Abstract Abstract 1567 The B-cell receptor (BCR) is critical to survival of normal B-cells, and regulates key aspects of cellular behavior. Of these, response to antigen determines pathways of normal B-cell maturation, including isotype switch events that occur by deletional class switch recombination (CSR), an irrevocable event, to yield IgG/A memory B-cells. Less frequently, CSR via a cryptic site generates IgD+ B-cells whereas IgM+IgD+ antigen experienced B-cells synthesize each isotype by an alternative transcript splicing mechanism. The role of the BCR in survival of malignant B-cells however is less well defined, in particular in response to antigen. Intriguingly, in Hairy cell leukemia (HCL), BCR assembly occurs with multiple surface immunoglobulin (sIg) isotypes (mult-HCL), many co-expressed on individual hairy cells (HCs) in an otherwise monoclonal tumor. Multiple isotypes appear to exclude deletional CSR events, and suggests a RNA processing mechanism of molecular assembly. This phenotype is rare even amongst malignant B-cells, and raises the question of the functional relevance of individual variant isotypes. It also potentially presents a model to dissect roles of multiple isotypes on single B-cells. To examine this, we investigated the BCR in CD19+CD11c+CD103+ mult-HCL cases (n=10), in which 2–4 differing sIg isotypes were present on most HCs, with single or, in 3 cases, dual sIgL expression. In all cases, IGHV genes were mutated, and confirmed monoclonality. Phenotype revealed 2 distinct subsets by sIg isotype co-expression, IgD+ve and IgD-ve. Using Ca2+ flux and ERK phosphorylation assays after cross-linking with specific anti-sIg antibodies, we observed a functional BCR in all mult-HCL examined, in both subsets (10/10 cases Ca2+, 6/6 cases ERK). However, striking differences emerged between the two subsets. In sIgD+ve mult-HCL, IgD mediated persistent Ca2+ flux, with flux also evident via >1 sIgH isotype. In marked contrast, in sIgD-ve mult-HCL Ca2+ flux was restricted to a single sIgH isotype, but not via IgM. Flux signals in this subset were transient. In most cases only a single sIgL transduced flux. We next evaluated BCR endocytosis after cross-linking individual isotypes and IgL. In 2 sIgD+ve cases, anti-IgD and anti-Igλ stimulation led to endocytosis of both sIgD and sIgλ, and in 1 case, where examined, anti-IgM stimulation endocytosed both sIgM and sIgλ. In 3 sIgD-ve cases, functional sIgH and sIgL induced endocytosis of the stimulated isotype, but again sIgM was dysfunctional, remaining immobilized on the cell surface. Ca2+ flux through endocytosed isotypes was correspondingly either significantly reduced or ablated in both subsets. In HCs, BCR endocytosis is clearly dependent on functional isotypes and IgL, and parallels events in normal B-cells. Lastly, we examined downstream effects of BCR signalling on cell viability, using soluble (sAb) and bound (bAb) anti-sIg antibodies. In a single IgD+ve mult-HCL case, both sAb and bAb anti-IgM yielded a significant level of apoptosis compared to control antibodies, whereas anti-IgD sAb resulted in no appreciable difference to level of spontaneous apoptosis, suggesting a disengagement of signals from this pathway. This disengagement was also observed in a separate HCL case expressing only IgD, and not in the mult-HCL cohort initially selected, where anti-IgD signals again did not increase levels of apoptosis. In IgD-ve mult-HCL (n=4), sAb and bAb specific cross-linking of IgG/A triggered significant apoptosis. These data demonstrate, for the first time, that mult-HCL retains a functional responsiveness via the BCR, suggesting an absence of anergic effects that may follow chronic antigen exposure in-vivo to self-antigen. Signals via sIgM/G/A isotypes, where functional, induce apoptosis in mult-HCL, whereas sIgD opposes such effects. Despite an apparently unique molecular mechanism of IgD expression in mult-HCL, this isotype appears to be hardwired in B-cells to mediate responses that differ from IgM. The persistent flux observed here indicates a more sustained and robust IgD signaling cascade, as also observed in B-cell models. These data reveal distinctive and opposing effects of individual isotypes on BCR mediated behavior in mult-HCL. While apoptotic responses appear to negate a role for antigen in tumor drive in-vivo, potential antigen engagement via IgD, if dominant leaves this question open. . Traditional methods for determining the solubility of oxygen in water are considered. Their advantages and disadvantages are shown. It is noted that especially great difficulties arise when measuring SO in a real chemical process, where the temperature and components of the system, as well as their ratios change over time. The new method for determining the solubility of oxygen based on thermodynamic calculation of the equilibrium phase and component composition of the oxygen-water system by minimizing the isobaric-isothermal potential of Gibbs using the software package "Selector" is proposed. The model is based on the exact equation of state for calculating the chemical potential of the vapor phase and the chemical potential of the liquid phase. The result is mathematical laws of dissolution of oxygen in water depending on a wide range of temperatures and pressures, which allows significantly facilitating and expanding the range of reliable control of the parameters of chemical-technological processes. The thermodynamic model allows calculations to be performed both in pure water and in acid and salt solutions, as well as taking into account the vapor phase and without it. The equation of Henry’s constant is given 2 1 X P A B T C T DT = ++ + 0.0181 exp( ln( ) ) for oxygen and air from temperatures in the range of 273...373 K and pressures up to 10 atm., which allows determining any intermediate values. To do this, it is necessary to substitute in the equation the current data of temperature and pressure, which are easily measurable. It is shown that within these ranges the deviation from the experimental values is minimal ∼1%, which allows using of the presented equations in technological calculations. The value of the temperature, at which the minimum solubility of oxygen is being observed, for pure oxygen is determined Tmin = 370.5K, for air Tmin = 370.3 K., the value of enthalpy changes sign.

Abstract The clue that surface Ig (sIg) is implicated in the pathogenesis of follicular lymphoma (FL) is that in FL the vast majority of the sIg variable regions are structurally modified by insertion of mannose residues into the antigen-binding sites. This is tumor-specific and reflects positive selection of sequence motifs for glycan addition introduced during somatic hypermutation. The termination at high mannoses is very unusual in cell surface molecules and it confers an ability to interact with local lectins expressed by macrophages. In this respect, the mannose cloaking of the sIg receptor resembles that of human immunodeficiency virus (HIV), which acquires a similar mannose coat to facilitate binding to macrophages. The lectin DC-SIGN is upregulated in FL, likely via local IL-4, and is the strong lectin candidate. To address the question of the functional implications of interaction between DC-SIGN and sIgM we used a FL-derived cell line (WSU-FSCCL) which expresses sIg-mannoses. We found that two recombinant derivatives of DC-SIGN (DC-SIGN-Fc or DC-SIGN-HA) bind to the sIgM but not to a cell line which express sIgM without mannose insertion. Although the mannoses are an integral part of the sIgM, and anti-IgM induces a Ca2+ flux, DC-SIGN binds but does not mimic anti-IgM. In the WSU-FSCCL cell line it remains on the surface IgM without inducing a Ca2+ response and without mediating endocytosis of the sIgM. However, DC-SIGN does act on the sIgM as revealed by the effects of pre-exposure of the cells to either DC-SIGN derivative, which, although there is no blocking of access of anti-IgM, alters the ability of the cell to respond to stimulation. Pre-exposure appears to partially paralyze the subsequent sIgM-induced Ca2+ flux indicating a lectin-mediated modification of sIgM function. On engagement by anti-IgM, sIg undergoes conventional endocytosis and this was confirmed in WSU-FSCCL. Inhibitors which block signaling did not affect endocytosis, indicating bifurcation of signaling and endocytic pathways. Knowing that DC-SIGN derivatives, again in contrast to anti-IgM, did not induce endocytosis of sIgM, we then asked if the paralysis of the sIgM-mediated Ca2+ flux by pre-exposure to lectin affected anti-IgM-induced endocytosis, and found that it did not. This confirms the separation of signaling and endocytosis and indicates that whatever change is occurring in sIgM due to lectin exposure does not remove the sIgM from the endocytic machinery. We then investigated primary FL cases, using a splenic FL and 2 lymph node FLs, all carrying N-glycosylation motifs and able to bind DC-SIGN. We focused on the DC-SIGN-HA derivative, which gives no detectable Ca2+ flux signal by itself. Binding of the lectin again paralyzed subsequent anti-IgM-induced Ca2+ flux in all cases (Fig.1A), confirming the results with the cell line. Further investigation revealed that the lectin prevents early events after antigen binding to the sIg receptor reducing SYK phosphorylation and leading to a reduced Ca2+ mobilization (Fig.1B). In conclusion, modulation of the B-cell receptor by mannose addition to the antigen-binding site allows lectin access from innate microenvironmental cells. The effect of this is to provide a low level/null signal without loss by endocytosis. The novel finding is that this interaction then lowers the function of the B-cell receptor and perhaps blocks potential interference by antigen. There may be a parallelism with reports of modification of T-cell receptor function by galectins, with both pointing to the role of post-translational modification adding another layer of control on the operation of the major immune receptors. In the case of FL, this has been exploited to maintain tumor cells in the hostile environment of the germinal center. Abstract Although most human B cells express receptors for Epstein Barr virus (EBV), few (usually less than 1%) are readily transformed into B lymphoblastoid cell lines after exposure to EBV. Transformable cells previously have been found to be mostly resting B lymphocytes. We recently developed a limiting dilution culture system which permits the growth of EBV-transformed B lymphocytes with high efficiency. Because in this system up to over 30% of peripheral blood- or tonsil-derived B cells respond to EBV, we re-examined the properties of EBV-transformable cells. Frequencies of transformable lymphocytes were determined by Poisson analysis. EBV-susceptible B cells committed to IgM, IgG, or IgA secretion were found to occur in the range of 3 to 27, 0.1 to 6, and 0.1 to 5 per 100 B cells, respectively. Under our culture conditions, a major proportion of the IgM-committed cells derived from large lymphocytes which appeared to have entered the cell cycle. This population contains most of the EBV-responsive cells detected and, therefore, most of the additional cells responding in our culture system. In contrast, precursors of IgG- or IgA-producing lymphoblast lines were small cells with DNA contents typical for the G0/G1 phases of the cell cycle. Anti-immunoglobulin antibodies were used in panning experiments to separate B cell subpopulations which expressed different immunoglobulin isotypes on their surface. In limiting dilution cultures of these purified B lymphocytes subsets, it was found that virtually all precursors of IgM-producing cell lines expressed surface IgM (sIgM) before their infection and transformation by EBV. The "cloning efficiency" of positively selected, large sIgM+ cells approached 100%. In contrast, sIgG or sIgA were found only on cells committed to the production of IgG or IgA, respectively. The expression of sIgD was examined by using sequential panning procedures. Virtually all IgM-committed lymphocytes and a subset of cells committed to IgA secretion were found among the sIgD+ cells. The majority of cells committed to IgA production and all IgG-committed cells were found in the sIgD- B cell population. Our findings indicate that the EBV-susceptible B cell subset of normal lymphocytes is heterogeneous with respect to cell size, cell cycle, sIg determinants, and efficiency of transformation. On the basis of our findings and previous reports, we propose a model in which transformability is a B cell-inherent property. Factors unrelated to the virus but present in our culture system appear responsible for the enhanced vulnerability to viral transformation in some cells which entered into the cell cycle.(ABSTRACT TRUNCATED AT 400 WORDS)

Terson's sign (TS) is classically defined as vitreous hemorrhage associated with subarachnoid hemorrhage of aneurysmal origin, being an important predictor of severity, indicating greater morbidity and mortality when compared to patients without the sign. The objective of this study is to review the relationship of Terson syndrome/Terson sign with the prognosis of aneurysmal subarachnoid hemorrhage. A search for original articles, research and case reports was performed on the PubMed, Scielo, Cochrane and ScienceDirect platform, with the following descriptors: Terson sign and subarachnoid hemorrhage. Retrospective, prospective articles and case reports published in the last 5 years and which were in accordance with the established objective and inclusion criteria were selected. Ten (10) articles were selected, in which the available results show an unfavorable prognostic relationship of TS and subarachnoid hemorrhage, because these patients had a worse clinical status assessed on the Glasgow scales ≤ 8, Hunt & Hess > III, Fisher > 3, in addition to intracranial hypertension and location of the aneurysm in the anterior communicating artery complex. The early recognition of this condition described by Albert Terson in 1900 brought an important contribution to neurosurgery, being recognized until nowadays.

Minority languages are often given less importance than national languages, especially when the languages differ in their modalities. In addition, the hearing majority often do not recognise sign languages, leading to prejudice and discrimination against them. Apart from the theoretical relevance of the study of sign languages for linguistic research, linguistic studies have proved crucial in reducing the social and cultural isolation of Deaf communities. At the same time, the specific sociolinguistics of sign languages must be considered when conducting such linguistic research. This paper presents relevant background information that needs to be considered when conducting research on the grammar of Slovenian Sign Language (SZJ), as well as the research topics in SZJ linguistics that have been addressed in recent decades.

The ability of Listeria monocytogenes to grow at refrigeration temperatures is critical for transmission of this foodborne pathogen. We evaluated the contributions of different transcriptional regulators and two-component regulatory systems to L. monocytogenes cold adaptation and cold growth. L. monocytogenes parent strain 10403S and selected isogenic null mutants in genes encoding four alternative σ factors (sigB, sigH, sigC, and sigL), two regulators of σB (rsbT and rsbV), two negative regulators (ctsR and hrcA), and 15 two-component response regulators were grown in brain heart infusion broth at 4°C with (i) a high-concentration starting inoculum (108 CFU/ml), (ii) a low-concentration starting inoculum (102 CFU/ml), and (iii) a high-concentration starting inoculum of cold-adapted cells. With a starting inoculum of 108 CFU/ml, null mutants in genes encoding selected alternative σ factors (ΔsigH, ΔsigC, and ΔsigL), a negative regulator (ΔctsR), regulators of σB (ΔrsbT and ΔrsbV), and selected two-component response regulators (ΔlisR, Δlmo1172, and Δlmo1060) had significantly reduced growth (P < 0.05) compared with the parent strain after 12 days at 4°C. The growth defect for ΔsigL was limited and was not confirmed by optical density (OD600) measurement data. With a starting inoculum of 102 CFU/ml and after monitoring growth at 4°C over 84 days, only the ΔctsR strain had a consistent but limited growth defect; the other mutant strains had either no growth defects or limited growth defects apparent at only one or two of the nine sampling points evaluated during the 84-day growth period (ΔsigB, ΔsigC, and Δlmo1172). With a 108 CFU/ml starting inoculum of cold-adapted cells, none of the mutant strains that had a growth defect when inoculation was performed with cells pregrown at 37°C had reduced growth as compared with the parent strain after 12 days at 4°C, suggesting a specific defect in the ability of these mutant strains to adapt to 4°C after growth at 37°C. Our data indicate (i) selected σ factors and two-component regulators may contribute to cold adaptation even though two-component regulatory systems, alternative σ factors, and the negative regulators CtsR and HrcA appear to have limited contributions to L. monocytogenes growth at 4°C in rich media, and (ii) inoculum concentration and pregrowth conditions affect the L. monocytogenes cold-growth phenotype.

<p>As a popular research direction in the field of intelligent transportation, various scholars have widely concerned themselves with traffic sign detection However, there are still some key issues that need to be further solved in order to thoroughly apply related technologies to real scenarios, such as the feature extraction scheme of traffic sign images, and the optimal selection of detection methods. For the purpose of overcoming these difficulties. This paper proposes a YOLO-based traffic sign detection framework. Firstly, a lightweight convolution attention mechanism is embedded into the backbone network to obtain the information of space and channel; Secondly, the multi-scale awareness module is used to replace large convolution with 3&times;3 convolution superposition to improve the receptive field area of the object in the model and enhance the feature fusion performance of the model; Finally, CIoU is used as the loss function of the bounding box to locate the experimental object with high precision. The experimental results show that on the CCTSDB data set, the MAP of this method reaches 91.0%, which is 3.5% higher than the original YOLOv5. Compared with other mainstream object detection algorithms, it has a certain degree of improvement, which proves the effectiveness of this method.</p> <p>&nbsp;</p>

Introduction: in this systematic literature review, the use of artificial intelligence in sign language translation for people with hearing and speech loss was analyzed. This review aims to identify the results of the application of artificial intelligence to sign language translation. Method: 462 articles, original and conference papers in SCOPUS, until June 2023, and relying on a selection process based on the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) statement, of which 26 studies met exclusion and inclusion criteria. Convolutional Neural Network (CNN) was the most widely implemented machine learning technique in the selected studies. Results: Many sign language systems were tested with various artificial intelligence algorithms and datasets on different continents to create new models and improve sign language translation accuracy. An increasing use of neural networks to achieve better sign language translation efficiency was identified, achieving results ranging from 90 % to 100 % accuracy. Conclusions: The application of artificial intelligence has greatly excelled in the field of Computer Science and has significantly improved the accuracy of sign language translation which has led to lower communication barriers between natural persons and persons with communication disabilities.

Traffic sign recognition is a key module of autonomous cars and driver assistance systems. Traffic sign detection accuracy and inference time are the two most important parameters. Current methods for traffic sign recognition are very accurate; however, they do not meet the requirement for real-time detection. While some are fast enough for real-time traffic sign detection, they fall short in accuracy. This paper proposes an accuracy improvement in the YOLOv3 network, which is a very fast detection framework. The proposed method contributes to the accurate detection of a small-sized traffic sign in terms of image size and helps to reduce false positives and miss rates. In addition, we propose an anchor frame selection algorithm that helps in achieving the optimal size and scale of the anchor frame. Therefore, the proposed method supports the detection of a small traffic sign with real-time detection. This ultimately helps to achieve an optimal balance between accuracy and inference time. The proposed network is evaluated on two publicly available datasets, namely the German Traffic Sign Detection Benchmark (GTSDB) and the Swedish Traffic Sign dataset (STS), and its performance showed that the proposed approach achieves a decent balance between mAP and inference time.

103 Background: Accumulating evidence pointed to the impact of the intestinal microbiota on ICI outcomes across various cancers. Although specific gut microbial species have been associated with beneficial responses ( i.e. Akkermansia muciniphila (Akk)), no consensus exists on a gut fingerprint predicting immunoresistance to clinical routine use. Methods: NCT04567446 provided whole genome sequencing (WGS) of longitudinal fecal samples from patients (pts) with advanced non-small cell lung cancer (NSCLC) during ICI (alone or with chemotherapy) in France and Canada. Topological Pearson networks clustered into species interacting groups (SIG) correlating with overall survival (OS; OS<12=NR; OS>12=R). Forty harmful (SIG1) and thirty-four beneficial (SIG2) WGS species were associated with NR and R to ICI. A monodimensional score (TOPOSCORE) based on SIG1/SIG2 ratio combined with Akk relative abundance was calculated and compared to machine-learning (ML) algorithms. Multivariate Cox analysis (MVA) adjusted for established risk factors (ATB, gender, age, ECOG, PD-L1, LIPI score). Intraindividual dynamics of the TOPOSCORE was evaluated in pts with at least two fecal samples. Three independent cohorts of NSCLC and genitourinary (GU) cancers pts validated the data. Results: In n=245 and n=148 NSCLC pts, we could classify pts into dysbiotic (SIG1+,33%) and eubiotic (SIG2+, 67%), using the TOPOSCORE. Pts falling within the SIG2+ exhibited a significantly prolonged OS than pts falling into SIG1+ (HR: (95% CI), 0.50 (0.36-0.71), p<0.0001). TOPOSCORE also predicted OS in 277 ICI-treated NSCLC and GU pts and compared to the state-of-the-art ML algorithms, held the highest percentage of correct predictions (63%). At MVA, TOPOSCORE was independently associated with OS (HR: 0.56 (0.39-0.81), p=0.002). Analyzing the intraindividual dynamics of the TOPOSCORE (n=67), we found that 74% of SIG2+ and 68% of SIG1+ individuals remained in their initial classification during ICI treatment. We finally scaled the calculation of the TOPOSCORE down to 24WGS (instead 75WGS) and set up a qPCR-based friendly-user test capable of accurately identifying the fecal presence of the bacteria of interest within 48 hrs. We confirmed (n=323) that OS was superior in those pts harboring a 24-bacteria-qPCR-based TOPOSCORE falling within the SIG2+ category (HR: 0.65 (0.48 to 0.87), p=0.0005). Conclusions: TOPOSCORE represents a robust biomarker predicting and following the dynamic of the immunoresistance to ICI across cancers on an individual basis. By converting the WGS TOPOSCORE to a qPCR-based test with a rapid turnaround time, it will be possible to adopt this score in routine clinical practice to improve pts stratification and ICI success rates guiding the selection of dysbiotic pts amenable to microbiota-centered interventions and eubiotic fecal microbiota transplantation donors. This study use descriptive qualitative method. The data were analyzed based on theory proposed by Barthes (1964) about the signifier and signified and about the meaning. The researcher used supporting theory by Wierzbicka (1996) about six color terms. The result of this study showed the verbal sign in form of written text and for the non-verbal sign showed in form of image. Much recent research has found that states generally comply with the treaties they sign. The implications of this finding, however, are unclear: do states comply because the legal commitment compels them to do so, or because of the conditions that led them to sign? Drawing from previous research in this Review on Article VIII of the IMF Treaty (Simmons 2000a), I examine the problem of selection bias in the study of treaty compliance. To understand how and whether international legal commitments affect state behavior, one must control for all sources of selection into the treaty—including those that are not directly observable. I develop a statistical method that controls for such sources of selection and find considerable evidence that the unobservable conditions that lead states to make the legal commitment to Article VIII have a notable impact on their propensity to engage in compliant behavior. The results suggest that the international legal commitment has little constraining power independent of the factors that lead states to sign.

In a previous article I have argued that the contribution of early Muslim scholars to Semiotics needs to be unearthed and highlighted. Semiotics is broadly defined as the study of signs or sign-use behaviour leading to communication and understanding between humans in society. Semiotics emerged in the early twentieth century as distinct from mainstream philosophy or philosophy of language. As an institutionalised academic discipline, semiotics has acknowledged the implicit contributions of earlier scholars from antiquity, the middle ages and early modernity. However, to date the contribution of Muslim scholars is conspicuously absent. Hence, in this article I intend to recapitulate perspectives from selected Muslim scholars in order to obtain a more balanced notion of sign-function and its potential implications. This is where the unique contributions of al-Ghazali (1058-1111) and Ibn Barrajan (n.d.-1141) come in, whose perspectives are reflected in key modern approaches to sign models and their interpretational tendencies. The article suggests that there is a need to explore the legacy of Muslim scholars in the field of semiotics in a much more systematic and comprehensive way, given that sign-based thinking (or the ayah doctrine) is a major theme in the Quran.

The aim of this theoretical article is to increase the cognitive value of the subject of Baby Sign Language, in particular their positive impact and significance for the development of a hearing child. Addressing this problem seems important because of constant interest in sign language for children, both among parents and scientists. Equally important is the argument of the lack of a sufficient number of Polish studies in this area. The article consists of a short introduction and four thematically and logically arranged parts. The first contains a synthetic description of the process of language acquisition and development as well as the communication of a young child. The second part presents the history of the Baby Sign Language, its basic assumptions and principles. The third part discusses the importance and benefits of using the Baby Sign Language in the development of a young child. The study ends with a short summary with final conclusions, which show, based on the presented empirical material, that the Baby Sign Language has a positive significance for the child's development in almost all its spheres, i.e. e.g. in the linguistic, cognitive, emotional and motor spheres.

e20517 Background: Changes in gut microbiota may affect benefit from immunotherapy (IO). Gut dysbiosis can be identified by a fecal metagenomic signature (TOPOSCORE SIG1+ vs SIG2+), and/or by low levels of sMAdCAM-1. Recent studies showed that peripheral thyroid hormones conversion (fT3/fT4 ratio) has a strong prognostic role in various cancers, with low fT3/fT4 ratio predicting worse outcomes. Correlation between gut dysbiosis and metabolic stress, such as altered fT3/fT4 conversion, has not been investigated yet. Methods: A total of 77 lung cancer pts treated at Gustave Roussy Cancer Center with IO-based treatment with available measurement of baseline fT3/fT4 ratio, metagenomic data defined by TOPOSCORE category SIG1+ vs SIG2+, and/or level of sMAdCAM1 (low vs high according to the median) from the ongoing ONCOBIOTICS multicentric prospective study (NCT04567446) were selected. fT3/fT4 low vs high groups were defined adopting the median value as cut-off. Correlation between the fT3/fT4 ratio categories with TOPOSCORE and sMAdCAM1 were tested by means of a 2-sided chi-square test. Results: Baseline fT3/fT4 ratio and TOPOSCORE and/or sMAdCAM-1 data were available for 62 and 57 patients, respectively. A significant correlation was observed between fT3/fT4 ratio and TOPOSCORE category, with a higher proportion of SIG1+ pts (64.5%) vs SIG2+ (35.5%) in fT3/fT4 low group, and higher rate of SIG2+ (61.3%) vs SIG1+ (38.7%) pts in fT3/fT4 high group (p = .042). Similarly, a significant association was observed between low levels of sMAdCAM-1 and the fT3/fT4 low group (p = .047). In a independent dataset of 42 NSCLC treated with 2nd line IO, pts with low fT3/fT4 ratio had a mPFS of 1.7 mos and a mOS of 2.4 mos compared to 6.8 and 18.7 mos respectively for fT3/fT4 high pts (PFS HR = 0.56, 95%CI 0.30-1.05, p = .040; OS HR = 0.50, 95%CI 0.26-0.99, p = .030). Conclusions: Our data confirm the prognostic value of fT3/fT4 in progressing metastatic NSCLC and its correlation with gut dysbiosis markers. These findings open the way to further research for exploring specific therapeutic applications, such as tailored nutritional support and microbiota-driven interventions. This paper presents a Sign Language Recognition System (SLRS) designed to facilitate seamless communication between individuals proficient in sign language and those who may not share this proficiency. The system employs a multi-faceted approach, integrating computer vision, machine learning, and signal processing techniques to accurately interpret and recognize sign language gestures. The methodology involves data collection from diverse signing styles, preprocessing to enhance data quality, feature extraction capturing key aspects of sign language expressions, and the selection and training of machine learning models. The system aims to represent sign language gestures effectively, providing a foundation for real-time recognition. Integration into hardware or software platforms ensures practical applications in various settings, including education, employment, and public spaces.

In this paper, 20 guided traffic signs mostly displayed around Malacca area were selected as project databased. Early hypothesis was made as the error for each usable image will increased as more interference introduced to the original image used. Three types of conditions which are hidden region, image brightness and image rotation were selected as an experiment to analyze the performance of each sign used. Each condition will perform a specific error to generate their mean value and in the same, image recognition will take place in the matchup process. By focusing on the result, it produces hidden region critically ascending mean error value at 62.5% = 0.07 and has average value at others points. For image brightness effect, it shows a higher mean error value collected at less brightness points and non-stable pattern at 10% to 60% brightness. As for rotation upshot, the values show a critically ascending for error value at 22.5% and slightly increase at 2% to 5% rotation point. For the recognition process, at 6.25% hidden region, almost 70% of images are correctly matched to its own classes while at 62.5% hidden region only 40% of images are correctly matched to its own classes and leaving 2 images to outperform. For -40% brightness, 45% of images are correctly matched to its own classes while at 60% brightness 65% of images are correctly matched to its own classes and leaving 1 image to outperform. Lastly, at 2.5 degree rotation, 85% of images are correctly matched to its own classes while at 25° rotation, 45% of images are correctly matched to its own classes and leaving 2 images to outperform. Finally, the error forms will affect the final output response of the detected traffic signs used.

In survival analysis, when a subset of subjects has extremely long survival, the two-part cure rate model has been commonly adopted. In the two-part model, the first part is for a binary response and describes the probability of cure. The second part is for a survival response and describes the probability of survival. Despite their intuitive interconnections, most of the existing works estimate the two parts without any constraint. The existing works on proportionality promote similarity in magnitudes (i.e. quantitative similarity) and can be too restrictive. In this study, for the two-part cure rate model, we propose imposing a sign-based penalty to promote similarity in signs (i.e. qualitative similarity). The proposed strategy can be more informative than those that neglect the two-part interconnections and be less restrictive than the existing proportionality works. Penalty is also imposed to select relevant variables and accommodate high-dimensional data. Numerical studies, including simulation and two data analyses, demonstrate the advantageous performance of the proposed approach.

Further, segmentation and classification are applied to datasets. This paper provides guidelines for the creation and selection of datasets.

The binding of human immunodeficiency virus (HIV) to C-type lectin receptors may result in either enhanced trans-infection of T-cells or virus degradation. We have investigated the efficacy of HIV-1 utilization of DC-SIGN, a C-type lectin receptor, in the setting of intrauterine or intrapartum mother-to-child transmission (MTCT). Viruses isolated from HIV-1-infected mothers at delivery and from their vertically infected children both shortly after birth and later during the progression of the disease were analysed for their use of DC-SIGN, binding and ability to trans-infect. DC-SIGN use of a child’s earlier virus isolate tended to be reduced as compared with that of the corresponding maternal isolate. Furthermore, the children’s later isolate displayed enhanced DC-SIGN utilization compared with that of the corresponding earlier virus. These results were also supported in head-to-head competition assays and suggest that HIV-1 variants displaying efficient DC-SIGN use are not selected for during intrauterine or intrapartum MTCT. However, viruses with increased DC-SIGN use may evolve later in paediatric HIV-1 infections.

The efficacy of various diagnostic imaging methods in determining the morphofunctional features of the affected kidney, the characteristics of kidney stones and its complications, as well as the sensitivity and specificity of conventional imaging methods (ultrasonography, simple KUB radiography, intravenous urography), functional (isotopic renography and dynamic renal scintigraphy) and complex (computed tomography and densitometric tomography associated with infrared spectroscopy) were assessed. Comparison of imaging data with the results of macroscopic evaluation of affected kidneys by stones during surgery allowed to specify the diagnostic contribution of each method with determining its influence on the selection of any applied treatment method. The article assesses an analysis of the main aspects related to diagnosis, the selection criteria for medical-surgical treatment, but especially the surgical techniques, selected from case to case, individualized with the introduction and implementation of numerous diagnostic procedures, treatments, prophylaxis, and metaphylaxis, during the years 2010–2021. The current study of kidney stone disease is complex scientific research, with a multilateral character, which is determined by the multitude of the examined aspects.

Abstract The extent to which genetic background can influence allelic fitness is poorly understood, despite having important evolutionary consequences. Using experimental populations of Arabidopsis thaliana and map-based population genetic data, we examined a multigeneration response to selection in populations with differentiated genetic backgrounds. Replicated experimental populations of A. thaliana with genetic backgrounds derived from ecotypes Landsberg and Niederzenz were subjected to strong viability and fertility selection by growing individuals from each population at high density for three generations in a growth chamber. Patterns of genome-wide selection were evaluated by examining deviations from expected frequencies of mapped molecular markers. Estimates of selection coefficients for individual genomic regions ranged from near 0 to 0.685. Genomic regions demonstrating the strongest response to selection most often were selected similarly in both genetic backgrounds. The selection response of several weakly selected regions, however, appeared to be sensitive to genetic background, but only one region showed evidence of positive selection in one background and negative selection in another. These results are most consistent with models of adaptive evolution in which allelic fitnesses are not strongly influenced by genetic background and only infrequently change in sign due to variation at other loci.

The aim of this study is to provide a detailed account for the phenomenon of movement epenthesis in Italian Sign Language (LIS). LIS displays at least two cases of epenthesis of movement, one affecting signs that involve contact with the body, the other affecting signs that do not (i.e. signs articulated in neutral space). The two cases of epenthesis of movement receive a unified analysis, once the mechanism of selection of the plane of articulation is spelled out. The general phenomenon of movement epenthesis is captured by a formal approach within a constraint-based framework, such as the one developed first for American Sign Language (ASL) in Brentari (1998). Cases of movement epenthesis in ASL will be discussed and compared to cases of LIS epenthesis

SummaryA general representation of multilocus selection is extended to allow recombination to depend on genotype. The equations simplify if modifier alleles have small effects on recombination. The evolution of such modifiers only depends on how they alter recombination between the selected loci, and does not involve dominance in modifier effects. The net selection on modifiers can be found explicitly if epistasis is weak relative to recombination. This analysis shows that recombination can be favoured in two ways: because it impedes the response to epistasis which fluctuates in sign, or because it facilitates the response to directional selection. The first mechanism is implausible, because epistasis must change sign over periods of a few generations: faster or slower fluctuations favour reduced recombination. The second mechanism requires weak negative epistasis between favourable alleles, which may either be increasing, or held in check by mutation. The selection (si) on recombination modifiers depends on the reduction in additive variance of log (fitness) due to linkage disequilibria (υ1 < 0), and on non-additive variance in log (fitness) (V′2, V′3,.. epistasis between 2, 3.. loci). For unlinked loci and pairwise epistasis, si = − (υ1 + 4V2/3)δr, where δr is the average increase in recombination caused by the modifier. The approximations are checked against exact calculations for three loci, and against Charlesworth's analyses of mutation/selection balance (1990), and directional selection (1993). The analysis demonstrates a general relation between selection on recombination and observable components of fitness variation, which is open to experimental test.

With a historical context as a foundation, the current trends, practices, and perspectives regarding the manual component of educating deaf children is examined, including Manually Coded English systems and American Sign Language. As decisions are considered regarding various approaches to sign communication, it is necessary to investigate issues that support and also question the appropriateness of any given language/system. In addition to the sign language/systems, an equally important aspect is the instructional strategy that supports sign usage, such as Total Communication, Simultaneous Communication, and Bilingual Education. Issues affecting the selection and use of sign language/systems conclude this article.

Objectives: This study compared feature selection by machine learning or expert recommendation in the performance of classification models for in-hospital mortality among patients with acute coronary syndrome (ACS) who underwent percutaneous coronary intervention (PCI).Methods: A dataset of 1,123 patients with ACS who underwent PCI was analyzed. After assigning 80% of instances to the training set through random splitting, we performed feature scaling and resampling with the synthetic minority over-sampling technique and Tomek link method. We compared two feature selection methods: recursive feature elimination with cross-validation (RFECV) and selection by interventional cardiologists. We used five simple models: support vector machine (SVM), random forest, decision tree, logistic regression, and artificial neural network. The performance metrics were accuracy, recall, and the false-negative rate, measured with 10-fold cross-validation in the training set and validated in the test set.Results: Patients’ mean age was 66.22 ± 12.88 years, and 33.63% had ST-elevation ACS. Fifteen of 34 features were selected as important with the RFECV method, while the experts chose 11 features. All models with feature selection by RFECV had higher accuracy than the models with expert-chosen features. In the training set, the random forest model had the highest accuracy (0.96 ± 0.01) and recall (0.97 ± 0.02). After validation in the test set, the SVM model displayed the highest accuracy (0.81) and a recall of 0.61.Conclusions: Models with feature selection by RFECV had higher accuracy than those with feature selection by experts in identifying patients with ACS at high risk for in-hospital mortality.

Abstract This article examines the social and interactional foundations of sign-creation among DeafBlind people in Seattle, Washington. Linguists studying signed languages have proposed models of sign-creation that involve the selection of an iconic gestural representation of the referent which is subjected to grammatical constraints and is thereby incorporated into the linguistic system. Drawing on 18 months of ethnographic fieldwork and more than 190 hours of video recordings of interaction and language use, I argue that a key interactional mechanism driving processes of sign-creation among DeafBlind people in Seattle is deictic integration. Deictic integration restricts the range of contextual values that the grammar can retrieve by coordinating systems of reference with patterns in activity. This process brings language into alignment with the world as it is perceived by the users of that language, making a range of potentially iconic relations available for selection in the creation of new signs.
