This is a feasibility study for the application of a novel concept of single-needle device for localized chemotherapy. Systemic chemotherapy has numerous and severe side effects. To conduct localized (electro)chemotherapy, we designed a novel device that does not currently exist on the market. Electrochemotherapy is based on the cell membranes temporary or permanent permeabilization using an electric current of defined characteristics. Electroporation can be reversible, when after a period of opened pores and membrane permeability increasing, membranes and cells return to their original state without damage. Electroporation can be an irreversible process when the pores on the membrane remain permanently open, electrolyte imbalance occurs resulting in cell death. Electrochemotherapy involves a combination of cytostatics and reversible electroporation, when pores on the cell membrane are temporarily opened and, during that short period, a large amount of cytostatic is entered into the cell, which is a macromolecule that would not normally penetrate the cell. After closing the pores, the cytostatic remains trapped in the cell in large quantities, multiplying its effect. In this paper, we present a feasibility study of electroporation application in irreversible mode without the use of cytostatics. Fresh porcine liver tissue was used to show that the constructed equipment was effective, thus opening the way for further investigations using reversible electroporation with the application of cytostatics, which would represent localized electrochemotherapy. We penetrated the virtual tumor area (liver metastases) with a specially designed needle with electrodes that generate an electric field and apply electroporation in the target tissue. We have shown that the constructed novel design single needle equipment for electroporation is effective on the experimental model of isolated porcine liver. Further steps in our study will be the testing of electrochemotherapy in an experimental animal model in vivo.

The purpose of this study is to evaluate the impact on the friction & wear of Clutch Plate material from different material composition. If we use the composite material the cost instead of the traditional material, weight can be reduced and the life of the brake material can be extended at low cost. In our research, material strength is experimentally investigated to predict compression, tensile & impact testing in both materials (E-glass & Jute fiber) to choose the better material of single plate clutch

The purpose of this paper is to present a taxonomy of single-subject experimental designs and discuss relevant examples that underscore the versatility and flexibility of this approach to clinical research. The proposed taxonomy serves as a heuristic model that may facilitate an understanding of single-subject experimental designs. Four general evaluation strategies employed in applied research—treatment-no treatment comparison, component assessment, treatment-treatment comparison, and successive level analysis—are discussed within this schema. Each of these evaluation strategies is related to commonly posed clinical research questions, and published examples of design options that address these questions are presented. Throughout the discussion basic considerations relating to appropriate design selection are reviewed.

Bayesian optimal experimental design is a sub-field of statistics focused on developing methods to make efficient use of experimental resources. Any potential design is evaluated in terms of a utility function, such as the (theoretically well-justified) expected information gain (EIG); unfortunately however, under most circumstances the EIG is intractable to evaluate. In this work we build off of successful variational approaches, which optimize a parameterized variational model with respect to bounds on the EIG. Past work focused on learning a new variational model from scratch for each new design considered. Here we present a novel neural architecture that allows experimenters to optimize a single variational model that can estimate the EIG for potentially infinitely many designs. To further improve computational efficiency, we also propose to train the variational model on a significantly cheaper-to-evaluate lower bound, and show empirically that the resulting model provides an excellent guide for more accurate, but expensive to evaluate bounds on the EIG. We demonstrate the effectiveness of our technique on generalized linear models, a class of statistical models that is widely used in the analysis of controlled experiments. Experiments show that our method is able to greatly improve accuracy over existing approximation strategies, and achieve these results with far better sample efficiency.

Based on the design idea of microcomputer protection, a set of three-section current protection, experimental device was designed by using MCU. Using MCU as the core control of experimental device, the circuits were designed, including the passive filter, current acquisition and signal output; real-ti me, current data were collected. When the current exceeded the setting value, the three-section protection had the single hop or linkage, thereby sending out a tripping signalto the simulated circuit breaker. By testing, the device could store data such as real-time acquisition of the current, alarm, tripping and closing in local database and cloud database, and realize remote monitoring and analysis of devices through cloud service., The device basically reached the goal of laboratory experiment teaching.

This study aimed to investigate the cerebral volumetric changes measurable by brain magnetic resonance imaging, as well as the different physiological variables documented through neurobiofeedback equipment, before and after 30 serial sessions of guided meditations in a sensory isolation tank. A pilot test was carried out, with a single subject, and it was concluded that serial sessions of meditations in flotation tanks changed the brain structure, triggered the relaxation response, generating greater physical, psychological and mental well-being for the research participant.

Single plate shear connections, often referred to as shear tabs, have gained considerable popularity in recent years due to their efficiency and ease of fabrication. Shear tab connections are primarily used to transfer beam end reactions to the supporting elements. The connection consists of a plate welded to a support at one edge and bolted to a beam web. Figure 1 shows typical applications of single plate shear connections. This paper presents the summary of a research project on the behavior and design of single plate shear connections. Based on experimental and analytical studies, a new design procedure is developed and presented.

Health problems are often idiosyncratic in nature and therefore require individualized diagnosis and treatment. In this paper, we show how single-case experimental designs (SCEDs) can meet the requirement to find and evaluate individually tailored treatments. We give a basic introduction to the methodology of SCEDs and provide an overview of the available design options. For each design, we show how an element of randomization can be incorporated to increase the internal and statistical conclusion validity and how the obtained data can be analyzed using visual tools, effect size measures, and randomization inference. We illustrate each design and data analysis technique using applied data sets from the healthcare literature.

Here we demonstrate a microfluidic-based analysis system based on single cell capture array, which can physically trap individual cell using micrometer-sized structures. A stable and in vivo-like microenvironment was built with the novel structure at the single-cell detection level. The microfluidic-based design can decouple single cells from fluid flow with the help of micropillars. The size and geometry of the cell jails are designed in order to discriminate between mother and daughter cells. It provides an experimental platform to efficiently monitor individual cell state for a long period of time. Furthermore, the parallel microfluidic array can ensure accuracy. In addition, finite element method (FEM) was employed to predict fluid transport properties for the most optimal fluid microenvironment.

Based on several structure optimization parameters for the description of the geometric features of a double oblique radial swirler, simplified structure design parameters and value interval were determined. And, the CFD analysis model of the swirler is established. Based on the approaches of multi-wheel space filling experimental designs, 17 design schemes were gained, then CFD numberical experiments on the cold single-phase flow field was developed to all design schemes. Finally, the optimal design scheme were found by numberical space filling experimental design method in a several turn.

Single-case experiments are increasingly popular in the behavioral sciences. Due to their flexibility, single- case designs can be customized to test a variety of experimental hypotheses. We were interested in using a single-case experimental approach to test whether pain thresholds can be influenced by Pavlovian classical conditioning. Following the example of earlier studies into this topic, we planned to measure whether participants would more frequently report specific electrocutaneous stimuli as painful when they were presented with specific vibrotactile stimuli that had previously been associated with painful electrocutaneous stimuli. First, we decided on a mean difference effect size measure derived from the Sensation and Pain Rating Scale ratings for the electrocutaneous stimuli provided by the participants. Next, we discussed several possible single-case designs and evaluated their benefits and shortcomings. Then, we ran pilot tests with a few participants based on the possible single-case designs. We also conducted a simulation study to estimate the power of a randomization test to test our hypothesis using different values for effect size, number of participants, and number of measurements. Finally, we decided on a sequentially replicated AB phase design with 30 participants based on the results from the pilot tests and the power study. We plan to implement this single-case design in a future experiment to test our hypothesis

Abstract This paper presents an attractive and a new voltage-mode quadrature oscillator using a single Fully Balanced-Voltage Differencing Buffer Amplifier (FB-VDBA) as the active element. The circuit structure is very simple, consisting of merely one FB-VDBA, one resistor and two capacitors. The circuit is implemented using the commonly available OPA860 which results in low output impedance and high current drive capability. The proposed circuit also has a suitable architecture for IC production. Experimental results which are matched well with the theoretical assumptions are given.

AbstractPerfectionism can maintain depression, anxiety and eating disorders, yet few studies have evaluated treatments for perfectionism. This study examined the effectiveness of individual cognitive behaviour therapy (CBT) in treating perfectionism in four adults with a diagnosis of either an anxiety disorder or depression. The study used an A-B single case experimental design series with follow-up, and a 3-week pre- and postbaseline phase. Treatment involved 8 sessions and a 2-week follow-up session. Visual inspection of data revealed downward trends in overall perfectionism and clinically significant decreases in perfectionism for two participants. No clinically significant reductions were observed in depressive or anxious symptomatology. CBT for perfectionism warrants further investigation.

Abstract Various alternatives for the design of clonal field trials in forestry were studied using simulated data to identify “optimal𠇍 or “near-optimal𠇍 scenarios for the estimation of genetic parameters. The simulated field site consisted of a rectangular grid on which 256 clones with 8 ramets each were installed. Estimates of genetic parameters were compared for (1) single-tree and four-tree row plots; (2) several experimental designs (completely randomized, randomized complete block, incomplete blocks of various sizes, and row–column); (3) no mortality versus 257% mortality; and (4) different patterns of environmental variability (only patches, only gradients, and both patches and gradients). Use of single-tree plots, on average, increased the correlations between true and predicted clonal values by 57% over four-tree row plots and increased genetic gain from selection. Starting with a parametric broad-sense heritability (H B 2) of 0.25 for a completely randomized design, the experimental designs resulting in the highest H B 2 were row–column designs for single-tree plots and incomplete blocks with 32 blocks per replication when four-tree row plots were used. These designs increased average heritability 107% and 147% over a randomized complete block design, respectively. The only effect of 257% mortality was an increase in the variability of some variance component estimates.

In this paper, the development of a fission-gas collecting and physical-analysis-enabling instrument was proposed for small-volume determination. Analysis specifications require a design capable of accurately and repeatably determining volumes in the range of 0.07–2.5 mL. This system relies on a series of gas expansions originating from a cylinder with known internal volume. The combined gas law is used to derive the unknown volumes from these expansions. Initial system designs included one of two known volumes, 11.85 ± 0.34 mL and 5.807 ± 0.078 mL, with a manifold volume of 32 mL. Results obtained from modeling this system’s operation showed that 0.07 mL can be determined with a relative expanded uncertainty greater than 300% (k = 2) for a single replicate, which was unacceptable for the proposed experimental design. Initial modeling showed that the volume connecting the known volume and rodlet, i.e., the manifold volume, and the sensitivity of the pressure sensor were key contributors to the expanded uncertainty of the measured rodlet volume. The system’s design limited the available options for pressure sensors, so emphasis was placed on the design of the manifold volume. The final system design reduced the manifold volume to 17 mL. These changes in design, combined with replicate analysis, were able to reduce the relative expanded uncertainty by ±12% (k = 2) for the 0.07 mL volume.

A novel cassava peeling machine that operates on the principle of the lathe was designed, fabricated and tested. The unique peeling principle uses a self-loading, self-adjusting single point cutting tool that can follow the contour of the cut cassava tuber sections or sized lots. The single point cutting tool allows the peeler to peel both freshly harvested cassava tubers and cassava tubers that are up to four days old effectively and efficiently. Test results show a peeling efficiency of over 70% for all categories of tubers and a minimal loss of flesh.

Demonstration of experimental control is considered a hallmark of high-quality single-case research design (SCRD). Studies that fail to demonstrate experimental control may not be published because researchers are unwilling to submit these papers for publication and journals are unlikely to publish negative results (i.e., the file drawer effect). SCRD studies comprise a large proportion of intervention research in special education. Consequently, the existing body of research, comprised mainly of studies that show experimental control, may artificially inflate efficacy of interventions. We discuss how experimental control evolved as the standard for high-quality SCRD; why, in the era of evidence-based practice, rigorous studies that fail to fully demonstrate experimental control are important to include in the body of published intervention research; the role of non-replication studies in discovering intervention boundaries; and considerations for researchers who wish to conduct and appraise studies that fail to yield full experimental control.
