Texto completo da fonteAdvanced age is accompanied by a decline of immune functions, which may play a role in low efficacy of vaccinations in elderly people. However, it is yet unclear at which level age-intrinsic mechanisms and external factors (namely the co-infections with persistent viruses) cooperate, contributing to immunosenescence. The aim of this work was to unravel the age-dependent interplay between the host immune system and 3 main herpesviruses: HSV-1, CMV ad EBV. To reach this scope, I focused on the effect of persistent infections on immunosenescence and of immunosenescence on the immune responses to infections. Three main questions were addressed: 1) May persistent viral infections influence the T-cell compartment and immune responsiveness in older adults? 2) How age affects the memory responses to herpesviruses? 3) How age affects the naïve responses to primary infections (SARS-CoV-2)? To this aim, I first assessed immunological parameters, related to CD8+ and CD4+ T-cell responsiveness, according to the serological status for common latent herpesviruses in two independent cohorts: 1. healthy individuals aged 19y to 95y and 2. individuals above 70y old enrolled in a primo-vaccination clinical trial (n= 137). Results show a prevalent effect of age and CMV infection on CD8+ and CD4+ naive T cells respectively. CMV seropositivity was associated with blunted CD4+ T-cell and antibody responses to primary vaccination against tick-borne encephalitis virus. The concomitant presence of different persistent infections is also responsible of the accumulation (inflation) of late-differentiated memory T cells in older subjects. Then, I measured HSV-, CMV- and EBV-specific cellular responses in middle-aged and elderly adults from Cohort 1 dissecting the secretory capacity of distinct memory subsets. Notably, an age-related increase of late-differentiated, HSV-specific memory CD8+ T cells is observed, suggesting that HSV-1 participate to the memory inflation of the CD8 compartment. In addition, a concomitant decrease of secretory capacity upon general TCR stimulation is noticed in early/intermediate differentiated memory CD8+ T cells. Finally, to confirm whether primary CD8+ T-cell responses are defective in advanced age and assess potential consequences, an in vitro approach to prime SARS-CoV-2-specific naive CD8+ T cells from healthy, unexposed donors of different age groups was exploited. Being an emerging infection, SARS-CoV-2 is the perfect model to study primary responses in unexposed subjects. Compared to younger adults, older individuals display a poor SARS-CoV-2-specific T-cell priming capacity in terms of both magnitude and quality of the response. In addition, older subjects recognize a lower number of epitopes. Collectively, these data implicate that immune ageing is associated with altered primary responses. This implies that elderly subjects present with a low SARS-CoV-2-specific cellular immunity. As virus-specific CD8+ T-cells have shown to be protective toward critical COVID-19 manifestations, an age-related suboptimal cellular immunity may contribute to the age-pattern of the disease. Moreover, the concomitant infection with CMV negatively affects, in elderly individuals, CD4-mediated response and thus both cellular and humoral immunity. This may further explain the reduced responses to emerging infections and de-novo vaccination with advanced age. Overall, recall T-cell responses are less affected by age. Nonetheless, herpesviruses cause an “inflation” of late-differentiated memory cells that could negatively impact on immunological memory towards other antigen specificities (e.g. previously administered vaccines) and is a sign of frequent viral reactivation in life. Altogether, these data provide insights on the changes in adaptive immunity over time and the associated decline in vaccine efficacy with ageing. This knowledge is important for the management of infectious diseases in elderly populations.

Carbazole based polyazomethines have been synthesized via polycondensation reactions between di-substituted carbazoles, bearing amino and formyl functionsin positions 3,6 or 2,7. Optical and electronical properties of such polymers have been studieddepending of the linkage position. A comonomer EDOT has then been integrated into the polymer chain, and impact of such insertion has been studied. Squaric and croconic acid base polymers have also been synthesized. By varying polymerization conditions, optoelectronic properties have been tuned, leading to the formation of polymers exhibiting a white emission. These polymers have then been integrated into OLED, as the active layer. Finally, more original polymers have been synthesized, using more original reactions or monomers such as by forming in situ benzobisthiazole. Other polymers integrating more originals monomers, such a tetrazine or divanillin, have been synthesized. Optoelectronic properties of such materials have been studied for the purpose of their integration into devices

The Presence and Use of the Native American and African American Oral Trickster Traditions in Zitkala-Sa's Old Indian Legends and American Indian Stories and Charles Chesnutt's The Conjure Woman My dissertation examines early Native American and African American oral trickster tales and shows how the pioneering authors Zitkala-Sa (Lakota) and Charles W. Chesnutt (African American) drew on them to provide the basis for a written literature that critiqued the political and social oppression their peoples were experiencing. The dissertation comprises 5 chapters. Chapter 1 defines the meaning and role of the oral trickster figure in Native American and African American folklore. It also explains how my participation in the Native American and African American communities as a long-time storyteller and as a trained academic combine to allow me to discern the hidden messages contained in Native American and African American oral and written trickster literature. Chapter 2 pinpoints what is distinctive about the Native American oral tradition, provides examples of trickster tales, explains their meaning, purpose, and cultural grounding, and discusses the challenges of translating the oral tradition into print. The chapter also includes an analysis of Jane Schoolcraft's short story "Mishosha" (1827). Chapter 3 focuses on Zitkala-Sa's Old Indian Legends (1901) and American Indian Stories (1921). In the legends and stories, Zitkala-Sa is able to preserve much of the mystical, magical, supernatural, and mythical quality of the original oral trickster tradition. She also uses the oral trickster tradition to describe and critique her particular nineteenth-century situation, the larger historical, cultural, and political context of the Sioux Nation, and Native American oppression under the United States government. Chapter 4 examines the African American oral tradition, provides examples of African and African American trickster tales, and explains their meaning, purpose, and cultural grounding. The chapter ends with close readings of the trickster tale elements embedded in William Wells Brown's Clotel; or, The President's Daughter (1853), Harriett Jacobs's Incidents in the Life of a Slave Girl (1861), and Martin R. Delany's Blake, or the Huts of America (serialized 1859 - 1862). Chapter 5 shows how Charles Chesnutt's The Conjure Woman rests upon African-derived oral trickster myths, legends, and folklore preserved in enslavement culture. Throughout the Conjure tales, Chesnutt uses the supernatural as a metaphor for enslaved people's resistance, survival skills and methods, and for leveling the ground upon which Blacks and Whites struggled within the confines of the enslavement and post-Reconstruction South. Native American and African American oral and written trickster tales give voice to their authors' concerns about the social and political quality of life for themselves and for members of their communities. My dissertation allows these voices a forum from which to "speak."

Texto completo da fonteDrawing from existing literature on price endings and luxury pricing, this thesis highlights the paradox of adopting odd prices (i.e., prices just below a round number) in a luxury context, where, intuitively, prices should mostly be round numbers. Price endings practices are investigated in the product category of luxury women handbags. In a first empirical analysis, different types of price endings and the determinants of their use by pricing managers are identified. In a second part, the results of two experiments measure customers’ perceptions of different facets of luxury and overall perceived luxury itself. In the last part, a conjoint analysis reveals customers’ preferences for odd price endings. The findings of this thesis challenge previous research on price endings practices, in that they show that well-know low-price, low-quality, low-prestige connotations typical of odd prices in a non-luxury context, might not subsist in a luxury environment, at least for the handbag product category and for an intermediate level of luxury

Texto completo da fonteBreast cancer is a very complex and heterogeneous disease. Among the different molecular subtypes, Triple-Negative (TN) breast cancers are particularly aggressive and of poor prognosis. TN tumours are characterized by a lack of estrogen receptors expression (ER), progesterone receptors expression (PR), the absence of Human Epidermal growth factor receptor 2 overexpression (HER2) of the frequent mutations on BRCA1 / 2 genes ("BRCAness" phenotype). In the absence of effective targeted therapies, many targeted therapies including poly-ADP-ribose polymerase inhibitors (anti-PARPs) are currently under development in preclinical and clinical studies. Based on the synthetic lethality concept, the anti-PARPs specifically target the BRCAness properties of TN tumors. In this context, these works were focused on the development of diagnostic tools for the optimization of TN tumours treatment with anti-PARPs. For this, firstly, 3D cell cultures formed with the Liquid Overlay technique as well as associated cytotoxicity tests were developed, from the TN breast cancer cell lines MDA-MB-231 and SUM1315. These two spheroid models were then optimized and standardized in a synthetic culture medium called OPTIPASS (BIOPASS). Secondly, the efficacy of a co-treatment combining anti-PARP1 Olaparib at low and high doses and fractioned radiotherapy (5x2 Gy) was analyzed on the two cell lines MDA-MB-231 and SUM1315 cultured in 2D and 3D conditions. These experiments clearly demonstrated a potentiating effect of Olaparib on radiotherapy (i) in presence of low doses of this anti-PARP (5 μM or inferior) (ii) at long term and (iii) in presence of the maximum fractionation (5x2 Gy). In addition, these two TN cell lines showed a heterogeneous sensitivity to the co-treatment. Thus, an in silico transcriptomic analysis revealed very different profiles of these highly metastatic and highly aggressive cell lines. Notably, the SUM1315 cell line presented a neuronal commitment, suggesting its cerebral metastatic origin. These promising results could open up new perspectives for the treatment of TN tumours brain metastases, which are very common in this subtype. Thirdly, in order to better characterize the mode of action of Olaparib on these spheroid models, a fluorescent derivative of Olaparib, Ola-FL, was synthesized and characterized. The analysis of Ola-FL penetration and distribution in MDA-MB-231 and SUM1315 spheroids showed a rapid and homogeneous distribution of the compound as well as its persistence after 3h of incubation, in all the depth of the spheroids and especially in the central hypoxic zones. Finally, the analysis of the co-expression of two major Multidrug Resistance (MDR) pumps, MRP7 and P-gp after the treatment of the two TN lines with Olaparib, revealed on 2D cultures, a relay type expression of the MRP7 and the P-gp. On spheroids treated with a low dose of Olaparib art long term (10 days), a basal expression of MRP7 and an overexpression of P-gp were detected in the peripheral residual cells of the spheroids. These results clearly highlighted the involvement of these efflux pumps in Olaparib resistance mechanisms, in these aggressive tumors. All the results resulting from the modeling of the action of Olaparib on MDA-MB-231 and SUM1315 spheroids suggest its greater efficacy at low dose and at long-term, especially in the hypoxic zones of the spheroids. This parameter might be probably at the origin of its potentiating effect with radiotherapy

Texto completo da fonteOs objectivos do presente trabalho consistiam na validação estrutural das versões portuguesas da Self-Assessed Wisdom Scale (SAWS; Webster, 2003) e da Rosenberg Self- Esteem Scale (RSES; Rosenberg, 1965) e na validação externa da versão portuguesa da SAWS, através da criação e teste de um Modelo Preditor da Auto-Estima (MPAE) em adultos idosos. Por métodos não probabilísticos, seleccionou-se uma amostra de 240 sujeitos autónomos e voluntários, com um leque etário entre os 65 e os 90 anos (M=72,7), residentes na área da Grande Lisboa e maioritariamente pertencentes ao género feminino (60,8%). Através da modelação de equações estruturais (LISREL 8.53), obteve-se um modelo tetrafactorial hierárquico de 21 itens da SAWS e um modelo unifactorial de sete itens da RSES, ambos bem ajustados e com boas qualidades psicométricas. Por fim, obteve-se um bom ajustamento do MPAE em adultos idosos, em que a sabedoria foi tida como preditora da Auto-Estima (AE), tendo-se concluído que a sabedoria exerce uma influência positiva sobre a AE global.
ABSTRACT: The aims of this work were to validate the portuguese versions of the Self-Assessed Wisdom Scale (SAWS; Webster, 2003) and of the Rosenberg Self-Esteem Scale (RSES; Rosenberg, 1965) and to test the external validity of the Portuguese version of the SAWS, through the creation and test of a Predictor Model of the Self-Esteem (PMSE) in old adults. Using non-probabilistic methods, we selected a sample of 240 autonomous and volunteer subjects, with an age range between 65 and 90 years old (M=72,7), living in Lisbon and mostly women (60,8%). Through structural equation modelling (LISREL 8.53), we obtained a 21-item hierarchical model with four factors of the SAWS and a seven-item model of the RSES, both well adjusted and with good psychometric qualities. At last, we obtained a good adjustment of the PMSE in the elderly, in which wisdom was predicting Self-Esteem (SE). We concluded that wisdom has a positive influence on global SE.
