Les légumineuses sont capables d'établir une interaction symbiotique avec des bactéries du sol, collectivement appelées rhizobia. Les facteurs Nod (NFs), molécules produites par le symbiote, sont indispensables à l'établissement de cette symbiose. Les études génétiques chez M. Truncatula ont révélé l'implication de deux récepteurs kinase de la famille des LysM-RLKs, NFP et LYK3, dans la perception des NFs. NFP est indispensable à toutes les réponses induites chez l'hôte par les NFs alors que LYK3 contrôle spécifiquement l'infection. Afin de mieux comprendre les mécanismes de signalisation encore mal connus en aval de LYK3, deux criblages de banques d'ADNc de M. Truncatula dans deux systèmes double-hybride ont été entrepris dans le but d'identifier des partenaires de ce récepteur. Un de ces criblages a permis l'identification d'un gène codant pour une E3 ubiquitine ligase de la famille des U-Box et nommé MtPUB1. MtPUB1 joue un rôle négatif dans les processus d'infection et de nodulation. Comme pour LYK3, ce rôle dépend de la nature des NFs produits par le symbiote. En parallèle, il a été montré par une approche ciblée que LYK3, NFP et DMI2 peuvent interagir avec une remorine, protéine membranaire contrôlant positivement l'infection chez M. Truncatula
Leguminous plants can establish symbiotic interaction with nitrogen fixing soil-born bacteria collectively referred as rhizobia. Nod factors (NFs) are rhizobia produced molecules essential to the establishment of this interaction. Genetic studies of the NFs perception in M. Truncatula led to the identification of two LysM receptor-like kinases, NFP and LYK3. NFP is necessary for all NFs induced responses while LYK3 specifically controls infection. Signalling events downstream LYK3 are poorly understood. To decipher this signalling pathway, two different yeast two-hybrid screens using M. Truncatula cDNAs and LYK3 kinase as bait were performed. One screen identified an E3 ubiquitin ligase of the U-Box family renamed MtPUB1. MtPUB1 plays a negative role in infection and nodulation, and as for LYK3, this role relies on the NFs structure produced by the rhizobia. In parallel, a second approach based on pairwise interaction assays identified a remorin protein as partner of all three symbiotic receptor-like kinases, NFP, LYK3 and DMI2

L’objectif de ce travail a été de développer deux approches analytiques dédiées à l’analyse toxicologique des nouveaux produits de synthèse (NPS) dans différentes matrices biologiques (sang, urine et cheveux). La première est basée sur le criblage non ciblé par chimiluminescence sur biopuces et chromatographie liquide couplée à la spectrométrie de masse haute résolution (LC-HRMS) et la deuxième correspond à un criblage ciblé par spectrométrie de masse en tandem (LC-MS/MS). Ces deux approches ont ensuite été appliquées dans des études observationnelles pour évaluer la consommation de NPS dans des populations à risques de surdosage, de pharmacodépendance ou de soumission chimique dans un contexte clinique ou médico-judiciaire.La dernière partie a été consacrée au développement d’un nouvel outil analytique de traitement des données issues de la LC-HRMS qui a permis d’étudier le métabolisme de 9 NPS in vitro sur des cultures de microsomes du foie humain (HLM) et in vivo sur des échantillons biologiques d’usagers de ces drogues. Cette dernière approche a permis la création d’une bibliothèque de spectres de haute résolution composée de 228 métabolites dont certains ont été proposés comme marqueurs pertinents d’exposition aux NPS dont ils sont issus.Ce travail a été concrétisé par la rédaction de 10 publications scientifiques et a permis d’initier plusieurs collaborations pluridisciplinaires
The aim of the present work was to develop two analytical approaches dedicated to the analysis of new psychoactive substances in different biological matrices (blood, urine and hair). The first approach is based on untargeted screening by both biochip array technology chemiluminescence assay and liquid chromatography coupled to high resolution mass spectrometry (LC-HRMS) and the second corresponds to a targeted screening by liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS). These two approaches were then applied in observational studies to assess the consumption of NPS in high risk populations (overdose, drug abuse, drug facilitated crimes) in clinical and forensic settings. The last part of the work was devoted to the development of a new analytical tool for LC-HRMS data processing which made it possible to study the metabolism of 9 NPS In vitro on human liver microsomes (HLM) and In vivo in biological samples from drug users. This approach has enabled the creation of HRMS spectral library containing 228 metabolites, some of which have been proposed as relevant markers of NPS exposure.This work has resulted on 10 scientific publications and allowed to initiate many multidisciplinary collaborations

Hepatitis C virus (HCV), a positive-sense RNA virus that chronically infects between 2.7 and 3.9 million Americans, is highly mutational, making the HCV infection difficult to treat. Thus, it is of high interest to search for highly conserved therapeutic targets within the HCV genome. Two such sequences are located within the 5' untranslated region (UTR) of HCV, being complementary for the microRNA miR-122, a liver microRNA essential for the production of the infectious virus. The use of peptide nucleic acids (PNAs) as therapeutic agents has become a promising area of study in recent years. In this study, we characterized the interactions between miR-122 and the HCV 5'UTR and designed PNAs to disrupt these interactions and thus, inhibit RNA replication and translation. Our results show that the PNAs effectively disrupt the interactions involving miR-122 and the 5'UTR, thereby increasing the possibility of a new therapeutic option against HCV.
