Usnea lichens are known for their beneficial pharmacological effects with potential applications in oral medicine. This study aims to investigate the extract of Usnea barbata (L.) Weber ex F.H. Wigg from the Călimani Mountains in canola oil as an oral pharmaceutical formulation. In the present work, bioadhesive oral films (F-UBO) with U. barbata extract in canola oil (UBO) were formulated, characterized, and evaluated, evidencing their pharmacological potential. The UBO-loaded films were analyzed using standard methods regarding physicochemical and pharmacotechnical characteristics to verify their suitability for topical administration on the oral mucosa. F-UBO suitability confirmation allowed for the investigation of antimicrobial and anticancer potential. The antimicrobial properties against Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 27353, Candida albicans ATCC 10231, and Candida parapsilosis ATCC 22019 were evaluated by a resazurin-based 96-well plate microdilution method. The brine shrimp lethality assay (BSL assay) was the animal model cytotoxicity prescreen, followed by flow cytometry analyses on normal blood cells and oral epithelial squamous cell carcinoma CLS-354 cell line, determining cellular apoptosis, caspase-3/7 activity, nuclear condensation and lysosomal activity, oxidative stress, cell cycle, and cell proliferation. The results indicate that a UBO-loaded bioadhesive film’s weight is 63 ± 1.79 mg. It contains 315 µg UBO, has a pH = 6.97 ± 0.01, a disintegration time of 124 ± 3.67 s, and a bioadhesion time of 86 ± 4.12 min, being suitable for topical administration on the oral mucosa. F-UBO showed moderate dose-dependent inhibitory effects on the growth of both bacterial and fungal strains. Moreover, in CLS-354 tumor cells, F-UBO increased oxidative stress, diminished DNA synthesis, and induced cell cycle arrest in G0/G1. All these properties led to considering UBO-loaded bioadhesive oral films as a suitable phytotherapeutic formulation with potential application in oral infections and neoplasia.

Medical research explores plant extracts’ properties to obtain potential anticancer drugs. The present study aims to formulate, develop, and characterize the bioadhesive oral films containing Usnea barbata (L.) dry ethanol extract (F-UBE-HPC) and to investigate their anticancer potential for possible use in oral cancer therapy. The physicochemical and morphological properties of the bioadhesive oral films were analyzed through Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), Atomic Force Microscopy (AFM), thermogravimetric analysis (TG), and X-ray diffraction techniques. Pharmacotechnical evaluation (consisting of the measurement of the specific parameters: weight uniformity, thickness, folding endurance, tensile strength, elongation, moisture content, pH, disintegration time, swelling rate, and ex vivo mucoadhesion time) completed the bioadhesive films’ analysis. Next, oxidative stress, caspase 3/7 activity, nuclear condensation, lysosomal activity, and DNA synthesis induced by F-UBE-HPC in normal blood cell cultures and oral epithelial squamous cell carcinoma (CLS-354) cell line and its influence on both cell types’ division and proliferation was evaluated. The results reveal that each F-UBE-HPC contains 0.330 mg dry extract with a usnic acid (UA) content of 0.036 mg. The bioadhesive oral films are thin (0.093 ± 0.002 mm), reveal a neutral pH (7.10 ± 0.02), a disintegration time of 118 ± 3.16 s, an ex vivo bioadhesion time of 98 ± 3.58 min, and show a swelling ratio after 6 h of 289 ± 5.82%, being suitable for application on the oral mucosa. They displayed in vitro anticancer activity on CLS-354 tumor cells. By considerably increasing cellular oxidative stress and caspase 3/7 activity, they triggered apoptotic processes in oral cancer cells, inducing high levels of nuclear condensation and lysosomal activity, cell cycle arrest in G0/G1, and blocking DNA synthesis. All these properties lead to considering the UBE-loaded bioadhesive oral films suitable for potential application as a complementary therapy in oral cancer.

Physical Vapor Deposition (PVD) is a commonly used technique for back side metallization (BSM) and under bump metallization (UBM). Cutting edge BSM and UBM stacks require good adhesion, low contact resistance and low intrinsic stress. Nickel Vanadium (NiV) is commonly used as a barrier layer in these BSM and UBM stacks. As a non-magnetic material, NiV is more suitable for magnetron sputtering than pure Ni, which is magnetic. NiV tends to be sputtered in a high stress state and high stress in thin films has often degraded adhesion, resulting in long term reliability issues, and bowing of thinned wafers. NiV films deposited by PVD using argon (Ar) process gas are usually under high tensile stress. In these cases, film stress can be controlled by various means during the PVD process: deposition temperature, deposition pressure, and RF bias applied to the wafers during deposition. This paper explores how changing the process gas from pure Ar to a mixture of argon and nitrogen gasses can control film stress. Stress neutral NiV(N) films were achieved by adding up to 50% of nitrogen to the Ar/N2 mixture. Stress, deposition rate, and sheet resistance were studied as a function of nitrogen percentage in the process gas mixture. Electrical and barrier properties of the resulting NiV(N) films were found to be comparable to or better than NiV films deposited with a pure Ar gas.

Oral squamous cell carcinoma (OSCC) is the most frequent oral malignancy, with a high death rate and an inadequate response to conventional chemotherapeutic drugs. Medical research explores plant extracts’ properties to obtain potential nanomaterial-based anticancer drugs. The present study aims to formulate, develop, and characterize mucoadhesive oral films loaded with Usnea barbata (L.) dry acetone extract (F-UBA) and to investigate their anticancer potential for possible use in oral cancer therapy. U. barbata dry acetone extract (UBA) was solubilized in ethanol: isopropanol mixture and loaded in a formulation containing hydroxypropyl methylcellulose (HPMC) K100 and polyethylene glycol 400 (PEG 400). The UBA influence on the F-UBA pharmaceutical characteristics was evidenced compared with the references, i.e., mucoadhesive oral films containing suitable excipients but no active ingredient loaded. Both films were subjected to a complex analysis using standard methods to evaluate their suitability for topical administration on the oral mucosa. Physico-chemical and structural characterization was achieved by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), thermogravimetric analysis (TGA), scanning electron microscopy (SEM), and atomic force microscopy (AFM). Pharmacotechnical evaluation (consisting of the measurement of specific parameters: weight uniformity, thickness, folding endurance, tensile strength, elongation, moisture content, pH, disintegration time, swelling rate, and ex vivo mucoadhesion time) proved that F-UBAs are suitable for oral mucosal administration. The brine shrimp lethality (BSL) assay was the F-UBA cytotoxicity prescreen. Cellular oxidative stress, caspase 3/7 activity, nuclear condensation, lysosomal activity, and DNA synthesis induced by F-UBA in blood cell cultures and oral epithelial squamous cell carcinoma (CLS-354) cell line were investigated through complex flow cytometry analyses. Moreover, F-UBA influence on both cell type division and proliferation was determined. Finally, using the resazurin-based 96-well plate microdilution method, the F-UBA antimicrobial potential was explored against Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 27353, Candida albicans ATCC 10231, and Candida parapsilosis ATCC 22019. The results revealed that each UBA-loaded film contains 175 µg dry extract with a usnic acid (UA) content of 42.32 µg. F-UBAs are very thin (0.060 ± 0.002 mm), report a neutral pH (7.01 ± 0.01), a disintegration time of 146 ± 5.09 s, and an ex vivo mucoadhesion time of 85 ± 2.33 min, and they show a swelling ratio after 6 h of 211 ± 4.31%. They are suitable for topical administration on the oral mucosa. Like UA, they act on CLS-354 tumor cells, considerably increasing cellular oxidative stress, nuclear condensation, and autophagy and inducing cell cycle arrest in G0/G1. The F-UBAs inhibited the bacterial and fungal strains in a dose-dependent manner; they showed similar effects on both Candida sp. and higher inhibitory activity against P. aeruginosa than S. aureus. All these properties lead to considering the UBA-loaded mucoadhesive oral films suitable for potential application as a complementary therapy in OSCC.

The influence of temperature (20–80°C) on the electrochemical behaviour of passive films anodically formed on UB6 stainless steel in phosphoric acid solution (5.5 M H3PO4) has been examined by using potentiodynamic curves, electrochemical impedance spectroscopy, and Mott-Schottky analysis. UB6 stainless steel in contaminated phosphoric acid is characterised by high interfacial impedance, thereby, illustrating its high corrosion resistance. The obtained results show that the films behave as n-type and p-type semiconductors in the potential range above and below the flat band potential, respectively. This behaviour is assumed to be the consequence of the semiconducting properties of the iron oxide and chromium oxide regions which compose the passive film.

NbxSiyNz thin film nanostructure was grown using the unbalanced magnetron sputtering (UBM) technique with varying Si content. Corrosion resistance was evaluated by potentiodynamic polarisation technique in a 3% NaCl solution. Microstructure was analysed by X-ray diffraction (XRD), scanning electron microscopy (SEM) and laser scanning microscopy. Chemical composition was ascertained by X-ray fluorescence (XRF) technique. The results showed that deposition rates increased with higher Si content. A microstructural change was observed for greater than 5% Si content through the transition from a crystalline to an amorphous structure in the thin films. Corrosion test results demonstrated that the thin films having the highest silicon content had better corrosion resistance.

This thesis covers the topic ”Thinning and Turbulence in Aqueous Films”. Experimental studies in two-dimensional systems gained an increasing amount of attention during the last decade. Thin liquid films serve as paradigms of atmospheric convection, thermal convection in the Earth’s mantle or turbulence in magnetohydrodynamics. Recent research on colloids, interfaces and nanofluids lead to advances in the developtment of micro-mixers (lab-on-a-chip devices). In this project a detailed description of a thin film experiment with focus on the particular surface forces is presented. The impact of turbulence on the thinning of liquid films which are oriented parallel to the gravitational force is studied. An experimental setup was developed which permits the capturing of thin film interference patterns under controlled surface and atmospheric conditions. The measurement setup also serves as a prototype of a mixer on the basis of thermally induced turbulence in liquid thin films with thicknesses in the nanometer range. The convection is realized by placing a cooled copper rod in the center of the film. The temperature gradient between the rod and the atmosphere results in a density gradient in the liquid film, so that different buoyancies generate turbulence. In the work at hand the thermally driven convection is characterized by a newly developed algorithm, named Cluster Imaging Velocimetry (CIV). This routine determines the flow relevant vector fields (velocity and deformation). On the basis of these insights the flow in the experiment was investigated with respect to its mixing properties. The mixing characteristics were compared to theoretical models and mixing efficiency of the flow scheme calculated. The gravitationally driven thinning of the liquid film was analyzed under the influence of turbulence. Strong shear forces lead to the generation of ultra-thin domains which consist of Newton black film. Due to the exponential expansion of the thin areas and the efficient mixing, this two-phase flow rapidly turns into the convection of only ultra-thin film. This turbulence driven transition was observed and quantified for the first time. The existence of stable convection in liquid nanofilms was proven for the first time in the context of this work.
Diese Diplomarbeit behandelt das Thema ”Dünnung und Turbulenz in wässrigen Filmen”. Experimente in zweidimensionalen Systemen erfuhren in den vergangenen Jahren zunehmend an Aufmerksamkeit. Dünne Flüssigkeitsschichten dienen als Modell für atmosphärische Konvektion, thermische Konvektion im Erdmantel oder Turbulenz in der Magnetohydrodynamik. Aktuelle Forschung im Bereich der Kolloide, Grenzflächen und Nanofluidik führt zu Fortschritten in der Entwicklung von Mikromixern (”lab-on-a-chip”). In diesem Projekt wird eine detaillierte Beschreibung eines Dünnfilmexperiments mit Fokus auf die besonderen Oberflächenkräfte vorgestellt. Die Auswirkung der Turbulenz auf die Dünnung von parallel zur Gravitationskraft orientierten Flüssigkeitsschichten wurde untersucht. Es wurde ein Experiment entwickelt, welches die Aufnahme von Dünnschichtinterferenzmustern unter kontrollierten Oberflächenbedingungen und Atmosphäre erlaubt. Der Messaufbau dient auch als Prototyp eines Mixers auf Basis von thermisch induzierter Turbulenz in Flüssigkeitsfilmen mit Dicken im Nanometerbereich. Die Konvektion wird durch das Platzieren eines gekühlten Kupferstabs in der Mitte des Films realisiert. Der Temperaturgradient zwischen Stab und äußerer Atmosphäre resultiert in einem Dichtegradienten in dem flüssigen Film, sodass durch unterschiedliche Auftriebskräfte Turbulenz erzeugt wird. In der vorliegenden Arbeit ist die thermisch getriebenen Konvektion an Hand eines neu entwickelten Verfahrens (Cluster Imaging Velocimetry - CIV) zur Ermittlung des strömungsrelevanten Vektorfelder (Geschwindigkeit und Deformation) charakterisiert worden. Auf Basis dieser Erkenntnisse wurde die im Experiment vorherrschende Strömung in Hinsicht auf ihre Mischungseigenschaften im Vergleich zu theoretischen Modellen untersucht und die Mischungseffizienz berechnet. Die gravitationsgetriebene Ausdünnung der Flüssigkeitsschicht unter Einfluss der Turbulenz wurde analysiert. Durch starke Scherkräfte kommt es lokal zur Bildung ultradünner Domänen bestehend aus ”Newton black film”. Diese Zweiphasenströmung geht durch das exponentielle Ausdehnen der dünnen Bereiche und die effiziente Mischung sehr schnell in eine Konvektion von ausschließlich ultradünnem Film im Gleichgewichtszustand über. Dieser turbulenzgetriebene Übergang wurde zum ersten Mal beobachtet und quantifiziert. Die Existenz stabiler Konvektion in flüssigen Nanofilmen ist zum ersten Mal im Rahmen dieser Arbeit belegt worden.

Es ist seit einigen Jahren bekannt, dass Elektronen unter bestimmten Bedingungen in dünne Filme eingeschlossen werden können, selbst wenn diese Filme aus Metall bestehen und auf Metall-Substrat aufgebracht werden. In Photoelektronenspektren zeigen diese Filme charakteristische diskrete Energieniveaus, und es hat sich herausgestellt, dass sie zu großen, technisch nutzbaren Effekten führen können, wie der oszillatorischen magnetischen Kopplung in modernen Festplatten-Leseköpfen.

In dieser Arbeit wird untersucht, inwieweit die der Quantisierung in zweidimensionalen Filmen zu Grunde liegenden Konzepte auf niedrigere Dimensionalität übertragbar sind. Das bedeutet, dass schrittweise von zweidimensionalen Filmen auf eindimensionale Nanostrukturen übergegangen wird. Diese Nanostrukturen sind zum einen die Terrassen auf atomar gestuften Oberflächen, aber auch Atomketten, die auf diese Terrassen aufgebracht werden, bis hin zu einer vollständigen Bedeckung mit atomar dünnen Nanostreifen. Daneben werden Selbstorganisationseffekte ausgenutzt, um zu perfekt eindimensionalen Atomanordnungen auf Oberflächen zu gelangen.

Die winkelaufgelöste Photoemission ist als Untersuchungsmethode deshalb so geeignet, weil sie das Verhalten der Elektronen in diesen Nanostrukturen in Abhängigkeit von der Raumrichtung zeigt, und unterscheidet sich darin beispielsweise von der Rastertunnelmikroskopie. Damit ist es möglich, deutliche und manchmal überraschend große Effekte der eindimensionalen Quantisierung bei verschiedenen exemplarischen Systemen zum Teil erstmals nachzuweisen. Die für zweidimensionale Filme wesentliche Rolle von Bandlücken im Substrat wird für Nanostrukturen bestätigt. Hinzu kommt jedoch eine bei zweidimensionalen Filmen nicht vorhandene Ambivalenz zwischen räumlicher Einschränkung der Elektronen in den Nanostrukturen und dem Effekt eines Übergitters aus Nanostrukturen sowie zwischen Effekten des Elektronenverhaltens in der Probe und solchen des Messprozesses. Letztere sind sehr groß und können die Photoemissionsspektren dominieren.

Abschließend wird der Effekt der verminderten Dimensionalität speziell für die d-Elektronen von Mangan untersucht, die zusätzlich starken Wechselwirkungseffekten unterliegen. Auch hierbei treten überraschende Ergebnisse zu Tage.
It has been known for several years that under certain conditions electrons can be confined within thin layers even if these layers consist of metal and are supported by a metal substrate. In photoelectron spectra, these layers show characteristic discrete energy levels and it has turned out that these lead to large effects like the oscillatory magnetic coupling technically exploited in modern hard disk reading heads.

The current work asks in how far the concepts underlying quantization in two-dimensional films can be transferred to lower dimensionality. This problem is approached by a stepwise transition from two-dimensional layers to one-dimensional nanostructures. On the one hand, these nanostructures are represented by terraces on atomically stepped surfaces, on the other hand by atom chains which are deposited onto these terraces up to complete coverage by atomically thin nanostripes. Furthermore, self organization effects are used in order to arrive at perfectly one-dimensional atomic arrangements at surfaces.

Angle-resolved photoemission is particularly suited as method of investigation because is reveals the behavior of the electrons in these nanostructures in dependence of the spacial direction which distinguishes it from, e. g., scanning tunneling microscopy. With this method intense and at times surprisingly large effects of one-dimensional quantization are observed for various exemplary systems, partly for the first time. The essential role of bandgaps in the substrate known from two-dimensional systems is confirmed for nanostructures. In addition, we reveal an ambiguity without precedent in two-dimensional layers between spacial confinement of electrons on the one side and superlattice effects on the other side as well as between effects caused by the sample and by the measurement process. The latter effects are huge and can dominate the photoelectron spectra.

Finally, the effects of reduced dimensionality are studied in particular for the d electrons of manganese which are additionally affected by strong correlation effects. Surprising results are also obtained here.

The presented work describes new concepts of fast switching elements based on principles of photonics. The waveguides working in visible and infra-red ranges are put in a basis of these elements. And as materials for manufacturing of waveguides the transparent polymers, dopped by molecules of the dyes possessing second order nonlinear-optical properties are proposed. The work shows how nonlinear-optical processes in such structures can be implemented by electro-optical and opto-optical control circuit signals. In this paper we consider the complete cycle of fabrication of several types of integral photonic elements. The theoretical analysis of high-intensity beam propagation in media with second-order optical nonlinearity is performed. Quantitative estimations of necessary conditions of occurrence of the nonlinear-optical phenomena of the second order taking into account properties of used materials are made. The paper describes the various stages of manufacture of the basic structure of the integrated photonics: a planar waveguide. Using the finite element method the structure of the electromagnetic field inside the waveguide in different modes was analysed. A separate part of the work deals with the creation of composite organic materials with high optical nonlinearity. Using the methods of quantum chemistry, the dependence of nonlinear properties of dye molecules from its structure were investigated in details. In addition, the paper discusses various methods of inducing of an optical nonlinearity in dye-doping of polymer films. In the work, for the first time is proposed the use of spatial modulation of nonlinear properties of waveguide according Fibonacci law. This allows involving several different nonlinear optical processes simultaneously. The final part of the work describes various designs of integrated optical modulators and switches constructed of organic nonlinear optical waveguides. A practical design of the optical modulator based on Mach-Zehnder interferometer made by a photolithography on polymer film is presented.
Die vorliegende Arbeit beschreibt neue Konzepte für schnelle photonische Schaltelemente. Diese Elemente basieren auf optischer Wellenleitung im sichtbaren und nahen infraroten Spektralbereich. Die Arbeit ist auf organische Wellenleiter aus transparenten, farbstoffdotierten Polymeren fokussiert, welche Chromophore mit besonders großen optischen Nichtlinearitäten zweiter Ordnung enthalten. Insbesondere wird dargestellt, wie nichtlineare optische Prozesse in derartigen Bauelementen genutzt werden können, wenn man elektrische oder optische Steuersignale einsetzt. Es wird der gesamte Her-stellungszyklus verschiedener integrierter photonischer Bauelemente betrachtet. Die Arbeit umfasst weiterhin eine detaillierte theoretische Analyse der Wellenausbreitung in Medien mit großer nichtlinearer optischer Suszeptibilität zweiter Ordnung bei hoher Lichtintensität. Unter Annahme optischer Materialkonstanten, welche den experimentell ermittelten Werten entsprechen, erfolgte eine quantitative Abschätzung für das Auftreten von Szenarien, bei denen messbare Abwei-chungen der Strahlausbreitung vom linearen Regime auftreten, z:B. das Auftreten von zeitlichen, räumlichen und spektralen Satteliten zu den im Material propagierenden Laserpulsen. Es gelang, we-sentliche Aspekte der Strahlpropagationsprozesse durch Anwendung von Jacobi-Integralfunktionen in geschlossener mathematischer Form darzustellen. Darüber hinausgehende theoretische Untersuchungen nutzten die Finite-Elemente-Methode, um die Verteilung des elektromagnetischen Feldes im Inneren optischer Wellenleiter für verschiedene Pro-pagationsmoden zu analysieren. Ein weiterer Teil der Arbeit beschäftigt sich mit organischen Kompositmaterialen, welche große opti-sche Nichtlinearitäten aufweisen. Mittels quantenchemischer Verfahren erfolgte eine detaillierte Un-tersuchung der Zusammenhänge zwischen der molekularen Struktur und den linearen sowie den nichtlinearen optischen Eigenschaften der untersuchten Farbstoffmoleküle und Polymere. In Bezug auf die Probenpräparation sind unterschiedliche Verfahren zur Schichtbildung und zur molekularen Orientierung miteinander verglichen und bewertet worden, da letztere eine Voraussetzung für das Auftreten nichtlinearer optischer Prozesse zweiter Ordnung bildet. Die Untersuchungen erstreckten sich auf Vakuumaufdampfschichten aus niedermolekularen Chromophoren und auf Polymerschich-ten, welche durch Vakuumabscheidung sowie durch Spincoating oder andere Flüssigphasenabschei-dungsverfahren hergestellt worden sind. Auf orientierenden Substraten (z.B. geriebene Schichten aus Polytetrafluoräthylen) konnte eine spontane Orientierung der deponierten Chromophore nachgewie-sen werden. Die Chromophore in Polymerschichten wurden durch Coronapolung orientiert. In der Arbeit ist zum ersten Mal vorgeschlagen worden, eine räumliche Modulation der nichtlinearen optischen Eigenschaften durch gezielte lokale Coronapolung oder andere Orientierungstechniken derart zu generieren, dass die Abfolge gepolter Domänen dem Bildungsgesetz einer modifizierten Fibonacci-Reihe mit gebrochen rationalen Zahlen gehorcht. Der Vorteil von optischen Wellenleitern mit dieser Struktur darin, dass diese mehrere unterschiedli-che nichtlineare optische Prozesse gleichzeitig unterstützen und somit eine Vielzahl neuartiger Phä-nomene auftreten kann, welche man in neuartigen photonischen Bauelementen, z.B. für die optische Informations- und Kommunikationstechnik nutzen kann. Der letzte Teil der Arbeit beschreibt den Aufbau verschiedener integrierter optischer Modulatoren und Schalter, die sich mit Hilfe das neu entwickelten Verfahrens herstellen lassen. Zukünftige Entwicklungen in der Photonik, insbesondere der optischen Informations- und Kommuni-kationstechnik werden im abschließenden Teil der Arbeit diskutiert.

Electron transfer phenomena in proteins represent one of the most common types of biochemical reactions. They play a central role in energy conversion pathways in living cells, and are crucial components in respiration and photosynthesis. These complex biochemical reaction cascades consist of a series of proteins and protein complexes that couple a charge transfer to different forms of chemical energy. The efficiency and sophisticated optimisation of signal transfer in these natural redox chains has inspired engineering of artificial architectures mimicking essential properties of their natural analogues. Implementation of direct electron transfer (DET) in protein assemblies was a breakthrough in bioelectronics, providing a simple and efficient way for coupling biological recognition events to a signal transducer. DET avoids the use of redox mediators, reducing potential interferences and side reactions, as well as being more compatible with in vivo conditions. However, only a few haem proteins, including the redox protein cytochrome c (cyt.c), and blue copper enzymes show efficient DET on different kinds of electrodes. Previous investigations with cyt.c have mainly focused on heterogeneous electron transfer of monolayers of this protein on gold. An important advance was the fabrication of cyt.c multilayers by electrostatic layer-by-layer self-assembly. The ease of fabrication, the stability, and the controllable permeability of polyelectrolyte multilayers have made them particularly attractive for electroanalytical applications. With cyt.c and sulfonated polyaniline it was for the first time possible that fully electro-active multilayers of the redox protein could be prepared. This approach was extended to design an analytical signal chain based on multilayers of cyt.c and xanthine oxidase (XOD). The system does not need an external mediator but relies on an in situ generation of a mediating radical and thus allows a signal transfer from hypoxanthine via the substrate converting enzyme and cyt.c to the electrode. Another kind of a signal chain is based on assembling proteins in complexes on electrodes in such a way that a direct protein-protein electron transfer becomes feasible. This design does not need a redox mediator in analogy to natural protein communication. For this purpose, cyt.c and the enzyme bilirubin oxidase (BOD, EC 1.3.3.5) are co-immobilized in a self-assembled polyelectrolyte multilayer on gold electrodes. Although these two proteins are not natural reaction partners, the protein architecture facilitates an electron transfer from the electrode via multiple protein layers to molecular oxygen resulting in a significant catalytic reduction current. Finally, we describe a novel strategy for multi-protein layer-by-layer self-assembly combining cyt.c with an enzyme sulfite oxidase (SOx) without use of any additional polymer. Electrostatic interactions between these two proteins with rather separated pI values during the assembly process from a low ionic strength buffer were found sufficient for the layer-by-layer deposition of the both biomolecules. It is anticipated that the concepts described in this work will stimulate further progress in multilayer design of even more complex biomimetic signal cascades taking advantage of direct communication between proteins.
Elektronentransferphänomene in Proteinen stellen den häufigsten Typ biochemischer Reaktionen dar. Sie spielen eine zentrale Rolle bei der Energieumwandlung in der Zelle und sind entscheidende Komponenten in der Atmung und Photosynthese. Diese komplexen Kaskaden biochemischer Reaktionen setzen sich aus einer Reihe von Proteinen und Proteinkomplexen zusammen, die den Energietransfer an verschiedene Formen chemischer Energie koppeln. Die große Effektivität und Selektivität des Signaltransfers in diesen natürlichen Redoxketten war Vorbild für die Entwicklung künstlicher Architekturen, die die wesentlichen Eigenschaften ihrer natürlichen Analoga nachahmen. Die Implementierung des direkten Elektronentransfers (DET) von Proteinen mit Elektroden war ein Durchbruch im Bereich der Bioelektronik. Sie lieferte einen einfachen und effizienten Weg für das Koppeln biologischer Erkennungsereignisse an einen Signalumwandler. Durch den DET können Redoxmediatoren vermieden und damit potentielle Grenzflächen und Nebenreaktionen reduziert werden. Ebenso wird damit die Kompatibilität für in vivo Bedingungen erhöht. Jedoch zeigen nur einige Hämproteine wie das Redoxprotein Cytochrom c (Cyt c) und blaue Kupferproteine einen effizienten DET auf verschiedenen Elektrodentypen. Bisherige Untersuchungen mit Cyt c konzentrierten sich hauptsächlich auf den heterogenen Elektronentransfer von Monoschichten dieses Proteins auf Gold. Ein wichtiger Fortschritt war die Herstellung von Cyt c Multischichten durch die elektrostatische Layer-by-Layer-Technik. Die einfache Herstellung, die Stabilität sowie die kontrollierbaren Permeationseigenschaften von Polyelektrolyt-Multischichten machte sie besonders attraktiv für elektroanalytische Anwendungen. So gelang es auch zum ersten Mal vollständig elektroaktive Multischichten aus Cyt c und Polyanilinsulfonsäure zu präparieren. Dieser Ansatz wurde hier erweitert, um eine analytische Signalkette auf der Basis von Multischichten aus Cyt c und Xanthinoxidase zu entwerfen. Das System bedarf keinen externen Mediator, es hängt jedoch von der in situ Generierung eines vermittelnden Radikals ab und erlaubt daher einen Signaltransfer von Hypoxanthin über ein substratumwandelndes Enzym und Cyt c zur Elektrode. Eine andere Art von Signalketten basiert auf der Assemblierung von Proteinen in Komplexen auf Elektroden in solcher Art und Weise, daß ein direkter Protein-Protein-Elektronentransfer möglich wird. Dieser Ansatz benötigt keinen Redoxmediator in Analogie zu Beispielen aus dem biologischen Signaltransfer. Zu diesem Zweck werden Cyt c und das Enzym Bilirubinoxidase mit einem selbst-assemblierenden Polyelektrolyten auf einer Goldelektrode koimmobilisiert. Obwohl diese zwei Proteine keine natürlichen Reaktionspartner sind, unterstützt die Protein-Architektur einen Elektronentransfer von der Elektrode über mehrere Proteinschichten zu molekularem Sauerstoff und ergibt einen signifikanten katalytischen Reduktionsstrom. Schließlich wird eine neue Strategie beschrieben für eine Selbstassemblierung von Proteinen ohne zusätzlichen Polyelektrolyten - am Beispiel der Kombination von Cyt c mit Sulfitoxidase. Es stellte sich heraus, daß die elektrostatische Wechselwirkung zwischen diesen zwei Proteinen mit ziemlich weit voneinander entfernt liegenden pI-Werten während des Assemblierungsprozesses durch einen Puffer mit geringer Ionenstärke ausreicht um die beiden Biomoleküle nach dem Layer-by-Layer-Prinzip auf einer Elektrode abzuscheiden. Es wird erwartet, daß das entwickelte Konzept von Multiprotein-Assemblaten auf Elektroden weitere Fortschritte bei dem Entwurf von Multischichten und sogar noch komplexeren biomimetischen Signalkaskaden anregen wird und dabei der Vorteil der direkten Kommunikation zwischen Proteinen genutzt wird.

In den vergangenen Jahren wurden stetig wachsende Produktionskapazitäten von Biokunststoffen aus nachwachsenden Rohstoffe nverzeichnet. Trotz großer Produktionskapazitäten und einem geeigneten Eigenschaftsprofil findet Stärke nur als hydrophile, mit Weichmachern verarbeitete thermoplastische Stärke (TPS) in Form von Blends mit z. B. Polyestern Anwendung. Gleiches gilt für Kunststoffe auf Proteinbasis. Die vorliegende Arbeit hat die Entwicklung von Biokunststoffen auf Stärkebasis zum Ziel, welche ohne externe Weichmacher thermoplastisch verarbeitbar und hydrophob sind sowie ein mechanisches Eigenschaftsprofil aufweisen, welches ein Potenzial zur Herstellung von Materialien für eine Anwendung als Verpackungsmittel bietet. Um die Rohstoffbasis für Biokunststoffe zu erweitern, soll das erarbeitete Konzept auf zwei industriell verfügbare Proteintypen, Zein und Molkenproteinisolat (WPI), übertragen werden. Als geeignete Materialklasse wurden Fettsäureester der Stärke herausgearbeitet. Zunächst fand ein Vergleich der Säurechlorid-Veresterung und der Umesterung von Fettsäurevinylestern statt, woraus letztere als geeignetere Methode hervorging. Durch Variation der Reaktionsparameter konnte diese optimiert und auf eine Serie der Fettsäurevinylester von Butanoat bis Stearat für DS-Werte bis zu 2,2-2,6 angewandt werden. Möglich war somit eine systematische Studie unter Variation der veresterten Fettsäure sowie des Substitutionsgrades (DS). Sämtliche Produkte mit einem DS ab 1,5 wiesen eine ausgprägte Löslichkeit in organischen Lösungsmitteln auf wodurch sowohl die Aufnahme von NMR-Spektren als auch Molmassenbestimmung mittels Größenausschlusschromatographie mit gekoppelter Mehrwinkel-Laserlichtstreuung (GPC-MALLS) möglich waren. Durch dynamische Lichtstreuung (DLS) wurde das Löslichkeitsverhalten veranschaulicht. Sämtliche Produkte konnten zu Filmen verarbeitet werden, wobei Materialien mit DS 1,5-1,7 hohe Zugfestigkeiten (bis zu 42 MPa) und Elastizitätsmodule (bis 1390 MPa) aufwiesen. Insbesondere Stärkehexanoat mit DS <2 sowie Stärkebutanoat mit DS >2 hatten ein mechanisches Eigenschaftsprofil, welches insbesondere in Bezug auf die Festigkeit/Steifigkeit vergleichbar mit Verpackungsmaterialien wie Polyethylen war (Zugfestigkeit: 15-32 MPa, E-Modul: 300-1300 MPa). Zugfestigkeit und Elastizitätsmodul nahmen mit steigender Kettenlänge der veresterten Fettsäure ab. Ester längerkettiger Fettsäuren (C16-C18) waren spröde. Über Weitwinkel-Röntgenstreuung (WAXS) und Infrarotspektroskopie (ATR-FTIR) konnte der Verlauf der Festigkeiten mit einer zunehmenden Distanz der Stärke im Material begründet werden. Es konnten von DS und Kettenlänge abhängige Glasübergänge detektiert werden, die kristallinen Strukturen der langkettigen Fettsäuren zeigten einen Schmelzpeak. Die Hydrophobie der Filme wurde anhand von Kontaktwinkeln >95° gegen Wasser dargestellt. Blends mit biobasierten Polyterpenen sowie den in der Arbeit hergestellten Zein-Acylderivaten ermöglichten eine weitere Verbesserung der Zugfestigkeit bzw. des Elastizitätsmoduls hochsubstituierter Produkte. Eine thermoplastische Verarbeitung mittels Spritzgießen war sowohl für Produkte mit hohem als auch mittlerem DS-Wert ohne jeglichen Zusatz von Weichmachern möglich. Es entstanden homogene, transparente Prüfstäbe. Untersuchungen der Härte ergaben auch hier für Stärkehexanoat und –butanoat mit Polyethylen vergleichbare Werte. Ausgewählte Produkte wurden zu Fasern nach dem Schmelzspinnverfahren verarbeitet. Hierbei wurden insbesondere für hochsubstituierte Derivate homogenen Fasern erstellt, welche im Vergleich zur Gießfolie signifikant höhere Zugfestigkeiten aufwiesen. Stärkeester mit mittlerem DS ließen sich ebenfalls verarbeiten. Zunächst wurden für eine Übertragung des Konzeptes auf die Proteine Zein und WPI verschiedene Synthesemethoden verglichen. Die Veresterung mit Säurechloriden ergab hierbei die höchsten Werte. Im Hinblick auf eine gute Löslichkeit in organischen Lösungsmitteln wurde für WPI die Veresterung mit carbonyldiimidazol (CDI)-aktivierten Fettsäuren in DMSO und für Zein die Veresterung mit Säu-rechloriden in Pyridin bevorzugt. Es stellte sich heraus, dass acyliertes WPI zwar hydrophob, jedoch ohne Weichmacher nicht thermoplastisch verarbeitet werden konnte. Die Erstellung von Gießfolien führte zu Sprödbruchverhalten. Unter Zugabe der biobasierten Ölsäure wurde die Anwendung von acyliertem WPI als thermoplastischer Filler z. B. in Blends mit Stärkeestern dargestellt. Im Gegensatz hierzu zeigte acyliertes Zein Glasübergänge <100 °C bei ausreichender Stabilität (150-200 °C). Zeinoleat konnte ohne Weichmacher zu einer transparenten Gießfolie verarbeitet werden. Sämtliche Derivate erwiesen sich als ausgeprägt hydrophob. Zeinoleat konnte über das Schmelzspinnverfahren zu thermoplastischen Fasern verarbeitet werden.
In recent years, a steadily growing production capacity of bioplastic based on renewable resources was noticed. Despite its huge production capacities and an appropriate property profile (ubiquitous occurrence, easy extraction), starch is only applied in addition of plasticizers in a hydrophilic, thermoplastic form in blends with e. g. polyesters. The same applies to bioplastics based on proteins. The actual study has the aim to develop starch-based bioplastics, which are hydrophobic, thermoplastic without the addition of any plasticizer and have mechanical properties to be a suitable alternative material in the area of food packaging. To obtain a variation of the raw materials for bioplastics, the concept shall be applied to two types of industrial available proteins, whey protein isolate (WPI) and Zein. Fatty acid esters of starch came out to be a suitable class of materials. Initially, the methods of esterifying acid chlorides and the transesterification of fatty acid vinyl esters were compared with the latter being more appropriate. Reaction parameters of this method were optimized and it was applied to a complete series of vinyl ester reagents (butanoate to stearate), leading to degree of substitution (DS)-values up to 2.2-2.6. With that, a systematic study of the variation of the fatty acid ester chain as well as the DS became possible. It came out that all products with a DS >1.5 showed a well-marked solubility in organic solvents, whereby solution NMR-studies as well as measurements of the molecular weight distributions by using size exclusion chroma-tography with multi-angle laser light scattering (SEC-MALLS) were possible. The different solution behavior was studied by dynamic light scattering (DLS). All soluble products could be formed into films via casting, where materials with a DS of 1.5-1.7 showed the highest values concerning tensile strength (up to 42 MPa) and Youngs modulus (up to 1390 MPa). Especially starch hexanoate with DS <2 and starch butanoate with a DS >2 revealed mechanical properties which are comparable to usually applied polymers for food packaging, e. g. polyethylene (tensile strength: 15-20 MPa, E-Mod: 300-1300 MPa). Tensile strength and Youngs modulus were reduced with increasing length of the esterified fatty acid. Wide-angle X-Ray scattering (WAXS) and infrared spectroscopy (ATR-FTIR) explained this tendency by an increasing intermolecular distance of the starch in the material. Glassy transitions of the materials were detected and showed a dependency on the type of esterified fatty acid and the DS. The crystalline structures of the esterified long-chain fatty acids revealed a melting peak. All films came out to be hydrophobic with contact angles against water >95°. The tensile strength and the Youngs modulus of the highly substituted products could be further improved by blending them with biobased polyterpenes as well as the acylated Zein. A thermoplastic processing without the use of any plasticizer additives was possible for products with a medium and high DS. Homogeneous, transparent testing specimens were obtained. The specific mechanical values were comparable with the casted films, although the highest values for the tensile strength and the elongation were lower. Investigations of the hardness showed comparable values to polyethylene. Selected samples were further processed to fibers by melt spinning. Especially starch esters with high DS revealed homogeneous fibers with a significant increase in the tensile strength compared to the film or testing specimen. Even fatty acid starch esters with a medium DS were processed by the melt-spinning, but their higher glassy transition lead to a reduced softening behavior. To transfer this concept to the class of proteins, different methods of synthesis were studied in the first step, which differed in their amount of acylation. The acylation using fatty acid chlorides lead to highest values. With regard to a well-marked organic solvent solubility, in the case of WPI the acylation with carbonyldiimidazol (CDI)-activated fatty acid was established. For Zein, the acid chloride acylation in pyridine gave the desired results. It came out the fatty acid acylated soluble WPI could not be thermoplastic processed without additional plasticizers. By using biobased oleic acid as additive, the potential of acylated WPI as a thermoplastic filler in blends with e. g. fatty acid esters of starch was shown. In contrast, fatty acid acyl derivatives of Zein revealed well marked glassy transitions <100 °C with an adequate thermal stability. While Zeinoleate could be formed into transparent films via solvent casting without any plasticizer additives, low amounts of tall oil enabled film-forming in the case of acyl derivatives with shorter fatty acids as well. All derivatives revealed a well-marked hydrophobicity. Finally, Zeinoleate was thermoplastically processed into fibers by melt-spinning without any further additives.

Piezoelectric materials are very useful for applications in sensors and actuators. In addition to traditional ferroelectric ceramics and ferroelectric polymers, ferroelectrets have recently become a new group of piezoelectrics. Ferroelectrets are functional polymer systems for electromechanical transduction, with elastically heterogeneous cellular structures and internal quasi-permanent dipole moments. The piezoelectricity of ferroelectrets stems from linear changes of the dipole moments in response to external mechanical or electrical stress. Over the past two decades, polypropylene (PP) foams have been investigated with the aim of ferroelectret applications, and some products are already on the market. PP-foam ferroelectrets may exhibit piezoelectric d33 coefficients of 600 pC/N and more. Their operating temperature can, however, not be much higher than 60 °C. Recently developed polyethylene-terephthalate (PET) and cyclo-olefin copolymer (COC) foam ferroelectrets show slightly better d33 thermal stabilities, but usually at the price of smaller d33 values. Therefore, the main aim of this work is the development of new thermally stable ferroelectrets with appreciable piezoelectricity. Physical foaming is a promising technique for generating polymer foams from solid films without any pollution or impurity. Supercritical carbon dioxide (CO2) or nitrogen (N2) are usually employed as foaming agents due to their good solubility in several polymers. Polyethylene propylene (PEN) is a polyester with slightly better properties than PET. A “voiding + inflation + stretching” process has been specifically developed to prepare PEN foams. Solid PEN films are saturated with supercritical CO2 at high pressure and then thermally voided at high temperatures. Controlled inflation (Gas-Diffusion Expansion or GDE) is applied in order to adjust the void dimensions. Additional biaxial stretching decreases the void heights, since it is known lens-shaped voids lead to lower elastic moduli and therefore also to stronger piezoelectricity. Both, contact and corona charging are suitable for the electric charging of PEN foams. The light emission from the dielectric-barrier discharges (DBDs) can be clearly observed. Corona charging in a gas of high dielectric strength such as sulfur hexafluoride (SF6) results in higher gas-breakdown strength in the voids and therefore increases the piezoelectricity. PEN foams can exhibit piezoelectric d33 coefficients as high as 500 pC/N. Dielectric-resonance spectra show elastic moduli c33 of 1 − 12 MPa, anti-resonance frequencies of 0.2 − 0.8 MHz, and electromechanical coupling factors of 0.016 − 0.069. As expected, it is found that PEN foams show better thermal stability than PP and PET. Samples charged at room temperature can be utilized up to 80 − 100 °C. Annealing after charging or charging at elevated temperatures may improve thermal stabilities. Samples charged at suitable elevated temperatures show working temperatures as high as 110 − 120 °C. Acoustic measurements at frequencies of 2 Hz − 20 kHz show that PEN foams can be well applied in this frequency range. Fluorinated ethylene-propylene (FEP) copolymers are fluoropolymers with very good physical, chemical and electrical properties. The charge-storage ability of solid FEP films can be significantly improved by adding boron nitride (BN) filler particles. FEP foams are prepared by means of a one-step procedure consisting of CO2 saturation and subsequent in-situ high-temperature voiding. Piezoelectric d33 coefficients up to 40 pC/N are measured on such FEP foams. Mechanical fatigue tests show that the as-prepared PEN and FEP foams are mechanically stable for long periods of time. Although polymer-foam ferroelectrets have a high application potential, their piezoelectric properties strongly depend on the cellular morphology, i.e. on size, shape, and distribution of the voids. On the other hand, controlled preparation of optimized cellular structures is still a technical challenge. Consequently, new ferroelectrets based on polymer-layer system (sandwiches) have been prepared from FEP. By sandwiching an FEP mesh between two solid FEP films and fusing the polymer system with a laser beam, a well-designed uniform macroscopic cellular structure can be formed. Dielectric resonance spectroscopy reveals piezoelectric d33 coefficients as high as 350 pC/N, elastic moduli of about 0.3 MPa, anti-resonance frequencies of about 30 kHz, and electromechanical coupling factors of about 0.05. Samples charged at elevated temperatures show better thermal stabilities than those charged at room temperature, and the higher the charging temperature, the better is the stability. After proper charging at 140 °C, the working temperatures can be as high as 110 − 120 °C. Acoustic measurements at frequencies of 200 Hz − 20 kHz indicate that the FEP layer systems are suitable for applications at least in this range.
Piezoelektrische Materialien haben große technische und wirtschaftliche Bedeutung für Anwendungen in Sensoren und Aktuatoren. Neben den traditionellen ferroelektrischen Keramiken und Polymeren bilden Ferroelektrete eine neue Gruppe der Piezoelektrika. Ferroelektrete sind reversible funktionelle Polymersysteme zur Umwandlung von elektrischer in mechanische Energie und umgekehrt. Sie zeichnen sich aus durch eine elastische zelluläre Struktur mit internen quasi-permanenten Dipolen. Der Mechanismus der Piezoelektrizität in Ferroelektreten wird dominiert von der Änderung der einzelnen Dipolmomente bei Einwirkung einer äußeren mechanischen Kraft. Insbesondere zelluläres Polypropylene (PP) war in den vergangenen zwei Jahrzehnten Gegenstand intensiver Forschung und Entwicklung im Hinblick auf die grundlegenden Eigenschaften und Anwendungen von Ferroelektreten. Einige bereits erhältliche kommerzielle Produkte nutzen die in geladenem zellulären PP erreichbaren hohen piezoelektrischen d33-Koeffizienten von 600 pC/N und mehr, sind aber durch eine relativ geringe maximale Betriebstemperatur von ungefähr 60 °C eingeschränkt. Die kürzlich entwickelten Ferroelektrete aus zellulärem Polyethylenterephthalat (PET) und zellulären Cyclo-Olefin-Copolymeren (COC) zeigen eine bessere Temperaturbeständigkeit (vor allem COC), allerdings gewöhlich auf Kosten von geringeren d33-Koeffizienten. Das Ziel der vorliegenden Arbeit ist es, temperaturbeständige Ferroelektrete mit für den Markt geeigneten piezoelektrischen Eigenschaften zu entwickeln. Physikalisches Schäumen ist eine beliebte Methode, um besonders reine Polymerschäume herzustellen. Häufig werden, wegen ihrer guten Löslichkeit in vielen Polymeren, Kohlenstoffdioxid (CO2) und Stickstoff (N2) im superkritischen Zustand als Treibmittel eingesetzt. Der Polyester Polyethylennaphtalat (PEN) hat ähnliche Eigenschaften wie PET, ist jedoch temperaturbeständiger. Ein Dreistufenprozess (Schäumen, Aufblähen und Strecken) wurde entwickelt, um PEN-Schäume für hochwertige Ferroelektrete herzustellen. Ungeschäumte PEN-Folien werden mit superkritischem CO2 unter hohem Druck gesättigt und anschließend unter geringem Druck bei Temperaturen nahe der Glastemperatur geschäumt. Um die Hohlräume zu vergrößern, wird der Schaum anschließend mittels Gasdiffusionsexpansion (GDE) aufgebläht. Nach zusätzlichem biaxialen Verstrecken erhält man die optimalen linsenförmigen Zellen, welche zu einer minimalen mechanischen Steifigkeit und einem maximalen piezoelektrischen d33-Koeffizienten des Ferroelektrets führen. Sowohl Korona- als auch Kontaktaufladung werden an zellulärem PEN erfolgreich eingesetzt. Die Lichtemission der dielektrisch behinderten Entladungen (DBDs) kann klar beobachtet werden. Korona-Aufladung in Gasen mit hohen dielektrischen Durchbruchsfestigkeiten, wie z.B. Schwefelhexafluorid (SF6), ermöglicht es, das Paschen-Durchbruchsfeld in den Hohlräumen und damit die erzielbare interne Ladungsdichte zu erhöhen. Dadurch können für zelluläres PEN piezoelektrische d33-Koeffizienten bis zu 500 pC/N erzielt werden. Piezoelektrischen Resonanzmessungen der Ferroelektrete liefern Steifigkeiten c33 im Bereich von 1 – 12 MPa, Antiresonanzfrequenzen von 0.2 – 0.8 MHz und elektromechanische Kopplungsfaktoren zwischen 0.016 und 0.069. PEN-Ferroelektrete zeigen eine bessere Temperaturstabilität als solche aus PP und PET. Der Anwendungsbereich von unbehandeltem PEN reicht bis etwa 80 – 100°C, jener von getemperten oder bei 120°C geladenen Proben bis etwa 110 – 120 °C. Akustische Messungen im Frequenzbereich von 2 Hz – 20 kHz zeigen die Eignung von PEN-Ferroelektretwandlern für Luftschallanwendungen. Fluoriertes Ethylen-Propylen (FEP) ist ein Fluorpolymer mit sehr guten physikalischen, chemischen und elektrischen Eigenschaften. Die Ladungsspeichereigenschaften von ungeschäumtem FEP können durch die Beimengung von Bornitrid deutlich verbessert werden. In dieser Arbeit wird zelluläres FEP mittels eines einstufigen Prozesses, dem schon erwähnten Schäumen mit überkritischem CO2, hergestellt. Die geladenen FEP-Proben weisen d33-Koeffizienten von bis zu 40 pC/N auf. Ermüdungstests zeigen eine sehr gute mechanische Stabilität von PEN- und FEP-Ferroelektreten. Zelluläre Polymerferroelektrete haben großes Potenzial für Anwendungen, und die Suche nach geeigneten zellulären Morphologien ist eng verknüpft mit dem technischen Aufwand ihrer Herstellung. Alternativ wurden Ferroelektrete mit Sandwich-Strukturen aus FEP-Folien entwickelt. Durch Laserverschmelzen eines FEP-Foliengitters mit zwei umgebenden FEP-Folien wird eine definierte, einheitliche zelluläre Struktur gebildet. Aus dielektrischen Resonanzspektren können effektive piezoelektrische d33-Koeffizienten bis zu 350 pC/N, effektive mechanische Steifigkeiten um 0.3 MPa, Antiresonanzfrequenzen um 30 kHz und elektromechanische Kopplungsfaktoren von etwa 0.05 abgeleitet werden. Proben, welche bei erhöhter Temperatur geladen werden, zeigen höhere Ladungsstabilitäten. Nach geeigneter Aufladung bei 140 °C kann die Arbeitstemperatur bis auf 110 – 120 °C gesteigert werden. Akustische Messungen im Frequenzbereich von 2 Hz – 20 kHz zeigen die Eignung von FEP-Sandwich-Strukturen für Luftschallanwendungen.

With the rise of nanotechnology in the last decade, nanofluidics has been established as a research field and gained increased interest in science and industry. Natural aqueous nanofluidic systems are very complex, there is often a predominance of liquid interfaces or the fluid contains charged or differently shaped colloids. The effects, promoted by these additives, are far from being completely understood and interesting questions arise with regards to the confinement of such complex fluidic systems. A systematic study of nanofluidic processes requires designing suitable experimental model nano – channels with required characteristics. The present work employed thin liquid films (TLFs) as experimental models. They have proven to be useful experimental tools because of their simple geometry, reproducible preparation, and controllable liquid interfaces. The thickness of the channels can be adjusted easily by the concentration of electrolyte in the film forming solution. This way, channel dimensions from 5 – 100 nm are possible, a high flexibility for an experimental system. TLFs have liquid IFs of different charge and properties and they offer the possibility to confine differently shaped ions and molecules to very small spaces, or to subject them to controlled forces. This makes the foam films a unique “device” available to obtain information about fluidic systems in nanometer dimensions. The main goal of this thesis was to study nanofluidic processes using TLFs as models, or tools, and to subtract information about natural systems plus deepen the understanding on physical chemical conditions. The presented work showed that foam films can be used as experimental models to understand the behavior of liquids in nano – sized confinement. In the first part of the thesis, we studied the process of thinning of thin liquid films stabilized with the non – ionic surfactant n – dodecyl – β – maltoside (β – C₁₂G₂) with primary interest in interfacial diffusion processes during the thinning process dependent on surfactant concentration 64. The surfactant concentration in the film forming solutions was varied at constant electrolyte (NaCl) concentration. The velocity of thinning was analyzed combining previously developed theoretical approaches. Qualitative information about the mobility of the surfactant molecules at the film surfaces was obtained. We found that above a certain limiting surfactant concentration the film surfaces were completely immobile and they behaved as non – deformable, which decelerated the thinning process. This follows the predictions for Reynolds flow of liquid between two non – deformable disks. In the second part of the thesis, we designed a TLF nanofluidic system containing rod – like multivalent ions and compared this system to films containing monovalent ions. We presented first results which recognized for the first time the existence of an additional attractive force in the foam films based on the electrostatic interaction between rod – like ions and oppositely charged surfaces. We may speculate that this is an ion bridging component of the disjoining pressure. The results show that for films prepared in presence of spermidine the transformation of the thicker CF to the thinnest NBF is more probable as films prepared with NaCl at similar conditions of electrostatic interaction. This effect is not a result of specific adsorption of any of the ions at the fluid surfaces and it does not lead to any changes in the equilibrium properties of the CF and NBF. Our hypothesis was proven using the trivalent ion Y3+ which does not show ion bridging. The experimental results are compared to theoretical predictions and a quantitative agreement on the system’s energy gain for the change from CF to NBF could be obtained. In the third part of the work, the behavior of nanoparticles in confinement was investigated with respect to their impact on the fluid flow velocity. The particles altered the flow velocity by an unexpected high amount, so that the resulting changes in the dynamic viscosity could not be explained by a realistic change of the fluid viscosity. Only aggregation, flocculation and plug formation can explain the experimental results. The particle systems in the presented thesis had a great impact on the film interfaces due to the stabilizer molecules present in the bulk solution. Finally, the location of the particles with respect to their lateral and vertical arrangement in the film was studied with advanced reflectivity and scattering methods. Neutron Reflectometry studies were performed to investigate the location of nanoparticles in the TLF perpendicular to the IF. For the first time, we study TLFs using grazing incidence small angle X – ray scattering (GISAXS), which is a technique sensitive to the lateral arrangement of particles in confined volumes. This work provides preliminary data on a lateral ordering of particles in the film.
Mit dem Heranwachsen der Nanotechnologie in den vergangenen zehn Jahren hat sich die Nanofluidik als Forschungsbereich etabliert und erfährt wachsende Aufmerksamkeit in der Wissenschaft, sowie auch in der Industrie. Im biomedizinischen Bereich, wo intrazelluläre Prozesse häufig komplexer, nanofluidischer Natur sind, wird sich vermehrt für ein detailliertes Verständnis von nanofluidischen Prozessen interessiert, z.B. für den Einfluss von Kolloiden verschiedenster Form oder elektrischer Ladung auf die Kanäle und auf das Fließverhalten oder die Auswirkungen der Einengung von Flüssigkeiten und Kolloiden in Nanometer Geometrien. In der vorliegenden Arbeit werden dünne flüssige Filme, hinsichtlich ihrer Funktion als nanofluidische Modelle untersucht. Im ersten Teil der Arbeit wurde die Fließgeschwindigkeit des Fluids aus dem dünnen Film, abhängig von der Konzentration der filmstabilisierenden Tensidmoleküle n – Dodecyl β – D – Maltoside ( β – C₁₂G₂) bei einer konstanten Elektrolytkonzentration von 0.2 mM NaCl untersucht. Mit einem theoretischen Modell konnte das Dünnungsverhalten nachgezeichnet werden. Es wurde eine kritische Tensidkonzentration gefunden, unter der die Oberflächen lateral mobil sind und über der sie sich wie fest verhalten. Dadurch konnten wir Aufschluss darüber erlangen, wie die Oberfläche des Films unter verschiedenen Bedingungen geschaffen ist, und das in Bezug zur Verteilungsdichte der Moleküle an den Oberflächen setzen. Im weiteren wurden komplexere, nanofluidische Systeme untersucht, wobei zum einen ~ 1 nm lange, stäbchenförmige, multivalent geladene Spermidin - Moleküle die punktförmigen Elektrolyte ersetzten. Es konnte eine deutliche Veränderung der Stabilität zwischen Filmen mit und ohne Stäbchen festgestellt werden. Die Filme, mit NaCl, blieben länger in dem metastabilen „Common Film“ (CF) Zustand als die Filme, die eine vergleichbare Konzentration von Spermidin Stäbchen beinhalteten. Die Ergebnisse deuteten auf eine zusätzliche Anziehungskraft durch Brückenbildung zwischen zwei geladenen Oberflächen durch gegensätzlich geladene Stäbchenförmige Moleküle hin. Es konnte gezeigt werden, dass dieser Effekt weder ein Ergebnis von spezifischer Ionenadsorption an die Filmoberfläche war, noch ein Unterschied in den Gleichgewichtszuständen von den Dicken der CFs und der Newton Black Films (NBFs) hervorrief, was auf die korrekte Annahme der Ionenstärke in der Lösung schließen ließ. Auch in Versuchen mit ebenfalls trivalenten Ionen YCl3 wurde festgestellt, dass kein vergleichbarer Überbrückungseffekt auftritt. Die Ergebnisse wurden mit theoretischen Simulationen verglichen und es wurde eine quantitative Übereinstimmung gefunden bezüglich der Größe des Systeminternen Energiegewinns durch den Überbrückungseffekt. Desweiteren wurde das Fließverhalten von Fluiden mit Kolloiden eingeengt in Nanometer Geometrien untersucht. Für zwei verschiedene Arten von Nanopartikeln (Fe3O4 stabilisiert mit Oleinsäure und polymerstabilisierte Goldpartikel) wurde eine Verlangsamung der Fließgeschwindigkeit festgestellt. Mit einem theoretischen Modell konnte das Fließverhalten nur für enorm erhöhte Viskositätswerte des Fluids erklärt werden. Die Viskositätserhöhung wurde mit Partikelaggregaten, die den Ausfluss behindern, erklärt und diskutiert, unter der Annahme eines nicht - Newtonischen Fließverhaltens der Dispersionen. Gleichermaßen wurde die strukturelle Anordnung der Partikel in den Filmen hinsichtlich ihrer vertikalen und lateralen Verteilung untersucht. In dieser Arbeit werden vorläufige Ergebnisse präsentiert, die noch weiteren Studien bedürfen. Mit Neutronenreflexion sollte die Anordnung der Partikel orthogonal zur Oberfläche im Film analysiert werden. Eine qualitative Analyse lässt schließen, dass bei einer höheren Konzentration von Partikeln in Lösung, sich auch eine erhöhte Konzentration von Partikeln im dünnen Film befindet. Leider konnten die Daten nicht hinsichtlich der Lage der Partikel analysiert werden. Zum ersten Mal wurden dünne flüssige Filme mit Kleinwinkelröntgenstreuung unter streifendem Einfall (GISAXS) analysiert. Mit Hilfe dieser Methode sollte eine laterale Anordnung der Partikel im Film untersucht werden. Es konnten erfolgreiche Messungen durchgeführt werden und mit Hilfe der rechnergestützten Analyse konnte eine Aussage gemacht werden, dass ~ 6 nm große Teilchen in ~ 43 nm Abstand sich im Film befinden. Die Aussage bezüglich der kleinen Teilchen könnte sich auf einzelne, kleinere Partikel beziehen, allerdings könnten auch die 43 nm eine relevante Strukturgröße darstellen, da es in der Dispersion gehäuft Aggregate mit dem Durchmesser in dem Größenbereich vorliegen. Zusammenfassend können sich mit dieser Arbeit die dünnen flüssigen Filme als eine wichtige Kernmethode der Untersuchung von nanofluidischen Prozessen, wie sie häufig in der Natur vorkommen, behaupten.

The piezoelectric and pyroelectric properties of oriented films possessing dipole moments are increasingly being used in pressure, acoustic, thermal and optical devices. The performance of these devices in many applications may be enhanced by thin-film technology.The developing Langmuir-Blodgett thin-film deposition technique offers the opportunity to obtain highly oriented and uniform organic-based films in the 10–5000 nm thickness range. Special techniques must be used, however, to assemble these molecules in such a way as to result in polar multilayer films. Several possible deposition techniques are investigated, with one resulting in a polar and pyroelectric film about 50 nm thick.

How does informal economic activity respond to increased financial inclusion? Does it become more entrepreneurial? Does access to new financing options change the gender configuration of informal economic activity and, if so, in what ways and what directions? We take advantage of nationwide data collected in 2010/11 and 2015/16 by India’s National Sample Survey Office on unorganized (informal) enterprises. This period was one of rapid expansion of banking availability aimed particularly at the unbanked, under-banked, and women. We find strong empirical evidence supporting the crucial role of financial access in promoting entrepreneurship among informal sector firms in India. Our results are robust to alternative specifications and alternative measures of financial constraints using an approach combining propensity score matching and difference-in-differences. However, we do not find conclusive evidence that increased financial inclusion leads to a higher likelihood of women becoming entrepreneurs than men in the informal sector.

Part art history essay, part experimental fiction, part theoretical manifesto on the politics of equivalence, Helicography examines questions of scale in relation to Robert Smithson’s iconic 1970 artwork Spiral Jetty. In an essay and film made to accompany the earthwork, Smithson invites us to imagine the stone helix of his structure at various orders of magnitude, from microscopic molecules to entire galaxies. Taking up this invitation with an unrelenting and literal enthusiasm, Helicography pursues the implications of such transformations all the way to the limits of logic. If other spirals, from the natural to the man-made, were expanded or condensed to the size of Spiral Jetty, what are the consequences of their physical metamorphoses? What other equivalences follow in turn, and where do their surprising historical, cultural, and mechanical connections lead? This book considers a number of forms in order to find out: the fluid vortices of whirlpools, hurricanes, and galaxies; the delicate shells of snails and the threatening pose of rattlesnakes; prehistoric ferns and the turns of the inner ear; the monstrous jaws of ancient sharks; a baroque finial scroll on a bass viol; a 19th-century watch spring; phonograph discs and spooled film; the largest open-pit mine on the planet. The result is a narrative laboratory for the “science of imaginary solutions” proposed by Alfred Jarry (whose King Ubu also plays a central role in the story told here), a work of fictocriticism blurring form and content, and the story of a single instant in time lost in the deserts of the intermountain west.

Using a firm-level panel dataset on private small- and medium-sized enterprises (SMEs) in Viet Nam’s manufacturing sector, this paper examines productivity dynamics of formal and informal firms. We decompose productivity changes into changes within and between formal and informal firms. We assess the contributions of firm entry and exit as well as informal–formal transitions. Our results show that productivity is considerably lower and misallocation more prevalent in the informal than in the formal sector. Yet, formalizing firms in Viet Nam make an important contribution to aggregate productivity growth among manufacturing SMEs, growing faster than other firms and increasing efficiency. We identify two ‘regimes’ of formalization. Until early 2010, more productive (previously) informal firms formalize. Policy changes and accelerated formalization then alter the characteristics of formalizers, as less productive firms become formal. While this formalization wave depresses average formal total factor productivity growth, the overall productivity effect is positive.

Sydney is most well known internationally for its spectacular harbour, opera house and stunning Northern Beaches. But the history of the city includes an avant-garde underbelly. In 1965 Sydney was a fertile breeding ground for counter-cultural, underground cinema. and one of Australia’s first avant-garde independent film-making, distribution and exhibition groups, ‘Ubu’, was established. The name, from Alfred Jarry’s 1896 subversive play, Ubu Roi, signposted their revolutionary aims. Ubu’s members experimented with film form and pushed the boundaries of cultural conventions. They screened their own work and that of international and local avant-garde film-makers like Bruce Connor, Norman McLaren, Paul Winkler and Dusan Marek. They lobbied against censorship and built a creative community. Numerous members later became important to the revival of the Australian feature film industry in the 1970s, including Bruce Beresford, Yoram Gross, Phillip Noyce and Peter Weir.

This chapter begins with Harry Dean and his half-brother Stanley McKnight Jr. going to their mother Ersel's funeral, both stoned on marijuana. Ersel's drinking, gambling and periodic disappearances had left both with bitter memories even though she and Harry Dean had reconciled before her death. Back in Los Angeles Harry Dean was living with actress Maggie Blye and rode through the New Hollywood wave with roles in key films such as Wise Blood (1979), Alien (1979), and The Rose (1979). As bogus preacher Asa Hawks in John Huston's Wise Blood he tapped into the hard-shell fundamentalism of his rural Kentucky roots. John Carpenter's Escape from New York (1981) and Christine (1983) introduced him to a new generation of fans as did The Rose (1979) and later Pretty in Pink (1986). On the set of Francis Ford Coppola's One from the Heart (1981) he met possibly the love of his life, Rebecca De Mornay, but she would later drop him for heartthrob Tom Cruise. It was his work with European directors Ulu Grosbard in Straight Time (1978) and Bertrand Tavernier in Death Watch (1980), however, that set the stage for the greatest roles of his career.

Raman spectroscopy (RS) [7, 8] It is used to examine the energy levels of molecules that cannot be well character-ized via infrared spectroscopy. Th e two techniques, however, are complimentary. In the RS, a sample is irradiated with a strong monochromatic light source (usu-ally a laser). Most of the radiation will scatter or "reflect off' the sample at the same energy as the incoming laser radiation. However, a small amount will scat-ter from the sample at a wavelength slightly shifted from the original wavelength. It is possible to study the molecular structure or determine the chemical identity of the sample. It is quite straightforward to identify compounds by spectral library search. Due to extensive library spectral information, the unique spectral finger-print of every compound, and the ease with which such analyses can be per-formed, the RS is a very useful technique for various applications. An important application of the RS is the rapid, nondestructive characterization of diamond, diamond-like, and amorphous-carbon films. 4.3.2.3. Scanning electron microscopy (SEM) / energy dispersive X-ra y analysis (EDX) [7, 8] The SEM produce s detailed photographs that provide important information about the surface structure and morphology of almost any kind of sample. Image analy-sis is often the first and most important step in problem solving and failure analy-sis. With SEM, a focused beam of high-energy electrons is scanned over the sur-face of a material, causing a variety of signals, secondary electrons, X-rays, photons, etc. - each of which may be used to characterize the material with re-spect to specific properties . The signals are used to modulate the brightness on a CRT display, thereb y providing a high-resolution map of the selected material property. It is a surface imaging technique, but with Energy Dispersive X-ray (EDX) it can identify elements in the near-surface region. This technique is most useful for imaging particles. 4.3.2.4. X-ray fluorescence (XRF) [7, 8] Incident X-rays are used to excite surface atoms. The atoms relax through the emission of an X-ray with energy characteristic of the parent atoms and the inten-sity proportional to the amount of the element present. It is a bulk or "total mate-rials" characterization technique for rapid, simultaneous, and nondestructive analysis of elements having an atomic number higher than that of boron. Tradi-tional bulk analysis applications include identifying metals and alloys, detecting trace elements in liquids, and identifying residues and deposits. 4.3.2.5. Total-reflection X-ray fluorescence (TXRF) [7, 8] It is a special XRF technique that provides extremely sensitive measures of the elements present in a material's outer surface. Applications include searching for metal contamination in thin films on silicon wafers and detecting picogram-levels o f arsenic, lead, mercury and cadmium on hazardous, chemical fume hoods." In this paper, a wafer-level packaging solution for pressure sensor microelectromechanical system (MEMS) is reported. Sensor and glass cap wafers are anodically bonded at a bonding temperature less than 400°C. Bubble free interfaces are obtained and the bond strength is higher than 20 MPa. Sensor and bottom silicon cap wafers are bonded at a temperature of 400–450°C with the assistance of a gold intermediate layer. The bond strenght is higher than 5 MPa. The via holes, used for feedthroughs leading out the circuit, on bottom silicon cap wafer are anisotropically formed in KOH etching solution. Aluminum layer is sputtered on the bottom silicon wafer for electrical connection, re-routing circuit and the seed layer of under bump metallization (UBM). During sputtering process, the sidewalls of via holes are also sputtered with aluminum film. At the same time, the metal pads on sensor wafer are also built up to connect with metallized via holes. It is found that the cavities are vacuum sealed. Sputtered Cr/Ni/Au layers are used for UBM layers. Finally, solder bumps can printed or plated on the UBM. The whole process leads to promising performance of the devices.

Tungsten-doped diamond-like carbon (DLC) coatings have been magnetron sputtered onto 52100 steel with chromium and chromium/tungsten carbide dual interlayers using a Hauzer Techno Coating HTC 1200 4 UBM unbalanced magnetron deposition system. Internal fixturing to the deposition chamber rotates parts to be coated with a two degree of freedom system. By design, at certain intervals during the deposition, the acetylene flow is linearly altered to change film characteristics throughout the film. AES sputter depth profiling analysis shows that the fixture rotational system, designed to uniformly coat parts, causes localized chemistry variations in the coating. For a given location, the AES depth profile also clearly documents the intervals when acetylene flow was constant and when the flow was ramped.

Computations of film cooling are presented based on the geometry of a UBC experimental turbine blade model. This model has a semi-circlar leading edge with four rows of laterally-inclined film cooling orifices positioned symmetrically about the stagnation line. The computational domain follows the physical domain and includes the curved blade surface as well as the coolant regions in the circular coolant orifices. The injection orifices are inclined spanwise at 30° to the blade surface. A multi-zone curvilinear grid is used to simulate the complex configuration. Grids are generated by a block-structured elliptic grid generation method which represents exactly the curved blade surface as well as the circular injection orifices. Computations over the cooled turbine blade model are carried out for overall mass flow ratios of 0.52 and 0.97. The relative mass flow ratios from each orifice are specified to match experimental values. Density ratios of coolant to free stream were taken to be unity (constant density). Comparison of predicted film cooling effectiveness with experimental data showed reasonable agreement.

Abstract New heterogeneous 3D integration schemes and continuing miniaturization of semiconductor packaging components, such as micropillars, are driving demand for substantive changes to conventional PFA (physical failure analysis). In particular, desired performance capabilities include the ability to nondestructively determine failures within seconds to minutes. New tools should be quantitative, have sufficient resolution to determine sub-micron sized defects and voids in TSVs at the wafer or package level. It should also measure thickness and their material composition of multilayer structures above the wafer surface, such as microbumps, or those below the surface including UBM and RDL. In this paper we are introducing a novel x-ray fluorescence microscope technique capable of solving the above applications in advanced packaging for PFA and process development. The same technique can also be applied in the front end metrology of new gate materials, 3D FinFET structures within test structures in patterned wafers. Characterization of sub nanoscopic changes (sensitivity of sub-angstrom) in film and dopants deposited in 3D structures will also be shown. With its high sensitivity for trace materials, contamination analysis of post hard mask residue, post metal etch residue especially in high aspect ratio structures is also possible.
