Анотація:
Impaired NO-cGMP signaling has been linked to several neurological disorders. NO-sensitive guanylyl cyclase (NO-GC), of which two isoforms—NO-GC1 and NO-GC2—are known, represents a promising drug target to increase cGMP in the brain. Drug-like small molecules have been discovered that work synergistically with NO to stimulate NO-GC activity. However, the effects of NO-GC stimulators in the brain are not well understood. In the present study, we used Förster/fluorescence resonance energy transfer (FRET)-based real-time imaging of cGMP in acute brain slices and primary neurons of cGMP sensor mice to comparatively assess the activity of two structurally different NO-GC stimulators, IWP-051 and BAY 41-2272, in the cerebellum, striatum and hippocampus. BAY 41-2272 potentiated an elevation of cGMP induced by the NO donor DEA/NO in all tested brain regions. Interestingly, IWP-051 potentiated DEA/NO-induced cGMP increases in the cerebellum and striatum, but not in the hippocampal CA1 area or primary hippocampal neurons. The brain-region-selective activity of IWP-051 suggested that it might act in a NO-GC isoform-selective manner. Results of mRNA in situ hybridization indicated that the cerebellum and striatum express NO-GC1 and NO-GC2, while the hippocampal CA1 area expresses mainly NO-GC2. IWP-051-potentiated DEA/NO-induced cGMP signals in the striatum of NO-GC2 knockout mice but was ineffective in the striatum of NO-GC1 knockout mice. These results indicate that IWP-051 preferentially stimulates NO-GC1 signaling in brain slices. Interestingly, no evidence for an isoform-specific effect of IWP-051 was observed when the cGMP-forming activity of whole brain homogenates was measured. This apparent discrepancy suggests that the method and conditions of cGMP measurement can influence results with NO-GC stimulators. Nevertheless, it is clear that NO-GC stimulators enhance cGMP signaling in the brain and should be further developed for the treatment of neurological diseases.

A diversificação de uso de porta-enxertos de citros é importante no Brasil devido à presença de diversos estresses abióticos e bióticos, além da busca por atributos horticulturais desejáveis como nanismo, alta eficiência de produção e indução de boa qualidade aos frutos. Para a seleção de um genótipo com potencial de uso como porta-enxerto, porém, o desempenho na fase de propagação também é relevante. Assim, caracterizaram-se os frutos e avaliou-se a propagação em ambiente protegido de porta-enxertos híbridos de citros obtidos ou selecionados pelo Programa de Melhoramento Genético de Citros da Embrapa: citrandarins ‘Indio’, ‘Riverside’ e ‘San Diego’, híbridos HTR-051, TSKC x (LCR x TR)-040 e 059, LVK x LCR-010 e 038, TSKC x CTTR-002, TSKC x CTSW-041 e LCR x TR-001, além das variedades comerciais citrumelo ‘Swingle 4475’, trifoliata ‘Flying Dragon’, limoeiro ‘Cravo Santa Cruz’ e tangerineira ‘Sunki Tropical’. Foram avaliados variáveis biométricas, fisiológicas e coeficientes técnicos. O delineamento experimental foi em blocos ao acaso, com quatro repetições e 50 plantas na parcela ou 20 frutos por genótipo, conforme a avaliação. Citrumelo ‘Swingle’ e LVK x LCR-010 apresentaram alta produção de sementes por fruto, enquanto HTR-051 e LCR x TR-001 produziram as menores quantidades. A poliembrionia foi superior para TSKC x CTTR-002, TSKC x (LCR x TR)–040, LCR x TR-001, citrandarins ‘Indio’, ‘Riverside’ e ‘San Diego’ e tangerineira ‘Sunki Tropical’, tendo trifoliata ‘Flying Dragon’ e LVK x LCR-010 apenas 45% de poliembrionia média. A emergência de citrandarin ‘Riverside’ foi mais rápida e uniforme em relação aos demais genótipos. O crescimento vegetativo da parte aérea e do sistema radicular foi superior para citrandarin ‘Riverside’, TSKC x (LCR x TR)-059, tangerineira ‘Sunki Tropical’, citrumelo ‘Swingle’ e limoeiro ‘Cravo’ e seus híbridos. Todos os híbridos de citros avaliados apresentam potencial de uso como porta-enxertos.

RESUMO O objetivo deste trabalho foi avaliar a produção de mudas de laranjeiras doces ‘Pera’ e ‘Westin’, tangerineira-tangor ‘Piemonte’ e limeira-ácida ‘Tahiti’ enxertadas em 14 porta-enxertos de citros em viveiro protegido. As mudas de laranjeiras-doces ‘Pera D-6’ e ‘Westin’, tangerineira-tangor ‘Piemonte’ e limeira-ácida ‘Tahiti CNPMF-02’ foram avaliadas em viveiro protegido após a enxertia, em 11 porta-enxertos híbridos: citrandarins ‘Indio’, ‘Riverside’ e ‘San Diego’, citrumelo ‘Swingle 4475’, HTR-051, TSKC x (LCR x TR)-040 e 059, LVK x LCR-010 e 038, TSKC x CTTR-002 e TSKC x CTSW-041, além de trifoliata ‘Flying Dragon’, limoeiro ‘Cravo Santa Cruz’ e tangerineira ‘Sunki Tropical’. Coletaram-se variáveis biométricas e fisiológicas, sendo o delineamento experimental em blocos ao acaso, em parcelas subdivididas, com 56 tratamentos, três repetições e dez plantas na parcela. Independentemente do porta-enxerto, a limeira-ácida ‘Tahiti CNPMF-02’ foi a copa mais vigorosa em viveiro, seguida da tangerineira-tangor ‘Piemonte’ e, por fim, pelas laranjeiras ‘Pera-D6’ e ‘Westin’. A tangerineira ‘Sunki Tropical’ induziu maior crescimento vegetativo e de sistema radicular em combinação com todas as copas estudadas. O trifoliata ‘Flying Dragon’ e o híbrido HTR-051 necessitam de maior período para a formação das mudas em função do menor vigor desses genótipos, em combinação com todas as variedades copas avaliadas. Não se observaram quaisquer sintomas de incompatibilidade entre as variedades no viveiro.

Purpose. The study examines the relationship between children's television (TV) viewing and physical fitness. Design. Cross-sectional data from questionnaires and objective measures were analyzed. Setting. Data were collected during the fall of 1990 from public elementary school students in a suburban California city. Subjects. Approximately 98% of eligible students participated. Of these, 10% were dropped due to missing data, yielding a final sample of 284 girls and 304 boys. Measures. Children reported their amount of TV viewing on a typical summer day; parents reported their child's TV viewing on a typical weekday during the school year. Cardiovascular fitness was the 1-mile run/walk. Body fat was both the child's body mass index (BMI) and skinfolds. Additional measures included muscular strength/endurance and flexibility. Results. Mile run/walk times were associated with both parental (η2 = . 051 and . 031 for boys and girls, respectively) and child reports (η2 = . 020 and . 028) of the child's amount of TV viewing. Parental reports, but not child reports, of the child's TV viewing were related to BMI (η2 = .041 and .058) and skinfolds (η2 = .050 and .029). Neither measure of children's TV viewing was related to muscular strength/endurance or flexibility. Conclusions. Children's TV viewing seems to be weakly and inconsistently related to various components of physical fitness. However, given the tracking of cardiovascular disease risk factors from childhood into adulthood and the high proportion of children who watch television, these relationships are worthy of further study.

In these two adult cases of transient loss of consciousness (TLoC), careful history-taking revealed a stereotyped prodrome of déjà vu suggestive of focal seizures. One patient had an implantable cardiac monitor fitted, the other a 48 hour Holter monitor. Each patient subsequently had a typical event of déjà vu followed by TLoC with rapid recovery and for both, the monitor showed cardiac asystole corresponding with this event. A permanent pacemaker (PPM) was inserted in both patients. This abolished TLoC. In its absence, déjà vu became more frequent, intense, and prolonged for one patient, resolving with levetiracetam. The other patient had a post-PPM episode of déjà vu followed by a generalised tonic-clonic seizure. It transpired that their lamotrigine had been withdrawn following PPM insertion. Seizure-freedom was achieved by restarting lamotrigine.Ictal asystole, a rare complication of focal epilepsy, has a mean ventricular standstill duration of 20 s. This can be benign or progress to post-ictal asystole −fatal in 54% of cases. Careful attention should be paid to prodromal symptoms suggestive of focal seizures (including ‘auras’) in patients presenting with what might otherwise sound like syncope. These patients may have epilepsy presenting with ictal asystole and require antiepileptic drugs alongside their cardiac investigations/interventions.

ABSTRACT The E1B-55K protein plays an important role during human adenovirus type 5 productive infection. In the early phase of the viral infection, E1B-55K binds to and inactivates the tumor suppressor protein p53, allowing efficient replication of the virus. During the late phase of infection, E1B-55K is required for efficient nucleocytoplasmic transport and translation of late viral mRNAs, as well as for host cell shutoff. In an effort to separate the p53 binding and inactivation function and the late functions of the E1B-55K protein, we have generated 26 single-amino-acid mutations in the E1B-55K protein. These mutants were characterized for their ability to modulate the p53 level, interact with the E4orf6 protein, mediate viral late-gene expression, and support virus replication in human cancer cells. Of the 26 mutants, 24 can mediate p53 degradation as efficiently as the wild-type protein. Two mutants, R240A (ONYX-051) and H260A (ONYX-053), failed to degrade p53 in the infected cells. In vitro binding assays indicated that R240A and H260A bound p53 poorly compared to the wild-type protein. When interaction with another viral protein, E4orf6, was examined, H260A significantly lost its ability to bind E4orf6, while R240A was fully functional in this interaction. Another mutant, T255A, lost the ability to bind E4orf6, but unexpectedly, viral late-gene expression was not affected. This raised the possibility that the interaction between E1B-55K and E4orf6 was not required for efficient viral mRNA transport. Both R240A and H260A have retained, at least partially, the late functions of wild-type E1B-55K, as determined by the expression of viral late proteins, host cell shutoff, and lack of a cold-sensitive phenotype. Virus expressing R240A (ONYX-051) replicated very efficiently in human cancer cells, while virus expressing H260A (ONYX-053) was attenuated compared to wild-type virus dl309 but was more active than ONYX-015. The ability to separate the p53-inactivation activity and the late functions of E1B-55K raises the possibility of generating adenovirus variants that retain the tumor selectivity of ONYX-015 but can replicate more efficiently than ONYX-015 in a broad spectrum of cell types.
