ABSTRACT Bacteroides thetaiotaomicron, a gram-negative obligate anaerobe, utilizes polysaccharides by binding them to its cell surface and allowing cell-associated enzymes to hydrolyze them into digestible fragments. We use the starch utilization system as a model to analyze the initial steps involved in polysaccharide binding and breakdown. In a recent paper, we reported that one of the outer membrane proteins involved, SusG, had starch-degrading activity but was not sufficient for growth on starch. Moreover, SusG alone did not have detectable starch binding activity. Previous studies have shown that starch binding is essential for starch utilization. In this paper, we report that four other outer membrane proteins, SusC through SusF, are responsible for starch binding. Results of 14C-starch binding assays show that SusC and SusD both contribute a significant amount of starch binding. SusE also appears to contribute substantially to starch binding. Using affinity chromatography, we show in vitro that these Sus proteins interact to bind starch. Moreover, protease accessibility of either SusC or SusD greatly increased when one was expressed without the other. This finding supports the hypothesis that SusC and SusD interact in the outer membrane. Evidence from additional protease accessibility studies suggests that SusC, SusE, and SusF are exposed on the cell surface. Our results demonstrate that SusC and SusD act as the major starch binding proteins on the cell surface, with SusE enhancing binding. SusF's role in starch utilization has yet to be determined, although the fact that starch protected it from proteolytic attack suggests that it does bind starch.

ABSTRACT An early step in the utilization of starch by Bacteroides thetaiotaomicron is the binding of starch to the bacterial surface. Four starch-associated outer membrane proteins of B. thetaiotaomicron that have no starch-degrading activity have been identified. Two of these, SusC and SusD, have been shown by genetic analysis to be required for starch binding. In this study, we provide the first biochemical evidence that these two proteins interact physically with each other. Both formaldehyde cross-linking and nondenaturing gel electrophoresis experiments showed that SusC and SusD interact to form a complex. Two other proteins encoded by genes in the same operon, SusE and SusF, proved not to be essential for starch utilization and actually decreased starch binding when they were present along with SusC and SusD. Consistent with this, nondenaturing gel analysis revealed that in a strain producing SusC, SusD, and SusE, the SusCD complex was partially destabilized. The strain producing SusC, SusD, and SusE also grew more slowly on starch than a strain producing SusC, SusD, SusE, and SusF (μmax, 0.29 and 0.37/h, respectively). Thus, SusE appears to interact with the SusCD complex. SusE also interacts with SusF, because SusE was less susceptible to proteinase K digestion when SusF was present, and nondenaturing gel analysis detected a complex formed by these two proteins. Our results indicate that SusC, SusD, SusE, and SusF form a protein complex in the outer membrane but that SusE and SusF are dispensable members of this complex.

Offline Arabic Handwriting Recognition (OAHR) has recently become instrumental in the areas of pattern recognition and image processing due to its application in several fields, such as office automation and document processing. However, OAHR continues to face several challenges, including high variability of the Arabic script and its intrinsic characteristics such as cursiveness, ligatures, and diacritics, the unlimited variation in human handwriting, and the lack of large public databases. In this paper, we introduce a novel context-aware model based on deep neural networks to address the challenges of recognizing offline handwritten Arabic text, including isolated digits, characters, and words. Specifically, we propose a supervised Convolutional Neural Network (CNN) model that contextually extracts optimal features and employs batch normalization and dropout regularization parameters. This aims to prevent overfitting and further enhance generalization performance when compared to conventional deep learning models. We employ a number of deep stacked-convolutional layers to design the proposed Deep CNN (DCNN) architecture. The model is extensively evaluated and shown to demonstrate excellent classification accuracy when compared to conventional OAHR approaches on a diverse set of six benchmark databases, including MADBase (Digits), CMATERDB (Digits), HACDB (Characters), SUST-ALT (Digits), SUST-ALT (Characters), and SUST-ALT (Names). A further experimental study is conducted on the benchmark Arabic databases by exploiting transfer learning (TL)-based feature extraction which demonstrates the superiority of our proposed model in relation to state-of-the-art VGGNet-19 and MobileNet pre-trained models. Finally, experiments are conducted to assess comparative generalization capabilities of the models using another language database , specifically the benchmark MNIST English isolated Digits database, which further confirm the superiority of our proposed DCNN model.

In this paper we describe an educational program for the development of the study method for students with specific learning disorders (SLD) and a first Italian application on a small sample of fifth grade students. This experience was aimed at collecting useful data precisely to review and improve the above-mentioned program. A particularly relevant problem was identified looking at the anthropological dimension of the capability learning and the need for students to acquire the learning to learn ability. Specifically, it is about the low levels of text comprehension in Italian schools and its consequences on the study activities. Within the inclusive perspective and taking into account the effective didactic strategies in the evidence-based education (EBE) perspective, the project created and tested a teaching kit (SUST) for the fifth classes of the primary school, at-tended by students with SLD. This experimental framework was conducted following the Design Based Research methodology. It also focused on the training of teachers within the intervention classes. The positive results of SUST application open a constructive scenario for the definition of intervention programs aimed at improving text comprehension and study skills of students with SLD. This program should be tested in a wider context of schools.

Abstract. Ship-based multi-axis differential optical absorption spectroscopy (MAX-DOAS) measurements were conducted along the marginal seas of China from 19 April to 16 May 2018 to measure the vertical profiles of aerosol, nitrogen dioxide (NO2), and nitrous acid (HONO). Along the cruise route, we found five hot spots with enhanced tropospheric NO2 vertical column densities (VCDs) in the Yangtze River Delta, Taiwan Strait, Guangzhou–Hong Kong–Macau Greater Bay Area, port of Zhanjiang, and port of Qingdao. Enhanced HONO concentrations could usually be observed under high-level aerosol and NO2 conditions, whereas the reverse was not always the case. To understand the impacts of relative humidity (RH), temperature, and aerosol on the heterogeneous reaction of NO2 to form HONO in different scenarios, the Chinese Academy of Meteorological Sciences (CAMS) and Southern University of Science and Technology (SUST) MAX-DOAS stations were selected as the inland and coastal cases, respectively. The RH turning points in CAMS and SUST cases were both ∼ 65 % (60 %–70 %), whereas two turning peaks (∼ 60 % and ∼ 85 %) of RH were found in the sea cases. As temperature increased, the HONO / NO2 ratio decreased with peak values appearing at ∼ 12.5∘C in CAMS, whereas the HONO / NO2 gradually increased and reached peak values at ∼ 31.5∘C in SUST. In the sea cases, when the temperature exceeded 18.0∘C, the HONO / NO2 ratio rose with increasing temperature and achieved its peak at ∼ 25.0∘C. This indicated that high temperature can contribute to the secondary formation of HONO in the sea atmosphere. In the inland cases, the correlation analysis between HONO and aerosol in the near-surface layer showed that the ground surface is more crucial to the formation of HONO via the heterogeneous reaction of NO2; however, in the coastal and sea cases, the aerosol surface contributed more. Furthermore, we discovered that the conversion rate of NO2 to HONO through heterogeneous reactions in the sea cases is larger than that in the inland cases in higher atmospheric layers (> 600 m). Three typical events were selected to demonstrate three potential contributing factors of HONO production under marine conditions (i.e., transport, NO2 heterogeneous reaction, and unknown HONO source). This study elucidates the sea–land and vertical differences in the forming mechanism of HONO via the NO2 heterogeneous reaction and provides deep insights into tropospheric HONO distribution, transforming process, and environmental effects.

The last decade has witnessed significant progress in the development of solar water splitting devices, with solar-to-hydrogen (STH) efficiency as high as 30% already demonstrated. However, two major challenges remain. First, the high-efficiencies (> 15%) have only been achieved using devices based on expensive and non-scalable III-V semiconductors. On the other hand, low-cost metal-oxide based devices, mainly using BiVO4 as the absorber, have only achieved STH efficiency of < 10%. Due to stability limitations, many of these metal-oxide based devices are operated in near-neutral pH electrolytes, which presents an additional mass transport challenge. Second, the majority of the demonstrated devices are still at the laboratory scale. Reports on large-area devices start to emerge, but they typically show much lower efficiencies. This is best illustrated in a recent review:[1] even when III-V semiconductor-based devices are considered, there is no report of devices with a semiconductor absorber area larger than 10 cm2 and STH efficiency > 10%. In this talk, we will discuss the scale-up of our photoelectrochemical water splitting devices based on a complex metal oxide photoabsorber. Factors other than the semiconductor photoabsorber itself are found to be responsible for a total voltage loss of > 500 mV and therefore limit the overall performance of the large-area device.[2] To properly address this limitation, we quantify and break down the different loss mechanisms associated with the device scale-up and the practical operational conditions.[3] Concentration overpotential due to pH gradient is found to be a major contributor to the performance loss, and we show using multiphase multiphysics simulations and in-situ fluorescence measurements that careful control of natural and forced convection can overcome this limitation.[3-5] In addition, we also explore the possibility to achieve efficient product separation in devices with and without separators. The product crossover, optical and Ohmic losses are quantified using a combination of experiments and simulations, and the optimization of the device working parameters and/or separator properties to achieve the minimum overall loss will be discussed.[6,7] References J. It became clear that bringing the Earth to an ecological balance is an archival task of humanity, but reaching an agreement on the need for comprehensive changes will not be easy. But the question has already been put on the agenda. The role of the «Club of Rome», World Environmental Forums and Reports of International Environmental Commissions in justifying the concept of sustainable development is analyzed. It was revealed that for most subjects of world politics in the development and implementation of the goals of sustainable development, the imitation of legislative functions, rhetoric prevails instead of real changes in the ecological and economic paradigm. It is emphasized the need for a radical change in the moral paradigm of human life, according to which only what is ecological is economic. The factors of the scientific, economic, technological, political-legal and worldview levels that universally inhibit the introduction of the model of sustainable development are indicated and analyzed.

Secondly, students' performance improved significantly, and they developed better attitudes towards learning English via cooperative learning strategy. Finally, in cooperative learning, students are motivated and less reluctant. Also, the study recommends the following: first, teachers of English should adopt cooperative learning methods in the teaching and learning process. Second, students should be provided with enough opportunities in the learning process, and more importantly, they should be encouraged to implement cooperative learning methods both in and out the classroom.

With the rapid development of computer technology and the need of college Eng-lish teaching, the automatic grading of college students' English writing level is an inevitable trend. This is the inevitable result of the combination of computer tech-nology and English teaching. The theoretical basis of this paper is the feedback of English writing and related theories introduced as follows. This paper analyzes the influence of automatic scoring and feedback system on English writing level via testing the variation of student composition’s grammar, structure, length and grade in a teaching experiment. The experimental results indicate that online feed-back is effective in evaluating students' English writing proficiency, which can help students improve their writing proficiency and independent writing ability. The students' attitudes and views towards automatic grading and feedback system were investigated with 118 students in the English experiment class of SUST as the test object. The research results can suggest a guide line in the improvement of college students’ writing competence and evaluation of the effect of automatic scoring feedback system.

This paper presents a new concept of a photovoltaic module. The increase in efficiency is obtained by using a three-dimensional form of flexible photovoltaic panels with an additional reflecting mirror. A flexible panel is distributed over the entire inner surface of the cylinder, while the mirror distributes the flux on its surface. The comparison of selected solutions for a mirror shape is realized with numerical simulations. The irradiance results for absorbed flux are discussed on the ground of possible improvements. Parthasarathy, Madhavan, Jiban Khuntia e Rulon Stacey. "Impact of Remote and Virtual Care Models on the Sustainability of Small Health Care Businesses: Perceptual Analysis of Small Clinics, Physician Offices, and Pharmacies in Colorado". Journal of Medical Internet Research 23, n.º 2 (25 de fevereiro de 2021): e23658. http://dx.doi.org/10.2196/23658.

Background Lockdowns and shelter-in-place orders during COVID-19 have accelerated the adoption of remote and virtual care (RVC) models, potentially including telehealth, telemedicine, and internet-based electronic physician visits (e-visits) for remote consultation, diagnosis, and care, deterring small health care businesses including clinics, physician offices, and pharmacies from aligning resources and operations to new RVC realities. Current perceptions of small health care businesses toward remote care, particularly perceptions of whether RVC adoption will synergistically improve business sustainability, would highlight the pros and cons of rapidly adopting RVC technology among policy makers. Objective This study aimed to assess the perceptions of small health care businesses regarding the impact of RVC on their business sustainability during COVID-19, gauge their perceptions of their current levels of adoption of and satisfaction with RVC models and analyze how well that aligns with their perceptions of the current business scenario (SCBS), and determine whether these perceptions influence their view of their midterm sustainability (SUST). Methods We randomly sampled small clinics, physician offices, and pharmacies across Colorado and sought assistance from a consulting firm to collect survey data in July 2020. Focal estimated study effects were compared across the three groups of small businesses to draw several insights. Results In total, 270 respondents, including 82 clinics, 99 small physician offices, and 89 pharmacies, across Colorado were included. SRVC and SCBS had direct, significant, and positive effects on SUST. However, we investigated the effect of the interaction between SRVC and SCBS to determine whether RVC adoption aligns with their perceptions of the current business scenario and whether this interaction impacts their perception of business sustainability. Effects differed among the three groups. The interaction term SRVC×SCBS was significant and positive for clinics (P=.02), significant and negative for physician offices (P=.05), and not significant for pharmacies (P=.76). These variations indicate that while clinics positively perceived RVC alignment with the current business scenario, the opposite held true for small physician offices. Conclusions As COVID-19 continues to spread worldwide and RVC adoption progresses rapidly, it is critical to understand the impact of RVC on small health care businesses and their perceptions of long-term survival. Small physician practices cannot harness RVC developments and, in contrast with clinics, consider it incompatible with business survival during and after COVID-19. If small health care firms cannot compete with RVC (or synergistically integrate RVC platforms into their current business practices) and eventually become nonoperational, the resulting damage to traditional health care services may be severe, particularly for critical care delivery and other important services that RVC cannot effectively replace. Our results have implications for public policy decisions such as incentive-aligned models, policy-initiated incentives, and payer-based strategies for improved alignment between RVC and existing models.

ABSTRACT The human gut microbe Bacteroides fragilis can alter the expression of its surface molecules, such as capsular polysaccharides and SusC/SusD family outer membrane proteins, through reversible DNA inversions. We demonstrate here that DNA inversions at 12 invertible regions, including three gene clusters for SusC/SusD family proteins, were controlled by a single tyrosine site-specific recombinase (Tsr0667) encoded by BF0667 in B. fragilis strain YCH46. Genetic disruption of BF0667 diminished or attenuated shufflon-type DNA inversions at all three susC/susD genes clusters, as well as simple DNA inversions at nine other loci, most of which colocalized with susC/susD family genes. The inverted repeat sequences found within the Tsr0667-regulated invertible regions shared the consensus motif sequence AGTYYYN4GDACT. Tsr0667 specifically mediated the DNA inversions of 10 of the 12 regions, even under an Escherichia coli background when the invertible regions were exposed to BF0667 in E. coli cells. Thus, Tsr0667 is an additional globally acting DNA invertase in B. fragilis, which probably involves the selective expression of SusC/SusD family outer membrane proteins.

Sucrose synthase (Sus) is a key enzyme of sucrose metabolism. Two Sus-encoding genes (Sus1 and Sus2) from Arabidopsis thaliana were found to be profoundly and differentially regulated in leaves exposed to environmental stresses (cold stress, drought or O2 deficiency). Transcript levels of Sus1 increased on exposure to cold and drought, whereas Sus2 mRNA was induced specifically by O2 deficiency. Both cold and drought exposures induced the accumulation of soluble sugars and caused a decrease in leaf osmotic potential, whereas O2 deficiency was characterized by a nearly complete depletion in sugars. Feeding abscisic acid (ABA) to detached leaves or subjecting Arabidopsis ABA-deficient mutants to cold stress conditions had no effect on the expression profiles of Sus1 or Sus2, whereas feeding metabolizable sugars (sucrose or glucose) or non-metabolizable osmotica [poly(ethylene glycol), sorbitol or mannitol] mimicked the effects of osmotic stress on Sus1 expression in detached leaves. By using various sucrose/mannitol solutions, we demonstrated that Sus1 was up-regulated by a decrease in leaf osmotic potential rather than an increase in sucrose concentration itself. We suggest that Sus1 expression is regulated via an ABA-independent signal transduction pathway that is related to the perception of a decrease in leaf osmotic potential during stresses. In contrast, the expression of Sus2 was independent of sugar/osmoticum effects, suggesting the involvement of a signal transduction mechanism distinct from that regulating Sus1 expression. The differential stress-responsive regulation of Sus genes in leaves might represent part of a general cellular response to the allocation of carbohydrates during acclimation processes.

The suspensor is the first differentiated structure produced during plant embryogenesis. In most angiosperms, the suspensor functions early in development to provide nutrients and growth regulators to the embryo proper. In Arabidopsis, the suspensor undergoes programmed cell death at the torpedo stage and is not present in mature seeds. We have identified at least 16 embryo-defective mutants of Arabidopsis that exhibit an enlarged suspensor phenotype at maturity. In this report, we focus on seven abnormal suspensor mutants, which define three genetic loci (sus1, sus2 and sus3). Recessive mutations at each of these loci disrupt morphogenesis in the embryo proper and consistently result in the formation of a large suspensor. Defects in the embryo proper appear by the globular stage of development; abnormalities in the suspensor are detected soon after at the heart stage. Storage protein and lipid bodies, which normally accumulate only in the embryo proper late in embryogenesis, are present in both the arrested embryo proper and enlarged suspensor. Therefore, cell differentiation in the embryo proper can proceed in the absence of normal morphogenesis, and the suspensor can be transformed into a structure with features normally restricted to the embryo proper. These observations are consistent with a model in which normal development of the embryo proper limits growth and differentiation of the suspensor. Altered development of the embryo proper in mutant seeds leads indirectly to proliferation of suspensor cells and expression of properties characteristic of the embryo proper. Ultimately, growth of the transformed suspensor is limited by the same genetic defect that disrupts development of the embryo proper. The availability of multiple alleles of sus1 and sus2, including T-DNA tagged alleles of each, will facilitate the cloning of these essential genes and molecular analysis of interactions between the embryo proper and suspensor early in development.

When it comes to policy issues, a legal scholar would traditionally study sustainable taxation from a “top-down” perspective, thus focusing on the legislator and on rational ways to steer economic life in a more sustainable direction. Here, we start at another end—we think of it as “the bottom-calling-the-top” perspective—in order to highlight (1) a relatively new business model and its merits from a circular economy perspective, namely the so called Product Service Systems; (2) how this model faces initial problems regarding especially foreseeability and that it might therefore have problems making its breakthrough; and (3) thus might need help from the legislator. Business models typically emanate from economic life rather than from political discourse, hence the bottom-up perspective and the bottom-to-the-top expression. Within the “bottom-calling-the-top” perspective, the focus lies on the needs of the economic actors carrying out their business. This text will contribute to the integration of a “bottom-up” perspective into the sustainable taxation discussion, and we will illustrate how a proper dose of bottom-up perspective might contribute to a more viable discussion.

The purpose of the article is to define an innovative concept of sustainable management based on the analysis of the institutional configuration of business ecosystem entities. The concept of sustainable (SUST) management is outlined as the creation of conditions for multi-entity management of territorial capital on the basis of digital platform technologies and the principles of partnership between government, business and the population. The institutional configuration of territorial business ecosystems is analyzed. It is determined that the business-ecosystem organization of economic activity is a flexible and dynamic system that creates opportunities for the integration of natural resources into economic circulation in territorial communities. It is established that there is a need for a radical restructuring of the institutional configuration of the spatial management system of natural resources in the direction of creating a system of multi-entity management of territorial capital. The main institutions and institutes of the spatial system of natural resources management of the territorial business ecosystem are outlined, in the field of local government, public-private partnership, corporate-platform and structural-project management, etc., within which economic actors of business ecosystems are endowed with appropriate functions and powers.. The structure of the business-ecosystem concept of sustainable management is detailed, including the basic formats of institutionalization of territorial assets management, informational and communicational platform component and management mechanisms of sustainable management, which are focused on creating a system of multi-entity management of sustainable development of territorial communities. Basic formats and forms of institutionalization of territorial assets management in the conditions of decentralization of power are proposed, including communication platforms of collaborative interactions between public authorities, business and population, as well as a set of mechanisms of structural and project management of sustainable development of territorial communities.

One of the topical issues is the study of the formation conditions, geological structure, chemical composition and methods of development of the "Duzdag" salt deposit, formed in the Babek region of the Nakhchivan depression on the territory of Azerbaijan, in order to meet the salt needs of the population. The field is divided into two sections: southern - Nakhchivan and Sust, located 4.5 km north-west of it. The physical and mechanical properties of both sections are close to each other. Losses are allowed during field development due to the fact that salt layers alternate with clay layers. The constantly growing demand for salt has predetermined the need to develop more advanced and progressive production methods based on the mechanization of technological processes, the use of new technologies and world experience. Depending on the formation of salt deposits, different methods of salt extraction are used in world practice. The main method of production of table salt in the world should be its extraction in the form of a solution and evaporation in the sun. The share of each of these methods is about 35%, and about 30% of the salt is extracted from an underground mine. The productive layer of the Nakhchivan rock salt deposit is 93-95% halite and is of high quality. The excess content of clay minerals in the field forces them to be used only in animal husbandry. Frosts drilled in the Nakhchivan rock salt deposit can be widely used in the treatment of liver diseases. The mountain has 130 million tons of natural salt reserves that are effective in treating respiratory ailments. Since the ice is horizontal, patients adapt to the underground part. To open new production facilities and treatment facilities in Duzdag, it is necessary to continue the installation of equipment that meets modern standards, the introduction of mines into a fully automated, controlled technological regime, equipped with a modern ventilation system. Resumo:
Carbon supported platinum catalysts for proton exchange membrane fuel cell (PEMFC) applications have been studied intensively in the scientific community.1,2 The catalytic activity of the catalyst depends on the characteristics of the carbon support material3 and on the Pt depositing method4,5. The aim of the study was to investigate the oxygen reduction reaction (ORR) on Pt nanoparticles deposited on peat-derived carbon. The Pt nanoparticles were deposited on the carbon support material by three different methods using NaBH4 (NBH), ethylene glycol (EG) and isopropyl alcohol (IA) as a reducing agent. The studied materials were characterized using N2 sorption, X-ray diffraction (XRD) and thermogravimetric analysis (TGA). Structure of the platinum nanocatalyst on carbon support was also studied using scanning electron microscopy with energy-dispersive X-ray analysis (SEM-EDX). For electrochemical characterization, the electrochemically active surface area (ECA) of the materials were measured in a three-electrode system (0,1 M HClO4) and in a completed PEMFC. The ORR kinetics of the materials were studied by the rotating disk electrode (RDE) method as well as in a PEMFC configuration. As a result, it was found that the higher the ECA of the material, the higher the catalytic activity. The catalytic activity of the synthesized materials increases in order: IA < NBH < EG. ECA of the materials increases in the same order. Also, the special surface area of the materials increases in the same order. The catalytic activities of the synthesized materials were compared to a commercial catalyst material, 60% Pt on HSA Ketjenblack. References O. Z. Sharaf and M. F.
