A 10-12 kD lymphokine, herein termed TCAF, was recently shown to be secreted from Th after crosslinking of their antigen/MHC (T3-Ti) receptors. TCAF stimulates resting T lymphocyte proliferation via binding to surface components of the T11 pathway. To determine whether TCAF could induce antigen-independent activation of the lytic machinery of cytotoxic cells, the present studies were conducted. In the presence of TCAF, both T8+ class I MHC-specific and T4+ class II MHC-specific cytotoxic T cell clones were induced to kill targets, including those lacking the appropriate MHC molecules. This effect was unique to TCAF, since IL-1, IL-2, IFN-gamma could not stimulate lytic activity. Furthermore, both T3+T11+ and T3-T11+ NK clones were triggered to lyse NK-resistant target cells. These findings suggest that TCAF can function in an antigen-independent fashion to amplify cytotoxic effector responses.

The management of advanced or recurrent endometrial cancers presents a challenge due to the development of resistance to treatments. The knowledge regarding the role of the tumor microenvironment (TME) in determining the disease’s progression and treatment outcome has evolved in recent years. As a TME component, cancer-associated fibroblasts (CAFs) are essential in developing drug-induced resistance in various solid tumors, including endometrial cancers. Hence, an unmet need exists to test the role of endometrial CAF in overcoming the roadblock of resistance in endometrial cancers. We present a novel tumor–TME two-cell ex vivo model to test CAF’s role in resisting the anti-tumor drug, paclitaxel. Endometrial CAFs, both NCAFs (tumor-adjacent normal-tissue-derived CAFs) and TCAFs (tumor-tissue-derived CAFs) were validated by their expression markers. Both TCAFs and NCAFs expressed positive markers of CAF, including SMA, FAP, and S100A4, in varying degrees depending on the patients, while they consistently lacked the negative marker of CAF, EpCAM, as tested via flow cytometry and ICC. CAFs expressed TE-7 and immune marker, PD-L1, via ICC. CAFs better resisted the growth inhibitory effect of paclitaxel on endometrial tumor cells in 2D and 3D formats compared to the resistance of the tumoricidal effect of paclitaxel in the absence of CAFs. TCAF resisted the growth inhibitory effect of paclitaxel on endometrial AN3CA and RL-95-2 cells in an HyCC 3D format. Since NCAF similarly resisted the growth inhibitor action of paclitaxel, we tested NCAF and TCAF from the same patient to demonstrate the protective action of NCAF and TCAF in resisting the tumoricidal effect of paclitaxel in AN3CA in both 2D and 3D matrigel formats. Using this hybrid co-culture CAF and tumor cells, we established a patient-specific, laboratory-friendly, cost-effective, and time-sensitive model system to test drug resistance. The model will help test the role of CAFs in developing drug resistance and contribute to understanding tumor cell-CAF dialogue in gynecological cancers and beyond.

AbstractMicrobiological burden of room-air in operating theatres is a known risk factor for surgical site infections. However, it is unclear how to best evaluate efficacy and efficiency under routine clinical conditions. Moreover, there still is a lack of data to assess the impact on infection rates. The aim of this study was to evaluate a temperature-controlled ventilation system (TcAF) under routine clinical conditions and assess its impact on infection rates. 10 clinical installations of the TcAF system Opragon (Avidicare AB, Sweden) were assessed during live surgeries according to the Swedish SIS TS 39: 2015 standard. Furthermore, a retrospective analysis of 1,000 consecutive cases of primary total joint arthroplasty (hip, knee) before and 1000 after installation of the TcAF system was performed. Endpoints for clinical outcome were length of stay and infection rates. Our results show that requirements of the Swedish standard were met or significantly exceeded by the TcAF system reliably and robustly ensuring "ultra-clean" air in the entire operating theatre and demonstrating the capability to reduce the risk of airborne microbial transmission under routine clinical conditions. The study shows positive impact on key clinical outcome parameters in line with previous research by Charnley and Lidwell.

Foram avaliados os efeitos da ingestão crônica de cafeína e exercício de natação sobre o tecido ósseo. Ratos Wistar jovens (n = 32) alocados em quatro grupos: sedentário controle (SCO), sedentário + cafeína (SCAF), treinados (TCO) e treinados + cafeína (TCAF), receberam 1 mg/ml de cafeína em água durante 30 dias (SCAF e TCAF) e foram submetidos a 21 dias de natação em intensidade moderada (TCO e TCAF). A tíbia esquerda foi submetida a ensaio biomecânico para determinação do coeficiente de rigidez, limite de elasticidade e carga máxima. A área de tecido ósseo primário e espessura do disco epifisário foram avaliadas no fêmur. Não foram observadas alterações nas propriedades biomecânicas e na deposição de tecido ósseo primário. O grupo SCAF apresentou maior espessura no disco epifisário distal. Os resultados sugerem que o treinamento moderado de natação e a ingestão de cafeína não provocaram alterações morfológicas e biomecânicas do tecido ósseo.

Abstract BACKGROUND Up to date, surgical outcome of multilevel thoracic ossification of posterior longitudinal ligament (T-OPLL) with high canal occupation ratio is less satisfactory. OBJECTIVE To explore the result of thoracic column antedisplacement and fusion (TCAF) in treatment of multilevel T-OPLL with high canal occupation ratio. METHODS A total of 5 patients who underwent TCAF procedure for T-OPLL were retrospectively reviewed. Parameters including extent of OPLL, thickness of the maximal OPLL (max-OPLL), maximal canal occupying ratio (max-COR) of OPLL, effective canal diameter (ECD) at the max-OPLL level, antedisplacement distance of thoracic columns, ASIA grades, Japanese Orthopedic Association (JOA) scores, and complications were collected and analyzed at preoperation and the last follow-up. RESULTS All patients (5 F, mean age 61.0 yr, mean follow-up 18.0 months) underwent TCAF successfully and no spinal cord injury or cerebrospinal fluid leakage occurred. The mean extent of OPLL was 2.8 vertebral bodies. The mean preoperative thickness of the max-OPLL was 5.9 mm. The average antedisplacement distance of thoracic columns was 5.6 mm. The mean ECD was improved from 6.5 mm to 10.9 mm, and the max-COR was improved from 50.7% to 7.1% at last follow-up. Two patients showed improvement in ASIA grades and JOA scores were significantly improved from 5.6 points to 10.4 points at final follow-up. The overall therapeutic results of 1 patient were classified into good and 4 into fair at last follow-up. CONCLUSION TCAF may be a safe and effective procedure in treatment of multilevel T-OPLL with high canal occupation ratio.

Objective to analyze the morphological structure of cardiac, blood pressure (BP), heart rate (HR) and heart rate variability (HRV) of rats subjected to physical training with supplementation of caffeine. Methods 60 rats were divided into 4 groups: control (CO), control with caffeine (CAF), trained control (TRE) and trained with caffeine (TCAF). All trained groups underwent 4 weeks of swimming, and all caffeine groups were supplemented by voluntary ingestion of caffeine diluted in drinking water. Results there were no changes to BP and HR between groups. Regarding HRV, there was a decrease in LFnorm (low frequency) and LF/HF ratio (low and high frequency) in TCAF and CAF compared to group (p<0.02 and p<0.03, respectively). An increase occurred in CAF compared to the CO in the component LFnorm (p<0.05). The results also showed an increase in the relative weight of heart in the TRE (p<0.04) and TCAF (p<0.03) compared to CO. Conclusion caffeine did not modify the hemodynamic responses. However, physical training resulted in a decrease in sympathetic response and an increase in relative heart weight.

Abstract The transient Galactic black hole candidate MAXI J0637–430 went through an outburst in 2019–2020 for the very first time. This outburst was active for almost 6 months from 2019 November to 2020 May. We study the spectral properties of this source during that outburst using archival data from NICER/XTI, Swift/XRT, and NuSTAR/FPM satellites/instruments. We have analyzed the source during six epochs on which simultaneous NICER–NuSTAR and Swift/XRT–NuSTAR data were available. Using both phenomenological and physical model fitting approaches, we analyzed the spectral data in the broad 0.7–70 keV energy band. We first used a combination of disk blackbody with power-law, disk blackbody with broken power-law, and disk blackbody with power-law and bulk motion Comptonization (BMC) models. For a better understanding of the accretion picture, e.g., understanding how the accretion rates change with the changing size of the perceived Compton cloud, we used the two-component advective flow (TCAF) model with the broken power-law, and TCAF with the combined power-law, BMC models. For the last three epochs, the diskbb+power-law and TCAF models were able to spectrally fit the data for acceptable χ 2/degrees of freedom (dof). However, for the first three epochs, we needed an additional component to fit spectra for acceptable χ 2/dof. From our analysis, we reported on the possible presence of another component during these first three epochs when the source was in the high soft state. This additional component in this state is best described by the BMC phenomenon. From the TCAF model fitting, we estimated the average mass of the source as 8.1 − 2.7 + 1.3 M ⊙ .

ABSTRACT Galactic short orbital period black hole candidate (BHC) XTE J1752−223 was discovered on 2009 October 21 by the Rossi X-ray Timing Explorer (RXTE). We study the spectral properties of this outburst using transonic flow solution based two component advective flow (TCAF) model. TCAF model fitted spectrum gives an estimation of the physical flow parameters, such as the Keplerian disc rate, sub-Keplerian halo rate, properties of the so-called Compton cloud, other than the mass of the source and normalization (N). N is a standardized ratio of emitted to observed photon flux in TCAF that does not include X-ray emission from jets. In the presence of jets, this ratio changes and this deviation is used to obtain the estimation of X-ray contribution from the jets. Nature of the jet is found to be compact during low luminous hard state and discrete or blobby during high luminous intermediate states. We find a correlation between the radio (5.5 GHz) and X-ray (2.5–25 keV) fluxes from different components. The radio (FR) and jet X-ray (Fouf) fluxes are found to be correlated within the acceptable range of the standard correlation (0.6 to 0.7). A similar correlation indices were reported by our group for three other short orbital period transient BHCs (Swift J1753.5−0127, MAXI J1836−194, and XTE J1118+480).

La consommation d’alcool au cours de la grossesse constitue une cause majeure de troubles du comportement et de handicap. Alors qu’il est possible pour un clinicien d’établir un diagnostic néonatal du syndrome d’alcoolisation fœtale, l’atteinte la plus sévère des troubles causés par l’alcoolisation fœtale (TCAF), une grande majorité des enfants échappe à un diagnostic précoce en raison de l’absence d’anomalies morphologiques évidentes. Plusieurs années de prise en charge sont alors perdues. Des avancées récentes ont permis d’établir l’existence d’un axe fonctionnel placenta-cerveau impliqué dans le contrôle de l’angiogenèse cérébrale, qui se trouve dérégulé chez les enfants exposés in utero à l’alcool. Une angiogenèse cérébrale normale étant un prérequis à l’établissement d’un neurodéveloppement correct, ces avancées ouvrent la voie à l’identification d’une nouvelle génération de biomarqueurs placentaires d’atteinte cérébrale pour le diagnostic précoce des enfants TCAF.

Abstract We study the recent outburst of the black hole candidate EXO 1846-031, which went into an outburst in 2019 after almost 34 yr in quiescence. We use archival data from the Swift/XRT, MAXI/GSC, NICER/XTI, and NuSTAR/FPM satellites/instruments to study the evolution of the spectral and temporal properties of the source during the outburst. The low-energy (2–10 keV) X-ray flux of the outburst shows multiple peaks, making it a multipeak outburst. Evolving type-C quasi-periodic oscillations are observed in the NICER data in the hard, hard-intermediate, and soft-intermediate states. We use the physical two-component advective flow (TCAF) model to analyze the combined spectra of multiple satellite instruments. According to the TCAF model, the accreting matter is divided into Keplerian and sub-Keplerian parts, and the variation in the observed spectra in different spectral states arises out of the variable contributions of these two types of accreting matter in the total accretion rate. Studying the evolution of the accretion rates and other properties of the accretion flow obtained from the spectral analysis, we show that the multiple peaks in the outburst flux arise out of the variable supply of accreting matter from the pile-up radius. We determine the probable mass of the black hole to be 10.4 − 0.2 + 0.1 M ⊙ from the spectral analysis with the TCAF model. We also estimate the viscous timescale of the source in this outburst to be ∼8 days from the peak difference of the Keplerian and sub-Keplerian mass-accretion rates.

ABSTRACT Spectral and timing properties of the stellar-mass black hole candidate XTE J1752-223 during its 2009-10 outburst are studied using RXTE PCA data in the 2.5–25 keV energy range. Low frequency quasi-periodic oscillations are seen during outburst. The spectral analysis is done using two types of models: one is the combined disc blackbody plus power-law model and the other is Transonic flow solution based Two Component Advective Flow (TCAF) model. Light-curve profiles and evolution of hardness ratios are studied using MAXI GSC and Swift BAT data. Based on the evolution of the temporal and the spectral properties, we find that the object evolved through the following spectral states: hard, hard-intermediate, and soft-intermediate/soft. From the TCAF model fitted spectral analysis, we also estimate the probable mass of the black hole in the range of 8.1−11.9 M⊙, and more precisely, the mass appears to be 10 ± 1.9 M⊙.

TRPM8 is a cold sensor that is highly expressed in the prostate as well as in other non-temperature-sensing organs, and is regulated by downstream receptor–activated signaling pathways. However, little is known about the intracellular proteins necessary for channel function. Here, we identify two previously unknown proteins, which we have named “TRP channel–associated factors” (TCAFs), as new TRPM8 partner proteins, and we demonstrate that they are necessary for channel function. TCAF1 and TCAF2 both bind to the TRPM8 channel and promote its trafficking to the cell surface. However, they exert opposing effects on TRPM8 gating properties. Functional interaction of TCAF1/TRPM8 also leads to a reduction in both the speed and directionality of migration of prostate cancer cells, which is consistent with an observed loss of expression of TCAF1 in metastatic human specimens, whereas TCAF2 promotes migration. The identification of TCAFs introduces a novel mechanism for modulation of TRPM8 channel activity.

The adjunctive use of healing gels following periodontal plastic surgery is not common in clinical practice, and no definitive benefits have yet been demonstrated. Case Presentation. A 33-year-old male patient with a central lower incisor class RT1 recession sought treatment due to sensitivity and dissatisfaction with the appearance of his smile. The patient had no history of periodontal disease; however, he was under orthodontic treatment contributing to the gingival recession and irregular gum contours. Treatment. The patient underwent two sequential surgical procedures. Initially, an apically repositioned flap (APF) was performed to correct the frenulum reducing flap tension and improving the gum line aesthetics. Subsequently, after 8 weeks, a tunneled coronally advanced flap (TCAF) was executed to further refine the gum contours and achieve root coverage. Postoperative Healing Protocol. To enhance the healing process and alleviate postoperative discomfort, a healing gel containing hyaluronic acid as the active molecule was applied to the surgical sites. The gel was expected to reduce pain perception and minimize the need for painkiller intake during the critical first week of recovery. The patient was asked to fill a pain chart for the initial 7 days, recording pain levels on a visual analogue scale (VAS 0-10) and the number of paracetamol tablets taken as painkillers. Results. After both the APF and TCAF surgeries, the patient reported pain levels with a mean VAS score of 4.33 and 4.25, respectively. The painkiller intake during the first week was noted to be 3 tablets for the APF and 2 tablets for the TCAF. Notably, the application of the healing gel with hyaluronic acid did not cause any adverse reactions, indicating its potential safety and efficacy in this context. Conclusion. The application of a healing gel containing hyaluronic acid after periodontal plastic surgery showed promising results in reducing postoperative pain and the need for painkillers during the initial week of recovery. However, further investigations through randomized clinical trials are required to establish the potential benefits and broader applicability of such healing gel applications in the context of periodontal plastic surgery.

AbstractIn wake of changing lifestyle and increasing surgical expertise, knee replacement surgeries have become common. Attempts at improving the postoperative range of motion have prompted high-flexion knee arthroplasties. Consequently, there is a need to study the anatomy of knee joint in high flexion. This study focused on a holistic description of terminal condylar articular facet (TCAF; part of the femoral articular area in contact with the tibia in high flexion) of medial and lateral condyles and their quantification. In total, 104 dry femora and 16 dissected cadaveric knee joints (10 adult and 6 fetal) were studied. In most cases, the medial TCAF showed a tongue- or wedge-shaped extension in its lateral half, which has been referred to as extended condylar facet (ECF). The surface area of the ECF was approximately 0.5 cm2. In adult cadaveric dissections, the fibrous capsule showed the lax attachment beyond the ECF, thus creating a cave in the medial tibiofemoral compartment. These features were absent in fetal dissection. The presence of an ECF has not been documented in dry bones and has not been quantified. The variability of the size of this extension supports the hypothesis of this being an acquired trait. This study is expected to provide factual data for implant design improvement and a new anthropological data on Indian femora.

Resumo:

Objetivou-se quantificar o crescimento vegetal e avaliar os índices fisiológicos de diferentes acessos de mamoeiro inoculados com o vírus PRSV-P e assim identificar acessos superiores frente a mancha anelar. O experimento foi realizado na Casa de Vegetação da Embrapa Mandioca e Fruticultura, localizada em Cruz das Almas - BA, em blocos inteiramente casualizados. Foram testados 24 acessos e uma cultivar, com três repetições e dois controles de cada acesso. Três plantas foram inoculadas com PRSV-P e duas deixadas como controles negativos. As avaliações de crescimento foram feitas aos 15, 30 e 60 dias após inoculação. Foram mensurados a altura de planta, diâmetro do caule, número de folhas e área foliar e os índices fisiológicos Taxa de crescimento absoluto (TCAP), Taxa de crescimento relativo da planta (TCRP), Taxa de crescimento absoluto foliar (TCAF) e Taxa de crescimento relativo foliar (TCRF). Os dados foram submetidos ao teste F da análise de variância. As médias das cultivares foram agrupadas pelo teste de Scott-Knott a 5% de probabilidade e as médias dos tratamentos (controle x inoculados) foram comparadas pelo teste de Tukey a 5% de probabilidade. A análise quantitativa de crescimento e os índices fisiológicos mostraram ser ferramentas eficientes para identificar diferenças no crescimento de diferentes materiais. O acesso CMF026 demonstrou maior estabilidade para as características número de folhas e área foliar ao longo das avaliações, sendo menos afetada para essas características pelo vírus da mancha anelar.

Resumo:

Cancer-associated fibroblasts (CAFs) within a solid tumor can support the progression of cancer. We studied the identification and characterization of patient-derived endometrial CAFs in the context of their clinical relevance in endometrial cancers. We established patient-derived primary cultures of CAFs from surgically resected tumors (TCAF) and tumor-adjacent normal (NCAF) tissues in 53 consented patients with success rates of 97.7% and 75%, respectively. A passage of CAF was qualified by the (1) absence of CK 8,18,19, EpCAM, CD45, and CD31, and (2) presence of SMAalpha, S100A4, CD90, FAP, TE-7, CD155, PD-L1, TGFB, PDGFRA (qRT-PCR, flow cytometry, Western blot, ICC). Out of the 44 established CAFs, 31 were aggressive (having an early, i.e., 4–7 week, establishment time and/or >3 passages) compared to 13 which were non-aggressive. A post-surgery-event (PSE) was observed in 7 out of 31 patients bearing aggressive CAFs, 2 of whom were also positive for CTCs, while none of the 13 patients bearing non-aggressive CAFs had events. A positive correlation was found between patients with grade 3 (p = 0.025) as well as stage 3/4 diseases (p = 0.0106) bearing aggressive CAFs and the PSE. Finally, aggressive TCAFs from patients with PSE resisted the effects of paclitaxel and lenvatinib on the growth of HUVEC and endometrial tumor cells. Our study is the first to report a correlation between the PSE and the aggressive nature of CAFs in endometrial cancers and provides an undeniable reason to study the in-depth mechanism of CAF function towards the development of treatment resistance in endometrial cancers.

Resumo:

The newly discovered galactic black hole candidate (BHC) MAXI J1348-630 showed two major outbursts in 2019, just after its discovery. Here, we provide a detailed spectral and temporal analysis of the less-studied second outburst using archive data from multiple satellites, namely Swift, MAXI, NICER, NuSTAR and AstroSat. The outburst continued for around two and a half months. Unlike the first outburst from this source, this second outburst was a ‘failed’ one. The source did not transition to soft or intermediate spectral states. During the entire outburst, the source was in the hard state with high dominance of non-thermal photons. The presence of strong shocks are inferred from spectral fitting using a TCAF model. In NuSTAR spectra, weak reflection is observed from spectral fitting. Low-frequency quasi-periodic oscillations are also detected in AstroSat data.

A new Ti-Cr based alloy has been developed to reach a TWIP (TWinning Induced Plasticity) effect as the main deformation mechanism. This new composition, involving Fe addition, was derived from a classical TRIP/TWIP alloy Ti-8.5Cr-1.5Al (wt%) (TCA). The main objective is to achieve an optimized strength/hardenability combination by limiting the TRIP (TRansformation Induced Plasticity) effect whose critical resolved shear stress lowers the plasticity threshold. This new alloy Ti-7Cr-1Al-xFe (wt%) (TCAF) displays excellent mechanical properties, with an increased yield strength (with respect to TCA alloy), a very high work-hardening rate and an extremely high fracture strength (UTS=1415MPa), while maintaining an excellent ductility (ε=0.38 at fracture). Both mechanical (tensile tests) and microstructural characterization at different scales (EBSD, XRD) have been performed, evidencing a dense network of fine {332}<113> mechanical twins as well as the presence of stress-induced martensite plates at twins intersections, as a secondary mechanism.

In a prospective, randomized trial Cancer and Leukemia Group B (CALGB) evaluated CAF chemotherapy (cyclophosphamide + doxorubicin + 5-fluorouracil [5-FU]) v CAF plus tamoxifen (TCAF) in advanced breast cancer. Patients were stratified by estrogen receptor (ER) status, dominant site of metastatic disease, menopausal status, and prior adjuvant therapy. Regardless of ER status or menopausal status, the addition of tamoxifen conferred no significant advantage in response rate, response duration, time to treatment failure (TTF) or survival over CAF alone. A secondary objective was to compare the response to CAF of ER positive (ER+) and ER negative (ER-) patients to determine if there was a differential response to cytotoxic chemotherapy. Response rates of ER+ and ER- patients to CAF were identical (56%), but the response duration, time to treatment failure, and survival of ER+ patients were significantly longer than ER- patients. This lack of differential response implies that chemotherapy and hormonal therapy may compete for the same pool of ER+ cells. It also suggests that chemotherapy kills breast cancer cells indiscriminately, regardless of ER status.

Formation of disinfection by-products (DBPs), including dichloroacetonitrile (DCAN), trichloroacetonitrile (TCAN), chloroform (TCM), dichloroacetic acid (DCAA), trichloroacetic acid (TCAA), chloropicrin (TCNM) and chloral hydrate (CH), from monochloramination of chironomid larvae was investigated at different contact times, pH levels and temperatures. Increased monochloramine dosage and prolonged reaction time resulted in an increase in most DBPs. Furthermore, the concentrations of TCAN, DCAN and CH initially increased over time before reaching a plateau. This is because of the hydrolysis of functional groups and the function of NH2Cl. The concentrations of DCAA, TCAA and TCM decreased as pH was increased from 5 to 10, and TCAN, DCAN and CH were not detected when the pH exceeded 8. As for TCNM, their concentration increased as the pH increased from 5 to 7, following a subsequent decrease at higher pH values. High temperatures led to higher DCAN and TCAN concentrations, while low temperatures resulted in higher TCNM and DCAA concentrations. Both high and low temperatures reduced the concentrations of TCAN and CH. Finally, concentrations of the four predominant DBPs (TCM, DCAN of DCAA and TCAA) all decreased as Cl/N ratio declined.

Resumo:

The 2015 Outburst of V404 Cygni is an unusual one with several X-ray and radio flares and rapid variation in the spectral and timing properties. The outburst occurred after 26 years of inactivity of the black hole. We study the accretion flow properties of the source during its initial phase of the outburst using Swift/XRT and Swift/BAT data in the energy range of 0.5–150 keV. We have done spectral analysis with the two component advective flow (TCAF) model fits file. Several flow parameters such as two types of accretion rates (Keplerian disk and sub-Keplerian halo), shock parameters (location and compression ratio) are extracted to understand the accretion flow dynamics. We calculated equipartition magnetic field Beq for the outburst and found that the highest Beq∼900 Gauss. Power density spectra (PDS) showed no break, which indicates no or very less contribution of the Keplerian disk component, which is also seen from the result of the spectral analysis. No signature of prominent quasi-periodic oscillations (QPOs) is observed in the PDS. This is due to the non-satisfaction of the condition for the resonance shock oscillation as we observed mismatch between the cooling timescale and infall timescale of the post-shock matter.

This research evaluates the economic-financial sustainability and adherence to compliance and accountability practices in civil society organizations (CSOs) with projects in the “Legal Amazon Biome”, supported with resources from the Amazon Fund, in the protection of environmental thematic areas. The object of each project is social and environmental responsibility and the primary data submitted for evaluation are recovered from the financial statements of the CSOs in the time horizon from 2015 to 2017. The Fund manager is the National Bank for Economic and Social Development (NBESD) and the governance , until then exercised by the Guiding Committee of the Amazon Fund (CAF) and Technical Committee of the Amazon Fund (TCAF), was discontinued in 2019, unilaterally by the Government of Brazil, resulting in an increase in the environmental degradation of the Biome by around 30% in 2019, compared to the previous year, in addition to the interruption of contributions from donors and support for projects. The research results, using positivist methodology, provide evidence that adherence to compliance and accountability practices is partial, contributing to fund management and to the literature in subsequent investigations, but CSOs are not self-sustainable and the available information does not allow evaluate the fulfillment of the object and of the UN's ESG agenda.

Abstract Background: In endometrial cancers (EC), the mechanistic contribution of PD-L1/L2 and PD-1 signaling of host’s stromal microenvironment (SME) [cancer-associated fibroblast (CAF) and immune cells] in relation to the tumor (T) cells is elusive. Aim: To understand the tumor-stroma-immune crosstalk in EC, we studied the compartmental pattern of PD-L1, PD-L2, and PD-1 expression in EC tissues and their matched CAFs from both T and tumor-adjacent normal (N) tissues. Method: Over 116 surgically resected T and N tissues were obtained from consented patients with EC at the Avera Cancer Institute (March ‘16 and September ’21). IHC expression was performed in T, N-epithelium, SME (mesenchyme) in the T and N tissues. The staining intensity and distribution pattern were evaluated by a pathologist using a standard scoring system (1% cut-off value). Since fibroblasts are the most abundant cells of SME, the PD-L1 was parallelly evaluated in the multiple passages of cultured T and N derived CAFs by the qRT-PCR, flow-cytometry, and ICC. Result: The expression of 3 immune checkpoint proteins in the epithelial and mesenchyme of N was limited to only rare and focal positivity for PD-L1. The epithelial compartments of the T tissue, in contrast, had 70% positivity for PD-L1, which was independent of differentiation status, presence of TILs, or lymphovascular invasion. PD-L1 positivity was 90% of the SME of tested samples, identified predominantly in the (1) immune component, including macrophages and lymphocytes, and (2) EpCAM-/SMA+/FAP+/S100A4+ CAFs. Both PD-L1 and PD-L2 were identified by flow, ICC, and qRT-PCR in NCAF and TCAF. The overall expression of PD-L2 was found to be qualitatively and quantitatively lesser than that of PD-L1, primarily contributed by SME/stromal macrophages and lymphocytes. PD-1 expression was restricted to the SME/stromal immune components, predominantly lymphocytes. The expression for PD-L1 and PD-L2 mRNA was higher in TCAF than NCAF in successive passages, which ICC and flow cytometry confirmed. Summary: The expression of immune checkpoint proteins, PD-L1, and PD-L2, are primarily tumor-driven and associated with the endometrioid (invasive) and serous adenocarcinomas of grades II and above with myometrial invasion. The expression of PD-L1 and PD-L2 are distributed in both tumor and SME/stromal components, while PD-1 expression is exclusively SME/stromal. Resumo:

In order to find the factors and their effects for the Ca-based absorbent for CO2-capture, the experimental method was used to research the limestone in the calcination/carbonation fixed bed process. Then measured the specific surface and specific pore volume respectively by the method of COULTER_SA_3100 nitrogen absorption specific surface and the method of BET and BJH through pore size analyzer. The result has showed that the microstructure of absorber and conversion ratio of carbonation X was affected by the operating parameters including calcination temperature (Tcal), CO2 concentration in calcination atmosphere, variable cyclic number N and carbonation temperature (Tcar). In this experiment, X reached its maximum when Tcal was 850°C, with the calcination temperature continued to rise, X reduced gradually. The increase of the concentration of CO2 will strengthen the role of sintering and reduce X in the calcination atmosphere. With the increase of the cyclic number N, sintering effect made the micro-structure of absorbent change, so that X decreased. Tcar close to the limit temperature was appropriate. In the limited temperature range, with the increase of N, conversion ratio of carbonation X declined slowly.

Objective. Trichloroacetic acid (TCAA), or trichloroethanoic acid, is a chemical analogue of acetic acid where three methyl group hydrogen atoms are replaced by chlorine. TCAAs are also abbreviated and referred to as TCAs, causing confusion with the psychiatric antidepressant drug class, especially among patients. TCAAs exist in dermatological treatments such as chemical peels or wart chemoablation medication. TCAA ingestion or overdose can cause gastric irritation symptoms including vomiting, diarrhea, or lassitude. This symptomatology is less severe than TCA overdose, where symptoms may include elevated body temperature, blurred vision, dilated pupils, sleepiness, confusion, seizures, rapid heart rate, and cardiac arrest. Owing to the vast difference in symptoms, the need for clinical intervention differs greatly. While overdose of either in a self-harm attempt can warrant psychiatric hospital admission, the risk of death in TCAA ingestion is far less. Case Report. A patient ingested TCAA in the form of a commercially available dermatological chemical peel as a self-harm attempt, thinking that it was a more injurious TCA. Conclusion. Awareness among physicians, particularly psychiatrists, regarding this relatively obscure chemical compound (TCAA) and its use by suicidal patients mistakenly believing it to be a substance that can be significantly more lethal (TCA), is imperative.

Foi estudada a variabilidade genética em progênies de bacabi (Oenocarpus mapora Karsten) e bacaba (Oenocarpus distichus Mart.) para caracteres de emergência e crescimento inicial de mudas. Foram coletados frutos maduros de 47 matrizes em quintais de produtores rurais e no Banco de Germoplasma da Embrapa Amazônia Oriental, envolvendo análise de 47 progênies de polinização aberta delineadas em blocos ao acaso, com quatro repetições e parcelas de 24 sementes. As variáveis avaliadas foram: a percentagem de emergência (PE), tempo médio de emergência (TME), taxa de crescimento absoluto da altura de plântulas (TCAA) e taxa de crescimento absoluto do diâmetro de plântulas (TCAD). As progênies apresentaram diferenças significativas (p≤0,01) para todos os caracteres da análise das duas espécies, com boa precisão experimental. Para a herdabilidade no sentido amplo foi verificado que no conjunto total de progênies e no conjunto de progênies de bacabi todos os caracteres estudados apresentaram estimativas de herdabilidade acima de 50%. As exceções foram para TME e TCAD, no conjunto de progênies de bacaba, com estimativas de herdabilidade abaixo de 50%. Os coeficientes "b" apresentaram a mesma tendência observada na herdabilidade, e suas magnitudes expressam também a variabilidade genética. Os ganhos de seleção estimados a partir das porcentagens de seleção pré-determinados mostraram valores altos para todos os caracteres, favorecidos pela pressão de seleção com uma intensidade de 10% para o total de progênies e apenas bacabi e 20% para as progênies somente de bacaba.

Although the von Neumann architecture-based computing system has been used for a long time, its limitations in data processing, energy consumption, etc. have led to research on various devices and circuit systems suitable for logic-in-memory (LiM) computing applications. In this paper, we analyze the temperature-dependent device and circuit characteristics of the floating gate field effect transistor (FGFET) source drain barrier (SDB) and FGFET central shallow barrier (CSB) identified in previous papers, and their applicability to LiM applications is specifically confirmed. These FGFETs have the advantage of being much more compatible with existing silicon-based complementary metal oxide semiconductor (CMOS) processes compared to devices using new materials such as ferroelectrics for LiM computing. Utilizing the 32 nm technology node, the leading-edge node where the planar metal oxide semiconductor field effect transistor structure is applied, FGFET devices were analyzed in TCAD, and an environment for analyzing circuits in HSPICE was established. To seamlessly connect FGFET-based devices and circuit analyses, compact models of FGFET-SDB and -CSBs were developed and applied to the design of ternary content-addressable memory (TCAM) and full adder (FA) circuits for LiM. In addition, depression and potential for application of FGFET devices to neural networks were analyzed. The temperature-dependent characteristics of the TCAM and FA circuits with FGFETs were analyzed as an indicator of energy and delay time, and the appropriate number of CSBs should be applied.

Pancreatic adenocarcinoma (PDAC) has low survival rates worldwide due to its tendency to be detected late and its characteristic desmoplastic reaction, which slows the use of targeted therapies. As such, the discovery of new connections between genes and the clinicopathological parameters contribute to the search for new biomarkers or targets for therapy. Transient receptor potential (TRP) channels are promising tools for cancer therapy or markers for PDAC. Therefore, in this study, we selected several genes encoding TRP proteins previously reported in cellular models, namely, Transient Receptor Potential Cation Channel Subfamily V Member 6 (TRPV6), Transient receptor potential ankyrin 1 (TRPA1), and Transient receptor potential cation channel subfamily M (melastatin) member 8 (TRPM8), as well as the TRPM8 Channel Associated Factor 1 (TCAF1) and TRPM8 Channel Associated Factor 2 (TCAF2) proteins, as regulatory factors. We analyzed the expression levels of tumors from patients enrolled in public datasets and confirmed the results with a validation cohort of PDAC patients enrolled in the Clinical Institute Fundeni, Romania. We found significantly higher expression levels of TRPA1, TRPM8, and TCAF1/F2 in tumoral tissues compared to normal tissues, but lower expression levels of TRPV6, suggesting that TRP channels have either tumor-suppressive or oncogenic roles. The expression levels were correlated with the tumoral stages and are related to the genes involved in calcium homeostasis (Calbindin 1 or S100A4) or to proteins participating in metastasis (PTPN1). We conclude that the selected TRP proteins provide new insights in the search for targets and biomarkers needed for therapeutic strategies for PDAC treatment.

In 2009, a disease survey was conducted in 97 commercial canola (Brassica napus L.) fields in Western Australia by the Department of Agriculture and Food, Western Australia (DAFWA). In about 20% of the fields from the northern agricultural region of Western Australia, small patches were observed where canola plants showed symptoms of stunting and wilting. These plants were collected and roots of affected plants were washed thoroughly and examined for the presence of root disease. Small galls and clublike structures were observed on the secondary roots and sometimes on the main root of the affected plants. Examination of thin free hand sections from the root galls revealed that several cortical cells were enlarged and full of resting spores. The diameter of resting spores ranged between 2.5 and 3.0 μm. Plasmodia and zoosporangia were also observed in the root hairs. The identity of Plasmodiophora brassicae Woronin was confirmed by PCR using a modified method of Cao et al. 2007 (1). DNA from spores and slices of the galls of 14 different samples were extracted using DNeasy plant mini kit (QIAGEN Australia) as per manufacturer's instructions. Samples were disrupted by placing them into MPBIO tube A and placed in the Fast Prep machine at speed of 6 ms–1 for 40 s. This was repeated twice. The species-specific primers TC1F (5′-GTGGTCGAACTTCATTAAATTTGGGCTCTT-3′)/TC1R (5′-TTCACCTACGGAACGTATATGTGCATGTGA-3′) and TC2F (5′-AAACAACGAGTCAGCTTGAATGCTAGTGTG-3′)/TC2R (5′-CTTTAGTTGTGTTTCGGCTAGGATGGTTCG-3′) were used (1). The primers TC1F and TC1R failed to produce a PCR product of 548-bp size but using the primers TC2F and TC2R the PCR reaction resulted in a 519- bp fragment. Seven out of 14 samples gave positive results for P. brassicae with primers TC2F and TC2R. This indicates that the P. brassicae pathotype from Western Australia may be different than the one found in Alberta, Canada. However, pathotypes of P. brassicae from brassica vegetables from Australia have been found similar to the populations of P. brassicae present in the United States (2). Pathogenicity of P. brassicae was tested by dipping roots of five 10-day-old canola plants var. Cobbler in a spore suspension (1 × 106 resting spores/ml). Roots of five control plants were dipped in sterile water. Five weeks after inoculation, small galls were observed on the roots of three inoculated plants and the control plants remained symptomless. Resting spores were recovered from the galls developed on the roots of affected plants. Presence of P. brassicae in the affected roots was further confirmed by PCR using the method described above. To our knowledge, this is the first report of club root of canola in Australia. Club root is reported from vegetable brassicas and white mustard (Sinapis alba L.) in Australia. Club root has become a serious disease of canola in Canada since its detection in Alberta in 2006 (3). The resting spores of the fungus can survive for several years in soil, and therefore, this disease could pose a significant threat to canola production in Western Australia. Terahertz coded-aperture imaging (TCAI) can overcome the difficulties of traditional radar in forward-looking and high-resolution imaging. Three-dimensional (3D) TCAI relies mainly on the reference-signal matrix (RSM), the large size and poor accuracy of which reduce the computational efficiency and imaging ability, respectively. According to the previous research on TCAI, traditional TCAI cannot reduce the heavy computational burden while the improved TCAI achieve reconstructing the target parts of different ranges in parallel. However, large-sized RSM still accounts for the computational complexity of traditional TCAI and the improved TCAI. Therefore, this paper proposes a more efficient imaging method named back projection (BP)-TCAI (BP-TCAI). Referring to the basic principle of BP, BP-TCAI can not only divide the scattering information in different ranges but also project the range profiles into different imaging subareas. In this way, the target parts in different subareas can be reconstructed simultaneously to synthesize the whole 3D target and thus decomposes the computational complexity thoroughly. During the pulse compression and projection processes, the signal-to-noise ratio (SNR) of BP-TCAI is also improved. This present the imaging method, model and procedures of traditional TCAI, the improved TCAI and the proposed BP-TCAI. Numerical experimental results prove BP-TCAI to be more effective and efficient than previous imaging methods of TCAI. Besides, BP-TCAI can also be seen as synthetic aperture radar (SAR) imaging with coding technology. Therefore, BP-TCAI opens a future gate combining traditional SAR and coded-aperture imaging.

Background: The aim of this study was to assess the prevalence and associated factors of Traditional, Complementary and Alternative Medicine’s (TCAM) use of chronic disease patients in a community setting in Myanmar. Materials and Methods: A cross-sectional community survey was conducted in the Kyauk Tan Township with the International Complementary and Alternative Medicine Questionnaire (I-CAM-Q). Results: Of the 1600 participants in the survey, the overall prevalence of any TCAM use (providers, products or self-care) was 95.1% (TCAM provider= 14.6%, TCAM products=65.0%, and self-help TCAM=86.2%) in the past 12 months. For all different types of TCAM providers, TCAM products and self-help TCAM more than 90% of participants perceived the TCAM as very or somewhat helpful. In multivariate logistic regression analysis, older age, no formal education, rural residence and having two or more chronic conditions were associated with any TCAM use. Conclusions: TCAM use, especially TCAM products and self-help TCAM, seem to be common in Myanmar.

Purpose: To evaluate the effect of a combination of Tarantula cubensis alcohol extract (TCAE) and capecitabine (CAP) in the treatment of azoxymethane (AOM)-induced colorectal cancer (CRC). Methods: Forty-two Wistar albino rats were divided into 7 groups with 6 rats in each group. The groups consisted of Control (C), Control+TCAE (C-TCAE), Control+CAP (C-CAP), Cancer control (CC), Cancer+TCAE (CC-TCAE), Cancer+CAP (CC-CAP) and Cancer+CAP+TCAE (CC-CAP+TCAE). To induce CRC, AOM (15 mg/kg) was administered to rats subcutaneously (sc) twice at a one-week interval to all the groups except control. From the 15th week, TCAE (0.2 mL/rat sc) was administered to CC-TCAE group every 3 days for 4 weeks, and CAP (40 mg/kg/day) was administered by gavage to CC-CAP group for 4 weeks. In CC-CAP+TCAE group, TCAE (0.2 mL/rat sc) was administered every 3 days for 4 weeks, and CAP (40 mg/kg/day) was administered gavage for 4 weeks. Animals were treated for 18 weeks. Aberrant crypt foci (ACF) were evaluated histopathologically among CC, CC-TCAE, CC-CAP, and CC-CAP+TCAE groups. β-catenin, CD15, Proliferating Cell Nuclear Antigen (PCNA), and Nuclear Factor kappa B (NF-κB) expression levels were immunohistochemically compared among all groups. Results: Histopathologically, ACF scores were significantly increased in CC group, while a significant decrease in the relevant scores (p < 0.001) was observed in CC-CAP and CC-CAP+TCAE treatment groups, and the lowest scores were in CC-CAP+TCAE group. Immunohistochemically, in CC group, β-catenin, Nuclear Factor kappa B (NF-κB), Proliferating Cell Nuclear Antigen (PCNA) and CD15 expressions were highly irregular. CC-CAP and CC-CAP+TCAE groups had significantly reduced expressions (p < 0.001), and the lowest expressions were in CC-CAP+TCAE group. Conclusion: The combined use of TCAE and CAP in treatment of CRC has a synergistic effect and increases the anticancer efficacy of TCAE, and CAP. More studies at the molecular level are needed in the future to demonstrate the clinical benefit of TCAE supplementation during the treatment of CRC with CAP.

Abstract Specific receptors for the beta-chemokine TCA3 have been identified on mouse monocyte/macrophage cell lines and on mouse mesangial cells. Using Scatchard plot analysis with 125I-labeled TCA3, a single high-affinity receptor (3-4 nM) was identified. Cells of the monocyte lineage express 1,400 to 8,600 TCA3 binding sites, while mesangial cells display 40,000 to 49,000 sites/cell. Competitive inhibition studies indicated that the TCA3 receptor is unique, although MCP-1, IL-8, and RANTES were weak competitors of TCA3 binding. We also established the functional activity of TCA3 and other chemokines on primary cultures of mouse mesangial cells. TCA3 treatment induces increased mesangial cell adhesiveness to fibronectin. TCA3 is also a chemoattractant for mesangial cells. In addition, TCA3 treatment stimulates [3H]thymidine uptake by mesangial cells. The combined results indicate that TCA3 and other chemokines interact with a broader range of target cells than previously considered.
