The impact of proton pump inhibitors (PPIs) on clinical outcomes with first-line immune checkpoint inhibitors (ICIs) in patients with metastatic melanoma was previously analyzed in the phase II study, CheckMate 069. This retrospective analysis utilized data from three phase II/III studies of first-line ICI therapy in untreated advanced melanoma: CheckMate 066, 067, and 069. All randomized patients with PPI use ≤ 30 days before initiating study treatment were included in the PPI-use subgroup. Possible associations between baseline PPI use and efficacy were evaluated within each treatment arm of each study using multivariable modeling. Approximately 20% of 1505 randomized patients across the studies reported baseline PPI use. The median follow-up was 52.6–58.5 months. Objective response rate (ORR), progression-free survival (PFS), and overall survival analyses provided insufficient evidence of a meaningful association between PPI use and efficacy outcomes with nivolumab-plus-ipilimumab, nivolumab, or ipilimumab therapy. In five of the six ICI treatment arms, 95% confidence intervals for odds ratios or hazard ratios traversed 1. Significant associations were observed in the CheckMate 069 combination arm between PPI use and poorer ORR and PFS. This multivariable analysis found insufficient evidence to support meaningful associations between PPI use and ICI efficacy in patients with advanced melanoma.

Abstract Quasiperiodic eruptions (QPEs) represent a novel class of extragalactic X-ray transients that are known to repeat at roughly regular intervals of a few hours to days. Their underlying physical mechanism is a topic of heated debate, with most models proposing that they originate either from instabilities within the inner accretion flow or from orbiting objects. At present, our knowledge of how QPEs evolve over an extended timescale of multiple years is limited, except for the unique QPE source GSN 069. In this study, we present results from strategically designed Swift observing programs spanning the past 3 yr, aimed at tracking eruptions from eRO-QPE1. Our main results are as follows: (1) the recurrence time of eruptions can vary from flare to flare and is in the range of 0.6–1.2 days; (2) there is no detectable secular trend in evolution of the recurrence times; (3) consistent with prior studies, their eruption profiles can have complex shapes; and (4) the peak flux of the eruptions has been declining over the past 3 yr, with the eruptions barely detected in the most recent Swift data set taken in 2023 June. This trend of weakening eruptions has been reported recently in GSN 069. However, because the background luminosity of eRO-QPE1 is below our detection limit, we cannot verify whether the weakening is correlated with the background luminosity (as is claimed to be the case for GSN 069). We discuss these findings within the context of various proposed QPE models.

e14595 Background: The PD-1 inhibitor Nivo and antiCTLA-4 Ipi in combination was registered by FDA and EMA for the treatment of metastatic melanoma.The accelerated approval of this indication was based on PFS data.OS data in previously untreated melanoma pts was reported only in the Phase II CheckMate 069 study.In the Nivo+Ipi group the 1YOS% was 73,4% and 63,8% at 2Y, in the Ipi group 1YOS% was 64,8%, 2YOS% was 53,6% at median follow up of 24,5 months.The median OS had not been reached, the outcome measures could not assess the long-term benefit of antitumor immun response. Methods: In order to measure the relative clinical values, we analyzed the 1Y, 2Y survival data and calculated the ARR (absolut risk reduction) and NNT values (the number of patients who need to be treated to prevent one death) of the Phase II CheckMate 069 trial Nivo+Ipi vs Ipi group, and the overall survival data in subgroup of pts who discontinued the immun-therapy due to treatment related AEs. We used GraphPad Software QuickCalcs/NNT for calculating the results of 95% CI. Results: In the CheckMate 069, the Nivo+Ipi group vs Ipi group ARR was 10,93, the NNT was 10(95%CI 3,6-17,8) at 1Y.The ARR was 13,19, the NNT value was 8(95%CI 3,3-24,1) at 2Y. 1 in every 8-10 pts will benefit from the Nivo+Ipi combined immuno-therapy in order to prevent one death. In the subgroup of patients (37%) who discontinued treatment, the ARR was 21,16 at 1Y and 21,76 at 2Y. Despite of discontinuation, 1 in every 5 pts will benefit from immuno-therapy at 1Y and 2Y, the NNT was 5 (95%CI 2,5-40,4) and 2Y (95%CI 2,3-127,2). Conclusions: These results of NNT 5-10 represent the exceptional survival benefit of combined immuno-therapy. The limitations of NNT bias the pts demographic parameters and the value depends on comparator drug. The risk reduction was not constant over the follow up time and the long term benefit of immuno-therapy is not linear.The NNT data provide measurement of relative clinical value and clinical impact.

ABSTRACT We report the Fermi Large Area Telescope (Fermi-LAT) detection of the γ-ray emission towards the young massive star cluster RSGC 1. Using the latest source catalogue and diffuse background models, we found that the diffuse γ-ray emission in this region can be resolved into three different components. The GeV γ-ray emission from the region HESS J1837-069 has a photon index of 1.83 ± 0.08. Combining with the HESS and MAGIC data, we argue that the γ-ray emission in this region likely originates from a pulsar wind nebula. The γ-ray emission from the north-west part (region A) can be modelled by an ellipse with the semimajor and semiminor axes of 0.5° and 0.25°, respectively. The GeV emission has a hard spectrum with a photon index of about −2 and partially coincides with the TeV source MAGIC J1835-069. The possible origin of the γ-ray emission in this region is the interaction of the cosmic rays (CRs) accelerated by SNR G24.7+0.6 or/and the OB cluster G25.18+0.26 with the surrounding gas clouds. The GeV γ-ray emission from the south-east region (region B) can be modelled as an ellipse with the semimajor and semiminor axes of 0.9° and 0.5°, respectively, and also reveals a hard γ-ray spectrum. We argue that the most probable origin is the interaction of the accelerated protons in the young massive star cluster RSGC 1 with ambient gas clouds, and the total CR proton energy is estimated to be as high as ${\sim}1\times 10^{50}\ \rm erg$.

Techniques applied to promote citrus propagation are of extreme relevance, since they assure high yield rates, as well as high genetic and phytosanitary quality. The aim of the present research is to assess the vegetative growth and survival of citrus cultivars subjected to different rootstocks through minigrafting in order to generate identical to the parents. Minigrafting of apical segments (1 and 2 cm long) of ‘Clementine’ tangerine budded onto rootstocks of citrandarins ‘Indio’ and ‘Riverside’ and of the combination between varieties ‘Pera’ sweet orange, ‘Sunki Madarin’ tangerine and ‘Santa Cruz Rangpur’ lemon budded onto rootstocks of citrandarins ‘Indio’, HTR-069 and LRF × (LCR × TR)-005 were evaluated. Assessments were conducted in greenhouse 120 days after the experiment were installed. The use of 2cm long segments facilitates minigrafting adherence to smaller caudal apices. The rootstock of HTR-069 presented the best survival responses among the assessed crowns. Rootstock of citrandarin ‘Indio’ enabled the best crown length development and graft diameter. Based on the results, minigrafting can be a new option for citrus propagation.

The structure of butorphanol hydrogen tartrate {(9R,13S,14S)-(-)-17-(cyclobutylmethyl)morphinan-3,14-diol (2S,3S)-(-)-hydrogen tartrate} (C21H29NO2 .C4H6O6) was solved by direct methods and refined anisotropically to the R value of 0.029 for 2 069 observed reflections. The title morphine analogue crystallizes in the triclinic space group P1 with lattice parameters a = 7.620(1), b = 9.140(1), c = 9.591(1) Å, α = 105.48(1), β = 112.91(1), γ = 84.29(1)°, Z = 1. The butorphanol B ring possesses the 3E envelope conformation with small 3H2 distortion, C and D rings have a regular chair conformation. The intramolecular N17-H17...O2 hydrogen bond is observed in crystal structure of the title compound. The butorphanol and hydrogen tartrate molecules are joined together by means of O2-H(O2)...O7, O1-H(O1)...O7' and O3-H(O3)...O8" hydrogen bonds to form networks.

RESUMO Objetivou-se estudar os mecanismos fisiológicos de percepção e tolerância ao estresse salino de híbridos de citros em cultivo hidropônico. O experimento foi realizado em ambiente protegido do Centro de Ciências e Tecnologia Agroalimentar - CCTA, da Universidade Federal de Campina Grande - UFCG, Pombal, PB. Foram avaliados, em blocos casualizados, em esquema fatorial (2x5), dois níveis de salinidade da água de irrigação (0,3 e 4,0 dS m-1) e cinco híbridos recomendados para uso como porta-enxerto [HTR - 069, HTR - 116, HTR - 127, LCR x TR - 001 e TSKC x (LCR x TR) - 040)], com três repetições. Os genótipos foram cultivados em sistema hidropônico adaptado em vasos de Leonard, utilizando solução de Hoagland. O início da aplicação da solução nutritiva com uso de águas salinas ocorreu aos 90 dias após a semeadura, quando se procedeu as avaliações quanto às trocas gasosas e fluorescência do PSII às 0, 24 e 48 horas após aplicação dos tratamentos a fim de determinar o efeito do estresse salino sobre os atributos fisiológicos. A fluorescência inicial, a condutância estomática e a eficiência no uso da água foram as variáveis que mais permitem evidenciar a percepção do estresse neste trabalho. O híbrido TSKC x (LCR x TR) - 040 foi o mais tolerante à salinidade, seguido do híbrido HTR - 069. O híbrido LCR x TR - 001 foi o mais sensível à salinidade, por ter sua atividade do PSII comprometida.

E-mail: walter.soares@embrapa.br 1 RESUMO Objetivou-se avaliar a morfofisiologia de genótipos de citros recomendados como porta-enxertos, em sistema hidropônico alternativo, a partir de vasos de Leonard a base de garrafa pets. O experimento foi desenvolvido em ambiente protegido, sendo estudados treze genótipos de citros [1 – Hibrido Trifoliado (HTR)-127; 2– Tangerineira ‘Sunki Comum’ (TSKC) x Citrange Argentino (CTARG)-019, 3– TSKC; 4 – Limoeiro Rugoso da Flórida (LRF); 5 - limoeiro ‘Cravo’ (LCR) x Poncirus trifoliata (TR) - 001; 6 – TSKC x Citrumelo Swingle (CTSW)-028; 7 - TSKC x CTSW-033; 8 - TSKC x CTSW-041; 9 - TSKC x (LCR x TR)-040; 10–HTR-116; 11 - limoeiro ‘Cravo Santa Cruz’ (LCRSTC); 12 - limoeiro ‘Volkameriano’ (LVK) e 13 - HTR–069], em blocos casualizados com quatro repetições e quatro plantas úteis por parcela. As plantas foram cultivadas em sistema hidropônico alternativo, a partir de um sistema de vasos de Leonard confeccionados com garrafas de polyethylene terephthalate (PET) e distribuição gravitacional da solução de Hogland® em fluxo contínuo, sendo as garrafas preenchidas com substrato comercial a base de pó de coco e as plantas cultivadas até os 75 dias após semeadura, quando se avaliou aspectos de crescimento e as trocas gasosas das plantas. O sistema hidropônico alternativo com uso de vasos de Leonard pode ser usado para o desenvolvimento morfofisiológico de mudas cítricas; Os genótipos TSKC x CTARG– 019, LRF, TSKC x (LCR x TR) – 040, LCRSTC, LVK e HTR – 069 possuem maiores potenciais fisiológicos e de crescimento vegetativo, sendo indicados para obtenção de mudas mais precoces. It were studied thirteen genotypes of citrus (1- HTR-127; 2- TSKC x CTARG-019; 3- TSKC; 4- LRF; 5- LCR x TR-001; 6 - TSKC x CTSW-028; 7- TSKC x CTSW–033; 8- TSKC x CTSW-041; 9- TSKC x (LCR x TR)- 040; 10- HTR-116; 11- LCRSTC; 12- LVK and 13- HTR-069), in a randomized block design, with four replications and four plants by parcel, crop on alternative hydroponically system using Leonard’s pots made with polyethylene terephthalate (PET), being the Hoagland’s solution distributed by gravitation in continuous rate, in west coconut, being plants cultivated for 75 days after sowing, when it was evaluated the growth and gas exchange aspects. The alternative hydroponic system with use of Leonard pots can be used for morphophysiological development of citrus seedlings; The genotypes TSKC x CTARG– 019, LRF, TSKC x (LCRxTR) – 040, LCRSTC, LVK e HTR – 069 have greater physiological and of vegetative growth potential, being suitable for obtaining early seedlings. The solar detachment rates of all stable atomic anions belonging to elements in the main body of the periodic table have been calculated. Multi-channel photodetachment effects owing to fine structure levels and temperature effects are included in the treatment. All told the solar detachment rates of 31 anions at 300 and 104 K are made available. The rates show strong correlation to the group number in the periodic table. The higher the group number of the element, the lower is its detachment rate. Caesium has the highest detachment rate of 82 s-1, while fluorine has the lowest rate of 0�069 S -1. The calculations shown here may be useful to researchers conducting experiments on ionospheric modification where negative ions which are not normally found in the atmosphere may appear in abundance.

AbstractThe Oxylog, a portable breath-by-breath gas analyser, was used on seven animals to determine standing metabolic rate, energy cost of walking on soils of different consistencies and efficiency of work ploughing and carting. The average standing metabolic rate of animals was 5·63 (s.e. 0·12) W/kg M00·75. The consistency of the soil on which animals worked had a marked effect on their energy cost of walking which was 1·59 (s.e. 0·069) on unploughed soil, 2·15 (s.e. 0·084) on ploughed soil and 1·0 (s.e. 0·10) J/m per kg live weight on laterite tracks. The efficiency of ploughing sandy soils (i.e. ratio of work done to energy used for work) was 0·32 and was not significantly different from the efficiency of carting with different loads. The efficiency of doing work was not influenced by the type of work performed, the draught force exerted or the walking speed.

Purpose. To analyse the process of heat exchange between the man and the environment, including production areas, as well as to develop and study the criterion of workplace comfort by the thermal factor. To achieve this goal, the following tasks are to be solved: - to consider the characteristics of the microclimate in the workplace, including that of office space using information technologies; - to develop a mathematical model of heat exchange between the human body and the environment; - to develop a criterion for the efficiency of functioning of microclimate normalization systems in the workplace. Methodology. To perform the tasks in the work, an analysis of the literature and sanitary and hygienic documentation on the specified problem is performed; analytical method is applied for studying the comfort of the sanitary and hygienic condition of the indoor air using a mathematical model of heat exchange between workers and the environment. Findings. Based on the theoretical analysis, the criterion of microclimate comfort is obtained, which differs from the known ones in that it takes into account all types of heat transfer from the human body to the environment depending on the nature and intensity of human activity, allows estimating the total value of heat flow depending on the environmental condition with fair accuracy for the practical purposes and can be used to substantiate the efficiency of methods and means of normalizing the microclimate when performing work using information technologies. Originality. A mathematical model of heat transfer in the man environment system is developed and theoretically substantiated. The criterion of microclimate comfort at workplaces is offered and mathematical relation to calculate it is obtained. Practical value. The results of calculations of the comfort criterion in the workplace can be used when developing tools and methods for normalizing the microclimate in the workplace and assessing the effectiveness of their implementation.
