The thesis is devoted to the solution of one of the current scientific and technical problems of modern medical instrument making - increasing the efficiency of the medical ozone generator by determining the parameters of the discharge chamber, the supply voltage and the oxygen consumption. Currently, ozone and its mixtures are widely used in the world for the wide range of applications. Different types of ozone generators depending on their application are used for ozone synthesis. These generators differ in size, the amount of ozone generated by the time span, etc. The peculiarity of the generators of medical ozone is that it is necessary to maintain the prescribed parameters of the resulting ozone-oxygen mixture throughout the entire duration of the procedure, and medical oxygen is used as the working gas. This allows us to implement ozone therapy techniques for the most effective treatment of patients. Typical parameters of the ozone-oxygen mixture, which the ozone generating plant should provide, are regulated in Ukraine "Methods for the application of ozone in medical practice", which were approved by the Ministry of Health in 2004. According to the modern version of these methods, the concentration of ozone in the ozone-oxygen mixture should vary in the range from 0.1 to 80 mg / l, and the flow rate of the mixture is from 0.1 to 1 l / min. Medical ozone generators are actively used in the offices of ozonotherapy and medical institutions in Ukraine, CIS countries, as well as Latin American countries. A large number of conferences devoted to ozone therapy are being held annually in the world in which it is further developed. The practical use of generators of medical ozone shows that most of the equipment used needs to be improved to provide work on the basis of modern medical solutions in treatment. That is, the development of a modern generator requires not only compliance with the parameters of modern requirements, but also a stock for development and the possibility of increasing the values of ozone concentration. In addition, in most medical ozone generators there is a discrepancy between the stated parameters of the ozone oxygen mixture and the value of ozone productivity. The performance of the ozone generator, in turn, depends on: the parameters of the discharge chamber, the parameters of the supply voltage, the parameters of the working gas. Many scientists, including Japanese, German and scholars from the CIS countries, are engaged in solving the problem of finding the optimal values of these parameters for increasing the efficiency of ozone generator operation. But at the moment there is still no universal model for calculating the parameters of the ozone generator. Thus, the task of increasing the efficiency of the medical ozone generator by determining the best parameters of its nodes, which will improve the quality of ozone therapy procedures, is an urgent and promising direction for the development of ozone generators. In the dissertation, for the first time, a mathematical model of the synthesis of ozone in a digital chamber of the medical ozone generator was constructed, which takes into account the influence not only of the parameters of electric energy, but also of the parameters of the discharge chamber and the working gas. The dependence of the influence of electric energy parameters on the synthesis of ozone in a discharge chamber was obtained on the basis of the conducted analysis and using computer simulation. The model of the ozone generation process in the barrier discharge has been further developed, which, in contrast to the existing one, takes into account not only the power supply parameters and camera dimensions, but also the processes occurring in the discharge gap. The method of calculation of the basic parameters of the ozone generator during its development, which allows increasing the efficiency of ozone synthesis, is proposed. The practical value of the results obtained is to justify the list of parameters that have the greatest impact on the performance of the medical ozone generator; engineering calculations of parameters of the generator of medical ozone, which allowed to optimize the design of the generator with the given parameters; the implementation of an experimental model of the medical ozone generator with an improved system of automatic regulation of gas consumption, which allowed to ensure the implementation of a wide range of procedures for ozone therapy. The practical results of the dissertation work were introduced at the Scientific-Production Enterprise "Econika" in the development and manufacture of modern generators of medical ozone, and also a prototype sample was developed, was tested at the medical center "Pulse Medika" and at the Institute of Medical Radiology N.A. Grigoriev.

The thesis is devoted to the development of a method for determining the parameters of a medical ozone generator to effectively increase its productivity by determining the parameters of its constituent parts, taking into account the influence of factors of different nature. On the basis of the study of the issues of constructing generators of medical ozone, the relevance of the study of ways to increase its efficiency was proved. A mathematical model based on the use of a single volume, which describes the processes occurring in the discharge chamber when the voltage is applied, is proposed. The level of influence of the parameters of the working gas and the geometry of the discharge chamber on the performance of the ozone generator, as well as on the stability of the parameters of the ozone-oxygen mixture, which were set. A method has been developed for calculating the parameters of a discharge chamber of a medical ozone generator, which makes it possible to obtain the most efficient design for given operating modes. A study has been carried out on the development of an automated working gas consumption system with improved characteristics, which makes it possible to carry out the main methods of ozone therapy. The efficiency of this system has been proved on the basis of the work of a medical ozone generator layout during the passage of medical approbation.

Повний текст джерелаColorectal cancer (CRC) is one of the most common cancer globally but above all the second leading cause ofdeath for oncological reasons. Despite medical research advances in anti-cancer treatments, many side effectspersist in patients as well as development of resistances to conventional treatments. The development of new anticancertherapeutic strategies is necessary in order to improve care of patients. Photodynamic therapy (PDT) usingphotosensitizers (PS) comes as an innovative therapeutic strategy severely restricting these undesirable sideeffects. PDT has been approved for treatment of some cancers due to the generation of cytotoxic reactive oxygenspecies only with photoactivated PS. However, low physiological solubility and lack of selectivity towards tumorsites are the main limitations of their clinical use. Indeed, targeted drug delivery is a crucial point in cancer therapy.Nanomedicine through the use of nanoparticles improves tumor-targeting because they are able to spontaneouslyaccumulate in solid tumors through an enhanced permeability and retention effect. The purpose of this study wasto prove added value of 5-(4-hydroxyphenyl)-10,15,20-triphenylporphyrin-xylan (TPPOH-X) vectorization bysilica nanoparticles (SNPs) in order to enhance anti-cancer efficacy through better tumor-targeting. It has beendemonstrated significant anti-cancer efficacy increase of TPPOH-X SNPs-PDT thanks to cellular uptakeimprovement relative to free TPPOH-PDT in 3 human CRC cell lines. Moreover, it has been characterized thatcell death induced by TPPOH-X SNPs-PDT is conducted via apoptosis and autophagy acts as a resistance pathwayto cell death. Furthermore, in vivo and without toxicity, TPPOH-X SNPs-PDT induce an elevated anti-cancerefficacy through improvement of tumor-targeting compared to free TPPOH-PDT. This study therefore highlightedthe added value of PDT and nanomedicine combination in order to improve future cancer treatments

Повний текст джерелаSleep regulations and mood disorders exert strong functional interactions, representing a major public health challenge. Light greatly influences physiology and behavior, including regulation of locomotion,sleep and mood. These non-visual effects are either, indirect through the phase shifting of circadian rhythms or direct. Our goal was to characterize the direct, non-circadian effects of light on behaviorand to assess the contribution of the different photoreceptors involved. To achieve this, melanopsin knockout mice were exposed to different luminance (<10 lux, 150 lux, 600 lux) without changing the phase of circadian rhythm. Our results indicate that light influences mood, locomotion and sleep, with< 10 lux leading to mood and sleep homeostasis alteration. These effects are primarily mediated by melanopsin-based phototransduction. If confirmed in humans, our observations will have applications for the clinical use of light as well as for societal lighting conditions

Повний текст джерелаLight influences physiology and behavior through both types of non-image-forming effects: i) indirect, synchronizing the circadian master clock located in the suprachiasmatic nucleus (SCN), ii) direct effects, independent from the circadian process though mechanisms poorly understood. Our studies in mice demonstrate that the direct influence of light constitutes a key mechanism of regulation for sleep, alertness and mood and is as important as the circadian process. The direct effects of light are mainly mediated through melanopsin, a retinal photopigment that projects to the different structures of the brain such as the SCN and the VLPO. The SCN, beyond their role as circadian clock are also a relay system for the direct effects of light. Further, we validated Arvicanthis ansorgei as a diurnal model for the study of sleep regulatory mechanisms. This is an important step in the translational approach from animal research to applications in humans. Various data suggest that the direct effects of light interact with the dopaminergic system. In the last part of this thesis, we evaluated the indication of bright light therapy in dopaminergic pathologies (Parkinson disease, restless legs syndrome, mood disorders). These advances open up new perspectives for possible applications of light therapy and may help improving societal lightening conditions

Повний текст джерелаThe impact of cold exposure in the context of physical exercise and recovery on physiological and perceptual variables was investigated.We have highlighted, by meta-analysis, that:- Exposure to cold during physical activity improves aerobic and anaerobic physical performance (effect sizes depending on the cooling technique and the cooled body area).- Using cryotherapy / cryostimulation and cold-water immersion following a physical activity reduces muscle soreness, perceived fatigue and blood concentrations of muscle damage and inflammatory markers.Our experimental work shows that:- Wearing a cooling vest during a vigorous physical exercise improves perceptual responses such as thermal comfort and reduces physiological stress (reduced cutaneous temperature, faster return to resting heart rate, improvement of the cerebral oxygenation).- A 3-min partial body cryotherapy exposure is an optimal time to improve the quality of sleep after training.- A 3-min whole body cryotherapy exposure improves the subjective (questionnaires) and objective (actimetry) sleep quality. The mechanisms related to this improvement seems to depend on the parasympathetic activity during deep sleep episodes.This work shows the effectiveness of the cooling uses in different contexts of physical activity and recovery processes. The biological mechanisms likely to interact in these recovery processes are presented

Повний текст джерелаDesde a era dos descobrimentos até aos dias de hoje que Portugal possui uma forte ligação com o mar, explorando-o nas mais diversas vertentes, entre as quais a económica e a de lazer. No entanto, relativamente à composição química, estas águas têm também características de elevada importância que as tornam ricas para fins terapêuticos e preventivos para o organismo, como é o caso da talassoterapia. A Talassoterapia consiste no uso medicinal da água do mar e do seu ambiente marinho como fonte de terapia. Deste modo, recorre à utilização da água do mar e dos seus produtos, sob supervisão médica, a uma temperatura aproximada de 38 °C. Comumente é usada como uma estratégia terapêutica em diversos países, inclusive Portugal, para a prevenção e tratamento de múltiplas patologias. Assim, o objetivo é estudar quais os efeitos tóxicos, genotóxicos, anti genotóxicos e de longevidade que os produtos thalion (TH), lamas marinhas (LM) e água do mar (AM) possam promover. Os efeitos tóxicos são determinados a partir do número de adultos eclodidos, pela sua longevidade e pela prolificidade celular das células da linha MG 63. A genotoxicidade dos produtos é determinada pelos danos no DNA nas células neuroblásticas de Drosophila melanogaster, nas células da linha celular MG 63 e pelas lesões nos omatídios de D. melanogaster, recorrendo ao teste do ensaio do “cometa” e ao teste de recombinação e mutação somática (SMART). Neste contexto foram testados produtos utilizados atualmente em diversos SPA’s para fins estéticos (regeneradores, tonificantes e hidratantes). Através da análise dos resultados dos diferentes ensaios realizados, verificou-se que os produtos TH, LM e AM testados não têm qualquer efeito genotóxico, averiguando-se também que o produto TH possui um efeito anti mutagénico. Nestes mesmos ensaios constatou-se também o aumento da prolificidade em D. melanogaster quando comparado com o grupo controlo. Estes dados sugerem que estes produtos possuem características peculiares, quando usados isolados ou combinados, que poderão ter grande importância na saúde para o tratamento de diversas patologias, na área da dermatologia e oncologia.
Since the age of discovery to the present day that Portugal has a strong connection to the sea, exploring it in several aspects, including the economic and leisure. However, regarding the chemical composition, these waters have also highly important characteristics that make them rich for therapeutic and preventive purposes to the body, as is the case of thalassotherapy. The SPA is the medical use of seawater and its marine environment as a source of therapy. Thus, makes use of seawater and their products under medical supervision at a temperature of about 38 ° C. Commonly is used as a therapeutic strategy in many countries, including Portugal, for the prevention and treatment of many pathologies. The objective is to study what a toxic, genotoxic, anti-genotoxic and longevity that Thalion products (TH), marine mud (LM) and seawater (AM) can promote. Toxic effects are determined from the number of adults hatched, the longevity and cellular prolificacy MG cell line 63. The genotoxicity of products is determined by DNA damage in Drosophila melanogaster neuroblastic cells, cells in the cell line MG 63 and causes for the omatídios D. melanogaster, using the test rehearsal "comet" and recombination and somatic mutation test (SMART). In this context were tested products currently used in many spas for aesthetic purposes (regenerative, toning and moisturizing). By analyzing the results of the various tests conducted, it was found that the product TH, LM and MA tested have no genotoxic effect, it is also ascertaining that the HT product has an anti-mutagenic effect. In these same studies have also observed increased prolificacy in D. melanogaster compared with the control group. These data suggest that these products have peculiar characteristics, when used alone or in combination, that may be of great importance in health for the treatment of various diseases in the field of dermatology and oncology.

Повний текст джерелаO turismo de saúde é uma terapêutica de bem-estar físico e mental, que complementa o descanso e o lazer com terapêuticas adequadas, como a talassoterapia, que visam ampliar os efeitos do bem-estar induzido pelo turismo. A talassoterapia constitui uma estratégia terapêutica que combina o recurso a banhos com Água do Mar (balneoterapia), um clima marinho e a exposição moderada à radiação solar (climoterapia), com tratamentos com lamas e soluções à base de algas (algoterapia). De facto, trata-se de uma estratégia terapêutica comummente utilizada em inúmeros países, Portugal inclusive, não só para a prevenção, como também para o tratamento de várias doenças. As propriedades biofísicas da Água do Mar, as propriedades mineralógicas e as características microbianas são fatores que contribuem para o tratamento e para a melhoria de diversas doenças. De resto, para além dos benefícios do ponto de vista medicinal, o recurso à talassoterapia apresenta, também, resultados a nível estético, nomeadamente a melhoria da permeabilidade e das funções da barreira epitelial. Tais terapias poderão, assim, ter uma função antienvelhecimento, bem como um importante papel na longevidade celular epitelial. Nesse sentido, os efeitos tóxicos e genotóxicos foram testados, recorrendo ao modelo de estudo Drosophila melanogaster, a técnicas de biologia molecular, como Somatic Mutation and Recombination Test (SMART), ao Single Cell Gel Electrophoresis assay (SCGE), mais conhecido como ensaio do cometa, e a testes com a linha celular de osteossarcoma MG-63. Na análise dos resultados, verificou-se a ausência de qualquer consequência prejudicial tóxica e genotóxica dos compostos testados. Os resultados mostraram, um aumento da longevidade das D. melanogaster, um aumento da prolificidade celular, como também, uma capacidade reparadora de danos dos produtos “3 Algas”, “Thalion” e “Banho Aromático”. Com efeito, esses estudos abrem novas portas para a criação de novos produtos com um suporte experimental científico e com uma maior eficiência antienvelhecimento e reparação de mutação, assegurando um potencial preventivo em doenças de pele como o cancro.
Health tourism is a therapeutic physical and mental well-being, which contemplates the rest and recreation with appropriate therapies such as thalassotherapy, which increase the effects of welfare-induced tourism. The Thalassotherapy is a therapeutic strategy that combines the use of baths with sea water (Balneotherapy), a marine climate, moderate exposure to sunlight (Climotherapy), treatments with sludge and algae-based solutions (Algotherapy). In fact, it is a therapeutic strategy commonly used in many countries, including Portugal, not only for the prevention and also for the treatment of various diseases. The biophysical properties of sea water, the mineralogical properties and microbial factors contributing to the treatment and amelioration of various diseases. Moreover, beyond the benefits of medicinal point of view, the use of Thalassotherapy presents results on an aesthetic level, including improving permeability and function of the epithelial barrier. These therapies can thus have an anti-aging function as well as a role in epithelial cell longevity. In this sense, the genotoxic effects were tested, using the study of Drosophila melanogaster model, molecular biology techniques, such as Somatic Mutation and Recombination Test (SMART) and the Single Cell Gel Electrophoresis assay (SCGE), better known as comet assay and tests with the osteosarcoma cell line MG-63. In analyzing the results, we verified the absence of any genotoxic detrimental effect of the tested compounds. The results also demonstrated reparative ability to damage some products tested. Indeed, these studies open new doors for the creation of new products with a scientific experimental support and greater efficiency and anti-aging repair mutation, ensuring preventive potential in skin diseases such as cancer.

Повний текст джерелаO turismo de saúde e bem-estar é um novo segmento do turismo, que está a ganhar cada vez maior expressão tanto à escala mundial como nacional. A presente investigação pretende analisar a satisfação do cliente da Villa Termal das Caldas de Monchique SPA Resort e a forma como ela é influenciada pela qualidade percebida e pelas emoções. De seguida analisa-se as consequências da satisfação, designadamente a fidelização e o compromisso. Foi realizado um inquérito adaptado do modelo elaborado e validado por Antunes (2004), a 134 clientes da Villa Termal, entre os 18 e os 96 anos de idade. Sendo que, 64,9% eram do género feminino, 51,5% eram clientes de Spa (termalismo de saúde e bem-estar) e 48% eram clientes do temalismo clássico. A análise dos dados recolhidos foi feita atrvés do programa informático SPSS, versão 17. Os resultados obtidos mostraram que existem uma correlação muito forte entre as variáveis “satisfação” e “qualidade percebida”, sendo este o factor com maior influência na satisfação do cliente da Villa Termal de Monchique. Constatou-se que a dimensão afectiva mais correlacionada com a satisfação é o entusiasmo com r = 0,536, e que as emoções negativas apresentam uma correlação negativa com a satisfação.
