha) and 2,4-D (561 g/ha) were not as effective at reducing seed production as other herbicide treatments.

Abstract Annexin A2 (ANXA2) promotes myeloma cell growth, reduces apoptosis in myeloma cell lines, and increases osteoclast formation. ANXA2 has been described in small cohorts of samples as expressed by myeloma cells and cells of the BM microenvironment. To investigate its clinical role, we assessed 1148 samples including independent cohorts of 332 and 701 CD138-purified myeloma cell samples from previously untreated patients together with clinical prognostic factors, chromosomal aberrations, and gene expression–based high-risk scores, along with expression of ANXA2 in whole BM samples, stromal cells, osteoblasts, osteoclasts, and BM sera. ANXA2 is expressed in all normal and malignant plasma cell samples. Higher ANXA2 expression in myeloma cells is associated with significantly inferior event-free and overall survival independently of conventional prognostic factors and is associated with gene expression–determined high risk and high proliferation. Within the BM, all cell populations, including osteoblasts, osteoclasts, and stromal cells, express ANXA2. ANXA2 expression is increased significantly in myelomatous versus normal BM serum. ANXA2 exemplifies an interesting class of targetable bone-remodeling factors expressed by normal and malignant plasma cells and the BM microenvironment that have a significant impact on survival of myeloma patients.

Abstract Introduction Sleep disturbances after a trauma forecast mental health difficulties such as post-traumatic stress symptoms (PTSS). COVID-19 has resulted in numerous stressors for youth (e.g., school closures, social isolation) that directly disrupt sleep and may have negative mental health consequences. However, investigations of sleep and adverse events in youth have primarily been limited to months or years after a trauma has occurred. How adolescent sleep patterns and mental health intersect during an ongoing adverse experience is unknown. Methods Adolescents from the United States (N = 503; 80.9% female; 13–18 years) participated in an online daily diary study within 3 weeks after COVID-19 was declared a national emergency. Participants completed one week of twice daily reports (5406 observations) on their nightly sleep (sleep timing, sleep quality, sleep onset difficulties, and nightmares) and daytime mental health symptoms (COVID-19 PTSS, positive affect, negative affect, and loneliness). Results Mixed models adjusting for age, sex, and socioeconomic status indicated that cyclical, bidirectional effects emerged, with daytime mental health symptoms predicting same-night sleep disturbances, and sleep disturbances predicting next-day mental health. Greater PTSS predicted sleep onset difficulties (Estimate = .02, SE = .004, t = 5.56 p < .001) and sleep onset difficulties predicted next-day PTSS (Estimate = .35, SE = .17, t = 2.04, p < .05). Greater daytime negative affect predicted greater nightmares (Estimate = .07, SE = .01, t = 4.97, p < .001), and nightmares predicted marginally more next-day negative affect (Estimate = .07, SE = .04, t = 1.66, p = .09). PTSS (Estimate = .01, SE = .003, t = 3.47, p < .001) and negative affect (Estimate = .05, SE = .03, t = 2.06, p < .05) both predicted poorer sleep quality the following night. Conclusion Findings suggest that mental health symptoms during the onset of the COVID-19 pandemic are associated with increased sleep disturbances. In turn, these sleep disturbances predict increased mental health symptoms. Overall, results provide a greater understanding of youth sleep patterns during COVID-19 and how sleep disturbances may inhibit resilience in the context of an ongoing stressor. Abstract Introduction: The significance of viral respiratory infections (VRI) and efficacy of anti-flu vaccine in patients (pts) with lymphoproliferative malignancies (LPM) have not been fully elucidated. The current large comprehensive study was designed to evaluate the incidence of respiratory infections in LPM pts, define clinical significance of VRI and assess the impact of anti-flu vaccine on RI prevention in this population. Methods: All consecutive pts, diagnosed either with non-Hodgkin lymphoma (NHL) or multiple myeloma (MM), followed at the Rambam Hematology Outpatient Unit between 01/2011 and 03/2012 (during or after anti-cancer therapy or untreated) were evaluated. Data regarding anti-lymphoma/MM therapy used, respiratory infections occurring during the study period, tests performed to assess these infections (focusing on viral ones), administration of anti-flu vaccine and infection-related outcome were recorded. Samples of nasopharyngeal aspirate (NPA) or bronchoalveolar lavage (BAL) were evaluated for VRI pathogens using both immunofluorescence and polymerase chain reaction (PCR) methods. Results: Five hundred and sixty two respiratory infection episodes were reported in 369 pts. In 224 (40%) episodes, VRI investigation was performed using either NPA (n=180) or BAL (n=44) samples. The decision regarding the sample source was taken based on the respiratory infection severity. The cohort screened for VRI included a larger proportion of pts receiving anti-cancer therapy during the study period than the non-screened cohort (70% vs 30%, respectively, p<0.01). The number of respiratory episodes requiring hospitalization for their respiratory infection was higher in the former cohort (54% vs 18%). One hundred fifty two screened episodes were community-acquired (50 required hospitalization) and 72 were hospital-acquired. Fever was the most common clinical symptom in screened episodes (n=111, 50%), followed by cough (n=91, 41%), dyspnea (n= 87, 39%), runny nose (n=47, 21%), respiratory distress (n=45, 20%): with decreased O2 saturation (n=35, 16%) and mechanical ventilation (n=10, 4%). In 12% and 23% of episodes, respectively, neutropenia and significant hypogammaglobinemia (IgG <500 mg/dl) were revealed. VRI were identified in 66/224 episodes (29%), 59 episodes were viral infections and 7 - co-infections (viral with another pathogen). Sixty viral episodes were diagnosed in pts with upper respiratory tract infections and 6 in pts with pneumonia. Cough, abnormal lung physical findings and hypogammaglobinemia were statistically more frequent in pts with viral infections vs those with other respiratory infections. Viral infections were treated according to pathogens recognized. Tamiflu was prescribed for influenza infections and ribavirin was given for part of the respiratory syncytial virus (RSV) infections. Supportive treatment only was given for parainfluensa and metapneumovirus infections. Twelve episodes of VRI, developed outside of hospital, required hospitalization (5%) and 2 of these episodes resulted in death. The most frequent viral infection was Influenza A (22 cases), followed by RSV (14 cases), parainfluenza and metapneumovirus (11 cases each). Less frequent viral pathogens were Influenza B (6 cases), and adenovirus (2 cases). Seven cases (10%) were accompanied with a secondary bacterial or fungal infection. Notably, 3 of the 10 pts diagnosed with RSV infection had a concurrent Aspergillus infection. Data regarding anti-flu vaccination were available in 701 pts; 431 pts (61%) were vaccinated. In a univariate analysis, anti-flu vaccination was found to have no impact on the respiratory infection frequency (51% vs 48.9%, p=0.752). Among pts with VRI, 49% did and 45% did not previously receive anti-flu vaccination (p=0.95). This vaccination had no effect on the VRI incidence in the investigated episodes. Conclusions: VRI, although frequent, are usually associated with a good outcome in LPM pts. Presence of cough and abnormal pulmonary examination were found to predict a positive screen for a viral pathogen. Influenza and RSV, both potentially treatable with Tamiflu and ribavirin, respectively, were responsible for 55% of VRI episodes, justifying a wider use of viral screening in LPM pts. Anti-flu vaccination has not proved its efficacy either in preventing viral infections or in reducing the entire incidence of respiratory infections in these pts. Disclosures No relevant conflicts of interest to declare.

e4360011The objective of this research was to analyze how official documents and educational legislation present the subject of ethnic-racial diversity, and also reflect on their impact on reality through the National Household Sample Survey (PNAD) data. To this end, we conducted an analysis of legislation and official documents created for Brazilian education, from 1988 to 2018, based on the Bardin Content Analysis technique. Subsequently, this analysis was compared with the data available in the PNAD to verify the impact of publications regarding 1) average years of study by color/race in the period; 2) average years of study by color/race, considering the situation of poverty (extremely poor, vulnerable, poor and non-poor); and 3) average years of study in regions of Brazil with a higher percentage of black people. The prescriptions related to the education of ethnic-racial relations of the 12 files analyzed were described, as well as their categories and characteristics. After this process, were established the relations between the categories and the PNAD data. The results allowed us to conclude that the reduction in the average time difference between blacks and whites was a little less than 5 months. Despite the relevance of the efforts, the implementation of policies aimed at combating racial inequality in education is still far from achieving the expected effectiveness. Keywords: Ethnic-Racial relations, Educational legislation, Racial inequalities, Content analysis.ReferencesAGUIAR, Márcia Angela Da S. e4176066In this paper we present a discussion about the implications of the theory of mental models in the teaching and learning process in Chemistry, with the purpose of discussing the use of stop motion animation as a data collection instrument to investigate the students' mental representations about chemical concepts, especially about their dynamic character. To achieve the objectives mentioned in the present work, we seek to relate the theory of mental models and their implications for the teaching and learning processes in chemistry, with the proposal of the triangulation of the three different representational levels of Chemistry (submicroscopic, symbolic and macroscopic), which are necessary to understand this Science. It is also noteworthy that this articulation was based on the main difficulties associated with the teaching and learning processes in Chemistry and was thought of the particularity involving the dynamics of the atomic-molecular level. The discussions carried out involving the different references used allowed us to discuss the potentialities associated with the use of stop motion as an instrument for investigating mental models on chemical concepts, highlighting the fact that it is a technique that is easy to apply, low cost and that allows the dynamic representation of chemical phenomena considering the submicroscopic level. Keywords: Educational Research, Educational Technology, Cognitive Psychology, Evaluation methods.ReferencesBACHELARD, G. Objective: Lower limb amputation is increasingly common yet outcomes are historically poor, with high rates of short-term mortality, revisions and poor quality of life. This observational study evaluates outcomes from a high-volume tertiary vascular referral centre. Methods: Retrospective electronic case note review was performed to elicit all major and minor amputations performed between 03/11/2014 and 31/05/2022 for peripheral arterial disease and diabetic foot sepsis. Cases of amputation for trauma or chronic pain were not included. Results: A total of 1,566 amputations (865 major, 701 minor) were carried out in 1,237 patients during the study period. Mortality was significantly higher for patients undergoing major amputation compared with minor amputation at 30 days (7.9%, n=61/773 vs 1.0%, n=6/575; p<0.0001) and at 1 year post-procedure (23.7%, n=183/773 vs 13.6%, n=78/575, p<0.0001). Revision rates were significantly lower for major amputations than for minor amputations at 30 days (2.4%, n=19/799 vs 13.2%, n=91/691; p<0.0001) and at 1 year post-procedure (10.0%, n=68/677 vs 26.3%, n=163/619, p<0.0001). Of the patients with major amputation fitted with a prosthesis, there was no significant difference in the numbers of patients ambulating (p=0.19) or the mean ambulatory level (p=0.08) between above-knee and below-knee amputation at 6 months post-procedure. Conclusions: Both major and minor lower limb amputations are associated with a high risk of revision and mortality within 1 year which should be discussed explicitly as part of the informed consent process.

Abstract Funding Acknowledgements Type of funding sources: None. Objectives The impairment of left atrial mechanics(LA) is previously described in hypertrophic cardiomyopathy (HCM). It is independently influenced by left ventricular(LV) mechanics and correlated to the severity of LV phenotype. We examined the alterations in LA structure and mechanics following surgical septal myectomy using vector velocity imaging. Methods Twenty-five HCM patients, 60% males with mean age (34.5 ± 12 years) were investigated before and six months after surgical myectomy using VVI. In addition to LA size and volume index (LAD, LAVI), LA reservoir function using peak systolic strain(εsys) & systolic strain rate(SRsys), and LA conduit(SRe) and LA contractile(SRa) functions were analyzed. Electrophysiologic properties of the LA were investigated using the difference between longest and shortest cycle; time to peak SRa(TTP-d) and the standard deviation(SD) of TTP-mean derived from regional SRa curves. Similar longitudinal εsys parameters were quantified in LV and right ventricle (RV) from apical views. The magnitude of changes in LA function (Δ) was expressed as the difference between pre and post myectomy values. Results Post myectomy, the studied cohort demonstrated atrial reverse remodeling and 68% of patients showed normalization of LAVI (<34ml/m2). All patients demonstrated a reduction of: LVOT gradient to < 20 mmHg, septal thickness, LV mass index, severity of mitral regurgitation and improvement of functional class from NYHA class III to class I. Following myectomy, although there was significant reduction of LV and RV systolic mechanics, there was a considerable increase in LA reservoir function (εsys:26 ± 13 vs 31 ± 16%, P<.03, SRsys:0.9 ± 0.4 vs 1.2 ± 0.5% s-1, P < 0.02) and contractile function (SRa: -0.8 ± 0.4 vs -1.2 ± 0.9 s-1, P < 0.04) respectively. LA Δ εsys was inversely correlated to pre-myectomy maximal wall thickness (r=-.494, P<.003) & LAVI (r=-.489, P<.01) and directly correlated to pre-myectomy LA εsys % (r=.639, P<.001) and LA SRa (r=.536, P<.008). Δ SRe showed direct correlation to LV εsys (r=.405, P< .04) and LA SRe (r=.635, P<.001) before surgery, respectively. ΔSRa was directly correlated to LA εsys % before surgery (r=.701, P<.0001). ΔTTP-SD and ΔTTP-d, showed inverse correlation to LV systolic mechanics before surgery (r=-.418, P<.038 and r=-.395, P<.050) respectively. Stepwise linear regression analysis identified global LV εsys% and RV εsys pre-myectomy as independent determinants of normalization of LAVI following myectomy, (β: 0.6, 95% CI: -4.22 - -1.96, p< 0.001) (β: 0.4, 95% CI: -4.06 - -0.57, p < 0.04) respectively. Conclusion Surgical myectomy is associated with improvement of LA mechanical properties and reverse remolding in HCM patient with symptomatic improvement despite subclinical reduction of LV and RV systolic mechanics.

Abstract Background Cystatin C is a low molecular weight non-glycosylated alkaline protein, with a molecular weight of 13kDa. It is formed by all nucleated cells in human at a constant rate. The only pathway of Cystatin C in vivo is excretion through glomerular filtration and absorbed by proximal renal tubular epithelial cells, which was decomposed finally. Therefore, the concentration of Cystatin C in serum is almost not affected by external factors, such as weight, age, gender, etc. Compared with creatinine and urea, it is an ideal endogenous marker for monitoring glomerular filtration function. Currently, the most methods on the market for testing Cystatin C are either latex enhanced immunoturbidimetric method, or immunoturbidimetric method. These methods usually use polyclonal antibodies from different animal species, which are sometime susceptible to the interference from rheumatoid factors reported in the literature, resulting in false positive results. The anti-RF interference ability of this reagent was enhanced by selecting the optimal antibody and optimizing reagent formulation. This study evaluated performances of the Cystatin C assay on Mindray BS-2000M system. Method Mindray Cystatin C assay is based on latex immunno-agglutination. Latex nanoparticles were coated with anti-Cystatin C antibody. Reagent of this assay contains R1 buffer and R2 with latex particles coupled with anti-Cystatin C antibodies. The assay starts with R1 and sample incubation for 5 min and then R2 is added. After another 5 min of incubation, agglutination occurs, leading to turbidity increasing. The turbidity is measured at 570 nm. Multi-level (5-point) calibration is used. Cystatin C concentrations are obtained from the calibration curve with signals from the assay with patient samples. Following basic assay performances were evaluated: precision, limit of detection, linearity, method comparison, interference and high dose hook. Results The precision study followed EP05-03 protocol. The repeatability and within-lab CVs on the BS-2000M system with 2 QC and 3 patient samples were ranged from 0.9% to 5.3%, respectively. Limit of detection was 0.1 mg/L and linearity was given up to 8.63 mg/L. The assay (y) shows good accuracy when compared with Roche Cystatin C (x) assay on the cobas 701: y = 1.0055x - 0.0268 (r2 = 0.9994, n = 109). The assay showed no significant with bilirubin up to 40 mg/dL, with hemoglobin up to 500 mg/dL, with lipids up to 500 mg/dL, and with rheumatoid factors up to 1200 IU/mL that is about 0.3% out of a total of 55 184 samples collected from our customers in China with RF measurements. No prozone was observed with the assay up to the high cystatin C concentration of 800 mg/L tested. Conclusion From this evaluation, we confirmed that Mindray Cystatin C assay can measure serum Cystatin C precisely and accurately, and with good performance against rheumatoid factors interference. It is suitable for use in routine clinical laboratories.

PurposeThe purpose of this paper is to examine career engagement and perceived employability during the school-to-work transition. We studied within-person changes in career engagement and perceived employability in the transition from higher education to the labor market. We investigated their dynamic reciprocal relationship to unravel whether career engagement or perceived employability is the leading indicator in the relationship in view of providing adequate support for students during the school-to-work transition.Design/methodology/approachWe conducted latent change score (LCS) analyses on a three-wave sample of 701 graduates in Flanders (Belgium). We collected data in July (right before graduation), November and May. LCS is a novel method allowing to simultaneously test change and reciprocal relationships.FindingsOur findings demonstrated how both career engagement and perceived employability changed (within-person) non-linearly during the school-to-work transition. As for their relationship, we found that perceived employability is the driving force in the relationship. Perceived employability fueled subsequent positive changes in career engagement, whereas career engagement did not lead to subsequent changes in perceived employability.Originality/valueOur study connects the career development and the graduate employability literature, and examines the school-to-work transition from preparation for the labor market to ten months after graduation. We also make an important methodological contribution, demonstrating the added value of LCS for studying employability in higher education. Our findings provide insights in how higher education institutions may support students in the school-to-work transition.

PurposeThe purpose of this study is to understand the effects of self-risk perception and health consciousness on the intention to purchase organic food. The study also explored external factors, such as social networking, culture and their impact on attitude, self-risk perception and purchasing organic food.Design/methodology/approachThis study uses a quantitative research method to collect data from New Zealand and Fiji (N = 701). The data analysis used the partial least squares path modeling technique (PLS-PM) to test the proposed model.FindingsThe empirical results revealed that self-risk perception positively influenced organic food intention. The results show that the health consciousness level strengthens the relationship between self-risk perception, beliefs, values and purchase intention. The health consciousness level dampens the relationship between social networking and purchase intention.Research limitations/implicationsThe findings of this study provide valuable insights into the guidance and promotion of Fijian and New Zealand's organic food industry. This study enables marketers to develop health-related promotional tactics to stimulate organic food sales. It gauges organic food promoters to use social media-oriented consumer networking to spread health awareness swiftly.Originality/valueThis comprehensive study extends the literature by scrutinizing the profundity of self-risk perception and health consciousness in influencing and explaining consumers' purchase intentions. Aside from ample growth in the study of organic food purchase intention, which commonly replicates simple relationships, this study ascertains deeper meaning and new relationships to understand the moderating role of health consciousness levels in organic food studies, expanding the theory of planned behavior.

The U.S. is the world’s leading producer of highbush blueberries (Vaccinium corymbosum L.), and Michigan is ranked in the top five production states (USDA NASS, 2022). In June and July of 2021, 268 blueberry stem blight samples were collected for a pathogen survey across 22 total fields in Van Buren and Ottawa counties in Michigan. Current season stems with symptoms of necrosis and wilting were collected. Stems were cut just below the necrotic area and cross-sections (2-3 mm long) were surface disinfested in 10% bleach for 1 min, rinsed twice in sterile distilled water, and dried on sterile paper towels. Stem cross-sections were plated onto potato dextrose agar (PDA) amended with 100 µg/ml streptomycin sulfate and 50 µg/ml ampicillin. Plates were incubated at 21°C under a 12-h photoperiod for 5-6 days. Outgrowing fungi with morphology similar to Diaporthe spp. were transferred to new PDA plates 2 consecutive times after 7 days of similar incubation to ensure single colony isolation. After 7 days, colonies consisted of white and light brown mycelia that were mostly flat, with some isolates that had partially raised mycelia towards the center of the plate. After 3-4 weeks, colonies turned brown and gray and produced dark brown pycnidia. Aseptate, hyaline, fusiform to ellipsoid, biguttulate alpha conidia measuring 5.4 to 7.6 x 2.6 to 3.7 µm (n = 60) were produced. No beta conidia were observed. In total, 3 isolates, representing 3 different farms (37-95 km apart) and cultivars (‘Duke’, ‘Jersey’, and ‘Bluecrop’), as well as 2 counties, were identified as Diaporthe through colony morphology (Gomes et al. 2013, Udayanga et al. 2014). Amplification and subsequent Sanger sequencing were performed for the internal transcribed spacer (ITS) region and portions of the translation elongation factor (TEF) 1-α, β-tubulin (TUB), and histone H3 (HIS) genes using primers ITS5/ITS4 (White et al. 1990), EF1-728F/EF1-986R (Carbone and Kohn 1999), T1/Bt-2b (Glass and Donaldson 1995), and CYLH3F/H3-1b (Glass and Donaldson 1995), respectively. Representative sequences were deposited in NCBI GenBank (accession no. OQ507870-OQ507872 for ITS, and OQ550272-OQ550278 for TEF, HIS, and TUB). BLASTn results revealed 97-100% identity for all 4 genes across other established D. eres isolates reported in Gomes et al. (2013). For example, JMK047 had 99.8% (577/578 bp), 99.7% (327/328 bp), 100% (701/701 bp), and 100% (439/439 bp) homology with ITS, TEF, TUB, and HIS sequences, respectively, of D. eres CBS 439.82 (accession no. KC343090, KC343816, KC344058, KC343574). Koch’s Postulates were fulfilled via pathogenicity tests on 2-year-old potted ‘Blueray’ plants with 2 isolates. Stems were surface sterilized with 1% bleach then 8-mm long pieces of bark were removed using a sterile razor blade to expose the cambium. Plugs of sterile PDA (negative control) or mycelia from 7-day old cultures on PDA (5-mm diameter) were placed onto the cambium layer and sealed with Parafilm. Six stems on unique plants were inoculated per treatment. Plants were grown in a 20.5°C greenhouse with a 14-hr photoperiod. After 3 weeks, the stems inoculated with D. eres isolates showed similar stem blight symptoms to those observed in the field while control stems remained healthy. Re-isolation and sequencing of the ITS region of 3 replicates per treatment with the protocol described above confirmed symptoms correlated with D. eres isolates. This is the first report of D. eres associated with stem blight of highbush blueberry in Michigan, and the second report in the U.S. (Lombard et al. 2014). Increasing prevalence of D. eres in U.S. blueberries may affect disease management programs. References Carbone, I., and Kohn, L. M. 1999. Mycologia 91:553. 10.1080/00275514.1999.12061051. Glass, N. The paper focuses on the filtration and electrical anisotropy coefficients and relationship between vertical and horizontal permeability in sandstone reservoir rocks. Field case study of DDB reservoir rocks. Petrophysical properties and parameters are estimated from core and log data from a Moscovian and Serpukhovian stages of Dnipro-Donetsk Basin (West-Shebelynka area well 701-Bis and South-Kolomak area well 31). Routine core analysis included estimation of absolute permeability, open porosity, irreducible water saturation and electrical resistivity (on dry and saturated by mineralized solution) of 40 core samples along two orthogonal directions. Shale fraction is estimated using well logging data in wells which are analyzed. The authors report that reservoir rocks are represented by compacted poor-porous (φ <10 %), low permeable (k<1mD) laminated sandstone with different ratios of clay minerals (Vsh from 0,03 to 0,7) and high volume of micaceous minerals (in some cases 20-30 %). Research theory. One of the main objectives of the work is to develop empirical correlation between vertical permeability and other capacitive and filtration properties for compacted sandstone reservoirs. A modified Kozeny-Carman equation and the concept of hydraulic average radius form the basis for the technique. Results. Coefficients of the anisotropy of gas permeability (IA) and electrical resistivity (λ) are defined based on the results of petrophysical studies. The experiments proved that IA lies in a range from 0,49 to 5 and λ from 0,77 to 1,06. Permeability and electrical resistivity anisotropy in most cases have horizontal distribution. It has been shown that in West-Shebelynka area sample №1 (depth 4933 m) there is probably no fluids flow in vertical direction and in samples №№3 and 15 fractures have the vertical orientation. We have also found that the values of electrical and filtration anisotropy for all samples of South-Kolomak area are similar, this characterized the unidirectionality in their filtration properties, as well as the fact that the motion of the fluid flow mainly in the horizontal direction. In the studied rocks the degree of anisotropy has been concluded to depend on the volume of clay and micaceous minerals, their stratification, fractures, density, and their orientation. New correlation between vertical permeability, horizontal permeability and effective porosity are developed for Late Carboniferous DDB intervals that are analyzed.

Internet of Things (IoT) applications are increasingly making impact in all areas of humanlife. Day by day, its chatty embedded devices have been generating tons of data requiring effectivenetwork infrastructure. To deliver millions of IoT messages back and fort with as few faults aspossible, participation of communication protocols like MQTT is a must. Lightweight blueprintand friendly battery are just two of many advantages of this protocol making it become a dominantin IoT world. In real application scenarios, distributed MQTT solutions are usually required sincecentralized MQTT approach is incapable of dealing with huge amount of data. Although distributedMQTT solutions are scalable, they do not adapt to fluctuations of traffic workload. This might costIoT service provider because of redundant computation resources. This leads to the need of a novelapproach that can adapt its size changes in workload. This article proposes such an elastic solutionby proposing a flexible MQTT framework. Our MQTT framework uses off-the-shelf componentsto obtain server’s elasticity while keeping IoT applications intact. Experiments are conducted tovalidate elasticity function provided by an implementation of our framework.
