Abstract Objective Monitoring the cellular immunity for Cytomegalovirus (CMV) in organ transplant recipients is a promising tool to support prevention strategies for posttransplant CMV infection or reactivation. Commercially available in vitro test systems differ substantially in their capacity to stimulate subpopulations of T-lymphocytes. We compared two assays for CMV immune monitoring in respect of their clinical practicability and significance. Methods Blood samples of 30 lung transplant recipients (LuTRs) were examined before transplantation and over a period of six months afterwards with T-Track® CMV (Lophius Biosciences GmbH, Regensburg) and QuantiFERON®-CMV (Cellestis GmbH, Darmstadt). The T-Track® CMV is based on ELISpot-technology, allowing quantification of interferon-gamma (IFNγ) secreting CD4+ and CD8+ T-cells after specific stimulation. Contrarily, the QuantiFERON®-CMV assay is restricted to detection of IFNγ secreted by CD8+ T-lymphocytes with ELISA. The data are evaluated in the context of transplant outcome and determination of viral load in plasma by qPCR. Results and Conclusion Both approaches provide similar results while exhibiting certain advantages and limitations. Early during immune suppressive therapy, QuantiFERON®-CMV generates often indeterminate results as depletion of T-cells is not taken into account. Although a comparatively large volume of blood is required, T-Track® CMV circumvents this drawback. In addition, T-Track® CMV reaches a higher sensitivity prior to LuTx and after onset of immunosuppression.

Abstract Abstract 4839 Functional characterizations of T lymphocytes are performed to gain understanding on their contribution in certain immunological situations, to monitor the course of diseases, and to track therapeutic interventions. Meanwhile the flow cytometric analysis of antigen-specific T cells examined for intracellular cytokine production and expression of activation markers after a short-term in vitro antigenic challenge is a well-established method for research applications. Despite the advantages of this approach to qualify samples on a single cell level and by multiple parameters, the broad use of this analysis for immune monitoring purposes is hampered. Screening of a lot of samples is time-consuming, requires many manual handling steps, and operator experience in flow cytometric analysis of stimulated T cell samples. To overcome these hurdles, we worked out a complete strategy to rapidly study cytokine and activation marker profiles in antigen-specific T cells of multiple samples by a semi-automated process. For the simultaneous analysis of multiple samples we examined for the T cell stimulation an antigen pre-coated 96-strip-well culture system. This flexible and ready-to-use format provides the opportunity to screen either for a single antigen or in parallel for up to twelve antigen specificities by combining 8-well-strips possessing different antigens. The coated antigens consist of pools of overlapping 15-mer peptides derived from a single viral protein of cytomegalovirus, Epstein-Barr-Virus, or adenovirus. The peptide pools have been designed for activation of the specific CD4+ as well as CD8+ T cells. They are solubilized and thereby accessible for T cell stimulation after addition of a cell sample, e.g. peripheral blood mononuclear cells, suspended in culture medium into the antigen-coated well. After a stimulation period of six hours the induced T cell response is comparable to the activation with a conventional lyophilized and reconstituted peptide pool. To reduce the time and work load for cell harvesting, fixation, permeabilization, and staining, we developed a protocol and reagents to allow a rapid and easy-to-handle intracellular staining procedure. Compared to conventional staining protocols, all steps are executed in the 96-strip-well culture plate, i.e. cell harvesting is dispensable. Without any washing step, cells are fixed and stained with defined reagent cocktails containing antibodies to identify virus-specific CD3+ CD4+ CD154+ and/or CD3+ CD8+ T cells and various Anti-cytokine-fluorochrome conjugates to evaluate the cytokine pattern. With these modifications, we drastically diminished the overall processing time for the staining of up to 96 samples to only 50 minutes. Furthermore, we integrated an automated flow cytometric analysis process. This includes the possibility to measure the samples in the 96-strip-well plates hands-free using pre-defined experiment settings and acquisition templates. We also applied an automated gating strategy for the data analysis. Finally, a report summarizes the results of the T cell response against several viral proteins for all samples tested, e.g. frequencies of cytokine+ CD154+ CD4+ and cytokine+ CD8+ T cell subsets are indicated. Hands-on time for the multi-sample acquisition and analysis is only minimal and the standardized reagents/protocol and sample analysis process decrease inter- and intra-assay variations. In summary, with our newly developed tools and protocols for in vitro T cell stimulation, staining of activation markers as well as intracellular cytokines, and automated flow cytometric analysis we have set up a fast and convenient procedure to routinely monitor antigen-specific T cell responses. The over-the-scope-clip (OTSC; Ovesco Endoscopy GmbH, Tuebingen, Germany) system is a newly designed method for the mechanical compression of large areas in the gastrointestinal tract. So far, indications for OTSC application are hemostasis of primary or postinterventional bleeding, closure of iatrogenic full-thickness or covered perforations. Recently closure of gastrointestinal tract fistulas using this device has been described. A 44-year-old man developed a gastrocutaneous fistula after surgical treatment for a perforated gastric ulcer. We describe the successful endoscopic closure of the fistula using the OTSC system. The patient's clinical followup was uneventful. Fistula closure was successfully implemented as it was documented by imaging and endoscopic examinations performed on the 2nd day and 6th week after the application of the clip. Endoscopic application of the OTSC device was safe and effective for the treatment of a gastrocutaneous fistula.

Background:Lower urinary tract symptoms (LUTS) are an underdiagnosed but frequent manifestation in systemic sclerosis (SSc) [1]. LUTS pathogenesis in SSc is undetermined, mainly involving dysautonomia, fibrosis and a possible antibody-mediated damage [2]. Divergently from general population, female sex and advanced age are not reported to significantly impact LUTS in SSc [2].Objectives:To evaluate the potential influence of gender and hormone-related factors in LUTS prevalence and severity among SSc patients (Pts).Methods:A population of 42 SSc Pts and 50 age- and sex-matched healthy subjects (HSs) was evaluated. SSc diagnosis was based on 2013 ACR/EULAR criteria. Demographic data, medications interfering with pelvic floor dynamics and general comorbidities commonly associated with LUTS – diabetes mellitus, chronic heart failure, chronic obstructive pulmonary disease, peripheral neuropathy, pelvic organ prolapse, fecal incontinence – were recorded. Validated self-reported questionnaires derived from the International Conference on Incontinence were used to assess prevalence and severity of LUTS, namely of urinary incontinence (UI) and overactive bladder (OAB) [3]. Data were analysed using non-parametric tests. Apvalue <0.05 and a confidence interval (CI) of 95% were considered statistically significant.Results:There were no significant differences in main demographic data between SSc Pts and HSs. Specifically, median age was 61 years (IQR 21-85)vs57 years (IQR 28-93) and female prevalence 83%vs84% in SSc PtsvsHSs, respectively. Amongst the female population, 83% of SSc Ptsvs84% of HSs was in post-menopausal state, with a median of 1 (IQR 0-3)vs1 (IQR 0-4) pregnancy by natural route, respectively. No woman of the study had received hormone replacement therapy or local hormonal therapies prior to the study. Similarly, there were not any significant differences in analysed comorbidities, while ongoing treatment was significantly different between the two populations, SSc patients more frequently receiving calcium channel blockers and glucocorticoids than healthy subjects (p< 0.001). In SSc Pts, statistically significant correlation was observed between stress UI and sex, with an increased female-to-male ratio (p< 0.005), but any significant difference was observed in US distribution depending on parity and menopausal state, nor on other analysed variables. Interestingly, female dominance has not resulted as a significant predictive factor for LUTS prevalence or severity in SSc Pts. In fact, in the regression analysis, SSc disease was the only significant predictor for LUTS (OR 3.45, 95% CI 1.41-7.95; p< 0.01), independently of other analysed variables, particularly of gender and hormone-related factors.Conclusion:This study confirms the absence of pathogenic female-gender participation in LUTS prevalence among SSc Pts. However, consistently with findings on general population, a significant increased prevalence of urinary symptoms, particularly of stress UI, in SSc female Pts has emerged [4]. The objective of this work has been to develop a robust calibration method for simultaneous multigas detection with a Fourier transform infrared (FT-IR) system. Calibration models for the identification and quantification of 23 gases in the presence of high concentrations of background gases such as water vapor, carbon dioxide, and methane have been obtained for an FT-IR instrument with 0.7-cm−1 resolution. The calibration models have been tested on a breadboard instrument for trace gas measurement in manned space missions. The results show that FT-IR combined with multivariate methods such as partial least-squares (PLS) and proper pretreatment of the infrared spectra used in calibration is well suited for this purpose. A procedure for baseline drift compensation has been introduced to make the system insensitive to baseline drift and variations in transmittance. This baseline drift compensation also reduces the need for background measurements. Further, a procedure for incorporating a priori information about the instrument signal-to-noise ratio (SNR) and the absorption strength of interfering absorption lines has been developed. Indoor air monitoring and industrial process monitoring are other possible application areas for these techniques. Parts of this work have been performed in a project for the European Space Agency (ESA) in cooperation with Kayser-Threde GmbH and Daimler-Benz Aerospace, Dornier GmbH.

This paper discusses engineering design ideas employed by P. Wirzius Heavy Assembly GmbH of Hinden, Germany, to transport heavy loads across states. In order to find a safe way to cross its bridges, Wirzius enlisted a specialist in getting heavy objects safely to their destinations, Greiner Vehicle Technology, an engineering company based in Neuenstein, Germany. Greiner says it designs systems for transporting large machinery by road or rail. Working with Wirzius, the Greiner company devised a plan to transfer some of the extraordinary vehicle weight to a temporary track system that would channel that part of the load directly to the bridge piers. Engineers working in 3D simulated the system in motion and avoided problems, like collisions, that would otherwise have needed correction later. Crews from Greiner and Wirzius tested the system at the former Butzweilerhof Airport in Ossendorf. A transport vehicle had to transport 295 tons of ballast across a 14S-meter track. The next day, they repeated the test as a demonstration for French motorway authorities.

Inorganic and organic geochemical and geostatistical studies of manganese ore deposits exposed at Daranna, near Kaoje have been carried out with the aim of characterizing and delineating the origin of the manganese ore deposits. Data were obtained from field observations and chemical analyses of 12 ore samples. Major and trace elements and organic geochemical analyses were conducted with Energy Dispersion X-ray Fluorescence Spectrometer and Rock-Eval pyrolysis method at Geo-data GmbH Garbsen, Germany. Results of the Major and trace elements, total organic carbon content abundances and the correlation among them imply that Daranna mineralizations are mainly of hydrothermal origin with little contribution from contemporaneous volcanic materials and this is confirmed by high Fe/Mn (8.274 to 24.066 wt%) and low Co/Zn ratios and trace element patterns. The significant geochemical characteristics such as high Mn content (22.660 to 62.330 wt%; average 45.919 wt%), low concentration of Fe (2.590 to 3.310 wt%; average 3.008 wt%) reveal a primary distal hydrothermal source for mineralization. The position of samples on Ni–Zn–Co and (Co/Zn)–(Co + Ni + Cu) diagrams and evaluation of these data reveals that hydrothermal activity was the main process with sedimentary influence responsible for mineralization in the Daranna manganese deposit.

In Austria last years sequenced according modern technology successfully bridges were made from UHPC or UHPFRC [1], [2], [3]. A couple of well-done examples of these bridges I presented on the occasion of Prof. Sparowitz birthday publication in 2017 [2]. Recently in Austria another example of a railway bridge consisting innovative material UHPFRC - created as a composite construction - was projected by ZKP ZT GmbH by order of Austrian Railway Company ÖBB. Innovative and new is the application of Ultra-High Performance Concrete in combination with Steel S355J2+N creating a composite action in highest utilization of both material strengths and auxiliary connected with a steel panel. Construction work was done from March to September 2017, the bridge was finished in October 2017 in single railway track Lavanttalbahn near city Wolfsberg in Carinthia.

Abstract This paper provides empirical insights and practical implications for the implementation process of agile leadership in multinational firms. Building on Häußler’s and Fischer’s Trafo-model our research shows that the dimensions “culture” and “leadership” have been important dimensions for the Advantest Europe GmbH in 2017 to boost the implementation of agile leadership. Concerning the dimension “culture” the most important insight was that agile leadership is more than agile methods such as Scrum or Kanban. Concerning the dimension “leadership” our results show for example that a lack of a common understanding of agile leadership and role conflicts of managers complicate the implementation of agile leadership. Our research has several implications for the practical implementation of agile leadership such as setting up a common understanding of agile leadership or developing clear mechanisms for delegation.

Valderi Luiz Dressler holds a chemistry degree (1992) and a master’s in chemistry (1994) from the Federal University of Santa Maria (UFSM), RS, Brazil, a PhD in chemistry (1999) from the Federal University of Santa Catarina, SC, Brazil and post-doctorates from Forschungszentrum Julich GmbH, Germany (2007 and 2010). Currently, he is a full professor and coordinator of the industrial chemistry and bachelor's chemistry degree courses at the UFSM. Dr. Dressler’s research activities mainly include trace analysis and environmental chemistry, involving the techniques of atomic absorption spectrometry (AAS), inductively coupled plasma optical emission spectrometry (ICP-OES), inductively coupled plasma mass spectrometry (ICP-MS), flow injection analysis (FIA), electrothermal vaporization (ETV) and laser ablation (LA). Furthermore, he performs chemical speciation analysis studies involving hyphenated techniques such as liquid and gas chromatography coupled to ICP-MS and also FIA systems coupled to AAS.

De el estúdio de 195 exudados vaginales enviados por el Servicio de Ginecologia de este hospital, durante el período 1988-1990, hemos seleccionado aquellos en los que el cultivo fue positivo para estreptococos, 58 (30%) de los cuales 26 (44.8%) correspondia a Streptococcus morbillorum, 9 (15.5%) a Gardnerella vaginalis, 5 (8.6%) a Enterococcus faecalis-durans, y a Streptococcus agalactiae, 3 (5.1%) a Streptococcus mitis y Streptococcus mitis, 2 (3-4%) a Streptococcus bovis y Streptococcus cremoris y 1 (1.7%) a Streptococcus salivarius, Streptococcus equinus y Strptococcus sanguis II respectivamente. En todos los casos se observo antecedentes de actuacción medico- quirurjica en el tracto genital, y en el 52.8% de los casos fuô concomitante con el diagnostico clinico-micologico de candidiasis vaginal. La ideittificaccion bacteriologica se realizo mediante el sistema API 20 STREP (sistema api bioMêríeux GmbH, Nütingen, Alemania) dando un patron tipico ("excelente identificacción") para el Streptococcus morbillorum.

Introduction. Anticoagulants such as argatroban and heparins (low-molecular-weight and unfractionated) play an immense role in preventing thromboembolic complications in clinical practice. Nevertheless, they can also have a negative effect on the immune system. This study is aimed at investigating the influence of these substances on polymorphonuclear neutrophils (PMNs), whose nonspecific defense mechanisms can promote thrombogenesis. Methods. Blood samples from 30 healthy volunteers were investigated, whereby PMNs were isolated by density gradient centrifugation and incubated with 0.8 μg/mL of argatroban, 1.0 U/mL of low-molecular-weight heparin (LMWH), 1.0 U/mL of unfractionated heparin (UFH), or without drug (control). A collagen-cell mixture was prepared and filled into 3D μ-slide chemotaxis chambers (IBIDI® GmbH, Germany). Stimulation was initiated by using a chemokine gradient of n-formyl-methionine-leucyl-phenylalanine (fMLP), and microscopic observation was conducted for 4.5 hours. The cells’ track length and track straightness, as well as the number of attracted granulocytes, level of ROS (reactive oxygen species) production, and NET (neutrophil extracellular traps) formation, were analyzed and categorized into migration distances and time periods. Results. All three anticoagulants led to significantly reduced PMN track lengths, with UFH having the biggest impact. The number of tracks observed in the UFH group were significantly reduced compared to the control group. Additionally, the UFH group demonstrated a significantly lower track straightness compared to the control. ROS production and NET formation were unaffected. Conclusion. Our data provide evidence that anticoagulants have an inhibitory effect on the extent of PMN migration and chemotactic migration efficiency, thus indicating their potential immune-modulatory and prothrombotic effects.

The estrogenic mycotoxin zearalenone (ZEN) is a common contaminant of animal feed. Effective strategies for the inactivation of ZEN in feed are required. The ZEN-degrading enzyme zearalenone hydrolase ZenA (EC 3.1.1.-, commercial name ZENzyme®, BIOMIN Holding GmbH, Getzersdorf, Austria) converts ZEN to hydrolyzed ZEN (HZEN), thereby enabling a strong reduction in estrogenicity. In this study, we investigated the efficacy of ZenA added to feed to degrade ZEN in the gastrointestinal tract of three monogastric animal species, i.e., pigs, chickens, and rainbow trout. For each species, groups of animals received (i) feed contaminated with ZEN (chickens: 400 µg/kg, pigs: 200 µg/kg, rainbow trout: 2000 µg/kg), (ii) feed contaminated with ZEN and supplemented with ZenA, or (iii) uncontaminated feed. To investigate the fate of dietary ZEN in the gastrointestinal tract in the presence and absence of ZenA, concentrations of ZEN and ZEN metabolites were analyzed in digesta of chickens and rainbow trout and in feces of pigs. Upon ZenA administration, concentrations of ZEN were significantly decreased and concentrations of the degradation product HZEN were significantly increased in digesta/feces of each investigated animal species, indicating degradation of ZEN by ZenA in the gastrointestinal tract. Moreover, upon addition of ZenA to the diet, the concentration of the highly estrogenic ZEN metabolite α-ZEL was significantly reduced in feces of pigs. In conclusion, ZenA was effective in degrading ZEN to HZEN in the gastrointestinal tract of chickens, pigs, and rainbow trout, and counteracted formation of α-ZEL in pigs. Therefore, ZenA could find application as a ZEN-degrading feed additive for these animal species.

Abstract Laser metal deposition (LMD) is an additive manufacturing process, which is used to generate large parts or for reconditioning machine components. During the process the welding tracks are affected by complex dynamic temperature-time curves that results in geometric deviations, which limits the build-up process. Therefore, complex structures are more difficult to manufacture with constant parameters. The present work describes an in-process monitoring system and first approaches of a control concept for LMD processes. It is set up using a newly developed laser triangulation sensor from Falldorf Sensor GmbH, which is trailing mounted. Due to the resistance to diffused radiation, e.g. from process light, it is possible to place the sensor close to the process zone which offers the possibility of a height control system. For the experiments, single tracks were welded from 316L powder by using a diode laser. The results show that the track width and height increase with rising laser power. Depending on the process parameter the height changes up to 29% and the width up to 58% if only the laser power is modified by 1800 W. The monitoring system forms a good basis for a later control concept, which can be used to make the process more stable in the case of complex structures.

Abstract Backround: The in vitro cytotoxic effects of six different clear aligner materials were evaluated using the MTT analysis. Methods: The clear aligner material samples [Duran (ScheuDental GmbH, Iserlohn, Germany), Zendura-Flx (Bay Materials LLC, Fremont, CA, USA), Taglus (Laxmi Dental Export Pvt. Ltd, Mumbai, India), Smart Track (Align Technology, San Jose, CA, USA), Zendura (Bay Materials LLC, Fremont, CA, USA), Essix C + (Essix® (Raintree Essix, Inc., 4001 Division St, Metairie, LA-USA)] were initially kept in a saline solution in airtight test tubes for 8 weeks at 37°C. According to the recommended ISO standards, the weights of the samples were divided by the volumes of the dilutions in the ratio of 0.1 g/ml. To evaluate the cytotoxicity of the samples, an MTT analysis was performed using a human gingival fibroblast cell line (HGF). To analyse the data, the Kruskal– Wallis test was applied (a=0.05). Results: Zendura was the most cytotoxic material resulting in 67.3 ± 16.20% cell viability, followed by Smart Track with 87.6 ± 5.53% cell viability. While Duran, Essix C + had 92.6 ± 26.34% and 94.9 ± 8.54% cell viability, Zendura-Flx, Taglus had 106.9 ± 12.76% and 113.183 ± 7.45% cell viability, respectively. Conclusion: While Zendura and Smart Track showed mild cytotoxicity, other materials showed greater cell viabilities. According to the ISO standards, the clinical use of each brand of aligners, except Zendura, may be considered reliable. Taking into account standard deviation, Zendura and Duran should be used with caution. The suppliers of aligners should adhere to the manufacturer’s recommendations since an increase in ion release might arise from material wear.

The antigen rapid diagnostic test (Ag-RDT) is an immunodiagnostic test that detects the presence of viral proteins (antigens) expressed by the COVID-19 virus in a sample from a patient’s respiratory tract. This study focused on evaluating the performance of self-conduct buccal and nasal swabs RTK-antigen test compared to nasopharyngeal swab RTK-based COVID-19 diagnostic assays, Panbio™ COVID-19 Ag Rapid Test Device (Nasopharyngeal) (Abbott Rapid Diagnostics Jena GmbH, Jena, Germany) used in hospitals for first-line screening. The sensitivity and specificity of the paired RTK-Ag test in detecting the an-tigen were calculated at 96.4% and 100%, respectively. Fisher exact tests showed the association between nasopharyngeal swabs RTK-Ag assay and buccal-nasal swabs RTK-Ag from ProdetectTM is significant (p-values < 0.001). The result showed that a self-conducted buccal and nasal RTK-antigen rapid test by the patients is comparable to the results obtained from a rapid test device conducted by trained medical personnel using a nasopharyngeal swab.

Anastomotic insufficiencies are severe complications of abdominal surgery, often leading to prolonged hospitalization, serious tissue inflammation, and even sepsis, along with the need for recurrent surgery. Current non-surgical treatments such as self-expanding metal stents (SEMSs) and endoscopic vacuum therapy (EVT) have limitations, including stent migration or perforation. This review evaluates the effectiveness of the VacStent GITM (Möller Medical GmbH, Fulda, Germany), a novel medical device combining SEMS and negative-pressure wound therapy in treating gastrointestinal leaks. Data were gathered from four prospective studies and compared with existing treatments. Studies on the VacStent GITM application demonstrate technical success and competitive closure rates in upper gastrointestinal leaks, with minimal complications reported. Comparative analyses with SEMS and EVT reveal promising and most importantly equally good outcomes while maintaining the possibility for sustained enteral nutrition and reducing the risk of stent migration. The VacStent GITM presents a promising alternative to current non-surgical treatments. Ongoing research aims to validate its efficacy in lower gastrointestinal leaks and comprehensively establish its role in leak management. Further investigation is necessary to confirm these findings and optimize treatment protocols. Future usages of the VacStent GITM in colonic anastomotic insufficiencies promise an effective approach and might be able to lower the rates of necessary implementations of a stoma.

The aim. Find out the nature of the changes of the hepatobiliary system in patients with intestinal bacterial overgrowth and study the possible mechanisms of their association.Materials and methods. 148 patients with intestinal bacterial overgrowth and intestinal dysbiosis were examined. The level of total cholesterol, cholestasis and cytolysis markers was determined in the blood using the analyzer «Labsystems» (Finland). Intestinal bacterial overgrowth syndrome was assessed using a hydrogen breath test with lactulose on the LactophaH2 apparatus of AMA (St. Petersburg). Intestinal dysbiosis was determined by plating feces on nutrient media. Bile acids in bile were determined on the AmazonX mass spectrometer (Bruker Daltonik GmbH, Bremen, Germany). Ultrasound examination of the abdominal cavity performed with the apparatus «SHIMADZU SDN-500» (Japan). Liver elastography was performed using the AIXPLORER apparatus (France).Results. The syndrome of intestinal bacterial overgrowth in 67% of cases was established in the presence of ileocecal insufficiency, in 33% of cases — with preserved ileocecal function. The combination of intestinal bacterial overgrowth syndrome and intestinal dysbiosis was detected in 81,8% of patients. The majority of the examined patients showed clinical symptoms of damage the hepatobiliary system and intestines, which was confirmed by change laboratory parameters — increase the level of total cholesterol, markers of cholestasis and cytolysis compared with the control group. In the study of bile acids in bile, decrease free (mainlycholic) and increase conjugated (glycodesoxycholic, taurodesoxycholic, glycocholic, taurocholic) bile acids was observed compared with the control group. In general, patients with the syndrome of intestinal bacterial overgrowth revealed the presence of non-calculous cholecystitis — in 11,5% of cases, I stage of cholelithiasis — in 25,7%, II stage of cholelithiasis — in 18,9%, non-alcoholic fatty liver disease on stage steatosis and steatohepatitis — in 43,9% of cases.Conclusion. Intestinal bacterial overgrowth syndrome is the beginning of bacterial translocation, which is the triggering factor in inflammation of the liver and biliary tract. In turn, diseases of the hepatobiliary system contribute to the development of intestinal dysbiosis by reducing the synthesis of bile acids with antibacterial action, as well as violations of their excretion. Thus, strong association of intestinal bacterial overgrowth syndrome with damage to the hepatobiliary system has been established.

Sensitive real-time detection of vapors produced by the precursors, reagents and solvents used in the illegal drugs manufacture represents a priority nowadays. Acetic anhydride (AA) is the key chemical used as acetylation agent in producing the illegal drugs heroin and methaqualone. This study was directed towards quick detection and quantification of AA in air, using two fast and very sensitive analytical techniques: photoionization detection (PID) and ion mobility spectrometry (IMS). Results obtained indicated that both PID and IMS can sense AA at ultra-trace levels in air, but while PID produces a non-selective response, IMS offers richer information. Ion mobility spectrometric response in the positive ion mode presented one product ion, at reduced ion mobility K0 of 1.89 cm2 V−1 s−1 (almost overlapped with positive reactant ion peak), while in the negative ion mode two well separated product ions, with K0 of 1.90 and 1.71 cm2 V−1 s−1, were noticed. Our study showed that by using a portable, commercial IMS system (model Mini IMS, I.U.T. GmbH Berlin) AA can be easily measured at concentrations of 0.05 ppmv (0.2 mg m−3) in negative ion mode. Best selectivity and sensitivity of the IMS response were therefore achieved in the negative operation mode.

AbstractSince 1994 the Alfred Wegener Institute (AWI) has operated an airborne radio-echo sounding system for remote-sensing studies of the polar ice caps in Antarctica and in Greenland. It is used to map ice thicknesses and internal layernigs of glaciers, ice sheets and ice shelves, and is capable of penetrating ice thicknesses of up to 4 km. The system was designed and built by AWI in cooperation with Aerodata Flugmeßtechnik GmbH, Technische Umversitat Hamburg-Harburg and the Deutsches Zentrum fur Luft- und Raumfahrt e.V. The system uses state-of-the-art techniques, and results in high vertical (5 m) as well as along-track (3.25 m) resolution. The radar signal is a 150 MHz burst with a duration of 60 or 600 ns. The peak power is 1.6 kW, and the system sensitivity is 190 dB. The short backfire principle has been adopted and optimized for antennae used on Polar2, a Dormer 228-100 aircraft, resulting in an antenna gain of 14 dB each. Digital data recording allows further processing. The quality of the recorded data can be monitored on screen and as online analogue plots during the flight.

Abstract LDN Labor Diagnostika Nord GmbH & Co. KG has developed an ELISA for the rapid semiquantitative or quantitative determination of histamine in different kinds of fish samples. Fresh/frozen tuna, canned tuna, fresh/frozen mahi mahi, canned sardines, and fish meal were used to validate the method under the requirements of the AOAC Research Institute Performance Tested MethodsSM Program. The results all fell within the defined acceptance criteria. Linearity was given throughout the 0–300 ppm measuring range. Cross reactivity to similarly structured food spoilage indicators (tyramine and cadaverine) was minimal, less than 1%. Overall recoveries for all tested matrixes were within the determined acceptable range (80–120%), and the repeatability of precision was less than 10% overall and less than 5% at the defect action level of 50 ppm set by the U.S. Food and Drug Administration. The LOQ for the ELISA was determined to be 1.27 ppm. No lot-to-lot differences were observed. A 2-year claimed shelf life, at 2–8°C, was confirmed through accelerated stability testing. After the introduction of minor procedural variations to the test, no differences in histamine levels were observed. Independent lab testing revealed that the ELISA works as well in the hands of minimally trained technicians as it does with expert developers. A strong correlation (r = 0.97) between the LDN ELISA and the AOAC 977.13 fluorometric method was observed. This study revealed that the LDN ELISA is equivalent to the AOAC Official Method977.13 for the precise and accurate measurement of histamine in fresh/frozen tuna, canned tuna, fresh/frozen mahi mahi, canned sardines, and fish meal.

OTSC naudojama su lanksčiais endoskopais šiais atvejais: 1) ūminiam kraujavimui stabdyti, 2) kiauriniam virškinamojo trakto defektui užverti, 3) audiniams suspausti ir suartinti, 4) endoskopinių ir chirurginių intervencijų komplikacijoms gydyti. Publikacijos tikslas – retrospektyvioji OTSC naudojimo virškinamojo trakto vientisumui atkurti analizė.RezultataiNuo 2013 m. kovo iki 2014 m. vasario Vilniaus universiteto Onkologijos institute ir Vilniaus universiteto Santariškių klinikose naudojant OTSC kabutes gydyti 9 ligoniai, iš jų 6 vyrai ir 3 moterys. Ligonių amžius – 35–86 metai. Indikacijos atlikti procedūrą: anastomozių nepakankamumas po stemplės rezekcijos, skrandžio rezekcijos ir gastrektomijos (4 atvejai), skrandžio ir dvylikapirštės žarnos perforacijos po daugybinių operacijų dėl nekrozinio pankreatito (2 atvejai), lėtinės fistulės po stemplės rezekcijos ir po stemplės užsiuvimo po perforacijos dėl svetimkūnio (2 atvejai) ir nusileidžiančiosios storosios žarnos perforacija atliekant endoskopinę polipektomiją (1 atvejis). Virškinamojo trakto defektas visiškai užvertas 7 iš 9 atvejų, užvėrimo adekvatumas patikrintas suleidus rentgenokontrastinės medžiagos ar metileno mėlio tirpalo iškart po procedūros. Iš viso 9 pacientams buvo dėta 12 kabučių. Vėlyvuoju laikotarpiu fistulė užsivėrė 7 iš 9 atvejų, vienu nekrozinio pankreatito atveju fistulė atsinaujino kitoje vietoje ir 1 atveju nepavyko užverti didelio anastomozės defekto.IšvadosOTSC kabutė – naujas efektyvus endoskopinis virškinamojo trakto fistulės, perforacijos ar anastomozės nepakankamumo gydymo būdas. The OTSC System could be used in flexible endoscopy for 1) acute bleeding, 2) full thickness wall closure, 3) compressionas well as approximation of tissue, and 4) management of complications after endoscopic or surgical interventions. The aim of the presentation is a retrospective analysis of the restitution of digestive tract integrity with OTSC System.Results2013.03–2014.02 in Vilnius University Hospital Santariskiu Klinikos and Institute of Oncology, Vilnius University, 9 patients were treated with the OTSC System. There were 6 males and 3 females, age range 35–86 years. Indications for the procedurewere: 4 anastomotic leaks and subacute fistulas after esophageal resection, gastrectomy and partial gastrectomy, 2 perforations due to necrotic pancreatitis after multiple operations – stomach and duodenum, 2 chronic fistulae after esophagealresection, suture of esophageal perforation due to a foreign body, and 1 acute perforation of the descending colon during colonoscopic polypectomy. Initial success was achieved in 7 out of 9 cases; the completeness of closure was checked radiologically or injecting a methylene-blue solution immediately after a clip application. The total number of clips was 12. At the follow-up, in 7 cases out of 9 no fistula/dehiscence was present, in one case of necrotizing pancreatitis a new fistula developed in another place and we didn’t succeed to close, one case with a large leak.ConclusionsThe OTSC System is a new efficient endoscopic means of closing a leak, fistula or perforation. A complete fistula / leak / perforationclosure was achieved in 7 out of 9 (78%) cases by using the OTSC System.

Virškinamojo trakto defektas visiškai užvertas 7 iš 9 atvejų, užvėrimo adekvatumas patikrintas suleidus rentgenokontrastinės medžiagos ar metileno mėlio tirpalo iškart po procedūros. Iš viso 9 pacientams buvo dėta 12 kabučių. The OTSC System could be used in flexible endoscopy for 1) acute bleeding, 2) full thickness wall closure, 3) compressionas well as approximation of tissue, and 4) management of complications after endoscopic or surgical interventions. The aim of the presentation is a retrospective analysis of the restitution of digestive tract integrity with OTSC System.Results2013.03–2014.02 in Vilnius University Hospital Santariskiu Klinikos and Institute of Oncology, Vilnius University, 9 patients were treated with the OTSC System. There were 6 males and 3 females, age range 35–86 years. Indications for the procedurewere: 4 anastomotic leaks and subacute fistulas after esophageal resection, gastrectomy and partial gastrectomy, 2 perforations due to necrotic pancreatitis after multiple operations – stomach and duodenum, 2 chronic fistulae after esophagealresection, suture of esophageal perforation due to a foreign body, and 1 acute perforation of the descending colon during colonoscopic polypectomy. Initial success was achieved in 7 out of 9 cases; the completeness of closure was checked radiologically or injecting a methylene-blue solution immediately after a clip application. The total number of clips was 12. At the follow-up, in 7 cases out of 9 no fistula/dehiscence was present, in one case of necrotizing pancreatitis a new fistula developed in another place and we didn’t succeed to close, one case with a large leak.ConclusionsThe OTSC System is a new efficient endoscopic means of closing a leak, fistula or perforation. A complete fistula / leak / perforationclosure was achieved in 7 out of 9 (78%) cases by using the OTSC System.

The study of the parameters of the upper respiratory tract has become one of the leading directions of medical research, due to the connection of this structure with the occurrence and course of many diseases of both childhood and adulthood. The study of normative indicators of this structure and its relationship with craniometric indicators should be the first step before further research of samples with pathologies. The purpose of the study is to establish correlations of cephalometric parameters of the upper respiratory tract in Ukrainian young men and young women with an orthognathic bite, regardless of the type of face. Primary lateral radiographs of 49 Ukrainian young men (aged 17 to 21 years) and 76 Ukrainian young women (aged 16 to 20 years) with an orthognathic bite and the absence of upper respiratory tract pathology taken from the database of the research center and the Department of Pediatric Dentistry age of National Pirogov Memorial Medical University, Vinnytsia, with the help of the licensed medical software OnyxCeph3™, version 3DPro (Image Instruments GmbH, Germany) and the diagnostic program “UniqCeph”, a cephalometric analysis of the upper respiratory tract was performed. In the “Statistica 6.0” license package, correlations between cephalometric indicators of the upper respiratory tract were assessed using non-parametric Spearman statistics. In Ukrainian young men, multiple, mostly direct, medium-strength (r= from 0.32 to 0.48) and strong (r= from 0.65 to 0.83) correlations were established between most of the characteristics of the upper respiratory tract or tongue; in young women, there are also predominantly direct, medium-strength (r= from 0.33 to 0.57) and strong (r= from 0.62 to 0.85) correlations between most of the characteristics of the upper respiratory tract or tongue, as well as medium-strength inverse (r= -0.30 and -0.40) and direct (r= 0.45 and 0.85) correlations between most characteristics of the soft palate. In addition, in Ukrainian young men, multiple inverse correlations of mainly medium strength (r= from -0.33 to -0.49) were established between the value of the PASmin distance and the UAA section with the value of the SPT distance and the SPA section, and between the value of the NL/PM-U angle and most of the characteristics the tongue, as well as multiple direct, mostly medium strength (r= from 0.30 to 0.55), connections between the characteristics of the upper respiratory tract itself and the hyoid bone or tongue and between the characteristics of the hyoid bone and the tongue; in young women, there are mainly straight lines of medium strength (r= from 0.33 to 0.55) between the value of the AH-CV distance and most of the characteristics of the upper respiratory tract itself, between the value of the AH-FH distance and all characteristics of the tongue, and between the value of the PM-U distance and by the distance VT and the section TA. Thus, the most pronounced manifestations of sexual dimorphism of connections are established between the characteristics of the soft palate, between the characteristics of the upper respiratory tract itself and the soft palate or tongue, as well as between the characteristics of the soft palate and the hyoid bone.

Sensitive real-time detection of vapors produced by toxic industrial chemicals (TICs) represents a stringent priority nowadays. Carbon disulfide (CS2) is such a chemical, being widely used in manufacturing synthetic textile fibers and as a solvent. CS2 is simultaneously a very reactive, highly flammable, irritant, corrosive, and highly toxic compound, affecting the central nervous system, cardiovascular system, eyes, kidneys, liver, skin, and reproductive system. This study was directed towards quick detection and quantification of CS2 in air, using time-of-flight ion mobility spectrometry (IMS); photoionization detection (PID) was also used as confirmatory technique. Results obtained indicated that IMS can detect CS2 at trace levels in air. The ion mobility spectrometric response was in the negative ion mode and presented one product ion, at a reduced ion mobility (K0) of 2.25 cm2 V−1 s−1. Our study demonstrated that by using a portable, commercial IMS system (model Mini IMS, I.U.T. GmbH Berlin Germany) one can easily measure CS2 at concentrations of 0.1 ppmv (0.3 mg m−3) in the negative ion mode, which is below the lowest threshold value of 1 ppmv given for industrial hygiene. A limit of detection (LOD) of ca. 30 ppbv (0.1 mg m−3) was also estimated.

Annotation. Changes in the size of the upper respiratory tract can lead to the emergence or aggravation, complications of already existing pathologies. In this regard, there is a need to study their sizes both in normal and pathological conditions. Cephalometric analysis is one of the important tools that allows you to assess the location of the structures of the upper respiratory tract and their parameters. The purpose of the study is to establish the peculiarities of correlations between cephalometric parameters of the upper respiratory tract in Ukrainian young men and young women with an orthognathic bite with a wide face type. On primary lateral radiographs of Ukrainian young men (aged 17 to 21 years) and young women (aged 16 to 20 years) with a wide facial type, orthognathic bite and absence of upper respiratory tract pathology, using the licensed medical software OnyxCeph³™, version 3DPro (company Image Instruments GmbH, Germany) and the diagnostic program “UniqCeph” carried out a cephalometric analysis of the upper respiratory tract, soft palate, hyoid bone and tongue. Correlations were evaluated using the Statistica 6.0 license package using Spearman's non-parametric statistics. In young men and young women with a wide face type, multiple, mostly straight, medium-strength reliable (r= from 0.42 to 0.56) and strong (r= from 0.62 to 0.82) correlations were established between most of the cephalometric characteristics of the upper respiratory tract itself, and only in young women – multiple straight correlations of average strength reliable (r=0.44 and r=0.53) relationships between most of the cephalometric characteristics of the hyoid bone. In addition, in young men with a wide face type, multiple straight correlations of mostly medium strength, reliable (r= from 0.47 to 0.52) and unreliable (r= from 0.30 to 0.41) relationships of the AH-CV, AH-MP or VT distances and most of the cephalometric characteristics of the upper respiratory tract itself, mainly the inverse of the average strength of reliable (r=-0.44 in both cases) and unreliable (r=-0.32 and r=-0.38) correlations of the value of most of the cephalometric characteristics of the soft palate (with the exception of the PM-U distance) and the tongue, as well as mostly straight correlations of medium strength unreliable (r= from 0.34 to 0.38) relationships between the values of most of the cephalometric characteristics of the hyoid bone (with the exception of the AH-CV distance) and the tongue; and in young women, there are direct, reliable, medium-strength (r=0.45 and r=0.53) and strong (r=0.63 in both cases) relationships between the value of the AH-CV distance and almost all cephalometric characteristics of the upper respiratory tract itself, as well as mainly direct unreliable average strength (r= from 0.37 to 0.38) correlations between the size of the PM-U or AH-FH distance and most of the characteristics of the tongue. Thus, the most pronounced manifestations of sexual dimorphism correlations are established between the cephalometric characteristics of the upper respiratory tract itself and the hyoid bone or tongue, as well as between the cephalometric characteristics of the soft palate and the tongue.

The pathogenicity of bacterial strains isolated from pathological processes and lesions of horses, strategies for their treatment and the choice of appropriate antimicrobials are frequently a challenging problem for private veterinarians who seek help in our laboratory. Therefore, the aim of this study was to map genera and species of Gram-negative aerobic and microaerophilic microorganisms isolated from pathological processes in horses and to identify the most effective antimicrobial agents for therapy based on antibiotic susceptibility. Between 2009 and 2014 a total of 449 clinical samples (n = 449) were examined; 229 (51%) of them were obtained from the respiratory tract, 121 (27%) from the skin, 40 (8.9%) from the digestive tract, 40 (8.9%) from the eyes, eight (1.8%) from the urinary system, six (1.3%) from the musculoskeletal system, four (0.9%) from the lymphatic system and one (0.2%) from milk. The examination was performed using conventional microbiological culture methods. The identification of isolates was confirmed using MALDI-TOF molecular phenotyping (Bruker Daltonics GmbH, Bremen, Germany). From the 276 Gram-negative isolates (prevalence of 61.5%), the most frequently detected strains were Enterobacter spp., Escherichia spp., Acinetobacter spp., Pseudomonas spp. and Actinobacillus spp. with prevalence rates of 7.6%, 6.7%, 6.7%, 6.0% and 5.8%. In addition, another 20 genera of microorganisms were detected. Susceptibility to antimicrobial agents was determined using the disc diffusion method. The most effective agents were gentamicin (94.1%), enrofloxacin (91.7%), colistin (87.0%), florfenicol (86.2%), neomycin (85.5%), streptomycin (82.4%) and tetracycline (78.5%). A good knowledge of the spectrum of bacterial species participating in pathological processes and lesions in horses and their antimicrobial susceptibility may be of great importance not only in treatment but also in deciding which prophylactic antibiotics to administer after surgical interventions.

Total knee arthroplasty (TKA) is a widely accepted and successful procedure for end-stage arthritis. Nevertheless, fast-track may be compromised by many factors, such as pain, edema, and blood loss. Cryotherapy has been advocated as a safe and effective strategy to improve the postoperative results, acting on pain, edema, and blood loss. This study is a prospective randomized controlled study, involving 50 patients after primary TKA. A power analysis was performed preoperatively. Twenty-four patients were addressed to a postoperative treatment with a continuous cold flow device (Hilotherm, Hilotherm GmbH, Germany). Twenty-six patients represented the control group, treated with crushed ice packs. All the patients shared the same analgesic strategy and the same rehabilitation protocol. Pain, analgesic consumption, active knee range of motion, drain output, transfusion requirement, and total blood loss were evaluated at different follow-ups (postoperative first, third, and seventh days). The two groups were homogenous for preoperative and intraoperative features. The groups showed no statistically significant differences in all the evaluated parameters. A modest reduction of knee volume was evident after 7 days from surgery (trend). No differences in blood loss were noticed. Continuous cold flow device in the acute postoperative setting after TKA did not show superiority in reducing edema, pain, and blood loss, compared with traditional icing regimen. Thus, due to the costs, it should be reserved to selected cases.

Abstract Introduction Leukemia stem cells (LSC) in chronic myeloid leukemia (CML) are important in disease progression and relapse. Conventional therapy with tyrosine kinase inhibitors (TKIs), although very effective at reducing bulk CML cells, frequently fail to eliminate LSC residing in the protective bone marrow niche. Thus, there is an unmet need for combination therapy that simultaneously targets bulk disease and eliminates LSC in order to prevent disease progression and relapse. Stem cell niches are often rich in hyaluronic acid. CD44, the main receptor for hyaluronic acid, and some of its splice variants are frequently overexpressed on cancer stem cells, including LSC. In this study we aimed to evaluate the in vivo anti-LSC activity of CD44 monoclonal antibody (mAb) RG7356. Methods and Results For anti-CD44 LSC inhibition studies, immunodeficient RAG2-/-γc-/- neonatal mice were intrahepatically transplanted with human BC LSC from imatinib responsive and resistant patients. Once engraftment was verified, mice were treated with control IgG1 (30mg/kg/week), dasatinib (25mg/kg/day), CD44 mAb (RG7356) (30mg/kg/week) or a combination of CD44 mAb (RG7356) and dasatinib. The mice were sacrificed after two weeks of therapy and bone marrow, spleen, peripheral blood and myeloid sarcomas were analyzed by FACS to assess CML LSC burden. In the imatinib responsive patient, CD44 mAb single agent therapy showed an 85% reduction of LSC in bone marrow, 98% reduction in spleen and 91% reduction in peripheral blood. CD44 mAb therapy thus showed an efficacy equal to that of dasatinib, which showed an 87%, 98% and 98% reduction in bone marrow, spleen and peripheral blood respectively. Combination therapy (CD44 mAb + dasatinib) completely eradicated any trace of CML LSC in all tissues analyzed. In the imatinib resistant patient, dasatinib very effectively reduced the number of myeloid sarcomas and blasts but did not inhibit LSC survival in bone marrow. In contrast, CD44 mAb single agent therapy effectively reduced the number of CML LSC to 51% in bone marrow, 96% in spleen and 93% in peripheral blood. Conclusions This study demonstrates that a human CD44 specific mAb, RG7356, significantly reduces CML LSC survival. Notably, TKI resistance of CML LSC, can be overcome by treatment with a human CD44 specific mAb, RG7356, as it sensitizes CML LSC residing in malignant niches to dasatinib. Abstract Vestibular voice includes participation of larynx structures which are absent in physiological process. Vestibular phonation may be desired when vocal folds are damaged as in paralytic dysphonia, or undesired in marginal hyperfunction. Vestibular voice may result from psychogenic dysphonia – phononeurosis. The aim of the study is perceptive evaluation of vestibular voice, objective larynx visualization, acoustic and aerodynamic examination. The study included 40 patients: 20 with vestibular voice, 20 with euphonic voice. Voice quality has been evaluated using perceptual GRBAS scale. Endoscopic and stroboscopic larynx examination used Endo-STROB-EL-Xion GmbH with visual tract. High-Speed Digital Imaging (HSDI) and High Speed (HS) camera registered true vocal folds vibrations. Acoustic evaluation of voice with DiagnoScope Specjalista, DiagNova Technologies included analysis of F0, Jitter, Shimmer, NHR, nonharmonic components. MPT has been analyzed. In examined group, hoarseness (95%), roughness (75%) and voice strain (55%) have been recorded. Endoscopy revealed edema of vestibular folds with dilation of vessels covering glottis. Stroboscopy and HSDI confirmed coexistence of hyperfunctional (95%) or paralytic (5%) dysphonia. Acoustic assessment revealed increase in Jitter, Shimmer, NHR and decrease in F0 and MPT. The vestibular voice is observed most frequently in women with hyperfunctional dysphonia (phononeuroses) or in paralytic dysphonia. Visualization techniques confirm the coexistence of vestibular folds hypertrophy and edema with vibration disorders. In the perceptual assessment, vestibular voice was hoarse, rough and strained. Acoustic examination showed increase of Jitter, Shimmer, NHR, presence of nonharmonic components and decrease of F0 and MPT.

The article presents the results of field experiments on the use of micro fertilizers on sunflower. The object of the study was a hybrid of sunflower Fausto from the company "Strube D&S GMBH". Field experiments were carried out with micro fertilizers of the German company "Lebozol", the Russian "Polydon Agro" and the Lithuanian "Acoustic Biotechnologies (brand Humic Land"). Leaf fertilizing with micronutrients and their treatment of sunflower seeds and their combination were studied. As studies have shown, the variant with the use of micronutrients for seed dressing against the background of leaf fertilizing showed the greatest effectiveness. Leaf fertilizing without treating seeds with trace elements did not significantly in-crease the yield and oil yield. Also, the oil content of the seeds according to the variants in the years of re-search did not differ significantly. In general, the condition of sunflower plants during the years of re-search before harvesting was satisfactory. At the same time, it should be noted that in 2022 abnormal pre-cipitation affected the quality of sunflower seeds. The analysis of the obtained results indicates the need for further search for optimal ways of using micronutrients to increase the yield of sunflower and its quali-ty. It should also be taken into account the fact that according to economic calculations, the costs of ap-plying micro fertilizers during seed treatment will not exceed 10 rubles/ ha, and with leaf fertilizing they will amount to 1300-2300 rubles/ ha. accordingly.

STDs are usually experienced by teenagers, adults, and the elderly due to deviant behavior such as free sex, anal sex, oral sex, or because of direct contact with sufferers through the genital tract, skin touch, vaginal fluids, sperm fluid and sexual intercourse that is not use condoms and other sexual safety devices. Factors that influence adolescent behavior are personal experience, the influence of others, cultural influences, mass media, educational institutions, religion and emotional factors. Therefore, it is necessary to conduct education and socialization on the prevention of sexually transmitted diseases using lecture methods, question and answer and discussion as well as aids or poster media used. From the results of the education and socialization activities, it is hoped that it can prevent sexually transmitted diseases and increase the generation of healthy living in adolescents at GMIH Eben Haezer Church, Ternate.

Background: It is clear that deficiencies of some trace elements cause marked alterations in lipid and lipoprotein metabolism. The mechanisms of their effects are not completely obvious and inspite of intense research, the role of these microelements need further elucidation. Thus, the objective of this study is to evaluate the correlations of serum concentrations of trace elements including copper with lipid profile parameters of adult men in Southern Rajasthan.Methods: Present study was conducted in Cardiology and Biochemistry Department in GMCH, Udaipur during the year 2019. A total of 150 MI cases/controls were selected. Blood sample was collected by vein puncture using an aseptic technique. The blood was allowed to settle at 37ºC and then centrifuged at 3000 rpm for 15 min. Serum was then separated and stored at -20ºC until analysis.Results: The study cases had significantly higher lipid values than those of controls (p<0.001). In cases the copper levels are higher than the controls. Copper levels were more in males 95.40±18.93 than in females 86.93±14.54 (p<0.05). Lipid levels were higher in urban population related to rural, TC (238.47 V/s 266.59), TG (219.83 V/s 202.40), HDL (64.39 V/s 54.19) and LDL (131.28 V/s 139.48) (p<0.05). Significant negative correlation was seen in study group between serum copper levels and TC (r=-0.288, p<0.05), TG (r =-0.236, p<0.05), HDL (r=-0.946, p<0.05) and VLDL levels (r=0.102, p<0.05). This indicates that as copper level reduces lipids increases.Conclusions:Our findings indicate the possible effect of Cu level in serum lipid profile and this effect may be due to the role of Cu as an antioxidant. The correlations between the serum concentrations of trace elements with lipid profile in physiological concentrations may not be the same as the changes observed during deficiencies of the trace elements as in hyperlipidemic patients

Background:Apremilast (APR) has been studied extensively in phase III randomized, controlled trials. However, real-world information is limited on the effectiveness and safety of APR in patients with psoriatic arthritis (PsA).Objectives:To assess the effectiveness and safety of APR in a large cohort of patients with active PsA from routine clinical practice settings in Germany.Methods:In this multicenter, prospective, non-interventional study, the primary endpoint was the proportion of patients reaching ≥1 point (≥20%) improvement from baseline (BL) in the Physician’s Global Assessment of Disease Activity (PhGA) score. Other endpoints included effects on swollen and tender joint counts (SJC/TJC), psoriasis-involved body surface area (BSA), enthesitis, dactylitis, Patient’s Global Assessment of Disease Activity score (PtGA), Psoriatic Arthritis Impact of Disease (PsAID) tool, pain, and pruritus. The current analysis is based on observed data through 1 year of study duration.Results:A total of 545 German patients were enrolled and followed up to 12 months of APR treatment; 488 patients were included in the safety analysis and 418 in the full analysis set. The mean age was 55 years, mean body mass index was 29.5 kg/m2, and 60% were female. The mean duration of psoriasis and PsA was 26 years and 18 years, respectively. At BL, 46.7% of patients had enthesitis based on the Leeds Enthesitis Index (LEI; mean [SD]: 2.9 [1.72]) and 23% had dactylitis (mean [SD]: 2.2±2.03); 74% of patients were biologic-naive. Effectiveness is shown after ~1 month (Visit 1 [V1]) and ~4 months (Visit 2 [V2]) and for up to 225 patients after 12 months (Visit 5 [V5]) of treatment (Table).Improvements were also seen in PtGA, overall pain, and pruritus. A subanalysis suggests APR was associated with greater benefits in biologic-naive patients compared with patients who previously received biologic therapy. Observed safety and tolerability through V5 were consistent with the known overall safety profile of APR. Common adverse events in clinical trials were similar, with a lower incidence of diarrhea (11.1%), nausea (7.0%), headache (3.9%), and respiratory tract infection (0.8%).Conclusion:Data from this large, real-world cohort of patients with PsA show the effectiveness of APR. In patients with up to 12 months of follow-up, APR was associated with rapid and maintained improvements in physician-assessed and patient-reported outcomes. ABSTRACT Having previously shown that CCL20/macrophage inflammatory protein 3α and tumor necrosis factor alpha (TNF-α) are released by polarized primary rat uterine epithelial cells (UEC) in response to Escherichia coli but not to Lactobacillus rhamnosus, we sought to determine if epithelial cells are responsive to pathogen-associated molecular patterns (PAMP), including lipopolysaccharide (LPS), lipoteichoic acid (LTA), and Pam3Cys, a bacterial lipoprotein analog. Epithelial cells were grown to confluence on Nunc cell culture inserts prior to apical treatment with PAMPs. In response to LPS, LTA, and Pam3Cys (EMC Microcollection GmbH, Tübingen, Germany), CCL20 levels increased (4- to 10-fold) while PAMPs caused increased TNF-α (1- to 4-fold) in the medium collected after 24 h of incubation. Both apical and basolateral secretion of CCL20 and TNF-α increased in response to PAMPs, but treatments had no effect on cell viability and integrity, as measured by transepithelial resistance. Time course studies of CCL20 and TNF-α release in response to Pam3Cys and LPS indicated that CCL20 release peaked between 2 and 4 h after treatment, whereas TNF-α release was gradual over the length of the incubation. Freeze-thaw and cell lysis experiments, along with actinomycin D studies, suggested that CCL20 and TNF-α are synthesized in response to PAMP stimulation. Taken together, these studies demonstrate that E. coli and selected PAMPs have direct effects on the production of CCL20 and TNF-α without affecting cell integrity. Since CCL20 is known to be both chemotactic and antimicrobial, the increase in apical and basolateral release by UEC in response to PAMPs suggests a new mechanism of innate immune protection in the female reproductive tract.

Abstract. In Germany, the Federal Company for Radioactive Waste Disposal (BGE) is responsible for the construction and operation of deep, geological repositories. BGE operates the Asse II mine and the Konrad and Morsleben repositories. Initially, these mines were excavated to extract raw materials. As a result, they have large void volumes on multiple levels. The stability of the unfilled cavities is limited in time so that backfilling measures are required for stabilisation. However, there are many requirements to consider when selecting backfill materials. BGE aims at making its locations climate neutral. Backfilling measures are particularly affected by this effort because of the large material volumes required and the greenhouse gas emissions that are released during material transport and during the entire material production chain. Bulk materials that are produced from the rock debris from the excavation of the respective mine have the smallest carbon dioxide (CO2) footprint. In order to use this rock debris, this material will be stored in underground excavations and large voids. In order to backfill the underground cavities in accordance with the requirements, the debris will be crushed in an underground crushing plant prior to backfilling. However, numerous cavities in the three mines mentioned above can only be backfilled with flowable, self-levelling and self-hardening materials. Generally, cement-based backfill materials and magnesia binders can be distinguished. The CO2 footprint of mixtures with cement increases primarily with the proportion of Portland cement clinker. For this purpose, recipes containing high proportions of supplementary cementitious materials such as pozzolan and latent hydraulic additives have been developed. Magnesia binders contain the binders magnesium oxide and/or magnesium hydroxide. Depending on the raw materials and the manufacturing process, different amounts of greenhouse gases are generated during their production. The classic approach is to use magnesium oxide, which is obtained by calcining magnesite (MgCO3). However, a lot of energy is required for this process, and the carbon dioxide content of the magnesite is released. Due to the extensive experience in the development of backfill materials and emplacement technologies, BGE has entrusted BGE TECHNOLOGY GmbH with the optimisation of the procedures for backfilling and the development of new backfill materials with the lowest possible CO2 footprint. Thus, raw materials and supply chains have been evaluated and are improved with regard to their environmental performance, taking into account all aspects of transport and logistics. The current focus of work is on the development of magnesia binders with natural brucite (magnesium hydroxide). Another positive aspect is the reduction of heat development of the backfill material during hardening. Furthermore, the reliability of supply for the implementation of the backfilling measures can be increased by a market analysis. The examples show that BGE and BGE TECHNOLOGY GmbH are on the right track to protect humans and the environment through the safe disposal of radionuclides and the realisation of sustainable, future-oriented technologies.

ObjectivesIn line with the child survival and gender equality targets of Sustainable Development Goals (SDG) 3 and 5, we aimed to: (1) estimate the age and sex-specific mortality trends in child-related SDG indicators (ie, neonatal mortality rate (NMR) and under-five mortality rate (U5MR)) over the 1960s–2017 period, and (2) estimate the expected annual reduction rates needed to achieve the SDG-3 targets by projecting rates from 2018 to 2030.DesignGroup method of data handling-type artificial neural network (GMDH-type ANN) time series.MethodsThis study used an artificial intelligence time series (GMDH-type ANN) to forecast age-specific childhood mortality rates (neonatal and under-five) and sex-specific U5MR from 2018 to 2030. The data sets were the yearly historical mortality rates between 1960s and 2017, obtained from the World Bank website. Two scenarios of mortality trajectories were simulated: (1) status quo scenarios—assuming the current trend continues; and (2) acceleration scenarios—consistent with the SDG targets.ResultsAt the projected rates of decline of 2.0% for NMR and 1.2% for U5MR, Nigeria will not achieve the child survival SDG targets by 2030. Unexpectedly, U5MR will begin to increase by 2028. To put Nigeria back on track, annual reduction rates of 7.8% for NMR and 10.7% for U5MR are required. Also, female U5MR is decreasing more slowly than male U5MR. At the end of SDG era, female deaths will be higher than male deaths (80.9 vs 62.6 deaths per 1000 live births).ConclusionNigeria is not likely to achieve SDG targets for child survival and gender equities because female disadvantages will worsen. A plausible reason for the projected increase in female mortality is societal discrimination and victimisation faced by female child. Stakeholders in Nigeria need to adequately plan for child health to achieve SDG targets by 2030. Addressing gender inequities in childhood mortality in Nigeria would require gender-sensitive policies and community mobilisation against gender-based discrimination towards female child.

Objective: Ultra-high-field B0 ≥ 7 tesla (7T) cardiovascular magnetic resonance (CMR) offers increased resolution. However, electrocardiogram (ECG) gating is impacted by the magneto-hydrodynamic effect distorting the ECG trace. We explored the technical feasibility of a 7T magnetic resonance scanner using an ECG trigger learning algorithm to quantitatively assess cardiac volumes and vascular flow. Methods: 7T scans were performed on 10 healthy volunteers on a whole-body research MRI MR scanner (Siemens Healthineers, Erlangen, Germany) with 8 channel Tx/32 channels Rx cardiac coils (MRI Tools GmbH, Berlin, Germany). Vectorcardiogram ECG was performed using a learning phase outside of the magnetic field, with a trigger algorithm overcoming severe ECG signal distortions. Vectorcardiograms were quantitatively analyzed for false negative and false positive events. Cine CMR was performed after 3rd-order B0 shimming using a high-resolution breath-held ECG-retro-gated segmented spoiled gradient echo, and 2D phase contrast flow imaging. Artefacts were assessed using a semi-quantitative scale. Results: 7T CMR scans were acquired in all patients (100%) using the vectorcardiogram learning method. 3,142 R-waves were quantitatively analyzed, yielding sensitivity of 97.6% and specificity of 98.7%. Mean image quality score was 0.9, sufficient to quantitate both cardiac volumes, ejection fraction, and aortic and pulmonary blood flow. Mean left ventricular ejection fraction was 56.4%, right ventricular ejection fraction was 51.4%. Conclusion: Reliable cardiac ECG triggering is feasible in healthy volunteers at 7T utilizing a state-of-the-art three-lead trigger device despite signal distortion from the magnetohydrodynamic effect. This provides sufficient image quality for quantitative analysis. Other ultra-high-field imaging applications such as human brain functional MRI with physiologic noise correction may benefit from this method of ECG triggering.

The aim of the study was to compare the hypolipidemic activity of the developed self-emulsifying drug delivery systems with simvastatin with reference samples of the substance and the finished drug product of industrial production. Materials and methods. The substance of simvastatin (India, s. DK40-2005021, 99.09 %, introduced into the composition of self-emulsifying compositions based on castor oil (Ukraine), polyethylene glycol 40 hydrogenated castor oil (India), Tween 80 (Ukraine), glycerol monostearate (Gustav Heess GmbH, Germany) or polyethylene glycol 100 stearate (ERCA, Italy). Reference samples were Simvastatin-Sandoz (Salutas Pharma, Germany, series LX5161) and simvastatin in pure form. The experimental animals were Syrian hamsters aged 2 months. Hyperlipidemia was modeled by means of an alimentary load. To assess the state of lipid metabolism in animals, the content of triacylglycerols, total cholesterol, low-density lipoprotein and high-density lipoprotein in the blood serum was determined by colorimetric enzymatic methods using the appropriate standard reagent kits "Triacylglycerols F" HP022.02, "Cholesterol F" HP026.02, "Cholesterol-LDL F" HP026.05 and "Cholesterol-HDL F" HP026.04 (LLC SPE "Filicit-Diagnostics", Ukraine) on a semi-automatic biochemical analyzer MapLab Plus (BSI, Italy). Results. The reference samples had similar dose-dependent efficacy parameters. At the same time, the test samples, also having similar dose-dependent effects, in absolute terms at the maximum concentration reduced the amount of low-density lipoprotein and total cholesterol more effectively than the reference samples. When using the test samples in their average concentration, the level of triglycerols was significantly reduced, which is rather a concomitant effect of simvastatin. Conclusions. The improvement of the overall efficacy of simvastatin when it is introduced into self-emulsifying drug delivery systems has been proved, which is associated with the modification of pharmacokinetic parameters by improving the solubility of the substance in the aqueous environment of the gastrointestinal tract

Carbohydrates play a key role in different reproductive events such as the sperm–oviductal cell interaction and sperm–oocyte recognition. In this way, α-d-mannosyl (Amari et al. 2006 Mol. Reprod. Dev. 59, 221–226) and α-2,3-sialic acid (Velasquez et al. 2006 Mol. Reprod. Dev. in press) residues contained in the zona pellucida have been identified as sperm receptors in bovine oocytes. The glycosidases, enzymes that remove carbohydrates, could play an important role in the reproductive tract, modulating decisive physiological events mediated by carbohydrates. However, the enzymatic activity level of these enzymes or its fluctuations throughout the estrous cycle in the bovine oviductal fluid (BOF) has not been studied. The objective of this work was to compare the enzymatic activity level of 7 different glycosidases in the oviductal fluid of cows at different stages of the estrous cycle. Oviducts were collected from the abattoir and classified according to the macroscopic aspect of the genital tract (Grippo et al. 1995 J. Reprod. Fertil. 105, 57–64) as early follicular (presence of growing follicles), late follicular (presence of a dominant follicle), early luteal phase (ovaries showing a corpus hemorrhagicum or a recent corpus luteum), and late luteal phase (old corpus luteum or corpus albicans). Oviductal fluid samples were collected by aspiration with an automatic pipette making simultaneous manual pressure from the isthmus toward the ampulla. Samples (9 per group) were centrifuged (7000g, 10 min) and supernatant was stored at −20°C until assay. Total activity levels were fluorimetrically measured at 450 nm, with the corresponding substrate conjugated to 4-methylumbelliferil for each enzyme (Abascal et al. 1998 Biochem. J. 333, 201–207) using a fluorometer Fluostar Galaxy (BMG LABTECH GmbH, Offenberg, Germany). Enzymatic assays were done in duplicate for 4 h at 37°C, and the reactions were stopped by adding glycine-calcium carbonate buffer. Fluorescences were corrected for tissue and substrate blanks. Fluorescence results of each enzyme and oviduct phase were analyzed using a 1-way ANOVA, with estrous cycle phase being the main factor. Results (mean counts of fluorescence) showed that the level of activity changes during the estrous cycle and the activity of some enzymes increases close to or after ovulation, suggesting a role of some glycosidases in the fertilization process. Preliminary assays for neuraminidase were negative for all samples. Future studies are necessary to identify the biological role played by the glycosidases present in the bovine oviductal fluid. Background: Acute bronchiolitis is a disorder of the lower respiratory tract that occurs mostly in between 1 month to 24 months. There are limited numbers of studies on magnesium sulphate nebulization in acute bronchiolitis. There is a desperate need for a standard treatment which can reduce the morbidity and mortality in acute bronchiolitis. The goal was to study the efficacy of magnesium sulphate nebulization in infants with acute bronchiolitis.Methods: It was a prospective open labelled randomized controlled study. Children in age group of 2-12 months admitted with clinical diagnosis of acute bronchiolitis in paediatric emergency ward, GMSH-16, Chandigarh. Study group patients were treated with 40 mg/kg magnesium sulphate nebulization diluted with 2-3 ml normal saline in addition to supportive treatment as oxygen and IV fluids, three doses of medication were given at 1 hour interval and it was not repeated after three doses. Control group patients were treated with oxygen therapy, IV fluids, symptomatic treatment for fever and supportive care.Results: Respiratory distress assessment instrument score and length of hospital stay was significantly lesser in study group as compared to control group. SpO2 was significantly more in study group as compared to control group.Conclusions: Magnesium sulphate nebulization is effective in improving respiratory distress and oxygen saturation, it also reduces duration of hospital stay in infants with mild to moderate bronchiolitis.

Woman’s reproductive tract often has problems, especially in women of childbearing age. One of them is Bacterial Vaginosis is a clinical condition that occurs a lot. BV infection is a polymicrobial infection caused by a decrease in the amount of Lactobaclilus with anaerobic bacteria that increases excessively. Alternative treatment for BV is given by using the results hydrolysates of konjac plant extracted in dosage forms Glucomannan Hydrolysates (GMH) containing glucose and mannose prebiotic which can support the growth development of Lactobacillus in the vaginal mucosa. This study was to determine the effect of Glucomannan Hydrolisates (GMH) + Metronidazole on levels of IL 23 cytokines in BV of reproductive age women. The method of this study used the True Experimental design with the type of research The randomized pretest-posttest was in vivo. The subjects selected in this study design used Randomized techniques. A sample of 7 people WUS with BV given GMH 300mg + Metronidazole 1000 mg for 9 days of use, To assess the levels of cytokine IL 23 using the ELISA method. Data analysis using a ratio scale was analyzed using the metric statistical test, normality test using Shapiro-Wilk test and comparative test using Repeated Measure ANOVA. The overall results of the analysis showed that the combination of GMH and Antibiotics Metronidazole could significantly increase the levels of cytokine IL 23 compared to other therapies to treat bacterial vaginosis in women of childbearing age

The transformation of our energy system will likely require the indirect electrification of the sectors heating, chemical industry and mobility by power-to-X processes, because not all processes can easily use electrical power directly. The high-temperature co-electrolysis using solid-oxide cells (SOCs) can produce syngas compositions desired by the downstream power-to-X process without the need of a reverse water-gas shift reactor and is therefore particularly well suited to produce synthesis gas as feedstock for various power-to-fuel and power-to-chemicals processes. Besides, a coupling of heat-producing downstream processes would increase system efficiency significantly.1 Besides the intrinsic degradation processes of the cells (e.g. nickel migration) and stacks (e.g. contact deterioration) any additional processes triggered by impurities in the feed gases are of relevance for any technical applications. Under certain circumstances, impurities can be introduced by Balance of Plant (BoP) components like the steam generators or preheaters that can release silicon species.2 Also, the feed gases themselves may contain harmful substances. This poisoning effect must be taken into account, because economically gases of lower qualities are preferred as purification steps increase the costs of the syngas. Depending on the exact source of carbon dioxide (CO2) the supplier uses, it contains different secondary components, of which sulfur species are of particular concern. In contrast to fuel-cell mode operation, in electrolysis mode operation hydrogen sulfide (H2S) is not the most likely candidate, but sulfur dioxide (SO2). The reasons for this is that CO2 may be obtained from combustion processes and that hydrogen sulfide is not stable in the atmosphere but has a short half-life time. Therefore, understanding the influence of SO2 contained in the supplied carbon dioxide on the performance of the co-electrolysis is of great importance. There is substantial knowledge about the impact of H2S in feed gases in fuel-cell mode, but the impact of sulfur dioxide in electrolysis mode is not as well investigated. Skafte et al. reported a severe effect in CO2 electrolysis with as low as 20 ppb H2S.3 In the context of this application, the relevant sulfur species present in the feed gases will be SO2. It should be noted, that the heterogenous reduction of SO2 with hydrogen or carbon monoxide is catalyzed by oxides with oxygen vacancies and mobility like CeO2 or YSZ and facilitated by the presence of transition metals.4 Intermediary steps and products beside H2S can include significant amounts of elemental S and COS and the formation of sulfided nickel seems likely. The formation of these products depends on the exact reaction conditions, e.g. temperature, H2/SO2 ratio, steam content, which makes the situation more complex as in fuel-cell mode with hydrogen as fuel. Jeanmonod et al. came to the conclusion, that even concentrations of SO2 as low as 0.5 ppm caused irreversible degradation in Ni/YSZ electrodes in co-electrolysis mode.5 This contribution will report about investigations performed on the degradation caused by sulfur dioxide and silicon species in sub-ppm concentrations in SOCs with Ni/YSZ and Ni/GDC fuel electrodes operated in co-electrolysis mode, using stacks in the Jülich F10 and F20 design. These targeted their impact, potential recovery, possible damage mechanisms and safe thresholds of sulfur dioxide in carbon dioxide. For this the authors performed long-term stationary operation, monitoring of poisoning experiments by electrochemical impedance spectrocopy and post-test analysis of cells, including SEM/EDX and chemical trace analyses. 2083 Background: PreCycle (NCT03220178), a multicenter, randomized phase IV Intergroup trial evaluates the impact of ePRO assessment on quality of life (QoL) in HR+/HER2- locally advanced or metastatic breast cancer patients (pts) treated by palbociclib (P) and an aromatase inhibitor or P+fulvestrant. Pts willing to use the web/APP-eHealth solution CANKADO are eligible. Patients are randomized (2:1, stratified by therapy line) to the active (CANKADO PRO-React) or inactive inform arm. Primary endpoint is time to deterioration (TTD) of QoL. Methods: The trial started in 2017 and is ongoing (81 centers); regular safety reports are routinely provided to the study sponsor. Analysis of distribution of serious adverse events (SAE) was initiated by the trial leadership and performed using the Oct 15, 2019 safety report. Data that could bias primary or secondary endpoints were not analyzed. Bayesian inference (non-informative prior) was used to estimate probabilities; no corrections for potential multiplicities were made. Results: At data cut-off, 261/281 randomized patients had received study medication and provided CANKADO documentation. At time of evaluation, a total of 40298 days were documented. CANKADO was used on 59% (+/-10%) of all days over a 2-year period. SAEs were observed in 26/175 (14.9%) of all active-arm patients vs. 18/86 (20.9%) of inform-arm patients (90% probability of reduction in inform patients). Total SAEs were 36 (active) vs. 27 (inform); corresponding SAE incidence per hundred patients was 20.6 vs. 31.4, a relative reduction of about one-third. Conclusions: CANKADO is well accepted and used regularly by pts in PreCycle, so far over a 2-year period. The present (unplanned) analysis suggests a potentially substantial, clinically relevant reduction in relative SAE incidence among 1stL pts using PRO-React, with a more modest decrease overall. This analysis is preliminary, representing a snapshot, and cannot provide a definitive explanation for the observed SAE reduction. PreCycle will continue to enroll patients in order to further evaluate the potential benefits of interactive eHealth support. This research aimed to substantiate the potential practicality of utilizing a matrix-like platform, a novel 3D-printed biomaterial scaffold, to enhance and guide host cells’ growth for bone tissue regeneration. The 3D biomaterial scaffold was successfully printed using a 3D Bioplotter® (EnvisionTEC, GmBH) and characterized. Osteoblast-like MG63 cells were utilized to culture the novel printed scaffold over a period of 1, 3, and 7 days. Cell adhesion and surface morphology were examined using scanning electron microscopy (SEM) and optical microscopy, while cell viability was determined using MTS assay and cell proliferation was evaluated using a Leica microsystem (Leica MZ10 F). The 3D-printed biomaterial scaffold exhibited essential biomineral trace elements that are significant for biological bone (e.g., Ca-P) and were confirmed through energy-dispersive X-ray (EDX) analysis. The microscopy analyses revealed that the osteoblast-like MG63 cells were attached to the printed scaffold surface. The viability of cultured cells on the control and printed scaffold increased over time (p < 0.05); however, on respective days (1, 3, and 7 days), the viability of cultured cells between the two groups was not significantly different (p > 0.05). The protein (human BMP-7, also known as growth factor) was successfully attached to the surface of the 3D-printed biomaterial scaffold as an initiator of osteogenesis in the site of the induced bone defect. An in vivo study was conducted to substantiate if the novel printed scaffold properties were engineered adequately to mimic the bone regeneration cascade using an induced rabbit critical-sized nasal bone defect. The novel printed scaffold provided a potential pro-regenerative platform, rich in mechanical, topographical, and biological cues to guide and activate host cells toward functional regeneration. The histological studies revealed that there was progress in new bone formation, especially at week 8 of the study, in all induced bone defects. In conclusion, the protein (human BMP-7)-embedded scaffolds showed higher regenerative bone formation potential (week 8 complete) compared to the scaffolds without protein (e.g., growth factor; BMP-7) and the control (empty defect). At 8 weeks postimplantation, protein (BMP-7) significantly promoted osteogenesis as compared to other groups. The scaffold underwent gradual degradation and replacement by new bones at 8 weeks in most defects.

BackgroundFew data are available on the progression and management of ILD, or the management of adverse events associated with drug treatment, in patients with lcSSc. SENSCIS-ON is an open-label extension trial that is collecting data on decline in forced vital capacity (FVC) and adverse events in patients treated with nintedanib over the long term.ObjectivesTo assess decline in FVC and adverse events in patients with lcSSc and ILD treated with nintedanib in SENSCIS-ON.MethodsPatients with SSc-ILD were eligible to enter SENSCIS-ON if they completed the randomized placebo-controlled SENSCIS trial (in which patients received trial drug until the last patient reached week 52 but for ≤100 weeks) or a drug–drug interaction (DDI) study of nintedanib and oral contraceptive (in which female patients received nintedanib for ≤28 days). Among patients with lcSSc, we analysed changes from baseline in FVC and adverse events over 52 weeks of SENSCIS-ON in patients who received nintedanib in SENSCIS and continued it in SENSCIS-ON (“continued nintedanib” group) and in patients who received placebo in SENSCIS and initiated nintedanib in SENSCIS-ON or who received nintedanib for a short time in the DDI study (“initiated nintedanib” group). Analyses were descriptive.ResultsThere were 98 patients with lcSSc in the continued nintedanib group and 127 patients with lcSSc (114 from SENSCIS, 13 from the DDI study) in the initiated nintedanib group. In these groups, respectively, mean (SD) FVC values at inclusion in SENSCIS-ON were 2449 (662) mL and 72.7(16.7) % predicted and 2508 (771) mL and 74.1 (17.4) % predicted. Mean (SE) changes in FVC from baseline to week 52 of SENSCIS-ON were −45.1 (19.1) mL in the continued nintedanib group, −41.5 (24.0) mL in the initiated nintedanib group, and −43.3 (15.3) mL in all patients with lcSSc, similar to the change in FVC in patients with lcSSc at week 52 of the SENSCIS trial (−39.1 [22.2] mL). The adverse event profile of nintedanib in SENSCIS-ON was consistent with that reported over 52 weeks of the SENSCIS trial (Table 1). Abstract Background Lefamulin is a first-in-class, oral and IV pleuromutilin antibiotic approved in the US, EU, and Canada for the treatment of community-acquired bacterial pneumonia (CABP) in adults. Lefamulin inhibits bacterial protein synthesis via a unique mechanism of action and its potency against S. aureus has been well established. We evaluated the in vitro activity of lefamulin against S. aureus from patients with cystic fibrosis (CF). Methods Unique isolates (n=224) were collected from the lower respiratory tract (LRT) of children (≤17 years old) with CF and LRT infection. Organisms were from qualified respiratory specimens and determined to be the probable cause of infection by the participant center. The isolates were collected in 2018-2020 from 22 medical centers in 11 countries and tested by broth microdilution methods at JMI Laboratories. Most isolates were from the US (43.3%), Spain (24.1%), France (20.5%), and Costa Rica (7.1%). Results Lefamulin was highly active against the CF S. aureus collection (MIC5090, 0.06/0.12 mg/L), with 99.6% of isolates inhibited at ≤0.25 mg/L, consistent with the susceptible [S] breakpoint published by the US FDA, CLSI, and EUCAST. Only 1 lefamulin-non-S (MIC, 1 mg/L) isolate was observed, a methicillin-susceptible (MSSA) collected in Costa Rica in 2018 and carrying a vga(A) gene. Lefamulin retained potent activity against methicillin-resistant (R) S. aureus (MRSA, n=52; MIC50/90, 0.06/0.12 mg/L), azithromycin-R (n=115; MIC50/90, 0.06/0.12 mg/L), levofloxacin-R (n=23; MIC50/90, 0.06/0.12 mg/L), clindamycin-R (n=11; MIC50/90, 0.06/0.12 mg/L), and gentamicin-R (n=9; MIC range of 0.03-0.12 mg/L) isolates as well as those isolates with multiple resistance phenotypes. Against MRSA, susceptibility to azithromycin was 23.5% and to levofloxacin 64.7%. All isolates were susceptible to vancomycin, linezolid and ceftaroline (Table). Among isolates from the US (n=97), the MRSA rate was 30.9% and all isolates were Lefamulin-S (MIC5090, 0.06/0.12 mg/L). Conclusion Lefamulin demonstrated potent in vitro antibacterial activity against S. aureus from children with CF, regardless of resistance phenotype. Lefamulin may represent a valuable treatment option for CF patients with S. aureus LRT infections. Disclosures Helio S. The sperm–egg interactions are species-specific forms of cell recognition and the binding event which are a necessary prerequisite for fertilization (Park et al. 2002 Anim. Reprod. Sci. 72, 83–94). Glycosidase enzymes that remove carbohydrates could play an important role in the reproductive tract, modulating decisive physiological events mediated by carbohydrates, which play a key role in sperm–oocyte recognition. The aim of this study was to analyze the presence of the glycosidases α-D-mannosidase, α-L-fucosidase, β-D-glucosaminidase, and β-D-galactosaminidase in intact and acrosome-reacted sperm from fertile matured boars. Sperm were washed three times in PBS by centrifugation at 800g for 10 min. The pelleted sperm were resuspended in the same buffer to obtain a final concentration of 250 � 106 spermatozoa mL–1. The acrosome reaction was induced by incubation of the sperm with 10 µm of calcium ionophore A23187 at 37�C for 30 min. Different enzymes were detected by incubating 8 µL (for α-Dmannosidase) or 80 αL (for the rest of the enzymes) of sperm sample with the corresponding substrate conjugated to 4-methylumbelliferil for 2 h at 37�C in PBS at pH 7.3. Fluorescences were read on a Fluostar Galaxy fluorimeter (BMG LabTech GmbH, Offenburg, Germany), using wavelengths of 340 and 450 nm for excitation and emission, respectively, and were corrected by subtracting tissue and substrate blanks. The results were analyzed using a one way ANOVA. An average of fluorescence units of 9685.86 � 1081.75, 7394.63 � 874.29, 3154.17 � 514.10, and 1666.40 � 117.86 was detected in the intact sperm sample for the α-D-mannosidase, α-L-fucosidase, β-D-glucosaminidase, and β-D-galactosaminidase, respectively. For the acrosome-reacted sperm sample (60–65% acrosome-reacted sperm in the samples measured by fluorescence microscope), an average of 9756.14 � 1011.45, 7026.93 � 771.48, 1185.70 � 277.51, and 1111.60 � 176.70 for α-D-mannosidase, α-L-fucosidase, β-D-glucosaminidase, and β-D-galactosaminidase, respectively. Statistically significant differences (P < 0.05) between intact and acrosome-reacted sperm were detected only for the β-D-glucosaminidase and β-D-galactosaminidase. These results suggest that the four different enzymes detected are mainly present in the sperm plasma membrane. Under the conditions used in this study, α-D-mannosidase is the main enzyme activity present in the sperm. Importantly, β-D-glucosaminidase and β-D-galactosaminidase activity detected in the intact sperm is decreased after the induction of the acrosome reaction.
