This work presents an experimental evaluation of various voice transformation techniques based on GMM models. These linear transforms, despite their quality obtained, they fail to some defects specially the oversmoothing effect, the problem of spectral distortion and the overfitting. In a first part, we proposed taking these issues into account to adapt the learning strategy of the conversion functions. The first main idea is to reduce the number of parameters describing the conversion function. The second idea considers the solutions based on linear transform are unstable face to the lack of the training data, hence the recourse to non-linear transform model like RBF. In a second part in some situations, we need to align non-parallel data from the source and target speakers, one solution consists to use a recursive representation of binary tree, whose depth depends on the learning data size. In the last part, to get a high voice quality, we have proposed a model of parameters issued from the PCA projection on the true envelope before applying the conversion methods

The goal of learning transfer is to apply knowledge gained from one problem to a separate related problem. Transformation learning is a proposed approach to computational learning transfer that focuses on modeling high-level transformations that are well suited for transfer. By using a high-level representation of transferable data, transformation learning facilitates both shallow transfer (intra-domain) and deep transfer (inter-domain) scenarios. Transformations can be discovered in data using manifold learning to order data instances according to the transformations they represent. For high-dimensional data representable with coordinate systems, such as images and sounds, data instances can be decomposed into small sub-instances based on coordinates. Coordinate-based transformation models trained using these sub-instances can effectively approximate transformations from very small amounts of input data compared to the naive transformation modeling approach. In addition, these models are well suited for deep transfer scenarios.

Resumo:
We investigate program optimization and program adaptation (or specialization) by the transformation of (constructive) synthesis proofs. Synthesis proofs which yield inefficient programs are transformed into analogous proofs which yield more efficient programs. These proofs are based on a Martin-Lof type theory logic and proved within the OYSTER proof refinement system [Martin-Lof 82]. The problems of automated program synthesis and verification have already been addressed within the proofs as programs paradigm [Constable et al. 86], [Bundy et al. 91]. By using constructive logic, the task of generating programs is treated as the task of proving a theorem. By performing a proof of a formal specification expressed in constructive logic, stating the input-output conditions of the desired program, an algorithm can be routinely extracted from the proof. We have implemented a system - the meta-level OYSTER proof transformation system (MOPTS) - for optimizing programs through the transformation of (OYSTER) synthesis proofs. The MOPTS has the desirable properties of automatability, correctness and various mechanisms for reducing the transformation search space, and various control mechanisms for guiding search through that space. A contribution afforded by proof transformations is that, in addition to program synthesis and verification, the problem of program transformation is also tackled by transposing the task to the proofs as programs paradigm. As with synthesis and verification, knowledge of theorem proving, and in particular automatic proof guidance techniques, can be brought to bear on the task. Furthermore, such transformations allow the human synthesizer to produce an elegant source proof, without clouding the theorem providing process with efficiency issues, and then to transform this into an opaque proof that yields an efficient target program. To accomplish program transformation through proof transformation, we have successfully, and for the first time, adapted a range of program transformation techniques to the proofs as program paradigm, notably: the tupling technique for 'merging' repeated (sub)computations, [Petrossi] [Chin 90], and the fold/unfold technique for transforming inefficient functional programs into equivalent, more efficient, functional programs by a process of unfolding and folding definitions [Darl 81]. Throughout the course of this thesis we shall highlight the benefits of our 'proofs as programs' approach to transformation, particularly with respect to search, correctness and automatability.

The draft of the Socio-economic Theory of Organizations allows to stamp the notions of transformation and change. This perspective allows to promote transformation within organizations with an autonomous theoretical approach, of socio-economic essence, in break with a comprehension of transformation as a form of change. Transformation est an endogenous and immanent socio-economic process. His process takes place in the facilities of the organization. It makes happen the change, all forms of which he inserts. Since then, the success of change, and more broadly any strategical or organizational implementation, must be leaned on the improvement of quality and effectiveness of functioning and management of the organization. To achieve change, the organization must develop its capacity permanently to be transformed. It assumes two prior conditions, on one hand to develop the capacity of study of transformation, on the other hand the permanente activation of process of me transformation. These three conditions can be united if the organization defines and implements a strategy of piloting of transformation. The strategy of transformation distinguishes itself from the strategy of change, because it consists in intervening in the facilities of functioning and management of the organization. Sur methodological plan, research implements the method of Research-intervention, said "Socio-economic Intervention", developed by the Institute of Socio-Economie from Firms and from Organizations (H. SAVALL and V. ZARDET, ISEOR, FRANCE). This method allows to conceive and to establish a strategy of transformation in the process of the common management of the organization and its functioning. Research shows that this fourth condition must be united to achieve change

This thesis employs phenomenological hermeneutic circle analysis, to investigate structure versus agency and adult identity change in lifelong learning. Achieving transformed agency and enhanced identity is argued to be about other ways of doing and other ways of seeing (Mezirow 2000:21). It proceeds by exploring if curricula employed in education can provide structure and/or the catalyst which allows ‘other’ to be revealed, agency to be regained, and to explicate what contribution curricula might make in transforming adult identities. Drawing upon the disciplines of sociology and psychology it provides holistic interpretations of participant accounts in the contemporary competitive world and explores the interstices in the duality of tensions between the utilitarian, and pragmatic adult, who employs education as a developmental pathway of choice. Interviews with six participants tell individual stories to provide holistic data of their erudition and experiences of cognitive and social change. Data are then employed to essentialise similarities, differences, themes, and congruent essences, and to distil factors which exemplify growth in understanding and expectations of the self. Growth in self-assurance and identity change capability is then contrasted with the fragility of adult identity; whereupon, this thesis critically positions fragility causation amongst the instrumental policies and forces of lifelong learning. Mezirow contends that agency is achieved by elaborating existing frames, learning new frames, transforming habits, and transforming points of view. This thesis moves to discuss the connected nature of these developmental factors and ‘glass ceilings’, and how immanent personal potential is (re) revealed to the adult self. Employing an archaeological hermeneutic research tool which suggests reflection is a central and developing feature in adult’s educational development the thesis finally contends that education is important in the personal delivery of agency over structure, and that curricula of any structurally legitimate form make a significant contribution to allowing persons to both flourish and confront a range of ‘other’ life circumstances and dilemmas.

Resumo:
Consumers are more and more interested in experiences cultivating a greater sense of purpose and meaningful relationships. In this context, there is a need for conscious and transformative holidays that provide means to change lifestyles, and to increase the positive impact made on destinations. However, we do not yet have a precise understanding of tourist transformation nor a conceptual model of how transformative tourism experiences occur. To address the consumers’ demand for meaningful and refreshing travel, research is needed to understand the dimensions of transformative tourism experiences and to define tourist transformation. To theoretically and empirically analyse transformative tourism experiences and how they are characterised, this thesis presents three studies. The first study of the thesis reviews the contexts in which transformative tourism research has emerged, and the main theories that have been employed to understand the phenomenon. Transformative learning and existential transformation were the two major conceptualisations found to explain changes in tourists. By adopting a multi-disciplinary approach, a range of transformative stimuli occurring across the tourism experience, namely peak episodes, disorienting dilemmas and physical performances, were identified and discussed. After a critical review, the first study proposes a tourist transformation conceptual model, and defines transformation as triggered by a combination of disruptive stimuli experienced throughout the holiday, which change the tourists’ value system and attitudes to create new meanings. Building on the comprehensive conceptual framework of tourist transformation proposed in the Conceptual Model, Study II explores the meanings attached to transformative travel by individuals as tourists, and investigates characteristics of their experiences that may facilitate or inhibit tourist transformation. Adopting a qualitative semi-structured interview approach, it revealed that interviewees viewed subjective tourist transformation as achieving greater self-efficacy, humility and personal enrichment. Study II findings suggest that transformation facilitators correspond to: interacting with locals and travellers, facing challenges, experiencing the sense of the place, long stays and post-travel reflection; while several aspects emerged as transformation inhibitors, such as short stays, repeated activities, and the lack of access to the residents’ lifestyle. When transformation emerged, it was perceived as permanent. Building on the findings of Study II, Study III further analyses gendered and subjective experiences of young travellers, who represent a crucial transition phase that has not yet been addressed within transformative tourism research. In the realm of transformative tourism research and tourism studies on gender, transformation is considered part of the empowering process that women mostly experience through travel. Yet, little is known about the same process from a male or comparative perspective. To address these research gaps, Study III adopts a double duoethnography approach aimed at analysing young male and female experiences of transformation through tourism. Data was collected from the candidate and another researcher’ personal experiences with transformative tourism, then analysed for gender-based similarities and differences. Overall, Study III findings suggest that both young female and male tourism experiences of transformation are characterised by becoming mentally stronger and more confident, as well as by exploring their identities during tourism experiences. The female process of transformation is marked by bodily feelings and self-consciousness, while the male experience is characterised by flow, adaptation, a sense of community with other travellers. The findings of the three studies composing the thesis advance knowledge in transformative tourism, and present important implications for operators and managers.

This thesis examines the socio-economic transformation processes in Poland and Romania and aims to embed them in broader structural and historical context of changes. The main argument is that transformation processes in the states of Central and Eastern Europe are constituted by the global processes of change within a social totality. The study has three main objectives. First, it aims to provide a theoretical framework challenging the mainstream approaches methodologically/ontologically to point out to their limits and account for the dialectical relationship between the global and the internal. Second, to develop an account of the international context surrounding the transformation processes to highlight the nature of the global and hence to emphasise the unity of transformation and integration processes. As such, it critically interprets the social purpose of the international institutions and the European Union involvement in the policy-making of the states in the region through the changing techniques of monitoring, reporting and the process of negotiations. Third, to provide an analysis of the transformation processes in Poland and Romania as processes of the internationalisation of the state which would effectively help in examining the constitutive role of the global in a dialectical relationship with the national level dynamics and changes. Integration with the global economy as well as Euro-Atlantic institutions was an integral element of the neo-liberal restructuring in Poland and Romania. Socio-economic transformation in the region with added dimensions of conditionality had important social consequences, thereby resulting in new forms of state-society relations.

This thesis aims to examine the cultural effect of transformation through the lens of procedural techniques applied to the human body and architecture. The body and architecture both operate as landscapes of transformation. Technological advancement has increasingly enabled cosmetic surgery and contemporary architectural techniques to encourage a cultural aesthetic of transformation. In both the human body and architecture, transformation offers a physical and psychological effect. As a cultural enterprise, how can architecture both benefit and contribute by engaging the cultural caprice of transformation? How can architectural procedural techniques define an urban project which engages the consumption of transformation? This thesis proposes a response to the cultural effect of transformation and is developed through the architectural process of testing procedural techniques. The thesis also responds to the notion of gradient as a residual witness of transformation, revealing the techniques by which the transformation was brought about. The first part of this thesis engages the idea that the human body is a landscape of transformation as cosmetic procedures continue to redefine the operation of its interiority. The second part of the thesis presents Central Park as a mechanical artifice and an operative site for the procedural techniques of transformation.

Abstract This study analyzes an attempt at integration of a pre-school class, a leisure-time center and an elementary school in Sweden. The integration was organized in the form of Vertical Track which implies a successive development of groups comprising children between six and twelve years old, pre-school teachers, recreation pedagogues, and schoolteachers. The integration was prompted by state governed reforms such as the 1992 law allowing six-year olds to start compulsory school. The study is based on cultural-historical activity theory and was carried out as participant observation and action research. The study addresses the question of the potentials and alternative goals for change and development of the present school pedagogy and classroom practice that integration implies. Special attention has been paid to what tools might potentially mediate in processes of integration. A research and educational program, the 5thD, was jointly created between researchers and teachers and located in a Vertical Track. The capacity of this complex tool as a mediator in the multicultural Vertical Track structure was explored. It is argued that the Vertical Track as an instantiation of the integration reform represents an arena for potential expansive transformation. However, in order for integration to have an impact on the pedagogical practice in schools, teacher interactions need to be mediated by communicative and conceptual tools. It is suggested that the 5thD program is an example of such tools. Keywords: Integration, pre-school teacher, recreation pedagogue, schoolteacher, contradiction, expansive learning, mediation, and tool.

Recent political events in South Africa have emphasized the importance of faculty diversity. Very little research has considered why it is the case that 20 years after the end of Apartheid, only 14% of professors are black. Or, why the University of Cape Town does not have a single black South African woman who is a full professor. Is it the case that black faculty are discriminated against during the hiring process or is it the case that black faculty depart at significantly higher rates than white faculty as sometimes suggested? Further, how do race, education and institutional factors interact in determining diversity levels? Survival analysis methods coupled with a novel data-set consisting of detailed administrative employee records, proxies for performance and various socio-economic variables are employed to test various hypotheses related to these questions. The findings suggest that the level of diversity is of secondary importance to an employee's race when determining the likelihood of survival at the University of Cape Town. In addition, findings suggest that the university is performing far better in terms of gender equality than racial equality. Overall, race matters, not only through a direct correlation between employee race and tenure length, but also through indirect effects where employees who differ significantly from others in their respective faculty (in terms of race, tenure, age, education and gender) face increased rates of departure from the work place.

Artist Statement My art has a common thread of transformation and renewal. It is often my response to the many different changing cycles in life, whether seasonal, emotional, or the process of aging. In addition, the spiritual connections to my faith and family are embedded into many of the works which tell a story of my life and my ever searching soul. Revelations of myself over time and the transformation of thoughts are visually exposed. However, they are revealed with a keen eye and hand which are meant to reach out to the viewer, so they may be transformed and discover something unique within themselves. My foundation in graphic design is clearly evident in my artwork. Fundamental design elements such as color, balance, movement, and texture are grounded into my sculptures and paintings. Similar imagery and design elements reappear throughout my work. Red, orange, copper, and gold recur in much of the work, often balanced with the cooler hues of blue and green. The emotionality of the warm palette reflects my Irish temperament and my identity as a redhead. My peaceful and calm nature can be seen in the cool colors. Sometimes, color is absent and only white is used which reflects periods where emotions are suspended and apprehension is present.

Traditions that shape our identity and beliefs are constantly undergoing transformation through our interactions with new cultures. At the same time, these interactions enhance our awareness of where we come from, and where we truly belong. In this thesis I investigate the history of jewellery in India through a contemporary perspective and interpretation. My project narrates my transformative journey from the rich tapestry of Indian traditions towards the contemporary western culture. The subject of Indian jewellery is vast and complex, the enquiry is just the beginning of the journey. Therefore, my goal is to discover linkages between the age-old traditions and the fast-changing world of today. I create a dialogue between traditional, ritualistic Indian jewellery and an irreverent choice of materials and methods that have been conceived from my experiences in the West. Through this dialog, I explore the push and pull between traditional ethos and modern sensibilities, and the transformation it engenders.

The subject of this thesis is the nature of Christian transformation as understood through the process of discipleship in the Wesleyan tradition and United Methodism in particular. A basic premise is that contemporary discipleship efforts are perceived as ineffective in spite of the numerous strategies that exist within the field of Christian education. The contention of this thesis is that the current situation is rooted in a failure to address the holistic and integrated nature of Christian transformation, which from a Wesleyan perspective is understood as the process of sanctification. This thesis explores a more holistic vision for discipleship, drawing upon methodology proposed by Richard Osmer to do theological reflection as it engages Wesleyan theology and transformative learning theory (TLT), a contemporary adult educational theory. The result is a contribution to the field of Christian education that has implications for disciple making ministries in the local congregation. Alasdair MacIntyre's theory of practice is developed as a means of accounting for the present incoherence within discipleship ministries, and to resource the development of a more holistic approach to the process of Wesleyan discipleship. As a result, discipleship is conceived of as a single complex practice comprised of four inseparably related and integrally connected dimensions: virtues, disciplines, ethos and telos. The theoretical framework also provides insight into contemporary discipleship efforts by systematically isolating each component and investigating the particular emphasis that is stressed, thus truncating the practice of discipleship. Putting this framework into conversation with TLT provides a way for theological reflection that can broker a cross-disciplinary dialogue between TLT and Wesleyan discipleship. The resulting discourse discerns which relevant aspects of TLT can be appropriated within a Wesleyan context and how TLT contributes to the field of Christian education. Contributions that Wesleyan discipleship can make to the field of TLT are also explored. The thesis develops an educational theory that views discipleship as a single coherent complex practice that is consistent with the process of sanctification in the Wesleyan tradition. Such a theory overcomes the current situation that results in isolating various discipleship efforts by prompting the field of Christian education to consider discipleship as sanctification that transforms persons and their contexts in holistic ways.

The chief aims of this practice-based research are to investigate the nature of attachment between person and object in respect to transformation; to find ways in which objects (jewellery) can be transformed to engender a wearer’s emotional attachment; and to demonstrate the different ways of applying such transformations that are pertinent to jewellery. This research has developed a certain type of transformation, which slowly makes traces on the surface of jewellery over time, and is used as an effective way to engender a wearer’s emotional response. The research is situated in the field of contemporary jewellery, and is specifically related to emotion and sentiment; a category of jewellery that relates to the wearer’s emotional feeling. The scope of the study extends to research on emotional design since this research focuses on user experiences in developing emotional relationships with the object and on how the designer/maker can promote the formation of such an attachment to the object. Two practical experiments have been conducted in this research to determine and construct appropriate and effective characteristics of transformation that engender an emotional relationship between the jewellery and its wearer, through the examination of the transformational character of emotional objects and the interactions that people have with transformational jewellery. These two experiments involve the process of making in order to provide a way of thinking through the hand manipulating a material. The use of this material thinking, develops a more broader understanding of the relationship between the transformational object and emotional attachment. Towards the end of the research, a definition of transformational jewellery is constructed that identifies its four important elements. It also provides two sets of practice work that demonstrate the findings and that facilitate the communication of the author’s tacit knowledge gained from the experiential knowledge. This research expands the field of contemporary jewellery by involving studies of emotional design and applying the element of transformation to create an emotional relationship between jewellery and its wearer. This specific transformation, which has been identified in both text and practical works, constitute the main contribution to knowledge in the field of contemporary jewellery.

Background: Research in digital transformation has previously focused on understanding top management and operational roles but the role of middle management has rarely been examined. This despite several studies showing that the middle manager has a central role in change processes. A few studies have shed light on how the middle manager influence and contribute to digital transformation, however, there are few empirical studies that examine how the middle manager works to drive change in digital transformation. Purpose: The purpose of this study is to create a deeper understanding about middle management’s role in digital transformation.  Methodology: This is a qualitative case study of a company in household services. It is based on five semi-structured interviews and a literature review. The study has adopted an abductive approach. Conclusion: The study shows that the middle manager acts as a connecting link between management and operational level. The results show that middle managers drive change in digital transformation by identifying problems and business opportunities, initiating change, interpreting and translating management language for employees, promoting and raising ideas from the operational activities and following up and modifying change processes to achieve better results. They have the opportunity to do this because of their intermediate position in the organization.

Model Driven Engineering (MDE) considers models as first class artifacts. Each model conforms to another model, called its metamodel which defines its abstract syntax and its semantics.Various kinds of models are handled successively in an MDE development cycle. They are manipulated using, among others, programs called model transformations. A transformation takes as input a model in a source language and produces a model in a target language. The developers of a transformation must have a strong knowledge about the source and target metamodels which are involved and about the model transformation language. This makes the writing of the model transformation difficult. In this thesis, we address the problem of assisting the writing of a model transformation and more generally of understanding how a transformation operates. We adhere to the Model Transformation By example (MTBE) approach, which proposes to create a model transformation using examples of transformation. MTBE allows us to use the concrete syntaxes defined for the metamodels. Hence, the developers do not need in-depth knowledge about the metamodels. In this context, our thesis proposes two contributions. As a first contribution, we define a method to generate operational transformation rules from transformation examples. We extend a previous approach which uses Relational Concept Analysis as a learning technique for obtaining transformation patterns from 1-1 mapping between models. We develop a technique for extracting relevant transformation rules from these transformation patterns and we use JESS language and engine to make the rules executable. We also study how we better learn transformation rules from examples, using transformation examples separately or by gathering all the examples. The second contribution consists in recovering transformation traces from transformation examples. This trace recovery is useful for several purposes as locating bugs during the execution of transformation programs, or checking the coverage of all input models by a transformation. In our context, we expect also that this trace will provide data for a future model transformation learning technique. We first address the trace recovery problem with examples coming from a transformation program. We propose an approach, based on a multi-objective meta-heuristic, to generate the textit{many-to-many} mapping between model constructs which correspond to a trace. The fitness functions rely on the lexical and structure similarity between the constructs. We also refine the approach to apply it to the more general problem of model matching

Pflug, Nicholas Craig. "Identification of bioactive products from environmental transformation of steroids". Diss., University of Iowa, 2017. https://ir.uiowa.edu/etd/5979.

Texto completo da fonte
Resumo:
For bioactive chemical classes, it is often assumed that environmental transformation eliminates associated ecosystem risks. However, for endocrine-active steroid hormones, modest changes in structure can have a significant influence on biological activity and thus, subtle environmental transformations can yield products with conserved, enhanced, or activity across different biological endpoints. The aim of this work was to explore the environmental fate of high potency, endocrine-active steroid hormones during natural or engineered water processes in order to test the hypothesis that steroid transformation products generated during these processes are likely to contribute to residual bioactivity often reported in water resources. Specifically, laboratory experiments were used to simulate chemical disinfection (e.g., chlorination) or natural processes (e.g., photolysis) to: (i) determine the rate and extent of steroid transformation, (ii) isolate and identify products that are formed, and (iii) evaluate products or product mixtures for biological activity. These experimental results can be used to help guide occurrence studies for any products of concern in the environment and also guide computational predictions or rationalizations of chemical reactivity. Ultimately, the goal is to expand upon our awareness and understanding of how these potent endocrine ligands behave in the environment and how they potentially affect ecosystem health. Chapter 2 discusses the reaction of glucocorticoids (GC)s with aqueous chlorine (effectively, HOCl) to simulate their fate during engineered drinking water and wastewater chemical disinfection. Numerous transformation pathways were unveiled, including interconversion of GCs (e.g., endogenous cortisol to synthetic prednisolone), production of known androgens in the adrenosterone class, and chlorination of GCs (e.g., formation of 9-chloro-prednisone). We also showed that other advanced processes (e.g., oxidation via ozonation) result in more complete degradation of such pollutants, and may be better alternatives to chemical disinfection at eliminating bioactive steroidal product formation. In Chapter 3, results of the direct photolysis of dienogest (DNG), a widely prescribed oral contraceptive agent, are presented to simulate its fate in natural sunlit surface waters and engineered photochemical treatment systems (e.g., UV disinfection systems). The major products (~ 80% of the converted mass in neutral aqueous solutions) were identified to be photohydrates resulting from photochemical-induced incorporation of water into parent DNG. These products were found to be prone to dehydration in the dark, and thus, a source of substantial DNG regeneration (~ 65% after 72 h in neutral solutions). Other minor, non-revertible products were also identified, including two known estrogens. Although minor in initial yield, these estrogens are likely to accumulate over time through repeated cycling between DNG and its photohydrates, and thus, dominate DNG long-term fate. It was also found that DNG undergoes an unusual photochemical rearrangement to produce a minor product with a novel tetracyclic ring system--the subject of Chapter 4. Further, the generality of this unique photorearrangement process was explored through extension to the photolysis of two other dienone pharmaceutical steroids (e.g., the androgens methyldienolone and dienedione). Surprisingly, despite the significant change in core steroidal structure, the rearrangement products retain some progesterone receptor (PR) and androgen receptor (AR) bioactivity (i.e., low-µM to sub-nm EC50 values). Again, these represent other non-revertible, minor products that are likely to accumulate over time, with likely adverse ecological consequences. Chapter 5 covers results arising from the direct photolysis of trenbolone acetate (TBA) metabolites in the presence of model nucleophiles (e.g., sodium azide, sodium thiosulfate, ammonium hydroxide, hydroxylamine, and humic acid), some of which would be expected to be present, along with TBA metabolites, in agriculturally-impacted water resources. Previous studies by our group revealed that TBA metabolites undergo photohydration-thermal dehydration cycling, like that described above for DNG photolysis. The objective of this study was to determine if other nucleophiles would outcompete water for photochemical incorporation across the TBA metabolite extended conjugation system. It was found that TBA metabolite photolysis results in photochemical (and at times thermal) addition of the nucleophile to the TBA metabolites. It was also found that the addition products undergo thermal elimination in the dark and contribute to TBA metabolite regeneration, and therefore, are expected to increase TBA metabolite persistence in water resources. Finally, Chapter 6 discusses the reactions of various trienone and dienone steroids with aqueous chlorine to simulate their fate during engineered drinking water and wastewater chemical disinfection. Single-step transformation pathways were unveiled for each steroid class, including 4-chlorination (trienones) and 9,10-epoxidation (dienones). Chlorination at position C-4 is known to enhance anabolic potency of androgenic steroids and the 9,10-epoxy products were found to undergo acid- or base-catalyzed ring-opening and aromatization to yield known estrogenic products. In addition, Chapter 7 provides conclusions and future directions, while Chapter 8 details the experimental methods and procedures used throughout this thesis. Collectively, the results presented herein confirm our overall hypothesis that steroid transformation products would be expected to contribute to residual biological activity often detected in water resources. Furthermore, the results indicate that the transformation of high potency pharmaceuticals does not automatically equate with reduction or elimination of hazards to exposed organisms, especially in cases where such compounds have potential to form products exhibiting diverse biological endpoints. More holistic approaches to risk assessment of such high potency environmental contaminants are needed in order to accurately assess the fate and effects of such emerging pollutant classes and their bioactive transformation products.

Bag&rsquo
settlements have been a part of dynamic spatial systems, which seasonally depend on and reciprocally interact with Anatolian towns throughout centuries. This thesis presents the transforming setting of &lsquo
bag&rsquo
settlements related to changing values and meanings through an ontological assessment. Therefore, the thesis assumes that the main values of spatial organization, farmland pattern, ecological formation, settlement character, socio-cultural structure and lifestyle of the inhabitants constitute the unique entity of &lsquo
bag&rsquo
settlements. In time, the pressures of changing socio-economic conditions have destroyed the interaction between Anatolian towns and &lsquo
bag&rsquo
settlements. The thesis explains the changing role of &lsquo
bag&rsquo
settlements, changing and conflicting land uses and the loss of unique &lsquo
bag&rsquo
character in the case of Mugla-Karabaglar. Karabaglar is a &lsquo
bag&rsquo
settlement in the southwestern Turkey, where Mugla town residents live seasonally. It is a third grade natural site, the unique character, the natural and cultural assets and the lifestyle of which must be preserved. However, with transformation of the main values, meanings and practices, Karabaglar could no longer perpetuated its initial existence of being. Karabaglar is significant for two reasons: it has natural and cultural beings that need to be conserved, and as it still goes through a transformation process. The master&rsquo
s thesis of the author evaluated conservation plan of Karabaglar. Differently, this doctoral thesis evaluates the dynamics of the spatio-temporal transformation process in Karabaglar. It presents the changing role and significance of Karabaglar within the town-country continuum. This situation brings forward the conservation problematic of the character, landscape and uniqueness of Karabaglar. Within this respect, the thesis contributes to the literature of &lsquo
bag&rsquo
settlements in terms of defining their being and changing role throughout the history.

Resumo:
The general purpose of this study was to examine transformations of human agency in natural contexts. Existing theoretical formulations have primarily been confined to laboratory investigations. Moreover, the principles generated by such theories have not been validated beyond the laboratory setting. With this purpose in mind, there were two immediate aims of the study. The first aim was to contribute to counselling theory by assessing five prominent theories of human agency and providing a basis from which to potentially establish more adequate theoretical formulations. The second aim was to contribute to counselling practice by providing concrete information and a more informed basis through which to enhance agency in clients. A multiple case study design integrating intensive interviewing and Q-methodology was utilized for the study. Ten individuals, five women and five men, ranging in age from 28 to 64, were identified through a network of contacts for participation in the study. Based upon convergence of qualitative evidence from interviews and quantitative evidence from Q-sorts, rich, detailed narrative accounts of transformation were constructed for each individual. Each account was validated by the individual for whom each was written and by an independent reviewer. Through a comparative analysis of the ten diverse accounts of transformation, extensive commonality was identified. Twenty-two common themes were extracted from the accounts that portrayed significant features of the transformation. Based on these themes, an abstract story of the common pattern revealed in the transformation was plotted. Individual aspects of each of the theories of agency were validated as well as qualified in some important ways. In addition, the results extended these theories in three main ways. First, the results indicated that transformations of human agency were complex wholes that involved a configuration of features rather than any one or two isolated features. Second, the findings indicated that context played a critical role in transformations of agency. And third, the results emphasized the important role of powerful emotions in the process of transformation. The results of this study also generated a beginning holistic portrait of transformation which has implications for counsellors in terms of understanding and facilitating transformations of agency in clients.

This thesis uses heuristic and alchemical hermeneutic methodologies and a depth psychological perspective to examine the metaphor and experience of tattoo. The history of tattoos and ideas around healing are explored, as well as the author’s own healing and transformation through multiple tattoo experiences. The author’s analysis illustrates how a tattoo may be not only representative of a snapshot of the psyche in a moment in time, but might also move the psyche toward healing through the exploration of the archetypal energy in the image. In addition, the author explores how the modification of the body has the capacity to change the emotional and psychological relationship to one’s body.

Resumo:
The aim of this study was to develop a genetic transformation system for the plant pathogenic fungus Rhizoctonia solani (teliomorph, Thanatephprus cucumeris Frank [Donk]). The availability of a transformation system would allow us to study gene exchange, epidemiology, and to use techniques such as gene disruption or gene silencing to investigate the role of fungal enzymes in pathogenesis. The approach adopted was to use Agrobacterium tumefaciens to transform the fungus as reports in the literature suggested that this was the most efficient and easiest method to use. As a preliminary test, Fusarium oxysporum was transformed using a binary vector (pBINAN) containing a hygromycin resistance gene under control of an ascomycete promoter and terminator. Hygromycin resistant transformants were obtained after co-incubation of fungal conidia with the bacterium. The presence of the transgene was confirmed by analysis of DNA. The number of transformants depended on the genetic background of the A tumefaciens. Strains AGLO or AGRO gave higher numbers of transformants compared to LBA4404. No transformants were obtained when the hygromycin gene was under control of a basidiomycete (pBINHL1), or a plant (CaMV35S) promoter. Since the basidiomycete promoter used in pBINHL1 originates from Ustilago maydis, the vector was tested by transformation of Ustilago cynodontis. Stable transformants of U cynodontis were obtained with this vector. A series of experiments were carried out on transformation of R. solani mycelium. Both the protoplast and the Agrobacterium transformation methods were tested. Parameters affecting protoplast production and regeneration were examined. Protoplast production varied with the age of the mycelium, with the osmotic stabilizer used, and with time of treatment with protoplasting enzymes. Regeneration of protoplasts was also affected by the osmotic stabilizer and the growth medium. Transformation of several isolates from different anastomosis groups (AG) was attempted by inducing protoplasts to take up DNA using polyethyleneglycol. Two plasmids were used; (1) pAN7-1 containing the resistance gene under control of an ascomycete promoter, and (2) pHL-1 in which the resistance gene is under control of a basidiomycete promoter. No transformants were obtained. Attempts were then made to transform mycelium and protoplasts using A tumefaciens. The experiments used both mycelium and protoplasts as the recipient. A number of small resistant colonies were obtained using binary plasmid (pBINHL1) in which mycelium was transformed with the resistance gene was driven by the basidiomycete promoter. On transfer to fresh medium these colonies would grow to about 2cm diameter, and then stop growing. On a second transfer to fresh medium they failed to show any growth. No resistant colonies were obtained from A. tumefaciens transformation of protoplasts. To improve transformation efficiency, a vector was constructed in which the hygromycin resistance gene was fused to an R solani laccase promoter sequence. No resistant colonies were obtained using this vector. Further experiments were carried out using a hygromycin resistance gene specially modified for expression in basidiomycetes by the insertion of artificial introns in the 5’ and 3’ untranslated regions, and a number of AT to CG conversions in the coding region. Most of the recipient isolates gave transformants with the unstable resistance phenotype. However, one AG 6 isolate gave transformants with a stable resistance phenotype. Of six transformants recovered from this isolate, five were shown by PCR and southern blotting to contain the transgene. In four of these transformants the resistance phenotype was stable in the absence of selection.

Resumo:
We consider naturally occurring, uncountable transformation semigroups S and investigate the following three questions. (i) Is every countable subset F of S also a subset of a finitely generated subsemigroup of S? If so, what is the least number n such that for every countable subset F of S there exist n elements of S that generate a subsemigroup of S containing F as a subset. (ii) Given a subset U of S, what is the least cardinality of a subset A of S such that the union of A and U is a generating set for S? (iii) Define a preorder relation ≤ on the subsets of S as follows. For subsets V and W of S write V ≤ W if there exists a countable subset C of S such that V is contained in the semigroup generated by the union of W and C. Given a subset U of S, where does U lie in the preorder ≤ on subsets of S? Semigroups S for which we answer question (i) include: the semigroups of the injec- tive functions and the surjective functions on a countably infinite set; the semigroups of the increasing functions, the Lebesgue measurable functions, and the differentiable functions on the closed unit interval [0, 1]; and the endomorphism semigroup of the random graph. We investigate questions (ii) and (iii) in the case where S is the semigroup Ω[superscript Ω] of all functions on a countably infinite set Ω. Subsets U of Ω[superscript Ω] under consideration are semigroups of Lipschitz functions on Ω with respect to discrete metrics on Ω and semigroups of endomorphisms of binary relations on Ω such as graphs or preorders.
