This study aimed to analyze the effect of uchikomi movement exercise using kettler rubber to improve male judoka's physical abilities. This study uses an experimental method. A total of 22 male judoka participated as study samples and were randomly divided into two groups. The experimental group (21.8±1.78 years, 1.70±0.06 m, 71.1±13.5 kg) and control group (21.4±2.30 years, 1.72±0.05 m, 67.4±7.76 kg). The exercise program was carried out for 8 weeks with a frequency of 3 times/week. After warming up and before core judo training, the experimental group was treated with uchikomi movement training using kettler rubber. Meanwhile, the control group continued regular judo training. The statistical analysis procedure used an ANOVA test to determine the difference and comparison of pre-test and post-test mean values in experimental and control groups with a significance level (p<0.05). The results showed that parameter difference in the average value of experimental and control groups was significant on the performance of leg power and VO2Max endurance. In comparison, no significant differences occur in handgrip strength, flexibility, and speed. This study concludes that uchikomi seoi-nage movement exercise using a Kettler rubber produces changes in the performance of judoka physical abilities. However, a significant difference occurred in the performance of leg power and VO2Max endurance

The female and male keys to all 21 currently recognized Microtimyobia spp. are provided. The host distribution of Microtimyobia spp. is analyzed. These mites serve as peculiar “markers” of particular host groupings and in many cases are useful tools to test some controversial hypotheses of cricetid phylogeny, including the intriguing problem of phylogenetic relationships of the Palearctic and Nearctic voles of the genus Microtus.

Abstract Tian, Y., Uchikawa, K., Ueda, Y., and Cheng, J. Comparison of fluctuations in fish communities and trophic structures of ecosystems from three currents around Japan: synchronies and differences. – ICES Journal of Marine Science, 71: . Features of three marine ecosystems affected by the Tsushima (TWC), Kuroshio (KC), and Oyashio (OC) currents were analysed based on fishery, oceanographic, and climate datasets during 1955–2010. Principal component (PC) analysis for catches of 25 indicator species showed evident decadal variation patterns with a step change in the first principal component (PC1) in the late 1980s, indicating synchronies in the dominant variation mode across the ecosystems. Step changes were also detected in the mid-1970s in PC2 and PC3 in OC, and around 1970 in PCs in KC and TWC. These indicate that the most marked change across the ecosystems occurred in the late 1980s, corresponding to the late 1980s climate regime shift, but OC also responded strongly to the mid-1970s regime shift, indicating different responses to regime shifts. The generalized additive model showed the PCs associated largely with water temperature in each region as well as climate indices, indicating the importance of regional oceanographic conditions. Ecosystem indicators such as the mean trophic level showed similarities between TWC and KC but differences with OC, indicating that trophic structures in TWC and KC were largely dependent on the mid-trophic (small pelagic species) level, while on demersal species in OC.

Resumo:
Aurihydrargyrumite, a natural Au6Hg5 phase, was found in Iyoki, Uchiko, Ehime Prefecture, Shikoku Island, Japan. Aurihydrargyrumite with a metallic silver luster occurs as a submicron- to 2 μm-thick layer on the outermost surface of the placer gold. A prismatic face may be formed by {001} and {100} or {110}. The streak is also silver white and its Mohs hardness value is ca. 2.5. Its tenacity is ductile and malleable, and its density, as calculated based on the empirical formula and powder unit-cell data, is 16.86 g·cm−3. The empirical formula of aurihydrargyrumite, on the basis of 11 Au + Hg, is Au5.95Hg5.05. Aurihydrargyrumite is hexagonal, P63/mcm, with the lattice parameters a = 6.9960(10) Å, c = 10.154(2) Å and V = 430.40(15) Å3, which is identical with the synthetic Au6Hg5 phase. The seven strongest lines in the powder X-ray diffraction (XRD) pattern [d in Å(I/I0)(hkl)] were 2.877(29)(112), 2.434(42)(113), 2.337(100)(104), 2.234(87)(211), 1.401(39)(314), 1.301(41)(404), and 1.225(65)(217). Aurihydrargyrumite forms through the weathering of mercury-bearing placer gold by involvement of self-electrorefining. This new mineral has been approved by the IMA-CNMNC (2017-003) and it is named for its composition, being a natural amalgam of gold (Latin: aurum) and mercury (Latin: hydrargyrum).

Resumo:
Abstract Background: CD137 (4-1BB) is a co-stimulatory molecule which exerts activation signals in multiple immune cell subsets. However, agonist antibodies targeting CD137 have not been clinically successful due to severe systemic toxicity. Therefore, we generated STA551, a tumor selective CD137 agonist antibody. STA551 binds to CD137 only in the presence of ATP, which is known to be present at high levels in tumor tissue, and activates immune cells such as T cells. Previous studies have shown that STA551 exhibits antitumor efficacy without inducing a systemic immune response in a murine model. Combination studies with anti-PD-L1 antibody have also shown synergistic efficacy without inducing a systemic immune response. However, it was unclear why they exhibit strong combination effects. In this study, we elucidated the mechanism of STA551 and anti-PD-L1 antibody efficacy through comprehensive analysis of gene expression using single cell RNA sequencing. Method & Result: In a colon 38-bearing human CD137 knock-in mouse model, combination therapy with STA551 and anti-PD-L1 antibody showed potent anti-tumor efficacy. From immunohistochemistry of tumor tissue, increased numbers of CD8+ T cells and elevated expression of PD-L1 molecules were observed. To perform a detailed mechanistic analysis, tumor tissue was collected after antibody administration and CD3 positive and CD3 negative fractions were separated and gene expression was comprehensively analyzed using single cell RNA-seq. Next, the fraction expressing Cd8a gene was extracted from CD3 positive fraction and clustered. One cluster was found to be markedly increased in STA551 and anti-PD-L1 antibody combination compared to vehicle or monotherapy treatment. Differential gene expression analysis revealed that the cluster was enriched for genes related to cytokines and chemokines. This result revealed that the combination of STA551 and anti-PD-L1 antibody induced highly activated CD8+T cells. Conclusion: The combination of STA551 and PD-L1 showed synergistic anti-tumor efficacy in a colon38-bearing human CD137 knock-in mouse model. Detailed mechanistic analysis showed that the combination of STA551 and PD-L1 induced highly activated CD8+T cells expressing chemokines and cytokines compared to monotherapy. This cluster also express chemokines, suggesting that other immune cells have been recruited. These results suggest that even the activation of only tumor tissue, which is a characteristic of STA551, can induce a strong immune response. Abstract Background: Claudin-6 (CLDN6), one of over 20 known human CLDN family members, is upregulated in many solid tumors but shows minimal or no expression in healthy adult tissues. This makes it an ideal target for cancer therapy. Although several therapeutic modalities targeting CLDN6 are currently under investigation—including T cell engagers (TCE), CAR-T, and antibody drug conjugates—a more potent therapeutic option is still desirable. TCEs are a promising avenue within cancer immunotherapy, but their current efficacy and safety must be improved, particularly against solid tumors. Methods and Results: To enhance immune activation and achieve successful clinical outcomes against solid tumors expressing CLDN6, we generated SAIL66, a CLDN6-targeting next generation TCE, using a new technological format called Dual-Ig®. SAIL66 is a tri-specific monoclonal antibody designed to bind to CLDN6 on cancer cells with one Fab arm and to both CD3 and CD137 with the other Fab arm. The preclinical characterization of SAIL66 was performed in a series of in vitro and in vivo studies which included comparisons to a conventional TCE targeting CLDN6 and CD3. SAIL66 demonstrated high specificity for CLDN6 despite its similarity to CLDN3, CLDN4, and CLDN9, suggesting a lack of off-target toxicity in patients. Using a Jurkat cell system harboring NFAT or NF-κB reporter genes, we confirmed that SAIL66 exerts CD3 and CD137 signal induction activity depending on CLDN6 expression. SAIL66 induced activation of T cells, release of cytokines, and lysis of CLDN6-positive cancer cells. In vivo studies with a humanized mouse (huNOG) model showed that SAIL66 had better anti-tumor efficacy in eliminating CLDN6-positive cancer cells than a conventional TCE. Flow cytometry analysis in the huNOG model revealed that SAIL66 also more significantly increased CD3+ T cell infiltration of tumor tissue and resulted in a smaller percentage of exhausted T cells in tumor. Moreover, transcriptome analysis in the huNOG model showed that both SAIL66 and the conventional TCE increase expression of genes associated with immune activation, with a durable increase observed especially with SAIL66. Further, we clarified that T cells repeatedly stimulated with SAIL66 had a greater ability to inhibit the growth of CLDN6-positive cancer cells than T cells stimulated with the conventional TCE in vitro. Conclusion and Discussion: Our study shows that SAIL66 has better anti-tumor efficacy against CLDN6-positive cancer cells than a conventional TCE. This is due to effective immune activation by SAIL66 which triggers CD3 signaling (signal 1) combined with CD137 co-stimulatory signaling (signal 2) in T cells. Thus, SAIL66 represents a promising therapeutic option for solid tumors expressing CLDN6. A clinical trial for CLDN6-positive cancer patient is now ongoing (NCT05735366). Performance is judged using a variety of methods to ensure uniformity between competitions. Uchikomi Fitness Test (UFT) could accomplished between morning qualifying and evening finals. The purpose of this study is to investigate the impact of different warm-up protocols on UFT at different times of the day in female judokas. Ten volunteer women who had been practising judo on a regular basis for more than 5 years and actively competed in international tournaments took part in this study. Judokas completed UFT, either after no-warm-up (NWU), specific warm-up (SWU), and linear+lateral warm-up (FWU) protocols for two times a day in the morning: 09:00–11:00 and in the evening: 16:00–18:00, on non-consecutive days. In conclusion, there was a significant increase in UFT scores (F = 9.89; p = 0.002), a + b (F = 4.42; p = 0.04) and heart rate (F = 28.99; p < 0.001) in the early evening compared to the morning. Increases in UFT performance were observed in the SWU protocol compared to the NWU and FWU protocols (p < 0.05). However, the interaction between time of day and warm protocol was not significant (p > 0.05). The UFT performance revealed diurnal variation, and the judokas’ performances may be favourably affected more in the late hours, particularly following SWU procedures.

Abstract Background Epidemiological evidence for the relationship between education and income and carotid intima-media thickness (CIMT) has been limited and inconsistent. The present cross-sectional study investigated this issue using baseline data from the Aidai Cohort Study. Methods Study subjects were 2012 Japanese men and women aged 34−88 years. Right and left CIMT were measured at the common carotid artery using an automated carotid ultrasonography device. Maximum CIMT was defined as the largest CIMT value in either the left or right common carotid artery. Carotid wall thickening was defined as a maximum CIMT value > 1.0 mm. Results The prevalence of carotid wall thickening was 13.0%. In participants under 60 years of age (n = 703) and in those aged 60 to 69 years (n = 837), neither education nor household income was associated with carotid wall thickening or with maximum CIMT. Among those aged 70 years or older (n = 472), however, higher educational level, but not household income, was independently related to a lower prevalence of carotid wall thickening: the multivariate-adjusted odds ratio for high vs. low educational level was 0.43 (95% confidence interval 0.21−0.83, p for trend = 0.01). A significant inverse association was observed between education, but not household income, and maximum CIMT (p for trend = 0.006). Conclusions Higher educational level may be associated with a lower prevalence of carotid wall thickening and a decrease in maximum CIMT only in participants aged 70 years or older.
