The excited states of four hemes, c-552, c-554, c-556, and c-559, in a c-type cytochrome subunit of the photosynthetic reaction center (PSRC) of Rhodopseudomonas (Rps.) viridis were studied using the symmetry-adapted cluster (SAC)-configuration interaction (CI) method. c-552 has two imidazole ligands, while the other three hemes have imidazole and methionine ligands. The electronic states of these four hemes are affected significantly by both the ligands and protein and water environments. The ligand effect classifies the four hemes into two groups, while the environmental effect is unique in each heme. The HOMO–LUMO energy separation of c-552 is much smaller than those of the other hemes, and therefore, the low-lying excited states of c-552 are more stable than those of the other hemes. The low-lying excited states and their oscillator strengths of c-559 are significantly modified by the environment when compared with c-556 and c-554. Some observed UV peaks of the PSRC of Rps. viridis were found to have been assigned reasonably to the excited states of these four hemes in a previous report. The calculated linear dichroism angles also supported our assignment.

Objective: The assessment of energy effectiveness of traction drives for carriages of rolling stock models 81-765/766/767 and 81-556/557/558 (“NEVA”) from various producers and drafting technical requirements for drive electric equipment. Stock 81-765/766/767 uses asynchronous engine of 170 kW wattage and 43 Hz field rated frequency, 81-556/557/558 — 167 kW with 70 Hz rated frequency. Methods: The comparative analysis of drive energy indicators of trains with asynchronous electric drives has been accomplished in accordance with the calculation methodology. Measuring section was set in accordance with GOST P 50850-96. There are defined the assumptions at motion of rolling stock with equal maximal acceleration in a runaway and maximal deceleration in a braking and constant deceleration in a run-out. Maximal acceleration and deceleration were calculated incompliance adhesion set coefficient, grip weight (mass of motor cars) and moment of inertia. The calculation by method of analytical integration of motion differential equation was held on runaway five intervals. Initial technical data of trains with rated load as well as calculated indicators at train motion on measuring section were accepted. Diagrams of train motion on section were built and comparative analysis of characteristics of stock carriage models 81-765/766/767 and 81-556/557/558 (“NEVA”) was pursued. Results: It was established during the comparison that engine overload by torque on shaft is the same with 1.5 coefficient to rated torque. Stocks have significantly different indicators as by unit expenses as well as by motion dynamics. It’s been also established that for to minimize energy costs for traction it’s required to develop maximally allowable deceleration at braking and maximally allowable acceleration at runaway. Practical importance: The necessity of entry into Technical Task for new rolling stock of the requirement on stock check on runaway possibility with maximal set acceleration.

Анотація:
Our study assessed miR-556-3p’s role in breast cancer cells. A total of 65 cases of breast cancer tissue samples were retrospectively analyzed to detect miR-556-3p level by PCR and analyze survival time and 30 normal breast tissues were included as a control group. Breast cancer cells were cultured followed by analysis of cell proliferation by MTT, cell invasion by transwell assay. miR-556-3p level was significantly upregulated in breast cancer patients compared to control group (P <0.05) and inversely associated with survival rate (P <0.05). In vitro experiments, cell activity and invasion were positively correlated with miR-556-3p level (P <0.05). In MCF-7 cell lines, miR-556-3p overexpression increased cell activity (P <0.05). Meanwhile, after miR-556-3p was overexpressed, the expression of DAB2IP, Erk, p-Erk in breast cancer cells was significantly reduced and increased after miR-556-3p was knocked down. In conclusion, miR-556-3p targets DAB2IP3′-UTR, promotes breast cancer cell proliferation, indicating that miR-556-3p might be involved in breast cancer pathogenesis and may be a new target for the treatment.

ABSTRACT N-(4-Chlorophenyl)-N′-(2,2,6,6-tetramethyl-piperidin-4-yl)-oxalamide (NBD-556) is a low-molecular-weight compound that reportedly blocks the interaction between human immunodeficiency virus type 1 (HIV-1) gp120 and its receptor CD4. We investigated whether the enhancement of binding of anti-gp120 monoclonal antibodies (MAbs) toward envelope (Env) protein with NBD-556 are similar to those of soluble CD4 (sCD4) by comparing the binding profiles of the individual MAbs to Env-expressing cell surfaces. In flow cytometric analyses, the binding profiles of anti-CD4-induced epitope (CD4i) MAbs toward NBD-556-pretreated Env-expressing cell surfaces were similar to the binding profiles toward sCD4-pretreated cell surfaces. To investigate the binding position of NBD-556 on gp120, we induced HIV-1 variants that were resistant to NBD-556 and sCD4 in vitro. At passage 21 in the presence of 50 μM NBD-556, two amino acid substitutions (S375N in C3 and A433T in C4) were identified. On the other hand, in the selection with sCD4, seven mutations (E211G, P212L, V255E, N280K, S375N, G380R, and G431E) appeared during the passages. The profiles of the mutations after the selections with NBD-556 and sCD4 were very similar in their three-dimensional positions. Moreover, combinations of NBD-556 with anti-gp120 MAbs showed highly synergistic interactions against HIV-1. We further found that after enhancing the neutralizing activity by adding NBD-556, the contemporaneous virus became highly sensitive to antibodies in the patient's plasma. These findings suggest that small compounds such as NBDs may enhance the neutralizing activities of CD4i and anti-V3 antibodies in vivo.

RATIONALE: Population and health services research can be performed by linkage analysis of administrative data. However, the robustness of study results is determined by the accuracy of the diagnostic coding.OBJECTIVES: To estimate the awareness, use and accuracy of theInternational Classification of Diseases, Ninth Revision(ICD-9) coding by physicians providing services for patients with Crohn’s disease (CD) and ulcerative colitis (UC).METHODS: All Ontario gastroenterologists and a 10% random sample of internists, pediatricians, pediatric or general surgeons, and family physicians were surveyed by postal questionnaire to estimate the frequency and 95% CI of using codes 555 or 556 when billing for CD- and UC-related services, respectively.Χ2tests were used for between-group comparisons.RESULTS: Of the physicians who were surveyed, 67.7% (416 of 614) responded; 258 of 391 (66%) who were still practising in Ontario saw patients with inflammatory bowel disease (IBD), and 54% had more than 10 IBD patients; 86.5% (95% CI 82.4% to 90.6%) were familiar with ICD-9 codes, and 91.4% (95% CI 88.1% to 95.6%) used the codes 555 (CD) or 556 (UC) for billing. Rates of ICD-9 use did not differ by sex but were used more frequently by those graduating after 1981 (P<0.02). Gastroenterologists used ICD-9 IBD codes 555 or 556 significantly more often than all other physicians (P=0.001). Most (more than 75%) Ontario physicians used ICD-9 IBD codes always or frequently when billing for IBD-related services. Few (10%) used these codes to bill for non-IBD-related problems.CONCLUSIONS: These data suggest that there is acceptable use and accuracy of ICD-9 diagnostic coding for CD and UC services – comparable with results from studies of other diseases. Administrative data may thus be used to undertake epidemiological studies in IBD in Ontario.

10542 Background: The mutations of KIT exon 11 are those most frequently observed in GIST, but there is a substantial level of heterogeneity in their nature and topography. We investigated the impact of the type of KIT exon 11 mutations on clinical presentation and outcome under imatinib in advanced GIST patients (pts) included in the BFR14 study. Methods: Among the 434 pts included, 167 were identified as having GIST with KIT exon 11 mutation (85% of all KIT mutations and 73% of all mutations available in the entire cohort). We analyzed the topography of the mutations and identified 3 groups (G) of mutations according to the first codon involved in the genetic alteration in which the precise topography was available (from codon 549 to 579). The distribution of these G in the different clinical presentations was investigated, as well as their predictive value for progression-free (PFS) and overall survival (OS). Results: Three G with distinct sites of altered codons were distinguished: G1 whose 1st mutated codon was <556 (n=41, 24.5%), G2 whose 1st mutated codon was 557 and 558 (n=74, 44.3%), including those with the deletion 557-558 (n=37, 22.1%), and G3 whose 1stmutated codon was 559 and beyond (n=52, 31.1%). All investigated parameters at inclusion, gender, tumor site and size, site of metastases, age, PS, CD34 expression, biological characteristics were well balanced between the three G of patients. Pts of G2 reached a statistically difference in terms of complete response under imatinib (p=0.028). In contrast, this G have the poorer PFS compared to the other two groups: the median PFS were 30.6 months (CI95%: 21.5;39.4), 49.4 months (CI95%: 33.5; 109.9) and 63.3 months (CI95%: 27.6; 84.7) for the G 2, 1 and 3 respectively (p=0.0176). OS was not significantly different in the 3 G of pts. Conclusions: Despite a favorable sensitivity to imatinib, GIST pts harbouring a genetic alteration initially involving the 557 and 558 codons develop statistically more secondary resistance than the others. These results deserve confirmation in other prospective series in advanced GIST and may be worth exploring in adjuvant series.
