ABSTRACT An early step in the utilization of starch by Bacteroides thetaiotaomicron is the binding of starch to the bacterial surface. Four starch-associated outer membrane proteins of B. thetaiotaomicron that have no starch-degrading activity have been identified. Two of these, SusC and SusD, have been shown by genetic analysis to be required for starch binding. In this study, we provide the first biochemical evidence that these two proteins interact physically with each other. Both formaldehyde cross-linking and nondenaturing gel electrophoresis experiments showed that SusC and SusD interact to form a complex. Two other proteins encoded by genes in the same operon, SusE and SusF, proved not to be essential for starch utilization and actually decreased starch binding when they were present along with SusC and SusD. Consistent with this, nondenaturing gel analysis revealed that in a strain producing SusC, SusD, and SusE, the SusCD complex was partially destabilized. The strain producing SusC, SusD, and SusE also grew more slowly on starch than a strain producing SusC, SusD, SusE, and SusF (μmax, 0.29 and 0.37/h, respectively). Thus, SusE appears to interact with the SusCD complex. SusE also interacts with SusF, because SusE was less susceptible to proteinase K digestion when SusF was present, and nondenaturing gel analysis detected a complex formed by these two proteins. Our results indicate that SusC, SusD, SusE, and SusF form a protein complex in the outer membrane but that SusE and SusF are dispensable members of this complex.

ABSTRACT Bacteroides thetaiotaomicron, a gram-negative obligate anaerobe, utilizes polysaccharides by binding them to its cell surface and allowing cell-associated enzymes to hydrolyze them into digestible fragments. We use the starch utilization system as a model to analyze the initial steps involved in polysaccharide binding and breakdown. In a recent paper, we reported that one of the outer membrane proteins involved, SusG, had starch-degrading activity but was not sufficient for growth on starch. Moreover, SusG alone did not have detectable starch binding activity. Previous studies have shown that starch binding is essential for starch utilization. In this paper, we report that four other outer membrane proteins, SusC through SusF, are responsible for starch binding. Results of 14C-starch binding assays show that SusC and SusD both contribute a significant amount of starch binding. SusE also appears to contribute substantially to starch binding. Using affinity chromatography, we show in vitro that these Sus proteins interact to bind starch. Moreover, protease accessibility of either SusC or SusD greatly increased when one was expressed without the other. This finding supports the hypothesis that SusC and SusD interact in the outer membrane. Evidence from additional protease accessibility studies suggests that SusC, SusE, and SusF are exposed on the cell surface. Our results demonstrate that SusC and SusD act as the major starch binding proteins on the cell surface, with SusE enhancing binding. SusF's role in starch utilization has yet to be determined, although the fact that starch protected it from proteolytic attack suggests that it does bind starch.

ABSTRACT The human gut microbe Bacteroides fragilis can alter the expression of its surface molecules, such as capsular polysaccharides and SusC/SusD family outer membrane proteins, through reversible DNA inversions. We demonstrate here that DNA inversions at 12 invertible regions, including three gene clusters for SusC/SusD family proteins, were controlled by a single tyrosine site-specific recombinase (Tsr0667) encoded by BF0667 in B. fragilis strain YCH46. Genetic disruption of BF0667 diminished or attenuated shufflon-type DNA inversions at all three susC/susD genes clusters, as well as simple DNA inversions at nine other loci, most of which colocalized with susC/susD family genes. The inverted repeat sequences found within the Tsr0667-regulated invertible regions shared the consensus motif sequence AGTYYYN4GDACT. Tsr0667 specifically mediated the DNA inversions of 10 of the 12 regions, even under an Escherichia coli background when the invertible regions were exposed to BF0667 in E. coli cells. Thus, Tsr0667 is an additional globally acting DNA invertase in B. fragilis, which probably involves the selective expression of SusC/SusD family outer membrane proteins.

In this article, we introduce a new notion of weakly soft susc-contraction in soft metric spaces and prove a soft fixed point theorem which assure the existence of soft fixed points for the type of weakly soft susc-contraction. Our results generalize many recent soft fixed point results in the literature.

Members of the genera Proteiniphilum and Petrimonas were speculated to represent indicators reflecting process instability within anaerobic digestion (AD) microbiomes. Therefore, Petrimonas mucosa ING2-E5AT was isolated from a biogas reactor sample and sequenced on the PacBio RSII and Illumina MiSeq sequencers. Phylogenetic classification positioned the strain ING2-E5AT in close proximity to Fermentimonas and Proteiniphilum species (family Dysgonomonadaceae). ING2-E5AT encodes a number of genes for glycosyl-hydrolyses (GH) which are organized in Polysaccharide Utilization Loci (PUL) comprising tandem susCD-like genes for a TonB-dependent outer-membrane transporter and a cell surface glycan-binding protein. Different GHs encoded in PUL are involved in pectin degradation, reflecting a pronounced specialization of the ING2-E5AT PUL systems regarding the decomposition of this polysaccharide. Genes encoding enzymes participating in amino acids fermentation were also identified. Fragment recruitments with the ING2-E5AT genome as a template and publicly available metagenomes of AD microbiomes revealed that Petrimonas species are present in 146 out of 257 datasets supporting their importance in AD microbiomes. Metatranscriptome analyses of AD microbiomes uncovered active sugar and amino acid fermentation pathways for Petrimonas species. Likewise, screening of metaproteome datasets demonstrated expression of the Petrimonas PUL-specific component SusC providing further evidence that PUL play a central role for the lifestyle of Petrimonas species.

Sucrose synthase (Sus) is a key enzyme of sucrose metabolism. Two Sus-encoding genes (Sus1 and Sus2) from Arabidopsis thaliana were found to be profoundly and differentially regulated in leaves exposed to environmental stresses (cold stress, drought or O2 deficiency). Transcript levels of Sus1 increased on exposure to cold and drought, whereas Sus2 mRNA was induced specifically by O2 deficiency. Both cold and drought exposures induced the accumulation of soluble sugars and caused a decrease in leaf osmotic potential, whereas O2 deficiency was characterized by a nearly complete depletion in sugars. Feeding abscisic acid (ABA) to detached leaves or subjecting Arabidopsis ABA-deficient mutants to cold stress conditions had no effect on the expression profiles of Sus1 or Sus2, whereas feeding metabolizable sugars (sucrose or glucose) or non-metabolizable osmotica [poly(ethylene glycol), sorbitol or mannitol] mimicked the effects of osmotic stress on Sus1 expression in detached leaves. By using various sucrose/mannitol solutions, we demonstrated that Sus1 was up-regulated by a decrease in leaf osmotic potential rather than an increase in sucrose concentration itself. We suggest that Sus1 expression is regulated via an ABA-independent signal transduction pathway that is related to the perception of a decrease in leaf osmotic potential during stresses. In contrast, the expression of Sus2 was independent of sugar/osmoticum effects, suggesting the involvement of a signal transduction mechanism distinct from that regulating Sus1 expression. The differential stress-responsive regulation of Sus genes in leaves might represent part of a general cellular response to the allocation of carbohydrates during acclimation processes.

Sanfilippo syndrome, or mucopolysaccharidosis type III (MPS III), is a rare lysosomal disease caused by congenital enzymatic deficiencies in heparan sulfate (HS) degradation, leading to organ dysfunction. The most severe hallmark of MPS III comprises neurological alterations, although gastrointestinal symptoms (GISs) have also been shown to be relevant in many patients. Here, we explored the contribution of the gut microbiota to MPS III GISs. We analyzed the composition and functionality of the gut microbiota in two MPS III siblings with the same mutation (c.544C > T, c.1080delC, in the SGSH gene) and the same diet, but with differences in their GISs, including recurrent diarrhea in one of them. Using 16S sequencing, we observed that the MPS III patients exhibited decreased alpha diversity and a lower abundance of Lachnospiraceae and Bifidobacteriaceae accompanied by a higher abundance of the Ruminococcaceae and Rikenellaceae families than the healthy control subjects. Comparing siblings, we found an increased abundance of Bacteroidaceae and a lower abundance of Ruminococcaceae and Akkermansiaceae in the GIS-free patient. This patient also had a higher relative abundance of Sus genes (SusA, SusB, SusE, and SusG) involved in glycosaminoglycan metabolism. We found higher HS levels in the stool of the two MPS III patients than in healthy volunteers, particularly in the patient with GISs. Functionally, whole fecal metabolites from the patient with GISs induced oxidative stress in vitro in healthy monocytes. Finally, the Bacteroides thetaiotaomicron strain isolated from MPS III stool samples exhibited HS degradation ability. Overall, our results reveal different microbiota compositions and functionalities in MPS III siblings, who exhibited differential gastrointestinal symptomatology. Our study may serve as a gateway to explore the impact of the gut microbiota and its potential to enhance the quality of life in Sanfilippo syndrome patients.

ABSTRACT The 6.10-Mb genome sequence of the aerobic chitin-digesting gliding bacterium Flavobacterium johnsoniae (phylum Bacteroidetes) is presented. F. johnsoniae is a model organism for studies of bacteroidete gliding motility, gene regulation, and biochemistry. The mechanism of F. johnsoniae gliding is novel, and genome analysis confirms that it does not involve well-studied motility organelles, such as flagella or type IV pili. The motility machinery is composed of Gld proteins in the cell envelope that are thought to comprise the “motor” and SprB, which is thought to function as a cell surface adhesin that is propelled by the motor. Analysis of the genome identified genes related to sprB that may encode alternative adhesins used for movement over different surfaces. Comparative genome analysis revealed that some of the gld and spr genes are found in nongliding bacteroidetes and may encode components of a novel protein secretion system. F. johnsoniae digests proteins, and 125 predicted peptidases were identified. F. johnsoniae also digests numerous polysaccharides, and 138 glycoside hydrolases, 9 polysaccharide lyases, and 17 carbohydrate esterases were predicted. The unexpected ability of F. johnsoniae to digest hemicelluloses, such as xylans, mannans, and xyloglucans, was predicted based on the genome analysis and confirmed experimentally. Numerous predicted cell surface proteins related to Bacteroides thetaiotaomicron SusC and SusD, which are likely involved in binding of oligosaccharides and transport across the outer membrane, were also identified. Genes required for synthesis of the novel outer membrane flexirubin pigments were identified by a combination of genome analysis and genetic experiments. Genes predicted to encode components of a multienzyme nonribosomal peptide synthetase were identified, as were novel aspects of gene regulation. The availability of techniques for genetic manipulation allows rapid exploration of the features identified for the polysaccharide-digesting gliding bacteroidete F. johnsoniae.

The suspensor is the first differentiated structure produced during plant embryogenesis. In most angiosperms, the suspensor functions early in development to provide nutrients and growth regulators to the embryo proper. In Arabidopsis, the suspensor undergoes programmed cell death at the torpedo stage and is not present in mature seeds. We have identified at least 16 embryo-defective mutants of Arabidopsis that exhibit an enlarged suspensor phenotype at maturity. In this report, we focus on seven abnormal suspensor mutants, which define three genetic loci (sus1, sus2 and sus3). Recessive mutations at each of these loci disrupt morphogenesis in the embryo proper and consistently result in the formation of a large suspensor. Defects in the embryo proper appear by the globular stage of development; abnormalities in the suspensor are detected soon after at the heart stage. Storage protein and lipid bodies, which normally accumulate only in the embryo proper late in embryogenesis, are present in both the arrested embryo proper and enlarged suspensor. Therefore, cell differentiation in the embryo proper can proceed in the absence of normal morphogenesis, and the suspensor can be transformed into a structure with features normally restricted to the embryo proper. These observations are consistent with a model in which normal development of the embryo proper limits growth and differentiation of the suspensor. Altered development of the embryo proper in mutant seeds leads indirectly to proliferation of suspensor cells and expression of properties characteristic of the embryo proper. Ultimately, growth of the transformed suspensor is limited by the same genetic defect that disrupts development of the embryo proper. The availability of multiple alleles of sus1 and sus2, including T-DNA tagged alleles of each, will facilitate the cloning of these essential genes and molecular analysis of interactions between the embryo proper and suspensor early in development.

ABSTRACT Commensal enteric bacteria are a required pathogenic factor in inflammatory bowel disease (IBD), but the identity of the pertinent bacterial species is unresolved. Using an IBD-associated pANCA monoclonal antibody, a 100-kDa protein was recently characterized from an IBD clinical isolate of Bacteroides caccae (p2Lc3). In this study, consensus oligonucleotides were designed from 100-kDa peptides and used to identify a single-copy gene from the p2Lc3 genome. Sequence analysis of the genomic clone revealed a 2,844-bp (948 amino acid) open reading frame encoding features typical of the TonB-linked outer membrane protein family. This gene, termed ompW,was detected by Southern analysis only in B. caccae and was absent in other species of Bacteroides and gram-negative coliforms. The closest homologues of OmpW included the outer membrane proteins SusC of Bacteroides thetaiotaomicron and RagA of Porphyromonas gingivalis. Recombinant OmpW protein was immunoreactive with the monoclonal antibody, and serum anti-OmpW immunoglobulin A levels were elevated in a Crohn's disease patient subset. These findings suggest that OmpW may be a target of the IBD-associated immune response and reveal its structural relationship to a bacterial virulence factor of P. gingivalis and periodontal disease.

Identification of the community structure of phytoplankton is an important link in studying the structure and function of aquatic ecosystems. However, research on the community structure of phytoplankton in the Golmud area is very scarce at present. To explore the composition of phytoplankton in the Golmud area, eukaryotic phytoplankton and environmental parameters were sampled in September 2020. The composition and diversity of the eukaryotic phytoplankton community were determined by microscopic observation and Illumina high-throughput sequencing technology. The results showed that a total of 40 species of eukaryotic phytoplankton from 4 phyla were identified by microscopic observation, and 154 species of eukaryotic phytoplankton from 7 phyla were obtained by high-throughput sequencing, mainly Bacillariophyta and Chlorophyta. The dominant species were Dunaliella sp., Teleaulax sp., Parvodinium mixtum, and Lindavia radiosa. The eukaryotic phytoplankton density in summer was 0.09–12.08 × 105 cells/L, the total biomass was 0.002–0.55 mg/L, and the concentration of chlorophyll-a was 0.00–0.91 μg/L. Multiple α diversity indices showed that the diversity of water in different salinities was in the order of freshwater > brackish > saline. The β diversity results showed that the eukaryotic phytoplankton community composition was more similar in water with the same salinity. The redundancy analysis results of water with different salinities showed that environmental factors susc as salinity, total phosphorus concentration, and dissolved oxygen affected the eukaryotic phytoplankton community structure, among which salinity had the greatest influence.

ABSTRACT The rag locus of Porphyromonas gingivalis W50 encodes RagA, a predicted tonB-dependent receptor protein, and RagB, a lipoprotein that constitutes an immunodominant outer membrane antigen. The low G+C content of the locus, an association with mobility elements, and an apparent restricted distribution in the species suggested that the locus had arisen by horizontal gene transfer. In the present study, we have demonstrated that there are four divergent alleles of the rag locus. The original rag allele found in W50 was renamed rag-1, while three novel alleles, rag-2 to rag-4, were found in isolates lacking rag-1. The three novel alleles encoded variants of RagA with 63 to 71% amino acid identity to RagA1 and each other and variants of RagB with 43 to 56% amino acid identity. The RagA/B proteins have homology to numerous Bacteroides proteins, including SusC/D, implicated in polysaccharide uptake. Monoclonal and polyclonal antibodies raised against RagB1 of P. gingivalis W50 did not cross-react with proteins from isolates carrying different alleles. In a laboratory collection of 168 isolates, 26% carried rag-1, 36% carried rag-2, 25% carried rag-3, and 14% carried rag-4 (including the type strain, ATCC 33277). Restriction profiles of the locus in different isolates demonstrated polymorphism within each allele, some of which is accounted for by the presence or absence of insertion sequence elements. By reference to a previously published study on virulence in a mouse model (M. L. Laine and A. J. van Winkelhoff, Oral Microbiol. Immunol. 13:322-325, 1998), isolates that caused serious disease in mice were significantly more likely to carry rag-1 than other rag alleles.

Tomato (Lycopersicon esculentum) line E427 has resistance genes to three races of Fusarium oxysporum f.sp. lycopersici derived from L. pennellii (L.pen) accession LA 716 and L. pimpinellifolium (L.pimp) accession PI 126915. E427 was crossed to susc. Bonny Best and F2 and backcross seed were obtained. Progeny were inoculated separately with Fusarium wilt races 1, 2, or 3. Lines with suspected recombination of resistance were selfed and re-inoculated until disease reactions were homozygous. Four lines were obtained with resistance to both races 2 and 3, but susceptible to race 1. These lines had the L.pen alleles at RFLP markers linked to I-3 on chromosome 7 and lacked L.pimp alleles linked to I and I-2 on chromosome 11. Complementation (F2) data indicated race 2 resistance on chromosome 7 was controlled by a single dominant gene. Three lines were resistant to race 2, but susceptible to races 1 and 3. These lines had L.pimp alleles at TG105 indicating the presence of I-2, and no L.pen alleles at markers linked to I-3. Three lines were resistant to race 1, but susceptible to races 2 and 3. All three had L.pimp alleles at TG523 confirming linkage to I on chromosome 11 and no L.pen alleles at markers tightly linked to I-3. However, one of the lines had L.pen alleles at CT113 on chromosome 7. This and F2 complementation data suggests the possible location of a race 1 resistant locus, I1. Two lines that were Fusarium wilt race 3 resistant and susceptible to race 1 had intermediate resistance to race 2. These two lines did not have the L. pennellii alleles at TG183, TG174, and CT43 near the I-3 locus indicating crossovers in this region reduced race 2 resistance.

A Gram-negative, aerobic, rod-shaped, non-motile, yellow-pigmented bacterium, KMM 9835T, was isolated from the sediment sample obtained from the Amur Bay of the Sea of Japan seashore, Russia. Phylogenetic analyses based on the 16S rRNA gene and whole genome sequences positioned the novel strain KMM 9835T in the genus Mariniflexile as a separate line sharing the highest 16S rRNA gene sequence similarities of 96.6% and 96.2% with Mariniflexile soesokkakense RSSK-9T and Mariniflexile fucanivorans SW5T, respectively, and similarity values of <96% to other recognized Mariniflexile species. The average nucleotide identity and digital DNA–DNA hybridization values between strain KMM 9835T and M. soesokkakense KCTC 32427T, Mariniflexile gromovii KCTC 12570T, M. fucanivorans DSM 18792T, and M. maritimum M5A1MT were 83.0%, 82.5%, 83.4%, and 78.3% and 30.7%, 29.6%, 29.5%, and 24.4%, respectively. The genomic DNA GC content of strain KMM 9835T was 32.5 mol%. The dominant menaquinone was MK-6, and the major fatty acids were iso-C15:0, iso-C15:1ω10c, and C15:0. The polar lipids of strain KMM 9835T consisted of phosphatidylethanolamine, two unidentified aminolipids, an unidentified phospholipid, and six unidentified lipids. A pan-genome analysis showed that the KMM 9835T genome encoded 753 singletons. The annotated singletons were more often related to transport protein systems (SusC), transcriptional regulators (AraC, LytTR, LacI), and enzymes (glycosylases). The KMM 9835T genome was highly enriched in CAZyme-encoding genes, the proportion of which reached 7.3%. Moreover, the KMM 9835T genome was characterized by a high abundance of CAZyme gene families (GH43, GH28, PL1, PL10, CE8, and CE12), indicating its potential to catabolize pectin. This may represent part of an adaptation strategy facilitating microbial consumption of plant polymeric substrates in aquatic environments near shorelines and freshwater sources. Based on the combination of phylogenetic and phenotypic characterization, the marine sediment strain KMM 9835T (=KCTC 92792T) represents a novel species of the genus Mariniflexile, for which the name Mariniflexile litorale sp. nov. is proposed.

Abstract Background: Succinylacetone (SUAC) is the primary metabolite accumulated in tyrosinemia type I—an inborn error of metabolism that, if untreated, can cause death from liver failure during the first months of life. Newborn screening laboratories measure SUAC in dried blood spot (DBS) samples to detect asymptomatic tyrosinemia type I. We used panels of SUAC-enriched DBSs to compare and evaluate the performance of these screening tests. Methods: We prepared sets of DBS materials enriched with predetermined SUAC concentrations and distributed samples of these materials, along with a screening practices questionnaire, to laboratories that perform SUAC tests. We compared their reported SUAC concentrations and questionnaire responses to identify screening practices that affect SUAC test outcomes. Results: Data from 2 pilot surveys showed large differences among laboratories in SUAC recoveries, reproducible within-laboratory recoveries, and stable performance of the DBS materials. Results from 257 proficiency test analyses contained a total of 6 false-negative misclassifications. Reported recoveries of added SUAC ranged from 0 to &gt;200%. Low-biased SUAC recoveries were associated with 1 method used by 5 laboratories. All laboratories that reported SUAC recoveries ≥100% used DBS matrix calibrators. Conclusions: The wide ranges of SUAC concentrations reported for pilot and proficiency testing specimens demonstrate a need to harmonize quantitative results among laboratories. Although DBS matrix calibrators are important for optimizing SUAC recoveries, the preparation of these calibrators is not standardized among laboratories. Certified DBS-based SUAC calibrators are needed for accuracy and harmonization.

Abstract Sucrose synthase (SuSy) is one of two enzyme families capable of catalyzing the first degradative step in sucrose utilization. Several earlier studies examining SuSy mutants in Arabidopsis failed to identify obvious phenotypic abnormalities compared with wild-type plants in normal growth environments, and as such a functional role for SuSy in the previously proposed cellulose biosynthetic process remains unclear. Our study systematically evaluated the precise subcellular localization of all six isoforms of Arabidopsis SuSy via live-cell imaging. We showed that yellow fluorescent protein (YFP)-labeled SuSy1 and SuSy4 were expressed exclusively in phloem companion cells, and the sus1/sus4 double mutant accumulated sucrose under hypoxic conditions. SuSy5 and SuSy6 were found to be parietally localized in sieve elements and restricted only to the cytoplasm. SuSy2 was present in the endosperm and embryo of developing seeds, and SuSy3 was localized to the embryo and leaf stomata. No single isoform of SuSy was detected in developing xylem tissue of elongating stem, the primary site of cellulose deposition in plants. SuSy1 and SuSy4 were also undetectable in the protoxylem tracheary elements, which were induced by the vascular-related transcription factor VND7 during secondary cell wall formation. These findings implicate SuSy in the biological events related to sucrose translocation in phloem.

Sus1 acts in nuclear mRNA export via its association with the nuclear pore-associated Sac3–Thp1–Cdc31 complex. In addition, Sus1 plays a role in transcription through its interaction with the Spt/Ada/Gcn5 acetyltransferase (SAGA) complex. Here, we have analyzed function and interaction of Sus1 within the SAGA complex. We demonstrate that Sus1 is involved in the SAGA-dependent histone H2B deubiquitinylation and maintenance of normal H3 methylation levels. By deletion analyses, we show that binding of Sus1 to SAGA depends on the deubiquitinylating enzyme Ubp8 and Sgf11. Moreover, a stable subcomplex between Sus1, Sgf11, and Ubp8 could be dissociated from SAGA under high salt conditions. In vivo recruitment of Sus1 to the activated GAL1 promoter depends on Ubp8 and vice versa. In addition, histones coenrich during SAGA purification in a Sus1–Sgf11–Ubp8-dependent way. Interestingly, sgf11 deletion enhances the mRNA export defect observed in sus1Δ cells. Thus, the Sus1–Sgf11–Ubp8 module could work at the junction between SAGA-dependent transcription and nuclear mRNA export.

Fusarinine C (FSC) has recently been shown to be a promising and novel chelator for 89Zr. Here, FSC has been further derivatized to optimize the complexation properties of FSC-based chelators for 89Zr-labeling by introducing additional carboxylic groups. These were expected to improve the stability of 89Zr-complexes by saturating the 8-coordination sphere of [89Zr] Zr4+, and also to introduce functionalities suitable for conjugation to targeting vectors such as monoclonal antibodies. For proof of concept, succinic acid derivatization at the amine groups of FSC was carried out, resulting in FSC(succ)2 and FSC(succ)3. FSC(succ)2 was further derivatized to FSC(succ)2 AA by reacting with acetic anhydride (AA). The Zr4+ complexation properties of these chelators were studied by reacting with ZrCl4. Partition coefficient, protein binding, serum stability, acid dissociation, and transchelation studies of 89Zr-complexes were carried out in vitro and the results were compared with those for 89Zr-desferrioxamine B ([89Zr]Zr-DFO) and 89Zr-triacetylfusarinine C ([89Zr]Zr-TAFC). The in vivo properties of [89Zr]Zr-FSC(succ)3 were further compared with [89Zr]Zr-TAFC in BALB/c mice using micro-positron emission tomography/computer tomography (microPET/CT) imaging. Fusarinine C (succ)2AA and FSC(succ)3 were synthesized with satisfactory yields. Complexation with ZrCl4 was achieved using a simple strategy resulting in high-purity Zr-FSC(succ)2AA and Zr-FSC(succ)3 with 1:1 stoichiometry. Distribution coefficients of 89Zr-complexes revealed increased hydrophilic character compared to [89Zr]Zr-TAFC. All radioligands showed high stability in phosphate buffered saline (PBS) and human serum and low protein-bound activity over a period of seven days. Acid dissociation and transchelation studies exhibited a range of in vitro stabilities following the order: [89Zr]Zr-FSC(succ)3 > [89Zr]Zr-TAFC > [89Zr]Zr-FSC(succ)2AA >> [89Zr]Zr-DFO. Biodistribution studies of [89Zr]Zr-FSC(succ)3 revealed a slower excretion pattern compared to [89Zr]Zr-TAFC. In conclusion, [89Zr]Zr-FSC(succ)3 showed the best stability and inertness. The promising results obtained with [89Zr]Zr-FSC(succ)2AA highlight the potential of FSC(succ)2 as a monovalent chelator for conjugation to targeted biomolecules, in particular, monoclonal antibodies.

Sudden unexpected postnatal collapse (SUPC) is a rare event, potentially associated with catastrophic consequences. Since the beginning of the 2000s, therapeutic hypothermia (TH) has been proposed as a treatment for asphyxiated neonates after SUPC. However, only a few studies have reported the outcome of SUPC after TH. The current study presents the long-term neurodevelopmental outcome of four cases of SUPC treated with TH in a single Italian center. Furthermore, we reviewed the previous literature concerning 49 cases of SUPC treated with TH. Among 53 total cases (of whom four occurred in our center), 15 (28.3%) died before discharge from the NICU. A neurodevelopmental follow-up was available only for 21 (55.3%) out of the 38 surviving cases, and seven infants developed neurodevelopmental sequelae. TH should be considered in neonates with asphyxia after SUPC. However, SUPC is a rare event, and there is a lack of comparative clinical data to establish the risk/benefit of TH after SUPC with different degrees of asphyxia. Analysis of large cohorts of newborns with SUPC, whether treated with TH or untreated, are needed in order to better identify infants who should undergo TH.

Purpose The purpose of this paper is threefold: first, to shed light on the increasing start-up competitions (SUCs) phenomenon; second, to provide an interpretive framework to understand whether the SUCs have the potential to be effective entrepreneurial learning environment; third, to analyse the different roles of public and private actors in organizing SUCs. Design/methodology/approach The paper presents a cross-section analysis of the Italian SUCs population. In total, 77 competitions are analysed on the basis of different criteria which should properly mirror their distinguishing structural features, helping understand the potential of SUCs as learning environments. Findings The recent increase in the number of SUCs has been mainly fuelled by private actors. Moreover, Italian SUCs show some features that make them rich learning environments. Private and public actors play different roles, as confirmed by statistical tests performed. Privately organized SUCs follow mainly a market-oriented approach, while publicly organized ones are more education oriented. Research limitations/implications The findings cannot be easily generalized mainly due to the peculiarities of the Italian context. Practical implications Soft forms of regulation should be defined to strengthen those features which could potentially support the entrepreneurial learning processes. In this view, SUCs should be part of a start-up friendly ecosystem where actors (startuppers, incubators, venture capitalists) are effectively coordinated with each other. Originality/value Despite the remarkable diffusion of SUCs, there are significant gaps in literature about this phenomenon. Given the lack of a systematic and comprehensive analysis of SUCs as effective entrepreneurial learning environments, the paper represents an important starting point.

Objective Hypothermia is considered the treatment of choice for newborns with hypoxic-ischemic encephalopathy. Even though currently no consensus exists regarding the opportunity to extend this treatment also to newborns with sudden unexpected postnatal collapse (SUPC), this treatment is frequently adopted. The aim of the study was to evaluate whether the availability of hypothermia also for collapsed newborns in a tertiary level center may impact the estimated incidence of SUPC. Study Design A retrospective study was performed comparing the number of newborns admitted with the diagnosis of SUPC in the tertiary referral Neonatal intensive care unit of Florence in the period before (2008–2010) and after (2011–2014) the introduction of hypothermia treatment for newborns with SUPC. Result After introducing active cooling also to newborns with SUPC, the number of newborns with SUPC admitted to our center augmented and, consequently, SUPC incidence apparently increased from 5.3 to 15.5/100,000 live births, even though this difference did not reach statistical significance (odds ratio = 0.340; 95% confidence interval = 0.071–1.639; p = 0.179). Conclusion The apparent increase of SUPC incidence is probably related to a more effective centralization of newborns in our referral center but suggests that the real incidence of SUPC is probably significantly higher than currently estimated.

PURPOSE: The aim of this economic assessment was to evaluate the impact of a new nurse-led model of care, the Symptom and Urgent Review Clinic (SURC), for patients with cancer experiencing disease- or treatment-related symptoms. METHODS: An economic assessment was undertaken to estimate costs of the SURC from the service funder perspective and to compare the cost with cost offsets stemming from the implementation of the SURC. The cost offsets focused on the changes in emergency department (ED) presentations and inpatient admissions during a comparable 6-month period before and after the SURC implementation. Costs were analyzed in 2018 Australian dollars, and return on investment was calculated by comparing the cost offsets in the ED and inpatient units with the cost of the SURC. RESULTS: After the implementation of the SURC, patients were less likely to present to the ED (7.2% v 8.5%; P = .01), and patients who did present to the ED were more likely to be admitted to inpatient units (78% v 71%; P = .03) for additional treatment. The post-SURC period had a net cost savings of $37,090 compared with the pre-SURC period. From the service funder perspective, the SURC achieved an investment return of $1.73 for every dollar invested in the new service. CONCLUSION: Our study establishes the economic credentials of a new care model using empirical linked hospital service data. The SURC presents a new cancer care service for policy consideration from an economic standpoint. It demonstrates an efficient approach to hospital resource allocation to deliver quality cancer care.

The analysis on codon usage bias of Riemerella anatipestifer (RA) RagB/SusD gene (GenBank accession No. NC_017045.1) may improve our understanding of the evolution and pathogenesis of RA and provide a basis for understanding the relevant mechanism for biased usage of synonymous codons and for selecting appropriate expression systems to improve the expression of target genes. In this study, the synonymous codon usage in the RagB/SusD gene of RA and 19 reference bacteroidetes have been investigated. The results showed that codon usage bias in the RagB/SusD gene was strong bias towards the synonymous codons with A and T at the third codon position. A high level of diversity in codon usage bias existed, and the effective number of codons used in a gene plot revealed that the genetic heterogeneity in RagB/SusD gene of bacteroidetes was constrained by the G + C content. The codon adaptation index (CAI), effective number of codons (ENC), and GC3S values indicated synonymous codon usage bias in the RagB/SusD gene of bacteroidetes, and this synonymous bias was correlated with host evolution. The phylogentic analysis suggested that RagB/SusD was evolutionarily closer to Ornithobacterium rhinotracheale and that there was no significant deviation in codon usage in different bacteroidetes. There are 25 codons showing distinct usage differences between RA RagB/SusD and E. coli, 30 between RA RagB/SusD and Homo sapiens, 26 codons between RA RagB/SusD and yeast. Therefore the yeast and E. coli expression system may be suitable for the expression of RA RagB/SusD gene if some codons could be optimized.

Abstract Background: Tyrosinemia type I (TYR 1) is a disorder causing early death if left untreated. Newborn screening (NBS) for this condition is problematic because determination of the diagnostic marker, succinylacetone (SUAC), requires a separate first-tier or only partially effective second-tier analysis based on tyrosine concentration. To overcome these problems, we developed a new assay that simultaneously determines acylcarnitines (AC), amino acids (AA), and SUAC in dried blood spots (DBS) by flow injection tandem mass spectrometry (MS/MS). Methods: We extracted 3/16-inch DBS punches with 300 μL methanol containing AA and AC stable isotope-labeled internal standards. This extract was derivatized with butanol-HCl. In parallel, we extracted SUAC from the residual filter paper with 100 μL of a 15 mmol/L hydrazine solution containing the internal standard 13C5-SUAC. We combined the derivatized aliquots in acetonitrile for MS/MS analysis of AC and AA with additional SRM experiments for SUAC (m/z 155–137) and 13C5-SUAC (m/z 160–142). Analysis time was 1.2 min. Results: SUAC was increased in retrospectively analyzed NBS samples of 11 TYR 1 patients (length of storage, 52 months to 1 week; SUAC range, 13–81 μmol/L), with Tyr concentrations ranging from 65 to 293 μmol/L in the original NBS analysis. The mean concentration of SUAC in 13 521 control DBS was 1.25 μmol/L. Conclusion: The inclusion of SUAC analysis into routine analysis of AC and AA allows for rapid and cost-effective screening for TYR 1 with no tangible risk of false-negative results.

Development of spherical ultrasonic motor (SUSM) for space is an aim of this investigation. SUSM is a motor of multi-DOF. It's necessary to clear the various conditions to use a motor at space. In this paper, to confirm the durability to the radiant heat from the sun, drive experiments of SUSM was conducted in high temperature condition. As a result, our SUSM maintained the drive performance in high temperature condition. Materials of piezoelectric elements and of adhesives used in SUSM for space were important elements to maintain its drive performance in space. Selection results of the piezoelectric element and the adhesive were also reported in detail.

Background: Although an orphan disease with still obscure aetiopathogenesis, Susac syndrome has to be considered as differential diagnosis in multiple sclerosis (MS), since its clinical presentation and paraclinical features including routine magnetic resonance imaging (MRI) findings partially overlap. Objective: We aimed to study a potential benefit of 7T MRI for (i) the differentiation between Susac syndrome and MS and (ii) the clarification of pathogenesis of Susac syndrome. Methods: Five patients suffering from Susac syndrome, 10 sex- and age-matched patients with relapsing–remitting MS (median Expanded Disability Status Scale (EDSS) score 1.5) and 15 matching healthy controls were investigated at 7 Tesla MRI. The protocol included T1-weighted MPRAGE, T2*-weighted FLASH, and TIRM sequences. Results: Almost all T2* FLASH lesions in patients with MS were centred by a small central vein (325 lesions; 92%) and often showed a small hypointense rim (145 lesions; 41%). In contrast, white matter lesions in Susac syndrome exhibited a perivascular setting significantly less frequently (148 lesions; 54%, p=0.002), and very rarely exhibited a hypointense rim (12 lesions; 4%, p=0.004). Furthermore, in addition to callosal atrophy, Susac patients showed cerebrospinal fluid-isointense lesions within the central part of corpus callosum that are not commonly seen in MS. Conclusion: At 7T MRI, plaques in MS patients and patients with Susac syndrome differed substantially with respect to morphology and pattern. Thus, lesion morphology at 7T (i) may serve as a marker to distinguish Susac syndrome from MS and (ii) reflects a different pathophysiological mechanism underlying Susac syndrome, for example microinfarction rather than demyelination.

Performance, durability, and efficiency limitations of proton exchange membrane fuel cells (PEMFCs), as one of the alternative clean energy resources, has been hindering the widespread adoption of these devices. Improving those parameters is closely entangled with identifying the degradation mechanism of each component in the electrodes i.e., catalyst – commonly platinum nanoparticles, catalyst support – carbon, and the ion conductive polymer – ionomer, as well as the effects that degradation can have on the porous network of the electrodes[1]. Degradation during typical automotive operation protocols like startup/shut down (SUSD)[2,3] is of a special interest. Therefore, it is essential to observe the changes of the microstructure, as whole, and each individual component’s response to the working condition, in order to understand degradation mechanisms and devise degradation mitigation strategies. Transmission electron microscopy (TEM) and scanning transmission electron microscopy (STEM) in conjunction with energy dispersive spectroscopy (EDS) have proven to be valuable tools in unravelling the complex changes of the microstructure and understanding of the degradation mechanisms[4]. Moreover, correlating the numerical microstructural descriptors, extracted by quantification of the TEM and EDS results, with the visual observations helped the research community to draw a connection between the degradation and microstructural changes[5]. However, the conventionally prepared TEM samples by slicing (microtomy) an epoxy-embedded piece of a catalyst coated membrane (CCM) have limitations in observing carbon and ionomer, and distinguishing them from each other and from the epoxy. Partial epoxy-embedding showed very promising results regarding distinguishing carbon from ionomer in selected regions of the CCM[6]. However, not a full CCM was observed. Therefore, we report for the first time, an epoxy-free microtomy approach for a whole CCM, successfully implemented on a fresh and degraded cathode layer of beginning-of-life (BOL) and SUSD samples, respectively. Electrochemical characterization of the samples presented here are discussed in previous publication[2]. The Pt catalyst, carbon, ionomer and porous network of the cathodes were clearly observable and distinguishable in both BOL and SUSD samples using high-resolution TEM and 3D electron tomography-TEM (ET-TEM) imaging. The microstructural comparison revealed the changes in Pt particles distribution – moving into the ionomer network rather than sitting on and inside the carbon support, disintegration of corroded carbon and dispersion of broken-off graphitic carbon pieces in the ionomer, and alteration of the ionomer network – formation of more ionomer filaments. In addition, ET-TEM of the BOL and SUSD samples confirmed the presence of Pt particles inside the amorphous core of the carbon particle and their migration during SUSD operation, respectively. Finally, the visual results were correlated with the microstructural descriptors quantified using our proprietary quantification technique. The numerical microstructure properties corroborated the observations that was made based on the TEM images observations. Therefore, implementation of whole-CCM-epoxy-free microtomy technique can be considered as a novel method in aiding the research community in understanding the degradation mechanism and microstructural properties alteration. Retino-cochleo-cerebral angiopathy or Susac syndrome is a rare autoimmune disease that selectively affects the vessels of the retina, the inner ear and the central nervous system. Differentiation of Susac syndrome and multiple sclerosis presents difficulties due to the similarity of MRI semiotics of these two diseases. This article presents two clinical cases of patients with Susac syndrome who were diagnosed with multiple sclerosis at the onset of the disease. Based on the analysis of our own clinical observations and literature data, the issues of differential diagnosis of Susac syndrome and multiple sclerosis are highlighted. For the first time a variant of the MRI picture transformation in Susac syndrome is presented.

Various start-up circuits (SUCs) for low input voltage self-powered direct current-type (DC-type) energy harvesters have been proposed in recent years, and their structures have been evolving and changing continuously. A general classification and systematic overview of SUCs are presented here. With the development of CMOS process technologies, recent advances in SUCs have paved the way for lower input voltage, simpler structures, lower cost, and better performance. So far, no works have given an overall description of SUCs for low input voltage self-powered energy harvesters. This work is intended to not only provide insights into the topologies of SUCs, but also make a conclusion which identifies their developing tendency.

Neste estudo procurou-se levantar os perfis sociodemográficos registrados nas ocorrências e tentativas de suic�dio ocorridas nos munic�pios de Anastácio e Aquidauana, Estado de Mato Grosso do Sul no per�odo de 2007 a 2016. Nesse sentido foram utilizadas as fontes do Instituto Brasileiro de Geografia e Estat�stica (IBGE), para coleta dos dados censitários sobre a população residente; Departamento de Informática do SUS (DATASUS), para coleta de dados de suic�dio nacional e estadual; Unidade Regional de Per�cia e Identificação – URPI / Pol�cia Civil, para o levantamento das ocorrências de suic�dios de Aquidauana e Anastácio; Vigilâncias Epidemiológicas do Munic�pio de Aquidauana e do Munic�pio de Anastácio, para coleta de dados sobre as tentativas de suic�dio; 1º Subgrupamento de Bombeiro Militar de Aquidauana, para coleta de dados sobre as tentativas de suic�dio de Aquidauana e Anastácio. Constatou-se a existência de subnotificação e a falta de padronização nos registros de tentativas de suic�dio. Em relação às ocorrências conclui-se que as crianças e adolescentes utilizam-se do enforcamento como principal método de tirar a própria vida. Abstract. Tourism in Burau District, East Luwu Regency has great potential for development. This is because most of the area is a coastal area and borders directly on Bone Bay. Two beaches in Burau District that are popular with tourists are Lemo Beach and Ujung Suso Beach. Ujung Suso Beach, which has a lot of potential to be managed, currently has not received special attention from the East Luwu Regency government. As a result of the government's lack of attention in the process of managing Ujung Suso Beach, it has a negative impact on its natural conditions, such as severe coastal erosion which causes the distance between the seashore and the road to become closer, and increasing piles of rubbish around the beach in the long term can damage the ecosystem on the beach. The Ujung Suso Beach tourist attraction requires supporting facilities. Therefore, currently a regional tourism management strategy is needed that can improve the quality of facilities and infrastructure, and the natural resources of Ujung Suso Beach. The data collected includes visitor data, tourism potential consisting of physical potential, social culture and previous management methods of Ujung Suso Beach. The results of the SWOT analysis show that the Ujung Suso Beach tourist attraction is at the coordinate point in quadrant II with the strategy used to prioritize, namely the WO strategy, where the strategy used to prioritize is taking advantage of opportunities to overcome weaknesses. Pantai Ujung Suso yang memiliki banyak potensi untuk di Kelola, saat ini belum mendapat perhatian khusus dari pemerintah Kabupaten Luwu Timur. Akibat dari kurangnya perhatian pemerintah dalam proses pengelolaan Pantai Ujung Suso berdampak buruk terhadap kondisi alamnya seperti parahnya erosi pantai yang menyebabkan jarak antara tepi laut denan jalan semakin dekat, tumpukan sampah yang semakin banyak banyak disekitar pantai dalam jangka waktu Panjang dapat merusak ekosistem di pantai tersebut. Objek Wisata Pantai Ujung Suso membutuhkan sarana yang mendukung, Oleh karena itu saat ini dibutuhkan strategi pengelolaan wisata Kawasan yang dapat memperbaiki kualitas sarana dan prasarana, dan sumber daya alam Pantai Ujung Suso. Data yang di kumpulkan meliputi data pengunjung, potensi wisata yang terdiri dari potensi fisik, social budaya dan metode pengelolaan Pantai Ujung Suso sebelumnya. Hasil analisis SWOT menunjukkan objek wisata Pantai Ujung Suso berada pada titik koordinat kuadran II dengan strategi yang di gunakan untuk diprioritaskan yaitu strategi WO dimana Rumusan strategi yang digunakan untuk di prioritaskan yaitu memanfaatkan peluang untuk mengatasi kelemahan.

Data yang dikumpulkan meliputi data pengunjung, potensi wisata yang terdiri dari potensi fisik, sosial budaya, serta ekonomi, dan metode pengelolaan Pantai Ujung Suso sebelumnya. Selanjutnya wawancara dan Indepth interview dilakukan untuk mengetahui bagaimana metode pengelolaan untuk pengembangan Pantai Ujung Suso dengan menggunakan SWOT. Hasil penelitian menunjukkan bahwa beberapa kekuatan yang merupakan potensi wisata Pantai Ujung Suso adalah social budaya masyarakat, ekosistem pantai, perekonomian, serta kemudahan akses. Selain kekuatan yang merupakan potensi Pantai Ujung Suso, ditemukan pula beberapa kelemahan seperti pengambilan terumbu karang liar oleh masyarakat, kebersihan lingkungan pantai yang semakin mengkhawatirkan akibat aktivitas pengunjung dan warung di sekitar pantai yang membuang sampah disekitar pantai, persepsi masyarakat terkait pengembanagan wisata adalah dengan mendatangkan wisatawan sebanyak-banyaknya. Partisipasi masyarakat dalam kegiatan wisata perlu dikembangkan dan didampingi. Selain itu wisata pantai Ujung Suso juga mempunayai peluang secara ekonomi dengan memberikan kesempatan kerja dan berusaha bagi masyarakat disekitar pantai.

Selain kekuatan yang merupakan potensi Pantai Ujung Suso, ditemukan pula beberapa kelemahan seperti pengambilan terumbu karang liar oleh masyarakat, kebersihan lingkungan pantai yang semakin mengkhawatirkan akibat aktivitas pengunjung dan warung di sekitar pantai yang membuang sampah disekitar pantai, persepsi masyarakat terkait pengembanagan wisata adalah dengan mendatangkan wisatawan sebanyak-banyaknya. Partisipasi masyarakat dalam kegiatan wisata perlu dikembangkan dan didampingi. Selain itu wisata pantai Ujung Suso juga mempunayai peluang secara ekonomi dengan memberikan kesempatan kerja dan berusaha bagi masyarakat disekitar pantai.

Enforced expression of GNG11, G-protein subunit γ 11, induces cellular senescence in normal human diploid fibroblasts. We here examined the effect of the expression of GNG11 on the growth of immortalized human cell lines, and found that it suppressed the growth of SUSM-1 cells, but not of HeLa cells. We then compared these two cell lines to understand the molecular basis for the action of GNG11. We found that expression of GNG11 induced the generation of reactive oxygen species (ROS) and abnormal nuclear morphology in SUSM-1 cells but not in HeLa cells. Increased ROS generation by GNG11 would likely be caused by the down-regulation of the antioxidant enzymes in SUSM-1 cells. We also found that SUSM-1 cells, even under normal culture conditions, showed higher levels of ROS and higher incidence of abnormal nuclear morphology than HeLa cells, and that abnormal nuclear morphology was relevant to the increased ROS generation in SUSM-1 cells. Thus, SUSM-1 and HeLa cells showed differences in the regulation of ROS and nuclear morphology, which might account for their different responses to the expression of GNG11. Thus, SUSM-1 cells may provide a unique system to study the regulatory relationship between ROS generation, nuclear morphology, and G-protein signaling.

During the COVID-19 pandemic, Susi Air as an airline service company experienced a decrease in the number of passenger flights and cargo flights. This study aims to determine the effect of COVID-19 on the number of passenger and cargo flights and the way Susi Air has stabilized the number of flights during the COVID-19 pandemic. The method in this study uses a combined method or mixed methods. This study uses two types of data, namely primary data in the form of observation data carried out directly by researchers in the field, and secondary data in the form of time series data on the number of passenger flights and cargo flights of Susi Air Airlines at Nusawiru Cijulang Airport and interview data conducted with two operational staff. Susi Air Airlines. The results of the study show that the COVID-19 pandemic affects the number of passenger flights with Susi Air Airlines with a significance value in the run test, which is 0.000, the COVID-19 pandemic affects the number of Susi Air cargo flights with a significance value in the t test, namely amounting to 0.000, as well as the way Susi Air has stabilized the number of flights during the COVID-19 pandemic, namely by utilizing Instagram social media as a promotional tool, conducting ticket sales cooperation through the Traveloka platform, establishing pioneering flight collaborations between Susi Air Airlines and the Provincial Government. , providing discounts on airline ticket prices, and providing Charter Medical Evacuation (Medevac Emergency Indonesia) services as one of its service products.

Sudden unexpected postnatal collapse (SUPC) of healthy newborns is a catastrophic event caused by cardiorespiratory collapse in a healthy newborn. The most common cause of SUPC is poor positioning of the newborn during skin-to-skin contact or breastfeeding when the newborn is not being observed by a health professional, attentive parent, or caretaker. Maternal/newborn health care professionals need to know about the essential information, definitions, incidence, risk factors, clinical presentation, outcomes, and prevention and management strategies to minimize the occurrence and impact of SUPC. A sample SUPC hospital policy is included in the manuscript.

Subcutaneous (subc) abdominal and femoral adipose tissue metabolism was studied in sedentary and endurance-trained premenopausal women. Both fat cell weight and lipoprotein lipase (LPL) activity were lower in the subc abdominal depot of trained women compared with controls. Epinephrine- and isoproterenol-stimulated lipolytic responses, as well as the beta-adrenergic sensitivity of subc abdominal adipocytes, were higher in trained than in sedentary women, whereas both the antilipolytic effect of brimonidine (UK-14304) and the alpha 2-adrenoceptor sensitivity were lower in endurance-trained than in sedentary subjects. Maximal lipolysis in the presence of postadrenoceptor agents was also enhanced in subc abdominal adipose cells of trained women compared with sedentary controls. Negative relationships were found between maximal lipolytic responses of subc abdominal fat cells to catecholamines or to postreceptor agents and body fatness as well as abdominal fat distribution indexes. It is concluded that 1) endurance-trained women are characterized by a preferential lipid mobilization from the subc abdominal fat depot, and 2) differences in the metabolic characteristics of subc abdominal adipocytes between trained and sedentary women may involve changes in both LPL activity and the lipolytic cascade that include modifications at receptor and postreceptor levels. However, these alterations appear to be largely resulting from the reduced adipose cell size rather than from exercise training per se.

Susac syndrome is a rare autoimmune disorder characterised by the clinical triad of encephalopathy, retinopathy (branch retinal artery occlusions) and hearing loss. The diagnosis of Susac syndrome may be difficult initially, and it is not uncommon for patients with Susac syndrome to be misdiagnosed with multiple sclerosis. In this case report, we describe a patient who was diagnosed as having multiple sclerosis for three years, with further deterioration after starting treatment with interferon beta-1a. The patient had the triad of encephalopathy, branch retinal artery occlusions and sensorineural hearing loss. She had the classic magnetic resonance imaging appearance, with normal magnetic resonance imaging of the spinal cord and absence of oligoclonal bands in the cerebrospinal fluid. Our patient responded well to treatment with a combination therapy and discontinuation of interferon beta-1a. Our observations raise awareness about the importance of the early and correct diagnosis of Susac syndrome, which usually affects young patients, with an excellent prognosis if treated aggressively at an early stage of the disease. Susac syndrome is underdiagnosed and is not uncommonly misdiagnosed as multiple sclerosis. Susac syndrome is a great mimicker of multiple sclerosis, and establishing diagnostic criteria for this syndrome is very useful. In any patient presenting with a progressive disabling neurological disorder associated with callosal lesions and/or hearing loss, and/or visual loss especially in women, Susac syndrome should be suspected.

Liberating the SUCs from limited resources is a challenge they face; otherwise excellence in the areas of instruction, research, extension, and production will be at stake. As such, they need to establish income generating projects or IGPs. A descriptive study evaluated the Income Generating Projects (IGPs) of the State Universities and Colleges (SUCs) in Region VIII. The detailed assessment of the IGPs of the SUCs focused on the profile of the IGP administrators and employees; profile of IGPs of SUCs in Region VIII; status of the IGPs of SUCs in region VIII as perceived by the IGP administrators, employees and clients in terms of setting up an IGP programs, organizational structure, operation performance measurement, profit sharing and accounting. All aspects of operations,namely, setting up an IGP program, organizational structure, operations, performance measurement, profit sharing and accounting, were rated as “often” as perceived and were found to have highly significant differences of perceptions. It was recommended that administrators and the entire management of the different SUCs in Region should strengthen the operations of IGPs through giving it as a priority and through conforming to the standards set in the proposed IGP Manual.

ABSTRACT Results from previous studies had suggested that Bacteroides thetaiotaomicron utilizes starch by binding the polysaccharide to the bacterial surface and subsequently degrading the polymer by using cell-associated enzymes. Most of the starch-degrading activity was localized to the periplasm, but a portion appeared to be membrane associated. This raised the possibility that some breakdown might occur in the outer membrane prior to exposure of the polysaccharide to the periplasmic polysaccharide-degrading enzymes. In this study, we show that SusG, an outer membrane protein which has been shown genetically to be essential for starch utilization, has enzymatic activity. Results of protease accessibility experiments support the hypothesis that SusG is exposed on the cell surface. Results of [14C]starch binding assays, however, show that SusG plays a negligible role in binding of starch to the cell surface. Consistent with this, SusG has a relatively high Km for starch and by itself is not sufficient to allow cells to grow on starch or to bind starch. Hence, the main role of SusG is to hydrolyze starch, but the binding of starch to the cell surface is evidently mediated by other proteins presumably interacting with SusG.

Information security is imperative for state colleges and universities (SUCs) to have a sound information security in protecting their information assets, enhancing institutional reputation and ensuring compliance with the law. This paper presents the level of implementation of information security in the state colleges and universities (SUCs) in the Philippines. Data used was based on questionnaire adapted from the Information Security Governance (ISG) Assessment Tool for Higher Education(EDUCAUSE/Internet2 Security Task 2004) and was distributed to a total of 30 individuals who are responsible in managing and keeping the information assets of the SUCS. Findings revealed that information security is not yet fully implemented among the SUCS, this is a clear indication of failure or disaster, and these institutions are vulnerable to information security threats. SUCs are encouraged to implement comprehensive information security program at a strategic level involving the board of regents and top executives for safety and security.
